Characterization and Molecular Targeting of a Mechanosensor Mechanism Controlled By the G-Quadruplex/I-Motif Molecular Switch in the MYC Promoter NHE III₁Sutherland, Caleb Daniel January 2015 (has links)
MYC is overexpressed in most types of tumors, but a means to selectively decrease its expression is yet to be found. Our recent findings on modulation of BCL2 gene expression through protein interactions with the BCL2 i-motif have provided a basis for further investigation of MYC gene control. It is proposed that the MYC i-motif could function by a similar molecular switch mechanism as in BCL2.Binding sites for heterogeneous nuclear ribonucleoprotein K (hnRNP K) within the MYC promoter also exist in the i-motif-forming sequence. Circular dichroism and bromine footprinting confirmed that this DNA sequence is able to form an i-motif, and systematic mutation of the cytosine residues in this sequence has revealed a 5:5:5 loop configuration. Indeed, all loops of the i-motif, when folded into a 5:5:5 loop configuration, contain the hnRNP K consensus sequence (CCCT). Previous studies show that hnRNP K binds to this i-motif-forming sequence, but it was assumed to be single-stranded. Binding studies revealed that hnRNP K has more binding affinity to its consensus sequence in the i-motif compared to a mutant sequence where the i-motif cannot form. Further investigation of the MYC promoter revealed an additional two runs of cytosine seven bases downstream of the MYC i-motif. Biophysical studies showed that the additional two runs were not involved in i-motif formation, however recent studies describe their importance for transcriptional activation. We found that hnRNP K preferred the longer 5CT sequence compared to the i-motif forming 4CT sequence when using a competitive binding assay. Utilizing luciferase reporters containing either the 4CT or 5CT sequence validated that hnRNP K required both the i-motif and 5th CT element for maximum transcriptional activation. Competition binding studies and bromine footprinting showed that hnRNP K bound to the downstream 5th CT element and the central and lateral loops of the i-motif.Additionally, we found that co-overexpression of Sp1 and hnRNP K induced a 10-fold increase in luciferase activity in the 5CT reporter only. We hypothesize that Sp1 continuously primes the promoter to initiate transcription inducing more negative superhelicity and increasing the melting of duplex DNA. This increased melting grants hnRNP K’s three KH domains access to the i-motif loops and the 5Th CT element. Confirmation by ChIP analysis validated that Sp1 overexpression causes an increase in hnRNP K occupancy at the MYC promoter. These findings provide new insight into the mechanisms of MYC transcriptional control by the i-motif and G-quadruplex.Recently, our group has demonstrated that two small molecules IMC-48 and IMC-76 can interact with the i-motif and can be an effective means to modulate BCL2 expression. Based on these results with the BCL2 i-motif, we employed a similar strategy and screened and identified small drug-like molecules that interact with MYC i-motif, using a FRET high-throughput assay. We then further validated that IMC-16 stabilizes the MYC i-motif through the interactions with the loops of the i-motif. No stabilization by IMC-16 treatment was observed with the MYC G-quadruplex and the BCL2 and PDGFRβi-motifs demonstrating selectivity for the MYC i-motif.Finally, we investigated the effects of IMC-16 on MYC expression in three lymphoma cell lines all expressing different levels of MYC. In the case of both Daudi and RAJI Burkitt’s lymphoma cell lines we demonstrated that selectively stabilizing the i-motif by IMC-16 could increase MYC expression. Furthermore, we demonstrated that the MYC G-quadruplex stabilizing compound GQC-05 and IMC-16, which stabilizes the MYC i-motif, have antagonistic effects on MYC expression, providing further evidence of a molecular switch mechanism in the NHEIII1. Directly targeting MYC expression through the i-motif offers advantages over targeting the G-quadruplex, because of the reduced stability and dynamic nature of the i-motif, additionally the i-motif is only found in DNA. The use of such i-motif interactive compounds is the first step into the development of new innovative approaches to treat cancers.

Hypertension et régulation de l'expression moléculaire de l'angiotensinogène par la ribonucléoprotéine hétérogène nucléaire KAbdo, Shaaban 06 1900 (has links)
Le diabète est une maladie chronique dont la principale
caractéristique est un niveau plasmatique élevé de glucose, qui est causé
soit par un défaut dans la production d’insuline, l’action de l’insuline, ou
les deux à la fois. Plusieurs études ont démontré que l’hyperglycémie
chronique peut mener à la dysfonction et même la défaillance de
plusieurs organes, dont le coeur, le système vasculaire, les yeux et les
reins, se traduisant par des infarctus du myocarde, des accidents
cérébro-vasculaires et des complications rétinales et rénales,
respectivement. La néphropathie diabétique (DN) est la principale cause
de déficience rénale et affecte près de 25-40% des patients diabétiques.
La DN est invariablement associée à un risque élevé d’accident cérébrovasculaire
et de dysfonction cardivasculaire. L’angiotensinogène (Agt) est
l’unique précurseur de tous les types d’angiotensines. En plus du
système rénine-angiotensine (RAS) sytémique, le rein possède son
propre système intrarénal et exprime tous les composants du RAS. L’Agt
est fortement exprimé dans les cellules du tubule proximal rénal (RPTC)
et y est converti en angiotensine II (AngII), le peptide biologiquement actif
du RAS. Les patients diabétiques présentent de hauts niveaux d’AngII et
une augmentation de l’expression des gènes du RAS, suggérant que
l’activation du RAS intrarénal joue un rôle important dans la progression
de la DN. Les mécanismes qui contrôlent la régulation du niveau rénal
d’Agt par l’hyperglycémie et l’insuline demeurent mal compris.
Le but global de cette thèse est de mieux comprendre les
mécanismes moléculaires qui contrôlent l’expression du gène Agt chez la
souris Akita (un modèle murin de diabète de type 1). Dans cette optique,
la première partie de la thèse se concentre sur deux facteurs de
transcription de la famille des ribonucléoprotéines nucléaires hétérogènes
(hnRNP). Chan et collaborateurs ont déjà identifié 2 protéines nucléaires
hnRNP F et hnRNP K, de 48kD et 70kD respectivement. HnRNP F et
hnRNP K forment un hétérodimère et se lient à l’élément de réponse à
l’insuline (IRE) présent dans le promoteur du gène Agt du rat et inhibent
la transcription du gène Agt in vitro. Afin de déterminer si hnRNP F / K
sont responsables de l’inhibition de l’expression rénale de Agt par
l’insuline in vivo, nous avons étudié des souris Akita males traités ou non
avec des implants d’insuline pour une période de 4 semaines. Des souris
non-Akita males ont été employées comme contrôles. Les souris Akita
développent de l’hypertension et de l’hypertrophie rénale. Le traitement à
l’insuline rétablit les niveaux de glucose plasmatiques et la pression
systolique (SBP), et atténue l’hypertrophie rénale, l’albuminurie (ratio
albumine/créatinine urinaire, ACR) et les niveaux urinaires d’Agt et AngII
chez les souris Akita. De plus, le traitement à l’insuline inhibe l’expression
rénale du gène Agt, tout en augmentant l’expression des gènes hnRNP
F, hnRNP K et ACE2 (enzyme de conversion de l’angiotensine-2). Dans
des RPTC in vitro, l’insuline inhibe Agt, mais stimule l’expression de
hnRNP F et hnRNP K en présence de hautes concentrations de glucose,
et ce via la voie de signalisation MAPK p44/42 (protéine kinase activée
par un mitogène). La transfection avec des petits ARN interférents
(siRNA) contre hnRNP F et hnRNP K prévient l’inhibition de l’expression
d’Agt par l’insuline dans les RPTC. Cette étude démontre bien que
l’insuline prévient l’hypertension et atténue les dommages rénaux
observés chez les souris Akita diabétiques, en partie grâce à la
suppression de la transcription rénale de Agt, via une augmentation de
l’expression de hnRNP F et hnRNP K.
La seconde partie de cette thèse change de focus et se tourne
vers le facteur Nrf2 (nuclear factor erythroid 2-related factor 2). Nrf2 est
un facteur de transcription qui contrôle les gènes de la réponse
antioxydante cellulaire en réponse au stress oxydant ou aux
électrophiles. Le but de cette étude est d’examiner l’impact de la
surexpression de la catalase (Cat) dans les RPTC sur l’expression du
gène Agt via Nrf2 et sur le développement de l’hypertension et des
dommages rénaux résultants chez les souris diabétiques Akita
transgéniques (Tg). Nos études ont démontré que la surexpression de
Cat dans les souris Akita Cat-Tg normalise la SBP, atténue les
dommages rénaux et inhibe l’expression des gènes Nrf2 et Agt dans les
RPTC. In vitro, le glucose élevé (HG) et l’oltipraz (un activateur de Nrf2)
stimulent l’expression de Nrf2 et Agt, et cet effet peut être bloqué par la
trigonelline (inhibiteur de Nrf2), des siRNA contre Nrf2, des antioxydants
ou des inhibiteurs pharmacologiques NF-κB et MAPK p38. La
suppression de sites de réponse à Nrf2 présents dans le promoteur du
gène Agt du rat abolit la stimulation par l’oltipraz. Finalement, des souris
males adultes non-transgéniques traitées avec l’oltipraz montrent une
augmentation de l’expression de Nrf2 et Agt dans leurs RPTC et cette
augmentation peut être normalisée par la trigonelline. Ces données
permettent d’identifier un nouveau mécanisme d’action de Nrf2, par la
stimulation du gène Agt intrarénal et l’activation du RAS, qui induisent
l’hypertension et les dommages rénaux par le glucose élevé et les
espèces réactives de l’oxygène chez les souris diabétiques. Nos
conclusions permettent de démontrer que l’insuline induit l’expression de
hnRNP F et hnRNP K, qui jouent ensuite un rôle protecteur en prévenant
l’hypertension. La surexpression de la catalase dans les RPTC vient
quant à elle atténuer l’activation de Nrf2 et ainsi réduit la SBP chez les
souris Akita. / Diabetes mellitus is a chronic metabolic disorder characterized by
high plasma glucose caused by an impairment of insulin production,
insulin action or both. Accumulating evidence has shown that chronic
hyperglycemia can lead to dysfunction and failure of multiple organs
including the heart, vascular system, eyes, and kidneys resulting in
myocardial infarction, stroke, and retinal and renal complications,
respectively. Diabetic nephropathy (DN) is the leading cause of end-stage
renal disease affecting approximately 25–40% of diabetic patients. DN is
invariably associated with an increased risk of stroke and cardiovascular
dysfunction. Angiotensinogen (Agt) is the sole precursor for all types of
angiotensins. In addition to systemic renin-angiotensin system (RAS), all
the components of the intrarenal RAS are expressed in the kidney. Agt is
highly expressed in the renal proximal tubular cells (RPTCs) and
converted into biologically active angiotensin II (Ang II). In Diabetics,
intrarenal Ang II level and RAS gene expression are upregulated,
suggesting that intrarenal RAS activation plays an important role in the
progression of DN. The mechanism (s) underlying the regulation of renal
Agt by hyperglycemia and insulin are not completely understood. The
overall aim of this thesis is to understand the molecular mechanism(s)
that regulate renal Agt gene expression in an Akita mouse (a mouse
model of type 1 diabetes). For this purpose, the first part of this thesis
focuses on two transcription factors from the heterogenous nuclear
ribonucleoprotein (hnRNPs) family. Previously, Chan’s group identified
two nuclear proteins hnRNP F and hnRNP K of 48kD and 70kD,
respectively. hnRNP F and hnRNP K form a heterodimer and bind to the
insulin-responsive element (IRE) in the rat Agt gene promoter inhibiting
Agt gene transcription in vitro. To determine whether hnRNP F / K
mediate insulin inhibition of renal Agt expression in vivo, we used adult
male Akita mice treated ± insulin implants for 4 weeks. Non-Akita mice
served as controls. The Akita mice developed hypertension and exhibited
renal hypertrophy. Insulin treatment normalized plasma glucose levels
and systolic blood pressure (SBP), attenuated renal hypertrophy,
decreased urinary albumin/creatinine ratio (ACR) and urinary Agt and
Ang II levels in Akita mice. Furthermore, insulin treatment inhibited renal
Agt expression but enhanced hnRNP F, hnRNP K and angiotensinconverting
enzyme-2 (ACE2) expression. In vitro, insulin inhibited Agt but
stimulated hnRNP F and hnRNP K expression in high-glucose media via
p44/42 mitogen-activated protein kinase signaling in RPTCs. Transfection
with hnRNP F and hnRNP K small interfering RNAs (siRNA) prevented
the insulin inhibition of Agt expression in RPTCs. This study
demonstrates that insulin prevents hypertension and attenuates kidney
injury, at least in part, through suppressing renal Agt transcription via
upregulation of hnRNP F and hnRNP K expression in diabetic Akita mice.
In the second part of the thesis we focused on the nuclear factor
erythroid 2-related factor 2 (Nrf2). Nrf2 is a transcription factor that
regulates cellular antioxidant gene defense against oxidative stress or
electrophiles. The purpose of this study is to investigate the impact of the
overexpressing catalase (Cat) in RPTCs on Agt gene expression via
Nrf2and the resulting effects on the development of hypertension and
renal injury in diabetic Akita transgenic (Tg) mice. Our studies
demonstrate that Cat overexpression normalizes SBP, attenuates renal
injury, and inhibits RPTC Nrf2 and Agt gene expression in the Akita Cat-
Tg compared to Akita mice. In vitro, high glucose (HG) and Oltipraz
stimulated Nrf2 and Agt gene expression; these changes were blocked by
Trigonelline (an inhibitor of Nrf2), siRNA against Nrf2, antioxidants, or
pharmacological inhibitors of NF-kB and p38 mitogen-activated protein
kinase. Moreover, deletion of Nrf2-responsive elements in the rat Agt
gene promoter abolishes the stimulatory effect of Oltipraz. Finally,non
transgenic adult male mice treated with the Nrf2 activator Oltipraz,
upregulated Nrf2 and Agt expression in mouse RPTs, an effect that was
normalized by Trigonelline. These data identify a novel mechanism via
which Nrf2 mediates the stimulation of intrarenal Agt gene expression and
activates the RAS through whichHG/reactive oxygen species (ROS)
induce hypertension and renal injury in diabetic mice. Our findings
demonstrate that the insulin induced hnRNP F and hnRNP K gene
expression play a protective role in the preventing hypertension. Catalase
overexpression, in RPT's, attenuates Nrf2 activation and lowers the SBP
in Akita mice.

Recherche de partenaires potentiels de la protéine Damaged-DNA Binding 2 dans la régulation de l’expression génique : le cas des heterogeneous ribonucleoprotein K et J dans la régulation du gène NFKBIA / Search for potential partners of Damaged-DNA Binding 2 protein in the regulation of gene expression : the case of heterogeneous ribonucleoprotein K and J in the regulation of NFKBIA geneDrouot, Guillaume 16 November 2018 (has links)
Le laboratoire a identifié récemment la protéine DDB2 comme ayant une activité dans la régulation de l’expression de gènes cibles tels que SOD2, BCL2 et NFKBIA, conférant ainsi à cette protéine un rôle dans le contrôle de la progression métastatique et dans la réponse thérapeutique des cellules tumorales mammaires. Cependant, l’activité réelle de DDB2 dans la transcription génique reste à définir, car sa structure ne permet pas d’expliquer son influence sur l’expression de ses gènes cibles. Cette activité peut être soit inhibitrice comme pour SOD2 et BCL2, soit activatrice comme NFKBIA qui code la protéine IκBα, suggérant que DDB2 doit s’associer avec des inhibiteurs ou des activateurs de la transcription génique, ou en favoriser le recrutement. Afin d’identifier ces partenaires potentiels, susceptibles de participer à son activité transcriptionnelle, nous avons développé une approche de « DNA pull-down », couplée à une analyse protéomique globale par spectrométrie de masse à partir des cellules tumorales mammaires MCF-7 surexprimant naturellement DDB2. Nous avons révélé la présence du complexe UV-DDB (DDB2, DDB1 et Cul4A) sur le promoteur des gènes SOD2 et NFKBIA. Nous avons également mis en évidence que ce complexe, connu pour participer aux premières étapes de la réparation de l’ADN lésé par des rayonnements UV, favorise le recrutement de l’histone acétyl transférase p300 sur le promoteur du gène NFKBIA, expliquant en partie le rôle activateur de DDB2 sur ce gène cible. Notre analyse protéomique à révéler, avec un score élevé, la présence des protéines hnRNP K et J sur le promoteur du gène NFKBIA et d’une manière indépendante de toute interaction physique avec DDB2. La forme J, très peu décrite, présente une affinité plus grande pour le promoteur du gène NFKBIA que la forme K. De plus, nous l’avons observée strictement nucléaire et liée à la chromatine. De manière intéressante, nous montrons que la forme J est surexprimée dans le noyau des cellules tumorales MDA-MB231 métastatiques, comparativement aux cellules T47D non métastatiques. Par la suite, nous avons évalué l’importance des hnRNP K et J dans la transcription du gène NFKBIA par rapport à DDB2 et un régulateur bien décrit tel que le facteur de transcription Sp1. Nos résultats indiquent que les protéines hnRNP K et J, lorsqu’elles sont surexprimées, jouent un rôle de répresseur du gène NFKBIA et ce même en présence des activateurs DDB2 et Sp1. L’ensemble de ce travail a contribué à montrer la présence de protéines, pouvant participer à l’activité transcriptionnelle de DDB2, telles que le complexe UV-DDB et p300. En dehors de tout partenariat avec DDB2, il a été mis en évidence une relation entre les hnRNP K et J et l’activité constitutive de NF-κB, en particulier avec la forme J, qui, par son expression corrélée à l’agressivité des cellules tumorales mammaires, présente un intérêt clinique potentiel en tant que marqueur prédictif de la progression métastatique, tout comme DDB2 / The laboratory has recently identified the DDB2 (Damaged-DNA Binding 2) protein as a regulator of target gene expression like SOD2, BCL2 and NFKBIA, thus conferring to this protein a role in control of metastatic progression and therapeutic response of breast cancer cells. However, the real activity of DDB2 in gene transcription remains to be defined because its structure cannot entirely explain its influence on target gene expression. This protein can act either as an inhibitor like for the SOD2 and BCL2 genes or as an enhancer like for the NFKBIA gene, encoding IκBα protein. This suggests that DDB2 must associate with, or promote recruitment, of inhibitors or activators of gene transcription. In order to search and identify potential partners that could participate in its transcriptional activity, we developed a “DNA pull-down” approach associated with a global proteomic analysis by mass spectrometry from MCF-7 breast cancer cells overexpressing naturally DDB2. With this approach, we reveal the presence of the UV-DDB complex composed by DDB2, DDB1 and Cullin 4A proteins on the SOD2 and NFKBIA gene promoters. We also highlighted that this complex, known for its role in first steps of UV-induced DNA lesion repair, promotes the recruitment of the p300 histone acetyl transferase on the NFKBIA gene promoter, which may explain, in part, the enhancer activity of DDB2 on this target gene. The proteomic analysis from the “DNA pull-down” also reveals, with originality, the presence of heterogeneous ribonucleoproteins K and J (hnRNP K and J) on the NFKBIA gene promoter with a high recovery score among many other proteins and independently of any physical interaction with DDB2. The J form, very poorly described, shows a higher affinity for NFKBIA gene promoter than the K form. Furthermore, we observed that the J form is strictly nuclear and mostly bound to chromatin, while the K form is also found in cytoplasm. Interestingly, we show that the J form is overexpressed in nucleus of metastatic breast cancer MDA-MB231 cells by comparison with non-metastatic breast cancer T47D cells. Then, we evaluated the importance of hnRNP K and J proteins in NFKBIA gene transcription compared with DDB2 and with a well-known regulator, the Sp1 transcription factor. Our results show that hnRNP K and J proteins, when they are overexpressed, play a repressor role on NFKBIA gene expression by binding on its promoter even in presence of DDB2 and Sp1 activators. Taken together, these data show that some proteins could participate in DDB2 transcriptional activity, like the UV-DDB complex and the p300 protein. Outside of any interaction with DDB2, this work highlights a relationship between the hnRNP K and J proteins, and NF-κB constitutive activity, especially with the J form. This latter has an expression correlated with aggressiveness of breast cancer cells and a potential clinical interest as a predictive marker of metastatic progression, like DDB2
