Regulation of Pancreatic α and β Cell Function by the Bile Acid Receptor TGR5

The discovery that bile acids act as endogenous ligands of the membrane receptor TGR5 and the nuclear receptor FXR increased their significance as regulators of cholesterol, glucose and energy metabolism. Activation of TGR5, expressed on enteroendocrine L cells, by bile acids caused secretion of GLP-1, which stimulates insulin secretion from pancreatic β cells. Expression of TGR5 on pancreatic islet cells and the direct effect of bile acids on the endocrine functions of pancreas, however, are not fully understood. The aim of this study was to identify expression of TGR5 in pancreatic islet cells and determine the effect of bile acids on insulin secretion. Expression of TGR5 was identified by quantitative PCR and western blot in islets from human and mouse, and in α (αTC1-6) and β (MIN6) cells. Release of insulin, glucagon and GLP-1 were measured by ELISA. The signaling pathways coupled to TGR5 activation were identified by direct measurements such as stimulation of G proteins, adenylyl cyclase activity, PI hydrolysis and intracellular Ca2+ in response to bile acids; and confirmed by the use of selective inhibitors that block specific steps in the signaling pathway. Our studies identified expression of TGR5 receptors in β cells and demonstrated that activation of these receptors by both pharmacological ligands (oleanolic acid (OA) and INT-777) and physiological ligand (lithocholic acid, LCA) induced insulin secretion. TGR5 receptors are also expressed in α cells and, activation of TGR5 by OA, INT-777 and LCA at 5 mM glucose induced release of glucagon, which is processed from proglucagon by the selective expression of prohormone convertase 2 (PC2). However, under hyperglycemia, activation of TGR5 in α cells augmented the glucose-induced increase in GLP-1 secretion, which in turn, stimulated insulin secretion. Secretion of GLP-1 from α cells reflected TGR5-mediated increase in PC1 promoter activity and PC1 expression, which selectively converts proglucagon to GLP-1. The signaling pathway activated by TGR5 to mediate insulin and GLP-1 secretion involved Gs/cAMP/Epac/PLC-ε/Ca2+. These results provide insights into the mechanisms involved in the regulation of pancreatic α and β cell function by bile acids and may lead to new therapeutic avenues for the treatment of diabetes.

L'objectif de la cosmologie observationnelle est d'utiliser les informations auxquelles les télescopes modernes nous donnent accès afin de connaître à la fois l'histoire, le contenu matériel et la géométrie de notre Univers. L'histoire de l'Univers nous renseigne sur la suite d'événements qui ont conduit celui-ci, à partir d'une soupe de particules homogènes et chaudes, à avoir aujourd'hui l'aspect qu'on lui connaît, c'est-à-dire en expansion et avec une structuration de la matière en agrégats plus ou moins gros selon l'échelle à laquelle on regarde. Il y a les étoiles et planètes à notre échelle, avec leur stupéfiante complexité biologique. A plus grande échelle, on trouve les galaxies, composées de plusieurs milliards d'étoiles organisées tantôt en forme spirale, tantôt en forme de gros ballons de rugby. Ensuite viennent les amas et les superamas de galaxies qui s'organisent dans l'espace sous la forme de gigantesques éponges avec de grandes zones vides séparant des régions denses. L'histoire de l'Univers tente donc de répondre à la question : comment en est-on arrivé là ? Cette question métaphysique en appelle aussitôt une autre : qu'arrivera-t-il après ? En cosmologie, cette question est liée au contenu et à la géométrie de l'Univers. La notion de géométrie de l'Univers, héritée de la théorie de la Relativité Générale d'A. Einstein, intègre la courbure spatio-temporelle de l'Univers dans son ensemble. Cette dernière est déterminée par la quantité et le type de matière qu'Il contient. On dit la géométrie ouverte si la courbure est négative, ce qui correspond à un Univers contenant peu de matière (par rapport à la densité critique qui est de quelques protons par m3). L'Univers est alors infini dans le temps et l'espace, son avenir n'est qu'expansion, dilution et refroidissement éternels. Si l'Univers contient juste assez de matière pour avoir la densité critique, on le dit plat puisque sa courbure est nulle, dans ce cas aussi, il est infini dans le temps et l'espace, avec un avenir similaire au cas ouvert. En revanche, si l'Univers est fermé, c'est à dire s'il contient suffisamment de matière pour que l'effet gravitationnel de cette dernière soit suffisant pour freiner son expansion, il finira par se recontracter, et, comme un film tournant à l'envers les galaxies se rapprocheront les unes des autres, la densité augmentera indéfiniment pour atteindre un big-crunch final similaire au big-bang initial. Connaître la géométrie de l'Univers est donc l'une des questions les plus essentielles de la cosmologie.

En conptant sur eux, nous avons d?velopp? un atelier corps biographique, dans lequel nous avons r?alis? 10 v?cus dans la p?riode qui a couvert le deuxi?me semestre de 2007 jusqu au deuxi?me semestre de 2008, en manifestant la r?flexivit? autobiographique, le ludique, la cr?ativit?, la sensibilit? et la r?flexivit? du v?cu. Nous avons utilis? comme instruments pour la construction des donn?s, l observation participante, les propres v?cus, le port folio, la technique du jeu de sable et l'enregistrement photographique. L analyse a montr? que le v?cu de la formation humaine ? partir des principes de la corpor?it?, implique dans un parcours vers soi-m?me dans lequel ?mergent les exp?riences qui nous ont constitu?s en tant qu ?tres existentiels. Pour finaliser, nous avons aper?u les r?percussions dans la vie professionnelle et personnelle de ces institutrices, qui comme moi pensaient avoir besoin d exposer leur luminosit? int?rieure pour que l on la red?couvre. Com elas, foi desenvolvido um ateli? corpogr?fico, no qual realizamos 10 viv?ncias corpogr?ficas, do segundo semestre de 2007 at? o segundo semestre de 2008. Durante essas viv?ncias foi evidenciada a reflexividade autobiogr?fica, a ludicidade, a criatividade, a sensibilidade e a reflexividade vivencial. Os instrumentos utilizados para a constru??o dos dados foram a observa??o participante, as pr?prias viv?ncias, o portf?lio, e o sandplay (t?cnica do Jogo de Areia) e o registro fotogr?fico. A an?lise mostrou que vivenciar a forma??o humana a partir dos princ?pios da corporeidade implica uma caminhada para si mesmo, em que emergem as experi?ncias que nos constitu?ram como seres existenciais. Ao final da pesquisa, foi poss?vel observar as repercuss?es na vida profissional e pessoal dessas docentes que, assim como eu pensava, possu?am uma luz interior que precisava ser redescoberta. Ao vivenciarem as tessituras, as docentes se mostraram implicadas, experienciando momentos de brincadeira, cria??o, frui??o, afeto e de a??o reflexiva, durante os quais se revelou a ess?ncia humana escondida, abrindo-se possibilidades para o sentir da humanesc?ncia. Por fim, foram revelados corpos brincantes de sabedoria e beleza, que, no encontro consigo mesmos e com o outro, puderam ser poieticamente corpografados

Para isto, foram utilizadas ferramentas metodol?gicas como revis?o bibliogr?fica e fonogr?fica, reuni?es, entrevista e an?lise da partitura final, visando ? performance. Desta Pesquisa resultaram: uma proposta interpretativa constru?da em contato direto com o compositor - que pode servir como primeiro par?metro para o in?cio de uma tradi??o de interpreta??o; uma edi??o da partitura com dedilhados e arcadas; e a estreia da Pe?a com registro audiovisual. A an?lise dos dados sobre a intera??o compositor-int?rprete permitiu concluir que o processo colaborativo foi ben?fico para ambas as partes: por um lado, houve melhoria na escrita da Obra, que se tornou mais idiom?tica; por outro, o contato direto com o compositor trouxe importante contribui??o ? constru??o da proposta interpretativa. A revis?o bibliogr?fica, a an?lise da partitura e a entrevista possibilitaram estabelecer que Danilo Guanais utiliza a est?tica Armorial de maneira pessoal, aliando-a a t?cnicas contempor?neas de composi??o, com frequente combina??o de modalismo, tonalismo e serialismo. / This work presents an artistic research on the interpretation of O Caldeir?o dos esquecidos for solo violin by Danilo Guanais, written in northeastern brazilian musical style, called Armorial. It also reports on the collaboration between performer and composer during the piece's composition and it?s preparation for performance. Methodological tools as bibliographic and phonographic review, face-to-face meetings, interview and score analysis were used. From this research resulted: an interpretation of the piece, developed in direct contact with the composer - that may become a first parameter, beginning an interpretation tradition; the score edition with bowings and fingerings; the premi?re and a record of the piece. Data analysis on the composer-performer collaboration concluded that the collaborative process was beneficial for both parties: on one hand, the composition was improved and became more idiomatic; on the other hand, the direct contact with the composer contributed substantially to the construction of the piece?s interpretation. Bibliographic review, score analysis, and interview showed that Danilo Guanais uses Armorial aesthetics in a personal way, combining it with contemporary techniques of composition. Modalism, tonalism and serialism combination is often used.

Structural characterization of JIP3 recruitment by Kinesin-1 / Caractérisation structurale du recrutement de JIP3 par la Kinésine-1

Le transport intracellulaire de cargos est un processus critique au sein des cellules eucaryotes, et notamment au niveau des neurones, pour contrôler différentes fonctions dont la maturation et la transmission synaptique. La kinésine-1 est un moteur moléculaire capable de transporter différents types de cargos, comme des organelles, des vésicules ou des assemblages macromoléculaires le long des microtubules. La kinésine-1 est un hétérotétramère constitué d’un homodimère de chaînes lourdes (KHC) associé à deux chaînes légères (KLC) ; les deux chaînes, KHC et KLC étant capables de recruter des cargos. L’un des premiers cargos de la kinésine-1 à avoir été identifiés sont les protéines JIP3/4 (JNK-Interacting Protein 3/4) ; elles jouent aussi un rôle de protéines adaptatrices pour le transport d’autres cargos de la kinésine-1. La kinésine-1 recrute les protéines JIP3/4 de deux façons distinctes et indépendantes (i) via KHC et (ii) via KLC. Le recrutement de JIP3/4 par KHC et KLC est capable, via des mécanismes moléculaires distincts, d’activer la motilité de la kinésine-1 et donc de contrôler le transport intracellulaire dans lequel elle est impliquée et les fonctions associées au sein des neurones.Au cours de mon travail de thèse, j’ai contribué à caractérisé par des approches bio-informatiques, biochimiques/biophysiques et structurales, les deux modes de recrutement des protéines JIP3/4 par la kinésine-1 : (i) via KHC et (ii) via KLC. Ce travail a permis d’apporter des nouveaux éléments pour comprendre le mode de recrutement de ces protéines cargos/adaptatrices par la kinésine-1, mais aussi de mieux comprendre les mécanismes moléculaires de son activation par les protéines JIP3/4. / The intracellular transport of cargos is a crucial process on eukaryotic cells, and notably in neurons, in order to regulate different functions as cell’s maturation and synaptic transmission. The Kinesin-1 is a molecular motor capable of transporting different types of cargos as organelles, vesicles and macromolecular assemblies along the microtubules. It is a heterotetramer composed by a homodimer of heavy chains (KHC) bound to two light chains (KLC), where both KHC and KLC are capable of cargos recruitment. One of the first identified cargos of Kinesin-1 is JIP3/4 (JNK-Interacting Protein 3/4), which are also adaptor proteins, intermediating the transport of other cargos. Kinesin-1 recruits JIP3/4 by two different and independent modes, (i) via KHC and (ii) via KLC. Therefore, JIP3/4 recruitment by KHC and KLC activates the motility of Kinesin-1, by distinct mechanisms, allowing the intracellular transport of cargos and the associated functions in neurons. During my PhD, I contributed to the characterization of the dual binding mode of Kinesin-1 and JIP3/4 by bioinformatical, biochemical/biophysical and structural approaches. This work allowed a better understanding of the cargos’ recruitment by Kinesin-1, as well as the molecular mechanisms of Kinesin-1 activation by JIP3/4.

According to the Law Lei de Diretrizes e Bases para a Educa??o Nacional, Lei n? 9.394 de 20 de dezembro de 1996 special education is described as a teaching form available since Child Education to Higher Education. It is constituted by material and human resources used for the guarantee of the access condition of people with studying disability. As what concerns the education of the deaf student user of Brazilian Sign Language (Libras), the presence of proficient professionals in Brazilian Sign Language is fundamental in the classroom and other educational spaces as part of the specialized support. As a result of this reality, it is seen the rise of the Libras interpreter as a professional whose education path is recent, strongly influent factor in their work conditions. The theoretical reference which gives this study its fundamentals is the critical history, where it is understood that the education and the education of the deaf person is a historical and cultural process, dated in time and social place (VIGOTSKI, 1997). In a particular way, the referent analysis to the Libras Interpreter education path occurs through the observation of the position occupied by this individual in society and their cultural and material conditions of education (BOURDIEU, 1975). As a theoretical-methodological approach, uses the qualitative research for the investigation of a field that involves human and social sciences (LOWY, 1985; GIL, 1999). The methodology used to build the data is a questionnaire composed by forty one questions and a space for comment available in a website during five months, accessed by a hundred and thirty one visitors, among them the twenty nine interpreters participant of this research. The analysis of the data considers three theme axes: education paths, working conditions and social economical reality. The results show that the education of these interpreters occurred from elements taken from the interaction with values of the deaf community, appropriation of the scientific knowledge and working relations. / A presente disserta??o tem como objetivo estudar as trajet?rias de forma??o e as condi??es de trabalho do int?rprete de Libras certificado pelo exame nacional de profici?ncia para interpreta??o de Libras, que atua em Institui??es de Educa??o Superior. De acordo com a Lei de Diretrizes e Bases para a Educa??o Nacional, Lei n? 9.394 de 20 de dezembro de 1996, a educa??o especial caracteriza-se como uma modalidade de ensino dispon?vel desde a Educa??o Infantil at? a Educa??o Superior. La propulsion a��robie �� grande vitesse est depuis longtemps identifi��e comme l'un des prochains sauts technologiques �� franchir dans le domaine des lanceurs spatiaux. Cependant, les v��hicules hypersoniques (HSV) fonctionnant dans des domaines de vitesse extr��mement ��lev��es, de nombreuses contraintes et incertitudes entravent les garanties des propri��t��s des contr��leurs. L'objet de cette th��se est d'��tudier la synth��se de commande d'un tel v��hicule.Pour commencer, il s'agit de d��finir un mod��le repr��sentatif d'un HSV exploitable pour la commande. Dans ce travail, nous construisons deux mod��les de HSV. Un pour la simulation en boucle ferm��e, et le second afin de poser pr��cis��ment le probl��me de commande.Nous proposons ensuite une synth��se de commande de la dynamique longitudinale dans le plan vertical de sym��trie. Celle-ci est robuste aux incertitudes de mod��lisation, tol��rante �� des saturations, et n'excite pas les dynamiques rapides n��glig��es. Ses propri��t��s sont ��valu��es sur diff��rents cas de simulation. Puis, une extension est propos��e afin de r��soudre le probl��me de commande simultan��e des dynamiques longitudinale et lat��rale, sous les m��mes contraintes.Ce r��sultat est obtenu par une assignation de fonction de Lyapunov, suite �� une ��tude des dynamiques longitudinale et lat��rale. Par ailleurs, pour traiter les erreurs de poursuite dues aux incertitudes de mod��lisation, nous nous int��ressons au probl��me de r��gulation asymptotique robuste par retour d'��tat. Nous montrons que cette r��gulation peut ��tre accomplie en stabilisant le syst��me augment�� d'un int��grateur de la sortie. Ceci constitue une extension de la structure de contr��le proportionnel-int��gral au cas des syst��mes non lin��aires.

Para isto, durante a fase do projeto, idealizamos a feitura de uma obra especialmente para materializar os interesses da presente pesquisa, onde foi determinada apenas a instrumenta??o a ser utilizada. Foi sugerida desde o pr?-projeto, a forma??o de quinteto de trompetes, utilizando os principais instrumentos da fam?lia (em Si?, D?, Mi?, piccolo, e fluguelhorn). A metodologia desenvolvida para esta pesquisa se deu por meio do cumprimento de uma agenda de encontros previamente elaborada, constitu?da de uma s?rie de etapas e organizadas em duas fases, onde abordamos a intera??o desde os primeiros impulsos criativos do compositor e sua desenvoltura criativa. Apresentamos tamb?m, como se deu a prepara??o de excertos da obra no decorrer de ensaios-experimentos, bem como a constru??o de uma interpreta??o e de resultados/consequ?ncias provenientes dessa intera??o. / The present work deals with the report on the interaction interpreter-composer, lined with individual biases - mediated by the interpreter-researcher Rudson Ricelli Lima da Silva - and collectives - of the composer Samuel Cavalcanti Correia with the group Trompetearte of EMUFRN. For this, during the project phase, we idealized the making of a work especially to materialize the interests of the present research, where only the instrumentation to be used was determined. From the pre-project, the trumpet quintet formation was suggested, using the main instruments of the family (in Si?, D?, Mi?, piccolo, and fluguelhorn). The methodology developed for this research was accomplished through the fulfillment of an agenda of meetings previously elaborated, constituted of a series of stages and organized in two phases, where we approach the interaction from the first creative impulses of the composer and his creative resourcefulness. We also present the preparation of excerpts from the work in the course of trial-experiments, as well as the construction of an interpretation and results / consequences from this interaction.
