Développement de biocapteurs ampérométriques pour la détermination de l’activité de la transcétolase et pour la détection d’inhibiteurs de cette enzyme / Development of amperometric biosensors for the determination of the activity of transketolase and for the detection of inhibitors of this enzyme

Depuis peu, des travaux ont montré que chez l’Homme, la transcétolase (TK, EC 2.2.1.1.) dont le cofacteur est la thiamine diphosphate (forme active de la vitamine B1), est une enzyme impliquée dans de nombreuses maladies telles que, le diabète, certains cancers, ou encore des maladies neurologiques, comme le syndrome de Wernicke-Korsakoff et la maladie d’Alzheimer. Pour des applications thérapeutiques, des inhibiteurs spécifiques de cette enzyme sont actuellement conçus et synthétisés dans les milieux académiques et industriels. Afin de déterminer l’activité de la TK (dans un but de diagnostic) d’une part, et de détecter des inhibiteurs potentiels de cette enzyme (dans un but thérapeutique) d’autre part, il est nécessaire de disposer de tests alliant rapidité, sensibilité et faible coût. Nous avons envisagé d’utiliser des biocapteurs ampérométriques qui combinent l’ensemble de ces avantages, et qui, de plus, n’ont jamais été mis en oeuvre avec la TK. Pour la détermination de l’activité des TK d’E. coli et humaine libres en solution, nous avons tout d’abord élaboré un premier biocapteur à galactose oxydase (GAOx, EC 1.1.3.9), dans lequel cette enzyme est immobilisée sur la laponite. Puis, dans le but de detecter des inhibiteurs de la TK, avec un système réutilisable, nous avons developpé un biocapteur à GAOx-TK d’E. coli, les deux enzymes étant co-immobilisées à la surface de l’électrode. Pour cela la TK a été immobilisée dans des Hydroxydes Doubles Lamellaires (HDL). Ce biocapteur bicouche et bi-enzymatique GAOx-TK, nous a permis d’évaluer l’effet d’inhibiteurs, tels que différents analogues du cofacteur et de substrats pris comme modèles. / Some recent studies have shown that human transketolase (TK, EC 2.2.1.1.), which thiamine diphosphate (active form of vitamin B1) is the cofactor, is involved in numerous disease such as diabete, some cancers and neurodegenerative diseases as Alzheimer’s disease and Wernicke-Korsakoff syndrome. For therapeutic purposes, TK inhibitors have been designed and synthesized in both academic and industrial fields. To determine TK activity (diagnostic) on the one hand, and to detect potential inhibitors of this enzyme (therapeutic) on the other hand, it is necessary to develop fast, sensitive and low cost assays. In this context, we designed some original amperometric biosensors that combine these advantages and were never studied with TK from now. We performed a first galactose oxidase (GAOx, EC 1.1.3.9) biosensor for E. coli and human TK activities detection. For that purpose, GAOx was immobilized on laponite matrix. Then, we designed a GAOx-TK biosensor by co-immobilization of GAOX and TK on the electrode surface that enabled the detection TK inhibitors with a reusable system. Thence, TK was immobilized in Layered Double Hydroxides (HDL). This bilayer and bi-enzymic biosensors, allowed us to determine the inhibitor potencies of several cofactors and substrates analogues as model compounds.

Intercalation Of Alkyl Surfactants In Layered Double Hydroxides : The Anchored Bilayer In Dispersions And The Condensed Phase

Bilayers formed by molecules that possess long alkyl hydrophobic tails are ubiquitous in the natural world manifesting both in biological systems as well as in chemistry. The lipid bilayer is an integral feature of cell membranes of living systems with functions that are of critical importance to the life of the cell. Long chain amphiphilic surfactant molecules can be introduced within the interlamellar region of layered inorganic host lattices to form anchored alkyl chainbilayerswithinthegalleries.Theintercalatedbilayerbearsastriking resemblance to lipid bilayers. However, unlike lipid bilayers where individual molecules can undergo lateral diffusion and also flip-flop between layers the anchored bilayer is characterized by the total absence of translational mobility. The degrees of freedom of the alkyl chains of the anchored bilayer are restricted to changes in conformation. This thesis describes a detailed investigation of the anchored bilayer formed by the intercalation of the anionic surfactant dodecyl sulphate (DDS) in a layered solid, Mg-Al Layered Double Hydroxide(Mg-AlLDH) using both experimental measurements and Molecular Dynamics (MD) simulations (Chapter 2). The thesis is organized as two parts. The first (Chapters 2 -4) deals with the anchored bilayer in the condensed phase -the conditions for the formation of the bilayer arrangement of the intercalated surfactant chains and the conformation and dynamics of the alkyl chains of the surfactant in the galleries of the layered solid. The surfactant intercalated Mg-AlLDH-DDS may be delaminated in nonpolar solvents to give colloidal dispersions of individual Mg-Al LDH sheets with the DDS surfactant chains remaining tethered to the inorganic sheets(Scheme 1).The second part of thesis(Chapters 5 -9)describe studies on the dispersions of the Mg-AlLDH-DDS in toluene. A summary of the results of the of the investigations of the anchored bilayer, formed by the intercalation of DDS ions in Mg-Al LDH, in the condensed and the dispersed phases is presented in the concluding chapter(Chapter10). Layered Double Hydroxides(LDH) are insulating lamellar solids of the general chemical formula[M’(1-x) Mx(OH)2], where M’ is a divalent metal ion and M a trivalent ion. Their structure may be derived from that of Brucite, Mg(OH)2, by isomorphous substitution of apart of the Mg2+ by trivalent ions like Al3+ with electrical neutrality maintained by interlamellar exchangeable ions. The studies reported in this thesis are on an Mg-Al LDH,Mg(1−x)Alx(OH)2, x ranging from 0.17 to 0.37. Dodecyl sulphate surfactant ions have been ion-exchange intercalated in Mg-AlLDH (Chapter 3). By varying the Mg-Al ratio, differing packing densities of the surfactant chains in the interlamellar space of the Mg-Al LDH-DDS are realized. At high packing densities the alkyl chains of the intercalated dodecyl sulphate ions anchored on opposing Mg-Al LDH sheets are arranged as bilayers while at lower packing densities the surfactant chains form a monolayer with the chains oriented flat in the galleries. This composition driven monolayer to bilayer transformation in the surfactant intercalated Mg-AlLDH-DDS is also reproduced by MD simulations. The simulations also indicate that there are profound differences in the factors that decide the arrangement of the surfactant chains. In the bilayer arrangement it is dispersive van der Waals interactions between the chains in opposing layers that is responsible for the cohesive energy of the solid whereas at lower packing densities, where a monolayer arrangement is favored, Coulomb interactions between the positively charged Mg-Al LDH sheets and the negatively charged head-group of the DDS anion dominate. The conformation and dynamics of the alkyl chains of the intercalated surfactant chains in both the monolayer and bilayer arrangements as well as the effect of packing density on these parameters is reported in Chapter 4. The conformation was studied using spectroscopic techniques, infra-red, Raman and 13C Nuclear Magnetic Resonance (NMR) while the dynamics by Variable Contact Time Cross Polarization Magic Angle Spinning(VCT -CPMAS) and2DWidelineSeparation(2DWiSe)NMR techniques. The results showed the expected trends; the concentration of gauche defects and the dynamics of the chains increase with decreasing packing density. There is, however a sharp increase in the gauche concentration and conformational mobilities of the intercalated surfactant chains associated with the bilayer to monolayer transformation. The results of the MD simulations, too, reflect these trends. The second part of thesis describes the delamination of the intercalated anchored bilayer (Mg-AlLDH-DDS) in non-polar solvents. Delamination results in a colloidal dispersion Of the anchored bilayer, isolated Mg-AlLDH sheets with the DDS chains tethered to them, as neutral nanosheets of nanometer thickness and micron size. With increasing concentration of the anchored bilayers in the solvent a gel state is realized. The sol to gel transformation of the dispersions of the anchored bilayer in toluene has been investi-gated. Frequency dependent rheology measurements (Chapter6) were used to investigate the visco-elastic properties of the dispersions and Small Angle X-ray Scattering (SAXS) measurements(Chapter 7) to understand the structure and shape of the nanosheets. The rheology experiments showed that the dispersions irrespective of their concentrations showed shear thinning. The SAXS results indicate a tactoid structure of the dispersions as well as in the gel phase. At higher concentrations, the X-ray scattering curves indicated that the layers stack loosely with an interlamellar space of ~ 39 Å , a value much larger than the interlayer lattice spacing of solid Mg-AlLDH-DDS( ~ 27 Å). The nature of interactions between solvent molecules and the anchored DDS chains were probed by 1Hand 2H NMR measurements(Chapter8). A clear association between the toluene molecules and the alkyl chains of the anchored surfactant was observed. 2D NOESY experiments established that there are toluene molecules in close proximity that interact with the methyl tail of the anchored surfactant. NMR measurements were also able to distinguish two types of solvent molecules based on their widely differing mobilities. MD simulations(Chapter9)of the dispersed anchored bilayer are able to reproduce the essential features of the experimental observations including the formation of a loosely bound lamellar structure. It also provides an explanation on how the spectroscopic observation of motional heterogeneity gives rise to the viscoelastic properties of the dispersed anchored bilayer.

Characteristics and mechanisms of phosphate removal by calcium-iron layered double hydroxides and their hydrolysis products / Synthèse et transformations minéralogiques des hydroxydes doubles lamellaires calcium-fer pour l'élimination des phosphates en milieu aqueux

La mise au point de nouveaux matériaux pour piéger les anions phosphate présents dans les eaux usées est une des clés pour lutter contre le phénomène d’eutrophisation des eaux de surface. Au cours de cette étude, les hydroxydes doubles lamellaires contenant des ions CaII et FeIII dans les feuillets cationiques et des anions chlorure et nitrate au sein de l’espace inter-feuillets (CaFe-HDL-Cl et CaFe-HDL-NO3) ont été utilisés pour piéger les phosphates. Ces deux variantes d’HDL ont été synthétisées par trois méthodes simples de co-précipitation. La capacité de piégeage de ces matériaux a été évaluée par des expériences de sorption en « batch » à l’équilibre et des mesures de cinétique dans une gamme de pH comprise entre 4 et 12. La cinétique de piégeage du phosphate par les HDL obéit à une loi du pseudo-second ordre. Les isothermes révèlent que les HDL synthétisés avec un rapport CaII : FeIII de 2 : 1 sont les plus efficaces pour piéger les phosphates. On mesure des capacités de piégeage très élevées de l’ordre de 385 et 402 mg g-1 pour les composés CaFe-HDL-Cl et CaFe-HDL-NO3, respectivement. Les données obtenues à l’équilibre ont pu être ajustées par des isothermes de type Langmuir et/ou Freundlich. Pour des concentrations en phosphate élevées ([PO43-] ≥ 100 mg L-1), la caractérisation des solides et des liquides par diverses techniques révèlent que la HDL se dissout et libèrent des cations solubles Ca2+ qui précipitent avec les phosphates pour former l’hydroxyapatite Ca5(PO4)3OH. A ce mécanisme principal s’ajoute un mécanisme de piégeage secondaire qui est l’adsorption des phosphates par un gel ferrique dopé en ions calcium qui est lui aussi issu de la transformation minéralogique de l’HDL. A faible concentration en phosphate ([PO43-] ≥ 100 mg L-1), l’hydroxyapatite ne se forme pas et l’adsorption des phosphates par ce gel ferrique dopé en ions calcium devient le mécanisme de piégeage principal. / The development of an efficient removal material for phosphate from wastewater to prevent the eutrophication in surface waters is very important. In this study, CaIIFeIII layered double hydroxide containing chloride and nitrate as intercalated anions (CaFe-LDH-Cl and CaFe-LDH-NO3) were used as removal materials for phosphate. Both forms were designed and synthesized at different CaII: FeIII molar ratios (2:1, 3:1, and 4:1) following three simple co-precipitation methods. The materials were evaluated for the removal of phosphate by batch equilibrium sorption experiments and kinetic measurements. The efficiency of the phosphate removal from aqueous solution was investigated in the range pH 4-12. As a result, it was found that pseudo-second-order kinetic model described well the phosphate removal. The isotherms of adsorption of PO43- ions showed that CaFe-LDH-Cl and CaFe-LDH-NO3 at CaII: FeIII molar ratio of 2:1 revealed the highest rate of phosphate uptake of 385 and 402 mg g-1, respectively, comparing to other CaII: FeIII molar ratios. Equilibrium data were well fitted with Freundlich and Langmuir isotherms. In the case of high phosphate concentration ([PO43-] ≥ 100 mg L-1), the comprehensive analysis of the phosphate-uptake products with different techniques revealed that CaFe-LDH dissolve first and then released Ca2+ ions reacting with PO43- anions to form Ca5(PO4)3OH (hydroxyapatite) which is the main mechanism for phosphate removal. Also, phosphate was removed partially via a subsidiary process other than precipitation of hydroxyapatite. The excess in phosphate removal was attributed to the formation of a disordered Ca doped ferrihydrite which removed phosphate via simple surface adsorption. In the case of initial low phosphate concentration ([PO43-] < 100 mg L-1), the analysis showed that PO43- removed mainly via simple surface adsorption over disordered Ca doped ferrihydrite which was the main component in the residual solid after partial Ca2+ releasing from LDH

Efeitos de insulto hipóxico-isquêmico neonatal em respostas comportamentais e bioquímicas analisadas em curto, médio e longo prazo em ratos

A hipóxia-isquemia (HI) neonatal é uma causa importante de deficits neurológicos. Estimativas relatam que 3-5 em cada 1.000 nascidos vivos ocorrem encefalopatia neonatal, sendo que, encefalopatia hipóxico-isquêmica (EHI) moderada ou grave acomete 0,5-1/1000 dos nascidos vivos. Aproximadamente 10 a 60% das crianças com EHI morrem, e pelo menos 25% dos que sobrevivem, apresentam deficiência permanente, como paralisia cerebral, epilepsia e dificuldades na aprendizagem. Evidências clínicas e experimentais sugerem que a exposição a estímulos nocivos no início da vida pode resultar em mudanças duradouras no processamento sensorial. Apesar da magnitude do problema, pouco se conhece sobre a modulação da dor e mecanismos envolvidos durante um processo hipóxico-isquêmico em neonatos e suas conseqüências ao longo da vida. Sendo assim, a presente tese objetivou avaliar respostas comportamentais e bioquímicas de filhotes de ratos submetidos à hipóxia/isquemia no 7°dia de vida, avaliados em diferentes idades. Ninhadas de ratos Wistar machos foram randomizadas em 6 grupos experimentais: controle, hipóxia-isquemia, isquemia, hipóxia, sham-ischemia, sham-hipóxia. O modelo experimental utilizado foi de HI unilateral descrito por Levine (1960) e adaptado por Rice e colaboradores (1981), realizando-se isquemia através da oclusão da carótida esquerda e hipóxia com câmara hipóxica de 92% de nitrogênio e 8% de oxigênio. Avaliaram-se os animais no 14° (P14), 30°(P30) e 60° (P60) dias de vida. Para avaliação da (s) resposta (s): nociceptiva térmica foram utilizados testes de tail-flick e placa quente; e alodinia mecânica utilizou-se teste de Von Frey eletrônico. Para as avaliações dos biomarcadores foram utilizados kits comerciais de BDNF e TNF por métodos ELISA e LDH por espectrofotometria. A ativação neuronal foi quantificada em lâminas de imunoistoquímica de c-fos em regiões CA1 e giro dentado de hipocampo bilateral. Foi observado que ratos com hypoxia e/ou isquemia tiveram diminuição irreversível de neurônios com ativação por c-fos em área CA1 do hipocampo, região ipsilateral do insulto. Maior sensibilidade em animais com HI em P14 foi observado no teste do Von Frey, mas não foi observada em P30 e P60. Independentemente do grupo experimental, a idade do animal desempenhou um papel significativo na resposta de comportamento nociceptivo, bem como sobre o BDNF e dos níveis séricos de TNF-α, mas não sobre os níveis séricos de LDH. Em conclusão, o insulto cerebral da HI induz uma redução significativa e irreversível sobre a ativação neuronal ipisilateral à lesão, com a evidência de uma alteração da resposta comportamental nociceptiva manifestada no curto, mas não a médio ou longo prazo. Biomarcadores de neuroplasticidade, inflamação e danos celulares apresentaram poucas alterações sobre o insulto cerebral, mas mudanças mais pronunciadas foram observadas com relação à idade. / Neonatal hypoxia-ischemia (HI) is an important cause of neurological deficits. It is estimated that 3-5 in 1000 newborns suffer neonatal encephalopathy, and moderate or severe hypoxic-ischemic encephalopathy (HIE) in 0,5-1/1000 of the newborns. Approximately 10 to 60% of the children with HIE die, and at least 25% of the survivors present permanent deficits such as cerebral palsy, epilepsy and learning difficulties. Clinical and experimental evidence suggest that the exposure to noxious stimuli at the beginning of the life might result in long lasting changes in sensorial processing. Despite the magnitude of the problem, few is known regarding pain modulation and its involved mechanisms during the HI process in neonates and its life lasting consequences. Hence, the present thesis aimed to evaluate behavioral and biochemical responses in rats’ litters submitted to HI during their seventh day of life, evaluated at different ages. Male Wistar rats’ litters were randomized into 6 experimental groups: control, HI, ischemia, hypoxia, sham-ischemia and sham-hypoxia. The experimental model was the one of unilateral HI described by Levine (1960) and adapted by Rice et al (1981), inducing ischemia by occlusion of the left carotid artery, and hypoxia using hypoxic chamber with nitrogen 92% and oxygen 8%. Animals were evaluated in their 14th (P14), 30th(P30) and 60th (P60) postnatal day. To assess the thermal nociceptive response the tail flick and hot plate test were employed; and mechanical allodynia was assessed by means of the digital Von Frey test. To evaluate biomarkers, commercial BDNF and TNF kits with ELISA, and LDH kits with spectrophotometry were used. Neuronal activation was quantified by c-fos immunohistochemistry in CA1 and dentate gyrus of bilateral hippocampi. It was observed that rats with hypoxia and/or ischemia had irreversible diminution of neurons with c-fos activation in the CA1 area of the hippocampus ipsilateral to the insult. Higher sensitivity in animals with HI at P14 was observed in the Von Frey test, but was not observed at P30 or at P60. Irrespective of the experimental group, animals’ age played a significant role in the nociceptive behavioral response, as well as on the BDNF and TNF-α serum levels, but not on the LDH serum levels. In conclusion, cerebral HI insult induces a significant and irreversible reduction on neuronal activation ipsilateral to the insult, with evidence of a significant alteration on the nociceptive behavioral response manifested in the short, but not in the medium or long term. Biomarkers of neuroplasticity, inflammation and cellular damage present few alterations after the cerebral insult, however more pronounced changes were observed related with age.

Alguns processos de descontaminação utilizam materiais sólidos para adsorção/sorção, com características apropriadas para a implantação em escala industrial, apresentando alta eficiência e baixo custo. Os Processos Avançados de Oxidação (AOP), como a fotocatálise heterogênea, também têm sido empregados na descontaminação de águas, convertendo os poluentes orgânicos em substâncias mais facilmente degradáveis, inertes ou inofensivas. Materiais multifuncionais como os Hidróxidos Duplos Lamelares (HDL) e seus derivados se mostram promissores tanto em processos adsortivos como fotocatalíticos. No presente trabalho foi dado enfoque ao processo de adsorção de um azocorante têxtil, utilizando HDL do sistema ZnAl-CO3 e o óxido misto obtido no tratamento térmico a 450°C. Os materiais apresentaram características interessantes e alta capacidade de remoção do corante. As isotermas de adsorção apresentaram boa correlação com o modelo de Langmuir e o cálculo de seus parâmetros revelou que a capacidade máxima adsortiva do material calcinado foi cerca de 6 vezes superior ao material não calcinado, 1587,1 mg⋅g-1 e 261,8 mg⋅g-1, respectivamente. A maior capacidade de adsorção/sorção foi atribuída ao melhoramento das propriedades texturais e ao comportamento da “regeneração” da estrutura HDL pelo precursor calcinado. Os resultados revelaram que o controle cinético do processo é uma etapa de adsorção química, e os parâmetros termodinâmicos calculados mostraram que o processo de adsorção é espontâneo e endotérmico. Os experimentos de conversão fotocatalítica de corante mostraram que os derivados da calcinação dos HDL são materiais promissores para serem utilizados nos AOP. A formação do nanocompósito ZnAl2O4/ZnO no material calcinado a 850°C, mostrou os melhores resultados de conversão, pela estratégia de acoplamento de bandas para separação de cargas, que diminui os problemas relacionados com a recombinação dos portadores de carga, além da maior absorbância no UV. Dessa forma, os resultados obtidos mostram a multifuncionalidade desses materiais, e fornecem novas evidências sobre a reconstrução de HDL pelo “efeito memória”, tópico de intensa discussão na literatura. / The contamination of water resources by several organic pollutants, including textile dyes, which have caused serious environmental problems. Solid materials for adsorption with appropriate features to be employed in industrial scale, such as high efficiency and low cost have been used in some decontamination processes. The Advanced Oxidation Processes (AOP) including heterogeneous photocatalysis, have also been used in water treatment, converting the organic pollutants into easily degradable, inert or harmless substances. Multifunctional materials, such as the Layered Double Hydroxides (LDH) and their derivatives, show potential in both adsorptive and photocatalytic processes. In this work the textile azo dye adsorption process using a ZnAl-CO3 LDH and the mixed oxide obtained from thermal treatment at 450 °C were focused. The materials presented interesting features and high removal capacity for the dye. The adsorption isotherms showed good correlation with the Langmuir isotherm model and the calculated parameters revealed that the maximum adsorption capacity of the calcined material was about 6 times higher than the non-calcined material, 1587.1 mg⋅g-1 and 261.8 mg⋅g-1, respectively. The higher adsorption capacity was assigned to the improvement of the textural properties and to the LDH-structure recovery from the calcined precursor. The results showed kinetic control of the process is a chemical adsorption step, and the calculated thermodynamic parameters showed the adsorption process is spontaneous and endothermic. The dye photocatalytic conversion experiments presented that the derivatives of the LDH calcination are promising materials to be used in the AOP. The formation of a ZnAl2O/ZnO nanocomposite from calcination at 850 °C showed the best conversion results from the coupling semiconductors for charge separation strategy that diminish some problems associated with charge carrier recombination and its higher UV absorbance. Thus, the results show the multi-functionality of these materials, giving some light to the discussion about LDH recovery by "memory effect" that remains a topic of intense debate in the literature. / FAPESP: 2013/25043-4

Efeitos de insulto hipóxico-isquêmico neonatal em respostas comportamentais e bioquímicas analisadas em curto, médio e longo prazo em ratos

A hipóxia-isquemia (HI) neonatal é uma causa importante de deficits neurológicos. Estimativas relatam que 3-5 em cada 1.000 nascidos vivos ocorrem encefalopatia neonatal, sendo que, encefalopatia hipóxico-isquêmica (EHI) moderada ou grave acomete 0,5-1/1000 dos nascidos vivos. Aproximadamente 10 a 60% das crianças com EHI morrem, e pelo menos 25% dos que sobrevivem, apresentam deficiência permanente, como paralisia cerebral, epilepsia e dificuldades na aprendizagem. Evidências clínicas e experimentais sugerem que a exposição a estímulos nocivos no início da vida pode resultar em mudanças duradouras no processamento sensorial. Apesar da magnitude do problema, pouco se conhece sobre a modulação da dor e mecanismos envolvidos durante um processo hipóxico-isquêmico em neonatos e suas conseqüências ao longo da vida. Sendo assim, a presente tese objetivou avaliar respostas comportamentais e bioquímicas de filhotes de ratos submetidos à hipóxia/isquemia no 7°dia de vida, avaliados em diferentes idades. Ninhadas de ratos Wistar machos foram randomizadas em 6 grupos experimentais: controle, hipóxia-isquemia, isquemia, hipóxia, sham-ischemia, sham-hipóxia. O modelo experimental utilizado foi de HI unilateral descrito por Levine (1960) e adaptado por Rice e colaboradores (1981), realizando-se isquemia através da oclusão da carótida esquerda e hipóxia com câmara hipóxica de 92% de nitrogênio e 8% de oxigênio. Avaliaram-se os animais no 14° (P14), 30°(P30) e 60° (P60) dias de vida. Para avaliação da (s) resposta (s): nociceptiva térmica foram utilizados testes de tail-flick e placa quente; e alodinia mecânica utilizou-se teste de Von Frey eletrônico. Para as avaliações dos biomarcadores foram utilizados kits comerciais de BDNF e TNF por métodos ELISA e LDH por espectrofotometria. A ativação neuronal foi quantificada em lâminas de imunoistoquímica de c-fos em regiões CA1 e giro dentado de hipocampo bilateral. Foi observado que ratos com hypoxia e/ou isquemia tiveram diminuição irreversível de neurônios com ativação por c-fos em área CA1 do hipocampo, região ipsilateral do insulto. Maior sensibilidade em animais com HI em P14 foi observado no teste do Von Frey, mas não foi observada em P30 e P60. Independentemente do grupo experimental, a idade do animal desempenhou um papel significativo na resposta de comportamento nociceptivo, bem como sobre o BDNF e dos níveis séricos de TNF-α, mas não sobre os níveis séricos de LDH. Em conclusão, o insulto cerebral da HI induz uma redução significativa e irreversível sobre a ativação neuronal ipisilateral à lesão, com a evidência de uma alteração da resposta comportamental nociceptiva manifestada no curto, mas não a médio ou longo prazo. Biomarcadores de neuroplasticidade, inflamação e danos celulares apresentaram poucas alterações sobre o insulto cerebral, mas mudanças mais pronunciadas foram observadas com relação à idade. / Neonatal hypoxia-ischemia (HI) is an important cause of neurological deficits. It is estimated that 3-5 in 1000 newborns suffer neonatal encephalopathy, and moderate or severe hypoxic-ischemic encephalopathy (HIE) in 0,5-1/1000 of the newborns. Approximately 10 to 60% of the children with HIE die, and at least 25% of the survivors present permanent deficits such as cerebral palsy, epilepsy and learning difficulties. Clinical and experimental evidence suggest that the exposure to noxious stimuli at the beginning of the life might result in long lasting changes in sensorial processing. Despite the magnitude of the problem, few is known regarding pain modulation and its involved mechanisms during the HI process in neonates and its life lasting consequences. Hence, the present thesis aimed to evaluate behavioral and biochemical responses in rats’ litters submitted to HI during their seventh day of life, evaluated at different ages. Male Wistar rats’ litters were randomized into 6 experimental groups: control, HI, ischemia, hypoxia, sham-ischemia and sham-hypoxia. The experimental model was the one of unilateral HI described by Levine (1960) and adapted by Rice et al (1981), inducing ischemia by occlusion of the left carotid artery, and hypoxia using hypoxic chamber with nitrogen 92% and oxygen 8%. Animals were evaluated in their 14th (P14), 30th(P30) and 60th (P60) postnatal day. To assess the thermal nociceptive response the tail flick and hot plate test were employed; and mechanical allodynia was assessed by means of the digital Von Frey test. To evaluate biomarkers, commercial BDNF and TNF kits with ELISA, and LDH kits with spectrophotometry were used. Neuronal activation was quantified by c-fos immunohistochemistry in CA1 and dentate gyrus of bilateral hippocampi. It was observed that rats with hypoxia and/or ischemia had irreversible diminution of neurons with c-fos activation in the CA1 area of the hippocampus ipsilateral to the insult. Higher sensitivity in animals with HI at P14 was observed in the Von Frey test, but was not observed at P30 or at P60. Irrespective of the experimental group, animals’ age played a significant role in the nociceptive behavioral response, as well as on the BDNF and TNF-α serum levels, but not on the LDH serum levels. In conclusion, cerebral HI insult induces a significant and irreversible reduction on neuronal activation ipsilateral to the insult, with evidence of a significant alteration on the nociceptive behavioral response manifested in the short, but not in the medium or long term. Biomarkers of neuroplasticity, inflammation and cellular damage present few alterations after the cerebral insult, however more pronounced changes were observed related with age.

Design, synthesis and biological evaluation of potential inhibitors of S100P, a protein implicated in pancreatic cancer

Pancreatic cancer is relatively uncommon. Despite its relative scarcity, it is the fourth-ranked cancer killer in the Western world with less than a 5% 5-year survival rate. The high mortality rate is due to the asymptomatic nature of the disease and the advanced stage at which it is usually diagnosed. S100P is a calcium-binding protein that has been shown to be highly expressed in the early stages of pancreatic cancer and has been proposed as a potential therapeutic target via the blocking of its interaction with its receptor RAGE, the receptor for advanced glycation end-products. In this thesis, computational techniques were employed on the NMR ensemble of S100P (PDB Accession code 1OZO) to identify potential inhibitors of the S100P-RAGE interaction in the hope of identifying a series of novel leads that could be developed into clinical candidates for the treatment of pancreatic cancer. In silico studies identified putative binding sites at the S100P dimeric interface capable of accommodating cromolyn, an anti-allergy drug shown to bind to the protein both in vitro and in vivo. Virtual screening of >1 million lead-like compounds using 3D pharmacophore models derived from the predicted binding interactions between S100P and cromolyn, identified 9,408 'hits'. These were hierarchically clustered according to similarities between chemical structures into 299 clusters and 77 singletons. Biological screening of 17 of the 'hits' identified from virtual screening stuidies, 4 of which were synthesised in-house, against pancreatic cancer cell lines identified five compounds that demonstrated an equal or greater capacity to reduce BxPC-3 S100P-expressing pancreatic cells' metastatic potential in vitro relative to cromolyn. Compound 24 in particular, showed significant (p<0.05) inhibition of invasion of these cells at a concentration of 100 μM that was comparable to cromolyn at the same concentration. This compound, structurally distinct from cromolyn, was successfully synthesised, purified and characterised in-house alongside 39 of its analogues. Biological screening of compound 24 and four of its analogues for anti-proliferative activity against BxPC-3 and Panc-1 pancreatic cancer cell lines showed all five compounds significantly (p < 0.0001) inhibiting proliferation in both cell lines at a concentration of 1 μM relative to the non-treated control. Hence, structurally distinct compounds that show promising inhibitory activity on the metastasis and proliferation of pancreatic cancer cells have been identified using a structure-based drug design methodology. These compounds, with further optimisation, could provide good starting points as therapeutic lead candidates for the treatment of pancreatic cancer.
