Cultivo in vitro e desenvolvimento pós-seminal de espécies de Bromeliaceae com potencial ornamental / In vitro culture and post-seminal development of Bromeliaceae with ornamental potential

As bromélias são valorizadas por suas características ornamentais, sendo o gênero Vriesea representativo neste setor. O aprimoramento de métodos de propagação in vitro destas plantas é altamente necessário a fim de suprir as necessidades do mercado, e evitar o extrativismo ilegal. Nesse contexto, o presente trabalho objetivou aprimorar o protocolo de propagação in vitro de espécies do gênero Vriesea, bem como aumentar o conhecimento global das espécies em estudo. Para tanto, sementes das espécies V. carinata, V. friburgensis, V. paraibica e V. simplex foram submetidas a processos de assepsia e introduzidas in vitro sob três temperaturas: 22 °C, 27 °C e 32 °C. Paralelamente, sementes das mesmas espécies foram semeadas em bandejas e mantidas em casa de vegetação. Através da microscopia eletrônica de varredura e ótica foi realizada a descrição morfo-anatômica do desenvolvimento pós-seminal das plântulas das mesmas espécies. Além disso, procurou-se adequar o meio de cultura às necessidades das mesmas espécies e de V. hieroglyphica, sendo testadas 3 doses de nitrogênio e 3 doses de magnésio. Também procurou-se avaliar a taxa de sobrevivência durante o processo de aclimatização de plântulas das espécies de Vriesea mencionadas (com exceção de V. hieroglyphica). Objetivou-se comparar características anatômicas e morfológicas de folhas das referidas espécies cultivadas in vitro e em casa de vegetação. Por fim, com o objetivo de estabelecer um protocolo de micropropagação para as espécies Vriesea carinata, V. paraibica, V. phillipo-coburgii; V. simplex e Aechmea nudicaulis, foram introduzidos in vitro explantes somáticos, após testes de assepsia. A partir dos experimentos citados foi verificado que a temperatura exerce uma forte influência nas taxas de germinação e mortalidade das sementes de Vrieseas in vitro, sendo que a temperatura de 32°C proporcionou as maiores taxas de mortalidade, mostrando-se prejudicial ao sucesso reprodutivo. A germinação em casa de vegetação apresentou altas taxas de mortalidade e taxas de germinação mais baixas do que in vitro. A descrição morfo-anatômica do desenvolvimento pós-seminal permitiu a caracterização de cinco estágios de desenvolvimento. Com relação ao experimento de nutrição mineral, foi evidenciado que as doses de nitrogênio e magnésio testadas acarretaram em menor acúmulo de cálcio e de potássio nas plantas, sendo que esse fato resultou em menor acúmulo de massa fresca. O experimento de aclimatização ficou inviabilizado devido ao ataque às plântulas por praga Fungus Gnats. Com a análise morfo-anatômica das folhas de plantas cultivadas in vitro e em casa de vegetação foi possível observar a presença de estruturas típicas de Bromeliaceae nas plantas cultivadas em ambas as condições: estômatos, tricomas escamiformes, mesofilo com epiderme unisseriada, parênquima aqüífero, feixes colaterais fechados e canais de aeração. Com relação à introdução in vitro a partir de explantes somáticos, pode-se afirmar que o uso de cefotaxima apresentou uma boa eficiência no combate à contaminação bacteriana em cultura de ápices caulinares. A escolha de ápice vegetativo de brotos laterais como explantes iniciais para a cultura das referidas espécies in vitro é uma boa opção. A otimização da propagação destas espécies in vitro poderá diminuir a pressão extrativista que estas vêm sofrendo e, ao mesmo tempo, abastecer o mercado ornamental / Bromeliads are valued for their ornamental characteristics and the genus Vriesea is representative in this sector. The improvement of in vitro propagation of these plants is highly necessary in order to meet market needs, and, at the same time, to prevent illegal extraction of this plants from their natural habitat. In this context, this study aimed to improve the protocol for in vitro propagation of species of Vriesea and increase the global knowledge of these by morpho-anatomical characterization of the development of the seedling and leaf. Seeds of V. carinata, V. friburgensis, V.paraiba and V. simplex were submitted to aseptic procedures and introduced in vitro under three temperatures: 22 ° C, 27 ° C and 32 ° C. Additionaly, seeds of these species were sown in trays and maintained in a greenhouse. The post-seminal development was described by light and scanning electron microscopy. In addition, the adjustment of the culture medium for these four species and V. hieroglyphica was tested, by testing three doses of nitrogen combined with three doses of magnesium. The acclimatization efficiency of these Vriesea species, except for V. hieroglyphica, after a prior culture in the presence and absence of IBA was done, in three commercial substrates to verify IBA effect in rooting and seedling survival. This study also aimed to compare anatomical and morphological characteristics of leaves of the species cultivated in vitro and in the greenhouse. Finally, in order to establish a micropropagation protocol for the species Vriesea carinata, V. paraiba, V. phillipo-coburgii; V. simplex and Aechmea nudicaulis, somatic explants were introduced in vitro after sterilization tests. From all the experiments cited it was observed that the temperature strongly influences germination and mortality rates of Vriesea germinating seeds in vitro. The temperature of 32 ° C provided the highest mortality rates, being harmful to the reproductive success of this species. The germination in the greenhouse showed higher mortality and lower germination rates than in vitro germination. The morpho-anatomical description of the post-seminal development allowed for the characterization of five stages of development. With regard to the mineral nutrition experiment, the doses of nitrogen and magnesium tested resulted in less accumulation of calcium and potassium in plants, resulting in less accumulation of fresh weight. The acclimatization experiment was lost by the attack of Fungus gnats. With the morpho-anatomical analysis of leaves of plants grown in vitro and in the greenhouse it was possible to observe the presence of typical structures of Bromeliaceae such as stomata, scales, mesophyll with uniseriate epidermis, water storage tissue, collateral vascular bundles and air channels. Finally, the use of cefotaxime proved efficient against bacterial contamination in in vitro establishment of shoot apex explants in vitro. The choice of shoot apices from lateral buds as initial explants for in vitro establishment of those species was a good alternative. Optimization of in vitro propagation of bromeliad species can reduce their extractivism pressure and, at the same time, supply the ornamental plant market

X. fastidiosa é uma bactéria fitopatogênica limitada ao xilema, que tem afetado um grande número de plantas no Brasil e no mundo. Muitos trabalhos já foram realizados sobre esta bactéria, mas pouco se conhece a respeito da adesão, colonização e expressão dos sintomas. Os objetivos deste trabalho foram: a) através do uso da microscopia eletrônica e de luz, determinar e relacionar o número de vasos colonizados de citros, cafeeiro e ameixeira com a sintomatologia em folhas; b) estudar a adesão, migração radial e colonização dos vasos do xilema do pecíolo de folhas de citros pela bactéria; c) estudar algumas variáveis experimentais que afetam a expressão dos sintomas em fumo; d) verificar os sítios de ligação da bactéria em cigarrinhas vetores; e) estudar a adesão e a formação do biofilme por X. fastidiosa em superfície de poliestireno, como uma nova metodologia. Os resultados mostraram em ameixeira e cafeeiro uma relação entre o número de vasos colonizados e a expressão de sintomas necróticos, relação esta que não pode ser observada para citros, o qual apresentava um número de vasos colonizados do pecíolo bem menor que o das outras duas espécies. No estudo da bactéria nos vaso do xilema de citros foi possível verificar as diversas fases do processo de colonização do xilema, bem como a capacidade da bactéria em degradar a parede celular primária da pontuação e migrar para os vasos adjacentes. Neste estudo foi também possível verificar respostas da planta à bactérias caracterizadas pela produção de cristais no lúmen dos vasos do xilema e o acúmulo de goma e hiperplasia de células nas folhas. No estudo com variedades de fumo verificou-se que a cultivar Havana apresentou expressão de sintomas mais intensa que as variedades TNN e RP1 e que o aparecimento dos mesmos não foi influenciado pelo volume de inóculo e pelo local de inoculação, mas sim pela adubação com sulfato de amônio, a qual retardou o aparecimento dos sintomas e reverteu os sintomas inicias em folhas após a aplicação. Em cigarrinhas, células bacterianas com morfologia similar as de X. fastidiosa, foram visualizadas aderidas pela parte lateral na câmara do cibário (sulco longitudinal, parede lateral e membrana do diafragma) de Acrogonia citrina, e Oncometopia facialis, no canal do apodeme de Dilobopterus costalimai e pela parte polar no precibário de O. facialis. Finalmente, no estudo da adesão de X. fastidiosa a película de poliestireno, os resultados revelaram as várias fases da formação do biofilme, aspectos da sua arquitetura, e indicaram que a técnica é uma ferramenta adequada para o estudo da formação do biofilme e também da morfologia das bactérias. Os resultados são discutidos em termos de modelos de adesão e colonização, da bactéria e importância para o conhecimento dos mecanismos de patogenicidade da bactéria em plantas e transmissão pelos vetores. / X. fastidiosa is a xylem-limited bacterium that has been affecting a high number of plants in Brazil and in the world. A lot of researches were already accomplished on this bacterium, but little is known regarding the adhesion, colonization and expression of the symptoms in plants. The objectives of this work were: a) through the use of electron microscopy and of light microscopy determine and to correlate the number of xylem colonized vessels of petiole of sweet orange, coffee and plum with chlorosis and leaf scorching in leaves; b) study the adhesion, radial migration and colonization of the vessels of the petiole xylem of sweet orange by the bacterium; c) study some experimental variables that affect the expression of symptoms in tobacco; d) verify the retention sites of the bacterium in sharpshooters; d) study the adhesion and biofilm formation by X. fastidiosa on polystyrene surface. The results showed a relationship between the number of colonized vessels in plum and coffee and the expression of necrotic symptoms. However, that relationship was not observed for sweet orange, which presented a number of colonized vessels smaller than the other two species. In the study of the bacterium in the xylem vessels of sweet orange it was possible to verify the several phases of the colonization process of the xylem as well as the ability of the bacterium to degrade the primary cell wall of the pit and migrate to adjacent vessels. It was also possible to verify responses of the plant to the bacterium characterized by the production of crystals in the lumen of the xylem vessels and gum accumulation and hyperplasia in the leaf cells. Regarding the tobacco varieties it was verified that the expression of symptoms is more intense in the cultivar Havana than in the cultivars TNN and RP1. It was also seen that symptoms expression was not influenced by the inoculum volume or the inoculation place, but it was altered by fertilization with ammonium sulfate, which delayed the beginning of the symptoms and reverted the symptoms in leaves after the application. In sharpshooters, bacterial cells exhibiting morphology similar to X. fastidiosa were visualized attached to the lateral side in the cibarium camera (longitudinal, lateral wall and membrane of the diaphragm) of Acrogonia citrina, and Oncometopia facialis, in the apodemal channel of Dilobopterus costalimai, and in the polar part in the pre-cibarium of O. facialis. Finally, in the study of the adhesion of X. fastidiosa on polystyrene surface, the results revealed the several phases of biofilm formation; aspects of its architecture, and it also indicated that the technique is an appropriate tool to study of the formation of biofilms and also of the bacterial morphology. The results are discussed regarding adhesion models, colonization, and distribution of the bacterium in the plant and the importance of knowing the pathogenicity mechanisms of X. fastidiosa and its transmission by the insect vectors.

The Mineralogical Composition of House Dust in Ontario, Canada

Despite increasing concern about the presence of heavy metals, pesticides and other toxins in indoor environments, very little is known about the physical and chemical composition of ordinary household dust. This study represents the first systematic investigation of the mineralogical composition of indoor dust in residential housing in Canada. Specimens of dust were obtained from homes in six geographically separate cities in the Province of Ontario: two located on the metamorphic and igneous rocks of the Precambrian Canadian Shield (Thunder Bay and Sudbury), the other four located on Palaeozoic limestone and shale dominated bedrock (Barrie, Burlington, Cambridge, and Hamilton). Forty samples of household vacuum dust were obtained. The coarse fraction (80 – 300 µm) of this dust was subjected to flotation (using water) to separate the organic components (e.g. insect fragments, dander), natural and synthetic materials (e.g. fibres, plastics) from the mineral residue. The mineral fraction was then analyzed using quantitative point counting, polarizing light microscopy, powder X-ray diffraction and scanning electron microscopy methods. Despite the great distances between the sampling localities and the distinct differences in bedrock geology, the mineral fraction of dust from all six cities is remarkably similar and dominated by quartz and feldspar, followed by lithic fragments, calcite, and amphibole. Some evidence of the influence of local geology can nevertheless be found. For example, a relatively higher proportion of sulphide minerals is observed in the two cities on the Canadian Shield where these minerals are clearly more abundant in the bedrock. Specimens from Sudbury, Canada’s largest mining centre located atop a nickel-sulphide mineral deposit, showed the highest sulphide contents. Quartz is the dominant mineral in all cities. All quartz grains have internal strain features and fluid inclusions that are indicative of a metamorphic-igneous provenance. In all cities, sand is used on the streets as an abrasive for traction during the icy winter season. This sand is obtained in all cases from local glaciofluvial deposits that were ultimately derived principally from the rocks of the Canadian Shield in the last Pleistocene glaciations that affected all of Ontario. Thus, tracking in sand is the most plausible mechanism by which quartz was introduced into these homes since sampling was done, in all cases, in the winter season. The results indicate that glacial deposits dominate the mineral composition of indoor dust in Ontario cities and that nature of the bedrock immediately underlying the sampling sites is relatively of minor importance.

Pollination biology of <i>Echinacea angustifolia</i> and <i>E. purpurea</i> (<i>Asteraceae</i>) in Saskatchewan

The goals of this research project were to identify the various insects observed to visit inflorescences of Echinacea angustifolia DC, and to rank these visitors according to their importance as pollinators of E. angustifolia in Saskatchewan. Studying nectar and the nectary is essential to understanding the interaction of disc florets with pollinators. Nectar-sugar production by disc florets of E. angustifolia and E. purpurea (L. Moench) was quantified from anthesis to cessation with production per disc floret peaking in the afternoon of the staminate phase (191.7 µg) and at midday of the first day of the pistillate phase (156.6 µg), respectively. Morphology of the disc-like floral nectaries of both Echinacea species was studied, as well as the ultrastructure of the nectary of E. purpurea. Modified stomata on the nectary rim are the most likely exits for nectar, but creases in the epidermis may also participate. The nectary of E. purpurea is vascularized by phloem alone, which occurred adjacent to the epidermis. Companion cells possessed wall ingrowths, and these cells may unload arriving sugar destined for either an apoplastic or symplastic pathway. Lobed nuclei were a key feature of secretory parenchyma cells, as was a predominance of mitochondria, suggesting that energy-requiring eccrine secretion predominates in E. purpurea. E. angustifolia exhibited a generalist pollination system, with pollinating insects belonging to the orders Coleoptera, Diptera, Hymenoptera, and Lepidoptera. The pollination efficiency of visitors was determined by single insect visits to bagged, virgin inflorescences followed by quantifying pollen tubes at the bases of receptive styles and/or calculating the percentage of shrivelled styles. It was determined that bumble bees (Bombus spp.) were efficient pollinators, indicating that they would likely contribute much to the pollination of E. angustifolia. Grasshopper bee flies (Systoechus vulgaris Loew) were plentiful but individually were not efficient pollinators, but taken together, they provided much pollination. Golden blister beetles (Epicauta ferruginea Say) were efficient pollinators but where yellow-petalled flowers occurred, their numbers on E. angustifolia decreased. Honey bees (Apis mellifera L.) were efficient pollinators and were present in low numbers without managed introduction. Pierid (2003) butterflies were regular visitors and efficient pollinators, and likely contributed significantly to E. angustifolia pollination. When introduced, the alfalfa leafcutter bee (Megachile rotundata Fabr.) preferred not to forage on E. angustifolia and as such, these solitary bees were not suitable as managed pollinators. In large agricultural plantings of E. angustifolia, however, native insects may not be capable of providing sufficient pollination for seed production when floral competition occurs.

Stability of sodium sulfate dicarbonate (~2Na₂CO₃• Na₂SO₄) crystals

Research on salts species formed by evaporation of aqueous solution of Na2 in the early 1930s. The thermodynamic, crystallographic and many other physical and chemical properties of most of the species formed from this solution has been known for decades. However, there was no complete information or reliable data to confirm the existence of a unique double salt that is rich in sodium carbonate, up until five years ago when a research identified the double salt (~2Na ₂ CO ₃ • Na ₂ SO ₄) from the ternary system Na₂CO ₃Na₂SO ₄ H₂O. Crystallization of this double salt so called sodium sulfate dicarbonate (~2Na ₂ CO ₃ • Na ₂ SO ₄) is known to be a primary contributor to fouling heat transfer equipment in spent-liquor concentrators used in the pulp and paper industry. Therefore, understanding the conditions leading to formation of this double salt is crucial to the elimination or reduction of an industrial scaling problem. In this work, double salts were generated in a batch crystallizer at close to industrial process conditions. X-ray diffraction, calorimetry, and microscopic observation were used to investigate the stability of the salts to in-process aging, isolation and storage, and exposure to high temperature. The results show that care must be taken during sampling on evaporative crystallization. Two apparent crystal habits were detected in the formation of sodium sulfate dicarbonate; the favored habit may be determined by calcium ion impurities in the system. The results also verify that sodium sulfate dicarbonate exists as a unique phase in this system and that remains stable at process conditions of 115-200℃

Το καρκίνωμα εκ πλακώδους επιθηλίου ή πλακώδες καρκίνωμα (ΠΚ) του δέρματος είναι ο δεύτερος πιο συχνός καρκίνος του δέρματος και εμφανίζεται συνήθως σε έδαφος ακτινικής υπερκεράτωσης (ΑΚ). Τα νεοπλασματικά κύτταρα εκφράζουν μια ποικιλία αντιγόνων προσελκύοντας με αυτό τον τρόπο λεμφοκύτταρα, τα διηθούντα τον όγκο λεμφοκύτταρα (Tumor Ιnfiltrating Lymphocytes-TILs) στο μικροπεριβάλλον του όγκου. Με βάση το ανοσοφαινοτυπικό τους προφίλ η πλειοψηφία των TILs εκφράζει το μόριο CD3 και αφορά σε Τ-λεμφοκύτταρα. Αυτά με την σειρά τους διαχωρίζονται σε CD8+ Τ-κυτταροτοξικά λεμφοκύτταρα και CD4+ Τ-λεμφοκύτταρα. Η παρουσία των κυττάρων αξιολογήθηκε με τη χρήση φωτονικού μικροσκοπίου με δυο διαφορετικά ημιποσοτικά συστήματα βαθμολόγησης για όλους τους μελετηθέντες Τ-κυτταρικούς υποπληθυσμούς καθώς και με ένα τρίτο σύστημα ποσοτικής καταμέτρησης για τα Τregs. Σκοπός ήταν να γίνει σύγκριση των μεθόδων ώστε να βρεθεί η καταλληλότερη μεθοδολογία αξιολόγησης της λεμφοκυτταρικής διήθησης στο πλακώδες καρκίνωμα του δέρματος, να υπολογιστεί ο αριθμός των Tregs στις μελετούμενες οντότητες χρησιμοποιώντας το βέλτιστο σύστημα βαθμολόγησης και να αναζητηθούν διαφορές στην πυκνότητα των λεμφοκυτταρικών υποπληθυσμών μεταξύ των οντοτήτων, οι οποίες μπορεί να έχουν παθογενετική ή θεραπευτική κλινική σημασία. / Squamous cell carcinoma (SCC) of the skin is the second most common skin cancer. It usually develops in a background of actinic keratosis (AK). Neoplastic cells express a variety of antigens, attracting lymphocytes in the tumor microenvironment (Tumor Ιnfiltrating Lymphocytes-TILs). Based on the immunophenotypic profile, the majority of TILs expresses the CD3 molecule, so they are T-lymphocytes. These in turn are divided into CD8+ cytotoxic T-lymphocytes and CD4+ T-lymphocytes. The expression of CD25 enables the separation of CD4+ lymphocytes in two further subgroups, the T-helper (CD4+/CD25-) and T-regulatory (CD4+/CD25+) cells. The most reliable marker for the detection of T-regulatory cells (Tregs) is considered to be the Forkhead box P3 (Foxp3). The increased number of TILs is associated with prognosis and treatment in a variety of tumors, including skin cancers, such as melanoma. The evaluation methology of lymphocytic infiltration, however, is not yet clarified and varies in different types of tumors. In the context of this thesis, the detection of T-lymphocytes was performed by immunohistochemistry using the appropriate antibodies (CD3, CD4, CD8, Foxp3). The presence of the cells was evaluated using light microscopy with two different semiquantitative scoring systems, for all the T-lymphocyte subpopulations, as well as with a third system of quantification of Tregs. The purpose was to compare the methods in order to find the most suitable methodology for the evaluation of the lymphocytic infiltration in squamous cell carcinoma of the skin, to evaluate the number of Tregs in the studied entities using the best method and examine for differences in the density of lymphocyte subpopulations.

Development, growth and ultrastructure of the floral nectar spur of Centranthus ruber (L.) DC (Valerianaceae)

The main objective of this research project was to study the growth and development of the floral nectar spur of Centranthus ruber (L.) DC. Nectar spurs are tubular floral outgrowths, generally derived from the perianth organs, which typically contain secreted floral nectar. The morphological characteristics of the spur, particularly the length, determine which floral visitors will be able to access the nectar reward pooled at the spur tip. Therefore, nectar spurs are ecologically important for the development of specialised pollinator interactions and have been demonstrated to act as key innovations in the evolution of some taxa. Morphological and anatomical characteristics of the spur and floral nectary were investigated using light and scanning electron microscopy. Ultrastructural features of the nectar spur, particularly the floral nectary within, were assessed using transmission electron microscopy. Nectar in C. ruber is produced by a trichomatous nectary which runs along the entire, inner abaxial surface of the spur. The nectary is aligned with the single vascular bundle which runs along the abaxial side of the spur, through the sub-nectary parenchyma, and back up the adaxial side. The secretory trichomes are unicellular and, in late development, they develop a thick layer of secondary wall ingrowths which vastly increases the surface area of the plasma membrane for nectar secretion. Elongate, non-secretory trichomes occupy the entire remaining circumference of the spur’s inner epidermis, but their density is reduced compared to the secretory trichomes. The cellular basis for spur growth is poorly characterized in the literature. Until recently, it was assumed that all nectar spurs grow by the constant production of new cells via up to three potential meristematic regions (the meristem hypothesis, Tepfer 1953). The cellular basis for spur growth in C. ruber was investigated by cell file counts and cell length and width measurements along the lateral side of nectar spurs in each of the developmental stages. DAPI stained spurs were also examined with Confocal/Apotome microscopy to determine the timing and position of cell division activity throughout spur development. It was determined that elongation of the spur epidermal cells contributes much more to spur growth than cell division. In early development, division is the primary driver of spur growth and the cells are isotropic. However, as development progresses, cell division activity slows down and the spur cells become increasingly anisotropic until anthesis. The patterns of nectar secretion were determined by assessing the volume, solute concentration and carbohydrate composition of the nectar throughout flowering phenology in two C. ruber plants. Nectar volumes and solute amounts rose initially, followed by an eventual decline in both as phenology progressed towards senescence. Because this study was conducted on greenhouse grown plants, it can be assumed that nectar was not removed by insects, suggesting that it is likely reabsorbed following secretion. High performance liquid chromatography (HPLC) analysis determined that C. ruber's nectar is sucrose dominant and that nectar composition remains stable following anthesis throughout floral phenology.
