Caractérisation Structurale et Biochimique de la Nucléoprotéine des virus grippaux de type A, B et D / Structural and Biochemical characterization of Nucleoprotein of innfluenza A, B and D viruses

Le virus de la grippe est un virus à ARN négatif appartenant à la famille des Orthomyxoviridae qui se compose de 7 membres dont les virus influenza A, B, C et D. Le génome viral comprend 7 à 8 particules ribonucléoprotéiques (RNP) au sein desquelles l’ARN viral (ARNv) est recouvert de multiples copies de nucléoprotéines (NP) et est associé à l’ARN polymérase virale via ses extrémités 3’ et 5’. Au cours de ce travail de thèse, nous nous sommes tout d’abord focalisés sur l’étude biochimique de NP A et NP B et avons pu mettre en évidence des comportements différents en ce qui concerne leurs propriétés d’oligomérisation en présence ou en absence d’ARN et en fonction de la concentration en sel. Pour la première fois nous avons pu observer une structure similaire aux RNP mais reconstituée uniquement à partir de NP A et d’un ARN de 12 nucléotides. Nous avons pu formuler l’hypothèse que 12 nucléotides de l’ARN serait fixés à la NP avec une forte affinité tandis que le reste de l’ARN fixerait la NP avec une affinité beaucoup plus faible. En parallèle nous avons résolu la structure cristallographique de la nucléoprotéine de la grippe de type D et réaliser la caractérisation de son interaction avec l’importine-α7 humaine. Enfin nous avons étudié la fixation de l’ARN sur NP D et mis en évidence l’importance de l’extrémité C-terminale dans le processus de fixation à l’ARN. Ces informations ont permis de formuler de nouvelles hypothèses quant au fonctionnement du virus de la grippe et permettre d’inscrire ce projet de thèse dans une dynamique globale de lutte contre ce virus. / Influenza virus is a negative RNA virus belongs to the Orthomyxoviridae family which consists of 7 members including influenza viruses A, B, C and D. The viral genome comprises 7 to 8 ribonucleoprotein particles (RNP) in which the viral RNA (vRNA) is coated with multiple copies of nucleoproteins (NP) and is associated with the viral RNA polymerase by its 3 'and 5' ends. In this thesis, we first focused on the biochemical study of NP A and NP B and we demonstrate that there are different behaviors with regard to their oligomerization properties in the presence or absence of RNA and as a function of the salt concentration. For the first time we were able to observe a structure very similar to RNP which was reconstituted only from NP A and a 12 nucleotide RNA. Thus, we formulate the hypothesis that 12 nucleotides of the RNA would bind NP with a very strong affinity while the rest of the RNA would bind NP with a lower affinity. In parallel, we solved the crystallographic structure of the nucleoprotein of influenza D virus and we characterized its interaction with human importin-α7. Finally, we studied the binding of RNA on NP D and we demonstrated the importance of the C-terminal end in the RNA binding process. This thesis project made it possible to formulate new hypotheses concerning the functioning of the influenza virus and to include this thesis project in a global dynamic of combating the influenza virus.

Polymeranaloge Carbanilierung von Cellulose / Beiträge zur Methodenentwicklung und Untersuchung von Depolymerisationsprozessen

Characterization of cellulose by its molecular weight distribution is afforded after polymeranalogeous dissolution. Additionally, a molecular dispersion of the polymer is a prerequisite. Common processes are dissolution of cellulose in dimethylacetamide-lithiumchloride, nitration and carbanilation. Degradation of the polysaccharide chains can occur in each of the mentioned processes. It is shown that degradation in pyridine occurs via beta-elimination at carbonyl groups along the cellulose chains. Carbanilierung in DMSO is much more pronounced. It comprises oxidation along the Pfitzner-Moffatt-mechanism and subsequent beta-elimination at the thus formed carbonyl-groups. This was elucidated with model systems and by investigation of the carbanilation in different media. Carbonyl groups of cellulose are masked through reaction with phenylisocyanate. This was shown with model. Therefore, the determination of carbonyl groups in cellulose-tricarbanilates is not possible. The separation of low-molecular weight byproducts was optimised. The influence of pretreatment and preactivation of cellulose-samples on the completeness of the conversion was studied. A standard protocol for the carbanilation of cellulose is provided. / Cellulose wird u.a. durch ihre Molmassenverteilung charakterisiert, deren Ermittlung ein polymeranaloges Verfahren zur molekulardispersen Auflösung des Polymers erfordert. Hierfür sind die Direktlösung, die Nitrierung und die Carbanilierung in Gebrauch. Bei allen Prozessen kann es zum Abbau der Polysaccharidketten kommen, wobei diesen Prozessen wenig Beachtung geschenkt wurde. In der Arbeit wird gezeigt, daß der Abbau bei der Carbanilierung in Pyridin durch Beta-Eliminierung an vorhandenen Carbonylgruppen erfolgt. Die Carbanilierung in DMSO fällt stets stärker aus als bei Einsatz von Pyridin und umfasst die Prozesse Oxidation nach dem Pfitzner-Moffatt-Mechanismus und anschließende Beta-Eliminierung an den neu gebildeten Carbonylgruppen. Dies wird durch Untersuchungen an Modellsystemen und am Polymer herausgearbeitet. Carbonylgruppen an Cellulose werden durch die Umsetzung mit Phenylisocyanat maskiert, was an Modellverbindungen gezeigt wurde (Bildung von Endioldicarbanilaten und carbanilierten Halbacetalen). Ihre Bestimmung in Cellulosecarbanilaten ist daher nicht möglich. Die Abtrennung von niedermolekularen Nebenprodukten der Umsetzung wurde optimiert. Der Einfluss der Vorbehandlung und Voraktivierung von Celluloseproben auf die Vollständigkeit der Umsetzung wurde eingehend untersucht. Es wird ein Standardverfahren zur Carbanilierung von Cellulose angegeben.

Suburban retrofitting : a useful strategy for swedish urban planners? / Suburban retrofitting : en användbar strategi för svenska stadsplanerare?

There is an on-going generational shift related to car culture, occurring in the developed world. After decades of increased driving, a decline can now be seen in twenty of the developed countries. Moreover, one out of five regional shopping malls in America is either dead or likely to fail within the next five years. This has given rise to a new urban planning strategy called “suburban retrofitting”. The strategy mainly concerns redevelopment of suburban areas; involving densification of the area’s built environment, a diversification of functions and improved public transportation to the area. The most common mode of public transportation used within suburban retrofitting is light rail. Most suburban retrofitting projects to date have been carried out on sites of dead shopping malls, and the literature focused on suburban retrofitting is, so far, mainly focused on American conditions. Thus the purpose of the thesis is to relate the theories about suburban retrofitting to the Swedish urban planning context, to see if these theories are useful and relevant for Swedish urban planners and policymakers. This question is highly dependent on whether these strategies can be considered to contribute to sustainable development. A case study is carried out about the redevelopment of a commercial area called Kungens Kurva, in Huddinge, south of Stockholm. The area will be denser, take on a city like character and be supplied with public transport. However, some important aspects of this project differ from the theories about suburban retrofitting: no housing is planned, substantial space for parking remains and shopping will still be the dominant activity in the area. The preconditions for suburban retrofitting differs greatly between Sweden and the U.S. Thus, the subjects for suburban retrofitting in Sweden might currently rather be the “million program” suburbs than commercial areas at the urban fringe. One of the reasons for this is that, in Sweden, commercial areas at the urban fringe are often still doing economically well. Which means that the preconditions for fundamentally changing the character of these areas – by introducing housing and diversifying the functions in the area – simply is not good enough. Other factors that make a radical transformation of commercial areas difficult are the strong regulations concerning noise and pollution. It is particularly the regulations concerning housing that make suburban retrofitting projects adjacent to highways problematic in Sweden. As for introducing a light rail system to the area, it is only a rational choice for the Swedish cities that already has such a system. The subway system has proven to be far more effective in diverting car trips to locations at the urban fringe. Therefore it might be a better choice for suburban retrofitting projects in Stockholm. / Det pågår för närvarande ett generationsskifte vad gäller bilkulturen, både i Sverige och i hela den industrialiserade världen. Efter decennier av ökande bilkörning kan man nu se en nedgång i tjugo av de utvecklade länderna. Dessutom har ett av fem externhandelsområden i USA redan lagts ned, eller är sannolikt att bli nedlagt inom de närmaste fem åren. Detta har gett upphov till en ny strategi inom stadsplanering som kallas för ”Suburban retrofitting”, eller ”förtortsomvandling”. Strategin syftar till att omvandla förortsmiljöer och innebär förtätning av den byggda miljön, en diversifiering av områdets funktioner och förbättrad kollektivtrafik till området. I förortsomvandlingsprojekt är den vanligaste typen av kollektivtrafik spårvägssystem. De flesta genomförda förortsomvandlingsprojekt har avsett nedlagda köpcentrum och den litteratur som berör ämnet är i huvudsak fokuserad på Amerikanska förhållanden. Syftet med examensarbetet är att relatera teorier om förortsomvandling till svensk stadsplanering för att se om dessa teorier är användbara och relevanta för svenska stadsplanerare och beslutsfattare. Vilket är en fråga som i sin tur är starkt beroende av huruvida dessa strategier kan anses bidra till en hållbar utveckling. I detta examensarbete genomförs en fallstudie som berör omvandlingen av externhandelsområdet Kungens Kurva, beläget söder om Stockholm. Området planeras att bli tätare, få en kvartersstruktur och förses med kollektivtrafik. Dock skiljer sig projektet från de teorier som finns i litteraturen om förortsomvandling i flera viktiga avseenden. Inga bostäder är planerade, bilen står fortfarande i centrum och shopping kommer fortfarande att vara den dominerande aktiviteten i området. Några av de viktigaste slutsatserna i examensarbetet är att förutsättningarna för förortsomvandling skiljer sig kraftigt mellan Sverige och USA. Framförallt är de ekonomiska förhållandena för externhandelsområden i Sverige fortfarande relativt bra. Dessutom har Sverige starka regleringar vad gäller partikelhalter och buller, vilket kan göra omvandlingsprojekt nära motorvägar svåra att genomföra. Särskilt om de avser bostäder, vilka är en nödvändig del av ett förortsomvandlingsprojekt om det ska anses bidra till en hållbar stadsutveckling. Vad gäller frågan om att införa spårvägsförbindelser till förortsmiljöer under omvandling är detta endast ett rationellt val om man redan har ett spårvägssystem, eller saknar spårbunden kollektivtrafik överhuvudtaget. I det fall man har ett tunnelbanesystem, som i Stockholm, är det sannolikt mer miljömässigt hållbart att utnyttja detta än att bygga spårväg. Det beror på att tunnelbanesystem är betydligt mer konkurrenskraftigt gentemot bilen avseende resor till perifera mål, eftersom tunnelbanetrafiken är separerad från andra trafikslag och därmed får en högre medelhastighet.

La pandemia está cambiando los hábitos y las actitudes de compra de los consumidores al rededor de todo el mundo. El negocio de malls en Arequipa en gran parte de la cuarentena no ha tenido ventas en local físico y enfrentan nuevos desafíos, por otro lado, las personas están repensando la compra dirigiéndola a sus necesidades básicas, a la compra en lugares cercanos a sus domicilios, hacia productos específicos y toman una actitud frente a la compra influenciadas diferentes variables. El objeto de esta tesis es validar un modelo a través de ecuaciones estructurales usando los criterios de lealtad, capital de marca, valor percibido y satisfacción hacia la actitud de compra en los estudiantes universitarios de Arequipa en tiempos de la covid-19 y post-covid 19. El modelo propuesto se aplicó a 389 estudiantes en el mes de diciembre del 2020; a partir de los datos obtenidos se ha determinado la validez y confiabilidad de cada uno de los elementos del modelo y posteriormente se ha reestructurado hacia el modelo final que se presenta en el trabajo. / Pandemic is changing consumers purchase habits and attitudes around the world. In quarantine shopping malls in Arequipa have not had sales in physical shop and they face new challenges. On the other hand, people are rethinking the purchase directing it to their basic needs, to purchase in places close to their homes, to acquire specific products and they take an attitude towards the purchase influenced by different variables. The purpose of this thesis is to validate a model through structural equations using the criteria of loyalty, brand equity, perceived value, and satisfaction towards the purchasing attitude of university students in Arequipa in times of covid-19 and post-covid 19. The proposed model was applied to 389 students in December 2020. From the data obtained, the validity and reliability of each element of the model has been determined and later it has been restructured towards the final model that is presented in this study. / Tesis
