Efeito da solubilidade da parede celular de Saccharomyces cerevisiae sobre a digestibilidade, produtos de fermentação microbiana e parâmetros imunológicos de cães adultos /

Orientador: Aulus Cavalieri Carciofi / Resumo: Derivados da parede celular de levedura têm sido estudados por seu efeito prebiótico. Recentemente, preparações purificadas e mais solúveis foram desenvolvidas, com vista a aumentar seu efeito fisiológico. Este estudo teve por objetivo avaliar os efeitos da inclusão de dois diferentes extratos da parede celular de leveduras, um convencional (PCL) e outro com maior porcentagem de mananoligossacarídeos solúveis (PCLs) sobre a digestibilidade, produtos de fermentação nas fezes e alguns parâmetros imunológicos de cães adultos hígidos. Foi empregada uma única formulação, desdobrada em três tratamentos: CON – controle, sem adição de parede celular de levedura; PCL – adição de 0,3% de parede celular de levedura convencional; PCLs - adição de 0,3% de parede celular de levedura com elevada solubilidade de mananoligossacarídeos (MOS). Foram utilizados 24 cães beagle adultos, com oito repetições por ração, em delineamento em blocos casualizados. Cada bloco teve duração 30 dias, sendo avaliados no início e no final do período as concentrações séricas de fator de necrose tumoral alfa e interleucinas 6 e 10, produção ex vivo de intermediários reativos do oxigênio e nitrogênio por células mononucleares e polimorfonucleares de sangue periférico, capacidade fagocítica de monócitos e neutrófilos e concentração de IgA nas fezes. Adicionalmente, foi avaliada a digestiblidade aparente dos nutrientes, produção e qualidade das fezes, ácidos graxos de cadeia curta e ramificada, lactato, amônia, pH e... Recently, more purified and soluble preparations were developed, attempting to increase their biological actions. This study evaluated the inclusion of two yeast cell wall preparations, one conventional (YCW), and another with higher solubility of the mannan oligosaccharide fraction (YCWs), on their effects on nutrient digestibility, fermentation products on feces, and some immunological parameters of dogs. A single food formulation was used, unfolded on the following treatments: CON – control, without yeast cell wall addition; YCW – addition of 0.3% of a conventional yeas cell wall extract; YCWs – addition of 0.3% of a yeast cell wall extraction with elevated mannan oligosaccharides solubility. Twenty-four adult beagle dogs were used, eight dogs per food, distributed on a randomized block design. Blocks lasted 30 days, and tumor necrosis factor alpha and interleukins 6 and 10, ex vivo production of hydrogen peroxide and nitric oxide by peripheral neutrophils and monocytes, phagocytic index, and fecal IgA were evaluated at the beginning and ending of each period. Microbial influence on intestinal development and mode of action of mannan oligosaccharides in broiler chicken

The effect of intestinal microbiota and dietary supplementation of mannan oligosaccharides (MOS) on mucosal architecture and digestive physiology in broiler chicks was examined. In experiment 1, pre-sterilized eggs (Ross x Ross 308) were placed in three HEPA (high efficiency particulate air)-filtered isolator units at day 19 of incubation. Germ-free chicks in one isolator were conventionalized by exposure to cecal contents from a laying hen. Bacterial contamination occurred in one germ-free isolator such that these birds were monoassociated by a bacterium within the Acinetobacter spp. resulting in 3 categories of microbial status including germ-free (GF, n=10), conventionalized (CV, n=19) and monoassociated (Mono, n=13) birds. Dietary treatments assigned to each isolator consisted of a negative control (NC, 0 g/kg of MOS in the basal diet) and MOS (2 g/kg of MOS in the diet) resulting in a 2X3 factorial treatment arrangement. At 7 d of age, body weight was recorded and birds were killed to permit collection of visceral organs, intestinal tissues and cecal contents. Body weight, relative length of small intestinal segments and relative bursa weight were significantly increased in CV birds. These birds also had increased crypt depth and lamina propria area. Dietary MOS increased villus height and villus surface area in CV birds compared with GF and Mono birds. Transcripts for all housekeeping genes tested in ileal tissue were increased by MOS such that transcripts were normalized to unit mass of total RNA. In comparison to birds fed the NC diet, MOS significantly increased the abundance of proliferative cell nuclear antigen (PCNA), toll-like receptor (TLR)-4, avian β-defensin (GAL)-6, interleukin (IL)-8, peptide transporter 1 (PepT1) and sodium-dependent glucose transporter (SGLT)-1 transcripts in ileum per unit total RNA. However, the effect of microbial status on selected gene expression profiles was surprisingly limited. A second experiment was conducted to confirm the conventionalization protocol produced a complex microbiota similar to conventionally reared birds. Twenty day-old broiler chicks (Ross x Ross 308) were assigned to one of two wire-floored battery cages provided the NC and MOS diets ad libitum and killed at 7 d of age. Terminal restriction fragment length polymorphism (TRFLP) analysis demonstrated that microbial diversity indices (Richness, Evenness, Shannon, and Simpson) were greater in conventionalized gnotobiotic birds compared to the conventionally reared birds confirming a successful conventionalization strategy in the gnotobiotic trial. These studies demonstrate that under good hygienic conditions, CV chicks thrive similar to GF animals. Based on responses to MOS observed in GF birds, evidence indicates that MOS, independent of changes in microbial composition, directly modifies host response parameters including innate immune activation, digestive and absorptive function.

Avaliação do desempenho de Suínos Alimentados com Mananoligossacarídeos (MOS) / Effects of mannan oligosaccharides on gilts and litters performance

O presente estudo buscou avaliar os efeitos dos Mananoligossacarídeos (MOS) como aditivo alimentar no desempenho de primíparas suínas em final de gestação e em lactação, bem como no desempenho e na saúde dos leitões até os 65 dias de idade. O experimento foi realizado no Laboratório de Pesquisa em Suínos (VNP-FMVZ-USP) Pirassununga SP. O delineamento experimental foi inteiramente casualizado em arranjo fatorial de tratamentos, sendo um fator o fornecimento dos MOS para fêmeas e o segundo para os leitões. Foram utilizadas 17 primíparas prenhes, tratadas a partir de 81 &plusmn;1,36 dias de gestação com 0,2% de MOS na dieta. Aos 82 &plusmn; 1,36 dias de gestação as fêmeas foram submetidas à coleta de sangue e à vacinação contra rinite atrófica progressiva, sendo pesadas quinzenalmente até a transferência para a maternidade, aos 109 &plusmn; 1,36 dias de gestação. Na segunda quinzena as fêmeas tratadas apresentaram uma vantagem numérica no ganho de peso médio diário (P=0,063). Durante o parto foram colhidas amostras de sangue e colostro para titulação de anticorpos contra o antígeno vacinal, sendo consideradas positivas 66,67% das fêmeas MOS e 42,86% das fêmeas controle. Os leitões aleitados por fêmeas tratadas tiveram um maior ganho numérico de peso na primeira semana (p=0,0614), significativo na segunda semana (p=0,047). Na terceira semana foi introduzido o segundo fator através do oferecimento de alimento sólido (0,4% de MOS). No período total, do nascimento aos 21 dias, foi observada uma vantagem numérica (p=0,0989) no ganho de peso a favor dos leitões de fêmeas MOS. As fêmeas, contudo, não diferiram quanto à variação de peso ou consumo durante a fase de lactação. Aos 23 &plusmn; 1,91 dias de idade dos leitões foi realizado o desmame abrupto com a transferência dos leitões para unidade de creche, onde foram alojados em gaiolas para 4 animais. O peso ao desmame foi maior nos leitões aleitados por fêmeas tratadas (p<0,0001), bem como em todas as pesagens semanais até a quinta semana pós-desmame. O consumo na primeira semana sofreu influência da suplementação de MOS nas fêmeas e do maior peso à desmama se mostrando superior nesses animais (P=0,049) influenciando diretamente o ganho de peso, superior na segunda semana (p=0,002). O MOS para os leitões aumentou o consumo de alimento (p=0,0784), e a conversão alimentar na segunda semana (p=0,0103). Aos 14 dias pós-desmame foram colhidas amostras de sangue para hemograma e realizada a aplicação oral de 108 UFC de Salmonella Typhimurium. Foram observadas diariamente a consistência das fezes por 28 dias e aferidas as temperaturas retais por 9 dias. Leitões tratados com MOS e os leitões oriundos de fêmeas MOS tenderam a apresentar o pico de hipertermia mais cedo que os demais (p=0,0629 e p=0,0976, respectivamente) e os leitões alimentados com MOS tenderam a ter uma temperatura de pico mais baixa (p=0,0989). Na última semana os leitões de fêmeas MOS apresentaram um maior consumo (p=0,0007) e uma incidência de Salmonella numéricamente inferior em linfonodos mesentéricos e nas fezes colhidas aos 36 dias pós-desafio. Pode-se concluir que a suplementação de MOS para primíparas prenhes e lactentes pode melhorar o desempenho e a saúde entérica de seus leitões nas fases de aleitamento e creche. / The present study evaluated the effects of mananoligossacarides (MOS) as a feed additive on the performance of primiparous sows in late gestation and lactation, as well as on performance and health status of their progeny up to 65 days of age. The experiment was conducted in the Laboratory of Swine Research (FMVZ/USP) Pirassununga SP. For this purpose a completely random factorial design was used, factors which corresponded to (1) feeding sows with MOS and (2) feeding piglets with MOS, characterizing 4 treatments: MM Feeding MOS to both sows and piglets, MC Feeding MOS only to the sows, CM Feeding MOS only to piglets, CC Control diets for both sows and piglets. A total of seventeen pregnant gilts were used, and divided into 2 groups: MOS (n=9) and Control (n=8). MOS gilts had 0,1% MOS added to their diets from 81 &plusmn; 1,36 days of gestation onward. On day 82 &plusmn; 1,36 blood was collected and vaccination against Progressive Atrophic Rhinitis was conducted. Animals were weighted biweekly until 109 &plusmn; 1,36 days of gestation, when transference to the farrowing unit was conducted. Between the first and second weighing, MOS gilts show a numerical advantage in daily weight gain (p=0,063). At farrowing, blood and colostrum samples were collected for determination antibodies titles against vaccine antigen, being considered positive 66,67% and 42,86% of MOS and Control sows, respectively. Piglets nursed by MOS sows had a numerical advantage in weight gain in the first week (p=0,0614), with statistical significance in the second week (p=0,047). On the third week, the second factor was introduced by the offering of solid feed (0,4% MOS). During the suckling period, from birth to 21 days of age, a numerical advantage in weight gain was observed for MOS sows piglets (p=0,0989). The sows themselves did not differ in weight change or feed consumption. At 23 &plusmn; 1,91 days of age, weaning was conducted, as piglets were transferred to nursing facilities and allocated in pens of 4 animals. Weight of MOS sows piglets was higher at weaning (p<0,001) and until the 5th week port-weaning. Feed consumption was affected by MOS supplementation of sows and by the higher weaning weight (p=0,0049), directly influencing weight gain, which was superior in the 2nd week. Feeding MOS to piglets enhanced feed consumption (p=0,0784) and feed conversion (p=0,0103) on the 2nd week. At 14 days of age, blood samples were collected for hemogram analysis and an oral dose of 108 CFU of Salmonella Typhimurium administered. Fecal consistency and rectal temperature were evaluated for 28 and 9 days, respectively. Piglets treated with MOS and MOS sows\' piglets tended to show a peek of hyperthermia earlier (p=0,0629 e p=0,0976, respectively). Piglets fed with MOS tended to show a lower temperature peek (p=0.0989). In the last week, MOS sows\' piglets show a higher feed consumption (p=0.0007) and a numerically inferior Salmonella incidence in mesenteric lymph nodes and feces 36 days after the bacterial challenge. In conclusion, supplementing primiparous sows with MOS during gestation and lactation can enhance their body weight gain and the performance and intestinal health of the offspring during the suckling and nursing period.

THE EFFECTS OF ACTIGEN® AND THREONINE SUPPLEMENTATION ON GROWTH PARAMETERS, IMMUNE FUNCTION, AND INTESTINAL HEALTH IN MONOGASTRICS

The objectives of these experiments were to evaluate the main effects and interactive effects between dietary supplementation of a mannan oligosaccharide based product, Actigen® (ACT), and different levels of threonine in monogastrics, as evidenced by growth parameters, immune function, and intestinal health characteristics. In nursery piglets, ACT supplementation decreased average daily feed intake (P=0.04), but had no effect on body weight or feed conversion ratio. There were no noted differences between threonine levels on performance traits. There were no consistent differences in complete blood count or cytokine gene expression profiles in the blood. The highest level of threonine, 77% true ileal digestible threonine:lysine (Thr:Lys), increased villus height (P=0.007) and villus height:crypt depth (P=0.01). The lowest level, 57% Thr:Lys, decreased villus surface area (P=0.04) and goblet cell density (P=0.04). Supplementation with ACT increased total goblet cell area (P=0.02) and density (P=0.05). There were no interactions observed between ACT and Thr:Lys levels. In broiler chicks, feeding a diet containing 0.56% threonine decreased body weight (PPPP=0.07). On d 7, birds supplemented with ACT had heavier spleens as a percentage of body weight (P=0.01) compared to no ACT. When adjusted for body weight, the 0.56% threonine fed birds had smaller spleens (P=0.05) on d 7 when compared to the other threonine levels. Humerus (PPP=0.04) and birds fed 0.56% threonine had a higher concentration of potassium (PP=0.03) and few goblet cells (P=0.04) on d 7 when compared to the other threonine levels. In d 21 jejunum, supplementation with ACT reduced apical width (P=0.03) and surface area (P=0.02). An interaction was observed between ACT and threonine level in the jejunum on d 21 on basal width (P=0.03) and surface area (P=0.02), indicating that in diets lacking ACT, excess threonine increased villus size. Overall, ACT and threonine acted primarily independently to modulate the intestinal architecture of both nursery piglets and broiler chicks. However, in broiler chicks ACT and threonine interacted to alter villus size. These results indicate that ACT and threonine have direct effects on the intestines of monogastrics.

Flavonóides e mananoligossacarídeos em dietas para frangos de corte

Foram utilizados 1500 frangos de corte da linhagem Cobb, distribuídos em um delineamento inteiramente casualizado com cinco tratamentos e seis repetições de 50 aves por parcela, para análise de desempenho nos períodos de 1 - 21, 22 - 42, 22 - 35, 36 - 42 e 1 - 42 dias. Os tratamentos consistiram em antibiótico, probiótico 1, probiótico 2, testemunha e flavonóide + mos. Aos 42 dias foram retiradas 24 aves por tratamento para as análises de rendimento de carcaça, de partes e qualidade de carne. A análise estatística dos dados foi realizada pelo SISVAR (Ferreira, 1998). Não houve diferença significativa entre os tratamentos para ganho de peso diário, consumo de ração, mortalidade, conversão alimentar e peso médio final nos períodos de 1 - 21, 22 - 42, 22 - 35, 36 - 42. No período de 1 - 42 dias também não houve diferença significativa entre os tratamentos para ganho de peso diário, consumo de ração, mortalidade e peso médio final mas houve diferença significativa para conversão alimentar, onde o tratamento flavonóide + mos apresentou o melhor índice. O rendimento de carcaça e de partes não foi alterado significativamente entre os tratamentos. The study aimed to evaluated to effect of the use of flavonoids + mannan oligosaccharides (mos) and others growth promoters on performance, carcass yield and cuts, meat quality of broilers reared from 1 - 42 days. This experimental design was the completely randomized. One thousand and fifty male Cobb broiler chicks were distributed in five treatment with six replicates of 50 birds each. Growth performance data were obtained from 1 - 21, 22 - 42, 22 - 35, 36 - 42 and 1 - 42 days period. The treatments were denominated as: antibiotic, probiotic 1, probiotic 2, negative control and flavonoids + mos. At 42 days of age, twenty four birds from each treatment were slaughtered to evaluate carcass yield and cuts, and meat quality. Experiment data were calculated and analyzed using SISVAR (Ferreira, 1998). There were no significant effects of treatments on growth performance data were obtained from 1 - 21, 22 - 42, 22 - 35, 36 - 42 days period. In the total period, 1 - 42 days, the results showed that there were no significant of treatment effect on mortality, weight gain, feed consumption, live weight and efficiency production factor but there were significant effect on feed convertion, the treatment flavonoids + mos was better when compared with other treatments. There were no significant effects of treatments on carcass yield, yield cuts, pH and color. There were significant effects of treatments on Tbars (this analysis show the oxidative stability) to breast and thig meat, the treatment flavonoids + mos. was better when compared with other treatments.

Piglets treated with MOS and MOS sows\' piglets tended to show a peek of hyperthermia earlier (p=0,0629 e p=0,0976, respectively). Piglets fed with MOS tended to show a lower temperature peek (p=0.0989). In the last week, MOS sows\' piglets show a higher feed consumption (p=0.0007) and a numerically inferior Salmonella incidence in mesenteric lymph nodes and feces 36 days after the bacterial challenge. In conclusion, supplementing primiparous sows with MOS during gestation and lactation can enhance their body weight gain and the performance and intestinal health of the offspring during the suckling and nursing period.

Flavonóides e mananoligossacarídeos em dietas para frangos de corte /

Orientador: Edivaldo Antônio Garcia / Resumo: O experimento foi realizado no setor de avicultura da Faculdade de Medicina Veterinária e Zootecnia, Câmpus de Botucatu, com o objetivo de avaliar os efeitos da inclusão de flavonóides juntamente com mananoligossacarídeos (mos) frente a outros aditivos sobre o desempenho, rendimento da carcaça e de partes e qualidade da carne de frangos de corte criados no período de 1 a 42 dias. Foram utilizados 1500 frangos de corte da linhagem Cobb, distribuídos em um delineamento inteiramente casualizado com cinco tratamentos e seis repetições de 50 aves por parcela, para análise de desempenho nos períodos de 1 - 21, 22 - 42, 22 - 35, 36 - 42 e 1 - 42 dias. Os tratamentos consistiram em antibiótico, probiótico 1, probiótico 2, testemunha e flavonóide + mos. Aos 42 dias foram retiradas 24 aves por tratamento para as análises de rendimento de carcaça, de partes e qualidade de carne. A análise estatística dos dados foi realizada pelo SISVAR (Ferreira, 1998). Não houve diferença significativa entre os tratamentos para ganho de peso diário, consumo de ração, mortalidade, conversão alimentar e peso médio final nos períodos de 1 - 21, 22 - 42, 22 - 35, 36 - 42. No período de 1 - 42 dias também não houve diferença significativa entre os tratamentos para ganho de peso diário, consumo de ração, mortalidade e peso médio final mas houve diferença significativa para conversão alimentar, onde o tratamento flavonóide + mos apresentou o melhor índice. The study aimed to evaluated to effect of the use of flavonoids + mannan oligosaccharides (mos) and others growth promoters on performance, carcass yield and cuts, meat quality of broilers reared from 1 - 42 days. This experimental design was the completely randomized. One thousand and fifty male Cobb broiler chicks were distributed in five treatment with six replicates of 50 birds each. Growth performance data were obtained from 1 - 21, 22 - 42, 22 - 35, 36 - 42 and 1 - 42 days period. The treatments were denominated as: antibiotic, probiotic 1, probiotic 2, negative control and flavonoids + mos. At 42 days of age, twenty four birds from each treatment were slaughtered to evaluate carcass yield and cuts, and meat quality. Experiment data were calculated and analyzed using SISVAR (Ferreira, 1998). There were no significant effects of treatments on growth performance data were obtained from 1 - 21, 22 - 42, 22 - 35, 36 - 42 days period. In the total period, 1 - 42 days, the results showed that there were no significant of treatment effect on mortality, weight gain, feed consumption, live weight and efficiency production factor but there were significant effect on feed convertion, the treatment flavonoids + mos was better when compared with other treatments. There were no significant effects of treatments on carcass yield, yield cuts, pH and color. There were significant effects of treatments on Tbars (this analysis show the oxidative stability) to breast and thig meat, the treatment flavonoids + mos. was better when compared with other treatments. / Mestre
