The effect of medium composition and ethanol toxicity on the growth of Saccharomyces cerevisiae strain W303-1A(a).

Published Article / The growth of Saccharomyces cerevisiae strain W303-1A(a) was evaluated in complex and chemically defined media. The use of chemically defined medium allowed the complete utilisation of glucose within 20 h. as well as all of the produced ethanol within 45 h. Maximum specific growth rates (µmax) were increased from 0.28 h-1 to 0.42 h-1 and the volumetric rate of ethanol production increased from 0.204 g l-1 h-1 to 0.597 g l-1 h-1. However, when the ethanol concentration exceeded a threshold value of 10 g l-1, the µmax value was significantly decreased. These observations suggest that ethanol metabolism related growth experiments for the relevant strain should be carried out in chemically defined medium with ethanol concentrations below 10 g l-1.

Storytelling for digital photographs: supporting the practice, understanding the benefit

The emergence of digital capture and editing technologies make providing a more detailed and coherent description of the experiences depicted in photos possible. Through the combination of photos, music and voice, people can compose digital stories of their life experiences. However, communicating an experience using photos to people who do not share the experience, and are not co-located is a difficult endeavor, even with effective digital editing tools. In this dissertation, I studied the online photo communication challenges that have arisen as a result of the transition from film to digital photography. I detail my studies of consumer desires and barriers related to online photo communication. Also, I present the design and user evaluation of the Storytellr system, which addresses those desires and barriers. The Storytellr system integrates storytelling activities with traditional photo activities to reduce the challenges of online photo communication. Through this work I contribute to the understanding of the challenges encountered by consumers who desire to engage in sharing life stories through photos over distance. I also contribute a method - integrating storytelling activities into photo activities - for enabling people to overcome those challenges using a process they find satisfying, and that produces an outcome that satisfies authors and viewers alike.

Composition as the creation of a performance, music as a vehicle for non-musical thought : six new works

This thesis comprises six new musical works composed between 2008 and 2015: ‘Struction (how I attempted to get the thoughts in my head into your head using only five instruments, five instrumentalists, metronome sound and MIDI') for amplified ensemble and pre-recorded soundtrack (2011); ‘My Life in Ventriloquism' for solo clarinet and pre-recorded soundtrack (2012); ‘Nightmusic' for solo violin (2012); ‘Replaceable Parts for the Irreplaceable You' for ensemble, pre-recorded soundtrack and video (2013); ‘Espial', a video work featuring string quartet (2014); and ‘Elbow Room' for amplified ensemble, pre-recorded soundtrack and video (2014). The works are presented in this thesis as musical scores (and other performance materials), accompanied by audio-visual documentation of performances. As a whole, these compositions reflect a period of practice-as-research into the role of metapraxis in musical performance and how it can be used to help convey non-musical thought through instrumental music. A commentary on this portfolio of new compositions begins by discussing two influential works — Mauricio Kagel's ‘Match' (1964) and ‘Failing: A Difficult Piece for Solo String Bass' (1975) by Tom Johnson — before examining each new work in detail in order to explicate the research and creative processes that led to their composition, to exteriorize a personal working practice and to document the reflection-on-practice which has furthered this research. The commentary details how I was able to write music on a variety of topics, including authority, technology and place, and concludes with some ideas for further research.

Veiksniai, įtakojantys sėmeninių linų morfogenezę in vitro / Factors affecting morphogenesis in vitro of linseed flax

Study aim is to study endogenic and exogenic factors preconditioning morphogenesis of linseed flax in vitro. Linseed flax studies in vitro were carried out at the Laboratory of Genetic – Biotechnological of Lithuanian University of Agriculture in 2004-2006. Factors which influence morphogenetic capability of linseed flax cultivars ‘Lirina’, ‘Barbara’ and ‘Szaphir’ in tissue culture were investigated. Two different types of explants – stem segments and hypocotyls – of the three genotypes were cultivated in nutrient media differing in their macro and micro salts concentration, vitamins and different levels of growth regulators. Our results showed that intensity of morphogenesis was affected not only by exogenous growth regulators, but also by the type of explant and genotype. Hypocotyls-derived callus gave the better results than that stem-segment-derived callus on all tested media. Four-weeks-old callus showed the maximum shoot regeneration frequency. The best morphogenetic capabilities were demonstrated for cultivar ‘Szaphir’. The largest number of shoots was produced by all the cultivars on MSB5 nutrient medium supplemented with cytokinin 2iP (2.0 mg 1-1). The master’s final work comprises 50 pages, including 3 tables, 10 figures and 66 literature references.
