Hydrogen peroxide-mediated oxidative stress disrupts calcium binding on calmodulin: more evidence for oxidative stress in vitiligo

No / Patients with acute vitiligo have low epidermal catalase expression/activities and accumulate 10 -3 M H 2O 2. One consequence of this severe oxidative stress is an altered calcium homeostasis in epidermal keratinocytes and melanocytes. Here, we show decreased epidermal calmodulin expression in acute vitiligo. Since 10 -3M H 2O 2 oxidises methionine and tryptophan residues in proteins, we examined calcium binding to calmodulin in the presence and absence of H 2O 2 utilising 45calcium. The results showed that all four calcium atoms exchanged per molecule of calmodulin. Since oxidised calmodulin looses its ability to activate calcium ATPase, enzyme activities were followed in full skin biopsies from lesional skin of patients with acute vitiligo (n = 6) and healthy controls (n = 6). The results yielded a 4-fold decrease of ATPase activities in the patients. Computer simulation of native and oxidised calmodulin confirmed the loss of all four calcium ions from their specific EF-hand domains. Taken together H 2O 2-mediated oxidation affects calcium binding in calmodulin leading to perturbed calcium homeostasis and perturbed L-phenylalanine-uptake in the epidermis of acute vitiligo.

Senile hair graying: H2O2-mediated oxidative stress affects human hair color by blunting methionine sulfoxide repair

No / Senile graying of human hair has been the subject of intense research since ancient times. Reactive oxygen species have been implicated in hair follicle melanocyte apoptosis and DNA damage. Here we show for the first time by FT-Raman spectroscopy in vivo that human gray/white scalp hair shafts accumulate hydrogen peroxide (H(2)O(2)) in millimolar concentrations. Moreover, we demonstrate almost absent catalase and methionine sulfoxide reductase A and B protein expression via immunofluorescence and Western blot in association with a functional loss of methionine sulfoxide (Met-S=O) repair in the entire gray hair follicle. Accordingly, Met-S=O formation of Met residues, including Met 374 in the active site of tyrosinase, the key enzyme in melanogenesis, limits enzyme functionality, as evidenced by FT-Raman spectroscopy, computer simulation, and enzyme kinetics, which leads to gradual loss of hair color. Notably, under in vitro conditions, Met oxidation can be prevented by L-methionine. In summary, our data feed the long-voiced, but insufficiently proven, concept of H(2)O(2)-induced oxidative damage in the entire human hair follicle, inclusive of the hair shaft, as a key element in senile hair graying, which does not exclusively affect follicle melanocytes. This new insight could open new strategies for intervention and reversal of the hair graying process.

TOWARD AN ENZYME-COUPLED, BIOORTHOGONAL PLATFORM FOR METHYLTRANSFERASES: PROBING THE SPECIFICITY OF METHIONINE ADENOSYLTRANSFERASES

Methyl group transfer from S-adenosyl-l-methionine (AdoMet) to various substrates including DNA, proteins, and natural products (NPs), is accomplished by methyltransferases (MTs). Analogs of AdoMet, bearing an alternative S-alkyl group can be exploited, in the context of an array of wild-type MT-catalyzed reactions, to differentially alkylate DNA, proteins, and NPs. This technology provides a means to elucidate MT targets by the MT-mediated installation of chemoselective handles from AdoMet analogs to biologically relevant molecules and affords researchers a fresh route to diversify NP scaffolds by permitting the differential alkylation of chemical sites vulnerable to NP MTs that are unreactive to traditional, synthetic organic chemistry alkylation protocols. The full potential of this technology is stifled by several impediments largely deriving from the AdoMet-based reagents, including the instability, membrane impermeability, poor synthetic yield and resulting diastereomeric mixtures. To circumvent these main liabilities, novel chemoenzymatic strategies that employ methionine adenosyltransferases (MATs) and methionine (Met) analogs to synthesize AdoMet analogs in vitro were advanced. Unstable AdoMet analogs are simultaneously utilized in a one-pot reaction by MTs for the alkylrandomization of NP scaffolds. As cell membranes are permeable to Met analogs, this also sets the stage for cell-based and, potentially, in vivo applications. In order to further address the instability of AdoMet and analogs thereof, MAT-catalyzed reactions utilizing Met and ATP isosteres generated highly stable AdoMet isosteres that were capable of downstream utilization by MTs. Finally, the development, use, and results of a high-throughput screen identified mutant-MAT/Met-analog pairs suitable for postliminary bioorthogonal applications.

De part son profil équilibré en acides aminés essentiels (AAE), la farine de poisson (FP) est la source protéique principale utilisée dans l’alimentation des crevettes d’élevage. Cependant, compte tenu des enjeux du développement durable de la production aquacole, son utilisation doit être réduite, et remplacée par d’autres sources protéiques comme les protéines végétales (PV), qui sont souvent carencées en lysine et méthionine mais riches en cystine. Les conséquences d’un tel changement alimentaire sont peu connues chez la crevette tigrée Penaeus monodon. Pour les évaluer, nous avons utilisédes aliments semi-purifiés reflétant les carences/excès en AA des PV pour estimer les besoins en protéine, lysine et méthionine pour l’entretien et la croissance. Tout en confirmant les données antérieures sur les besoins pour la croissance des stades postlarves, nous avons pu préciser la contribution de l’apport en ces deux acides aminés pour l’entretien. Au niveau métabolique, la variation de l’apport protéique (10, 30, 50% protéine brute) et la carence en méthionine (-30% par rapport au besoin) entraînent une modification de l’activité des enzymes du catabolisme des AA, mais pas celle des voies de reméthylation et transsulfuration. En revanche, et pour la première fois chez la crevette, nos résultats démontrent une épargne de la méthionine par la cystine (et la choline), soulignant l’importance de l’apport en AA soufrés totaux (methionine + cystine). Nos résultats illustrent aussi l’importance qu’il convient d’accorder à la disponibilité des AAE dans les études de remplacement de la FP par un mélange de PV pour améliorer l’utilisation azotée chez la crevette P. monodon. / Due to its well balanced essential amino acid (EAA) profile, fishmeal (FM) is the major protein source used in the formulation of aquafeed for cultured shrimp. To sustain farming systems, its incorporation, however, must be reduced and substituted by other protein sources less well nutritionally balanced, such as plant protein ( PP) which are often low in lysine and methionine but rich in cystine. The metabolic consequences of such a shift in dietary profile are not well known for the black tiger shrimp, Penaeus monodon. To describe these consequences, we used semi-purified diets limiting in lysine and methionine (to reflect PP profile) to determine juvenile requirements of protein, lysine and methionine for both maintenance and growth, applying a factorial approach. Our results confirm the previous data on growth requirement for post-larvalstages of P. monodon while also providing new data on maintenance requirements. At the metabolic level, a variation in the dietary protein level (10, 30, 50 % crude protein) and methionine (adequate or 30% lower) resulted in a significant change in the activity of transdeaminating enzyme, but not those of remethylation and transsulfuration. Nevertheless, we found for the first time that methionine utilisation for body protein accretion can be spared by cystine and choline (up to 50%) in this species, illustrating the importance to consider total sulphur AA supply. Our data also show that full consideration should be given to AA availability in order to develop practical diets with low FM levels for P. monodon.

4-Alkyl-L-proline derivatives (APD) are specialized precursors involved in the biosynthesis of at least three groups of different natural compounds: some pyrrolo-1,4-benzodiazepines with antitumor activity, bacterial hormone hormaomycin and clinically used lincosamide antibiotic lincomycin. These compounds share a biosynthetic pathway encoded by 5 or 6 homologous genes present in the biosynthetic gene clusters of the producing organisms. Similarities in biosynthesis and differences between APD structures of these compounds could be used to prepare a hybrid producing strain of biologically more effective lincomycin derivative. Unusual amino acid 4-propyl-L-proline (PPL) is the APD precursor of lincomycin. The originally proposed scheme of the PPL pathway does not comply with our current knowledge. Therefore, it was necessary to revise this scheme according to new results. The first two steps of the PPL pathway are functionally proved. Probing the next step was the main aim of this work. The protein LmbW was overproduced and its methyltransferase activity was confirmed in vitro. LmbW is able to directly methylate intermediate of second step of the pathway while the originally scheme proposed methylation at a later stage of biosynthesis. Effets cellulaires et moléculaires de l’invalidation conditionnelle du gène MTR au niveau du foie et du cerveau de souris / Cellular and molecular effects of conditional MTR gene knockdown in liver and mouse brain

L’enzyme méthionine synthase (MTR) catalyse la reméthylation de l’homocystéine en méthionine, le précurseur du donneur universel de groupe méthyle S-Adenosylmethionine (SAM), impliqué dans des mécanismes de régulations épigénétiques. Des polymorphismes de MTR sont associés à des défauts métaboliques et des défauts de développement embryonnaire. Afin d’étudier les conséquences d’une déficience en MTR, nous avons généré des modèles murins d’invalidation conditionnelle du gène MTR de manière constitutive ou inductible dans le foie et dans le cerveau. L’invalidation constitutive ou inductible ciblée dans le foie pendant l’embryogenèse n’est pas viable, suggérant un rôle limitant de la méthionine synthase sur le développement précoce et l’organogenèse en lien probable avec les conséquences sur la prolifération cellulaire. Dans les périodes post-natales, nous avons utilisé le modèle inductible complété par une hépatectomie pour étudier les altérations de la régénération hépatique liée aux effets sur le stress cellulaire ainsi que l’expression et l’activation des cyclines. Le KO dans le cerveau induit principalement une perte des fonctions de mémorisation de l’apprentissage hippocampo-dépendant. Au total, nos résultats illustrent les effets différents de l’invalidation de MTR en fonction de l’organe considéré. Le foie est un organe très plastique avec une capacité de régénération très importante. Les effets sur les étapes de l’organogénèse et sur l’inhibition de la régénération confirment l’hypothèse du rôle majeur et limitant de la méthionine synthase dans la régulation du cycle cellulaire. Le modèle d’invalidation au niveau du cerveau confirme le rôle très important de la voie de reméthylation de l’homocystéine catalysée par la méthionine synthase, rôle qui a déjà été illustré par d’autres travaux sur les rats carencés en donneur de méthyle et sur la souris transgénique KO cd320 / The enzyme methionine synthase (MTR) catalyzes the remethylation of homocysteine to methionine, the precursor of the methyl donor S-universal Adenosylmethionine (SAM), involved in epigenetic regulation mechanisms. We generated mouse models with conditional invalidation of the mtr gene in a constitutive or inducible manner to delete the gene expression specifically in the liver and brain. Constitutive invalidation during embryonic life is not sustainable when targeted to the liver, suggesting a limiting role of methionine synthase in early organogenesis and probably on cell proliferation. We performed hepatectomy to study regeneration-related effects on the cellular stress and found dramatic effects on cell proliferation through altered expression and activation of cyclins. The constitutive model in brain highlighted the behavioral anomalies related to a loss of learning and memory. This suggested major effects in the hippocampus. Overall, our findings highlighted the specific effects of the invalidation of methionine synthase in both organs. The liver is a plastic member with a very high regenerative capacity. The effects on organogenesis and inhibition of regeneration confirm the hypothesis for a major role of methionine synthase in cell cycle regulation. The invalidation model in the brain confirms the important role of the remethylation pathway catalysed by methionine synthase, a role which has been shown by other studies in rats deprived in methyl donors and in cd320 KO transgenic mice

Rôles de la méthionine sur le métabolisme hépatique de la truite arc-en-ciel (Oncorhynchus mykiss) : focus sur les mitochondries / Roles of methionine on hepatic metabolism of rainbow trout (Oncorhynchus mykiss) : focus on mitochondria

L’impératif d’une aquaculture durable conduit à orienter l’alimentation des poissons vers la substitution de la farine de poisson par des produits végétaux renouvelables. Toutefois, ce remplacement est souvent limité par des niveaux trop faibles en méthionine dans les matières premières végétales. Ainsi la supplémentation en méthionine de ces nouveaux régimes à base de végétaux est essentielle, mais requière une bonne connaissance de son rôle pour adapter les apports aux conditions physiologiques des poissons tout en prenant en compte les contraintes économiques et environnementales de production. Dans ce contexte, cette thèse avait pour principal objectif de caractériser les effets induits par une carence en méthionine sur le métabolisme mitochondrial de la truite arc en ciel. Les résultats obtenus dans notre première étude montrent que l'alimentation de truites avec un régime déficient en méthionine entraîne une baisse des performances de croissance associée à une baisse de l’intégrité mitochondriale et une diminution du statut oxydatif dans le foie. Nous démontrons également que ces perturbations s’accompagnent de l’induction d’un processus de dégradation des mitochondries par autophagie (appelé mitophagie) ainsi que d’une augmentation du stress du Réticulum Endoplasmique (RE) et de l’apoptose. Ces données originales publiées dans le journal Scientific Reports mettent ainsi en évidence les liens étroits qui existent entre différentes fonctions cellulaires pour faire face à un déséquilibre nutritionnel en méthionine. Outre cet effet à court terme, nous démontrons également, dans un seconde étude, qu’une carence en méthionine alimentaire pendant une courte période (2 semaines) lors des premiers repas des alevins entraîne une induction à long terme de facteurs liés à la mitophagie. Ces résultats, soumis à publication dans Journal of Experimental Biology, démontrent ainsi pour la première fois la mise en place d’un processus de programmation de cette fonction cellulaire par une carence précoce en méthionine. L’enrichissement en H3K4me3 et H3K36me3 des foies des poissons issus d’alevins carencés en méthionine par rapport aux poissons témoins suggère une implication de mécanismes épigénétiques dans ces effets. Enfin, dans une troisième étude qui se détache de la thématique principale de la thèse et qui a fait l’objet d’une publication dans le journal Frontiers in Physiology, nous nous sommes attachés à préciser les interactions existante entre l’autophagie, l’homéostasie du RE et le métabolisme intermédiaire. Dans l’ensemble, ces données approfondissent notre compréhension du rôle de la méthionine alimentaire au niveau cellulaire et soulignent le potentiel de cet acide aminé en tant que levier pour appliquer de nouvelles stratégies alimentaires, comme la programmation nutritionnelle, afin d’optimiser la nutrition et la santé des poissons d'élevage. / The expansion of the aquaculture industry in combination with limited availability and high prices of fishmeal has prompted feed producers to include more plant proteins in the aquaculture feeds. However, replacement of fish meal with plant proteins is often limited by the level of methionine in the alternative plant protein sources. Understanding of the different roles of methionine in fish is therefore essential to develop new diets and/or feeding strategies that are in tune with optimal fish growth, environmental and economic constraints. In this context, the main objective of this thesis was to characterize the effects induced by methionine deficiency on the hepatic mitochondrial metabolism in rainbow trout. The results obtained in our first study show that feeding trout with a methionine deficient diet leads to a decrease in growth performance associated with a decrease in both mitochondrial integrity and oxidative stress in the liver. We also demonstrate that these defects are accompanied by the induction of an autophagy-dependent mitochondrial degradation process (called mitophagy) as well as an increase in Endoplasmic Reticulum (ER)-stress and apoptosis. These original data published in Scientific Reports thus highlight the existence of close interactions between different cellular functions to cope to a dietary methionine deficiency. In addition to this short-term effect, we also demonstrate in a second study (submitted for publication in the Journal of Experimental Biology), that early nutritional stimulus during two weeks with a methionine deficient diet resulted in a long term programming of mitophagy. The enrichment of H3K4me3 and H3K36me3 in the liver of fish from methionine-deficient fry compared to their control counterparts suggests that epigenetic mechanisms are involved in these effects. Finally, in a third and last study, recently accepted for publication in Frontiers in Physiology, we sought to clarify, in primary culture of trout hepatocytes, the existing interactions between autophagy, ER homeostasis and intermediate metabolism under amino acid deprived conditions. Together, the results obtained in the present thesis extended our understanding of the role of dietary methionine at cellular level and emphasize the potential of this amino acid to apply new feeding strategies, such as nutritional programming, to optimize the nutrition and health of farmed fish.

Avaliação do padrão nutricional e níveis séricos de ácidos graxos nas gestantes portadoras de fetos com gastrosquise / Evaluation of the nutritional pattern and serum fatty acid levels in pregnant women with fetuses with gastroschisis

Objetivo: avaliar a ingestão de nutrientes no período pré-concepcional e níveis séricos de ácidos graxos, durante a gestação, em gestantes portadoras de fetos com gastrosquise e gestantes portadoras de fetos normais. Métodos: estudo prospectivo caso-controle realizado no período de Julho de 2013 a Julho de 2015 no setor de Medicina Fetal do Hospital das Clínicas. O grupo gastrosquise (GG) foi constituído de 57 gestantes com gestações únicas, idade gestacional inferior a 34 semanas e feto com gastrosquise isolada. O grupo controle (GC) foi constituído de 114 gestantes portadoras de fetos normais pareadas de acordo com idade materna (± 2 anos), idade gestacional (± 2 semanas) e mesma classificação de índice de massa corpórea (IMC) no período pré-concepcional. Os dados referentes ao consumo dietético das gestantes foram obtidos a partir do questionário de frequência e consumo alimentar (QFCA) e o cálculo da ingestão dos nutrientes (macronutrientes; micronutrientes, ácidos graxos e aminoácidos) foi obtido a partir de programas específicos: Dietwin Profissional 2.0® and Virtuanutri®. Para a avaliação de níveis séricos de ácidos graxos (AG), as gestantes foram submetidas à coleta de sangue na entrada no estudo e no momento do parto. A comparação de AG foi realizada durante a gestação e no momento do parto. Com o objetivo de avaliar se as diferenças entre os grupos eram mais frequentes na primeira ou na segunda metade da gestação, uma nova análise foi realizada subdividindo o período gestacional 25 semanas e < 34 semanas. Resultados: no período pré-concepcional, a media diária de calorias ingerida foi maior (2382,43 vs. 2198,81; p = 0,041) no GG em comparação com GC. O consumo médio de metionina (763,89 vs 906,34; p = 0,036), treonina (1248,34 vs. 1437,01; p = 0,018) e crômio (54,66 vs. 59,49 p = 0,014) foi menor no GG em comparação ao GC. Na análise de ácidos graxos, observa-se que o total AG (p = 0,008), AG insaturados (p = 0,002) e a razão C18:1n9/C18:00 (p = 0,021) foi menor no GG em comparação ao GC durante a gestação; entretanto, a razão C16:00 / C18:2n6 (p = 0,018) foi maior no GG em comparação ao GC no mesmo período. Total AG (p = 0,044) e AG insaturados (p = 0,024) foi menor no GG em comparação ao GC no período <= 25 . AG insaturados (p = 0,025) e a razão C18:1n9/C18:00 (p = 0,013) foi menor no GG em comparação ao GC no período > 25 semanas e < 34 semanas. Conclusão: gestantes portadoras de fetos com gastrosquise apresentam dieta de baixa qualidade nutricional, com alto valor calórico e pobre em aminoácidos essenciais, no período pré-concepcional, e baixos níveis séricos de ácidos graxos durante a gestação / Objective: To evaluate the nutrients intake during the preconceptional period and the serum fatty acid levels during the gestation period of pregnant women with fetuses with gastroschisis and pregnant women with normal fetuses. Methods: A prospective case-control study was conducted at the Fetal Medicine Unit at Hospital das Clínicas from July 2013 to July 2015. The gastroschisis group (GG) comprised 57 pregnant women with singleton pregnancies of less than 34 weeks with fetuses with isolated gastroschisis, and the control group (CG) comprised 114 pregnant women with normal fetuses matched for maternal age (± 2 years), gestational age (± 2 weeks), and the same preconceptional body mass index (BMI). Nutritional assessments related to the preconceptional period were obtained using the Food Consumption Frequency Questionnaire and nutrient intakes (macronutrient, micronutrient, fatty acid and amino acid) were calculated using nutrition programs: Dietwin Profissional 20 ® and Virtuanutri ®. For the evaluation of serum fatty acid levels (FA), a blood sample was collected from each subject at the time they entered the study and at the time of delivery. The FA comparison was performed during gestation and at the time of delivery. In order to evaluate whether the differences between both groups were more frequent in the first or second half of gestation, a new analysis was performed, subdividing gesta 25 weeks and < 34 weeks. Results: during the preconceptional period, the median daily calorie intake was higher (2382.43 versus 2198.81; p = 0.041) in the GG than in the CG. The median intakes of methionine (763.89 versus 906.34; p = 0.036), threonine (1248.34 versus 1437.01; p = 0.018) and chromium (54.66 versus 59.49 p = 0.014) were lower in the GG than in the CG. By analyzing the serum fatty acid levels, total FA (p = 0.008), unsaturated FA (p = 0.002) and the C18:1n9/C18:00 ratio (p = 0.021) were lower in the GG than in the CG during gestation; however, the C16:00 / C18:2n6 ratio (p = 0.018) was higher in the GG than in the CG during the indicated period. Total FA (p = 0.044) and unsaturated FA (p = 0.024) were lower in the GG than in the CG at period <= 25 w k , and unsaturated FA (p = 0.025) and the C18:1n9/C18:00 ratio (p = 0.013) were lower in the GG than in the CG at period > 25 weeks and < 34 weeks. Conclusion: Pregnant women with fetuses with gastroschisis have low-nutritional-quality diet, which is both high in calories and poor in essential amino acids during the preconceptional period, and have low serum FA levels during pregnancy

Com o objetivo de avaliar se as diferenças entre os grupos eram mais frequentes na primeira ou na segunda metade da gestação, uma nova análise foi realizada subdividindo o período gestacional 25 semanas e < 34 semanas. Resultados: no período pré-concepcional, a media diária de calorias ingerida foi maior (2382,43 vs. 2198,81; p = 0,041) no GG em comparação com GC. O consumo médio de metionina (763,89 vs 906,34; p = 0,036), treonina (1248,34 vs. 1437,01; p = 0,018) e crômio (54,66 vs. 59,49 p = 0,014) foi menor no GG em comparação ao GC. Na análise de ácidos graxos, observa-se que o total AG (p = 0,008), AG insaturados (p = 0,002) e a razão C18:1n9/C18:00 (p = 0,021) foi menor no GG em comparação ao GC durante a gestação; entretanto, a razão C16:00 / C18:2n6 (p = 0,018) foi maior no GG em comparação ao GC no mesmo período. Total AG (p = 0,044) e AG insaturados (p = 0,024) foi menor no GG em comparação ao GC no período <= 25 . AG insaturados (p = 0,025) e a razão C18:1n9/C18:00 (p = 0,013) foi menor no GG em comparação ao GC no período > 25 semanas e < 34 semanas. Conclusão: gestantes portadoras de fetos com gastrosquise apresentam dieta de baixa qualidade nutricional, com alto valor calórico e pobre em aminoácidos essenciais, no período pré-concepcional, e baixos níveis séricos de ácidos graxos durante a gestação / Objective: To evaluate the nutrients intake during the preconceptional period and the serum fatty acid levels during the gestation period of pregnant women with fetuses with gastroschisis and pregnant women with normal fetuses. Methods: A prospective case-control study was conducted at the Fetal Medicine Unit at Hospital das Clínicas from July 2013 to July 2015. The gastroschisis group (GG) comprised 57 pregnant women with singleton pregnancies of less than 34 weeks with fetuses with isolated gastroschisis, and the control group (CG) comprised 114 pregnant women with normal fetuses matched for maternal age (± 2 years), gestational age (± 2 weeks), and the same preconceptional body mass index (BMI). Nutritional assessments related to the preconceptional period were obtained using the Food Consumption Frequency Questionnaire and nutrient intakes (macronutrient, micronutrient, fatty acid and amino acid) were calculated using nutrition programs: Dietwin Profissional 20 ® and Virtuanutri ®. For the evaluation of serum fatty acid levels (FA), a blood sample was collected from each subject at the time they entered the study and at the time of delivery. The FA comparison was performed during gestation and at the time of delivery. In order to evaluate whether the differences between both groups were more frequent in the first or second half of gestation, a new analysis was performed, subdividing gesta 25 weeks and < 34 weeks. Results: during the preconceptional period, the median daily calorie intake was higher (2382.43 versus 2198.81; p = 0.041) in the GG than in the CG. The median intakes of methionine (763.89 versus 906.34; p = 0.036), threonine (1248.34 versus 1437.01; p = 0.018) and chromium (54.66 versus 59.49 p = 0.014) were lower in the GG than in the CG. By analyzing the serum fatty acid levels, total FA (p = 0.008), unsaturated FA (p = 0.002) and the C18:1n9/C18:00 ratio (p = 0.021) were lower in the GG than in the CG during gestation; however, the C16:00 / C18:2n6 ratio (p = 0.018) was higher in the GG than in the CG during the indicated period. Total FA (p = 0.044) and unsaturated FA (p = 0.024) were lower in the GG than in the CG at period <= 25 w k , and unsaturated FA (p = 0.025) and the C18:1n9/C18:00 ratio (p = 0.013) were lower in the GG than in the CG at period > 25 weeks and < 34 weeks. Conclusion: Pregnant women with fetuses with gastroschisis have low-nutritional-quality diet, which is both high in calories and poor in essential amino acids during the preconceptional period, and have low serum FA levels during pregnancy

Espectroscopia Raman dos AminoÃcidos L-metionina e DL-alanina e de Nanotubos de Carbono. / Raman spectroscopy of L-methionine and DL-alananine amino acids and Carbon Nanotubes

Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / No presente trabalho foram realizadas medidas de espalhamento Raman polarizado em cristais de L-metionina e de DL-alanina (dois aminoÃcidos) e em diversas amostras de nanotubos de carbono de parede simples (SWNT). As medidas de espalhamento Raman em cristais de L-metionina foram realizadas no intervalo espectral entre 50 cm-1 e 3100 cm-1 desde a temperatura ambiente atà a temperatura de 17 K. No intervalo de temperatura estudado a estrutura da L-metionina manteve-se estÃvel. à temperatura ambiente tambÃm foram realizadas medidas Raman em altas pressÃes hidrostÃticas. A mÃxima pressÃo atingida foi de 4,7 GPa e diversas modificaÃÃes nos modos associados Ãs unidades CO2, NH3, CC, CS, CH, CH2 e CH3 sugerem que a L-metionina sofre um transiÃÃo estrutural de fase em torno de 2,1 GPa com histerese de aproximadamente 0,8 GPa. No cristal de DL-alanina foram realizadas medidas de espalhamento Raman no intervalo de temperatura de 15 K a 295 K. Embora nenhuma mudanÃa significativa tenha sido observada neste intervalo de temperatura, os resultados sÃo importantes para se entender o comportamento de uma molÃcula fundamental na constituiÃÃo das proteÃnas. Medidas de espalhamento Raman em SWNTâs foram realizadas em amostras preparadas pela tÃcnica de arco voltÃico, utilizando-se vÃrios catalisadores metÃlicos. As amostras foram divididas em duas sÃries: A primeira com os catalisadores à base de MnNiCo e a segunda à base de FeNiCo. Em ambas as sÃries observou-se que a inserÃÃo de CÃrio (Ce) foi responsÃvel por tornar a distribuiÃÃo de diÃmetros do subconjunto ressonante com a energia 2,41 eV mais estreita. AlÃm disso, o mÃximo da distribuiÃÃo à deslocado para o azul, provavelmente em conseqÃÃncia da seleÃÃo de tubos de menor diÃmetro dentro do subconjunto estudado. A inserÃÃo de ZircÃnio (Zr) à segunda sÃrie nÃo trouxe mudanÃas significativas. Foram realizadas medidas de espalhamento Raman em funÃÃo da pressÃo hidrostÃtica em uma amostra comercial de SWNT. Como fluido transmissor foram utilizadas soluÃÃes de dois surfactantes: o dodecil sulfato de sÃdio (SDS) e o Ãcido plurÃnico F127 (F127). Devido à baixa relaÃÃo sinal-ruÃdo, nÃo foi possÃvel estudar o comportamento dos modos de respiraÃÃo radial (RBM), mas pela descontinuidade do grÃfico da freqÃÃncia em funÃÃo da pressÃo dos modos tangenciais em aproximadamente 2 GPa à provÃvel que os nanotubos sofram uma transiÃÃo de fase estrutural nessa pressÃo, com deformaÃÃo da seÃÃo circular dos tubos como predito por estudos teÃricos. Dois conjuntos de amostras contendo diferentes nÃveis de inserÃÃo de lÃtio tambÃm foram estudados por espectroscopia Raman. Cada conjunto era formado por uma amostra sem lÃtio, uma com lÃtio e a terceira com inserÃÃo parcial de lÃtio (obtida pela lavagem da amostra que contÃm lÃtio). Nos dois conjuntos a inserÃÃo foi eficiente, contudo o mecanismo de inserÃÃo à diferente de uma sÃrie para outra. Na primeira sÃrie o catalisador utilizado para a inserÃÃo de lÃtio foi LiNi0,5Co0,5O2. Com este composto o lÃtio à intercalado intersticialmente e pode ser removido quase que completamente pela lavagem da amostra. Jà na segunda sÃrie o composto utilizado foi LiCO3/NiO/CoO o que fez com que o lÃtio fosse intercalado dentro dos tubos de modo a nÃo ser removido pela lavagem da amostra. / This work describes polarized Raman scattering measurements in L-methionine and in DL-alanine (two amino acids) crystals and in several samples of single-walled carbon nanotubes (SWNT). In L-methionine crystal the Raman spectra were obtained from 17 K to 295 K in the spectral range from 50 cm-1 to 3100 cm-1, but no indication of a phase transition was observed. At room temperature, Raman scattering measurements were also performed for pressure up to 5 GPa. Several changes observed in the spectra were interpreted as due to structural phase transition undergone by L-methionine crystal at ~ 2.1 GPa. The results for decompression show that the phase transition is reversible with a hysteresis of ~ 0.8 GPa. In DL-alanine crystal the Raman spectra were obtained at temperatures from 15 K to 295 K over the spectral range 50 cm-1 - 3100 cm-1. No evidence of structural phase transition was found in this range of temperature, although information about diverse modes of the crystal were furnished. Samples of SWNTâs studied were prepared with metallic catalysts using the arc voltaic method. MnNiCo was the main compound of the first series and FeNiCo, the main compound of the second. In both sets it was observed that Cerium (Ce) insertion induces in the sub-set, probed with the 2.41 eV excitation energy, a narrowing of the diameter distribution favoring the tubes with smaller diameter. The Raman scattering measurements in a commercial sample of SWNTâs show a discontinuity at about 2 GPa. The discontinuity was represented by a changing in the slope of the frequency versus pressure for tangential modes. The measurement was performed twice, using two different solutions of surfactants, sodium dodecil sulfate (SDS) and plurocic acid F127 (F127), and the results were similar. The anomaly was interpreted as due to the deformation of the tubes as predicted theoretically. Two sets of three samples of SWNTâs containing different levels of lithium insertion were also analyzed by Raman spectroscopy. Each set of sample was formed by a sample with lithium, a sample without lithium and a sample with partial insertion of lithium. The results show that the lithium is efficiently intercalated with both lithium containing compound (LiCO3/NiO/CoO and LiNi0.5Co0.5O2), but the mechanism of intercalation differs from one to the other. The intercalation is unstable when lithium is intercalated interstitially (LiNi0.5Co0.5O2) and it can be removed almost completely by washing the sample, but if the lithium is intercalated inside the tubes (LiCO3/NiO/CoO) it can not be removed by the same process.
