In this context, the study of field cancerization, a phenomenon involved in the initial stages of oral carcinogenesis, has gained relevance for the establishment of biomarkers that may identify individuals exposed to carcinogens with the greatest risk for developing oral cancer. The first article of this study aimed to compare cytopathological and histopathological characteristics of the clinically normal mucosa adjacent to oral squamous cell carcinoma. The nuclear area of cells obtained by cytological smear of this region was also analyzed and compared to that of individuals without lesions but exposed to smoking and/or alcohol and of patients not exposed to these risk factors. Ninety patients of both sexes over 40 years old were included. In patients with carcinoma, in addition to exfoliative cytology, tissue was obtained from the area adjacent to the tumor for histopathological examination. In smears stained with Papanicolaou, the nuclei of 50 intermediate cells were measured. Both histological sections and cytological smears were classified as low or high-risk. The sensitivity, specificity and accuracy of the cytopathological diagnosis in relation to the histopathological diagnosis, considered the gold standard, were 100, 75 and 75.86%, respectively. The mean nuclear area was significantly lower (p<0.05) in the patients not exposed to the risk factors in relation to the others. The cyto-histopathological comparison of the area adjacent to oral cancer showed good sensitivity, specificity and accuracy. In conclusion, this article demonstrated that nuclear area can be used to detect early cellular changes in the oral mucosa exposed to carcinogens and that mean percentage of nuclei larger than 100 ?m2 is the most indicated method for the assessment of these changes. The second article aimed to assess genetic damage and cell proliferation rate in the field of cancerization, i.e., the clinically normal mucosa adjacent to oral carcinoma. Cytologic smears from the same scrapes used in the first article were stained with silver and with the Feulgen reaction. The mean number of AgNORs/nucleus and micronuclei (MN) was significantly higher (p<0.05) in the Tobacco/Alcohol and Oral Cancer Groups than in the Control Group. Conversely, the mean number of NBUDs was higher in the Control Group compared with the other groups. The number of AgNORs/nucleus and MN/1,000 cells provide evidence of initial oral carcinogenesis at field cancerization areas. Cutoff values for inclusion of individuals exposed to carcinogens in longitudinal monitoring were ? 3.38 AgNORs/nucleus and/or ? 3 MN/1,000 cells. A prospective model including the biomarkers assessed in this study was proposed. / A citopatologia bucal pode ser aplicada como m?todo de monitoramento em indiv?duos expostos a fatores de risco ao c?ncer de boca. O campo de canceriza??o, representando as etapas iniciais da carcinog?nese bucal, torna-se uma ?rea atrativa ao estudo de biomarcadores que poder?o ser utilizados para identificar indiv?duos expostos a carcin?genos com maior risco ao desenvolvimento do c?ncer bucal. O primeiro artigo deste estudo visou comparar as caracter?sticas citopatol?gicas e histopatol?gicas da mucosa clinicamente normal adjacente ao carcinoma espinocelular bucal. Al?m disso, a ?rea nuclear das c?lulas obtidas desta regi?o, por meio de esfrega?o citol?gico, foi mensurada e comparada ? das c?lulas de indiv?duos sem les?o expostos ao fumo e/ou ?lcool e a de pacientes n?o expostos a estes fatores de risco. Foram inclu?dos 90 pacientes de ambos os sexos com idade superior a 40 anos. Nos pacientes com carcinoma, al?m da citologia esfoliativa, foi obtido material para exame histopatol?gico da ?rea adjacente ao tumor. Nos esfrega?os, corados com Papanicolaou, foram mensurados os n?cleos de 50 c?lulas intermedi?rias. Tanto os cortes histol?gicos, quanto os esfrega?os citol?gicos foram classificados em baixo ou alto risco. Ao associar as caracter?sticas citopatol?gicas e histopatol?gicas, verificou-se sensibilidade, especificidade e acur?cia de 100%, 75% e 75,86%, respectivamente. A m?dia da ?rea nuclear foi menor no grupo n?o exposto ao fumo e ao ?lcool, com diferen?a significativa (p<0,05) em rela??o aos demais. A associa??o cito-histopatol?gica da ?rea adjacente ao c?ncer bucal apresentou boa sensibilidade, especificidade e acur?cia. Al?m disso, constatou-se que a ?rea nuclear ? pass?vel de ser utilizada para detectar altera??es celulares precoces na mucosa bucal exposta a carcin?genos, sendo a m?dia percentual de n?cleos com mais de 100 ?m2 o m?todo de avalia??o mais indicado. O segundo artigo objetivou avaliar o dano gen?tico e a velocidade de prolifera??o celular no campo de canceriza??o, ou seja, na mucosa clinicamente normal adjacente ao c?ncer bucal. Os esfrega?os citol?gicos foram corados com a t?cnica das AgNORs e rea??o de Feulgen, utilizando o mesmo raspado citol?gico obtido no primeiro artigo. A m?dia das AgNORs/n?cleo e do micron?cleo (MN) dos grupos fumo/?lcool e carcinoma bucal foi superior, com diferen?a estatisticamente significativa (p<0,05) em rela??o ao grupo controle. Este, por sua vez, obteve m?dia superior de bot?es nucleares (NBUDs) em rela??o aos grupos fumo/?lcool e carcinoma bucal. Ambos os marcadores, n?mero das AgNORs/n?cleo e MN, evidenciaram a fase inicial da carcinog?nese bucal, representada no campo de canceriza??o. Valores iguais ou superiores a 3.38 AgNORs/n?cleo e/ou 3 MN/1000 c?lulas foram identificados como ponto de corte ideal para incluir um indiv?duo exposto a carcin?genos no monitoramento longitudinal. Um modelo prospectivo dos marcadores foi sugerido.

Pesquisa do Estado da Bahia - FAPEB / The use of anabolic androgenic steroids (AAS) has grown among practitioners of recreational bodybuilding, with significant contributions of "Designer Steroids" (DS), EAA designed aiming muscle hypertrophy in healthy subjects. The abusive use of AAS in general is associated with adverse effects, one of the most worrisome is cancer development. Given that cancer is a genetic disease resulting from changes in genes critical in maintaining genomic stability and in the control of proliferation and differentiation, biomarkers able to identify these changes can take predictive value and contribute to reducing the high rates of morbidity and mortality by this disease. The aim of this study was to evaluate the effectiveness of the Cytokinesis Block Micronucleus Test (CBMN) in human lymphocytes in identifying risk groups for cancer development in users of AAS. Was collected 5ml of blood from 15 AAS users bodybuilders (G1), 20 nonusers bodybuilders (G2) and 20 nonusers sedentary (G3). Lymphocytes were cultured with blocking of cytokinesis and subsequent processing for making slides. MN analysis was performed on a minimum of 1000 binucleated lymphocytes. The occurrence of MN was significantly higher (p <0.05) in individuals of G1 compared to G2 and G3. The results indicate the sensitivity of CBMN in human lymphocytes in the identification of chromosomal damage in consequence of AAS using, however, further studies are needed to determine their potential to predict the cancer risk in users of AAS. / O uso de esteroides anabolizantes androg?nicos (EAA) tem crescido entre praticantes recreativos de muscula??o, com contribui??es significativas dos Designer Steroids (DS), EAA projetados para fins de hipertrofia muscular em indiv?duos saud?veis. O uso abusivo de EAA em geral est? associado a efeitos adversos, sendo um dos mais preocupantes o desenvolvimento de c?ncer. Tendo em vista que o c?ncer ? uma doen?a gen?tica resultante de altera??es em genes cr?ticos na manuten??o da estabilidade gen?mica e do controle da prolifera??o e diferencia??o, biomarcadores capazes de identificar estas altera??es podem assumir valor preditivo e contribuir para diminui??o das altas taxas de morbidade e mortalidade por esta doen?a. Among the many chemicals that have been identified as mutagenic action, include the Androgenic Steroids (AAS), hormones widely used in the search for improved physical performance and increased muscle mass. Objective: In this context, the aim of the development of this study was to evaluate the potential of androgenic anabolic steroid nandrolone decanoate, testosterone propionate and testosterone cypionate to induce chromosome damage, apoptosis and necrosis, using the micronucleus test in exfoliated cells from the oral mucosa of users of AAS with a view to its application as a tool in cancer prevention. Method: The study sample consisted of 55 volunteers, male, divided into two (02) groups, matched for age: 25 subjects (G1) users of nandrolone decanoate, testosterone propionate and testosterone cypionate (alone or simultaneously ) and 30 subjects in the control group (G2). The collection methodology and cytological analysis followed the protocol of Tolbert et al. (1992) and Thomas et al. (2009), which includes, in addition to micronuclei, the computation of degenerative nuclear changes indicative of apoptosis (cariorr?xis, condensed chromatin and pyknosis) and necrosis (karyolysis, cariorr?xis, condensed chromatin and pyknosis). Statistical analysis of the endpoints analyzed (micronucleus, cariorr?xis, condensed chromatin, karyolysis, pyknosis and broken eggs) was performed using the conditional test to compare proportions in situations of rare events. Results: Statistical analysis revealed no significant difference in the occurrence of micronuclei, karyolysis and broken eggs between groups. The occurrence of apoptosis was significantly greater in cells from control individuals. This fact has contributed to the reduction of environmental quality, and commitment the health of living beings that inhabit these ecosystems, including the human population. In this backdrop of reduced water quality, is the Lucrecia dam, which is a major surface water reservoirs by volume of semi-arid region of Rio Grande do Norte, and that has shown contamination by heavy metals, cyanobacteria toxic and the natural presence of Radon. The population that use this source has been showing high rates of cancer, popularly associated with the consumption of this water, with a prevalence about three times higher compared to the whole state of Rio Grande do Norte. Based on this, the present study aimed to evaluate the mutagenic potencial of surface water from the Lucrecia dam, using the Micronucleus Test in Tradescantia pallida (Trad-MN) and in human peripheral blood lymphocytes (CBMN) assay, as well as identify the concentrations of some heavy metals in this water. Water samples were collected on a dry season and a rainy season, in two distinct points. Moreover, in order to bring a completely view about the relationship of man-health-environment in this local, through the knowledge of knowing / acting environmental from residents of Lucrecia, and the use and perceptions they have about the dam of your city, a study of Environmental Perception was carried out with local residents. The results obtained for the both micronucleus test, showed significant results for the three points analyzed. The strongest mutagenic effect was observed in the dry season for both assays. Chemical analyses detected an increase of heavy metal levels in different points and season above the maximum allowed by legislation. Regarding the study of Environmental Perception with local residents, it was observed the knowledge of the environment that the residents have, as well as the strong ties and perceptions with the dam of the city. Thus, the combination of these two aspects (the genetic toxicity tests conducted in the dam together with analysis of environmental perception with the residents of Lucrecia) allowed to draw a more complete diagnosis on the local situation / A qualidade h?drica de diversos mananciais tem sido reduzida devido ? percola??o de contaminantes, seja de origem antr?pica ou natural, aumentando consideravelmente o n?vel de compostos genot?xicos nos ecossistemas aqu?ticos. Tal fato vem contribuindo para a redu??o da qualidade ambiental, bem como para o comprometimento da sa?de dos seres vivos que habitam esses ecossistemas, inclusive o homem. Visto isso, o presente estudo teve como objetivo avaliar o potencial mutag?nico da ?gua superficial do a?ude de Lucr?cia por meio do teste de micron?cleo em Tradescantia pallida e em cultura de Linf?citos Humanos, assim como identificar as concentra??es de metais pesados presentes nesta ?gua. A coleta de ?gua foi realizada em dois pontos amostrais, no per?odo de seca e no per?odo chuvoso. Ademais, com o intuito de trazer um panorama mais completo da rela??o homem-sa?de-ambiente nesta localidade, atrav?s do conhecimento do saber/agir ambiental dos moradores de Lucr?cia, assim como o uso e percep??es estes t?m sobre o a?ude do seu munic?pio, foi realizado um estudo de Percep??o Ambiental com os moradores locais. Os resultados obtidos para o teste de Micron?cleo, para ambos os modelos utilizados, mostraram resultados significantes para os pontos coletados. O per?odo de seca apresentou uma m?dia maior de micron?cleos quando comparado ao per?odo chuvoso. Foi observado tamb?m concentra??es acima do permitido pela legisla??o brasileira de alguns metais pesados. Com rela??o ao estudo de Percep??o Ambiental com os moradores locais, a an?lise dos dados permitiu observar os conhecimentos sobre meio ambiente que os moradores possuem, assim como a forte liga??o e as concep??es que os mesmos possuem com o a?ude do munic?pio. An?lise da estabilidade gen?tica de c?lulas-tronco mesenquimais humanas

The human umbilical cord as a cell source for cell therapy will help to avoid several ethical, political, religious and technical issues. One of the main issues with SC lines from different sources, mainly those of embryonic origin, is the possibility of chromosomal alterations and genomic instability during in vitro expansion. Cells isolated from one umbilical cord exhibited a rare balanced paracentric inversion, likely a cytogenetic constitutional alteration, karyotype: 46,XY,inv(3)(p13p25~26). Important genes related to cancer predisposition and others involved in DNA repair are located in 3p25~26. Titanium is an excellent biomaterial for bone-implant integration; however, the use can result in the generation of particulate debris that can accumulate in the tissues adjacent to the prosthesis, in the local bone marrow, in the lymph nodes, liver and spleen. Subsequently may elicit important biological responses that aren?t well studied. In this work, we have studied the genetic stability of MSC isolated from the umbilical cord vein during in vitro expansion, after the cryopreservation, and under different concentrations and time of exposition to titanium microparticles. Cells were isolated, in vitro expanded, demonstrated capacity for osteogenic, adipogenic and chondrogenic differentiation and were evaluated using flow cytometry, so they met the minimum requirements for characterization as MSCs. The cells were expanded under different concentrations and time of exposition to titanium microparticles. The genetic stability of MSCs was assessed by cytogenetic analysis, fluorescence in situ hybridization (FISH) and analysis of micronucleus and other nuclear alterations (CBMN). The cells were able to internalize the titanium microparticles, but MSCs preserve their morphology, differentiation capacity and surface marker expression profiles. Furthermore, there was an increase in the genomic instability after long time of in vitro expansion, and this instability was greater when cells were exposed to high doses of titanium microparticles that induced oxidative stress. It is necessary always assess the risks/ benefits of using titanium in tissue therapy involving MSCs, considering the biosafety of the use of bone regeneration using titanium and MSCs. Even without using titanium, it is important that the therapeutic use of such cells is based on analyzes that ensure quality, security and cellular stability, with the standardization of quality control programs appropriate. In conclusion, it is suggested that cytogenetic analysis, FISH analysis and the micronucleus and other nuclear alterations are carried out in CTMH before implanting in a patient / C?lulas mesenquimais estromais multipotentes, tamb?m conhecidas como c?lulas-tronco mesenquimais humanas (CTMH), s?o c?lulas multipotentes utilizadas em v?rias pesquisas de terapia celular, pois apresentam a capacidade de se diferenciar em m?ltiplas e diferentes linhagens, t?m grande capacidade de autorrenova??o e de expans?o in vitro, excelentes propriedades imunossupressoras e s?o capazes de secretar mol?culas bioativas que exercem efeitos tr?ficos. O cord?o umbilical ? uma fonte de CTMH cuja extra??o n?o necessita de um procedimento invasivo, al?m de n?o envolver controv?rsias ?ticas, pol?ticas e religiosas. Um dos problemas que envolvem linhagens de CTMH de diferentes fontes ? a possibilidade de ocorr?ncia de altera??es cromoss?micas e instabilidade gen?tica, que podem aparecer durante a expans?o in vitro. Al?m disso, as CTMH de um dos cord?es apresentaram uma altera??o cromoss?mica constitucional: invers?o parac?ntrica no bra?o curto do cromossomo 3, cari?tipo: 46,XY,inv(3)(p13p25~26). Em 3p25-26, est?o localizados v?rios genes de grande import?ncia biol?gica, como genes envolvidos com o reparo de DNA e outros respons?veis pelo desenvolvimento de tumores. O tit?nio ? um dos materiais mais utilizado para fabrica??o de implantes ortop?dicos e dent?rios, e ? considerado um excelente biomaterial, entretanto, as part?culas derivadas de pr?teses acumulam-se nos tecidos periprost?ticos e na medula ?ssea local, disseminam-se para linfonodos, f?gado e ba?o. As implica??es biol?gicas em longo prazo da dissemina??o sist?mica de part?culas de metais e seus efeitos cito e genot?xicos n?o est?o bem caracterizados. Neste trabalho investigamos a estabilidade gen?tica de CTMH isoladas da veia do cord?o umbilical durante a expans?o in vitro, ap?s a criopreserva??o, e em diferentes condi??es de cultivo, na presen?a e na aus?ncia de tit?nio, antes e ap?s o aparecimento de c?lulas senescentes no cultivo. As c?lulas foram isoladas, expandidas, diferenciadas em osteoblastos, adip?citos e condroblastos e analisadas com citometria de fluxo para comprovar que s?o c?lulas-tronco mesenquimais. As CTMH foram tratadas com diferentes doses/ tempo de exposi??o ? micropart?culas de tit?nio. A avalia??o da estabilidade gen?tica das CTMH foi realizada atrav?s da an?lise do cari?tipo, de hibrida??o in situ por fluoresc?ncia (FISH) e da an?lise do micron?cleo e outras altera??es nucleares (CBMN). Ficou estabelecido que as CTMH foram capazes de internalizar as micropart?culas de tit?nio, mas as c?lulas mant?m sua capacidade de prolifera??o, diferencia??o e preservam os mesmos marcadores de membrana. Al?m disso, demonstrou-se que existe um aumento na instabilidade gen?tica com o decorrer do tempo de expans?o in vitro, e esta instabilidade foi maior na presen?a de grande concentra??o de micropart?culas de tit?nio que induzem estresse oxidativo. ? necess?rio sempre avaliar os riscos/ benef?cios da utiliza??o do tit?nio na terapia tecidual envolvendo CTMH, considerando a biosseguran?a da utiliza??o da regenera??o ?ssea guiada que utiliza CTMH e tit?nio. Mesmo n?o se utilizando o tit?nio, ? importante que o uso terap?utico de tais c?lulas seja baseado em an?lises que garantam a qualidade, seguran?a e estabilidade celular, com a padroniza??o de programas de controle de qualidade adequados. Inclusive, estudos revelam que a defici?ncia desta vitamina aumenta a forma??o de micron?cleos, estruturas citogen?ticas indicadoras de danos no DNA. Al?m disso, esta defici?ncia ? modulada pelos polimorfismos gen?ticos associados ao metabolismo do ?cido f?lico, comprometendo o desempenho das fun??es de estabilidade do genoma e, portanto, est?o sendo associados com o desenvolvimento de v?rios dist?rbios, dentre eles as fissuras orais. A partir deste contexto, foi conduzido um estudo transversal do tipo caso-controle n?o pareado com o objetivo de avaliar a frequ?ncia de biomarcadores de instabilidade gen?mica, sua rela??o com polimorfismos gen?ticos do metabolismo do folato, e se essas vari?veis est?o associadas com o desenvolvimento das FL/PNS em crian?as do Rio Grande do Norte, Brasil. Com o devido consentimento dos participantes, realizou-se uma entrevista com os respons?veis para obten??o de dados epidemiol?gicos, como tamb?m procedeu-se a coleta sangu?nea das crian?as para a realiza??o dos testes. Foi executado o ensaio do micron?cleo com bloqueio da citocinese para calcular a frequ?ncia de micron?cleos (MN), pontes nucleoplasm?ticas (PN) e brotos nucleares (BN). A partir da extra??o do DNA gen?mico, avaliou-se os polimorfismos da metilenotetrahidrofolato redutase C677T e A1298C, metionina sintase A2756G, metionina sintase redutase A66G e do receptor de folato reduzido A80G pela t?cnica de rea??o em cadeia da polimerase associada ao polimorfismo de tamanho do fragmento de restri??o (PCR-RFLP). As crian?as com FL/PNS apresentaram maior frequ?ncia basal de MN, PN e BN que o grupo de crian?as controle (p < 0,001), e nenhum dos polimorfismos avaliados modificaram significativamente a frequ?ncia destes biomarcadores. Al?m disso, crian?as com FL/PNS apresentaram 2,3 vezes mais risco de exibir altas frequ?ncias de MN (p = 0,043) segundo modelo de regress?o log?stica bin?ria. A alta instabilidade do genoma nas crian?as com fissuras orais sugere que os eventos genot?xicos, em particular os que promovem quebras na dupla fita do DNA e n?o s?o devidamente reparados formando micron?cleos, representam fatores relevantes no desenvolvimento das fendas labiopalatinas n?o-sindr?micas. / The non-syndromic clefts of the lip and/or palate (NSCL/P) are common birth defects in humans characterized by an incomplete development of cellular structures that separate the nasal cavity from the oral cavity, and they are caused due to an interaction between genetic and environmental factors. Data from case-control and dietary intervention studies indicates that maternal supplementation with multivitamins containing folic acid prevents the formation of oral clefts. It is known that folic acid plays a role in essential cellular functions, such as nucleotides synthesis for DNA repair, which contribute to the protection of genome integrity from damage events generated by endogenous and/or exogenous factors. In fact, studies show that a deficiency in this vitamin increases micronuclei formation, a cytogenetic structure that indicates misrepair of damaged DNA. Furthermore, this deficiency is modulated by genetic polymorphisms associated with folic acid metabolism, affecting the performance of genome stability functions and therefore, it has been associated with the development of various disorders, including oral clefts. From this context, it was planned a unpaired case-control cross-sectional study in order to assess the frequency of genome instability biomarkers, their relationship with genetic polymorphisms in folate metabolism, and if these variables are associated with the development of NSCL/P in children from a Northeast region at Brazil. For this research, it was recruited 48 NSCL/P patients and 18 control children, respectively, at the Pediatric Hospital Professor Heriberto Ferreira Bezerra (HOSPED)/ UFRN and at schools in Natal city. With the participants consent, they were interviewed with a standard questionnaire to obtain epidemiological data, and the children underwent a blood sampling for the tests. It was performed the cytokinesis-block micronucleus assay to estimate the frequency of micronuclei (MN), nucleoplasmic bridges (NPB) and nuclear buds (NB). Also, from the genomic DNA extraction, it was evaluated by PCR-RFLP the polymorphisms of methylenetetrahydrofolate reductase C677T and A1298C, methionine synthase A2756G, methionine synthase reductase A66G and reduced folate carrier A80G. Children with NSCL/P had higher baseline frequency of MN, NPB and NB than the control group (p < 0.001), and none of the evaluated polymorphisms significantly modified the frequency of these biomarkers. In addition, children with clefts had 2.3 times more risk of displaying high frequency of MN (p = 0.043) according to the binary logistic regression model. The high genomic instability in children with oral clefts suggests that genotoxic events that promote double strand breaks on DNA and are not properly repaired, thus originating micronuclei, represent significant factors in the development of non-syndromic cleft lip/ palate.

The calcium carbonate (CC), in proportions of 1%, 5% and 10%, was added to the GIC powder. A polytetrafluoroethylene matrix (10 mm diameter and 3 mm height) was used to make the samples, and four groups were obtained (n = 4): G1 ? only GIC (control); G2 ? GIC with 1% CC; G3 ? GIC with 5% CC; G4 ? GIC with 10% CC. The preparation of the samples was carried out in accordance with ISO 10993-12. The MTT test was used to evaluate the cell cytotoxicity, and the micronucleus and comet tests were performed to evaluate the genotoxicity, by using a mouse fibroblast cell culture of the NIH/3T3 lineage. According to MTT test, the samples with 1% and 5% CC showed a higher cytotoxic potential, and the samples with 10% CC presented a cellular viability index comparable to the GIC. The micronucleus test showed that GIC with 10% CC produced an improvement in cell proliferative potential (IPBC).CIV with 5% CC had a reduction in IPBC that did not compromise the material from the genotoxic point of view. In the comet test, groups with the addition of CC had a small increase in genotoxic potential compared with GIC. It was concluded that the addition of 10% CC to the GIC had a low cytotoxic potential and it is feasible for use in the cellular environment, and the addition of 1%, 5% and 10% CC to the GIC did not induce genetic damage. / Este estudo teve o objetivo de avaliar a biocompatibilidade do cimento de ion?mero de vidro (CIV) convencional modificado por carbonato de c?lcio de conchas marinhas por meio dos testes de citotoxicidade e genotoxicidade celular. O carbonato de c?lcio (CC), nas propor??es de 1%, 5% e 10%, foi adicionado ao p? do CIV. Utilizando uma matriz de politetrafluoretileno (10 mm de di?metro e 3 mm de altura), foram confeccionadas as amostras, sendo obtidos quatro grupos (n=4): G1 ? apenas CIV (controle); G2 ? CIV com 1% de CC; G3 ? CIV com 5% de CC; G4 ? CIV com 10% de CC. A prepara??o das amostras foi realizada de acordo com a norma ISO 10993-12. The objective of this study, using the micronucleus test on the bone marrow of mice and the comet assay on human lymphocytes, was to evaluate the genotoxic and cytotoxic effects of four filling pastes that are used in this therapy: zinc oxide, calcium hydroxide P.A., mineral trioxide aggregate and an iodoform paste (Guedes-Pinto paste). To perform the micronucleus test, male Swiss mice (Mus musculus) were divided into groups of ten animals each: four groups were each exposed to one of the filling pastes, administered intraperitoneally at dilutions of 1/10, 1/50, 1/500 and 1/1000. Cyclophosphamide was used as the positive control. The negative controls used were the dilution vehicles: dimethylsulfoxide (DMSO) for the Guedes paste and zinc oxide; and phosphate-buffered saline solution (PBS) for the calcium hydroxide P.A. and mineral trioxide aggregate. The animals were sacrificed 24 h and 48 h after the treatment. The bone marrow was extracted, in order to calculate the micronucleus occurrence rate in 1000 polychromatic erythrocytes (PCE) in each of the animals, under an optical microscope (1000 X), in a blinded test. Cytotoxicity was evaluated by determining the PCE/NCE (normochromatic erythrocyte) ratio in 200 erythrocytes/animal. For the comet assay, human lymphocytes were cultured in different dilutions of each of the filling pastes (1:500, 1:750, 1:1000 and 1:2000), for 3 h at 37 ?C, under an atmosphere containing 5% CO2. Two positive controls were used: methyl-methanesulfonate (0.4 ?M) for the calcium hydroxide P.A. and mineral trioxide aggregate; and doxorubicin (0.6 ?M) for the Guedes paste and zinc oxide. Two negative controls were also used here: distilled water for the calcium hydroxide P.A. and mineral trioxide aggregate; and DMSO for the Guedes paste and zinc oxide. Comets were identified by means of fluorescence microscopy (400 X), and 100 of them were counted on each of the three slides that were analyzed for each drug test. The statistical analysis on the results from the micronucleus test was performed using the conditional test for comparing proportions in situations of rare events. Analysis of variance, followed by the Tukey test, was used to assess the PCE/NCE ratio obtained from the different treatments and also to compare the means from the DNA damage indices that were obtained through the comet test, using the Prisma software, version 4.0. The micronucleus occurrence rate was significantly higher among the animals treated with Guedes paste, at all the dilutions tested and at both sacrifice times and also among the animals treated with zinc oxide and sacrificed 48 h after treatment at the dilutions of 1:50, 1:500 and 1:1000. Cytotoxic effects from these pastes were detected in the animals sacrificed at both times. Calcium hydroxide P.A. and mineral trioxide aggregate did not present any cytotoxic or genotoxic effects. The results obtained from the comet assay also showed that zinc oxide and Guedes paste presented genotoxicity, whereas calcium hydroxide P.A. and mineral trioxide aggregate did not. These results show that there is a need to reassess the use of zinc oxide and Guedes paste and provide encouragement for conducting additional studies to evaluate the genotoxicity and cytotoxicity of these pastes. / A terapia pulpar de dentes dec?duos se constitui no ?ltimo recurso de preven??o da perda dent?ria, mas, dentre os produtos empregados para tal, as denominadas pastas obturadoras, nenhum ? considerado como ideal. O objetivo deste trabalho foi avaliar, com o uso do teste de micron?cleo em medula ?ssea de camundongo e do ensaio cometa em linf?citos humanos, os efeitos citot?xicos e genot?xicos de quatro pastas obturadoras utilizadas nesta terapia: ?xido de zinco, hidr?xido de c?lcio P.A., agregado tri?xido mineral e uma pasta iodoformada (pasta Guedes-Pinto). Para realiza??o do teste de micron?cleo, camundongos Swiss (Mus musculus), machos, foram divididos em grupos de dez animais, que foram expostos ?s pastas obturadoras, administradas via intraperitoneal nas dilui??es de 1/10, 1/50, 1/500 e 1/1000. Ciclofosfamida foi utilizada como controle positivo. Os controles negativos foram: dimetilsulf?xido (DMSO) para a pasta Guedes-Pinto e ?xido de zinco; e solu??o salina tamponada (PBS) para o hidr?xico de c?lcio P.A. e agregado trioxido mineral. Avalia??o do potencial genot?xico e citot?xico associado a queima artesanal da castanha de caju no Munic?pio de Jo?o C?mara

Despite the social and economic importance of the cashew nut, its production is still carried out artisanally. One of the main problems encountered in the cashew production chain are the conditions under which the roasting of the nut occurs to obtain the kernel from the shell. In the present study was conducted a biomonitoring of the genotoxic and cytotoxicity effects associated with the elements from the cashew nut roasting in Jo?o C?mara - RN, semi-arid region of Brazil. To assess the genotoxic was used the bioassay of micronucleus (MN) in Tradescantia pallida. In addition, it was performed a comparative between the Tradescantia pallida and KU-20 and other biomarkers of DNA damage, such as the nucleoplasmic bridges (NBP) and nuclear fragments (NF) were quantified. The levels of particulate matter (PM1.0, PM2.5, PM10) and black carbon (BC) were also measured and the inorganic chemical composition of the PM2.5 collected was determined using X-ray fluorescence spectrometry analysis and the assessment of the cytotoxicity by MTT assay and exclusion method by trypan blue. . For this purpose, were chosen: the Amarel?o community where the roasting occurs and the Santa Luzia farm an area without influence of this process. The mean value of PM2.5 (Jan 2124.2 &#956;g/m3; May 1022.2 &#956;g/m3; Sep 1291.9 &#956;g/m3) and BC (Jan 363.6 &#956;g/m3; May 70.0 &#956;g/m3; Sep 69.4 &#956;g/m3) as well as the concentration of the elements Al, Si, P, S, Cl, K, Ca, Ti, Cr, Mn, Fe, Ni, Cu, Zn, Se, Br and Pb obtained at Amarel?o was significantly higher than at Santa Luzia farm. The genotoxicity tests with T. pallida indicated a significant increase in the number of MN, NBP and NF and it was found a negative correlation between the frequency of these biomarkers and the rainfall. The concentrations of 200 &#956;g/mL and 400 &#956;g/mL of PM2.5 were cytotoxic to MRC-5 cells. All together, the results indicated genotoxicity and citotoxicity for the community of Amarel?o, and the high rates of PM2.5 considered a potential contributor to this effect, mainly by the high presence of transition metals, especially Fe, Ni, Cu, Cr and Zn, these elements have the potential to cause DNA damage. Other nuclear alterations, such as the NPBs and NFs may be used as effective biomarkers of DNA damage in tetrads of Tradescantia pallida. The results of this study enabled the identification of a serious occupational problem. Accordingly, preventative measures and better practices should be adopted to improve both the activity and the quality of life of the population. These measures are of fundamental importance for the sustainable development of this activity. / O Brasil ? o terceiro maior produtor mundial de castanha de caju e apesar da import?ncia social e econ?mica, sua produ??o ainda ? realizada de forma artesanal. Um dos maiores problemas da cadeia produtiva do caju s?o as condi??es nas quais ocorre a queima artesanal da castanha para se obter a am?ndoa. No presente estudo foi realizado um biomonitoramento do potencial genot?xico e avalia??o da citotoxicidade associada aos elementos oriundos da queima artesanal da castanha de caju no munic?pio de Jo?o C?mara - RN, semi-?rido brasileiro. Para a avalia??o genot?xica foi utilizado o bioensaio de micron?cleo (MN) em Tradescantia pallida. Al?m disso, foi realizado um comparativo quanto a sensibilidade da T. pallida frente ao clone KU-20 e outros biomarcadores de danos no DNA, tais como as pontes nucleoplasm?ticas (PNP) e fragmentos nucleares (FN) foram quantificados. A avalia??o citot?xica se deu pelo ensaio MTT e m?todo de exclus?o por tripan blue. As concentra??es de material particulado (MP1,0, MP2,5, MP10) e black carbon (BC) foram determinadas e a composi??o inorg?nica do MP2.5 definida pela t?cnica de fluoresc?ncia de raios-X. Foram definidos dois pontos testes: Comunidade do Amarel?o (local de queima da castanha de caju) e Fazenda Santa Luzia (sem influ?ncia da atividade). Os valores m?dios obtidos para o MP2,5 (Jan - 2124,2 &#956;g/m3; Mai 1022,2 &#956;g/m3; Set 1291,9 &#956;g/m3) e BC (Jan 363,6 &#956;g/m3; Mai 70 &#956;g/m3; Set 69,4 &#956;g/m3), bem como a concentra??o dos elementos Al, Si, P, S, Cl, K, Ca, Ti, Cr, Mn, Fe, Ni, Cu, Zn, Se, Br e Pb obtidos no Amarel?o foram significativamente maiores que na Fazenda Sta. Luzia. Os testes de genotoxicidade com T. pallida indicaram um aumento de 2-7 vezes maior na frequ?ncia de MN para o Amarel?o. Os outros biomarcadores tamb?m apresentaram sua frequ?ncia aumentada. Al?m disso, verificou-se uma correla??o negativa entre a freq??ncia de MN, PNP e FN com a precipita??o pluviom?trica. As concentra??es de 200 &#956;g/mL e 400 &#956;g/mL do MP2,5 em suspens?o foram citot?xicas para as c?lulas MRC-5. O conjunto dos resultados indicaram genotoxicidade e citotoxicidade para a comunidade do Amarel?o, sendo as altas concentra??es de MP2,5 um dos prov?veis contribuintes para esse efeito, principalmente pela elevada presen?a de metais de transi??o, sobretudo Fe, Ni, Cu, Cr e Zn, que potencialmente causam les?es no DNA. Outras altera??es nucleares, como PNP e FN podem ser utilizadas como biomarcadores efetivos de danos no DNA em t?trades de T. pallida. Os conjunto dos resultados possibilitaram a identifica??o de um problema ocupacional grave, com s?rios riscos aos trabalhadores que exercem a atividade. Diante disto, a ado??o de medidas preventivas e de melhores pr?ticas, s?o de fundamental import?ncia para o desenvolvimento sustent?vel da atividade e melhoria na qualidade de vida dos trabalhadores.
