Atualmente, existem evidências de que a exposição crônica a concentrações relativamente baixas de mercúrio poderia estar iniciando processos genotóxicos (dano ao DNA) em humanos. Porém, foram realizados até agora poucos estudos epidemiológicos com populações amazônicas expostas que não incluíram uma comparação com uma população controle. O objetivo do presente estudo foi identificar a técnica mais adequada para analisar gentoxicidade mercurial em populações amazônicas e estabelecer os valores normais de genotoxicidade em uma população ribeirinha amazônica. Para a realização dos testes in vitro foram aplicadas e comparadas duas técnicas tradicionais de detecção de genotoxicidade (micronúcleos e aberrações cromossômicas). Culturas primárias de linfócitos sangüíneos de voluntários de Belém foram expostas a concentrações relativamente baixas de metilmercúrio (1-500μg/l ou 0,004-2 μM). O índice mitótico (proporção de células em metáfase) originado com a técnica de detecção de aberrações cromossômicas revelou-se como o parâmetro mais sensível à genotoxicidade mercurial. Após a identificação da técnica e o parâmetro mais sensível à genotoxicidade mercurial, essa técnica foi aplicada para estudar uma população ribeirinha amazônica que funcionasse como controle para os estudos de genotoxicidade mercurial que estão sendo feitos. Foi selecionada a população de Panacauera, e a média do índice mitótico encontrado nos indivíduos dessa população foi de 0.077 ± 0.045. Os valores de índice mitótico detectados apresentaram uma variabilidade que não esteve relacionada com a idade ou o sexo. Quando esses valores foram comparados com os valores de Brasília Legal (comunidade exposta ao metilmercúrio) registrados na literatura, foi verificado que para alguns grupos o índice mitótico de Brasília Legal foi inferior ao de Panacauera, o que indicaria uma inibição da progressão do ciclo celular e/ou uma perda da capacidade proliferativa causada pela intoxicação mercurial. Estes resultados apóiam a idéia de que o índice mitótico poderia servir como parâmetro essencial para o diagnóstico precoce do dano causado pela exposição mercurial e contribuem para o escasso conhecimento epidemiológico sobre as conseqüências que está tendo a exposição crônica de mercúrio nas populações da Amazônia. / Mercury is an important environmental pollutant for the world and, specially, for the Amazon. Presently, there are some evidences about chronic exposure to relatively low concentrations of mercury initiating genotoxic processes (DNA damage) in humans. However, to date, few epidemiological studies were carried out with Amazonian populations exposed to mercury, but no study included a population as a control to compare. The aim of this study was to identify the technique more adequate for analyzing mercury genotoxicity in Amazonian populations and to establish control values of genotoxicity in an Amazonian riverside population. To carry out in vitro tests, two traditional methods to detect genotoxicity (micronuclei and chromosomal aberrations) were applied and compared. Primary cultures of blood lymphocytes of volunteers from Belém, were exposed to relatively low concentrations of methylmercury (1-500μg/l or 0,004-2 μM). Mitotic index (proportion of cells in metaphase) originated with the method of detection of chromosomal aberrations was the parameter more sensitive to mercury genotoxicity. After identification of the method and the parameter more sensitive to mercury genotoxicity, this method was applied to study an Amazonian riverside population as a control for studies about mercury genotoxicity. Panacauera was selected as control population and mitotic index for this population was 0.077 ± 0.045. Detected values of mitotic index showed variability, not related to age or sex. When these values were compared to the values of Brasilia Legal (community exposed to methylmercury) registered in literature, mitotic index of Brasilia Legal for some groups was below mitotic index of Panacauera, pointing to an inhibition of the cell-cycle progression and/or loss of proliferative capacity due to mercury intoxication. These results support the idea that mitotic index may serve as an essential parameter for the early diagnosis of the damage provoked by mercury exposure, and they contribute to the epidemiological knowledge about the consequences of the chronic exposure with mercury in Amazonian populations.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / A mutagenicidade do material particulado é atribuída primeiramente aos hidrocarbonetos policíclicos aromáticos (HPA). Investigamos a atividade mutagênica do material particulado (MP2,5) em amostras coletadas em três pontos da cidade do Rio de Janeiro. As coletas foram realizadas com auxílio de um amostrador de grande volume na Avenida Brasil, no campus da Universidade do Estado do Rio de Janeiro e no Túnel Rebouças em filtros de fibra de vidro. Metade de cada filtro foi submetido à extração por sonicação com o solvente diclorometano. Seis HPA foram identificados e quantificados por cromatografia gasosa com espectrometria de massa (GC/MS). Após a análise química as concentrações dos HPA obtidos foram correlacionados ao fatores físicos, além de ser realizado avaliação de risco para cada HPA estudado. Linhagens de Salmonella typhimurium (TA98 e derivadas TA98/1.8-DNP6, YG1021 e YG1024) foram utilizadas no ensaio de mutagenicidade e tratadas (10-50 g/placa) com extrato orgânico na presença e na ausência de metabolização exógena. Células de raiz de cebola foram tratadas com extratos orgânicos nas concentrações (5-25g/mL). A alta umidade encontrada no Túnel Rebouças pode ter influenciado na deposição de cinco dos seis HPA estudados em material particulado. Além disso, em diferentes condições de tráfego, motoristas de ônibus que cruzam a Avenida Brasil e o Rebouças túnel estão expostos ao risco induzidos por HPA na ordem de 10-6. Mutagenicidade foi detectada tanto na presença quanto na ausência de metabolização, para as linhagens YG1021 e YG1024 nos três pontos, sugerindo a presença de nitro e amino derivados de HPA. As amostras do Túnel Rebouças apresentaram os maiores valores para rev/g e rev/m3. Estes resultados podem estar relacionados ao longo trajeto e a restrita ventilação. Efeito citotóxico foi detectado pelo ensaio Allium cepa nos três pontos de monitoramento. Além disso os extratos orgânicos provenientes das coletas da Avenida Brasil, UERJ e do Túnel Rebouças induziram efeito clastogênico em células de raiz de Allium cepa / The mutagenicity of airborne particles from combustion is attributed primarily to polycyclic aromatic hydrocarbons (PAHs). We investigated the mutagenic activity of particulate matter (PM2.5) samples collected from three sites in Rio de Janeiro. Samples were collected using a high-volume sampler at Avenida Brasil, the campus of the Rio de Janeiro State University, and Rebouças tunnel. Half of each filter was submitted to sequential extraction by sonication with dichloromethane. Six PAHs were quantified by gas chromatography/mass spectrometry (GC/MS). Salmonella typhimurium TA98 and the derivative strains TA98/1.8-DNP6, YG1021 and YG1024 used in mutagenicity assays were treated (10-50 g/plate) with and without exogenous metabolization. Onion root cells were treated with organic extracts concentrations (5-25g/mL). The high humidity detected in the Rebouças Tunnel may have influenced the deposition of five of the six PAHs studied in particulate matter. Moreover, in different traffic conditions can put the bus drivers that cross the Brasil Avenue and Rebouças tunnel at the risk of exposure induced by HPA in the order of 10-6. Independently of exogenous metabolization, mutagenicity was detected for strains YG1021 and YG1024 at all the sites, suggesting the presence of nitro and amino derivatives of PAHs. Rebouças tunnel presented the highest values for rev/g and rev/m3. These could be related to the fact that this long, enclosed passageway and restricts ventilation. Cytotoxic effect was detected by the Allium cepa test in the three monitoring sites. Also organic extracts from Brazil Avenue, UERJ and Rebouças Tunnel showed clastogenic effect in Allium cepa roots

Dentre os efluentes domésticos, podemos citar uma gama de poluentes, tais como químicos de diversas categorias, além de contaminações por agentes biológicos diversos. Já os efluentes industriais contêm poluentes orgânicos e/ou inorgânicos, dependendo da atividade industrial. Baseando-se nestes dados, este trabalho teve como objetivo investigar, por meio de ensaios biológicos com dois organismos-teste, a possível presença de contaminantes com potencial citotóxico, genotóxico e mutagênico, que são despejados ao longo do rio Preto, inclusive na Represa Municipal de São José do Rio Preto. O material biológico utilizado neste estudo constituiu-se de sementes de Allium cepa (cebola) e peixes da espécie Oreochromis niloticus (Tilápia). Coletas de águas foram realizadas, sazonalmente, nos meses de agosto de 2006 e 2007 (estação seca) e março de 2007 e 2008 (estação chuvosa), em seis pontos distintos: Ponto 1 (P1), 8 km antes do represamento; Ponto 2 (P2), 1 km antes do represamento; Ponto 3 (P3), local de despejo do esgoto; Ponto 4 (P4), margem oposta do despejo do esgoto; Ponto 5 (P5), saída do represamento; Ponto 6 (P6), 1 km após o represamento. Análises químicas foram realizadas para todas as coletas realizadas. Para a realização do estudo, 100 sementes de Allium cepa foram submetidas à germinação, em placa de Petri, em amostras de águas coletadas nos seis diferentes pontos do rio Preto, em água ultra pura (controle negativo) e em uma substância reconhecidamente aneugênica (Trifluralina - controle positivo), sempre à temperatura ambiente... / Due to increasing population and industrial expansion observed in recent decades, the environment has received a significant increased burden of domestic industrial and agricultural sewerage, which can cause severe impacts on ecosystems, and a potential damage to human health as well. A wide range of harmful pollutants can be found in domestic effluent, such as chemicals from various categories, in addition to contamination by various biological agents. On the other hand, industrial effluents contain organic and / or inorganic pollutants, depending on industrial activity. Based on these data, this study aimed to investigate, by means of biological tests with two test-organism, the possible presence of contaminants with cytotoxic, genotoxic and mutagenic potential, which are dumped along the Preto river, an important river that flows in the region of Sao Jose do Rio Preto/SP. The biological material used in this study consisted of seeds of Allium cepa (onion) and one specie of fish (Tilapia: Oreochromis niloticus). Water samples were taken seasonally in August 2006 and 2007 (dry season) and March 2007, and 2008 (rainy season), in six distinct sites: Site 1 (S1), 8 km before the damming, Site 2 (S2), 1 km before the damming, Site 3 (S3), place of sewerage discharge; Site 4 (S4), opposite margin of sewage discharge, Site 5 (S5), end of the damming; Site 6 (S6) 1 km after damming. Chemical analyses were performed for all collected samples. For the study, 100 seeds of A. cepa were submitted to germination in Petri dishes with samples water from six different sites of the Preto river, Ultra pure water (negative control), and with an aneugenic substance (Trifluralin - positive control). Firefly luciferases and genes are among the most used reporter gene in bioluminescent biosensors. Firefly luciferases are pH-sensitive, exhibiting a red shifted bioluminescence spectra on the presence of metals, high temperatures and acidic pH, being this last property considered unusual for the most analytical applications. Nowadays most luminescent biosensors to metals and pH are fluorescent, and few of them are ratiometric based on spectral changes. Bioluminescent biosensors despite been less common, have same advantages as low background and does not need UV light irradiation, bring no photodamage to the cell. The aim of this project was: (I) study the applicability of the use of Macrolampis sp2 firefly luciferase and other pH-sensitive luciferases as spectral intracellular ratiometric pH biosensor; (II) apply these pH ratiometric biosensors in mammalian cells to investigate its physiology in real time and (III) developing, by engineering the pH sensor region of Macrolampis sp2 firefly luciferase, a ratiometric biosensor specific to metals. We obtained a relation between the pH and the ratio of the bioluminescence at green and red region of the spectra, allowing estimate ratiometrically the intracellular pH of bacteria and mammalian cells. Besides that, we confirmed its use to cellular image of pH and observed that occurs an alkalinization of the cytosol and nucleus during cell division and apoptosis. We demonstrate the applicability of firefly luciferases, in special the luciferase of Macrolampis sp2, as a ratiometric biosensor to metals and intracellular pH. The existence of a linear relationship between the concentrations of metals like cadmium, mercury and zinc, and the ratio of the bioluminescence at green and red region of the spectra, allowed also estimate ratiometrically, for the first time, concentrations of metals less than 100 µM, enabling the use of firefly luciferases as bioavailability indicator to toxic and potential toxic metals. / Bioluminescência, a emissão de luz fria e visível por organismos vivos é amplamente utilizada em biossensores. Luciferases de vagalumes e seus genes estão entre os genes repórteres mais utilizados em biossensores bioluminescentes. Luciferases de vagalumes são sensíveis ao pH, apresentando um deslocamento do espectro de bioluminescência para o vermelho na presença de metais, em temperaturas elevadas e pH ácido, sendo esta última propriedade considerada sem utilidade para a maioria das aplicações analíticas. Atualmente a maioria dos biossensores luminescentes para metais e pH são fluorescentes, poucos deles são raciométricos baseados nas mudanças espectrais. Biossensores bioluminescentes apesar de serem menos comuns, possuem algumas vantagens como baixo background e não necessitam de irradiação com luz UV, não causando fotodanos às células. Os objetivos desse projeto foram: (I) investigar a aplicabilidade de uso da luciferase do vagalume Macrolampis sp2 e outras luciferases pH-sensitivas como biossensor espectral raciométrico intracelular de pH; (II) aplicar esses biossensores raciométricos de pH em células de mamíferos para investigar sua fisiologia em tempo real e (III) desenvolver, por engenharia da região sensora de pH da luciferase de Macrolampis sp2, um biossensor raciométrico específico para metais. Obtivemos uma relação entre o pH e a razão da bioluminescência nas regiões do verde e do vermelho do espectro, permitindo estimar raciometricamente o pH intracelular de bactérias e células de mamíferos. Além disso, confirmamos seu uso para imagem celular de pH, e observamos que ocorre uma alcalinização do citosol e núcleo durante os processos de divisão celular e apoptose. Demonstramos a aplicabilidade das luciferases de vagalumes, em especial a luciferase de Macrolampis sp2, como biossensor raciométrico para metais e pH intracelular. Half of each filter was submitted to sequential extraction by sonication with dichloromethane. Six PAHs were quantified by gas chromatography/mass spectrometry (GC/MS). Salmonella typhimurium TA98 and the derivative strains TA98/1.8-DNP6, YG1021 and YG1024 used in mutagenicity assays were treated (10-50 g/plate) with and without exogenous metabolization. Onion root cells were treated with organic extracts concentrations (5-25g/mL). The high humidity detected in the Rebouças Tunnel may have influenced the deposition of five of the six PAHs studied in particulate matter. Moreover, in different traffic conditions can put the bus drivers that cross the Brasil Avenue and Rebouças tunnel at the risk of exposure induced by HPA in the order of 10-6. Independently of exogenous metabolization, mutagenicity was detected for strains YG1021 and YG1024 at all the sites, suggesting the presence of nitro and amino derivatives of PAHs. Rebouças tunnel presented the highest values for rev/g and rev/m3. These could be related to the fact that this long, enclosed passageway and restricts ventilation. Cytotoxic effect was detected by the Allium cepa test in the three monitoring sites. Also organic extracts from Brazil Avenue, UERJ and Rebouças Tunnel showed clastogenic effect in Allium cepa roots

Adaptive Evolution und Screening bei Cyanobakterien

Ziel dieser Arbeit war die Erhöhung der Temperaturtoleranz des Cyanobakteriums Synechocystis sp. PCC 6803 mittels ungerichteter Mutagenese und adaptiver Evolution. Trotz des erneuten Interesses an Cyanobakterien und Mikroalgen in den letzten Jahren, gibt es nur relativ wenige aktuelle Studien zum Einsatz dieser Methoden an Cyanobakterien. Zur Analyse eines mittels Mutagenese erzeugten Gemischs an Stämmen, ist es von großem Vorteil Hochdurchsatz-Methoden zur Kultivierung und zum Screening einsetzen zu können. Auf Basis eines Pipettierroboters wurde solch eine Plattform für phototrophe Mikroorganismen neu entwickelt und folgend stetig verbessert. Die Kultivierung erfolgt in 2,2ml Deepwell-Mikrotiterplatten innerhalb einer speziell angefertigten Kultivierungskammer. Schüttelbedingungen, Beleuchtung, Temperatur und CO2-Atmosphäre sind hierbei vollständig einstellbar.Die Plattform erlaubt semi-kontinuierliche Kultivierungen mit automatisierten Verdünnungen von hunderten Kulturen gleichzeitig. Automatisierte Messungen des Wachstums, des Absorptionsspektrums, der Chlorophyllkonzentration, MALDI-TOF-MS sowie eines neu entwickelten Vitalitätsassays wurden etabliert. Für die Mutagenese wurden die Letalität- und die nicht-letale Punktmutationsrate von ultravioletter Strahlung und Methylmethansulfonat für Synechocystis charakterisiert. Synechocystis wurde mit den so ermittelten optimalen Dosen mehrfach behandelt und anschließend einer in vivo Selektion unterzogen. Somit wurde dessen Temperaturtoleranz um bis zu 3°C erhöht. Über die Screeningplattform wurden die thermotolerantesten monoklonalen Stämme identifiziert. Nach einer Validierung wurde das vollständige Genom der Stämme sequenziert. Hierdurch wurden erstmals Mutationen in verschiedenen Genen mit der Langzeittemperaturtoleranz von Synechocystis in Verbindung gebracht. Bei einigen dieser Gene ist es sehr unwahrscheinlich, dass sie mittels anderer Verfahren hätten identifiziert werden können. / The goal of this work was the increase of the thermal tolerance of the cyanobacteria Synechocystis sp. PCC 6803 via random mutagenesis and adaptive evolution. Even with the renewed interest in cyanobacteria in the recent years, there is relatively limited current research available on the application of these methods on cyanobacteria. To analyse a mixture of various strains typically obtained through random mutagenesis, a method allowing high-throughput miniaturized cultivation and screening is of great advantage. Based on a pipetting robot a novel high-throughput screening system suitable for phototrophic microorganisms was developed and then constantly improved. The cultivation was performed in 2,2 ml deepwell microtiter plates within a cultivation chamber outfitted with programmable shaking conditions, variable illumination, variable temperature, and an adjustable CO2 atmosphere. The platform allows semi-continuous cultivation of hundreds of cultures in parallel. Automated measurements of growth, full absorption spectrum, chlorophyll concentration, MALDI-TOF-MS, as well as a novel vitality measurement protocol, have been established. Prior to the mutagenesis, the lethality and rate of non-lethal point mutations of ultraviolet radiation and methyl-methanesulphonate were characterized for Synechocystis. The thus determined optimal dosages were applied to Synechocystis followed by in vivo selection in four rounds of mutagenesis, thereby raising its temperature tolerance by 3°C. The screening platform was used to identify the most thermotolerant monoclonal strains. After validation, their whole genomes were sequenced. Thus mutations in various genes were identified which promote the strains'' thermal tolerance. For some of the genes it is very unlikely that their link to high thermal tolerance could have been identified by other approaches.

Identification of avian pathogenic E. coli (APEC) genes important for the colonization of the chicken lung and characterization of the novel ExPEC adhesin I

Aviäre pathogene E. coli (APEC) sind extraintestinale Pathogene, die beim Huhn systemische Infektionskrankheiten hervorrufen. Zur Identifizierung Gene, die an der Kolonisierung des Wirtes beteiligt sind, wurde ein Lungen-Infektionsmodell in 5 Wochen alten SPF Hühnern etabliert. In dem Modell wurden 1.800 mittels Signature-tagged-Mutagenese (STM) hergestellten Mutanten des APEC Stamms IMT5155 (O2:K1:H5; ST-Komplex 95) auf ihre Fähigkeit zur Kolonisierung getestet. Die Untersuchung führte zur Identifizierung Gene, einschließlich Adhäsin-, LPS- und Kapsel-bildenden Genen, sowie Genen mit putativer Funktion. Die STM-Analyse erlaubte zudem die Identifizierung eines zuvor nicht charakterisierten putativen Fimbrien-bildenden Adhäsins (Yqi). Das Genprodukt wurde vorläufig als ExPEC Adhäsin I (EA/I) bezeichnet. Eine Deletion des EA/I-Gens führte zu einer Reduzierung der Adhäsionsfähigkeit des Stammes IMT5155 in vitro und in vivo. Eine Komplementierung des EA/I-Gens in trans resultierte in einer Wiederherstellung des Adhäsions¬vermögens in vitro. Das EA/I-Protein (~39 kDa) wurde als Fusionsprotein in vitro exprimiert, und mittels SDS-PAGE und Western Blot nachgewiesen. Durch Überexpression des EA/I-Operons in dem Fimbrien-negativen E. coli-Stamm AAEC189 konnten mittels elektronenmikroskopischer Aufnahmen Fimbrien-bildende Strukturen auf der äußeren Membran dargestellt werden. Das Vorkommen des yqi in den untersuchten extraintestinal pathogenen E. coli (ExPEC), bei gleichzeitigem Fehlen in allen untersuchten intestinal pathogenen E. coli bestätigt die Bezeichnung ExPEC Adhäsin I. Die Prävalenz des EA/I-Gens war am stärksten assoziiert mit Stämmen der B2-Phylogenetische-Gruppe und des ST95-Komplexes des Multi-Lokus-Sequenz-Typisierungs (MLST)-Schemas. Sequenzanalysen ergaben zudem erste Hinweise auf eine positive Selektion des EA/I-Gens innerhalb dieses Komplexes. In der vorliegenden Arbeit gelang somit die Identifizierung und Charakterisierung des neuen ExPEC Adhäsin I. / The extraintestinal pathogen, avian pathogenic E. coli (APEC), known to cause systemic infections in chickens, is responsible for large economic losses in the poultry industry. To identify genes, involved adhesion and colonization, a lung colonization model of infection was established in 5-week old specific-pathogen free (SPF) chickens, and Signature-tagged mutagenesis (STM) was applied to this model by generating and screening a total of 1,800 mutants of an APEC strain IMT5155 (O2:K1:H5; ST complex 95). This led to the identification of new genes of interest, including adhesins, genes involved in capsule and LPS formation, and genes of putative function. Among the many genes identified was one coding for a novel APEC fimbrial adhesin (Yqi) not described for its role in APEC pathogenesis. Its gene product was temporarily designated ExPEC Adhesin I (EA/I). Deletion of the ExPEC adhesin I gene resulted in reduced colonization ability by APEC strain IMT5155 both in vitro and in vivo. Complementation of the adhesin gene restored its ability to colonize epithelial cells in vitro. The ExPEC adhesin I protein (~ 39 kDa) was expressed as a fusion protein in vitro as shown by SDS-PAGE and western blotting. Electron microscopy of an afimbriate strain E. coli AAEC189 over-expressed with the putative EA/I gene cluster revealed short fimbrial like appendages protruding out of the bacterial outer membrane. We observed that the adhesin coding gene yqi is prevalent among extraintestinal pathogenic E. coli (ExPEC) isolates and absent in all of the intestinal pathogenic E. coli strains tested, thereby validating the designation of the adhesin as ExPEC Adhesin I. In addition, prevalence of EA/I was most frequently associated with the E. coli phylogenetic group B2 and ST95 complex of the multi locus sequence typing (MLST) scheme, with evidence of a positive selection within this complex. This is the first report of the newly identified and functionally characterized ExPEC adhesin I.

FTIR-spektroskopische Untersuchungen am Phytochrom Agp2

In der vorliegenden Arbeit wurde der lichtinduzierte Reaktionszyklus des bakteriellen Phytochroms Agp2 aus Agrobacterium tumefaciens mit FTIR‑ und UV‑Vis‑Spektroskopie untersucht. Der Photorezeptor besteht aus einem photosensorischen Modul und einer signalgebenden Histidin-Kinase-Domäne. Das photosensorische Modul bindet das Tetrapyrrol Biliverdin als Chromophor. Der Grundzustand von Agp2 (Pfr, 750 nm) ist gegenüber dem lichtaktivierten Zustand (Pr, 700 nm) rotverschoben, weshalb Agp2 den Bathyphytochromen zugeordnet wird. Die Untersuchungen erfolgten unter Verwendung von Isotopenmarkierung, H/D-Austauschexperimenten und ortsspezifischer Mutagenese. Daraus ließen sich folgende molekulare Änderungen charakterisieren, welche im Reaktionszyklus von Agp2 erfolgen: Die lichtinduzierte Isomerisierung des Chromophors führt zu einem Übergang vom Pfr- in den Pr-Zustand, wobei zwei Intermediate, Lumi‑F und Meta‑F, durchlaufen werden. Neben der Konformationsänderung des Chromophor‑D‑Rings ist auch die C‑Ring-Propionsäureseitenkette an der Photoreaktion beteiligt. Die C-Ring-Propionsäureseitenkette ist im Pfr-Zustand protoniert und wird im Übergang von Meta-F zu Pr deprotoniert. Der Pr-Zustand weist eine pH-Abhängigkeit auf, welche auf die pH-abhängige Ladung des Histidins 278 der Chromophortasche zurückzuführen ist. Je nach Ladung des Histidins 278 wird die Keto‑ bzw. Enolform der C(19)=O‑Gruppe des D‑Rings stabilisiert. Die Keto/Enol-Tautomerie ist auf eine innerhalb des Chromophors erfolgende Protontranslokation zurückzuführen und moduliert die Relaxation in den Pfr-Zustand. Änderungen der Amid-I-Absorption im Pfr-Pr-Übergang werden der Umstrukturierung der Tongue-Region des photosensorischen Moduls von einer Alpha-helikalen zu einer Beta‑Faltblatt-Struktur zugeordnet. Diese Strukturänderung wird als möglicher Weg der proteininternen Signaltransduktion zwischen photosensorischem und signalgebendem Modul vorgeschlagen. / In this thesis the light-induced reaction cycle of the bacterial phytochrome Agp2 from Agrobacterium tumefaciens was investigated using FTIR and UV‑vis spectroscopy. The photoreceptor comprises a photosensitive module and a signalling histidine kinase domain. The photosensitive module binds the biliverdin tetrapyrrol as chromophore. The Agp2 ground state (Pfr, 750 nm) is red-shifted in comparison with its light-activated state (Pr, 700 nm). Therefore, Agp2 is assigned to the group of bathy phytochromes. The investigations were conducted using isotopically labelled protein, labelled chromophore as well as hydrogen‑deuterium (H‑D) exchange and site-directed mutagenesis. Based on these the following molecular changes could be characterized that occur in the reaction cycle of Agp2: The light-induced isomerization of the chromophore leads to a transition from the Pfr to the Pr state, involving two intermediates, Lumi-F and Meta-F. Besides conformational changes of the chromophore D-ring, the C-ring propionic side chain is involved in the photoreaction as well. The C-ring propionic side chain is protonated in the Pfr state and gets deprotonated in the Meta-F to Pr transition. The Pr state exhibits pH‑dependent alterations which can be explained by pH dependent polarity changes of histidine 278 in the chromophore pocket. Depending on the charge of histidine, the D‑ring C(19)=O group is stabilized either in keto or enol form. The keto/enol tautomerism involves a proton translocation within the chromophore and modulates the relaxation to the Pfr state. The changes in the amide I region in the Pfr-Pr transition are associated with an alpha‑helix to beta‑sheet secondary structure change of the PHY domain tongue‑region. This structural change is proposed as the potential path of signal transduction between the photosensitive and the signalling module.
