The Production of Organic Nitrates in Portland Oregon and the Columbia River GorgeNeill, Holly Ann 08 April 2013 (has links)
This work studied the production of aerosol-phase organic nitrates in both Portland and the Columbia River Gorge (CRG). Ozone and NOx species were investigated for correlation with organic nitrate aerosol, as they function as precursors to the production of organic nitrates. These ambient gas-phase measurements were collected in the same locations as high-volume (Hi-Vol) filters samples, in an urban and rural gorge setting to investigate correlations at the origin of the pollution plume and downwind. A novel Soxhlet extraction method for Hi-Vol filters was developed based on literature and EPA standard methods. Analysis for nitrate production was done by segregating data based on times when the wind blew out of Portland and down the CRG versus times when flow was not westerly. Filters were then compared to ambient gas-phase measurements and derived NO3 radical production rates to look for trends. Wind direction had a strong influence on the concentrations of precursor molecules in the CRG. On days with a westerly wind direction into the gorge, concentrations of the measure aerosol organic nitrates were similar at both sides. This suggests some contribution of a broader regional production of organic nitrates. There was some correlation between the production rate of NO3 radicals and the measured organic nitrate aerosol, suggesting a role for NO3 + VOC production of organic nitrates that later partition to the aerosol phase. This information will better illuminate the fate of nitrogen downwind of pollution sources. The information will also help to create a better understanding of the way topography and meteorological conditions can influence the flow of pollution. Understanding the downwind oxidative chemistry that happens in the CRG would better support both pollution prevention and mitigation efforts.

Le présent travail a pour but d'évaluer les potentialités électrocatalytiques du cuivre sous différentes formes pour les réactions de réduction du dioxyde de carbone et de réduction de l'ion nitrate.<p><p>Dans la première partie du travail, nous avons comparé, à l’aide de la voltampérométrie, d’électrolyses à potentiel contrôlé et d’analyses chromatographiques, l'activité d'électrodes polycristalline et monocristallines de Cu et d'alliages AuxCuy de différentes orientations cristallines pour l'électroréduction de CO2.<p>Nous avons pu établir une étroite corrélation entre l'activité de l'électrode de Cu et l'arrangement atomique de sa surface. L'activité et la sélectivité pour CH4 de Cu décroissent selon la séquence :Cu (111) > Cu (100) > Cu (poly). La réduction du CO2 et la formation de CH4 sont favorisées sur les surfaces lisses et denses à l'échelle atomique.<p>La sélectivité des alliages AuxCuy est considérablement orientée vers CO quand la fraction superficielle de Au augmente. L'alliage de composition 50-50 conduit à la formation exclusive de CO.<p><p>La seconde partie de la thèse est consacrée à l'étude de l'activité d'électrodes massives de Cu polycristallin et monocristallin d’orientation (111) et de dispersions de Cu pour la réduction électrochimique de NO3-. <p>Nous avons mis en évidence l'importance des conditions de pH pour le déroulement des processus à l'électrode de cuivre. En milieu acide, NO3- est directement réduit en NH4+ alors qu’en milieu neutre, les réactions de réduction de NO3- en NO2- et de NO2- en NH4+ s’observent successivement en fonction du potentiel.<p>L'activité des dispersions de Cu dans un film de polyaniline dépend fortement des conditions de dépôt. Le cuivre incorporé au film sous sa forme réduite est plus actif que le cuivre incorporé au film initialement oxydé. Par ailleurs, nous avons montré que la concentration de H+ dans le polymère est limitée. En conséquence, le processus de réduction de NO3- sur le cuivre dispersé dans un film de polyaniline est modifié en milieu acide.<p>L’utilisation d’un film de poly-3,4-éthylènedioxythiophène déposé sur une surface de Pt s'est quant à elle révélée inadéquate comme support pour l'incorporation de Cu dans le cadre de l'étude de la réduction de NO3- en milieu acide.<p> / Doctorat en Sciences / info:eu-repo/semantics/nonPublished

Ce travail de thèse concerne la préparation et caractérisation des électrodes de carbone vitreuses (GC) modifiées par les films de groupements aryl (substitués) ou par phtalocyanines des métaux (MPc) et l'évaluation de leurs activités catalytiques envers la réduction électrochimique des nitrates dans les milieux aqueux acides. Les techniques adoptées pour la modification de surface du substrat (le GC) consistent à la réduction électrochimique des sels de diazonium ou à l'abrasion physique de la surface contre la poudre des MPc. En faisant intervenir un mécanisme complexe, y compris les phénomènes d'adsorption du réactif et du produit sur la surface, l’électroréduction de sels de diazonium entraîne au greffage des groupements aryls sur la surface. Les analyses voltamétriques détaillées ont rendu possible de conclure, décisivement pour la première fois, que la réduction commence à se faire sur la surface entière et, selon la concentration du diazonium et/ou l'efficacité du greffage, peut continuer à se faire à traverse les ouvertures microscopiques générés parmi les groupements aryl greffés sur la surface durant l'étape précédente de réduction (...) / This thesis work concerns about the preparation, characterization and catalytic activities evaluation of (substituted) aryl and metallophthalocyanines films-modified glassy carbon (GC) electrodes towards nitrate electrochemical reduction in acidic aqueous media. The surface modification techniques adopted consisted of the electroreduction of 4-substituted aryl diazonium salts and the metallophthalocyanine (MPc) powder abrasive adsorption. Through a complex mechanism involving the reactants and products adsorptions on the substrate surface, the electroreduction of aryl diazonium salts leads to the covalent attachment of mono as well as multilayers of aryl groups on the substrate surface. Detailed voltammetric investigations enabled to conclude, decisively for the first time, that the diazonium cation reduction begins to take place on the bulk (whole) surface and, depending upon the concentration and/or the products grafting efficiency, may continue to take place across the microscopic pinholes formed among the aryl groups grafted on the surface during the previous reduction step, thereby explaining the origin of the two reduction peaks in the voltammograms on GC surface. Electrochemical characterization of 4-nitrophenyl(NP)-modified surfaces in various types of aqueous media shed light over a number of mechanistic aspects of the process. Some new electrochemical evidences of the complications of surface coverage determination of redox centers from their electrochemical responses and of the role of aqueous electrolyte species transport on the responses have been presented. Some new phenomena or observations such as identification of the regions of votammograms corresponding to aminophenyl and hydroxyaminophenyl formation of the surface bound NP groups reduction, identification of the method of surface bound NP groups surface coverage estimation from the total width at half wave maximum (or electron transfer coefficient) of the responses, identification of the mechanistic aspects governing the differences of voltammetric behaviors of surface bound NP layers and the solution phase nitrobenzene, electrochemical (and XPS) evidences of the presence of azo type functionalities in the aryl films prepared from aryldiazonium electroreduction were also noticed. Barrier characters of the aryldiazonium electroreduction-derived N,N-dimethylaminophenyl-, nitrophenyl- and aminophenyl-modified surfaces towards ferricyanide, hexaammineruthenium and proton electroreductions in aqueous media indicate to the existence of electrostatic interactions among the surface bound and the solution phase ionic species. However, upon negative potential applications (such as those of water or nitrate reductions) surface attached films are partly or completely lost from the surface, as evidenced by the analytical scale measurements as well as from potentiostatic electrolysis of nitrate reduction in acidic aqueous media. Concerning the MPc-modified GC surfaces, the analytical scale measurements showed that among the phthalocyanines of copper, Iron and Nickel, the one of Cu is an optimizing material for the nitrate electrochemical reduction in, not previously reported, acidic aqueous media. The catalytic activity of MPc powders towards nitrate reduction in these media, which varies in the order CuPc > FePc > NiPc > GC appears to be related to the mental center and not with the phthalocyanine ring

Ce travail porte sur la destruction quantitative et d'une manière respectueuse pour l'environnement de solutions concentrées en nitrates par deux procédés différents. Dans les deux cas, la solution de nitrates est d'abord réduite électrochimiquement en ammoniums sur électrode de cuivre avec une sélectivité élevée, ceci quel que soit le pH de la solution d'électrolyse. Dans le premier procédé, l'ammonium est ensuite oxydé en azote à l'aide d'ions hypochlorites générés électrochimiquement. Une excellente sélectivité réactionnelle en azote de 91,5% est obtenue avec des rendements chimiques et faradiques élevés pour la réaction de réduction des nitrates en azote, accompagnée d'une consommation énergétique basse. Le deuxième procédé est un couplage électrochimique / biologique où les solutions d'ammonium seront utilisées comme substrat azoté pour produire du biohydrogène via des boues traitées thermiquement. Une consommation complète de la solution d'ammonium provenant de la réduction des nitrates est obtenue. Un rendement maximal de 0,35 mole H2/mole de glucose est atteint en utilisant des boues activées collectées d'un bassin d'aération contre 1,1 mole H2/mole de glucose produit dans le cas des boues prélevées d'un digesteur anaérobie. / This work deals with the quantitative and environmentally friendly destruction of concentrated nitrates solutions using two different processes. In both cases, the nitrates solution was firstly reduced electrochemically into ammonium on a porous copper electrode. Whatever the initial pH of the electrolytic solution, a high ammonium selectivity was obtained. In the first process, the ammonium was subsequently oxidized to nitrogen gas by hypochlorite ions generated electrochemically. An excellent selectivity of 91.5% with high current efficiency and high chemical yield toward the nitrogen formation was recorded, with a low power consumption. The second method is an electrochemical / biological coupling process where the obtained ammonium solution will be used as a nitrogen source to produce biohydrogen (H2) via heat-treated sludge cultures. A complete assimilation of the ammonium solution resulting from the electroreduction of nitrate was obtained. A maximum hydrogen yield of 0.35 mol H2/mole glucose was achieved using activated sludge collected from an aeration tank versus 1.1 mole H2/mole glucose produced in the case of sludge taken from an anaerobic digester.

Nitrates in a catchment cleared of alien woody legumes in relation to ground water quality in the Atlantis aquifer (South Africa)Van der Merwe, Nicolette 12 1900 (has links)

neighbouring farm were studied to determine the effects of long term invasion of the
legume Acacia saligna on the soil nutrient content of a soil previously vegetated with
fynbos. The effect of the removal of this alien legume on general soil properties and
groundwater quality were also studied. The changes in nitrates and nitrites (NOX) due to
the invasion and removal of the alien legume were investigated in more detail than
changes in other soil nutrients. In addition to that emphasis was placed on the effect of
vegetation clearing on groundwater quality, specifically relating to potential
contamination with nitrates.
This study was initiated after Conrad et al., (1999) found increased NOX concentrations
in ground water while studying the effects of pig farming on ground water nitrogen (N)
near a site cleared of Acacia saligna by Working for Water (WFW). Since many sites are
scheduled for removal of this alien vegetation it was deemed necessary to study the
effects that clearing alone had on groundwater quality. It was suspected that there would
be an increase in soil and groundwater NOX with vegetation removal due to the inputs
from the legume alien invader.
Soil sampling was done continuously throughout the rainy season of 2007 (From May to
December) on three adjacent sites separated by some 50 m of distance, consisting of a
natural fynbos site and two Acacia saligna sites. The sites were selected approximately
on the same contour line to prevent interaction through lateral water flow. One of the
Acacia sites was cleared by the Working for Water programme in the usual manner
leaving slash on the ground. Soil samples were collected at regular intervals throughout
the season from all three sites (fynbos, Acacia and cleared site) using a Jarrett soil auger.
They were airdried (to achieve full oxidation of mineral N) and sieved though a 2mm
sieve. Soils were analysed by atomic absorption spectroscopy for basic cations and by ion
chromatography for anions, including nitrates and nitrites. Total carbon and nitrogen was
determined by combustion, pH (1M KCl and H2O) and EC (1:5 H2O) were also measured. The present study was part of a wider investigation into the quality of
groundwater, modelling flow and contaminant transport (Jovanovic et al., 2008).
The largest changes in soil properties were observed in the top (0-5 cm) layer. The fynbos
site had a lower nutrient status by comparison to the Acacia site and the cleared site. The
sum of cations from the soils of the fynbos site at a soil depth of 0-5cm was 100mg/kg
and soils from the cleared and Acacia sites were about 190mg/kg. The Acacia site had a
higher NOX status and experienced a larger NOX fluctuation during the rainy season than
the fynbos site; minimum NOX values were similar (below 10mg/kg) but the Acacia site
had a maximum NOX value of near 60mg/kg and the fynbos site just below 30mg/kg.
There was little difference in general soil characteristics (exchangeable cations, pH and
EC, total soil C and N) during the first season after clearing, between the Acacia and the
cleared sites.
The effect of soluble nitrogen changes due to alien legume invasion and removal on
groundwater quality, relating to NOX, during the first season after clearing, was
determined. It was found that the Acacia site had higher NOX concentrations than the
fynbos site. At 0-5cm the fynbos site NOX was less than 30mg/kg and the Acacia site was
between 30 and 110mg/kg for most of the season, with values lower than 30mg/kg for the
last four sampling dates only. N concentrations on the cleared site behaved in a similar
manner to the uncleared Acacia site, but generally N values were lower on the cleared
site, there were only two sampling dates where the cleared site had higher NOX values
than the Acacia site at 0-5cm. The average groundwater N in NOX under the cleared site
was 4.34 mg/l, and under the Acacia site 3.78mg/l, these values are both below the level
determined for water contamination with nitrates. However, the increase in ground water
nitrate levels after A. saligna clearing was significant.
It was concluded that there is a change in the nutrient status of soil with Acacia invasion
and again with removal. NOX migrates to the groundwater to a larger degree once vegetation has been removed, although during this study the nitrate pollution threshold of
10 mg/l nitrate N was not reached. / AFRIKAANSE OPSOMMING: Die sanderige gronde van die Riverlands Natuur Reservaat, naby Malmesbury, en die
aangrensende plaas was bestudeer om die effek van die langtermyn indringing van die
peulgewas, Acacia saligna, op die voedingswaarde van ‘n voorheen fynbos begroeide
grond, sowel as die effek van die verwydering van die indringer op die algemene
samestelling van die grond en grondwater kwaliteit. Oplosbare stikstof veranderings
(NOX) wat plaasvind as gevolg van die indringing en verwydering van die indringer
peulgewas, was in meer diepte bestudeer as die ander elemente. Klem was ook geplaas op
die effek van die verwydering van plantegroei op grondwater kwaliteit, met spesifieke
verwysing na potensiële nitraat besoedeling.
Die studie was beplan na Conrad et al., (1999) ‘n toename van NOX konsentrasies in
grondwater ontdek het, tydens ‘n studie van die effek wat vark boerdery het op

A. saligna area was skoongemaak deur die Working for Water program op die gewone

geneem met gereelde intervalle gedurende die seisoen op al drie areas (fynbos, A. saligna, en skoongemaakte area) deur die gebruik van ’n Jarret grondboor. Die monsters
was lugdroog gemaak (om volle oksidasie van die mineraal N te verseker) en toe gesif
deur ‘n 2mm sif. Die gronde was analiseer deur atoom absorpsie vir basiese katione en
deur ioon chromatografie vir anione, insluitende nitriete en nitrate. Totale koolstof en

inhoud en het ‘n groter NOX fluktuasie ervaar gedurende die reën seisoen as die fynbos
area. Die minimum NOX waardes was soortgelyk (minder as 10/mg/kg), maar die Acacia
area het ‘n maksimum NOX waarde van omtrent 60mg/kg terwyl die fynbos area se
maksimum net minder as 30mg/kg gehad het. Daar nie veel verskil tussen die algemene
grond eienskappe (uitruilbare katione, pH en EC, totale C en N) van die Acacia en
skoongemaakte areas gedurende die eerste seisoen na die indringer verwyder is nie.
Die effek van oplosbare stikstof veranderings as gevolg van die peulgewas se indringing
en verwydering op grondwater kwaliteit, met verwysing na NOX, gedurende die eerste
seisoen na verwydering van die indringer was bepaal. Dit was bevind dat die Acacia area
‘n hoër NOX konsentrasie as die fynbos area het. Op ‘n diepte van 0-5cm was die fynbos
NOX laer as 30mg/kg terwyl die Acacia area tussen 30 en 110mg/kg vir die grootste
gedeelte van die seisoen was met waardes laer as 30mg/kg vir die laaste vier datums
waarop monsters geneem was. Veranderings in N op die skoongemaakte area en die
Acacia area was soortgelyk, maar oor die algemeen was N waardes laer op die
skoongemaakte area. Daar was slegs op twee datums gevalle gevind waar die
skoongemaakte area ‘n hoër NOX inhoud as die Acacia area gehad het by ‘n diepte van
0-5cm. Die gemiddelde N in NOX in die grondwater onder die skoongemaakte area was 4.34mg/l en by die Acacia area was dit 3.78mg/l; hierdie waardes is onder die drempel
vir nitraatbesoedeling in grondwater. Die nitraat inhoud van die grondwater het wel
beduidend toegeneem as gevolg van verwydering van A. saligna.
Die gevolgtrekking was dat daar ‘n verandering plaasvind in die voeding status van grond
met Acacia saligna indringing en weer met verwydering. NOX migrasie na die
grondwater verhoog met verwydering van plantegroei. In hierdie studie was die nitraat
besoedeling drempel van 10mg/l nitraat N nie bereik nie.

In situ denitrification of nitrate rich groundwater in Marydale, Northern CapeIsrael (Clarke), Sumaya 12 1900 (has links)
Thesis (MScAgric (Soil Science))--University of Stellenbosch, 2007. / South Africa is a water scarce country and in certain regions the quantity of surface water is insufficient to provide communities with their domestic water needs. In many arid areas groundwater is often the sole source of water. This total dependence means that groundwater quality is of paramount importance. A high nitrate concentration in groundwater is a common cause of water being declared unfit for use and denitrification has been proposed as a potential remedy.
In groundwater of the Marydale district in the Northern Cape Province, nitrate levels are high enough to be of concern for domestic and livestock consumption. A review of the literature indicates that bacterial denitrification of groundwater can be achieved in situ by using a suitable energy substrate. The technology has been tested elsewhere in the world but more certainty is needed on whether it is a feasible option for local groundwater remediation using local, cost-effective energy substrates and exploiting bacterial populations present naturally in the regolith.
The objective of this study was to perform denitrification experiments by laboratory incubation using soil and groundwater samples collected in Marydale in order to determine; 1) The effectiveness of different carbon sources; 2) The effect of using soil sampled at different depths; 3) The effect of C:N ratio of the carbon substrate; and 4) The quality of resultant water.
Various experiments were set up using 10 g soil and 40 mL groundwater with different concentrations of carbon sources (sawdust, glucose, maize meal and methanol). All experiments were done under a nitrogen atmosphere to exclude oxygen and temperature was kept constant at 23 °C. Indicator parameters were selected based on literature review, and major cations and anions and some metals were analysed for initially and at selected times during each experiment to evaluate whether major ion chemistry was changing over time. Parameters analysed in supernatant solutions after varying periods of time to indicate progress of denitrification and reduction included nitrate, nitrite, sulfate, alkalinity, chloride, acetate, basic cations, ammonium, pH, electrical conductivity, dissolved organic carbon, heteThe Marydale groundwater in some boreholes is of predominantly NaCl type and the nitrate concentration of 19-32 mg/L as N exceeds ideal limits for drinking water of 6mg/L as N . Two soil materials were sampled at different depths from a red sand overlying calcrete (Plooysburg form, Family Py1000).
The incubation experiments showed denitrification was complete within a period of between 1 and 6 weeks depending on the carbon substrate and C:N used. Higher rates of nitrate removal were achieved where greater C:N was used. Readily degradable carbon substrates e.g. glucose showed rapid denitrification, while sawdust, a slowly degradable substrate, effected slower denitrification, hence it was concluded that intermediately degradable carbon substrates e.g. wheat straw may prove more suitable. Use of shallower soil material containing initially higher nitrate levels resulted in better denitrification rates, however, both soil materials effected denitrification.. Heterotrophic plate counts increased with time, this presence and growth of heterotrophic bacteria confirmed that conditions were optimum for growth and denitrification and that inoculation with bacteria is not a requirement for in situ denitrification. Dissolved organic carbon (DOC) concentration could be directly correlated to the initial input of carbon substrate as soil and groundwater lacked organic material. Results showed that reaction products such as acetate and nitrite, and basic cation concentrations were elevated in the supernatant solution in preliminary experiments. This was interpreted to be attributed to incomplete oxidation of organic material and excess soluble and available carbon for reaction. Cation concentrations were interpreted to have resulted from a decrease in pH brought on by organic acids produced during denitrification. The method used showed specificity, as the only parameters affected by the denitrification experiment were DOC, alkalinity, nitrite, nitrate, and the heterotrophic plate count. The DOC and HPC did not comply with acceptable levels for drinking water. Removal of HPC by boiling or chlorinating is required to ensure that the resultant water composition is of potable quality.
For further research with slowly degradable carbon sources it is recommended that a C:N ratio of more than 12 should be employed, and monitoring should focus on soluble carbon nitrate, nitrite, and heterotrophic plate count.
The study confirmed that denitrification of this groundwater with a range of carbon sources is possible within a short period of anaerobic contact with local soil material. With sufficient knowledge of the characteristics of the soil and groundwater in the area, establishment of a working in situ denitrification plant is probably feasible.

2. Nustatyti nėščių moterų ir moterų, kurios augina kūdikius iki 6 mėn. amžiaus, požiūrį į naudojamo vandens kokybę ir tyrimo rodiklius.
3. Įvertinti šachtinių šulinių geriamojo vandens kokybės gerinimo galimybes.
Tyrimo metodika. Anketinė nėščių moterų ir kūdikius iki 6 mėn. amžiaus auginančių moterų (n=167, atsako dažnis 100 proc.) apklausa. Šachtinių šulinių geriamojo vandens kokybės rodiklių tyrimo protokolų analizė (n = 270) ir visuomenės sveikatos centro ir biuro specialistų, rajono ekologo (n=3, atsako dažnis 100 proc.) interviu. Apklausos duomenys analizuoti „SPSS 15.0 for Windows“ programa. Hipotezės tikrintos, taikant χ² testą, nustatčius reikšmingumo lygį p<0,05.
Rezultatai. Tyrimo metu buvo nustatyta, kad respondentės, naudojančios užterštų šachtinių šulinių vandenį, dažniausiai gyvena kaimo vietovėse, turi mažas metines pajamas, yra bedarbės. 95,8 % respondenčių gavo informaciją apie šachtinių šulinių vandens kokybę ir patarimus, kaip apsaugoti save ir kūdikius. Moterys, turinčios žemesnį išsilavinimą, informavus apie vandens tyrimo rezultatus bei riziką sveikatai, vartoja užterštą geriamąjį vandenį nitratais ir nesiima jokių veiksmų jo kokybei gerinti. Objectives :
1. Estimate the quality of the dug wells for drinking water.
2. Evaluate the standpoint of pregnant women and the women growing up babies sunder the age of six (6) months to the dug wells water quality indicators.
3. Estimate the opportunities of improvements for the quality of drinking water.
Research methods. Questions for pregnant women and the women raising infants under six months Cn =167, response rate was 100 percent survey. The analysis of the dug wells for drinking water quality indicators (n=270) made by the public health center, the office professionals the region ecologist (n=3, response rate 100 percent) interview. Survey data, was analysed at “SPSS15.0 for Windows” program. The Hypothesis is verified by using the test χ2. According to it the significance level is p<0,05.
Results. The study found out that the respondents using contaminated water from dug wells, are unemployed. 95.8 % of them have received the right information about the quality of the water in their dug wells. They also have get the advices how to protect themselves and their children. The women with lower education were informed about the test results of their water, about health risks using the water, they still consumpt contaminated by nitrates drinking water and don’t take any actions to improve its quality. Modelování znečištění povrchových vod v CHKO Křivoklátsko s využitím GIS / Modelling of surface water pollution in Krivoklatsko protected area using GISŠereš, Michal January 2014 (has links)
Environmental modelling is lately becoming to be one of the most important decision support systems. Hydrological models present significant potential for use especially in area of surface and groundwater protection. In the diploma thesis, the Soil and Water Assessment Tool (SWAT) was used to evaluate nitrate surface water pollution in nine catchments situated in Křivoklátsko protected area during the period of 2003 - 2010. Modelling process consist of assorted consequent steps. First the hydrological model of each catchment was created. Afterwards the sensitivity analysis was conducted to select most sensitive model parameters. Calibration process was carried out after the parameters selection using SWAT-CUP automatic calibration tool. Model results were visualized and evaluated. The results of the model suggested, that the area of interest was not widely endangered by nitrate pollution. Within each catchment the critical source areas of nitrate pollution were identified. It was observed that these areas are related with intensive agricultural areas. Nitrate leaching ranged from 6 kg.ha-1 to 10 kg.ha-1 per year in agriculture areas. Some areas exceed this amount. Nitrato de amônio e nitrato de potássio no desenvolvimento in vitro de embriões somáticos de pupunheira / Ammonium nitrate and potassium nitrate in the peach palm somatic embryos in vitro developmentSantos, Thaís Lobo dos 18 January 2010 (has links)
O experimento foi conduzido com o objetivo de avaliar a influência da interação entre o nitrato de amônio e nitrato de potássio sobre as respostas morfogênicas de embriões somáticos de pupunheiras in vitro a fim de otimizar o protocolo de micropropagação da espécie. As concentrações utilizadas foram 0, 825, 1650, 2475 e 3300 mg L-1 de nitrato e amônio e 0, 950 , 1900, 2850 e 3800 mg L-1 de nitrato de potássio, combinadas entre si, perfazendo um total de 25 tratamentos. Os tratamentos foram preparados a partir da solução completa de Murashige e Skoog, devidamente modificado para as proporções desses íons no suprimento de nitrogênio. Utilizaram-se duzentos e cinqüenta embriões somáticos com características morfológicas homogêneas, isentos de raízes e folhas, obtidos a partir de microplantas mantidas em sala de crescimento com temperatura e luminosidade controladas. Foram aferidos dados do comprimento da parte aérea e da raiz, o número de raízes, brotações e folhas, a porcentagem de ramificação da raiz, a porcentagem de raízes finas, medianas e grossas, em quatro períodos de cultura, a cada 60 dias, totalizando 240 dias de cultivo. Ao final desse período, avaliou-se também o teor de proteínas totais solúveis, o teor de clorofila pelo índice SPAD e os teores de macro e micronutrientes nas microplantas. Utilizou-se delineamento estatístico inteiramente casualizado e os dados foram submetidos ao teste de Bartlett a 5% e análise de variância (ANOVA) a 1% e 5% de probabilidade de erro ou foi elaborada uma matriz de correlação de Pearson a 1% e 5% de probabilidade de erro, conforme o caso. Os resultados permitiram inferir que aos 240 dias de cultivo as microplantas passam a investir mais em formação de parte aérea e aumentam a porcentagem de raízes finas, e funcionais. Os tratamentos que melhor favoreceram a formação de proteínas foram aqueles com concentração de 2475 mg L-1 de NH4NO3 ou com 2850 mg L-1 de KNO3. Os maiores índices SPAD ocorreram nos tratamentos com até 1650 mg L-1 de NH4NO3 combinado com até 1900 mg L-1 de KNO3. Diferentes combinações dos sais de NH4NO3 e KNO3 podem favorecer a absorção de cada nutriente. Conclui-se que os resultados obtidos no trabalho podem contribuir com a melhoria do protocolo de micropropagação de B. gasipaes, na medida em que permitem estabelecer o melhor tratamento para maximização de uma resposta específica desejada para cada momento do processo de cultivo dos embriões somáticos de pupunheiras, como enraizamento, crescimento da parte aérea, formação de proteínas e formação de brotações, facilitando dessa forma a otimização da produção de microplantas com características desejáveis e, conseqüentemente, sua aclimatização e desenvolvimento ex vitro. / This study aimed to evaluate the influence of the interaction between ammonium nitrate and potassium nitrate on the morphogenetic responses of peach palm somatic embryos in vitro cultivated, to optimize the micropropagation protocol for this species. The concentrations used were 0, 825, 1650, 2475 and 3300 mg L-1 of ammonium nitrate and 0, 950 , 1900, 2850 and 3800 mg L-1 of potassium nitrate, in all possible associations, totalizing 25 treatments. The treatments were prepared with the complete solution of Murashige and Skoog, with modified proportions of these ions in relation to the nitrogen supply. Two hundred and fifty somatic embryos with homogeneous morphological characteristics, without roots and leaves, obtained from microplants from a controlled temperature and luminosity room, were used in the experiment. Every 60 days, during 240 days, the length of shoot and root, the number of roots, propagules and leaves and the root architecture were measured. At the end of 240 days of cultivation, it was also analyzed the total soluble proteins, the foliar chlorophyll by a chlorophyll meter equipment (SPAD-502) and the macronutrients and micronutrients concentrations in the microplants. The experiment was conducted in a randomized design. The data were subjected to Bartlett´s test at 5% and variance analysis (ANOVA) at 1% and 5% of probability of error, or it was made a correlation matrix at 1% and 5% of probability of error, according to each analysis. The results showed that at 240 days of cultivation the microplants spent more energy building the shoot part and raised the percentage of thin, functional root. The best treatments for proteins formation were those with 2475 mg L-1 of NH4NO3 or with 2850 mg L-1 of KNO3. The highest SPAD index occurred in the treatments with at most 1650 mg L-1 of NH4NO3 associated with at most 1900 mg L-1 of KNO3. Different associations of NH4NO3 e KNO3 may favor the absorption of each nutrient. We conclude that the results obtained may contribute to the optimization of the B. gasipaes micropropagation protocol, as it is possible to establish the best treatment for maximization of a specific answer for each moment of the somatic embryos cultivations process, such as rooting, shoot growth, propagules and protein formation, and thus increase the optimization of microplants production with wanted characteristics and hence its acclimatization and ex vitro development.

Ce travail de thèse s’inscrit dans une démarche d’approfondissement des connaissances du fonctionnement des aquifères karstiques. Pour cela, le site pilote des Sources du Toulon, siège d’une alimentation multiréservoirs, a été choisi et offre une fenêtre d’observation privilégiée sur les relations hydrogéologiques entre les aquifères de la marge Nord du Bassin aquitain. Elles sont utilisées depuis 1832 pour l’alimentation en eau potable de la ville de Périgueux, et constituent actuellement son unique ressource.L’objectif de ce travail a été de tester une approche multi-traceurs et un suivi haute-résolution pour caractériser le fonctionnement et la vulnérabilité d’un aquifère karstique multicouche.L’utilisation d’une approche couplée hydrodynamique et hydrochimie a mis en évidence l’alimentation des Sources du Toulon par une ressource profonde et captive en complément d’un aquifère libre plus superficiel. Ces informations ont été intégrées dans le contexte hydrogéologique global de la zone d’étude pour proposer une nouvelle délimitation du bassin d’alimentation des Sources du Toulon.L’utilisation d’un suivi haute-résolution a permis de préciser le fonctionnement hydrogéologique du système karstique étudié. Il en ressort des fonctionnements différents suivants les crues, avec des transferts de masse pouvant s’étendre de quelques jours à quelques semaines.L’apport des isotopes des nitrates (δ15N-NO3- ; δ18O-NO3-) a permis d’identifier deux origines des nitrates mesurés dans les eaux de surface et souterraines : les fertilisants minéraux de synthèse utilisés pour l’agriculture, et les rejets d’assainissement individuels.L’évaluation temporelle de la vulnérabilité spécifique des Sources du Toulon à certains marqueurs de contamination tels que les particules, les éléments bactériologiques et les nitrates a permis de distinguer plusieurs masses d’eau superficielles et subsuperficielles participant à l’alimentation pendant les crues, et jouant un rôle différent dans le transfert des contaminants. / This work is included in an approach for a better knowledge of karst aquifers. For this, the Toulon Springs pilot site was chosen, and provides the opportunity to study relationships between multilayered karst aquifers of the northern edge of the Aquitaine sedimentary basin. Toulon Springs are major regional springs and are located in Périgueux (Dordogne County, France). They have been supplying water to the metropolitan area of Périgueux since 1832.This work aim to test an innovative multitracer approach coupled with a physicochemical high-resolution auto-monitoring to characterize functioning and vulnerability of a multilayered karst aquifer.The use of a coupled hydrodynamic and hydrochemical approach has highlighted Toulon Springs supply by a deep and captive aquifer, in addition to a more subsuperficial aquifer. This information has been incorporated into the global hydrogeological situation of the study area to propose a new delineation of Toulon Springs hydrogeological cathment.The use of a high-resolution monitoring enables to specify the hydrogeological functioning of the studied karst system. The analysis performed on several flood events, identify that mass transfer can range from a few days to a few weeks.The use of nitrate isotope (δ15N-NO3- ; δ18O-NO3-) enables to identify two main nitrate sources in both surface water and groundwater: (i) inorganic fertilizer; (ii) sewage from individual house.The last part of this work was to analyse Toulon Springs temporal vulnerability during floods to marker of contamination such as particles, dissolved organic carbon, nitrate, and bacteria. Hence, several water type from surface runoff, unsaturated zone and saturated zone were identify as responsible of contaminant transfer: (i) water from saturated zone is responsible of vulnerability to mineral particles ; (ii) water from unsaturated zone is responsible of vulnerability to nitrare ; (iii) water from surface runoff is responsible of vulnerability to bacteria contamination.
