For this, a literature review on issues related to that goal was developed, from which relevant publications to the theoretical foundation of the proposed research model have been identified. Based on the literature review and using the content analysis method, which consists of systematic procedures that provide quantitative or qualitative indicators, it was possible to develop a maturity model of project portfolio management aligned to the Portfolio Management methodology of the Information Technology Resources System (MGPP-SISP) under the Ministry of Planning, Budget and Management. The model was evaluated to a focal group of experts to collect the perspectives and experiences of the participants. The present study proposes a project portfolio management maturity model adapted to MGPP-SISP for organizations of the Brazilian Federal Direct Public Administration. / O objetivo principal desta pesquisa ?? propor um modelo de maturidade de gest??o de portf??lio de projetos adaptado ?? Administra????o Direta P??blica Federal Brasileira. Para isso, foi desenvolvida revis??o de literatura sobre temas relacionados a esse objetivo, a partir da qual foram identificadas publica????es relevantes ?? fundamenta????o te??rica do modelo de pesquisa proposto. Com base na revis??o de literatura e utilizando o m??todo de an??lise de conte??do, que ?? composto por procedimentos sistem??ticos que proporcionam o levantamento de indicadores quantitativos ou qualitativos, foi poss??vel elaborar um modelo de maturidade em gest??o de portf??lio de projetos alinhado ?? metodologia de gest??o de portf??lio do Sistema de Administra????o dos Recursos de Tecnologia da Informa????o (MGPP-SISP) subordinado ao Minist??rio do Planejamento, Or??amento e Gest??o. In addition, it allows students to be attended in their specificities to build their identity from the confrontation with the differences and the coexistence with the other. The aim of this work is to carry out an exploratory study on the modalities of specialized educational service provided by private schools in the Federal District to students diagnosed with Attention Hyperactivity Disorder (ADHD) and investigate how this service is being carried out in schools of the private education network. Based on the theoretical framework and literature review, a qualitative research was conducted, with a semi-structured interview, and the data collection was done in seven private schools in Federal District, in the perspective of the inclusion of students with ADHD, whose participants were Counselors and/or Academic Advisors. The work consisted in an investigation of the strategies that are used in each one of these schools in the specialized educational assistance, as well as the factors that hamper the inclusion and learning of those students with ADHD. Also, some issues were discussed such as medicalization of students with this diagnosis, curricular adaptations, family participation, orientation for teachers, continued formation and the contributions that psychoanalysis can bring to professionals who work with these students in order to help them to rethink their practice. By analysing the data, were presented the general characteristics of the sample and the conceptions and practices in the Specialized Educational Assistance in private schools of the Federal District, whose main results point to the need for greater compliance in private schools with the laws that refer to Inclusion, including the implementation of educational strategies that improve the learning of students with ADHD, as well as greater investment and care in the continued formation of the teachers who will work with these students. / O Atendimento Educacional Especializado ?? um servi??o da Educa????o Especial que identifica, elabora e organiza recursos que ajudam os alunos com defici??ncia a superarem as dificuldades que se apresentam ao longo do processo educativo em escolas regulares. Al??m disso, possibilita que o aluno seja atendido em suas especificidades para que possa construir a sua identidade a partir dos confrontos com as diferen??as e da conviv??ncia com o outro. O objetivo deste trabalho ?? realizar um estudo explorat??rio sobre as modalidades de atendimento educacional especializado prestado por escolas particulares do Distrito Federal aos alunos com diagn??stico de Transtorno de D??ficit de Aten????o e Hiperatividade - TDA/H e investigar como este atendimento est?? sendo realizado em escolas da rede particular de ensino. In this way, when we speak in search of knowledge, we are reminded of the various ways of studying, and thinking about the current context with the evolution of technologies, where schools are increasingly adhering to studies aligned with the use of Information and Communication Technologies (ICT). ICTs are used as tools for learning aid. With this intention indeed it is believed that aligning the bases of traditional classes with distance classes, making use of a platform for extra moments in the classroom, enables students to acquire information relevant to a certain content. The MOODLE platform is a Free Software tool and is adopted in most of the Education Institutions to promote Distance Education (EaD). Knowing these principles is what you do if you base this study, in addition to take the student out of that old routine of a classroom with the teacher writing on the board or projecting information from your computer. First of all, we will make an initial milestone where we will count on a test called Pre-test, which will be the basis for us to know what learners bring with them. Apprentices will be introduced to the platform where they will have one more tool to search for information. Bringing the students into the MOODLE platform so they can acquire knowledge and thus arouse their curiosity to seek self-learning. The final mark will be with the application of the Post test, where we will analyze within all The collaborative process, if indeed the student has evolved. The help of the teacher-tutor is extremelly important at this moment because it is this figure that will begin to instigate the students to self-discover. This study aims to analyze the learning behavior and evolution of the apprentices of the Instituto Federal do Amap? (IFAP), Campus Laranjal do Jari, in the discipline subject of Computer Networks, using as a method the classroom classes in the classroom concomitantly with the distance classes. / Este estudo caracteriza-se como uma pesquisa qualitativa onde buscamos analisar com profundidade o conhecimento dos aprendizes, tamb?m de car?ter descritivo onde descrevemos os passos de como ocorreram os processos colaborativos dentro da plataforma. Dessa forma, quando falamos em busca de conhecimento, nos vem em mente as v?rias formas de estudar, e considerando o contexto atual das evolu??o das tecnologias, percebemos que as escolas est?o cada vez mais aderindo os estudos alinhados com o uso das Tecnologias da Informa??o e Comunica??o (TIC). As TICs s?o utilizadas como ferramentas para aux?lio na aprendizagem. Nesse sentido, acredita-se que ? indispens?vel alinhar as bases das aulas tradicionais ?s aulas ? distancia, utilizando uma plataforma para que fora sala de aula os alunos possam adquirir informa??es pertinentes a um determinado conte?do. Al?m de apresentar-se como proposta inovadora nas pr?ticas educativas, pois tira o aluno da velha rotina da sala de aula com o professor escrevendo no quadro ou projetando as informa??es de seu computador. Para tanto, o marco inicial da pesquisa foi a aplica??o de um Pr?-teste, que serviu de base para saber o que os aprendizes traziam de informa??o. Em seguida esses alunos utilizaram a plataforma MOODLE e as ferramentas dispostas para a realiza??o de atividades, a fim de que pudessem desenvolv?-las de forma aut?noma em busca do forma??o t?cnica e intectual. O marco final foi a aplica??o do P?s-teste, onde analisou-se, dentro do o processo colaborativo, a progress?o do conhecimento do aluno, quanto ? apropria??o de conceitos referentes a ?rea de estudo. O aux?lio do professor-tutor foi fator relevante na intera??o do aluno com a plataforma. Portanto, a pesquisa atendeu aos objetivos propostos de analisar o comportamento e a evolu??o do aprendizado dos alunos do Instituto Federal do Amap? This research seeks to analyze and consider the mechanisms that ensure accessibility and inclusion of people with disabilities in vocational education based on existing laws and also measure the permanence of conditions that student with disabilities in Technical Education, Technology, undergraduate and graduate students in IFAM campus Mau?s. This process was seen as a major challenge, as many campi the IFAM not have the Call Center for People with Special Needs (NAPNE). However, the IFAM campus Mau?s, site of this research, the NAPNE was officially implemented in May 2015, which for this study was considered a major breakthrough in training, awareness and support for school teachers for special education in the perspective of education inclusive. Specifically, this study sought to identify the deaf students at IFAM CMA; describe the architectural accessibility and characterize the didactic and pedagogical accessibility in IFAM CMA. The methodology was qualitative type Case Study. The data collection instruments were: participant observation, record in a diary, questionnaire and interviews. The data were analyzed based on content analysis of Laurence Bardin (2011). The following categories were raised that will make the results of chapters: Accessibility - incentives and barriers to education for deaf students at IFAM CMA; The perception of teachers about the challenges of raising a deaf student at IFAM CMA; The perception of deaf students about accessibility in IFAM CMA and the influence of this on their learning process. This research finds that were found dichotomies of from architectural accessibility, passing by positioning, attitudinal, professional education, support for this work and ensuring all teaching resources, and communication accessibility to the deaf student to be guaranteed their stay the federal technical education. / Esta disserta??o apresenta o resultado da investiga??o sobre a acessibilidade de estudantes surdos no Instituto Federal do Amazonas (IFAM) campus Mau?s, o qual recebe educandos surdos nos cursos profissionalizante desde a sua inaugura??o em 2010. Esta pesquisa buscou analisar e considerar os mecanismos que garantem a acessibilidade e a inclus?o das pessoas com defici?ncia auditiva na educa??o profissionalizante com base nas legisla??es vigentes e tamb?m mensurar as condi??es de perman?ncia desse estudante na Educa??o T?cnica, Tecnol?gica, gradua??o e p?s-gradua??o no IFAM campus Mau?s. Este processo foi entendido como um grande desafio, pois muitos campi do IFAM n?o possuem o N?cleo de Atendimento ?s Pessoas com Necessidades Especiais (NAPNE). Entretanto, no IFAM campus Mau?s, locus desta pesquisa, o NAPNE foi oficialmente implantado em maio de 2015, o que consolida a import?ncia desse estudo para a forma??o, a sensibiliza??o e o apoio aos professores regentes da educa??o especial na perspectiva da educa??o inclusiva, uma vez que se buscou identificar os alunos surdos no IFAM CMA e descrever a acessibilidade destes neste contexto. A metodologia utilizada foi a Pesquisa Qualitativa do tipo Estudo de Caso e an?lise de conte?do na perspectiva de Bardin (2011). Os instrumentos de coleta de dados foram: observa??o participante, registro em caderno de campo, question?rio e entrevistas, que foram gravadas em ?udio e v?deo. Como resultado das an?lises deste estudo foram levantadas as categorias: Acessibilidade ? est?mulos e barreiras ? educa??o de estudantes surdos no IFAM CMA; A percep??o dos professores sobre os desafios de educar um estudante surdo no IFAM CMA; A percep??o dos estudantes surdos sobre a acessibilidade no IFAM CMA e a influ?ncia desta no seu processo de aprendizagem. Esta pesquisa constata que foram as contradi??es que dizem respeito ? acessibilidade arquitet?nica, perpassando pelo posicionamento atitudinal do profissional da educa??o, ao apoio a este profissional e ? garantia de todos os recursos pedag?gicos e de acessibilidade comunicacional ao estudante surdo, para que seja assegurada sua perman?ncia na educa??o t?cnica federal.

The aim of this dissertation lies in establishing an o-minimal de Rham cohomology theory for smooth abstract-definable manifolds in an o-minimal expansion of the real field which admits smooth cell decomposition and defines the exponential function, by following the classical de Rham cohomology. We can specify the o-minimal cohomology groups and attain some properties as the existence of Mayer-Vietoris sequence and the invariance under smooth abstract-definable diffeomorphisms. However, in order to obtain the invariance of our o-minimal cohomology under abstract-definable homotopy we must, working in a tame context that defines sufficiently many primitives, assume the validity of a statement related to Bröcker\'s problem. / O objetivo desta tese reside em estabelecer uma cohomologia de De Rham o-minimal para variedades definíveis abstratas lisas em uma expansão o-minimal do corpo ordenado dos reais, a qual admite decomposição celular lisa e define a função exponencial, seguindo a cohomologia de De Rham clássica. Além de especificarmos os grupos da cohomologia de Rham o-minimal, obtemos algumas propriedades, como a existência da sequência de Mayer-Vietoris e a invariância sob difeomorfismos definíveis abstratos lisos. Todavia, a fim de lograrmos a invariância de nossa cohomologia o-minimal sob homotopia definível abstrata devemos, além de trabalhar num contexto moderado no qual muitas primitivas são definidas, assumir a validade de uma asserção relacionada ao problema de Bröcker.

Investigation of the Kinetics of Tet(O)-mediated Tetracycline Resistance

Widespread tetracycline resistance (TcR) has limited the clinical use of Tc for the treatment of bacterial infections. Tet(O) protein is present in many bacteria and is the major transmissible TcR determinant in Campylobacter jejuni, a common cause of acute bacterial diarrhea worldwide. Tet(O) protects ribosomes against the inhibition of protein synthesis by Tc. Tet(O) binds to the ribosome at a similar site as EF-G, a structural homologue of Tet(O) with GTPase activity that is required for protein elongation. EF-G interfered with the kinetics of Tet(O)-mediated Tc release suggesting that EF-G competes with Tet(O) for ribosome binding. Indirect assessment of EF-G and Tet(O) binding to 70S ribosomes by GTP hydrolysis was unable to clearly demonstrate competition for binding. This thesis contributed to the further understanding of the kinetics of Tc release by Tet(O), and may facilitate the development of novel strategies to overcome Tet(O)-mediated TcR in bacteria which cause human infections.
