Designing bioinspired materials with tunable structures and properties from natural and synthetic polymers

Varadarajan, Anandavalli 08 August 2023 (has links) (PDF)
Biological systems are composed of complex materials which are responsible for performing various functions, such as providing structural support, mobility, functional adaptation to the environment, damage repair, and self-healing. These complex materials display excellent mechanical properties and can rapidly adapt to external stimuli. Thus, nature inspires in terms of source materials, functions, and designs to develop new-generation structural and functional materials. Polymers (natural or synthetic) are excellent sources of developing materials to mimic the functions of soft segments in biological systems. This dissertation focuses on synthesizing and characterizing two different materials with tunable structures and properties: complexes from natural polysaccharides or polyelectrolytes and bioinspired hydrogels from synthetic polymers. Oppositely charged polyelectrolytes can form polyelectrolyte complexes (PECs) due to the electrostatic interactions. The structure and properties of PECs can be tuned by varying the salt concentration, as the addition of salt can facilitate associative phase separation. PECs were prepared from two biopolymers, positively charged chitosan and negatively charged alginate. Rheological experiments for the complexes displayed a tunable shear modulus with changing salt concentrations. The microstructural study conducted using small-angle X-ray scattering provided insights regarding the length scales of these complexes, and the results follow the observed rheological and phase behavior. Elastic biopolymers such as resilin display remarkable mechanical properties, including high stretchability and resilience, which many species exploit in nature for mechanical energy storage to facilitate their movement. Such properties of resilin have been attributed to the balanced combination of hydrophilic and hydrophobic segments present in the chain. In this work, we synthesized hydrogels with hydrophilic and hydrophobic components to mimic the properties of resilin. With this system, we determined the tensile, retraction (ability to revert to the original state after stretching), and swelling properties when (i) the concentration of the hydrophobic polymer was varied and (ii) additional hydrophobic components were included. The stretchability, stiffness, and strength of the gels varied as the compositions were altered. The fundamental understanding of the structure-property-function relationship for materials presented in this work provides insights into engineering materials for applications such as tissue engineering, drug delivery, wound healing, artificial muscles, soft robotics, and power amplification.

Development, Characterization, and Fundamental Studies on Molecular Ionic Composites and PBDT Hydrogels

Zanelotti, Curt Joseph 28 January 2022 (has links)
This dissertation aims to develop, characterize, and fundamentally understand a new class of materials termed "molecular ionic composites" (MICs). MICs show promise as next-generation solid electrolytes for batteries. MICs form when mixing a rigid polyanion with purely ionic fluids, and they behave mechanically as a solid but contain a high density of ions that move nearly as in a neat liquid. Specifically, prototypical MICs are based on solutions of the rigid-rod polyelectrolyte poly(2,2'-disulfonyl-4,4'-benzideneterephthalamide) (PBDT), which forms a double helix, combined with imidazolium-based ionic liquids (ILs). The IL comprises 75-97 wt% of the final solid, even though the Young's modulus can reach ~ 2 GPa at 80 wt% IL. We propose that these properties are driven by a biphasic internal structure in MICs corresponding to IL-rich "puddles" (an interconnected liquid phase) and PBDT-IL associated "bundles" where IL ions form the collective electrostatic associations that cause the MICs to be a solid. Through this dissertation I will discuss a wide variety of MICs that have been created through the use of two different formation processes, the "ingot" method and the "solvent casting" method, which allow for the use of many different ionic fluid sources to further tune MIC properties. The following chapters build to the fundamental knowledge and our current understanding of the wide variety of materials that can be created from PBDT and IL. / Doctor of Philosophy / Battery electrolytes, biosensors, and hydrogels all depend on new materials for next-generation applications. For these new materials to be used characterization on the interactions, morphological restrictions, and/or what unique internal structures used to generate their properties must be performed. Through This analysis using common polymeric characterization techniques these materials can be further optimized. This dissertation highlights a new class of materials termed "molecular ionic composites" (MICs) which are formed from a rigid double helical polymer, poly(2,2'-disulfonyl-4,4'-benzideneterephthalamide) (PBDT), and fluids composed entirely of ions, including ionic liquids (ILs). These composite systems feature a unique combination of properties including high thermal stability, mechanical stability, and excellent ionic conductivity, all of which are highly tunable through the amount of PBDT incorporated or the fluid ion types. Chapters 3, 4, 5, and 6 present fundamental investigations of MICs to determine how tunable they are, the processes by which they form, and the various ways we can fabricate them. Chapter 7 describes the creation of another impressive material formed from PBDT-low-polymer-content hydrogels. These studies are intended to provide deeper understanding of the behaviors of these unique materials and how they may be used in the future.

Development and Characterization of Advanced Polymer Electrolyte for Energy Storage and Conversion Devices

Wang, Ying 09 January 2017 (has links)
Among the myraid energy storage technologies, polymer electrolytes have been widely employed in diverse applications such as fuel cell membranes, battery separators, mechanical actuators, reverse-osmosis membranes and solar cells. The polymer electrolytes used for these applications usually require a combination of properties, including anisotropic orientation, tunable modulus, high ionic conductivity, light weight, high thermal stability and low cost. These critical properties have motivated researchers to find next-generation polymer electrolytes, for example ion gels. This dissertation aims to develop and characterize a new class of ion gel electrolytes based on ionic liquids and a rigid-rod polyelectrolyte. The rigid-rod polyelectrolyte poly (2,2'-disulfonyl-4,4'-benzidine terephthalamide) (PBDT) is a water-miscible system and forms a liquid crystal phase above a critical concentration. The diverse properties and broad applications of this rigid-rod polyelectrolyte may originate from the double helical conformation of PBDT molecular chains. We primarily develop an ionic liquid-based polymer gel electrolyte that possesses the following exceptional combination of properties: transport anisotropy up to 3.5×, high ionic conductivity (up to 8 mS cm⁻¹), widely tunable modulus (0.03 – 3 GPa) and high thermal stability (up to 300°C). This unique platform that combines ionic liquid and polyelectrolyte is essential to develop more advanced materials for broader applications. After we obtain the ion gels, we then mainly focus on modifying and then applying them in Li-metal batteries. As a next generation of Li batteries, the Li-metal battery offers higher energy capacity compared to the current Li-ion battery, thus satisfying our requirements in developing longer-lasting batteries for portable devices and even electric vehicles. However, Li dendrite growth on the Li metal anode has limited the pratical application of Li-metal batteries. This unexpected Li dendrite growth can be suppressed by developing polymer separators with high modulus (~ Gpa), while maintaining enough ionic conductivity (~ 1 mS/cm). Here, we describe an advanced solid-state electrolyte based on a sulfonated aramid rigid-rod polymer, an ionic liquid (IL), and a lithium salt, showing promise to make a breakthrough. This unique fabrication platform can be a milestone in discovering next-generation electrolyte materials. / Ph. D.

Déchloration réductive par les nanoparticules de fer zéro-valent : une solution innovante pour la réhabilitation des aquifères souterrains contaminés par le trichloroéthylène / Reductive dechlorination by nanoscale zero-valent iron particles : an innovative solution for the remediation of groundwaters contaminated with trichlorethylene

Kaifas, Delphine 27 March 2014 (has links)
Recent advances in nanotechnology have led to the development of new materials with unique physicochemical properties such as nanoscale zero valent iron particles (nZVI). These nanoparticles proved their efficiency to degrade chlorinated organic compounds such as trichlorethylene (TCE), but their migration in porous media is often limited. To overcome this problem, a solution is to modify their surface by adsorption of organic molecules. However, this coating modifies the reactivity towards TCE, which can potentially affect the treatment efficiency. Thus, the first part of this PhD focuses on the reactivity of nZVI (bare or modified by anionic polyelectrolytes) towards TCE. This reactivity was evaluated through the TCE degradation kinetics rates and its transformation products. In addition, groundwaters often contain reducible species that can react with dissolved Fe0. These last species may affect the reactivity of nZVI towards the target pollutant and therefore the remediation efficiency. The second part of this PhD focuses on the effect of two electron acceptors (CrVI and NO3-) on the reactivity of bare and modified nZVI.Finally, the third part of this PhD presents the assessment of the reactivity of nZVI towards TCE in a "real" case, in order to validate the remediation process. A pilot study and in situ application have been carried out on an industrial site which groundwater is contaminated with TCE (targeted pollutant) with high levels of CrVI and NO3-.

Emprego da flotação lamelar de alta taxa e convencional como pós-tratamento do efluente de sistema constituído de reator anaeróbio seguido de reator aeróbio tratando esgoto sanitário / Lamella design (high rate) and conventional flotation units applied to the post-treatment of the effluent from a system made up anaerobic reactor followed by aerobic reactor treating domestic sewage

Moretti, Renata Cristina 20 May 2005 (has links)
The aim of this research was the study of the lamella design (high rate) and conventional dissolved air flotation (DAF) process potentiality of application as the final part of a domestic sewage sequential treatment system made up anaerobic reactors (UASB) followed by aeration tank. The work, developed at the Jardim das Flores WWTP of Rio Claro city, was separated in three phases. In the first phase, using the flotatest apparatus, the most suitable values of chemical (synthetic cationic polymer, arrow-root starch, ferric chloride and ferric chloride combined with polymer or arrow-root starch) dosages, air to solids ratio (A/S), as well as rapid mix and flocculation time and mean velocity gradient values, were investigated. In this phase, the best DAF performances were obtained with the application of cationic polymer, alone and associated to ferric chloride. The use of ferric chloride was essential to obtain better phosphorus removal results. The mixed liquor flotation using ferric chloride alone as primary coagulant presented extremely unsatisfactory performance, with the formation of flocks showing bad characteristics of flotability. Moreover, it was observed that flotation of this kind of effluent had requested gentler mixture conditions. In many cases, the best results were obtained by means of rapid mix or flocculation step elimination. In the second and third phases, using the lamella design and conventional flotation units respectively and cationic polymer as the only flocculation/flotation aid (dosage around 1 g of polymer/kg TSS), the most suitable values of Overflow Rate (OR), Solids Application Rate (SAR) and amount of air supplied to the flotation process, expressed as the A/S ratio or air dosage (AD, in g of air/'M POT.3' of influent) depending on influent total suspended solids concentration (TSSinfl), were investigated. The flotation essays results showed that the A/S ratio is more suitable for the air supply estimation in flotation of concentrated suspensions (TSSinfl above 1100 mg/L in this study). Contrarily, the parameter AD is more representative in cases of flotation of diluted suspensions. Using the lamella design unit, fed with aeration tank mixed liquor presenting good bio-flocculation conditions, it was possible to obtain adequate results applying OR as high as 350 'M POT.3'/('M POT.2'.day) combined with SAR values between 120 and 250 kg of TSS/('M POT.2'.day), since the adequate amount of air has been provided for the process (AD values between 15,0 and 18,0 g of air/'M POT.3' of influent), for TSSinfl between 800 and 1100 mg/L. For the lamella flotation, it was noticed that the best performance has been directly associated to lower values of TSSinfl. The recommendations for flotation using the conventional DAF unit had to be more conservative, taking into account the poor bio-flocculation conditions presented by the mixed liquor during the third phase of work. In theses essays, the more adequate results were obtained by applying OR around 200 'M POT.3'/('M POT.2'.day), SAR under 200 kg of TSS/('M POT.2'.day) and A/S ratio around 0,020 (AD of 21,0 g of air/'M POT.3' of influent), for TSSinfl around 1400 mg/L

Arranjos supramoleculares de drogas em lípides sintéticos e/ ou polieletrólitos: estabidade coloidal e atividade in vitro / Supramolecular assemblies of drugs in synthetic lipid and/ or polyelectrolytes: colloid stability and in vitro activity

Vieira, Débora Braga 15 April 2008 (has links)
The formation, colloid stability and activity in vitro against Candida albicans of several supramolecular assemblies composed of drug, cationic lipid and/or polyelectrolytes were systematically evaluated by means of dynamic light scattering for particle sizing, zeta- potential analysis, UV-visible spectroscopy, effect of drug on gel-to-liquid-crystalline phase transition of the cationic bilayer and determination of drug loading. Supramolecular assemblies of antifungal drugs such as miconazole and amphotericin B were obtained by solubilization of the drugs in cationic bilayer fragments composed of dioctadecyldimethylammonium bromide or coverage of drug particles with a layer of the quoted cationic lipid. As a model of anticancer drug, cisplatin was sandwiched between two oppositely charged polyelectrolytes: chitosan and carboxymethylcellulose. Cisplatin induced reduction in particle size acting as a cross-linker between polyelectrolytes. For amphotericin B, at low and high molar proportion drug to cationic lipid, similar supramolecular assemblies were coated by polyelectrolytes such as carboxymethylcellulose, poly(diallyldimethylammonium) and polylysine yielding cationic nanoparticles that presented optimal colloid stability and fungicidal activity. Miconazole became attached at the hydrophobic edges of bilayer fragments at room temperature and/or, upon an increase in temperature, inserted in the bilayer core. Curiously, this last formulation in the cationic lipid yielded a synergistic action against Candida albicans. Dioctadecyldimethylammonium bromide (DODAB) is an excellent antimicrobial agents per se. Its mechanism of antimicrobial action was compared to the one for hexadecyltrimethylammonium bromide (CTAB). Adsorption of these compounds on the cells decreased going from CTAB to DODAB bilayer fragments and to large vesicles. Absence of leakage of small phosphorylated compounds, proteins or DNA from fungus indicated a mechanism of action different from cell lysis. Adsorption of the cationic compounds changed the sign of the cell zeta-potential from negative to positive. There was a clear relationship between positive charge on fungus and death.

Comparaison de la capacité de différenciation en cellules endothéliales, de deux types de cellules souches mésenchymateuses issues de la gelée de Wharton et de la moelle osseuse / Comparison of the endotheliale differentiation of mesenchymal stem cells isolated from the Wharton's jelly and the bone marrow

Rammal, Hassan 14 February 2014 (has links)
The incidence of cardiovascular disease remains a major public health problem. Despite the development of endovascular therapies, surgical treatment is necessary for many patients. The low availability of natural vessels, autologous or not, and the mechanical and biological limits of artificial substitutes, led to the use of a new domain: vascular engineering. In recent years, the concept has emerged and evolved. He could afford to offer new types of synthetic vascular substitutes and / or biological, in particular through the use of stem cells, offering interesting perspectives in the field of vascular engineering. The purpose of this study was to obtain a reliable and reproducible protocol to generate functional endothelial cells (ECs) from mesenchymal stem cells (MSCs) derived from the umbilical cord Wharton's jelly (WJ) and bone marrow (BM). Nevertheless, their differentiation into vascular cells needs a culture surface functionalization; our group demonstrated the potential use of polyelectrolyte multilayer made of poly(allylamine hydrochloride): PAH, and poly(styrene sulfonate): PSS, in promoting cells adhesion, proliferation and differentiation. Cells were cultured on (PAH-PSS)4 films or collagen type I (used as control), in the presence of angiogenic growth factors. Cells differentiation into EC was followed through the expression of endothelial markers (PCR and western blot); cell functionality was checked through their ability to incorporate acetylated LDL (Low Density Lipoprotein), to produce NO (Nitric oxide) and to express the von Willebrand factor (vWF). After 14 days of stimulation, only WJ-MSCs were able to generate functional ECs demonstrating the potential of combining WJ-MSCs and (PAH-PSS)4 films in vascular tissue engineering field

Study of the separation by organic solvent nanofiltration of diluted solutes using commercial, dense and porous membranes and their derivatives by deposition of polyelectrolyte nanolayers / Fractionnement par nanofiltration organique de mélanges liquides modèles de milieux de métathèse.

Morshed, Mahbub 16 July 2019 (has links)
The objective of this study was to improve the OSN separation performance of commercial membranes for metathesis applications in which highly diluted catalysts are used. In this work, commercial polymeric membranes were first studied to characterize their performance in organic media using very dilute solute-solvent binary mixtures. Based on a literature review, it was shown that the PERVAP4060 membrane, of which PDMS is the dense active layer, was a promising candidate for organic solvent nanofiltration (OSN). As a porous membrane, the AMS and PAN commercial supports have also been taken into account. In this study, we considered the modification on the surface to improve the separation properties of polymeric OSN membranes. Ar/O2 plasma and/or polyelectrolytes multilayers were used for the preparation of new prototype membranes. Unmodified and modified membranes were tested under OSN conditions using binary feed mixtures. Several highly dilute solutes, organophosphorus ligand R-BINAP, phase transfer catalyst ToABR, and linear alkanes have been studied. Both R-BINAP and ToABR were used in the range of 0.0001 to 0.5% by weight, and most experiments were subsequently performed with 0.05% solute concentrations in toluene. It has been shown that PDMS can retain 80% R-BINAP and about 93% ToABr in toluene. After modification by the LBL deposition, the rejection is improved with the modified PERVAP4060 membranes, leading to an 88% rejection of R-BINAP with a deposit of 10 PAH / PSS polyelectrolyte bilayers at the surface and this rejection being able to reach 95% when the number of bilayers is 20. ToABr rejection increases to 97% with the ten bilayered membranes. The performance of the membrane was studied under different pressures of between 1 and 40 bar; the high rejection, still observed in these OSN conditions, strongly supports a solution-diffusion transfer mechanism through the PDMS. The treatment of ternary mixtures mimicking the catalyst/solute/ solvent mixture corresponding to the hydroformylation has also been studied; no evidence of coupling was detected, and the highest retention remained unchanged. On the other hand, the improvement of the rejection also observed from the porous membranes after modification. The rejection of C44 in the AMS was reached 75% after modification by tention10 bilayers of PDDA / PSS, whereas it was only 25% before modification. In the modified PAN, the rejection of the solutes obtained is in the range of 37 to 50%, whereas it was only 3 to 7% by weight before modification. The disadvantage of the porous membrane, however, is the sharp decrease in flux after the deposition of the multiple layers.

Complexes de polyélectrolytes : Morphologies, cinétique et thermodynamique d'assemblage / Polyelectrolytes complexes : Morphologies, kinetics and thermodynamics of the assembly

Haddou, Marie 15 March 2019 (has links)
Complexes of polyelectrolytes (PECs) are oppositely charged polymers assemblies held together by electrostatic interaction. As the non-covalent assembly of macromolecules/colloids in solution has been thoroughly investigated over the last decade with a particular emphasis put on the morphology of the final aggregates as a function of the building blocks-chemistry, the objective of this thesis is to come to a better understanding of the influence of interaction strength between polyelectrolytes on complexes morphologies, as well as kinetics and thermodynamics of the complexation. For this study, weakly and strongly interacting systems given rise to coacervate and precipitate phases has been elected. Those systems have been studied on a wide charge ratio scale (Z[+]/[-]) to span several physical states of the PECs (soluble and colloidal PECs, dense phases). Morphologies of assemblies are assessed by microscopy (optical, fluorescence, atomic force) and scattering techniques (neutron, light). Kinetics and thermodynamics of the complexation process are studied by use of a stopped-flow mixing device and an isothermal titration calorimeter. Results gave us keys to discriminate strongly interacting systems from weakly interacting ones. In particular, the ‘strong systems’ are showed to be non-equilibrium assemblies, ie. the formulation pathway or the way these different macromolecules are coming into contact is then a key step. Another important aspect that could explain the complexation process behavior is the hydration energy of the polyelectrolytes. Calorimetry measurements have been performed for numerous polyelectrolytes and then correlated to complexes properties.
