Utvärdering av antibakteriell aktivitet hos växtextrakt utvunna från svenska örter

Infections caused by antibiotic resistant pathogenic bacteria is an increasing issue inhealthcare, and development of new antimicrobial substances could contribute to combat the continued spread. Plants have historically been used in traditional medicine, have intrinsic defense mechanisms against microbes, and could therefore be a source for new antimicrobial agents. At the Department of Chemistry and Biomedicine at Linnaeus University, Kalmar, a total of 18 extracts made from 9 different combinations of Swedish-growing plants were available.The purpose of the current thesis was to investigate possible antimicrobial effects of the plant extracts in vitro, on a selection of Gram-positive (n=3) and Gram-negative (n=4) bacterial strains. Initial screening of all 18 extracts was performed with agar-based methods including agar well diffusion and direct application on inoculated agar. Detection of concentration-dependent antimicrobial effects was performed with four extracts on Staphylococcus aureus and Enterococcus faecalis. At inhibitory concentrations, viability was estimated as colony forming units/ml (CFU/ml).Screening showed that 11 of 18 extracts affected the growth of at least one of the strains tested. Gram-positive species were affected to a greater extent than Gram-negatives. Estimation of concentration dependency showed inhibitory effects at 50 mg/l in the most potent extracts. Viability estimation revealed an average reduction for both S. aureus and E. faecalis, as compared to the positive control. In conclusion, the study showed possible antimicrobial effects of several extract-bacteria combinations, disclosing potential substances for further investigations.

Qualidade do mexilhão Perna perna submetido ao processo combinado de cocção, congelamento e armazenamento / The quality of the mussel (Perna perna) processing by cooking, freezing and storage

Os mexilhões cultivados no litoral Norte de São Paulo, município de Ubatuba, são comercializados in natura, constituindo risco à população. Com o crescimento da atividade é possível sugerir a implantação de uma unidade de processamento de mexilhões que promova um aumento do tempo de armazenamento, facilitando a comercialização e permitindo a exportação, além de fornecer ao consumidor um produto de melhor qualidade. Os mexilhões foram submetidos ao processamento por cocção, congelamento e armazenamento, sendo então determinados o ponto de congelamento, a velocidade de congelamento a as curvas de congelamento do mexilhão semidesconchado. A qualidade microbiológica e físico-química do produto foi avaliada. O beneficiamento do mexilhão iniciou-se com a cocção por imersão em água à ebulição por 10 minutos. Após a retirada das conchas, os mexilhões foram congelados individualmente IQF (Individually Quick Frozen) a -20ºC e armazenados a -18ºC durante 90 dias. A curva de congelamento do mexilhão apresentou forma geral típica, com o ponto de congelamento situando-se na faixa de zero a -1,5ºC; a velocidade de congelamento variou de 2 cm/h a 3,3 cm/h, conforme a disposição dentro da câmara de congelamento. Os resultados físico-químicos mostraram que não houve diferença significativa no valor nutricional dos mexilhões in natura, processados e armazenados, apresentando os teores médios de 7,4mg/100g de proteínas, 5,8 mg/100g de carboidratos e 1,4 mg/100g de lipídeos. Os valores encontrados para BNVT, TMA e pH no mexilhão in natura foram, 4,3 mg/100g; 2,0 mg/100g e 6,2, respectivamente, estando dentro dos limites estipulados pela legislação de 30mg/100g para BNVT e 4 mg/100g de TMA. Após o processo de cocção e congelamento houve um aumento no valor do pH para 6,9, enquanto o BNVT manteve-se na média. Todas as amostras de mexilhão in natura atenderam aos padrões microbiológicos estabelecidos pela legislação (RDC, nº12 de 02 de janeiro de 2001). Salmonella sp e Vibrio parahaemolyticus não foram isoladas em nenhuma das amostras de mexilhões in natura, cozidos, congelados e armazenados. O tratamento térmico foi efetivo no controle dos coliformes fecais, redução de coliformes totais, Staphylococcus coagulase+ e psicrotróficos. O processo de congelamento reduziu a contagem de coliformes totais e Staphyloocccus coagulase+, mantendo-se inalterados durante o armazenamento a -18ºC por 90 dias. Concluiu-se que o beneficiamento do mexilhão pelo processo combinado de cocção, congelamento e armazenamento assegura a qualidade físico-química e microbiológica do produto, podendo ser adotado como padrão para industrialização. / The mussels cultivated in the coast North of São Paulo, city of Ubatuba, are commercialized in natura, constituting risk to the population. With the growth of the activity it is possible to suggest the implantation of a unit of mussel processing that promotes an increase of the storage time, facilitating the commercialization and allowing the exportation, besides supplying to the consumer a product of better quality. The mussels had been submitted to the processing for cooking, freezing and storage, being then determined the freezing point, the speed of freezing to the curves of freezing of the semidesconchado mussel. The microbiological quality and physicochemical of the product were evaluated. The processing of the mussel was started with the immersion in boiling water per 10 minutes. After the withdrawal of the shells, the mussels had been frozen individually IQF (Individually Quick Frozen) at -20ºC and stored at -18ºC during 90 days. The curve of freezing of the mussel presented typical general form, with the freezing point placing from zero to -1,5ºC; the freezing speed varied from 2 cm.h-1 to 3,3 cm.h-1, as the disposal

Avaliação da cinética populacional de Leifsonia xyli subsp. xyli em resposta a fatores do hospedeiro e do ambiente / The analysis of population kinetics of leifsonia xyli subsp. xyli in response to factors of host and environment

O raquitismo das soqueiras (RSD), causado pela bactéria Leifsonia xyli subsp. xyli (Lxx) é uma das principais doenças na cana-de-açúcar, contudo, as informações sobre a biologia do patógeno, bem como sobre sua interação com cana são limitadas. Desta forma, três estudos foram conduzidos com o objetivo de verificar a interferência de fatores do hospedeiro e do ambiente na cinética populacional da Leifsonia xyli subsp. xyli (Lxx) in planta e in vitro. Num primeiro, foi quantificada, através de PCR em tempo real (qPCR), a colonização da bactéria de plantas de duas variedades submetidas a estresse por restrição hídrica e por injúria mecânica infligida através de corte da planta. Num segundo, o desenvolvimento in vitro da bactéria foi avaliado, por 10 dias, em resposta à adição ao meio de cultivo de fluido vascular de plantas de cana. Já o terceiro estudo avaliou o efeito do hormônio ácido abscísico (ABA) na cinética populacional de Lxx em cana. No primeiro estudo, os resultados apontaram efeito positivo tanto do estresse hídrico como da injúria no título bacteriano em plantas da variedade CB 49-260. Já na variedade RB83 5486, não houve efeito significativo destes agentes de estresse no título bacteriano. No segundo estudo, o maior desenvolvimento da população de Lxx ocorreu em meio de cultivo sem adição de fluido vascular, ao passo que o desenvolvimento da população em presença de fluido da variedade resistente não inoculada com Lxx foi maior, quando em presença de fluido de plantas da mesma variedade inoculadas com a bactéria. A adição de fluido vascular da variedade suscetível, por outro lado, inibiu substancialmente o desenvolvimento das culturas bacterianas. A análise cromatográfica do fluido vascular indicou diferenças na concentração de compostos do fluido das duas variedades, bem como entre plantas da mesma variedade inoculadas ou não com a bactéria. No terceiro estudo, apesar das variações nos títulos da Lxx detectados ao longo do tempo, tanto nas plantas testemunhas como nas plantas tratadas com ABA, não houve diferenças significativas (p>0,05) entre os tratamentos dentro de um período de avaliação de 4 meses. Em seu conjunto, os resultados, além de fornecerem informações inéditas sobre a interação entre Lxx e cana, ainda permitiram direcionar futuros estudos a fim de aprimorar o conhecimento sobre os mecanismos de colonização do hospedeiro desta bactéria. / Ratoon Stunting Disease (RSD) caused by the bacterium Leifsonia xyli subsp. xyli (Lxx) is a major disease in sugarcane; however, information on the biology of this pathogen and on its interaction with sugarcane is limited. Three studies were conducted to analyze effects of the host and environment on the population kinetics of Leifsonia xyli subsp. xyli (Lxx) in plant and in vitro. In the first treatment, we used real time PCR (qPCR) to quantify bacterial colonization in two sugarcane varieties subjected to water stress and mechanical injury. In the second treatment, we evaluated the in vitro growth of the bacteria for 10 days after the addition of 15% of sugarcane vascular fluid to the culture medium. The third treatment evaluated the effect of the application of abscisic acid (ABA) on Lxx colonization in sugarcane. In the first treatment, the results showed an increase in Lxx colonization in the variety CB 49-260 exposed to water stress and mechanical injury. For the variety RB83 5486, there was no significant effect of these stresses on the bacterial colonization. In the second treatment, the greatest development of Lxx population occurred in the culture medium without the addition of vascular fluid (control), while bacterial population in the presence of fluid from resistant variety non-inoculated with Lxx was greater, compared to the growth in the presence of plant fluids of the same variety inoculated with the bacteria. The addition of vascular fluid of the susceptible variety, on the other hand, significantly inhibited bacterial growth. The chromatographic analysis of the vascular fluid indicated differences in the compounds concentration in the fluid of the two varieties, as well as in plants of the same variety inoculated or not with the bacteria. In the third study, despite variations in Lxx colonization detected over time, the control plants and plants treated with ABA showed no significant differences (p> 0.05) among treatments during the trial period of four months. The results, in addition to providing new information about the interaction between Lxx and sugarcane, allowed to direct further studies on mechanisms of host colonization of this bacterium.

Influence of pathogenic bacterial determinants on genome stability of exposed intestinal cells and of distal liver and spleen cells

Most bacterial infections can be correlated to contamination of consumables such as food and water. Upon contamination, boil water advisories have been ordered to ensure water is safe to consume, despite the evidence that heat-killed bacteria can induce genomic instability of exposed (intestine) and distal cells (liver and spleen). We hypothesize that exposure to components of heat-killed Escherichia coli O157:H7 will induce genomic instability within animal cells directly and indirectly exposed to these determinants. Mice were exposed to various components of dead bacteria such as DNA, RNA, protein or LPS as well as to whole heat-killed bacteria via drinking water. Here, we report that exposure to whole heat-killed bacteria and LPS resulted in significant alterations in the steady state RNA levels and in the levels of proteins involved in proliferation, DNA repair and DNA methylation. Characterization of Dickeya solani strains and identification of bacterial and plant signals involved in induction of virulence / Caractérisation de souches de Dickeya solani et identification de signaux bactériens ou végétaux impliqués dans l'induction de gènes de virulence

Les bactéries pectinolytiques des genres Pectobacterium (ancien nom Erwinia carotovora) et Dickeya (ancien nom Erwinia chrysanthemi) sont les agents des maladies de la jambe noire et de la pourriture molle. Ils provoquent des dommages aux cultures et des pertes économiques élevées. Les pertes causées par les bactéries pectinolytiques sont évaluées à environ 2 à 10% du rendement de pommes de terre, en fonction de l'année. En 2009, les pertes en pommes de terre en Europe ont été estimées à 250 millions d'euros. Au cours des dernières années, des souches de Dickeya ont été de plus en plus souvent isolées de plantes malades en Pologne, en France et d'autres pays européens. Le genre Dickeya est un groupe très diversifié, qui, selon la nomenclature actuelle contient sept espèces: D. aquatica, D. chrysanthemi, D. dadantii, D. dianthicola, D. paradisiaca, D. solani et D. zeae. Les résultats récents, obtenus dans différents pays européens, indiquent qu'un nouveau groupe de souches de Dickeya peut infecter efficacement les plantes de pomme de terre et causer des symptômes de la maladie en climat tempéré. Les souches de D. solani sont considérés comme plus agressives que les autres bactéries causant la jambe noire. Une analyse préliminaire a suggéré qu’elles ont besoin de plus faibles températures optimales pour le développement de la maladie ainsi que de niveaux d'inoculum inférieurs pour la propagation de l'infection. Elles semblent avoir une plus forte capacité à coloniser les racines de plantes de pomme de terre et à se propager à travers le système vasculaire de la plante. Les souches de D. solani produisent une large gamme d’enzymes dégradant de la paroi cellulaire végétale, qui sont les principaux facteurs de virulence. Les objectifs de l'étude étaient les suivants: 1) la caractérisation phénotypique et génotypique des souches de D. solani isolées dans des pays ayant des conditions climatiques différentes: Pologne, Finlande et Israël, 2) l'étude de l’influence d'extraits de pomme de terre sur l'expression de quelques gènes sélectionnés de D. solani: pelD, pelL, tssk, lfaA, 3) la génomique comparative de dix souches de D. solani, basée sur 4 génomes séquencés pour cette étude et 6 séquences génomiques disponibles dans la base de données GenBank. En conclusion, toutes les études génomiques ont montré que les souches de D. solani forment un groupe très homogène. Cependant, leur analyse phénotypique révèle une certaine variabilité entre les souches provenant de différentes conditions climatiques. La raison des variations observées dans les traits phénotypiques peut être liée à la régulation de l'expression des gènes codant les facteurs de virulence qui peuvent être influencés par la température, le pH, la carence en fer ou en oxygène et la disponibilité en azote, ainsi que par la présence de composés spécifiques des tissus végétaux. / Dickeya solani is a species consisting of newly emerged plant pathogenic bacteria that cause blackleg and soft rot diseases. They are responsible for great damages to potato plantations in most of European countries. D. solani strains produce a wide range of plant cell-wall degrading enzymes which are the main virulence factors. The aims of the study were: 1) phenotypic and genotypic characterizations of the D. solani strains isolated in countries with different climatic conditions: Poland, Finland and Israel, 2) study of the potato tuber extract influence on the expression of a few selected D. solani genes : pelD, pelL, tssK, lfaA,3) comparative genomics of ten D. solani strains, performed on 4 genomes sequenced for this study and 6 genome sequences available in the GenBank databases. The results showed that the strains from different climatic conditions have identical profiles in rep-PCR (with three different primers) and in Restriction Fragments Lenght Polymorphism-Pulse Field Gel Electrophoresis. However, they do differ phenotypically, especially in the activity of plant cell-wall degrading enzymes. Polish strains have higher activities of pectinolytic, cellulolytic and proteolytic enzymes than Finnish and Israeli strains. D. solani mutants in the pelD, pelL, tssK, lfaA genes were constructed by site-specific mutagenesis. The highest induction by plant extracts was observed for the lfaA gene. The expression of pelL is also induced by plant derived signal(s), but not that of pelD and tssK. Comparative genomics helped to elucidate the D. solani pangenome. The 10 D. solani strains genomes are coding for a total of 41 947 proteins which were grouped into 5 045 Orthologous Groups, 3 809 belonging to the core genome, 413 to the accessory genome and 823 to the unique genome. Some pathogenicity-related genes as well as their regulators were selected on the basis of the knowledge available for D. dadantii 3937, the most studied Dickeya strain, which belongs to a closely related species. Analysis of their protein sequence showed no difference in the sequence of those genes within the 10 genomes. All the genetic studies proved that D. solani strains form a very homogenous group. On the other hand, the phenotypic analysis showed some variability among strains from different climatic conditions. The observed variations in the phenotypic traits can results from a different regulation of the expression of the genes encoding virulence factors which are influenced by temperature, pH, iron deprivation, oxygen and nitrogen availability, as well as by the presence of plant compounds.

Most HC and HUS outbreaks have been attributed to strains of entero-hemorrhagic serotype O157:H7, but there is growing interest on the risk to human health associated with the non-O157 STEC serotypes carried by beef products contaminated by ruminant feces. This study aimed to determine the occurrence of STEC in a cattle slaughterhouse located in the State of Minas Gerais. Sampling was carried out in two lots; Lot A with 75 bovines and Lot B with 51 animals in three stages of the slaughtering line: removal of skin after bleeding, carcass after sawing, and feces during evisceration. Samples were collected from ten animals of each lot. The samples were enriched in modified EC broth (mEC) and modified EC added with novobiocin (mECn) followed by isolation of typical colonies on Sorbitol MacConkey Medium. Two hundred and forty-one bacterial isolates obtained from the two lots, were identified as E. coli. Of these, 104 were derived from the skin, 93 from fecal material and 44 from carcasses. Polymerase chain reaction detected the stx gene in 18 isolates of E. coli. These isolates were obtained from six animals: four were of fecal origin and come from lot A; two were from carcasses from lot B. Enrichment in mECn resulted in isolates originated from one animal, whereas in mEC, the isolates were originated from five bovines. Isolates stx positive were analyzed for the hlyA gene encoding enterohemolysin. Four hlyA positive isolates originated from two bovines, one from lot A and another from lot B. Two displayed the entero- hemolytic phenotype and were isolated from the carcass. Ten of the 18 potentially STEC isolates showed cytotoxic effect on Vero cells. Eight isolates from three different animals had the stx PCR amplicons sequenced. The translated sequences obtained from stx of three isolates from the carcass and of one isolate from feces corresponded to Stx2, showing 98% to 99% identity, with references at the GeneBank (NCBI). The sequences inferred from four amplicons obtained from isolates of faecal origin corresponded to Stx1, with 94% to 96% identity. The presence of STEC in the feces and carcasses of cattle at a slaughterhouse in the state of Minas Gerais emphasizes the importance of establishing a HACCP system and the need for implementation, evaluation, and validation of control measures to minimize the risk of cross-contamination during and after slaughter. / Escherichia coli Shiga-toxigênica, STEC, tem emergido como patógeno que pode causar infecções de origem alimentar e doenças severas e potencialmente fatais, como Colite Hemorrágica (CH) e Síndrome Urêmico Hemolítica (SUH). A maioria dos surtos de CH e SUH tem sido atribuída a estirpes do sorotipo entero-hemorrágico O157:H7, entretanto existe interesse crescente sobre o risco à saúde humana associado com os sorotipos STEC não-O157 veiculados em produtos de carne bovina, contaminados por fezes de ruminantes. Este trabalho teve como objetivos determinar a ocorrência de STEC em bovinos em matadouro frigorífico, localizado no Estado de Minas Gerais. A amostragem foi realizada em dois lotes, lote A com 75 bovinos e lote B com 51 bovinos, em três etapas da linha de abate: pele, após a sangria; carcaça, após a serragem; e fezes, durante a evisceração. Dez animais de cada lote foram amostrados. As amostras foram enriquecidas em caldo EC modificado, mEC e EC modificado acrescido de Novobiocina, mECn, seguindo-se o isolamento de colônias típicas em meio MacConkey sorbitol. Duzentos e quarenta e um isolados bacterianos obtidos nos dois lotes foram identificados como E. coli. Desses, 104 foram provenientes da pele, 93 do material fecal e 44 das carcaças. Por reação em cadeia de polimerase, foi detectado o gene stx em 18 isolados de E. coli. Esses isolados foram provenientes de seis bovinos, sendo quatro do lote A (origem fecal) e dois do lote B (carcaça). O enriquecimento em meio mECn resultou em isolados originados de um bovino, enquanto no mEC os isolados se originaram de cinco bovinos. Os isolados stx positivos foram posteriormente analisados para o gene hlyA, codificador de entero-hemolisina. Quatro isolados, positivos para o gene hlyA, foram provenientes de dois bovinos, um do lote A (origem fecal) e outro do lote B (carcaça). Desses, dois isolados da carcaça apresentaram fenótipo entero-hemolítico. Dez dos 18 isolados potencialmente STEC demonstraram ter efeito citotóxico em células Vero. Oito isolados, provenientes de três bovinos distintos, tiveram os amplicons stx seqüenciados. As seqüências traduzidas de três isolados da carcaça e de um das fezes corresponderam a Stx2, com 98% a 99% de identidade, com referências no GeneBank (NCBI). As seqüências inferidas de quatro amplicons, de origem fecal, corresponderam, com 94% a 96% de identidade, a Stx1. A presença de STEC nas fezes e carcaça de bovinos em matadouro frigorífico no Estado de Minas Gerais enfatiza a importância de implementação do sistema HACCP, e a necessidade de implementar, avaliar e validar medidas de controle que minimizem os riscos de contaminação cruzada durante o processo de abate.

Em seu conjunto, os resultados, além de fornecerem informações inéditas sobre a interação entre Lxx e cana, ainda permitiram direcionar futuros estudos a fim de aprimorar o conhecimento sobre os mecanismos de colonização do hospedeiro desta bactéria. / Ratoon Stunting Disease (RSD) caused by the bacterium Leifsonia xyli subsp. xyli (Lxx) is a major disease in sugarcane; however, information on the biology of this pathogen and on its interaction with sugarcane is limited. Three studies were conducted to analyze effects of the host and environment on the population kinetics of Leifsonia xyli subsp. xyli (Lxx) in plant and in vitro. In the first treatment, we used real time PCR (qPCR) to quantify bacterial colonization in two sugarcane varieties subjected to water stress and mechanical injury. In the second treatment, we evaluated the in vitro growth of the bacteria for 10 days after the addition of 15% of sugarcane vascular fluid to the culture medium. The third treatment evaluated the effect of the application of abscisic acid (ABA) on Lxx colonization in sugarcane. In the first treatment, the results showed an increase in Lxx colonization in the variety CB 49-260 exposed to water stress and mechanical injury. For the variety RB83 5486, there was no significant effect of these stresses on the bacterial colonization. In the second treatment, the greatest development of Lxx population occurred in the culture medium without the addition of vascular fluid (control), while bacterial population in the presence of fluid from resistant variety non-inoculated with Lxx was greater, compared to the growth in the presence of plant fluids of the same variety inoculated with the bacteria. The addition of vascular fluid of the susceptible variety, on the other hand, significantly inhibited bacterial growth. The chromatographic analysis of the vascular fluid indicated differences in the compounds concentration in the fluid of the two varieties, as well as in plants of the same variety inoculated or not with the bacteria. In the third study, despite variations in Lxx colonization detected over time, the control plants and plants treated with ABA showed no significant differences (p> 0.05) among treatments during the trial period of four months. The results, in addition to providing new information about the interaction between Lxx and sugarcane, allowed to direct further studies on mechanisms of host colonization of this bacterium.

Écologie de Pseudomonas syringae dans un bassin versant : vers un modèle de transfert : des habitats naturels aux agro-systèmes / The ecology of Pseudomonas syringae in watersheds : towards a model of transfer from natural habitats to agrosystems

Caractériser la dissémination des bio-agresseurs est un enjeu majeur pour la gestion et la prédiction des maladies en santé des plantes. Face aux limites des approches usuelles en pathologie végétale, une nouvelle vision a été proposée abordant les paradigmes d’histoire de vie des agents phytopathogènes en dehors des limites du système hôte-pathogène. Parmi ces agents phytopathogènes, les études sur P.syringae sont celles qui ont contribuées le plus à ce nouveau courant de pensée et dont on connaît le mieux l’histoire de vie en relation avec ses réservoirs « non hôtes ». L’espèce est détectée dans de nombreux compartiments du cycle de l’eau, des précipitations jusqu’aux rivières et eaux d’irrigation, en passant par les plantes sauvages et le manteau neigeux. L’ensemble de ces observations ont soulevé de nouvelles questions sur la manière dont P. syringae se dissémine au travers de ces environnements et sur les processus impactant sur la dynamique des populations à l’échelle d’un bassin versant. Ces recherches se sont donc intéressées à ses processus dans des précipitations jusqu’aux cours d’eau alpins dans l’optique d’acquérir des données pour la modélisation des flux de P. syringae. Elles ont mis en évidence les populations résidentes de la litière et la survie sa survie dans le sol, processus jamais identifiés à l’histoire de vie de P. syringae. Elles ont également caractérisé (i) les conditions propices à son transport via les précipitations, (ii) le rôle du manteau neigeux comme réservoir et protecteur des populations des prairies alpines et (iii) ont mis en évidence la chimie de l’eau comme indicateur témoin de la dynamique des populations des les rivières. Ces observations suggérant un transport de P. syringae dans le sol, nous l’avons quantifiée à travers des études de terrain et des simulations en laboratoire.Enfin, l’ensemble des données de ces recherches couplées à des outils SIG et des modèles météorologiques et hydrologiques ont permis de proposé un modèle sur les flux de P.syringae des habitats naturels vers les agro-systèmes. / The characterization of the spread of bio-agressors spread is a major issue for themanagement of plant health and the prediction of disease emergence. Given thelimitations of conventional approaches in plant pathology, a new vision has beenproposed addressing paradigms life history of plant pathogens outside the limits of thecrop host-pathogen system. Among the plant pathogens, studies on P. syringae are thosethat have contributed the most to this new way of thought for which life history inrelation to "non host" reservoirs has been highlighted. The species is found in manycompartments of the water cycle, from precipitation to rivers and irrigation water, wildplants and snowpack. All these observations have raised new questions about how P.syringae spreads through these environments and on the processes impactingpopulation dynamics at the scale of a watershed. This research was therefore interestedin these processes with the objective to acquire data for modeling the tranfer of P.syringae through the watershed. They highlighted the resident populations of litter andtheir survival in the soil, processes never identified in association with the life history ofP. syringae. They also revealed (i) the conditions for transport via precipitations, (ii) therole of snowpack as a reservoir and protector of the populations in alpine meadows and(iii) showed that water chemistry can be used as an indicator of the populationdynamics in headwaters. These observations suggested a transport of P. syringae via thesoil that we subsequently characterized through field studies and laboratorysimulations. Finally, all data from this research combined with GIS tools andmeteorological and hydrological models have permitted us to propose a model of theflux of P. syringae of natural habitats to agricultural systems.

Qualidade do mexilhão Perna perna submetido ao processo combinado de cocção, congelamento e armazenamento / The quality of the mussel (Perna perna) processing by cooking, freezing and storage

Os mexilhões cultivados no litoral Norte de São Paulo, município de Ubatuba, são comercializados in natura, constituindo risco à população. Com o crescimento da atividade é possível sugerir a implantação de uma unidade de processamento de mexilhões que promova um aumento do tempo de armazenamento, facilitando a comercialização e permitindo a exportação, além de fornecer ao consumidor um produto de melhor qualidade. Os mexilhões foram submetidos ao processamento por cocção, congelamento e armazenamento, sendo então determinados o ponto de congelamento, a velocidade de congelamento a as curvas de congelamento do mexilhão semidesconchado. A qualidade microbiológica e físico-química do produto foi avaliada. O beneficiamento do mexilhão iniciou-se com a cocção por imersão em água à ebulição por 10 minutos. Após a retirada das conchas, os mexilhões foram congelados individualmente IQF (Individually Quick Frozen) a –20ºC e armazenados a -18ºC durante 90 dias. A curva de congelamento do mexilhão apresentou forma geral típica, com o ponto de congelamento situando-se na faixa de zero a –1,5ºC; a velocidade de congelamento variou de 2 cm/h a 3,3 cm/h, conforme a disposição dentro da câmara de congelamento. Os resultados físico-químicos mostraram que não houve diferença significativa no valor nutricional dos mexilhões in natura, processados e armazenados, apresentando os teores médios de 7,4mg/100g de proteínas, 5,8 mg/100g de carboidratos e 1,4 mg/100g de lipídeos. Os valores encontrados para BNVT, TMA e pH no mexilhão in natura foram, 4,3 mg/100g; 2,0 mg/100g e 6,2, respectivamente, estando dentro dos limites estipulados pela legislação de 30mg/100g para BNVT e 4 mg/100g de TMA. Após o processo de cocção e congelamento houve um aumento no valor do pH para 6,9, enquanto o BNVT manteve-se na média. Todas as amostras de mexilhão in natura atenderam aos padrões microbiológicos estabelecidos pela legislação (RDC, nº12 de 02 de janeiro de 2001). Salmonella sp e Vibrio parahaemolyticus não foram isoladas em nenhuma das amostras de mexilhões in natura, cozidos, congelados e armazenados. O tratamento térmico foi efetivo no controle dos coliformes fecais, redução de coliformes totais, Staphylococcus coagulase+ e psicrotróficos. O processo de congelamento reduziu a contagem de coliformes totais e Staphyloocccus coagulase+, mantendo-se inalterados durante o armazenamento a –18ºC por 90 dias. Concluiu-se que o beneficiamento do mexilhão pelo processo combinado de cocção, congelamento e armazenamento assegura a qualidade físico-química e microbiológica do produto, podendo ser adotado como padrão para industrialização. / The mussels cultivated in the coast North of São Paulo, city of Ubatuba, are commercialized in natura, constituting risk to the population. With the growth of the activity it is possible to suggest the implantation of a unit of mussel processing that promotes an increase of the storage time, facilitating the commercialization and allowing the exportation, besides supplying to the consumer a product of better quality. The mussels had been submitted to the processing for cooking, freezing and storage, being then determined the freezing point, the speed of freezing to the curves of freezing of the semidesconchado mussel. The microbiological quality and physicochemical of the product were evaluated. The processing of the mussel was started with the immersion in boiling water per 10 minutes. After the withdrawal of the shells, the mussels had been frozen individually IQF (Individually Quick Frozen) at -20ºC and stored at -18ºC during 90 days. The curve of freezing of the mussel presented typical general form, with the freezing point placing from zero to -1,5ºC; the freezing speed varied from 2 cm.h-1 to 3,3 cm.h-1, as the disposal
