Origins and use of the stochastic and sound-evoked extracellular activity of the auditory nerveBrown, Daniel January 2007 (has links)
[Truncated abstract] The present study investigated whether any of the characteristics of the compound action potential (CAP) waveform or the spectrum of the neural noise (SNN) recorded from the cochlea, could be used to examine abnormal spike generation in the type I primary afferent neurones, possibly due to pathologies leading to abnormal hearing such as tinnitus or tone decay. It was initially hypothesised that the CAP waveform and SNN contained components produced by the local action currents generated at the peripheral ends of the type I primary afferent neurones, and that changes in these local action currents occurred due to changes in the membrane potential of these neurones. It was further hypothesised that the lateral olivo-cochlear system (LOCS) efferent neurones regulate the membrane potential of the primary afferent dendrites to maintain normal action potential generation, where instability in the membrane potential might lead to abnormal primary afferent firing, and possibly one form of tinnitus. We had hoped that the activity of the LOCS efferent neurones could be observed through secondary changes in the CAP waveform and SNN, resulting from changes in the membrane potential of the primary afferent neurones. The origins of the neural activity generating the CAP waveform and SNN peaks, and the effects of the LOCS on the CAP and SNN were experimentally investigated in guinea pigs using lesions in the auditory system, transient ischemia and asphyxia, focal and systemic temperature changes, and pharmacological manipulations of different regions along the auditory pathway. ... Therefore, the CAP and SNN are altered by changes in the propagation of the action potential along the primary afferent neurones, by changes in the morphology of the tissues surrounding the cochlear nerve, and by changes in the time course of the action currents. If the CAP waveform is not altered, the amplitude of the 1kHz speak in the spontaneous SNN can be used as an objective measure of the spontaneous firing rate of the cochlear neurones. However, because the SNN contains a complex mixture of neural activity from all cochlear neurones, and the amplitude of the spontaneous SNN is variable, it would be difficult to use the spontaneous SNN alone as a differential diagnostic test of cochlear nerve pathologies. To record extratympanic electrocochleography (ET ECochG) from humans, a custom-designed, inexpensive, low-noise, optically isolated biological amplifier was built. Furthermore, a custom-designed extratympanic active electrode and ear canal indifferent electrode were designed, which increased the signal-to-noise ratio of the ECochG recording by a factor of 2, decreasing the overall recording time by 75%. The human and guinea pig CAP waveforms recorded in the present study appeared similar, suggesting that the origins of the human and guinea pig CAP waveforms were the same, and that experimental manipulations of the guinea pig CAP waveform can be used to diagnose the cause of abnormal human ECochG waveforms in cases of cochlear nerve pathologies.

Lors de la phonation, des phénomènes complexes mettent en jeu des interactions entre l'écoulement d'air provenant des poumons, les cordes vocales et des résonateurs acoustiques (trachée, conduit vocal et nasal). Cette thèse se focalise sur la validation expérimentale d'un des modèles de cordes vocales les plus utilisés: le modèle à deux masses. Les interactions de ce modèle avec l'écoulement d'air et les résonateurs acoustiques sont d'abord décrites théoriquement. Des outils d'analyses des descriptions théoriques permettant d'en extraire des paramètres pertinents en parole sont explicités. Un banc expérimental, basé sur des maquettes de cordes vocales, est ensuite utilisé pour tester la validité des prédictions théoriques obtenues avec le modèle. Enfin, deux exemples d'applications de l'utilisation du modèle à deux masses et ses interactions sont donnés.

Effects of Specific Cochlear Pathologies on the Auditory Functions : Modelling, Simulations and Clinical ImplicationsSaremi, Amin G. January 2014 (has links)
A hearing impairment is primarily diagnosed by measuring the hearing thresholds at a range of auditory frequencies (air-conduction audiometry). Although this clinical procedure is simple, affordable, reliable and fast, it does not offer differential information about origins of the hearing impairment. The main goal of this thesis is to quantitatively link specific cochlear pathologies to certain changes in the spectral and temporal characteristics of the auditory system. This can help better understand the underlying mechanisms associated with sensorineural hearing impairments, beyond what is shown in the audiogram. Here, an electromechanical signal-transmission model is devised in MATLAB where the parameters of the model convey biological interpretations of mammalian cochlear structures. The model is exploited to simulate the cell-level cochlear pathologies associated with two common types of sensorineural hearing impairments, 1: presbyacusis (age-related hearing impairment) and, 2: noise-induced hearing impairment. Furthermore, a clinical study, consisting of different psychoacoustic and physiological tests, was performed to trace and validate the model predictions in human. The results of the clinical tests were collated and compared with the model predictions, showing a reasonable agreement. In summary, the present model provides a biophysical foundation for simulating the effect of specific cellular lesions, due to different inner-ear diseases and external insults, on the entire cochlear mechanism and thereby on the whole auditory system. This is a multidisciplinary work in the sense that it connects the ‘biological processes’ with ‘acoustic modelling’ and ‘clinical audiology’ in a translational context.

A construtibilidade como requisito para avaliação de componentes para a edificação: o caso do Elemento de Integração alvenaria/esquadriaZucchetti, Lais January 2010 (has links)
Atualmente a utilização de sistemas modulares na indústria da construção tem crescido significativamente, e, dentre estes, destaca-se o sistema construtivo de alvenaria estrutural. Entretanto, diversos estudos apontam para um grande número de manifestações patológicas relacionadas a este sistema, em especial com a interface alvenaria/ esquadria como um dos locais de maior incidência, além de representar um gargalo construtivo, pela necessidade de realização de inúmeras atividades dependentes. Dentro deste contexto, a equipe do projeto COMPOHIS (FINEP) desenvolveu uma nova proposta de solução a estas questões, denominada Elemento de Integração (EI). Para a implementação deste novo elemento, foram necessárias avaliações que garantissem o comportamento adequado do produto nas construções e, um dos requisitos considerados foi a construtibilidade, visto que sua consideração e implementação trazem benefícios relacionados ao custo, tempo e qualidade das construções. Desta forma, o objetivo principal desta pesquisa foi avaliar a construtibilidade do EI. Para que esta avaliação fosse realizada, foram identificados, a partir de revisão bibliográfica, vinte e sete requisitos de construtibilidade. Dentre estes, foram selecionados vinte e dois e, agrupados em dez requisitos básicos. Estes dez requisitos foram utilizados na análise ao longo do desenvolvimento da pesquisa, considerando protótipos virtuais e físicos (construídos com diferentes materiais). Mais especificamente, foram utilizados quatro protótipos virtuais e três protótipos físicos do EI, sendo um em MDF e dois em GRC, e duas modelagens de seqüências construtivas, sendo uma com o EI e a outra com o método tradicional de construção. Sendo que, na última fase, foi construída uma parede experimental real com o EI e, submetida ao ensaio estrutural. Os principais resultados demonstram a adequação e aplicabilidade, formal, funcional e conceitual do EI ao sistema construtivo considerando os requisitos selecionados, com algumas ressalvas quanto à conexão entre o Elemento e a alvenaria (devido às fissuras identificadas nesta interface), quanto à dificuldade de manutenção dos componentes em GRC (considerando a necessidade de execução de substituições), quanto à exclusão de ajustes em canteiro, com relação à necessidade de cuidado com as arestas do EI e, finalmente com relação à segurança (pela instabilidade do EI logo que assentado). / Nowadays, the use of modular systems in the construction industry has grown substantially and, among those, the structural masonry system gains relevance. However, several studies point to a high incidence of pathological manifestations related to this system, particularly the interface between masonry and windows being one of the places with the highest number of problems, besides being a constructive bottleneck, since several activities depend on it. Within this context, the COMPOHIS project developed new solutions to these problems, named Element of Integration (EI). However, to implement this new element, performance evaluations were needed to ensure the adequate performance of the product in buildings, and one of these performance requirements that must be considered is the constructability of the solution. The use of this requirement brings benefits related to cost, time and quality of constructions. Thus, the main objective of this research is to evaluate the constructability of the EI. For this evaluation, 27 constructability requirements were identified in the literature. From those, 22 were selected and grouped into 10 major requirements. These 10 requirements were considered for analysis along the preparation and execution phases of this work, and the analysis was carried out considering physical and virtual prototypes (built with different materials). More specifically, 4 virtual prototypes and three physical prototypes of EI we used - one in MDF and two in GRC - and two sequences of constructive modeling, one with the EI and the other with the traditional method of construction. The last GRC prototype was used to build a wall, which was used for a structural resistance test. The main results demonstrated the formal, functional and conceptual adequacy of EI to the selected requirements, with some reservations about the connection between the element and the structural masonry, due to cracks observed in this interface, the difficulty of maintenance of GRC components, considering the need for repairs and replacements, the need to use protective elements at the edges of the GRC components and finally, in regard to safety, due to the instability of the EI when set in place.

Observation, modélisation et simulation des vibrations des maquettes de plis vocaux : applications à des configurations pathologiques / Observation, modelling and simulation of the vibrations of a vocal folds replica with applications to pathological configurationsHermant, Nicolas 09 October 2014 (has links)
Les travaux de cette thèse portent sur la compréhension des phénomènes physiques sous-jacents à la production normale ou pathologique de la voix. Certaines formes de pathologies pouvant concerner des affections structurelles des cordes vocales (polypes, paralysie, ...) et altérer de façon plus ou moins conséquente la mise en vibration des cordes vocales. Une partie de ces travaux porte ainsi sur l'observation expérimentale de la production de parole pathologique, grâce notamment à la mise au point d'une maquette auto-oscillante de cordes vocales artificielles. Celle-ci reprend la structure de latex remplie d'eau sous pressions des maquettes déjà développées au Gipsa-lab mais permet une plus grande indépendance des paramètres de contrôle. Un accent particulier des manipulations expérimentales est également porté sur la caractérisation mécanique des structures vibrantes, dont la connaissance est indispensable à la reproductibilité et la répétabilité des mesures ultérieures sur la maquette, ainsi qu'à l'analyse des résultats expérimentaux avec couplage aéroacoustique et la validation de modèles numériques associés.D'un côté, un calcul du comportement vibratoire des maquettes basée sur une méthode éléments finis a été mise oeuvre. Le modèle numérique développé utilise ainsi une formulation du couplage hydro-élastique entre le latex et l'eau qui constituent les maquettes et offre la possibilité de prendre en compte les grandes déformations et les pré-contraintes liées au gonflement du latex soumis à une forte pression d'eau. Finalement, un modèle d'écoulement glottique couplé à un modèle mécanique distribué de type "masse-raideur" a été utilisé et étendu pour simuler des comportements pathologiques tels que des asymétries (entre les deux cordes vocales et au sein d'une corde) ou la présence d'une masse ajoutée (kyste, polype). Un procédure d'optimisation de ce modèle sur une configuration expérimentale de référence permet alors de comparer les calculs aux observations des seuils d'oscillation sur les maquettes en présence d'une masse ajoutée. / My PhD work has focused on understanding physical phenomena related to speech production in healthy and pathological conditions. Some pathologies affect the structure of the vocal folds which could lead to a more or less substantial alteration of their vibrations. Part of this work is experimental modeling of speech production applied to pathologies, with the development of a self-oscillating vocal fold replica. This replica is made of water-filled latex, such as have already been developed at Gipsa-lab, but it allows a larger independence within its control parameters and it can be used to reproduce pathological conditions. A particular interest is brought to mechanical characterization of these vibrating structures. This characterization is of primary importance when it comes to repeatability and reproducibility of measurements, as for the analysis of experimental results with aeroacoustic coupling and the validation and improvements of corresponding numerical models.Simulations of the vocal fold vibrating behavior based on the finite element method have been developed. The model uses a formulation of the hydro-elastic coupling between the latex and the water which the replica is made of. It also offers the possibility to take into account the large deformations and the prestress within the latex induced by water pressure. Another numerical approach is made using a theoretical laryngeal flow model coupled with a distributed mass-spring model and a set of resonators. Calculations of the dynamical system stability offer the possibility for comparisons with the experimental setup through a finite set of parameters. This model is adapted to simulate pathological conditions such as asymmetries and the presence of growths (cysts, polyps). Finally, an optimization calculation of this model on an experimental reference configuration of the replica allows us to compare the calculations with the measurements on this replica with the presence of a polyp.

Several segments of society are, nowadays, worried about keeping alive the
history of our cities. This work presents a case study about Olavo Bilac State Institute
of Education, in Santa Maria, RS, Brazil, in order to show its current situation,
through an analysis of the outer facades of two buildings which are part of the main
complex built in the early twentieth century. We started with a bibliographic review
which exposes a view about education in Brazil and in Rio Grande do Sul, In
addition, we contextualized the Institutes of Education inside the State, emphasizing
their history and architectonic characteristics. We also presented decisive items to
understand the further steps of the research, such as pathological manifestations and
degradation factors in relevant historical buildings. Therefore, such data are part of
the case study which provided an overall view about the buildings, as well as allowed
the achievement of an investigation about the causes of these damages. The results
showed that a great part of the facades is endangered with some pathology, or the
ones which are not visibly endangered are an alert to the bilaquian population and
Santa Maria s society to put this property, which is under governmental trust, at risk.
Finally, with the data obtained through the survey and mapping of the existing
pathologies or damages, we propose a preservation plan for future actions which can
use this material as a support for the interventions, with high or low significance. / A preocupação em manter viva a história de nossas cidades é vista e
percebida por diversos setores da sociedade. Este trabalho apresenta um estudo de
caso, elaborado sobre o Instituto Estadual de Educação Olavo Bilac, na cidade de
Santa Maria/RS a fim de mostrar as condições atuais do mesmo, através de uma
análise sobre as fachadas externas dos dois prédios do complexo principal datado
do começo do século XX. Partiu-se com a revisão bibliográfica expondo um
panorama acerca da educação no Brasil e no Estado do Rio Grande do Sul, além de
realizar uma contextualização dos institutos estaduais disseminados no interior do
estado, evidenciando sua história e características arquitetônicas. Apresentou-se
ainda, itens determinantes para compreensão das etapas posteriores da pesquisa,
como as manifestações patológicas e os fatores de degradação em edificações de
relevância histórica. Neste âmbito, tais dados fizeram parte do estudo de caso que
forneceu uma visão geral sobre as edificações bem como ainda, possibilitou que
fosse realizada uma investigação acerca das causas destes danos. Os resultados
mostraram que existe grande parte das fachadas das edificações comprometidas
com alguma patologia, ou as que não foram detectadas mais visivelmente
encontram-se como um alerta para a população bilaquiana e sociedade
santamariense por colocarem este bem, que é tombado municipalmente, em risco.
Por fim com os dados obtidos através do levantamento e mapeamento das
patologias e/ou danos existentes, propôs-se um plano de preservação para que
ações futuras possam ter este material como auxilio em intervenções sejam elas de
maior ou menor significância.

Previous issue date: 2018-04-20 / Norteado pelo método analítico e interpretativo da Sociologia do Conhecimento de Karl Mannheim, o presente trabalho defende enquanto hipótese central que, no caso especificamente brasileiro do século XXI, as igrejas evangélicas neopentecostais apresentam um maior poder competitivo e de flexibilização adaptativa às profundas e radicais transformações socioculturais desencadeadas pela radicalização dos processos de modernização em tempos de globalização contemporânea do capitalismo, agindo nas subjetividades humanas (no “ego”), ao dar-lhes exatamente aquilo que lhes falta enquanto agentes competitivos: a força motivacional da fé para o agir social como potencial empreendedor “homem de negócios” bem sucedido, assim como da força do ânimo para ser virtuoso e vitorioso nos mercados concorrenciais cada vez mais acirrados, devido, sobretudo, ao sentido radical da ideia de conversão presente nas comunidades evangélicas neopentecostais. A força e o poder do sentido da noção paulina de conversão presente entre os neopentecostais oferecem aos seus fiéis um conjunto de respostas utilitárias e pragmáticas aos seres humanos que se encontram imersos e submersos nas sociedades narcísicas e hipercompetitivas do “ego”. Trata-se do poder de nascer de novo, do desejar ser outro, concedendo aos fiéis convertidos às expectativas com relação ao desenvolvimento dos sensos de autoconfiança no próprio “eu”. Logo, tratando-se de formas de religiosidades interessadas em contribuírem para a felicidade do indivíduo neste mundo, e não mais em um além-mundo ultraterreno. E, por isso mesmo, trata-se de uma forma de religiosidade dessacralizada. Paradoxalmente, no entanto, se as comunidades evangélicas neopentecostais apontam a presença do mal (por meio da personificação da figura do diabo) nas próprias relações sociais que foram degeneradas pelo individualismo excessivo das sociedades modernas contemporâneas (desde a ansiedade, vazios substanciais do “eu”, desânimo, vícios, uso de medicamentos psicotrópicos e psicoestimulantes, senso de autoculpabilidade pelo fracasso, sentimentos de impotência quanto a ser capaz e eficiente o suficiente para o cumprimento de seus deveres, divórcio, união homoafetivas e homossexualismo, depressão nervosa, etc.), ela não apenas efetua um diagnóstico da realidade social centrado na perversidade das figuras dos ateus, dos agnósticos, dos homossexuais, dos divorciados, das prostitutas, dos viciados e etc., mas, também apresenta um efeito terapêutico: o da negação da própria modernidade, por meio da anulação e não reconhecimento do processo de dilatação de novas demandas por direitos, construindo, portando, uma identidade religiosa potencialmente imunizadora e conservadora. / Based on the analytical and interpretative method of the Sociology of Knowledge of Karl Mannheim, the present work defends as central hypothesis that, in the specifically Brazilian case of the 21st century, the neo-Pentecostal evangelical churches present a greater competitive power and adaptive flexibilization to the deep and radical socio-cultural transformations Unleashed by the radicalization of modernization processes in times of contemporary globalization of capitalism. Acting on human subjectivities (in the "ego"), by giving them exactly what they lack as competitive agents: the motivational force of faith for social action as a successful entrepreneurial "business man", as well as for strength of mind To be virtuous and victorious in increasingly competitive markets, due above all to the radical sense of the idea of conversion present in Neo-Pentecostal evangelical communities. The power and power of the sense of the Pauline notion of present conversion among NeoPentecostals offer their believers a set of utilitarian and pragmatic responses to human beings who are immersed and submerged in the narcissistic and hypercompetitive societies of the "ego." It is the power to be born again, to wish to be another, by giving the converted faithful the expectations of developing self-confidence in self. Therefore, these are forms of religiosities interested in contributing to the happiness of the individual in this world, and no longer in an outer world beyond. And, for this very reason, it is a form of desacralized religiosity. Paradoxically, however, if the Neo-Pentecostal evangelical communities point to the presence of evil (through the personification of the devil's figure) in the very social relations that have been degenerated by the excessive individualism of contemporary modern societies (from anxiety, substantial emptiness of the "I" , Discouragement, addictions, use of psychotropic medications and psychostimulants, assumption of self-blame for failure, feelings of impotence as well as being able and efficient enough to fulfill their duties, divorce, union homoafetivas and homosexuality, depression and etc.) It not only makes a diagnosis of the social reality centered on the perversity of the figures of atheists, agnostics, homosexuals, divorcers, prostitutes, addicts, etc., but as they also have a therapeutic effect: that of denying one's own Modernity, through the annulment and non-recognition of the process of expansion of new demands for rights. Building, carrying, a potentially immunizing and conservative religious identity.

However, to implement this new element, performance evaluations were needed to ensure the adequate performance of the product in buildings, and one of these performance requirements that must be considered is the constructability of the solution. The use of this requirement brings benefits related to cost, time and quality of constructions. Thus, the main objective of this research is to evaluate the constructability of the EI. For this evaluation, 27 constructability requirements were identified in the literature. From those, 22 were selected and grouped into 10 major requirements. These 10 requirements were considered for analysis along the preparation and execution phases of this work, and the analysis was carried out considering physical and virtual prototypes (built with different materials). More specifically, 4 virtual prototypes and three physical prototypes of EI we used - one in MDF and two in GRC - and two sequences of constructive modeling, one with the EI and the other with the traditional method of construction. The last GRC prototype was used to build a wall, which was used for a structural resistance test. The main results demonstrated the formal, functional and conceptual adequacy of EI to the selected requirements, with some reservations about the connection between the element and the structural masonry, due to cracks observed in this interface, the difficulty of maintenance of GRC components, considering the need for repairs and replacements, the need to use protective elements at the edges of the GRC components and finally, in regard to safety, due to the instability of the EI when set in place.

Les stress oxydant et carbonylé interviennent dans le mécanisme physiopathologique de la dégénérescence maculaire liée à l'âge (DMLA) et de la maladie de Stargardt, constituant ainsi une cible thérapeutique intéressante. Une molécule lead de type lipophénol a préalablement été mise en évidence. Cette molécule est composée d’une partie phénolique de type alkylphloroglucinol, et lipidique de type oméga-3 (acide docosahexaènoïque, C22:6, ω3, DHA). Afin d’améliorer l’activité antioxydante de ce lipophénol, la synthèse totale de deux DHA deutérés en positions bis-allyliques a été réalisée. Leurs couplages au phloroglucinol alkylé a ensuite conduit aux dérivés lipophénoliques deutérés correspondants. L’impact des deutériums sur la diminution de la toxicité cellulaire et de la peroxydation lipidique a ensuite été mis en évidence lors d’une évaluation biologique in vitro réalisée en cellules de l’épithélium pigmentaire rétinien. / Age Related Macular Degeneration (AMD) and Genetic Stargardt’s disease share a common cytotoxic mechanism involving carbonyl and oxidative stress. Hence both stresses represent promising therapeutic targets. A lipophenol lead molecule has been previously highlighted. This molecule is composed by an alkylphloroglucinol moiety and a lipid part which is a polyunsaturated fatty acid, the docosahexaenoic acid (DHA, C22:6, ω3). In order to improve the antioxidant activity of this lipophenol, the total synthesis of two deuterated DHA on bis-allylic positions was performed. The cross-coupling between the deuterated DHA and the alkylphloroglucinol moiety led to the corresponding deuterated lipophenols. The impact of the deuterium to limit the cell toxicity and the lipid peroxidation was highlighted thanks to in vitro biological studies performed on retinal pigment epithelium cell line.
