Solubilité aqueuse, coefficient de partage octanol-eau et pression de vapeur de contaminants alimentaires organiques de la famille des phthalates et alkylphénols : détermination expérimentale et modélisation / Aqueous solubility, octanol-water partition coefficient and vapor pressure of organic food packaging contaminants : experimental determination and modeling

Cette étude s’attache à la caractérisation physico-chimique des contaminants des emballages alimentaires organiques, précisément les phtalates et les alkylphénols, en terme de solubilité aqueuse, coefficient de partage octanol-eau et pression de vapeur. Cette caractérisation se situe dans le cadre des règlementations REACH concernant l’identification des substances chimiques toxiques. Les mesures expérimentales de solubilité aqueuse sont effectuées par la méthode dynamique de saturation dans un intervalle de température [298.15-328.15K], celles du coefficient de partage octanol-eau avec la méthode « shake-flask » à 298.15. Les mesures de pression de vapeur sont réalisées avec la méthode dynamique de saturation dans un intervalle de température [313.15-423.15K] et validées avec la méthode statique. A partir de ces mesures, les coefficients de partage air-eau et octanol-air sont déterminés. En addition des mesures expérimentales, une prédiction de ces propriétés est effectuée avec les modèles thermodynamiques : UNIFAC originale, UNIQUAC, NRTL et COSMO-sac-dsp. A l’issue de ces résultats, une évaluation quantitative et qualitative de chacun des modèles est effectuée. Cette évaluation facilitera l’optimisation des paramètres de chacun des modèles des deux familles dans le but de créer un modèle de prédiction du phénomène de migration de l’emballage vers l’aliment / The aim of this study is the physical-chemical characterization of the organic food packaging contaminants, particularly phthalates and alkyl phenols, in terms of aqueous solubility, octanol-water partition coefficient and vapor pressure. This characterization falls within the scope of REACH regulations for the identification of chemical substances. The aqueous solubility measurements are performed using the dynamic saturation method in a temperature range of [298.15 – 328.15K], those of octanol-water partition coefficient with the “shake-flask” method at 298.15K. The vapor pressure measurements are carried out with the “dynamic saturation method” in a temperature range between 313.15 and 423.15K, and validated with the static method. These measurements are used in the determination of air-water and octnol-air partition coefficients. Beside experimental measurements, these poperties are predicted using thermodynamic models: UNIFAC originale, UNIQUAC, NRTL and COSMO-sac-dsp. A qualitative and quantitative evaluation is performed for each model. This evaluation will facilitate the optimization of the models’ parameters concerning phthalates and alkyl phenols in order to generate a model for the contaminants migration process

Nanosized delivery systems are more efficiently accumulated at the inflammatory site, targeting specifically macrophages to resolve inflammation locally and reduce systemic adverse effects. The aim of this research was to prepare pharmaceutical formulations based on polymeric nanoparticles. Three types of poly(lactide-co-glycolide) - two linear and one branched polymer - together with the acidoresistant polymer cellulose-acetate phthalate (CAF) in various ratios were used to prepare nanoparticles by nanoprecipitation method. Rhodamine B was used as model active substance. The effect of acidoresistant component content on size and zeta potential of the nanoparticles was evaluated. Furthermore, dissolution tests were performed at both acidic and physiological pH. It was found that CAF doesn't have any significant effect on size of the particles and their stability....

Baseado nesses fatos, o objetivo deste trabalho foi o monitoramento do trânsito gastrintestinal (TG) do sistema proposto até sua chegada ao cólon, através de uma técnica não invasiva, livre de radiação, a Biosusceptometria de Corrente Alternada (BAC), que pudesse avaliar o trânsito gastrintestinal (TG) do SMM em voluntários saudáveis em jejum e no estado alimentado. Foram também realizadas análises do perfil de liberação in vivo da TC. Os resultados obtidos mostraram que o sistema desenvolvido foi capaz de produzir campo magnético de intensidade suficiente para ser detectado e que a BAC foi capaz de monitorar o trânsito gastrintestinal do SMM até sua chegada ao cólon. O estado pós-prandial influenciou significativamente o esvaziamento gástrico, sendo o tempo médio de 107,5 min ± 43,37 min no estado de jejum e de 260min ± 67,64 min no estado alimentado. Além do esvaziamento gástrico mais demorado no estado alimentado, o trânsito pelo intestino delgado e a chegada ao cólon também foram influenciados pelo estado pós-prandial. Analyses of the in vivo TC release profile were also carried out. The results showed that the developed system was able to generate magnetic field strong enough to be detected and that ACB was efficient to monitoring the MMS transit until its arrival in the colon. The postprandial state influenced significantly the gastric emptying, being 260.0 min ± 67.64 min the mean time for the fed state and 107.5 min ± 43.37 min for the fasting state. Additionally to a longer gastric emptying time, the transit within the small intestine and the arrival in the colon were also influenced by the postprandial state. Cellulose acetate / plasticizer systems : structure, morphology and dynamics / Systèmes d'acétate de cellulose plastifiés : structure, morphologie et dynamique

Les polysaccharides sont l'une des principales options à retenir pour progresser dans l'utilisation ou la conception de polymères renouvelables. Depuis les années cinquante, le développement industriel de ce type de polymères s'était considérablement réduit du fait de l'avènement des polymères synthétiques. Cependant, cet intérêt a cru considérablement ces dernières années en raison de la sensibilisation du public sur la limite des ressources fossiles. Ces biopolymères sont donc devenus un sujet d'importance, tant sur le plan industriel que sur celui de la recherche fondamentale. Toutefois, les systèmes à base de polysaccharides sont le plus souvent transformés via l'utilisation d'importantes quantités de solvants (y compris l'eau), ce qui globalement pénalise le procédé associé en l'affligeant d'une charge environnementale supplémentaire. Par la voie ‘fondue', le développement de polymères thermoplastiques à base de dérivés de la cellulose est un véritable défi, qui concerne autant le mode de transformation de ces systèmes que le niveau des propriétés du matériau final. Pour exemple, la température de dégradation de l'acétate de cellulose (CA) (dont le degré de substitution 2,5 est développé par le Groupe Solvay) est si proche de sa température de fusion que son procédé de mise en oeuvre ne peut être envisagé qu'avec l'ajout d'une quantité importante de plastifiants externes (entre 20 et 30 en poids selon le type d'additif). Le comportement d'un mélange CA-plastifiant est principalement régi par un «réseau» de très fortes interactions polaires, dont la force et la densité dépendent de 3 paramètres spécifiques: le degré de substitution de CA, la typologie de plastifiant et la quantité de plastifiant. Pour expliquer les différents mécanismes de plastification, il est donc important pour nous d'étudier et de comprendre les propriétés dynamiques (en ce qui concerne les phénomènes de relaxation) de ce type de systèmes et comment les trois leviers que nous avons identifiés peuvent influencer ou moduler les différentes interactions échangées dans les mélanges / Polysaccharides are one of the main options to the on-going move towards the use of renewable polymers. The industrial interest in this type of polymers drastically shrunk by the advent of synthetic polymers in the fifties, but is currently reviving due to the public awareness on the limit of fossil resources. These biopolymers are nowadays offering a challenging and industrially profitable playground for researchers. However, current polysaccharides based materials are mostly processed with extensive use of solvents (including water) making the total process an environmental burden despite the advantage of the starting material. Development of thermoplastic cellulose-based materials is very challenging regarding both final material properties and polymer processing. The degradation temperature of Cellulose Acetate (CA) (degree of substitution 2.5) is so close to its melting temperature that it can only be processed with a significant amount of external plasticizers (between 20 et 30 wt.% depending on the type of the additive). Behavior of a CA-plasticizer blend is mainly governed by a ‘network’ of high polar interactions, the strength and the density of which clearly depend of 3 specific parameters: the CA’s degree of substitution, the typology of the plasticizer, the amount of plasticizer. In an attempt to explain the different plasticization mechanisms, it is thus of utmost importance for us to study and understand the dynamic properties (regarding the relaxation phenomena) of this kind of systems and how the three levers that we identified can influence or modulate the different interactions exchanged within the blends

Il est maintenant établi que les facteurs alimentaires participent à la survenue des maladies inflammatoires chroniques de l’intestin (MICI). Le traitement thermique des aliments génère des substances néoformées telles que les produits de Maillard (MRPs) dont les effets sur la santé sont controversés. D’autres contaminants retrouvés dans les aliments sont issus des emballages. L’ensemble de ces molécules ont été associées à des réactions inflammatoires mais peu de données existent sur leurs conséquences sur la sphère digestive. L’objectif de cette thèse était d’étudier l’effet du traitement thermique des aliments sur la modulation d’une colite chez la souris. Dans un premier temps, nous avons dosé quelques marqueurs des MRPs dans deux régimes : peu et fortement chauffés. Ensuite, nous avons évalué l’impact de ces matrices sur la réponse inflammatoire chez des souris soumises à deux modèles de colites (DSS, TNBS). Enfin, nous avons observé les conséquences de l’effet cumulé de l’ingestion de MRPs et d’un contaminant d’emballage (le phtalate DEHP) chez les animaux atteints d’une colite. Le niveau de traitement thermique de la matrice était proportionnel aux teneurs en produits avancés et terminaux de la réaction de Maillard. Un fort traitement thermique a permis de prévenir la réponse inflammatoire chez les animaux soumis aux colites mais nous n’avons observé aucune modification de la réponse inflammatoire colique suite à l’ingestion couplée de MRPs et de DEHP. A terme, ces résultats descriptifs pourraient, sous réserve d’en identifier les mécanismes, servir à l’élaboration de recommandations nutritionnelles pour les patients souffrant de MICI. / It is acknowledged that alimentary factors participate in the etiology of inflammatory bowel diseases (IBD). On the other hand, food heat treatment generates neoformed compounds such as Maillard Reaction Products (MRPs) with controversial effects on human health but also results in contamination coming from packaging. All of these compounds are incriminated in inflammatory reactions but there are few data related to their consequences on gut. This work was aimed at determining the effect of food heat treatment on the modulation of an experimental colitis in mice. At first, we assessed some markers of MRPs in two moderately and highly heated diets. We then evaluated the consequences of these diets on the inflammatory response of animals submitted to two models of experimental colitis (DSS, TNBS). At last, we observed the consequences of cumulative effects of oral repeated exposure to MRPs and to a packaging contaminant the phthalate DEHP in DSS colitic mice. We revealed that the thermic level treatment was proportional to level of advanced and terminal MRPs and that the highest heat treatment resulted in the prevention of both the colitis models. By contrast, we observed no change in the inflammatory response following the coupled exposure to DEHP and the highly treated diet. While being descriptive, those results are promising and, after understanding the mechanisms involved, could be of interest in the conception of nutritional recommendations for IBD patients.

Mécanismes d’action des perturbateurs endocriniens bisphénol A et phtalates sur le développement du testicule fœtal / Mechanisms of action of endocrine disruptors bisphenol A and phthalates on the fetal testis development

Depuis plusieurs années, un nombre conséquent d’études décrivent une augmentation de l’incidence de pathologies liées à la fonction de reproduction masculine. Ces anomalies ont été regroupées sous le terme de « syndrome de dysgénésie testiculaire ». Ce syndrome aurait pour origine les effets délétères de polluants environnementaux sur le développement du testicule en période fœtale. Parmi ces polluants environnementaux, les phtalates et le bisphénol A (BPA) sont les plastifiants les plus produits et les plus répandus dans les objets de consommation courante. De nombreuses études leur sont consacrées et ont permis de les classer au rang de perturbateurs endocriniens en mettant notamment en cause leurs effets reprotoxiques. Mon travail de thèse est une étude des effets de ces deux perturbateurs endocriniens sur le développement du testicule fœtal.Nous avons réalisé une première étude concernant les effets du BPA sur le développement du testicule fœtal. Grâce au modèle de culture organotypique, nous avons développé notre étude dans trois espèces : le rat, la souris et l’Homme. Nous démontrons que le BPA diminue la sécrétion de testostérone dans le testicule fœtal humain à partir d’une concentration de 10-8M, alors que chez le rat et la souris, la sécrétion de testostérone n’est affectée qu’à partir de 10-5M de BPA. Nous avons également démontré une diminution de l’expression du gène de l’Insl-3, dans ces mêmes conditions. Ceci nous a permis de mettre en évidence une différence de sensibilité entre les espèces. Pour tenter de comprendre les mécanismes par lesquels le BPA exerce son effet toxique, nous avons comparé ses effets à ceux du DES, autre perturbateur endocrinien œstrogénomimétique. Contrairement au BPA, le DES diminue la sécrétion de testostérone fœtale chez les rongeurs, et non chez l’Homme. Ce résultat suggère l’implication de deux voies de signalisation différentes pour ces deux xéno-œstrogènes. Cette hypothèse est d’ailleurs renforcée par l’étude que nous avons SourceMécanismes d’action des perturbateurs endocriniens bisphénol A et phtalates sur le développement du testicule fœtal / par Thierry N’Tumba-Byn ; sous la direction de Virginie Rouiller-Fabre, Université Paris Sud, 2013 [Thèse de Biologie de la Reproduction et du Développement]réalisée sur des souris invalidées pour le récepteur des œstrogènes ERα, dans lesquelles l’effet anti-androgénique du BPA persiste, contrairement à celui du DES.Parallèlement, nous avons recherché les mécanismes d’action des phtalates et de leur métabolite actif le plus répandu, le MEHP (mono-2-éthyl-hexyl phtalate). Dans la continuité de plusieurs travaux réalisés dans notre laboratoire sur les effets du MEHP, nous avons tenté de comprendre les mécanismes par lesquels le MEHP induit un effet pro-apoptotique dans les cellules germinales mâles. Nous avons mis en évidence une augmentation de l’expression du gène Stra8 dans les cellules germinales traitées au MEHP. Ce résultat nous suggère que le MEHP pourrait induire une différenciation erronée des cellules germinales mâles. De plus, nous avons recherché les récepteurs et la voie de signalisation activée par le MEHP. Nous observons que les agonistes des récepteurs PPARα et de PPARγ entrainent dans les cellules germinales les mêmes phénotypes que le MEHP, à savoir une augmentation du taux d’apoptose et de l’expression du gène Stra8. / For several years, an increase in the incidence of pathologies connected to the male reproductive functions has been described in numerous studies. These anomalies are classified under the term “testicular dysgenesis syndrome”. This syndrome might find its origins in the deleterious effects of environmental pollutants on the testis development in fetal period. Among theses environmental pollutants, phthalates and bisphenol A (BPA) are the most produced plasticizers found in products of common use. Many studies were performed in order to determine their effects, and allowed to classify them as endocrine disruptors because of their reprotoxic effects. My thesis work is a study of the effects of these two endocrine disruptors on the fetal testis development.Our first study focuses on the effects of BPA on the fetal testis development. Using the organotypic culture model, we developed our study in three species: rat, mouse and human. We demonstrated that BPA decreases the testosterone secretion in the human fetal testis from a 10-8M concentration, while in rat and mouse the testosterone secretion is only affected by 10-5M BPA. We also demonstrated a decreased Insl-3 gene expression, in the same conditions. These results allowed us to evidence a difference of sensibility between species. To understand the mechanisms involved in the BPA toxic effect, we compared it with the effect of DES, another endocrine disruptor with estrogenic activity. Unlike BPA, DES decreases the fetal testosterone secretion in rodents and not in human. This result suggests the involvement of two different signalisation pathways for these two xenoestrogens. This hypothesis is reinforced by the study that we performed in mice invalidated for the estrogen receptor ERα. In those mice, the anti-androgenic effect of BPA is maintained, unlike DES effect.In parallel, we investigated the mechanisms of action of phtalates and particularly of their most prevalent active metabolite, the MEHP (mono-2-ethyl-hexyl phthalate). Following previous studies performed in our laboratory concerning the effects of MEHP, we intended to understand the mechanisms by which MEHP induces the apoptosis in male germ cells. We evidenced an increase in Stra8 gene expression in MEHP treated germ cells. This result suggests that MEHP might induce a wrong differentiation in male germ cells. Furthermore, we investigated the receptors and the signalisation pathway activated by MEHP. We observe that PPARα and PPARγ receptors agonists induce the same phenotypes as MEHP, namely an increase in the apoptosis and in Stra8 gene expression in germ cells.

Nos travaux in vitro ont permis de faire le lien entre une exposition à des micropolluants (le mono(2-ethylhexyl) phtalate, un perturbateur endocrinien, et le benzo(a)pyrene, un carcinogène) et ces observations cliniques. Les biomarqueurs de sécrétion hormonale (hPL et hCG hyperglycosylée) et de dégénérescence (activation du purinorécepteur P2X7) que nous avons identifiés permettent de détecter l’exposition et le risque suite à une exposition à des polluants. / Prenatal exposure to pollutants is commonly evaluated using placenta as a barrier between mother and fetus. Here, we consider placenta as a target organ for toxic agents. To achieve this, we selected a trophoblastic cell model, which is adapted to toxicological studies. In clinical studies, pregnancy pathologies are associated to changes in human placental lactogen (hPL) and human chorionic gonadotropin (hCG) secretions. Our in vitro work links exposure to micropollutants (mono(2-ethylhexyl)phthalate, an endocrine disruptor, and benzo(a)pyrene, a carcinogen) and clinical observations.
