A Araucaria angustifolia é uma espécie vegetal de vital importância para a manutenção da Floresta Ombrófila Mista (Floresta de Araucária), ecossistema no qual ocorre naturalmente. A araucária apresenta elevado valor sócio-econômico e ambiental e, devido à intensa exploração predatória a que foi submetida, hoje se encontra criticamente ameaçada de extinção. A destruição dessa preciosa espécie vegetal ocasionaria graves consequências ao ecossistema, que poderiam envolver a redução da diversidade de inúmeros animais, plantas e micro-organismos, os quais estão intimamente associados e apresentam grande dependência de A. angustifolia. Pesquisas envolvendo particularmente a caracterização de rizobactérias promotoras do crescimento de plantas (RPCP), nunca foram realizadas em A. angustifolia. Assim, o objetivo do presente estudo foi realizar o isolamento, a seleção e a caracterização de rizobactérias com potencial de promoção do crescimento e biocontrole de fungos fitopatogênicos em A. angustifolia. Os isolados obtidos foram selecionados quanto à capacidade para a promoção do crescimento por meio da produção de ácido indolacético (AIA), solubilização de fosfato inorgânico, produção de fosfatases, fixação assimbiótica de N2, este último através da análise de acúmulo total de nitrogênio em meio de cultura e análise de redução do acetileno (ARA). Também foram selecionadas rizobactérias através da avaliação de mecanismos indiretos de ação, como produção de sideróforos e antagonismo a Fusarium oxysporum, Cylindrocladium candelabrum e C. pteridis, fitopatógenos de espécies arbóreas. Os isolados mais promissores foram caracterizados através da análise do perfil de ácidos graxos da membrana celular (FAME), BOX-PCR e sequenciamento parcial do gene 16S rRNA. Foram isoladas 97 rizobactérias, sendo submetidas aos testes fenotípicos. Todos os isolados apresentaram ao menos uma característica positiva para os mecanismos de promoção do crescimento avaliados, excetuando-se a produção de quitinases. Dentre os isolados obtidos, 18 produziram AIA, 27 foram capazes de solubilizar fosfato inorgânico, 37 foram positivos para sideróforos e 83 foram hábeis produtores de fosfatases. Com relação à fixação assimbiótica de N2, 20 isolados foram positivos para a formação de película em meio de cultura livre de nitrogênio, no entanto, nenhum diferiu significativamente do controle, sem nitrogênio, quando analisado o acúmulo deste elemento em meio de cultura. Por outro lado, 3 isolados foram capazes de reduzir acetileno a etileno. Quarenta e cinco isolados formadores de endósporos apresentaram antagonismo a F. oxysporum. Os isolados B4, B15, B27, B35, B36, B42 e RISP2, caracterizados como Bacillus spp., destacaram-se como bons antagonistas aos fitopatógenos avaliados. Também foi verificada a presença de estirpes com múltiplos mecanismos de ação. A análise genotípica dos isolados mais promissores através da técnica de BOX-PCR apresentou a formação de dois grandes grupos, expressando nitidamente o agrupamento dos isolados formadores de endósporo. A técnica FAME e o sequenciamento do gene 16s rRNA concordaram em caracterizar os melhores isolados como pertencentes às famílias Bacillaceae (9 isolados), Enterobacteriaceae (11) e Pseudomonadaceae (1). Até onde sabemos, este é o primeiro estudo a relacionar a espécie Ewingella americana a diversos mecanismos de promoção do crescimento. Osresultados obtidos demonstram grande potencial de aplicabilidade dessas rizobactérias como biofertilizantes e no controle biológico de fitopatógenos em A. angustifolia. / Araucaria angustifolia is a plant species of vital importance for the maintenance of the Mixed Ombrophylous Forest (Araucaria Forest) ecosystem in which it occurs naturally. Araucaria has a high socio-economic and environmental value, and due to its intense predatory exploitation, it is critically endangered. The destruction of this precious plant species would cause serious consequences to the ecosystem, which could involve a reduction of the diversity of many animals, plants and microorganisms, which are closely associated and reveal a great dependence on A. angustifolia. Studies involving plant growth promoting rhizobacteria (PGPR) and their characterization have never before been performed in A. angustifolia. The objective of this study was to isolate, select and characterize rhizobacteria with potential for growth promotion and biocontrol of plant pathogenic fungi in A. angustifolia. The isolates were selected by means of their potential to produce indole acetic acid (IAA), to solubilize inorganic phosphate, to produce phosphatase, and for asymbiotic N2 fixation. The latter was estimated by analyzing the accumulation of nitrogen in the culture medium and by the acetylene reduction assay (ARA). Rhizobacteria were also selected through the evaluation of indirect action mechanisms such as siderophore production and antagonism to Fusarium oxysporum, Cylindrocladium candelabrum and C. pteridis, all pathogens of trees. The most promising isolates were characterized by analyzing the fatty acid methyl ester (FAME), by BOX-PCR and partial sequencing of the 16S rRNA gene. Ninety seven rhizobacteria were isolated and subjected to the phenotypic tests. All isolates presented at least one positive feature, characterizing them as potential PGPR, except for the production of chitinases. Among these isolates, 18 produced IAA, 27 were able to solubilize inorganic phosphate, 37 were positive for siderophores and 83 were phosphatases producers. Regarding the asymbiotic N2 fixation, 20 isolates were effective in the pellicle formation in nitrogen free culture medium, however, none differed significantly from the control, when analyzed for the accumulation of N in the culture medium. Furthermore, three isolates were able to reduce acetylene to ethylene. Forty five isolates were endospore formers and antagonistic to F. oxysporum. The isolates B4, B15, B27, B35, B36, B42 and RISP2, characterized as Bacillus spp. stood out as good antagonists of the evaluated plant pathogens. We also observed the presence of strains with multiple mechanisms of action. Genotypic analysis of the most promising isolates by the BOX-PCR technique showed the formation of two major groups, expressing clearly the grouping of isolates forming endospores. Both techniques, FAME and PCR, resulted in the same grouping of the most effective isolates as belonging to Bacillaceae (9 isolates), Enterobacteriaceae (11) and Pseudomonadaceae (1). As far as we know, this is the first study to include the bacterium Ewingella americana among the PGPR. The results demonstrate a potential application of these rhizobacteria as biofertilizers and for biological control of plant pathogens in A. angustifolia.

Duração do período de molhamento em vinhedo de \'Niagara Rosada\' e sua relação com a ocorrência de míldio (Plasmopara viticola) / Leaf wetness duration in a \'Niagara Rosada\' vineyard and its relationships with downy mildew (Plasmopara viticola) occurrence

Atualmente, a videira Niagara Rosada é a variedade de uva de mesa mais cultivada no Estado de São Paulo, Brasil. Contudo, sua produtividade e custo de produção têm sido afetados pela ocorrência de doenças fúngicas, principalmente o míldio, causado pelo fungo Plasmopara viticola. Para o controle dessa doença, têm sido feitas pulverizações com fungicidas, as quais muitas vezes em excesso. Dentre as variáveis meteorológicas que influenciam a ocorrência de doenças fúngicas nas plantas, a duração do período de molhamento (DPM) é uma das mais importantes. A presença de molhamento sobre a superfície das plantas provê a água requerida pelos patógenos para o processo de germinação e infecção do tecido foliar. Visando a subsidiar sistemas agrometeorológicos de alerta fitossanitário, cuja finalidade é possibilitar a racionalização do uso de fungicidas, os objetivos do presente estudo foram: determinar a posição da videira com a DPM mais longa e sua relação com a DPM medida na posição padrão, a 30 cm de altura sobre o gramado; avaliar a estimativa da DPM sobre o gramado por diferentes modelos a partir de dados meteorológicos obtidos em uma estação meteorológica padrão e verificar as relações entre a DPM estimada para o gramado e a DPM medida no vinhedo; e correlacionar a epidemiologia do míldio (P. viticola) durante o ciclo da videira Niagara Rosada, sem controle químico, com as variáveis DPM medida no vinhedo (posição da videira com a DPM mais longa), DPM estimada na posição padrão sobre o gramado pelo melhor modelo, assim como com outras variáveis meteorológicas. A DPM foi medida em posição padrão sobre o gramado e em quatro diferentes posições da videira: topo da planta com a face superior do sensor voltada para sudoeste e nordeste (Topo-SW e Topo-NE) e altura dos cachos de uva com a face superior do sensor voltada para sudoeste e nordeste (Dossel-SW e Dossel-NE). A epidemiologia do míldio foi avaliada sem controle químico, utilizando-se escala de notas de 0 a 4, com nove níveis de severidade, para as folhas e cachos da videira. No estudo da variabilidade espacial da DPM, não houve diferença significativa entre a parte mais alta (1,6 m) e a parte mais baixa (1,0 m) da videira, assim como entre as faces sudoeste e nordeste das plantas. Ao se analisar a relação entre a DPM sobre o gramado e a DPM nas diferentes posições da videira obtida por meio de regressão linear simples, observou-se uma boa correlação, com R2 = 0,88. Na avaliação dos modelos de estimativa da DPM, o modelo CART foi o que teve o melhor desempenho sobre o gramado, sendo que esta estimativa também apresentou uma boa correlação com a DPM medida no interior do vinhedo. Isso permitiu concluir que é possível estimar a DPM no vinhedo de Niagara Rosada a partir de medidas ou estimativas da DPM na estação meteorológica padrão. Na modelagem do desenvolvimento do míldio, a DPM esteve presente nas melhores correlações com a severidade do míldio na videira Niagara Rosada, mostrando a grande importância desta variável para a ocorrência de doenças fúngicas nos vinhedos. / Nowadays, the \'Niagara Rosada\' grapevine is the most cultivated table grape variety in the State of São Paulo, Brazil. However, yield and production cost of this grapevine have been affected by fungal diseases, mainly downy mildew, caused by Plasmopara viticola fungus. For controlling this disease, producers have been applied an excessive number of sprays with fungicides. Among the meteorological variables that influence the occurrence of fungal plant diseases, leaf wetness duration (LWD) is one of the most important. The wetness presence on plant surface provides the water required by the phatogens to germinate and to infect leaf tissues. Aiming to subsidize the plant disease warning systems, which has as purpose to rationalize the use of fungicides in the vineyards, the objectives of the present study were: to determine the canopy position of the Niagara Rosada table grape with longer LWD and its correlation with measured standard LWD over turfgrass; to estimate LWD over turfgrass considering different models with data from a standard weather station, and to evaluate the correlation between estimated LWD over turfgrass and LWD measured in the vineyard; and to correlate downy mildew occurrence in the vineyard, without chemical control, with measured LWD at the vineyard (canopy position with longer LWD), with estimated LWD in standard condition over turfgrass for the best model, and with other meteorological variables. LWD was measured in standard condition over turfgrass and in four different canopy positions of the vineyard: at the top of the plants, with sensors facing southwest and northeast (Top-SW and Top-NE), and at the grape bunches height, with sensors facing southwest and northeast (Bottom-SW and Bottom-NE). The downy mildew epidemiology during the grapevine cycle was evaluated without chemical control, using scores ranging from 0 to 4, with nine severity levels, for leaves and bunches of grapevine. When the spatial variability of LWD was studied, no significant difference was observed between the top (1.6 m) and the bottom (1.0 m) of the canopy and also between the southwest and northeast face of the plants. The analysis of the relationship between standard LWD over turfgrass and crop LWD in different positions of the grape canopy showed a define correlation (R2 = 0.88). Among the LWD estimative methods, CART was the one with the best performance to estimate LWD over turfgrass. The results from this model also presented a good correlation with measured LWD inside the vineyard, showing that LWD can be estimated for this crop with data from a nearby standard weather station. For the downy mildew modeling, LWD also present the best correlations with disease severity in the \'Niagara Rosada\' vineyard, showing the great importance of this variable for fungal diseases occurrence in this crop.

Development of the polygalacturonase inhibiting protein (PGIP) for delivery of foreign proteins to the surfaces of plant cells

Polygalacturonase-inhibiting proteins (PGIPs) are cell wall-associated plant proteins that inhibit endopolygalacturonases (endo-PGs) from phytopathogenic fungi. For proteins to confer resistance to invading plant pathogens, it is preferred that they are either associated with the plant cell wall or secreted into the intercellular spaces where they can act almost immediately upon pathogen attack. The bactericidal efficacy of the Hen Egg White Lysozyme (HEWL) has previously been unequivocally demonstrated in transgenic plants; however, most of the protein remains intracellular. It was hypothesized that bean PGIP1, that has previously been expressed correctly in transgenic tomato plants and was found to inhibit the endopolygalacturonase activity of Stenocarpella maydis in a reducing sugar assay, would deliver the HEWL protein to the intercellular spaces due to its inherent translocation to the plant cell wall by means of a translational fusion between bean pgip1 and hewl genes. In this study, the efficacy of such a translational fusion was determined. The bean pgip1-hewl fusion was inserted into the binary vector pCAMBIA2300 and transformed into Nicotiana tabacum cv. LA Burley 21 plants by Agrobacterium-mediated transformation. Phenotypically normal transgenic plants were produced. Stable transgene insertion into the transgenic N. tabacum genomes was verified by PCR and Southern blot analyses. To demonstrate the efficacy of the bean PGIP1-HEWL fusion, independent homogenate and intercellular fluid protein extracts were prepared from transgenic N. tabacum leaf material. Protein extracts prepared so as to enrich for PGIP activity were tested in vitro for inhibition of S. maydis endo-PGs whereas protein extracts for HEWL activity were tested for lysis of Micrococcus luteus cells. Biochemical assays showed that bean PGIP1-HEWL inhibited S. maydis endo-PGs and cleaved M. luteus cell walls sufficiently to suggest that the PGIP1- HEWL fusion was structurally and functionally stable. Total protein extracts from the PGIP-HEWL and HEWL transgenic plants showed similar levels of HEWL specific activity, whereas intercellular fluid samples from PGIP-HEWL transgenic plants showed high activity in contrast to HEWL plants. With the success of showing protein activity in vitro of HEWL in intercellular spaces, bean PGIP1 can be recommended as a vehicle for delivery of other proteins to cell surfaces. Orientador: Edson Luiz Furtado / Resumo: A seringueira (Hevea brasiliensis) é a terceira cultura florestal mais plantada no Brasil, e o estado de São Paulo é responsável por mais de 60% da produção de látex do país. É uma espécie nativa da região amazônica que possui hábito caducifólio, e durante o período de reenfolha é suscetível ao ataque de fitopatógenos foliares, que prejudicam e até limitam seu estabelecimento em determinadas regiões. Dentre os principais patógenos foliares da seringueira, destacam-se os fungos causadores de oídios, microrganismos biotróficos que infectam tecidos jovens, e podem causar desfolha, principalmente em condições de viveiro e jardim clonal. O agente causal do oídio da seringueira é descrito como Erysiphe quercicola, porém, devido a insuficientes descrições taxonômicas, morfológicas e moleculares desta doença, seu agente causal permanece em discussão, além da fase sexual deste patógeno carecer de qualquer registro neste hospedeiro. Portanto, o objetivo deste trabalho é caracterizar o oídio da seringueira, utilizando caracterizações morfológicas, análises moleculares e testes de patogenicidade. No período de julho a dezembro de 2019, foram coletadas amostras foliares de seringueira em diferentes estágios fenológicos infectadas com oídio na região de Botucatu, São Paulo. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Rubber tree (Hevea brasiliensis) is the third most planted forest crop in Brazil, and the state of São Paulo is responsible for more than 60% of the country's latex production. It is a species native to the Amazon region that has a deciduous habit, and during the re-leafing period it is susceptible to the attack of leaf phytopathogens, which harm and even limit its establishment in certain regions. Among the main leaf pathogens of the rubber tree, there are the fungi that cause powdery mildew, biotrophic microorganisms that infect young tissues, and can cause defoliation, especially in nursery and clonal garden conditions. The causal agent of rubber mildew is described as Erysiphe quercicola, however, due to insufficient taxonomic, morphological and molecular descriptions of this disease, its causal agent remains under discussion, in addition to the sexual phase of this pathogen lacking any record in this host. Therefore, the objective of this work is to characterize powdery mildew mildew, using morphological characterizations, molecular analyzes and pathogenicity tests. From July to December 2019, leaf samples of rubber trees were collected at different phenological stages infected with powdery mildew in the Botucatu region, São Paulo. Producción y utilización Biotecnológica de nuevas proteínas antifúngicas de hongos filamentosos

Tesis por compendio / [ES] Los péptidos antimicrobianos (AMP) son una alternativa prometedora para el desarrollo de nuevos antifúngicos que puedan sustituir a los fungicidas usados en agricultura. Sin embargo, el alto coste de la síntesis química y la dificultad para su producción a gran escala han limitado su aplicación. Las proteínas antifúngicas (AFP) son AMP naturales, pequeñas, catiónicas, secretadas y ricas en cisteína con gran potencial para el control de hongos fitopatógenos. Las AFPs se encuentran en hongos filamentosos, son estables y pueden producirse en grandes cantidades. Sin embargo, el papel biológico en su hongo productor no se conoce en profundidad. En esta tesis, se estudió la diversidad de AFPs en genomas de hongos ascomicetos y se propuso una nueva clasificación en tres clases (A, B y C). Penicillium digitatum es el principal patógeno postcosecha de cítricos y codifica solo una AFP en su genoma de clase B (AfpB), mientras que Penicillium expansum, el principal patógeno postcosecha de manzana, codifica una AFP de cada clase (AfpA, AfpB y AfpC). En este trabajo describimos la producción biotecnológica y la caracterización de estas cuatro AFPs. Se ha caracterizado el papel biológico del gen afpB en P. digitatum mediante estudios de expresión génica y generación de mutantes nulos y de expresión constitutiva. Los resultados indicaron que afpB es prescindible para la biología y el ciclo vital del hongo, aunque la expresión del gen afpB bajo el promotor constitutivo gpdA de Aspergillus nidulans es perjudicial para su crecimiento y virulencia. Sorprendentemente, ni la cepa parental ni las cepas constitutivas produjeron cantidades detectables de AfpB a pesar de la alta expresión del gen codificante. El modelado molecular y el diseño racional permitieron predecir la estructura terciaria de AfpB y diseñar péptidos sintéticos para mapear motivos antifúngicos en su secuencia primaria. Confirmamos que los bucles catiónicos L2 y L3 mostraron actividad antifúngica moderada y que pueden actuar sinergísticamente. Con el objetivo de producir AfpB mediante biotecnología, usamos un casete de expresión de AFPs basado en las regiones promotora y terminadora del gen paf de Penicillium chrysogenum, hongo que produce naturalmente grandes cantidades de su propia proteína PAF. Este casete funcionó en P. digitatum y permitió la producción homóloga de AfpB. Los datos también mostraron que las secuencias del péptido señal (SP) y el pro-péptido de la SP-Pro-AfpB no determinan la producción de proteína. También demostramos la estabilidad térmica y la resistencia a la proteólisis de AfpB, y aportamos datos que sugieren que la estructura terciaria no es necesaria para la actividad antifúngica. Similar a lo descrito en P. digitatum, ninguna de las tres AFPs se detectó en los sobrenadantes de cultivo en medio rico de P. expansum. Sin embargo, AfpA se produjo en grandes cantidades en cultivos de medio mínimo de P. expansum. Para completar el repertorio de AFPs, produjimos las tres AFPs de P. expansum (AfpA, AfpB y AfpC) en P. chrysogenum con el casete paf. Las tres proteínas de P. expansum se produjeron, purificaron y caracterizaron con éxito. Ninguna de las AFPs producidas en este trabajo fue citotóxica frente a eritrocitos de mamíferos. AfpA de P. expansum seguida de AfpB de P. digitatum fueron las AFPs más activas contra hongos filamentosos, incluyendo patógenos de plantas y humanos, productores de micotoxinas y sus propios hongos productores, una característica previamente no descrita en las AFPs. Además, la AfpA de P. expansum y la AfpB de P. digitatum protegieron frente a la infección causada por el hongo Botrytis cinerea en plantas de tomate, y AfpA de P. expansum protegió frente a P. digitatum en frutos de naranja. Hem caracteritzat el paper biològic del gen afpB en P. digitatum mitjançant estudis d'expressió gènica i la generació de mutants nuls i d'expressió constitutiva. Els resultats van indicar que afpB és prescindible per a la biologia i el cicle de vida d'este fong, encara que l'expressió del gen afpB davall el promotor constitutiu gpdA d'Aspergillus nidulans és perjudicial per al seu creixement i virulència sobre fruits cítrics. Sorprenentment, ni el cep parental ni els ceps constitutius van produir quantitats detectables d'AfpB malgrat l'alta expressió del gen afpB. El modelatge molecular i el disseny racional van permetre predir l'estructura terciària d'AfpB i dissenyar pèptids sintètics per a identificar motius antifúngics dins de la seqüència primària. Confirmarem que les estructures catiòniques L2 i L3 mostren activitat antifúngica i que poden actuar de forma sinèrgica. Amb l'objectiu de la producció biotecnològica d'AfpB, utilitzarem un casset d'expressió d'AFPs basat en les regions promotora i terminadora del gen paf de Penicillium chrysogenum, el qual produïx naturalment grans quantitats de la seua pròpia proteïna PAF. Este casset va funcionar en P. digitatum i va permetre la producció homòloga d'AfpB. Les dades també van mostrar que les seqüències del pèptid señal (SP) i el propèptid de la SP-Pro-AfpB no determinaren la producció de proteïna. També demostrarem l'extrema estabilitat tèrmica i la resistència proteolítica d'AfpB, i proporcionem dades que suggerixen que l'estructura terciària no és necessària per a l'activitat antifúngica. Semblant a P. digitatum, cap de les tres AFPs es van detectar en els sobrenadants de medi de cultiu ric de P. expansum. Al contrari, AfpA es va produir en grans quantitats en cultius de P. expansum en medi mínim. Per a completar el repertori d'AFPs, vam produir les tres AFPs de P. expansum (AfpA, AfpB i AfpC) en P. chrysogenum mitjançant l'ús del casset paf. Així, les tres proteïnes de P. expansum es van produir, purificar i caracteritzar amb èxit. Cap de les AFPs produïdes en este treball va ser citotóxica front eritròcits de mamífer. AfpA de P. expansum seguida d'AfpB de P. digitatum van ser les AFPs més actives contra fongs filamentosos, incloent patògens de plantes i humans, productors de micotoxines i els seus propis productors, una característica prèviament no descrita per a les AFPs. A més, AfpA de P. expansum i AfpB de P. digitatum van protegir front la infecció causada pel fong Botrytis cinerea en plantes de tomaca, i l'AfpA de P. expansum va protegir front P. digitatum en fruits de taronja. Estos resultats confirmen la nostra hipòtesi anterior de que les AFPs són bones candidates per al desenvolupament d'antifúngics en protecció / [EN] Antimicrobial peptides (AMPs) are promising antifungal alternatives to the fungicides used in agriculture. However, the high cost of chemical synthesis and the difficulties of large-scale production have limited their application. Antifungal proteins (AFPs) are a group of natural, small, cationic, secreted, cysteine-rich AMPs that offer a great potential to develop new biomolecules for the control of phytopathogenic fungi. AFPs are naturally present in filamentous fungi, are very stable, and can be produced in large amounts. However, the biological role of these AFPs in their producer fungus is still unclear. In this thesis, we first studied the diversity of AFPs in ascomycetous genomes and proposed a new classification in three different classes (A, B and C). Penicillium digitatum is the main citrus postharvest pathogen and encodes only one AFP from class B in its genome (AfpB), while Penicillium expansum is the main pome postharvest pathogen and encodes one AFP from each class (AfpA, AfpB and AfpC). In this work, we report the identification, efficient biotechnological production and characterization of these four AFPs. We characterized the biological role of the afpB gene in P. digitatum by the study of its gene expression pattern and the generation of null and constitutive expression mutants. Results indicated that afpB is dispensable for the biology and life cycle of this fungus, although expression of the afpB gene under the constitutive Aspergillus nidulans gpdA promoter is detrimental to growth and virulence to citrus. Surprisingly, neither the wild type nor the constitutive strains produced detectable amounts of AfpB in spite of the high afpB gene expression. Molecular modeling and rational design allowed us to predict the AfpB tertiary structure and design synthetic peptides to map antifungal motifs within the AfpB primary sequence. We confirmed that the cationic exposed loops L2 and L3 showed moderate antifungal activity and that they can act synergistically. With the objective of the biotechnological production of AfpB, we used an AFP expression cassette based on the promoter and terminator regions of the well-studied paf gene from Penicillium chrysogenum, which naturally produces high amounts of its own protein PAF. This paf cassette worked efficiently in P. digitatum and allowed the homologous production of AfpB. Data also showed that the signal peptide (SP) and pro-peptide sequences of the translated SP-Pro-AfpB do not determine protein production. We also demonstrated the thermal stability and resistance to proteolytic cleavage of the P. digitatum AfpB, and provided data that suggest that tertiary structure is not required for antifungal activity. Similar to P. digitatum, none of the three AFPs were detected in supernatants of cultures of P. expansum in rich medium. By contrast, AfpA was produced with very high yields in P. expansum cultures in minimal medium. To complete the repertoire of AFPs from P. expansum we produced the three AFPs from P. expansum (AfpA, AfpB and AfpC) in P. chrysogenum with the use of the paf cassette. With this combined approach, the three P. expansum proteins were successfully produced, purified and characterized. None of the four AFPs produced in this work were cytotoxic against mammal erythrocytes. The P. expansum AfpA followed by the P. digitatum AfpB were the most active AFPs against filamentous fungi, including plant and human pathogens, mycotoxin-producer fungi, and their own producers, a feature that had not been previously described for AFPs. Moreover, AfpA from P. expansum and AfpB from P. digitatum protected against fungal infection caused by Botrytis cinerea in tomato plants, and additionally the P. expansum AfpA protected against P. digitatum in orange fruits. These results confirm our previous hypothesis that AFPs are good candidates for the development of antifungals in plant protection and postharvest conservation, but also in clinic or food preservation. / Garrigues Cubells, SM. Ultrastructural investigations of powdery mildew of rose caused by sphaerotheca pannosa (Wallr.) lev

The purpose of this study was to evaluate the ultrastructural aspects of the host-pathogen relationship between the fungus Sphaerotheca pannosa (Wallr.) Lev. and a susceptible horticultural variety of floribunda rose "Eutin". Attention was given to the penetration of hyphal tubes, and their subsequent development into: haustorial elements in epidermal cells of rose. In addition, changes in the morphology of infected epidermal cells were described. Thin sections of rose leaves infected with S. pannosa were examined in a Hitachi electron microscope, model HS-7, after glutaraldehyde-acrolein fixation, OsO4-uranyl acetate staining, and embedding in Epon plastic. The components of S. pannosa haustoria consisted of a haustorial body with several lobes. The body, and the lobes, were irregular in shape, and were characteristically surrounded by thin haustorial walls. Small amounts of sheath enclosed the haustorial walls, or was found lacking around some haustorial lobes. A slender neck attached the haustorium to the surface hyphal cell, and was considered an extension of that cell, and not the haustorium. At the base of the neck, a septum which contained a central pore, separated the haustorium from the surface hyphal cell, making the haustorium an independent cell. The haustorial protoplasts were similar to hyphal cells on the surface. They contained mitochondria, ribosomes, endoplasmic reticulum, lipid bodies, and vacuoles. Muclei were not detected. Penetration of the epidermal wall of rose by hyphal tubes was accompanied by cellular proliferations on the inner surfaces of the host wall. These proliferations showed layering, and some formed collars around the penetrating tubes. Although the patterns of the proliferations differed from the normal wall layers of the host, one of the layers of the collar exhibited strands of microfibrils. Some haustoria were completely surrounded by cellular proliferations which were similar in electron density to collar material. It was thought that these proliferations were extensions of the collar which formed in response to breakdown of parasitic control. It appeared that the plasma membrane of the host was the sheath membrane, and that it also lined the cavity of the collar.
