Este trabalho descreve o desenvolvimento de um algoritmo para avaliar o grau de corrosão em paredes de oleodutos, através de ultrassom. O algoritmo desenvolvido será implementado em um circuito digital, fazendo parte de um sistema que poderá ser embarcado em pigs ultrassônicos para inspeção de dutos à procura de corrosão. O modelo matemático conhecido como filtro casado é usado como base do algoritmo, além de um método de detecção de picos para localização dos sinais de eco de ultrassom, provenientes das reflexões do sinal nas paredes dos dutos. Um dos problemas é a definição do sinal (e do filtro) que será utilizado, mas por meio de sinais sintetizados (signal design) uma grande gama de alternativas foi analisada e apenas algumas foram selecionadas para testes em experimentos. Por último os melhores sinais foram utilizados na inspeção de um corpo de prova feito à partir de um duto corroído, e os resultados são analisados. A detecção da primeira parede do oleoduto apresenta resultados promissores em comparação com a medição tridimensional das mesmas paredes. O sistema foi capaz de reconstruir o perfil da parede interna do duto com elevada verossimilhança, mesmo em regiões severamente corroídas. A detecção da segunda parede apresenta resultados satisfatórios para baixos níveis de corrosão, porém alguns problemas foram encontrados para níveis mais altos de corrosão, prejudicando a medição direta da espessura da parede nestas condições. / This work describes the development of an algorithm to evaluate the degree of corrosion in pipeline walls, using ultrasound. The developed algorithm will be implemented in a dedicated digital circuit that can be embedded in ultrasonic pigs for pipeline corrosion inspection. The algorithm is based on the mathematic model known as matched filter, besides a peak detection method for the location of ultrasound echoes, originated from the signal reflections in the pipeline walls. One problem is the determination of the signal (and filter) that should be used, but using a signal design approach a large number of possibilities could be analyzed and only a few was tested in experiments. Finally, the best signals were used to inspect a test specimen made of a corroded pipeline section, and the results were analyzed. The first wall detection shows promising results when compared to the results of a three-dimensional inspection of the same walls. It was possible to reconstruct the profile of the internal wall with good likelihood, even at badly corroded areas. The second wall detection shows some satisfactory results on low corrosion levels, but show some problems with high corrosion levels, compromising the direct measurement of the wall of the pipeline.

Improving Pig Performance and Efficiency by Attenuating Transport and Immune Stress Responses through L-Tryptophan Supplementation

<p>  </p> <p>In commercial settings, piglet weaning and transportation occur concurrently due to the expansive application of multi-site production systems across the United States and the combination of these events can be defined as an early life stressor. Early life stress is known to reduce pig performance, efficiency, and immune resilience contributing to reduced welfare and increased production losses. To combat the deleterious effects of stress on pigs, the essential amino acid tryptophan (Trp) supplemented above current National Research Council (NRC 2012) recommended levels, improves neuroendocrine responses to stress, lowers plasma cortisol and norepinephrine concentrations, and improves hypothalamic-pituitary-adrenal axis recovery time following stress. However, there are discrepancies concerning the Trp requirement for nursery pigs, suggesting the 2012 recommendations for Trp may be inaccurate for optimizing growth performance and health in modern pigs. Three experiments were conducted to determine the effects of supplemented standard ileal digestible Trp above NRC (2012) recommended levels on performance, feed efficiency, immune vigor, and stress tolerance. The objective of experiment 1 was to eliminate or reduce short- and long-term, transport-induced reductions in piglet feed efficiency and growth by supplementing Trp above NRC (2012) recommendations pre-weaning and/or during the nursery phase. An oral gavage of Trp or a control milk carrier was provided to pre-allotted piglets beginning at day 5 of lactation and continuing to weaning. At weaning all pigs were blocked by sex, weaning weight and pre-wean treatment and randomly assigned to transport and post-wean treatments. Pigs were fed in four nursery phases with diets containing 1X or 2X NRC recommended concentrations of SID Trp and a common grower diet fed in 6 phases during the grow-finish period. At market, loin characteristics were measured via ultrasound and carcass data was collected from the packing plant. Pre-weaning Trp supplementation had no effect on pre-weaning growth performance; however, post-wean Trp increased overall body weight and average daily gain in nursery pigs when Trp was supplemented pre-wean. The objective for experiment 2 was to mitigate the adverse effects of early life transport stress on subsequent immune challenges by providing supplemental Trp during the nursery period. At weaning, pigs were transported for 8 hours and assigned to treatments of vaccine-induced immune challenge and dietary treatment. Pigs were fed standard nursery diets, in four phases, over 35 days with pigs receiving 1X or 2X the NRC (2012) recommended Trp concentration. Half the pigs on each dietary treatment were subjected to a 3-wk vaccine challenge consisting of circovirus, mycoplasma, and influenza vaccines administered in wk 2, 3, and 4 post-wean, respectively. At market, loin characteristics were measured via ultrasound and carcass data was collected.  At the culmination of the nursery period, unchallenged pigs supplemented with Trp were heavier compared to control pigs. This resulted from an overall improvement in average daily gain for 2X Trp fed pigs. However, increasing Trp did not affect market weight, loin eye area, or lean percent in market pigs, although 2X Trp increased back fat. The objective of experiment 3 was to determine what ratio of SID tryptophan, relative to lysine, maximizes growth performance and feed efficiency in weaned pigs during the nursery period. Pigs were blocked by sex and weaning weight and randomly allotted to one of five dietary treatments with Trp increasing stepwise by 0.33%-units, respectively. Pigs were fed standard nursery diets, in four phases, over 35 days with pigs receiving 1X, 1.33X, 1.66X, 2X, or 2.33X the NRC (2012) recommended Trp concentration.  Overall, there were no differences in performance or efficiency across dietary treatments during a 35-day nursery period in unstressed healthy pigs.  In conclusion, increasing Trp in swine diets prior to and/or following stress events can mitigate stress-related perturbations in performance and efficiency.      </p>

DEVELOPMENT OF A DOMESTIC ANIMAL MODEL OF ENDOMETRIOSIS

Endometriosis is a reproductive disease affecting women in their prime reproductive years characterized by the presence of endometrial glands and stroma in ectopic locations. Animal models have been proven to be indispensable not only for the study of the disease but also to develop better non-invasive diagnostic imaging modalities. A major limitation with the diagnosis of endometriosis is the lack of a specific and sensitive non-invasive diagnostic test. Our objective was to develop a domestic animal model of endometriosis suitable for serial diagnostic imaging procedures and assessment of therapies. Two major studies were conducted to achieve this objective. First study involved in vitro whole tissue-explant culture and surgical induction of endometriosis in dog, pig and sheep to choose the most suitable model. For in vitro co-culture, dog, pig and sheep endometrium was placed on visceral peritoneum for 24 to 72 h to assess the degree of attachment and adhesion characteristics of endometrium (epithelium, glandular and stromal cells). Surgical induction of endometriosis was tested in dog, sheep (n=5 each) and pig (n=4) using autologous endometrial (n=4 grafts per animal) and fat grafts sutured to visceral (urinary bladder surface in dog and pig, uterus in sheep) and parietal (abdominal wall) peritoneum. Sham surgeries were performed in control group animals (dog and sheep n=5, pig n=3) using fat grafts alone. Animals were euthanized between 80-110 days post-surgery. Size, gross characteristics and histopathologic features of endometriotic lesions were recorded. During in vitro explant culture, surface epithelial, stromal and glandular cells of endometrium were capable of attaching to visceral peritoneum within 24 hours with and without an intact layer of mesothelial lining in dog, pig and sheep. The proportion of successful endometrial attachments were greater at 24h compared to 72h (15/18 vs. 7/18, p=0.008; data combined among species) with intermediate attachment at 48h (12/15). Following surgical induction, there was no difference (p>0.05) in proportion of successful tissue grafts placed on serosal surface of visceral vs. parietal peritoneum in dog (10/10 vs. 10/10), pig (7/8 vs. 8/8) or sheep (7/10 vs. 8/10). A variety of outcomes (endometriotic cysts with sero-sangunous fluid, solid lesions, vesicles, absence of lesions) were found. The proportion of cystic lesions was greater (p<0.01) in dog (19/20 grafts) than in pig (8/16) and sheep (5/20). Further, the area of endometriotic lesions at euthanasia was larger (0.89 ± 0.11 cm2) compared to that at the time of surgery (0.50 ± 0.09 cm2) in dog, whereas, the size of lesions decreased by half or more (p<0.05) in pig and sheep. Combined among grafting sites (visceral and parietal peritoneum) and species, a greater proportion (p=0.015) of surgical sites had adhesions in treatment (12/14) versus control group animals (5/13). The wall of majority of endometrial cysts in dogs were characterized by simple cuboidal/columnar epithelium, endometrial glands (normal, dilated and cystic), subepithelial capillary network and presence of stromal and smooth muscle cells. Hemorrhage and/or hemosiderin-laden macrophages were observed in the cystic lesions in dog. Development of a greater proportion of growing lesions in the form of endometriotic cysts in dogs compared to sheep and pig led us to conclude dog as a better suitable domestic animal model for endometriosis than sheep and pig. Second study involved assessing the usefulness and limitations of ultrasonography, magnetic resonance imaging (MRI) and positron emission tomography-computed tomography (PET-CT) in detecting cystic endometriotic lesions in dog and sheep. Surgical induction of endometriois was performed in dogs (n=5) and sheep (n=5) using autologous endometrial grafts (n=4 grafts per animal) and fat grafts sutured to visceral peritoneum (urinary bladder in dogs, uterus in sheep) and parietal peritoneum (ventral abdominal wall). Weekly ultrasonography was performed from Week 1-9 post-surgery and day of euthanasia (Week 14-15). T1 and T2 weighted weighted MRI images (n=2 each for dog and sheep) and PET-CT (n=3 dog, n=1 sheep) using 18F-fluorodeoxyglucose (18F-FDG) as radiolabel was performed between Week 13-15 post-surgery in dog and sheep. Gray-scale B-mode ultrasonography was able to detect endometriotic cysts (0.25-1.75cm) on urinary bladder and abdominal wall in dogs; endometrial grafts Week 1 post-surgery appeared as a homogenous, hypoechoic masses, following which they grew larger with evidence of cyst formation by Week 5. Cysts were undetectable from Week 10-13, whereas they appeared as homogenous masses with a hypoechoic fluid-filled cavity with diffuse hyperechoic echoes and low vascularisation (Color-Doppler imaging) by Week 14-15. In sheep, endometrial grafts were detected as hypoechoic masses Week 1 post-surgery that became smaller until no detectable lesions were visible beyond Week 6-7. Cysts in dogs and sheep appeared hyperintense on T2 and hypointense on T1 weighted images. 18F -FDG PET-CT did not show hypermetabolic activity in endometriotic cysts in dogs and sheep. In conclusion, MRI appeared to provide the most definitive diagnostic images of endometriotic cysts in dogs and sheep, particularly for lesions in sheep which were not evident by ultrasonography. However, ultrasonography was sufficient to characterize most endometriotic cysts in dogs. Further research needs to be carried out to develop specific PET tracers for endometriosis.

Zusammenhang zwischen Bestands-, Gruppengröße und Indikatoren des Tierwohls in der konventionellen Schweinemast / Relationship between herd-, group size and indicators of animal welfare in conventional pig fattening farms

In der Vergangenheit hat der Tierschutzgedanke in der Gesellschaft zunehmend an Bedeutung gewonnen. Viele Verbraucher verbinden moderne landwirtschaftliche Nutztierhaltung primär mit hohen Tierzahlen auf engem Raum und daraus vermeintlich resultierenden Defiziten im Tierschutz. Der Begriff „Massentierhaltung“ wurde zum Synonym für wenig tiergerechte Haltungssysteme. Er wird vor allem in den Medien stark diskutiert und ist eindeutig negativ besetzt. Neben der Bestandsgröße steht besonders die Gruppengröße in der Kritik. Die Kenntnisse über die kausalen Beziehungen zwischen Tierwohl/Tierverhalten und konventioneller Tierhaltung bzw. Bestands- und Gruppengrößen sind dabei ausgesprochen gering, und zwar sowohl auf theoretischer wie auf empirischer Ebene. In der Literatur vorhandene Studien beziehen lediglich einzelne Teilaspekte des Tierschutzes und nur bestimmte Tierarten in ihre Auswertungen mit ein. Insbesondere für die Mastschweinehaltung, die in der aktuellen Tierschutzdebatte besonders im Fokus steht, gibt es nur wenige wissenschaftliche Untersuchungen, die sich mit dem Einfluss der Bestands- bzw. Gruppengröße auf das Tierwohl beschäftigen. Aus diesem Grund sollten im Rahmen der vorliegenden Studie belastbare Aussagen über den Zusammenhang von Tierwohlindikatoren und der Bestands- sowie Gruppengröße abgeleitet werden. Die Bestandsgröße wurde definiert als die gesamte Anzahl Tiere einer Nutzungsrichtung an einem Standort. Die Gruppengröße beschreibt die Anzahl Schweine pro Bucht bei konstantem Platzangebot pro Tier. Darüber hinaus sollte Auskunft über die Tiergerechtheit konventioneller Schweinemastbetriebe gegeben werden. Dies könnte zu einer wesentlichen Versachlichung der gesellschaftlichen Diskussion beitragen. Insgesamt wurden 60 Schweinmastbetriebe mit Hilfe des Welfare Quality®-Protokolls für Schweine bewertet. Unter Berücksichtigung der Betriebsgrößenentwicklung in der deutschen Schweineproduktion und in Anlehnung an die Einteilung der Betriebe nach dem Bundes-Immissionsschutzgesetz (BImSchG, 2013) wurden die untersuchten Betriebe in drei Bestandsgrößen-Kategorien (jeweils 20 Betriebe/Kategorie) eingeteilt: 1) klein = < 1.500 Mastplätze, 2) mittel = 1.500 bis 3.000 Mastplätze und 3) groß = 3.000 Mastplätze. Es wurden nur konventionell arbeitende Betriebe mit den am weitesten verbreiteten Haltungsverfahren (Planbefestigter Boden, Zwangsbelüftung, Automatische Fütterung) in die Untersuchungen einbezogen. Die teilnehmenden Betriebe hielten durchschnittlich 2.641 Schweine, wobei die Zahl der Mastplätze zwischen 260 und 11.000 variierte. Die Mehrheit der Betriebe (92%) hielt die Tiere auf Voll- und 8% auf Teilspaltenböden. Die Gruppengröße variierte von 10 bis 350 Schweinen pro Bucht (Mittelwert: 20 Schweine/Bucht, Median 16 Schweine/Bucht). Um den Einfluss der Gruppengröße auf das Tierwohl betrachten zu können, wurden drei Gruppengrößen-Kategorien definiert: klein: < 15 Schweine/Bucht, n = 207 Buchten; mittel: 15 bis 30 Schweine/Bucht, n = 257; groß: > 30 Schweine/Bucht, n = 136. Im Durchschnitt betrug das Platzangebot 0,83 m2/Schwein (0,31 m2 bis 2,5 m2/Schwein). Das Welfare Quality® Protokoll ist ein vornehmlich tierorientiertes Indikatorensystem, das der umfassenden Beurteilung des Wohlergehens von Nutztieren auf Betriebsebene dient. Es beinhaltet vier verschiedene Stufen, über die aus den ursprünglich 34 Indikatoren in einem hierarchischen Aggregierungsprozess 12 Kriterien, 4 Grundsätzen und schließlich ein einziger Gesamtscore gebildet wird. Die Betriebe wurden auf dieser Basis als „ausgezeichnet“, „verbessert“, „akzeptabel“ oder „nicht klassifiziert“ eingestuft. Die drei Bestandsgrößen- sowie Gruppengrößen-Kategorien wurden anhand der Mittelwerte aus der Tierwohl-Bewertung des Protokolls miteinander vergleichen. Der Betrieb wurde als zufälliger Effekt berücksichtigt. Annähernd 80% der Betriebe wurden als „verbessert“ klassifiziert; alle anderen Betriebe als „akzeptabel“. Grundsätzlich ist das Tierwohl-Niveau damit als eher niedrig einzustufen. Die Bestandsgröße hatte keinen signifikanten Effekt auf die Ausprägung des Gesamtscores. Auf der Grundsatz- und auf der Kriterienebene konnten keine signifikanten Unterschiede zwischen den Bestandsgrößen-Kategorien abgesichert werden. Der Grundsatz „Gute Fütterung“ erzielte mit Abstand die höchste Punktzahl, obwohl das dazugehörige Kriterium „Abwesenheit von Durst“, aufgrund teilweise erheblicher Mängel bei der Wasserversorgung nicht die volle Punktzahl erreichte. Das Kriterium „Liegekomfort“ des Grundsatzes „Gute Haltung“ wird mit Hilfe des Verschmutzungsgrades und dem Vorkommen von Bursitis bewertet. Im Mittel waren auf den 60 Betrieben, unabhängig von der Bestandgröße, 34,7% der Schweine von moderater Bursitis betroffen. Damit war Bursitis der am häufigsten vorkommende Indikator. Der Verschmutzungsgrad der Tiere nahm dagegen signifikant mit steigender Bestandsgroße zu (P < 0,05): in kleinen Betrieben betrug der Anteil moderat verschmutzter Tiere 10,7%, in mittleren 14,7% und in großen Beständen 20,6%. Hierbei ist das Fütterungssystem als möglicher Einflussfaktor zu diskutieren. Der Score für das Kriterium „Bewegungsfreiheit“ betrug im Durchschnitt 72 von 100 erreichbaren Punkten. Unter Berücksichtigung der Tierschutz-Nutztierhaltungsverordnung (2006) wurde festgestellt, dass etwa 40% der Buchten überbelegt waren. Der Grundsatz „Gute Gesundheit“ erzielte mit durchschnittlich 29,1 von 100 Punkten den schlechtesten Wert aller Grundsätze. Das dazugehörige Kriterium „Abwesenheit von Verletzungen“ bezieht unter anderem den Indikator Wunden ein. Mit einem Durchschnitt von 10,5% moderat verwundeten Schweinen über alle 60 Betriebe ist der Anteil vergleichsweise hoch. Die Schlachtbefunde fließen in das Kriterium „Abwesenheit von Krankheiten“ ein. Der Anteil Tiere mit veränderten Lungen war in kleinen Betrieben (4,2%) tendenziell geringer als in mittleren (11,6%) und größeren Betrieben (10,7%). Bei dem Schwänzekupieren und Kastrieren der Ferkel wurde kein Anästhetikum verwendet. Dadurch erzielte das Kriterium „Abwesenheit von Schmerzen“ eine geringe Punktzahl. Der Grundsatz „Artgemäßes Verhalten“ erreichte im Schnitt ebenfalls nur einen geringen Score von knapp über 30 von 100 Punkten. Ursächlich hierfür waren Defizite beim Erkundungsverhalten (Kriterium „Sonstiges Verhalten“) und die schlechte Bewertung des „emotionalen“ Zustandes der Tiere. Intensive Haltungssysteme werden oft hinsichtlich der mangelnden Ausübung von arttypischen Normalverhalten kritisiert. Aus den Ergebnissen lässt sich schlussfolgern, dass die Bestandsgröße als Indikator für unzureichendes Wohlergehen nicht geeignet ist. Eine hohe Anzahl an Mastschweinen pro Bestand deutet nicht automatisch auf eine niedrige Tierwohlbewertung hin. Hinsichtlich des Zusammenhangs zwischen den drei Gruppengrößen-Kategorien und ausgewählten Indikatoren des Welfare Quality® Protokolls wurde festgestellt, dass die Gruppengröße einen signifikanten Effekt auf den Verschmutzungsgrad der Schweine (P < 0,05) hat. In der größten Gruppengrößen-Kategorie (15,8%) waren signifikant mehr Tiere moderat verschmutzt als in der kleinen Gruppengrößen-Kategorie (10,4%). Der Anteil moderat verwundeter Schweine nahm ebenfalls mit steigender Gruppengröße signifikant zu. In der größten Gruppengrößen-Kategorie (16,3%) war der Anteil signifikant höher als in der mittleren (11,3%) sowie der kleinen Gruppengrößen-Kategorie (8,5%). Sowohl die Indikatoren Verschmutzungsgrad und Verwundung stellen wichtige Parameter bezüglich tiergerechter Haltungssysteme da. Das Vorkommen von Schweinen mit moderater Bursitis wurde dagegen nicht von der Gruppengröße beeinflusst (P > 0,5). Allerdings wurde in großen Gruppen signifikant häufiger eine gute Mensch-Tier-Beziehung beobachtet. Damit war keine der untersuchten Gruppengrößen-Kategorien bezüglich der Indikatoren des Welfare Quality® Protokolls konsistent überlegen. Vielmehr scheint die Bedeutung der Gruppengröße, ebenso wie die der Bestandsgröße, hinsichtlich des Wohls der Tiere überschätzt zu sein. Gleichzeitig muss allerdings festgestellt werden, dass die Gesamtsituation u.a. auf Grundlage der erfassten Prävalenzen der Technopathien als unbefriedigend bezeichnet werden muss. Die Diskussion um Tierschutz und Tierwohl muss weiter geführt werden. Es müssen andere Faktoren wie z.B. das Management oder das Haltungssystem in den Fokus der Untersuchungen genommen werden, um das Tierwohl auf den Betrieben nachhaltig zu verbessern.

The high health pigs gave the best results when factors like A.D.I., A.D.G., F.C.R., margin over feed cost and carcass weight are compared with the results of the other two treatments. The difference in achievement between the disease infected pigs and the high health pigs were not the same for all the production factors but it was still in favour of the high health pigs. The trial results show just how dynamic pig production has become. From the results it is very clear that there is not any place anymore for the old conventional type of housing in any modern piggery. The time has also come for every pig farmer to look at and evaluate the health status of his piggery and to consider changing over to a high health herd. From a production as well as an economic viewpoint it makes good sense.

Einleitung: Die Kationen-Anionen Bilanz (DCAB) des Futters modifiziert den Säure-Basen Status von Tieren und findet Anwendung in der Prävention von Milchfieber bei Kühen, MMA bei Sauen und Urolithiasis bei Haustieren. Durch die Veränderung des Kationen-Anionen-Verhältnisses in Futterrationen können biologische Prozesse beeinflusst werden, der Stickstoffmetabolismus. Ziel der Untersuchung: Die vorliegende Studie befasst sich mit der Frage, ob eine kaliumbedingte Variation der DCAB des Futters für wachsende Schweine einen Einfluss auf bestimmte Parameter des Säure-Basen-Haushaltes und möglicherweise auch auf die Stickstoffbilanz hat. Materialien und Methoden: Dem Versuch standen insgesamt 38 männlich kastrierte Mastschweine (Dreirassen-Kreuzung von Pietrain x Deutsches Edelschwein x Deutsche Landrasse) mit einem Einstallungsalter von ca. 12 Wochen und einer Lebendmasse von 17,3 kg bis 30,3 kg zur Verfügung. In randomisierter Reihenfolge erfolgte die Zuteilung der Schweine zu den acht Versuchsfuttern; Rohproteingehalt von 140 g kg-1 Futter und 200 g kg-1 Futter, sowie je 4 g, 10 g, 14 g oder 20 g Kalium kg-1 Futter. Die Versuchsfutter wiesen eine konstante Konzentration an Natrium und Chlorid auf. Die Hauptfutterkomponenten waren Mais und Weizen. Die beiden Rohproteingehalte wurden durch unterschiedliche Sojaextraktionsschrot- und Maisklebermengen gewonnen. Über Kaliumhydrogencarbonat (KHCO3) und Kaliumchlorid (KCl) resultierte die Einstellung der genannten Kaliumkonzentrationen sowie der vier DCAB-Stufen von -125 mEq kg-1, 66 mEq kg-1, 168 mEq kg-1, und 342 mEq kg-1 Futter. In einer Adaptionsphase von 15 Tagen gewöhnten sich die Scheine an das Versuchsfutter und die Umgebung. Die Einstallung erfolgte in Einzelboxen und die Versuchstiere erhielten Wasser ad libitum. Während der anschießenden zwei Bilanzphasen von je fünf Tagen wurden die Schweine in Bilanzkäfigen gehalten. Zwischen den beiden Bilanzphasen kam es zu einer fünftägigen Pause ohne Änderung der Fütterung. In der Bilanzzeit wurden der gesamte Harn und Kot der Tiere gesammelt sowie der dazugehörige pH-Wert kontinuierlich bestimmt. Harn- und Kotaliquots wurden für Stickstoff- und Elektrolytanalysen einbehalten. Jede fünftägige Bilanz endete mit der Gewinnung einer Blutprobe von jedem Schwein aus der Vena jugulares zur Bestimmung von Kalium, Natrium, Chlorid, pH-Wert, Hydrogencarbonat, Basenüberschuss und Aminosäuren. Ergebnisse: Kalium hat einen Einfluss auf den Harn pH-Wert. Analog zur steigenden Kaliumaufnahme (DCAB↑) wurden die Harn pH-Werte basischer (-125 mEq kg-1 Futter = Ø 5,93; 342 mEq kg-1 Futter = Ø 8,37). Die Blut pH-Werte, die im Durchschnitt bei 7,21 lagen, wie auch die Hydrogencarbonat- und Basenüberschusskonzentration, reagierten aufgrund der renalen Kompensation nicht wesentlich auf die unterschiedlichen DCAB im Futter. Die dazugehörigen Kot pH-Werte waren bei -125 mEq kg-1 Futter und 66 mEq kg-1 Futter um 0,16 höher als bei den anderen beiden DCAB-Stufen. Die Stickstoffaufnahme variierte zwischen 0,90 g kg-1KM d-1 und 1,22 g kg-1KM d-1 aufgrund der beiden Rohproteingehalte (14 und 20 %) in den Versuchsrationen. Eine Senkung der DCAB im Futter bewirkte eine Verbesserung der Stickstoffverdaulichkeit von 86,1 % auf 89,9 % (p<0,05). Hingegen zeigten die Diäten mit der kaliumärmsten Konzentration die höchsten renalen Stickstoffexkretionen von 442 mg kg-1KM d-1 gegenüber den anderen drei Kaliumkonzentrationen (345 mg kg-1KM d-1). In Folge dessen ergibt sich eine Stickstoffretentionssteigerung mit zunehmender DCAB im Futter. Jedoch wurde bei 66 mEq kg-1 Futter (Kalium 10 g kg-1 Futter) die höchste Stickstoffretention von 643 mg kg-1KM d-1 festgestellt. Die Untersuchung der Blutproben ergab keine Beeinflussung der Summe aller Aminosäuren, die bei ø 44,66 mg dl-1 lag. Die Summe der essentiellen Aminosäuren war bei einer DCAB von 66 mEq kg-1 Futter im Blut geringer als bei den übrigen Variationen. Einige Parameter der Elektrolytbilanzen waren zwischen den Futtervariationen verschieden: Bei dem Versuchsfutter mit einer DCAB von -125 mEq kg-1 Futter (Kalium 4 g kg-1 Futter) schieden die Schweine Na: 2,83 mg kg-1KM d-1 und Cl: 1,54 mg kg-1KM d 1 weniger mit dem Kot und Na: 7,05 mg kg-1KM d-1 weniger mit dem Harn aus gegenüber den weiteren Versuchsgruppen. Die renale Chloridexkretion zeigte keine Variabilität. Die renale, als auch die fäkale Kaliumausscheidung nahm analog zur DCAB des Futters zu (DCAB im Futter: -125 mEq kg-1; 66 mEq kg-1; 168 mEq kg-1; 342 mEq kg-1; K-Abgabe in mg kg-1KM d-1, renal: 74,0; 273,3; 431,1; 609,1; fäkal: 24,5; 31,2; 32,6; 44,0). In der Gesamtheit betrachtet ergibt sich für die Natrium- und Chloridretention keine richtungsweisende Beeinflussung im Zusammenhang mit der DCAB der Versuchsrationen. Die Kaliumretention hingegen stieg von 66,5 mg kg-1KM d-1 (-125 mEq kg-1 Futter) auf 167,0 mg kg-1KM d-1 (342 mEq kg-1 Futter) an, was nicht von den Kaliumkonzentrationen im Blut wiedergegeben wurde. Entsprechendes gilt für die Natrium- und Chloridkonzentrationen im Blut. Schlussfolgerungen: In der Alkalisierung des Harns zeigt sich, dass der DCAB des Futters Einfluss auf den Säure-Basen Status nimmt. Der systemische pH-Wert blieb aufgrund der Puffersysteme des Organismuses weitestgehend unberührt. Durch die KHCO3-Zulagen wurde das intragastrale bzw. das intestinale pH-Milieu verändert, was sich in der schlechteren Verdaulichkeit von Stickstoff bei höherer DCAB wiederspiegelt. Die Stickstoffretention steht in keinem Zusammenhang mit der Stickstoffverdaulichkeit. Mit dem DCAB von 66 mEq kg-1 Futter bzw. K: 10 g kg-1 Futter wurde die beste Retention für Stickstoff beobachtet. Die täglichen Gewichtszunahmen und die Futterverwertungen der Versuchsschweine konnten dies allerdings nicht reflektieren. Anzumerken sei, dass für einen eindeutigen Effekt auf die tägliche Zunahme eine längere Beobachtungsphase notwendig wäre (Sprung der täglichen Zunahmen von 520 g für -125 mEq kg-1 Futter auf das Niveau von 692 g für 66 mEq kg-1 Futter und mehr). Eine Empfehlung in Anlehnung an diese Studie wäre ein DCAB-Wert um die 66 mEq kg-1 Futter. Wird dieser Wert erhöht sinkt die Stickstoffverdaulichkeit auf der anderen Seite verschlechtert sich die Stickstoffretention bei Verringerung der DCAB. / Initiation: The dietary cation-anion balance (DCAB) of the feed modifies the acid-base balance and is used in the prevention of milk fever in cows, MMA in sows and urolithiasis in pets. The modification of the cation-anion ratio in diets can take an impact on biological processes inducting nitrogen metabolism. Objectives of investigations: This study objectively clarifies, whether potassium-based variation of the DCAB of the food has an influence on certain parameters of the nitrogen balance and the acid-base balance. Materials and Methods: The trial covered a total of 38 male castrated pigs (three racial crossing Pietrain x Large White x German Landrace) with a housing-age of approximately 12 weeks and a live weight of 17.3 kg to 30.3 kg. In randomized order, the pigs were allocated to the eight experimental feed: crude protein content of 140 g per kg feed and 200 g per kg feed, as well as 4, 10, 14 or 20 g of potassium per kg feed. The sodium and chloride concentrations in the feed were kept constant. The main food components were corn and wheat. The two crude protein levels were determined by various soybean meal and corn gluten quantities. Potassium hydrogen carbonate (KHCO3) and potassium chloride (KCl) were used to establish the four DCAB levels of -125 mEq kg-1, 66 mEq kg-1, 168 mEq kg-1, and 342 mEq kg-1 feed. In an adaptation period of 15 days pigs were accustomed to food and environment. They were kept in individual pens and were given water ad libitum. During the following two trial phases of five days each, the pigs were kept in balance cages. Between the two trial periods, there was a break of five days (no diet change). During the trial period all urine and excrement of the animals was collected, and the respective pH-value was continuously measured. Aliquots of urine and faeces were used in nitrogen and electrolyte analyses. At the end of each five-day record a blood sample from the jugular vein was taken from each pig for determination of potassium, sodium, chloride, pH-value, hydrogen carbonate, base excess, and amino acids. Results: Potassium has a significant influence on renal pH values. Analogous to increasing potassium intake (DCAB ↑), the urine pH value turned more basic (-125 mEq kg-1 feed = 5.93; 342 mEq kg-1 feed = 8.37). The blood pH levels, which averaged at 7.21, as well as the hydrogen carbonate concentration and base excess concentration, did not respond to the different DCAB in the feed because of the renal compensation. The associated feces pH values at -125 mEq kg-1 feed and 66 mEq kg-1 feed were higher by 0.16 than at the other two DCAB levels. The nitrogen intake varied between 0.90 g kg-1BM d-1 and 1.22 g kg-1BM d-1, based on both crude proteins (14 % and 20 %) in the experimental feeds. A reduction of DCAB in the feed resulted in an improvement of the nitrogen digestibility from 86.1 % to 89.9 % (< 0.05). However, diets with the lowest concentration of potassium showed the highest renal nitrogen excretions of 442 mg kg-1BM d-1 compared to the other three concentrations of potassium (345 mg kg-1BM d-1). As a consequence, nitrogen retention increases with increasing DCAB in the feed. However, the highest nitrogen retention of 643 mg kg-1KM d-1 was found with a 66 mEq kg-1 diet (potassium 10 g kg-1 feed). The examination of blood samples revealed no influence on the sum of the amino acids, which was 44.66 mg dl-1. The sum of the essential amino acids was reduced at a DCAB of 66 mEq kg-1 in blood, similar to the other variations. Some parameters of the electrolyte balances were different between the feed variations: In the experimental diet with a DCAB of -125 mEq kg-1 diet (potassium 4 g kg-1 feed), the pigs eliminated Na: 2.83 mg kg- 1BM d-1 and Cl: 1.54 mg kg– 1BM d-1 less in the feces and Na: 7.05 mg kg- 1KM d-1 less in the urine with respect to the other experimental groups. Renal chloride excretion showed no variability. The renal and fecal excretion of potassium increased proportionally to the DCAB of the feed (DCAB in the feed: -125 mEq kg-1, 66 mEq kg-1, 168 mEq kg-1; 342 mEq kg-1; K output in mg kg- 1BM d-1, renal: 74.0; 273.3; 431.1; 609.1; fecal: 24.5; 31.2; 32.6; 44.0). When viewed against the totality of results for the sodium and chloride retention, there were no trend-setting influences in connection with the DCAB of the experimental diets. The potassium retention, however, increased from 66.5 mg kg- 1BM d-1 (-125 mEq kg-1 feed) to 167.0 mg kg- 1BM d-1 (342 mEq kg -1 feed), which was not reproduced from the potassium concentrations in the blood. The same applied to the sodium and chloride concentrations in the blood. Conclusions: The alkalization of the urine shows that the DCAB of the feed influences the acid-base status. The systemic pH remained largely unaffected due to the buffer systems of the organism. The intragastric, respectively the intestinal, pH medium was changed by the addition of potassium hydrogen carbonate, which is reflected in the poorer digestibility of nitrogen at higher DCAB. The nitrogen retention is not related to the nitrogen digestibility. The best retention of nitrogen was observed with the DCAB of 66 mEq kg-1 feed (K: 10 g kg-1 feed). The daily weight gain and feed utilizations of the pigs certainly could not reflect this. It should be noted, however, that a longer observation period would be necessary for a clear effect on daily gain (jump of the daily weight gain from 520 g of -125 mEq kg-1 feed to the level of 692 g for 66 mEq kg-1 feed and more). A recommendation based on this study would be a DCAB value of 66 mEq kg-1 feed. If this value increases, the nitrogen digestibility decreases; on the other hand, the nitrogen retention deteriorated with reducing DCAB.

Porcine circovirus type 2 is a single stranded DNA virus that belongs to the genus Circovirus in the family Circoviridae. This virus is associated with many kinds of diseases in pigs and causes significant economic losses in swine-breeding. In this study, two approaches of vaccination were tested in order to develop an effective vaccine against PCV2. The first approach was to test DNA vaccines. For this purpose, eukaryotic expression plasmids encoding two form of PCV2 Cap protein were constructed. The expression plasmids encoding murine TNF-α and IFN-α1 were also prepared for co-immunization with antigen encoding plasmid to enhance the immune response. The second approach is based on the previous finding that chimeric pentamers of VP1 mouse polyomavirus capsid protein fused with PCV2 can induce protective immunity against PCV2. These chimeric pentamers were further modified by AA substitutions in PCV2 Cap immunodominant epitope in order to enhance protective antibody response directed against PCV2. The chimeric pentamers and DNA vaccines were tested for ability to induce antibody immune response against PCV2 in mice. Efeito da infecção pelo Toxoplasma gondii na expressão de genes associados à resposta imune em tecidos de suínos / Effect of Toxoplasma gondii infection on immune related tissue gene expression in pigs

A toxoplasmose é uma zoonose de ampla distribuição mundial afetando homens e diversas espécies animais. Levantamentos sorológicos indicam elevados índices de infecção, porém relatos de doença severa é rara. A infecção pelo T. gondii induz uma intensa resposta imune mediada pelo interferon-&#947; (IFN-&#947;) que rapidamente controla a multiplicação parasitária. Com o objetivo de explorar a resistência da espécie suína à toxoplasmose como modelo de estudo para a compreensão dos mecanismos de defesa a infecção, foram realizadas infecções orais com 4,5 x 105; oocistos (cepa VEG) e colheitas de amostras de linfonodo hepato-esplênico (LN HS), mesentérico (LN M) e íleo-cólico (LN IC), fígado, sangue, íleo, jejuno, baço e timo aos 2, 4, 7 e 14 dias pós-infecção (DPI). Analisou-se a expressão de 69 genes ligados a resposta de defesa às infecções pela Real-Time RT-PCR*. LN M, LN HS e fígado foram as amostras que apresentaram a maior quantidade de genes e maior intensidade de ativação enquanto que células mononucleares de sangue periférico (CMSP) e timo apresentaram reduzida resposta à infecção. A expressão e a produção de IFNG foram mais altas nas amostras de LN M e LN HS comparadas às amostras de LN traqueo-bronquial e CMSP, indicando diferentes níveis de resposta local. Intensa indução de resposta inata e inflamatória foi observada em vários tecidos, envolvendo os genes IL1B, IL6, IFNA1, TNF, ORM1, MYD88, TLR2, TLR4; estimulação de resposta Th1, mediada por IFNG incluiu os genes IRF1, IL23A, IL18, STAT1, SOCS1, SOCS3, ICSBP1, TBX21; balanceada pela expressão de citocinas regulatórias IL10, TGFB1 e TGFB3. Uma proeminente indução de ARG1, INDO and SLC11A1 indica ativação de mecanismos de proteção do hospedeiro envolvendo metabolismo de aminoácidos (arginina, triptofano) e ferro. A presença de parasitas foi detectada no LN M aos 2DPI pela Real-Time PCR e pela imunohistoquímica. Aumento do número de parasitas no 4DPI foi seguida de intensa resposta inflamatória, edema e necrose tecidual no 7DPI principalmente no fígado e no LN M. Elevados níveis de AST sérico no 7DPI confirmam a lesão de hepatócitos. A diminuição da inflamação e da quantidade de parasitas no 14DPI sugerem controle da proliferação parasitária. Elevados níveis de haptoglobina e óxido nítrico séricos detectados aos 4DPI (&#945;=0,05) indicam respectivamente a ativação de proteínas de fase aguda e de macrófagos. A análise de citometria de fluxo mostra elevação da porcentagem de células CD2/CD16 DP sugerindo envolvimento de células NK e não foram observadas alterações na porcentagem de células CD4+/CD8+, CD3+/CD25+. A estimulação na expressão gênica, a produção de IFN-&#947;, as determinações séricas e lesões histológicas apresentam padrão similar, com início de resposta no 2DPI, elevação no 4DPI e 7DPI e diminuição no 14DPI. A análise da expressão de mRNA nos tecidos revelou alguns dos mecanismos de defesa do hospedeiro ativados durante a infecção pelo T. gondii. Observou-se uma equilibrada ativação de citocinas pró- e anti-inflamatórias envolvendo uma intrincada rede de mecanismos co-estimulatórios e regulatórios coordenando a resposta do tipo Th1. *Siglas dos genes segundo o Human Gene Nomenclature Committee (HGNC) / Toxoplasmosis is a widespread zoonosis affecting humans and large range of animal species worldwide. Serological surveillance indicates high infection rates, but rarely is associated to severe disease. T. gondii (Tg) infection induces a strong IFN-&#947; dominated immune response that rapidly controls parasite replication. Pig resistance to toxoplasmosis was explored as an experimental model to evaluate host defense mechanisms. Pigs were fed 4.5 x 105 oocysts (VEG strain) and hepato-splenic (HS LN), mesenteric (MLN) and ileal lymph nodes (I LN), liver, blood, ileum, jejunum, spleen and thymus samples were collected at 2, 4, 7 and 14 days after infection (DAI). Gene expression analysis for a panel of 69 immune related genes were performed by Real-Time RT-PCR*. Most intense response were detected in MLN, HS LN and liver samples, whereas low changes were observed in periferic blood monocytes (PBMC) and thymus. IFNG mRNA expression and protein production were higher in MLN and HSLN cells than in tracheo-bronchial LN and PBMC, suggesting different levels of local response. Intense innate and inflammatory induction were observed in most of the tissues involving IL1B, IL6, IFNA1, TNF, ORM1, MYD88, TLR2, TLR4; stimulation of IFNG dominated Th1 response included IRF1, IL23A, IL18, STAT1, SOCS1, SOCS3, ICSBP1, TBX21; balanced by expression of regulatory cytokines IL10, TGFB1 and TGFB3. Prominent ARG1, INDO and SLC11A1 upregulation indicate activation of host amino acid (arginine and tryptophan) and iron protective mechanisms. Tg parasites were detected by Real-Time PCR and immunohistochemistry in MLN as early as 2DAI. Increase of parasite burden at 4DPI was followed by an intense inflammatory response, edema and tissue necrosis in liver and LNM at 7DPI. Increased serum AST levels at 7DPI confirmed liver damage. Reduced parasite numbers and inflammatory response suggest control of parasite replication at 14 DPI. High serum haptoglobin and nitric oxide at 4DAI (&#945;=0.05) indicate acute phase protein and macrophage activation, respectively. Flow citometry analysis showed increased percentage of CD2/CD16 DP suggesting involvement of NK cells, whereas no changes were observed at CD4+/CD8+, CD3+/CD25+ cells. Gene expression stimulation, IFN-&#947; production, serum determinations and histological lesions showed similar pattern, of induction at 2DPI, increasing at 4DPI and 7DPI and decreasing at 14DPI. Tissue mRNA expression analysis revealed some of the host defense mechanisms activated during T. gondii infection. A balance of pro- and anti-inflammatory cytokine activation, involving an intricated co-stimulatory and regulatory mechanisms that coordinates Th1 response was observed.*Gene names according to Human Gene Nomenclature Committee (HGNC)
