Influence of peptidoglycan metabolism on immunomodulatory properties of Lactobacillus casei / Influence du métabolisme du peptidoglycane sur les propriétés immunomodulatrices de Lactobacillus casei

Le peptidoglycane (PG) est le composant majeur de la paroi des bactéries à Gram positif. Il assure la forme et l’intégrité de la cellule bactérienne. Le PG ou des fragments dérivés sont connus pour être des inducteurs du système d’immunité innée de l’hôte, en particulier au travers des récepteurs Nod2. Au cours de ce travail, nous avons étudié l’influence du métabolisme du PG sur les propriétés immunomodulatrices de Lactobacillus casei BL23, en étudiant principalement sa capacité à moduler la réponse des cellules dendritiques humaines. Nous avons tout d’abord caractérisé les hydrolases du PG (PGHs) majeures de L. casei BL23. Une recherche in silico a révélé que L. casei possède un système de PGHs relativement complexe comprenant treize enzymes putatives avec des domaines catalytiques variés. Une analyse protéomique d’extraits de paroi de L. casei BL23 a permis de détecter la production de sept d’entre elles pendant la croissance bactérienne. Quatre d’entre elles ont été étudié plus en détails. La PGH la plus fortement exprimée, Lc-p75, a une activité de -D-glutamyl-L-lysyl-endopeptidase et est responsable de la séparation des cellules après division. De plus, Lc-p75 associée à la paroi est localisée au niveau des septa cellulaires. Il s’agit également de l’une des protéines majeures secrétée dans le surnageant de culture de L. casei BL23. Lc-p75 possède la particularité d’être une glycoprotéine. La PGH Lc-p40 possède un domaine CHAP doué d’une activité endopeptidase avec un site de clivage situé au niveau des ponts interpeptidiques du PG. Lc-p40 parait localisée au niveau de la paroi latérale des cellules de L. casei. Lc-p45 est une deuxième -D-glutamyl-L-lysyl-endopeptidase avec un rôle dans le maintien de la forme de la bactérie. Enfin nous avons caractérisé deux enzymes de prophages, Lc-Lys et Lc-Lys2, codée par le génome de L. casei BL23, qui possède toute deux un domaine de liaison au PG d’un nouveau type qui possède la particularité de lier spécifiquement le D-Asp amidé présent dans les ponts interpeptidiques du PG de L. casei BL23. La délétion des deux gènes qui codent pour les endopeptidases Lc-p75 et Lc-p45 chez L. casei BL23 conduit à l’absence de disaccharide dipeptide dans la structure du PG du mutant, tandis que la délétion de Lc-p75 seulement conduit à une réduction de la quantité du disaccharide-dipeptide. Ce disaccharide dipeptide est un ligand des récepteurs Nod2. Les deux mutants obtenus par délétion de Lc-p75 ou bien par délétion des deux endopeptidases ont été comparés avec la souche sauvage BL23 pour leur capacité à activer in vitro des cellules dendritiques humaines dérivées de monocytes sanguins. Suite à l’activation des cellules dendritiques par les souches de L. casei, quatre cytokines pro-inflammatoires, les interleukines IL-6, IL-8, IL-12 et le TNF- ont été produites. La quantité de chaque cytokine sécrétée en réponse aux mutants simple Lc-p75 et double Lc-p75/Lc-p45 était diminuée par rapport à celle induite par la souche sauvage L. casei BL23.En conclusion, L. casei BL23 est doté d’un équipement complexe en PGHs. Les PGHs caractérisées au cours de ce travail présentent des caractéristiques uniques et jouent un rôle important dans la division des bactéries ainsi que dans le maintien de leur morphologie. Nos résultats indiquent que la souche sauvage de L. casei Bl23 et les mutants dérivés obtenus par inactivation d’enzymes à activité endopeptidase, qui diffèrent à la fois au niveau de leur contenu enzymatique ainsi qu’au niveau de la structure de leur PG, ont des effets différents sur les cellules dendritiques humaines, avec un caractère anti-inflammatoire plus élevé pour les mutants / Peptidoglycan (PG) is the major component of the Gram-positive bacteria cell wall. It ensures bacterial cell shape and integrity. PG or PG-derived fragments have been shown to stimulate the host innate immune system, through Nod-2 receptors. In this work, we studied the influence of PG metabolism on immunomodulatory properties of Lactobacillus casei BL23, mainly its ability to modulate the response of human dendritic cells (DCs).We have first characterized the main peptidoglycan hydrolases (PGHs) of L. casei BL23. In silico search revealed that L. casei BL23 has a rather complex PGH complement including thirteen predicted PGHs with various catalytic domains. Proteomic analysis of bacterial cell wall extracts revealed the expression of seven of them during bacterial growth. We characterized four of them in details. Lc-p75 is the major PGH with a γ-D-glutamyl-L-lysyl-endopeptidase specificity and is responsible for daughter cell separation. Lc-p75 associated to the cell wall localizes at the cell septa. It is also one of the major secreted proteins of L. casei found in culture supernatant. Besides, we showed that L. casei Lc-p75 is a glycosylated protein. Lc-p40 is a PGH with a CHAP-domain endowed with endopeptidase hydrolytic specificity toward peptidoglycan cross-bridges and appears to localize on lateral cell wall. Lc-p45 is a second γ-D-glutamyl-L-lysyl endopeptidase with a role in cell shape maintenance. We further demonstrated that two prophage endolysins Lc-Lys and Lc-Lys2, encoded in L. casei BL23 genome, share a common novel type peptidoglycan-binding domain that recognizes specifically D-Asn cross-bridge, present in L. casei BL23 peptidoglycan.Deletion of the two endopeptidases, Lc-75 and Lc-p45, resulted in a complete loss ofdisaccharide-dipeptide, which is a ligand of Nod-2 receptor, in the muropeptide structure of L. casei BL23, whereas deletion of Lc-p75 gene led only to a reduction of disaccharide dipeptide. The two PGH-mutants, obtained by deletion of Lc-p75 gene alone or both Lc-p75 and Lc-p45 endopeptidase genes were compared with wild type L. casei BL23 for their capacity to stimulate signaling pathways in vitro in DCs derived from human monocytes. As a consequence of DC activation by L. casei strains, four pro-inflammatory cytokines IL-6, IL-8, IL-12 and TNF-α were produced. The concentrations of secreted cytokines in response to the single Lc-p75 and Lc-p75/p45 double mutant were lower than those induced by wild type L. casei BL23.In conclusion, L. casei BL23 has a complex PGH complement. The PGHs described in this work present unique features and play important role in cell division and morphology of L. casei. Our results indicate that wild type L. casei and endopeptidase-negative mutants, which differ in their PGH content and in their PG structure, have distinct effects on human DCs, with a higher anti-inflammatory character of the endopeptidase-negative mutants.

Investigations on the gut microbiota of salmonids and the applications of probiotics-based feed additives

A series of investigations were conducted in order to characterise the GIT microbiota of salmonids and to determine the effect of microbial modulating feed additives on the intestinal microbiota, immunity and growth of salmonids. The first experiment, Chapter three, used PCR-DGGE and 16S rRNA gene sequence analysis of cultivable bacteria were used to investigate the GIT microbiota of brown trout. 16S rRNA gene sequence analysis demonstrated that Citrobacter freundii and Carnobacterium maltaromaticum were the predominant culturable viable bacteria and lactic acid bacteria, respectively in all regions of the GIT. DGGE revealed complex communities with a diverse range of microbes from the Firmicutes and Proteobacteria phyla. The latter chapters focused not only identifying the gut microbiota of salmonids, but also on the ability of probiotics and prebiotics to modulate these communities. In Chapter four, rainbow trout were fed a commercial diet supplemented with P. acidilactici for four weeks. P. acidilactici was detected in the GIT of the probiotic group by multiple methods and P. acidilactici was able to persist for at least 24h at the cessation of probiotic feeding. Histological appraisal on the intestine revealed significantly higher microvilli density in the posterior mucosa and a higher density of goblet cells in the anterior mucosa of the probiotic fed fish. RT-PCR results demonstrated that IL-1β, IL-8 and IgT gene expression were up-regulated in the P. acidilactici fed fish at the end of the study. Whilst mRNA of PCNA, HSP70 and casp-3 were down-regulated in the probiotic group at both sampling points. In Chapter five, the efficacy of dietary administration of P. acidilactici and short chain fructooligosaccharide (scFOS) on Atlantic salmon (Salmo salar L.) was evaluated at 63 and 132 days. Compared to the control group, total bacterial cell counts in all regions of the intestine with exception of the anterior digesta were significantly lower in the synbiotic group at the mid sampling point. PCR-DGGE revealed that species richness, diversity and the number of OTUs were significantly higher in the synbiotic group in the anterior digesta at the mid sampling point. Intestinal microvilli and villi length were increased in the anterior intestine of the synbiotic fed group at the end sampling point. IEL levels were increased in the synbiotic group in the posterior intestine at both sampling points. The expression of immunological genes were significantly up-regulated in the synbiotic fed salmon. In Chapter six, rainbow trout were fed three diets fishmeal (FM), soybean meal (SBM) and PlantMix diets supplemented with or without P. acidilactici for 12 weeks. At both sampling points, with exception of fish fed FM, LAB levels were significantly higher in all probiotic groups compared to the control groups. Serum lysozyme activity was significantly higher in fish fed FM and SBM diets containing P. acidilactici than that of fish fed the control diets. This body of research demonstrates that P. acidilactici can modulate immune response via up-regulation of immune genes as well as modulate IEL and goblet cell levels. Despite these benefits, P. acidilactici had no detrimental effects on growth performance.

Fluorescence and bioluminescence imaging of the intestinal colonization of Enterococcus mundtii ST4SA and Lactobacillus plantarum 423 in mice infected with Listeria monocytogenes EGde

Thesis (MSc)--Stellenbosch University, 2015. / ENGLISH ABSTRACT: Lactic acid bacteria (LAB) are common inhabitants of the human gastro-intestinal tract (GIT). Some LAB, especially lactobacilli, are well known for their application in fermented foods and probiotic properties. These microorganisms exert many beneficial effects on human health, such as digestion and assimilation of food and preventing pathogens colonising the GIT. Furthermore, some selected probiotic strains are believed to perform a critical role in the treatment of gastro-intestinal disorders, lactose intolerance and in the stimulation of the immune system. Despite the ever increasing consumer interest in probiotic LAB, the mechanisms by which they exert their beneficial effects and the activities of probiotics in the GIT often remain poorly understood. Understanding survival mechanisms of LAB in the GIT, especially the interaction between LAB and pathogens, would be facilitated by the direct in vivo monitoring of these processes. Using the mCherry fluorescence gene, we successfully constructed Lactobacillus plantarum 423 and Enterococcus mundtii ST4SA reporter strains. With this study we showed that fluorescence imaging can be used to detect Lb. plantarum 423 and Ent. mundtii ST4SA in the GIT of mice. The two species colonized the cecum and colon for at least 24 h after one oral administration. To our knowledge, this is the first report on fluorescence imaging of LAB expressing mCherry in a mouse model. Using a bioluminescence marker system, we evaluated the impact of Lb. plantarum 423 and Ent. mundtii ST4SA on orally acquired Listeria monocytogenes infection and the ability of the probiotics to compete with the pathogen in the GIT of mice. Challenging Lb. plantarum 423 and Ent. mundtii ST4SA that were already established in the GIT of mice with L. monocytogenes EGDe had no effect on the survival and persistence of the probiotic strains. We demonstrated that the colonization of mice with Lb. plantarum 423 and Ent. mundtii ST4SA, or a combination of the strains, protected the animals against colonization of the GIT by L. monocytogenes EGDe. Enterococcus mundtii proved more effective than Lb. plantarum 423 in reducing the number of L. monocytogenes EGDe in the mouse model. / AFRIKAANSE OPSOMMING: Melksuurbakterieë (MSB) kom algemeen in die mens se spysverteringkanaal (SVK) voor. Verskeie MSB, veral lactobacilli, is bekend vir hul gebruik in gefermenteerde voedsel en as probiotika. Die bakterieë het baie eienskappe wat die mens se gesondheid kan bevoordeel, insluitend vertering en assimilasie van voedsel en voorkoming van kolonisering van die SVK deur patogeniese bakterieë. Sekere probiotiese rasse speel ook ʼn belangrike rol in die behandeling van SVK versteurings, laktose intoleransie en die stimulering van die immuun stelsel. Alhoewel die belangstelling in probiotiese bakterieë toeneem, is daar min inligting bekend oor die meganismes wat MSB gebruik om hulle voordelige eienskappe in die SVK uit te voer. Die oorlewing van MSB in die SVK, veral die interaksies tussen MSB en patogene, kan met behulp van ʼn in vivo moniteringsisteem bestudeer word. Deur die mCherry fluorisensie geen in Lactobacillus plantarum 423 en Enterococcus mundtii ST4SA te kloneer, het ons daarin geslaag om ʼn effektiewe verklikker sisteem te ontwikkel en kon die voorkoms en migrasie van die twee spesies in die SVK van muise bestudeer word. Lactobacillus plantarum 423 en Ent. mundtii ST4SA het veral die blindederm en kolon gekoloniseer. Beide rasse het na ʼn enkele mondelingse toediening vir ten minste 24 h in die SVK oorleef. Sover ons kennis strek, is hierdie die eerste verslag van fluoriserende MSB wat met behulp van die mCherry geenproduk in die SVK bestudeer is. Deur gebruik te maak van ʼn bioliggewende verklikker sisteem, het ons die vermoë van Lb. plantarum 423 en Ent. mundtii ST4SA om met Listeria monocytogenes in die SVK te kompeteer, bestudeer. Listeria monocytogenes het geen invloed gehad op die kolonisering van Lb. plantarum 423 en Ent. mundtii ST4SA nie. Deur die muise vooraf met Lb. plantarum 423 en Ent. mundtii ST4SA te koloniseer (in kombinasie of met net een van die twee rasse), kon ons daarin slaag om kolonisering van die SVK met L. monocytogenes te voorkom. In die muis model wat in hierdie studie gebruik is, was Ent. mundtii ST4SA meer effektief as Lb. plantarum 423 in die verlaging van Listeria selgetalle.

Otimização das propriedades reológicas e sensoriais de iogurtes probióticos enriquecidos com proteínas lácteas / Optimization of the rheological properties of probiotic yoghurts supplemented with milk proteins

Este estudo teve como objetivo otimizar as propriedades reológicas dos iogurtes probióticos suplementados com leite em pó desnatado (SMP), concentrado proteico de soro (WPC) e caseinato de sódio (Na-Cn), usando desenho experimental tipo simplex centroide, para modelagem de misturas. O estudo das interações entre os ingredientes (proteína concentrada de soro, caseinato de sódio e leite em pó) incluiu sete experimentos sendo três com os ingredientes puros, dois correspondentes a misturas binárias e um para a uma mistura ternária. Em paralelo aos sete experimentos delineados, foi realizado ensaio controle com leite não suplementado. Os leites foram fermentados a 42 °C até pH 4,5 empregando-se as culturas Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus e Bifidus lactis. A cinética de acidificação foi determinada bem como as análises físico-químicas, contagem de bactérias viáveis e características reológicas dos iogurtes. Modelos foram ajustados para os resultados (respostas cinética, contagem de bactérias viáveis e parâmetros reológicos) através de três métodos de regressão (linear, quadrático e cúbico especial) e aplicados às misturas. Os resultados deste estudo mostraram que a fortificação da base de leite com proteína concentrada de soro, caseinato de sódio e leite em pó desnatado, para o mesmo nível de teor de proteína, afetou o perfil de acidificação, a contagem de bactérias viáveis e reologia de iogurtes probióticos. Além disso, foi possível estimar o perfil de acidificação e a contagem de bactérias no leite suplementado com proteínas do leite em pó, concentrado proteico de soro e caseinato de sódio por metodologia de superfície de resposta, bem como calcular aproximadamente as propriedades viscoelásticas dos iogurtes probióticos. Pode-se ressaltar que a otimização das propriedades reológicas dos iogurtes probióticos suplementados com proteínas do leite depende da fonte de proteína utilizada. Por fim, a substituição parcial de 55% de leite em pó desnatado por concentrado proteico de soro e caseinato de sódio simultaneamente poderá reforçar as propriedades reológicas dos iogurtes probióticos, considerando a cinética de acidificação e a contagem de bactérias viáveis. Os resultados foram validados ao longo da vida de prateleira dos produtos, confirmando os valores estimados pelos modelos para a cinética de acidificação, a contagem de bactérias viáveis e as propriedades viscoelásticas, além da avaliação sensorial e da análise de microestrutura. / This study aimed to optimize the rheological properties of probiotic yoghurts supplemented with skim milk powder, whey protein concentrate and sodium caseinate throughout an experimental design type centroid simplex for mixture modeling. It included seven trials: three with the pure ingredients, three corresponding to the binary mixtures and one to the ternary one. A control experiment was accomplished with non supplemented milk. Milk was fermented at 42°C until pH 4.5 by Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus, and Bifidus animalis subsp. lactis. The kinetics of acidification was followed as well as physicochemical analyses, enumeration of viable bacteria and rheological characterization of the yoghurts. Models were adjusted to the results (kinetic responses, counts of viable bacteria and rheological parameters) through three regression models (linear, quadratic and cubic special) applied to mixtures. The results showed that the addition of milk proteins affected acidification profile and counts, and enhanced rheological properties of probiotic yoghurt. The estimated results were validated during shelf-life and were confirmed by for the kinetics of acidification, bacterial counts of viable and viscoelastic properties in addition to sensory evaluation and analysis of microstructure.

Efeitos clínicos, microbiológicos e imunológicos do probiótico Bifidobacterium animalis subsp. lactis com terapia adjuvante no tratamento não cirúrgico da periodontite crônica: estudo clínico controlado e aleatorizado / Clinical, microbiological and immunological effects of probiotic Bifidobacterium animalis subsp. lactis as adjuvant therapy in the non-surgical treatment of chronic periodontitis: a randomized controlled clinical trial

O propósito deste estudo foi avaliar o efeito adjuvante da terapia probiótica (TProb) no tratamento periodontal não cirúrgico em pacientes com Periodontite Crônica generalizada (PCg). Em um estudo clínico aleatorizado, duplo-cego e placebo-controle, 41 pacientes com PCg foram tratados com TProb associada à raspagem e alisamento radicular (RAR Grupo Teste) ou apenas RAR (Grupo Controle). Os pacientes do Grupo Teste consumiram pastilhas contendo Bifidobacterium animalis subsp. lactis (B. lactis) HN019 durante 4 semanas. Os pacientes do Grupo Controle receberam pastilhas placebo (sem probiótico). Parâmetros clínicos periodontais foram avaliados no baseline (período pré-intervenção) e em 30 e 90 dias após a RAR. Nestes mesmos períodos foram coletadas amostras de placa subgengival para contagem de 40 espécies bacterianas (checkerboard DNA-DNA hybridization) e detecção do genoma de B. lactis HN019 (Reação em cadeia da polimerase em tempo real - q-PRC), amostras do fluido crevicular gengival para avaliação dos níveis de Interleucina (IL)-1beta, IL-8 e IL-10 (Imunoensaios multiplex) e amostras de saliva para avaliação dos níveis de Imunoglobulina A (IgA) (Nefelometria). Foram realizadas, também, biópsias de tecido gengival para determinação da expressão de beta-defensina (BD)-3 e receptores do tipo Toll (TLR)-4 (reações imunohistoquímicas - estreptavidina-biotina-peroxidase). Os dados obtidos foram estatisticamente analisados (p<0,05). O Grupo Teste apresentou, aos 90 dias, redução de profundidade de sondagem (PS) e ganho de inserção clínica significativamente maiores que aqueles do Grupo Controle. O Grupo Teste também apresentou menor quantidade de bolsas periodontais moderadas (aos 30 e 90 dias) e profundas (aos 90 dias) que o Grupo Controle (p<0,05), bem como menor quantidade de pacientes (p<0,05) apresentando 3 ou mais sítios com PS &ge; 6 mm ou PS = 5mm e sangramento à sondagem (SS) positivo. O Grupo Teste apresentou menor SS (aos 30 e 90 dias) e menor IP (aos 30 dias) quando comparado ao Grupo Controle (p<0,05). Na análise microbiológica, o Grupo Teste apresentou proporções significativamente menores de espécies microbianas dos complexos vermelho (ao 30 e 90 dias) e do complexo laranja (aos 30 dias), bem como maiores proporções de espécies do complexo azul (aos 90 dias) quando comparado ao Grupo Controle. A análise por meio de q-PCR mostrou que o Grupo Teste apresentou um aumento no número de cópias/&mu;L do genoma da cepa probiótica B. lactis HN019 no biofilme subgengival aos 30 e 90 dias (p<0,05). Na análise imunológica, o Grupo Teste apresentou menor razão (valores ajustados ao baseline) de IL-1&beta; (aos 30 e 90 dias) e de IL-8 (aos 30 dias) do que o Grupo Controle (p<0,05). Apenas o Grupo Teste apresentou valores de IL-10 significativamente maiores que aqueles do baseline aos 30 dias. Não foram observadas diferenças entres os grupos nas razões de IgA aos 30 e 90 dias. Na análise imunohistoquímica, o Grupo Teste apresentou expressões significativamente maiores de BD-3 e TLR-4 em sítios doentes quando comparado ao Grupo Controle aos 30 dias. Pode-se concluir que a utilização do probiótico B. lactis HN019 como recurso adjuvante à RAR promove benefícios clínicos, microbiológicos e imunológicos adicionais no tratamento de pacientes com PCg / The purpose of this study was to evaluate the adjuvant effect of probiotic therapy (ProbT) in non-surgical periodontal treatment in patients with generalized chronic periodontitis (GCP). In this randomized, double-blind, placebo-control, 41 patients with GCP were treated with ProbT associated with scaling and root planing (SRP - Test Group) or SRP only (Control Group). The patients in the Test Group consumed lozenges containing Bifidobacterium animalis subsp. lactis (B. lactis) HN019 during 4 weeks. The patients in the Control Group received placebo (lozenges without probiotic). Periodontal clinical parameters were evaluated at baseline (pre-intervention) and at 30 and 90 days after SRP. Samples of subgingival plaque were collected to count 40 bacterial species (checkerboard DNA-DNA hybridization) and detection of genome of B. lactis HN019 (polymerase chain reaction in real time - q-PCR). Gingival crevicular fluid samples were collected for assessing levels of interleukin (IL)-1beta, IL-8 and IL-10 (multiplex immunoassays). Saliva samples were collected to measure levels of immunoglobulin A (IgA) (nephelometry). Also, biopsies of gingival tissues were performed to determine the expression of beta-defensina (BD)-3 and Toll-Like receptors (TLR)-4 (immunohistochemical reactions - streptavidin-biotin-peroxidase). The data obtained were statistically analyzed (p < 0.05). The Test Group presented, at 90 days, reduction of probing pocket depth (PPD) and clinical attachment gain significantly higher when compared to the Control Group. The Test Group also presented a lower number of moderate (at 30 and 90 days) and deep (90 days) periodontal pockets than the Control Group (p < 0.05), as well as a lower number of subjects (p < 0.05) with 3 or more sites with PPD &ge;6 mm or PPD = 5mm and bleeding on probing (BOP) positive. The Test Group presented lower BOP (at 30 and 90 days) and lower PI (at 30 days) when compared to the Control Group (p<0.05). In the microbiological analysis, the Test Group showed significantly lower proportions of microbial species of red (at 30 and 90 days) and the orange (at 30 days) complexes, as well as higher proportions of species of blue complex (at 90 days) when compared to the Control Group. q-PCR analysis showed that the Test Group presented an increase in the number of copies/&mu;L of the genome of the probiotic strain B. lactis HN019 in subgingival biofilm at 30 and 90 days (p < 0.05). In the immunological analysis, the Test Group presented a lower ratio of IL-&beta;1 (at 30 and 90 days) and IL-8 (30 days) than the Control Group (p < 0.05). Only the Test Group presented values of IL-10 significantly higher than those of the baseline at 30 days. No differences were observed between the groups on the ratios of IgA at 30 and 90 days. In the immunohistochemical analysis, the Test Group showed significantly higher expression of BD-3 and TLR-4 in sites with periodontitis when compared to the Control Group at 30 days. It can be concluded that the use of probiotic B. lactis HN019 as an adjuvant to SRP promotes clinical, microbiological and immunological additional benefits in the treatment of patients with GCP

O objetivo do presente estudo foi investigar as implicações da suplementação com levedura viva Saccharomyces cerevisiae e de um programa de treinamento físico de baixa intensidade sobre a digestibilidade dos componentes da dieta (MS, PB, EE, FDN, FDA, MO e amido), a resposta glicêmica e insulinêmica, perfil sérico de triglicerídeos, colesterol total e frações (HDL-C; LDL-C; VLDL-C), bem como mensurar a produção dos ácidos graxos de cadeia curta (AGCC) butírico, acético e propiônico, quantificar levedura viva nas fezes, avaliar o pH fecal, a população microbiana nas fezes e os níveis séricos de endotoxinas, além de mensurar a frequência cardíaca dos animais em exercício físico e na recuperação. Foram utilizados dez cavalos da raça Puro Sangue Árabe, machos, castrados, com idade média 72±7,5 meses e peso médio de 473±34,75 kg, alojados em baias individuais, alimentados com dieta constituída de 2% do PC em MS/dia, divididos em 0,75% de concentrado comercial multiparticulado e 1,25% de feno de gramínea (Cynodon dactylon sp. cv Tifton-85). Os tratamentos foram divididos em controle (sem adição de levedura) e suplementados (adição de 15g/dia de levedura viva Saccharomyces cerevisiae) em uma fase sem exercício e outra fase com exercício físico. O delineamento experimental utilizado foi inteiramente casualizado com esquema fatorial 2x2 e os dados obtidos foram submetidos à análise de variância pelo teste F. Para os coeficientes de digestibilidade observou-se efeito de exercício para os coeficientes de digestibilidade da MS, MO, FDN, FDA e PB (P<0,05). Para a variável EE observou-se interação da inclusão de levedura e exercício físico (P<0,1). O amido não apresentou efeito de tratamentos (P>0,1). Para os níveis plasmáticos de glicose observou-se efeito de exercício (P=0,015). Os níveis séricos de insulina não apresentaram efeito de tratamentos (P>0,1). Para triglicérides e VLDL-C não foi observado efeito de tratamentos (P>0,1). O CT, HDL-C e LDL-C apresentaram efeito de interação da inclusão de levedura e exercício (P<0,1). Para a produção de AGCC nas fezes com base na matéria original (MO) não houve efeito para os ácidos acético e butírico (P>0,1), no entanto, observou-se efeito da inclusão de levedura na produção do ácido propiônico (P=0,052). A quantificação de levedura viva nas fezes demonstrou efeito de interação da inclusão de levedura e exercício (P<0,001). Os resultados de pH apontaram efeito de interação da inclusão de levedura e exercício físico para as faixas de horário 1 (P=0,050) e 2 (P=0,080) e efeito de exercício para as faixas 3 (P<0,007) e 4 (P=0,001). Os resultados do perfil bacteriano nas fezes apontaram efeito de exercício para Fibrobacter succinogenes (P=0,083) e de interação da inclusão de levedura e exercício para Lactobacillus genus (P=0,020); não foi observado efeito de tratamentos para Ruminococcus flavenfaciens (P>0,1). Para os níveis séricos de endotoxinas, observou-se efeito de interação da inclusão de levedura e exercício (P=0,689). A FC dos animais em exercício demonstrou efeito de tempo durante o exercício (P<0,001). Para a FC de recuperação, foi possível observar interação da inclusão de levedura e exercício físico (P=0,020). A inclusão de levedura viva S. cerevisiae na dieta de equinos influencia a produção de AGCC propiônico. O exercício físico de baixa intensidade influencia os coeficientes de digestibilidade da MS, MO, FDN, FDA e PB, os níveis plasmáticos de glicose e a quantificação de Fibrobacter succinogenes nas fezes. A inclusão de levedura viva S. cerevisiae na dieta e o exercício físico influenciam o coeficiente de digestibilidade do EE, níveis séricos de HDL-C e LDL-C, a quantificação de levedura viva nas fezes, os níveis séricos de endotoxinas, o pH fecal, a quantificação de Lactobacillus genus nas fezes e a frequência cardíaca de recuperação pós-exercício. O tempo de exercício em cada velocidade influencia a frequência cardíaca dos animais durante o exercício físico / The aim of this study was to investigate the implications of supplementation with live yeast Saccharomyces cerevisiae and a low-intensity exercise training program on digestibility of dietary components (DM, CP, EE, NDF, ADF, MO and starch), the glycemic and insulin response, serum profile of triglycerides (TC), total cholesterol and fractions (HDL-C, LDL-C, VLDL-C), measure the production of short chain fatty acids (SCFA) butyric acid, acetic and propionic, quantify yeast live in feces, evaluating faecal pH, microbial population in faeces and serum levels of endotoxins, in addition to measuring the heart rate of the animals exercise and recovery. Ten horses in the Purebred Arabian race, castrated male, mean age 72 ± 7.5 months and average weight of 473 ± 34.75 kg were used, housed in individual pens, fed diet consisting of 2% of the BW in DM / day, divided into 0.75% multiparticulate commercial concentrate and 1.25% grass hay (Cynodon dactylon sp. cv Tifton -85). The treatments were divided into control (no addition of yeast) and supplemented (plus 15 g / day of live yeast Saccharomyces cerevisiae) in one phase and another phase with and without exercise, respectively. The experimental design was completely randomized in a factorial 2x2 and the data were submitted to analysis of variance by F test. For the digestibility coefficients it was observed exercise effect for the digestibility coefficients of DM, OM, NDF, ADF and PB (P<0.05). For the variable EE it was observed interaction of yeast inclusion and exercise (P<0.1). Starch no effect of treatments (P>0.1). For plasma glucose levels it was observed effect of exercise (P=0.015). Serum insulin levels showed no treatment effect (P>0.1). For triglycerides and VLDL-C was not observed treatment effect (P>0.1). The TC, HDL-C and LDL-C showed interaction effect of adding yeast and exercise (P<0.1). For the production of SCFA in the faeces based on this matter (OM) there was no effect for acetic and butyric acids (P>0.1), however, it was observed effect of including yeast in the production of propionic acid (P=0.052). The quantitation of live yeast in the feces demonstrated interaction effect of adding yeast and exercise (P<0.001). The pH values indicated yeast inclusion of the interaction effect and exercise for time range 1 (P=0.050) and 2 (P=0.080) and exercise effect to the time range 3 (P<0.007) and 4 (P=0.001). The results of the bacteriological profile in feces showed exercise effect to Fibrobacter succinogenes (P=0.083) and interaction of the yeast include the genus Lactobacillus and exercise (P=0.020). It wasnt observed for treatment effects Ruminococcus flavenfaciens (P>0.1). For serum levels of endotoxin it was observed interaction effect of adding yeast and exercise (P=0.689). The CF of animals in exercise demonstrated time effect during exercise (P<0.001). For CF recovery, it was observed interaction of yeast inclusion and exercise (P=0.020). The inclusion of live yeast S. cerevisiae in horse\'s diet influences propionic SCFA production. The low-intensity exercise influences the digestibility coefficients of DM, OM, NDF, ADF and PB, the plasma levels of glucose and quantification of F. succinogenes in the stool. The inclusion of live yeast S. cerevisiae in horses diet and exercise influence EE digestibility, serum levels of HDL-C and LDL-C, quantification of live yeast in the stool, serum levels of endotoxin, fecal pH and quantifying the genus Lactobacillus in stool and heart rate of post-exercise recovery. The exercise time at each speed influences the heart rate of the animals during exercise

Com o desenvolvimento e crescimento na produção equina, passou-se a utilizar o concentrado em substituição à parte da forragem na alimentação, principalmente por grãos de cereais ricos em amido, a fim de suprir as necessidades energéticas do animal. O amido não degradado no intestino delgado pode atingir o ceco e cólon, acarretando em mudanças na microbiota e consequentes distúrbios gastrointestinais. O objetivo desse estudo foi avaliar o efeito da suplementação de levedura Saccharomyces cerevisiae sobre a digestibilidade aparente, identificar as implicações nas respostas glicêmicas, insulinêmicas, valores de pH fecal e avaliar o nível de inclusão de levedura para cavalos alimentados com dietas de alta e baixa relação concentrado:volumoso. Foram utilizados oito animais da raça Mini-horse, machos, castrados, com idade média de 36 meses e peso médio inicial de 113±12 kg, em dois experimentos, sendo no experimento 1 utilizado dieta de baixa proporção concentrado:volumoso (BC), constituída de 30% concentrado comercial peletizado e 70% de feno de gramínea, e no experimento 2 utilizado dieta de alta proporção de concentrado:volumoso (AC), constituída de 70% concentrado comercial peletizado e 30% de feno de gramínea, cuja variação foi a inclusão de Saccharomyces cerevisiae, com níveis de 0g (controle), 10g, 20g, e 30g, de produto com concentração de 5x108 UFC/g. O delineamento experimental utilizado foi em quadrado latino duplo 4x4 com medidas repetidas no tempo e os dados obtidos foram submetidos à análise de variância e regressão polinomial com nível de significância de 5%. Para a dieta BC, não ocorreu diferenças na digestibilidade dos nutrientes e na resposta glicêmica e insulinêmica, contudo foi observado efeito quadrático (P=0,0087) para pH fecal, porém os valores permaneceram dentro do considerado ideal para o equilíbrio da microbióta. Com relação à dieta AC, foi observado efeito quadrático nos coeficientes de digestibilidade aparente da MO, FDN, FDA e PB (P=0,0023, P=0,0334, P=0,0238, P=0.0224), respectivamente, assim como para as concentrações plasmáticas de insulina (P=0,0012), no entanto, os valores encontrados não foram diferentes ao tratamento controle. Sendo assim, não foi possível avaliar o efeito da suplementação de levedura Saccharomyces cerevisiae para cavalos alimentados com dietas de baixa e alta relação concentrado:volumoso. / The development and growth in equine production increased the use of concentrate to replace part of the forage in the diet, mainly cereal grains rich in starch, in order to meet the energy needs of the animal. The starch that is not degraded in the small intestine can reach the cecum and colon, resulting in changes in the microbiota and consequently in gastrointestinal disturbance. The objective of this study was to evaluate the effect of supplementation of the yeast Saccharomyces cerevisiae on an apparent digestibility, identify the implications for glycemic and insulinemic responses and fecal pH values, and evaluate the level of inclusion of yeast into horses fed with high and low ratio concentrate: roughage diets. Eight Mini-horses were used: gelding, male, mainly 36- month old, and initial average weight of 113 ± 12 kg in two experiments. It was used in the experiment 1 a low ration concentrate: roughage diet (LC), consisting of 30% commercial concentrate pellets and 70% grass hay, and in the experiment 2, it was used a high ration concentrate: roughage diet (HC) consisting of 70% commercial concentrate pellets and 30% grass hay, which change was the inclusion of Saccharomyces cerevisiae, with levels of 0 g (control), 10g, 20g, and 30g of product concentration of 5x10 8 CFU/g. The experimental design was a double 4x4 Latin Square with repeated measures over the time and the data were subjected to analysis of variance and polynomial regression with a significance level of 5%. In the LC diet, there were no differences in nutrient digestibility, glycemic and insulin response, however it was observed a quadratic effect (P = 0.0087) for fecal pH, but values remained within the considered ideal for the balance of the microbiota. In the AC diet, a quadratic effect was observed in the apparent digestibility of OM, NDF, ADF and CP (P = 0.0023, P = 0.0334, P = 0.0238, P = 0.0224) respectively, as well as for plasma insulin concentrations (P = 0.0012), however, the values were not different to control. Therefore, it was not possible to evaluate the effect of supplementation of yeast Saccharomyces cerevisiae for horses fed with low and high ratio concentrate: roughage diets.

Influência do jejum alimentar, probióticos e antibiótico na população de enterobactérias, bactérias ácido lácticas, Bacillus e Salmonella sp. em cecos e papos de frangos de corte / Influence of feed withdrawal, probiotics and antibiotic on the enterobacteria, lactic acid bacteria, Salmonella and Bacillus populations in the crop and caeca of broiler chickens

No presente trabalho foram realizadas análises de enterobactérias e Salmonella em cecos e papos de frangos de corte criados sem reaproveitamento de cama e com baixa densidade, submetidos a períodos de jejum de 3, 6, 9, 12, 15 e 18 horas; também foram feitas análises das mesmas bactérias, incluindo esporos de Bacillus subtilis e bactérias ácido lácticas em papos e cecos de frangos de corte alimentados com probióticos (bactérias lácticas, Bokashi e esporos de B. subtilis) e antibiótico (bacitracina de zinco) nas rações. Não houve diferença significativa entre as populações de enterobactérias nos períodos de jejum analisados, e não foram isoladas cepas de Salmonella nos diferentes tratamentos. Houve diminuição significativa na população de enterobactérias nos cecos em relação ao grupo controle quando os frangos foram alimentados com bactérias ácido lácticas nas rações. Com Bokashi e antibiótico nas rações, a população de enterobactérias nos cecos foi maior que no grupo controle. Nos papos, não houve diferença significativa entre as populações de enterobactérias para os diferentes tratamentos, mas o tratamento com Bokashi foi o que obteve menor população das enterobactérias nesse habitat. As contagens de bactérias ácido lácticas e esporos de B. subtilis dos cecos e papos analisados não sofreram alteração significativa devido aos diferentes aditivos presentes nas rações, quando comparadas ao grupo controle. Novamente não foram isoladas cepas de Salmonella nos cecos e papos dos frangos analisados. A ausência de Salmonella pode ter explicação no modo de criação dos frangos, com baixa densidade (12 aves/m2) e sem reaproveitamento de cama, pois foi um resultado encontrado nas duas fases do trabalho. A diminuição de enterobactérias nos cecos por parte das bactérias ácido lácticas administradas na ração pode ter sido devido à exclusão competitiva, apesar da população dessas bactérias não ter aumentado significativamente neste habitat. O tratamento com administração de Bokashi obteve menor contagem de enterobactérias no papo possivelmente devido ao fato de ser um produto fermentado, com pH baixo, que poderia agir prontamente no papo. A administração de esporos de B. subtilis não surtiu efeito nas populações das bactérias analisadas nos cecos e papos dos frangos de corte. / This study analyzed the population of enterobacteria and Salmonella in the crop and caeca of broiler chickens submitted to 3, 6, 9, 12, 15 and 18 hours of feed withdrawal, and raised without reutilization of the litter, in low density. The same bacteria were analyzed, plus Bacillus subtilis spores and lactic acid bacteria, in the crop and caeca of broiler chickens fed rations with probiotics (lactic acid bacteria, Bokashi and B. subtilis spores) and antibiotic (zinc bacitracin). There were no significant differences between enterobacteria populations on the analyzed withdrawal periods, and Salmonella and antibiotic (zinc bacitracin). There were no significant differences between enterobacteria populations on the analyzed withdrawal periods, and B. subtilis spores populations from the caeca and crops of analyzed broiler chickens did not differ from the control group when the different additives were fed through the ration. Again there were no Salmonella isolates in the analyzed crop and caeca. The absence of Salmonella may have an explanation on the way that chickens were raised, with low density (12 birds/m2) and no litter reutilization, because this result was found on the two phases of the study. The lower caeca enterobacteria population found when the broilers were fed lactic acid bacteria may have been due to competitive exclusion, even though the lactic acid bacteria counting did not increase significantly in this habitat. Treatment with Bokashi administration had lower counting of enterobactérias in the crop, possibly due to the fact that Bokashi is a fermented product, with low pH, that could act readily in the crop. The administration of B. subtilis spores did not have any effect on the populations of the analyzed bacteria, in the crop and caeca of broiler chickens.

Desenvolvimento de uma formulação probiótica para a prevenção e tratamento da mastite em bovinos / Development of a probiotic formulation for the prevention and treatment of bovine mastitis

O Brasil é o quinto maior produtor de leite do mundo, entretanto sua produtividade é considerada baixa. Um dos motivos desta baixa produtividade é a alta incidência da mastite no rebanho. A mastite é a reação inflamatória da glândula mamária, causada principalmente pela invasão e ação de espécies de micro-organismos patogênicos. O principal tratamento consiste no uso de antibióticos de aplicação intramamária. Entretanto, o uso constante de antibióticos apresenta problemas graves como o surgimento de cepas de bactérias resistentes e a presença de resíduos destes medicamentos no leite. Para reduzir esses problemas, formas alternativas de prevenção e tratamento da mastite têm sido estudadas, como o uso de preparações contendo espécies probióticas. Pesquisas recentes têm demonstrado o potencial de algumas destas espécies no tratamento e prevenção da mastite bovina tendo em vista sua atividade inibitória sobre os patógenos responsáveis por esta infecção. Desta forma, a proposta deste estudo foi desenvolver e avaliar a estabilidade de uma preparação probiótica na forma de emulsão de aplicação intramamária contendo 108 UFC/mL de um \"pool\" constituído de cinco cepas de Lactobacillus (L. acidophilus ATCC 4356, L. casei ATCC 7469, L. fermentum ATCC 9338, L. plantarum ATCC 8014 e L. paracasei Nestlé ST11). Para tanto, 12 formulações de emulsão base foram desenvolvidas e avaliadas em ensaios preliminares de estabilidade (centrifugação e estresse térmico), sendo a formulação contendo monestearato de glicerila, lanolina anidra, óleo mineral, Tween 80® e Spam 80® selecionada para formulação da preparação probiótica. Adicionalmente, selecionou-se como agente conservante sorbato de potássio visando aumentar a vida útil da preparação probiótica. A preparação, assim desenvolvida, foi submetida ao teste de estabilidade físico-químico por 90 dias a 50°C ± 2°C. Observou-se que o aspecto da preparação se manteve estável, mas com redução nos parâmetros pH, densidade e aumento na viscosidade. A manutenção da viabilidade dos Lactobacillus na preparação foi estudada em um ensaio de 120 dias, sendo avaliada a influência dos fatores: temperatura de estocagem, adição de sacarose e sorbato de potássio. Observou-se influência significativa da temperatura demonstrando a importância da refrigeração na manutenção na viabilidade dos Lactobacillus. Verificou-se também que, sob refrigeração, a adição de sacarose promoveu redução de 0,5 ciclo logarítmico em relação a população inicial de células viáveis. Observou-se redução do pH da preparação probiótica avaliada nestas condições. Estes resultados permitem concluir que a refrigeração da preparação é indispensável para a manutenção da viabilidade das bactérias probióticas. A adição de 2% de sacarose à preparação, mantida sob refrigeração, se mostrou eficiente na manutenção da população de células viáveis ao final de 120 dias de ensaio, contendo 108 UFC/g, característica desejável para um produto probiótico. / Brazil is the world\'s fifth largest milk producer, though its productivity has been considered low. One of the reasons for this low productivity is the high incidence of mastitis in the herd. Mastitis is the inflammation of the mammary gland, mainly caused by the invasion and action of pathogenic microorganisms. The treatment is accomplished through the use of intramammary application of antibiotics. However, the constant use of antibiotics has serious problems such as the emergence of resistant strains of bacteria and the presence of residues of these drugs in milk. To reduce these problems, alternative means of prevention and treatment of mastitis has been studied as the use of preparations containing probiotic species. Recent studies have demonstrated the potential of some of these species in the treatment and prevention of bovine mastitis due to their inhibitory activity on pathogen species responsible for the infection. Thus, the purpose of this study was to develop and evaluate the stability of probiotic preparation in the form of intramammary application emulsion containing 108 CFU/mL of five Lactobacillus strains (L. acidophilus ATCC 4356, L. casei ATCC 7469 L. fermentum ATCC 9338, ATCC 8014, L. plantarum and L. paracasei ST11 Nestle). Therefore, 12 basic emulsion formulations were developed and evaluated in preliminary tests for stability (thermal stress and centrifugation), and only one was selected. Additionally, potassium sorbate was selected as preservative in order to increase the shelf life of the preparation. The developed preparation was subjected to study the physicochemical stability for 90 days at 50 ° C ± 2 ° C. The results showed that the aspect of the preparation remained stable, but decreasing in pH and density and increasing in viscosity. The maintenance of the viability of the lactobacillus in the preparation was studied in a 120-day test, and flowing factors were considered: storage temperature, sucrose addition and the potassium sorbate addition. It was observed that there was significant differences in the temperature factor demonstrating the importance of cooling in maintaining the viability of Lactobacillus. It was also observed that sucrose decreased by 0,5 log cycle compared to initial population of viable cells in the refrigerated preparation. Regarding the pH, it was noted a decrease in the different formulations evaluated. These results show that the cooling of the preparation is indispensable for maintaining the viability of probiotic bacteria. The addition of 2% sucrose to the preparation proved efficient with a slower decrease in viability maintaining the population of viable cells at the end of the 120 days of the assay.

Otimização das propriedades reológicas e sensoriais de iogurtes probióticos enriquecidos com proteínas lácteas / Optimization of the rheological properties of probiotic yoghurts supplemented with milk proteins

Este estudo teve como objetivo otimizar as propriedades reológicas dos iogurtes probióticos suplementados com leite em pó desnatado (SMP), concentrado proteico de soro (WPC) e caseinato de sódio (Na-Cn), usando desenho experimental tipo simplex centroide, para modelagem de misturas. O estudo das interações entre os ingredientes (proteína concentrada de soro, caseinato de sódio e leite em pó) incluiu sete experimentos sendo três com os ingredientes puros, dois correspondentes a misturas binárias e um para a uma mistura ternária. Em paralelo aos sete experimentos delineados, foi realizado ensaio controle com leite não suplementado. Os leites foram fermentados a 42 °C até pH 4,5 empregando-se as culturas Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus e Bifidus lactis. A cinética de acidificação foi determinada bem como as análises físico-químicas, contagem de bactérias viáveis e características reológicas dos iogurtes. Modelos foram ajustados para os resultados (respostas cinética, contagem de bactérias viáveis e parâmetros reológicos) através de três métodos de regressão (linear, quadrático e cúbico especial) e aplicados às misturas. Os resultados deste estudo mostraram que a fortificação da base de leite com proteína concentrada de soro, caseinato de sódio e leite em pó desnatado, para o mesmo nível de teor de proteína, afetou o perfil de acidificação, a contagem de bactérias viáveis e reologia de iogurtes probióticos. Além disso, foi possível estimar o perfil de acidificação e a contagem de bactérias no leite suplementado com proteínas do leite em pó, concentrado proteico de soro e caseinato de sódio por metodologia de superfície de resposta, bem como calcular aproximadamente as propriedades viscoelásticas dos iogurtes probióticos. Pode-se ressaltar que a otimização das propriedades reológicas dos iogurtes probióticos suplementados com proteínas do leite depende da fonte de proteína utilizada. Por fim, a substituição parcial de 55% de leite em pó desnatado por concentrado proteico de soro e caseinato de sódio simultaneamente poderá reforçar as propriedades reológicas dos iogurtes probióticos, considerando a cinética de acidificação e a contagem de bactérias viáveis. Os resultados foram validados ao longo da vida de prateleira dos produtos, confirmando os valores estimados pelos modelos para a cinética de acidificação, a contagem de bactérias viáveis e as propriedades viscoelásticas, além da avaliação sensorial e da análise de microestrutura. / This study aimed to optimize the rheological properties of probiotic yoghurts supplemented with skim milk powder, whey protein concentrate and sodium caseinate throughout an experimental design type centroid simplex for mixture modeling. It included seven trials: three with the pure ingredients, three corresponding to the binary mixtures and one to the ternary one. A control experiment was accomplished with non supplemented milk. Milk was fermented at 42°C until pH 4.5 by Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus, and Bifidus animalis subsp. lactis. The kinetics of acidification was followed as well as physicochemical analyses, enumeration of viable bacteria and rheological characterization of the yoghurts. Models were adjusted to the results (kinetic responses, counts of viable bacteria and rheological parameters) through three regression models (linear, quadratic and cubic special) applied to mixtures. The results showed that the addition of milk proteins affected acidification profile and counts, and enhanced rheological properties of probiotic yoghurt. The estimated results were validated during shelf-life and were confirmed by for the kinetics of acidification, bacterial counts of viable and viscoelastic properties in addition to sensory evaluation and analysis of microstructure.
