Garantia da qualidade de ensaios mecânicos de materiais metálicos

A implantação de Sistemas de Gestão da Qualidade em ambiente laboratorial vem se tornando uma necessidade frequente, devido à demanda dos clientes por resultados confiáveis e rastreáveis. No Brasil, a Coordenação Geral de Acreditação do Inmetro (Cgcre) é o órgão responsável pela acreditação de laboratórios segundo a NBR ISO/IEC 17025, sendo o Laboratório de Metalurgia Física (LAMEF) da Universidade Federal do Rio Grande do Sul acreditado para a realização de ensaios de materiais metálicos desde 2010. Em 2013, tendo passado por um processo de aumento de escopo de acreditação para os ensaios mecânicos, evidenciou-se o não atendimento ao item de garantia da qualidade da NBR ISO/IEC 17025 e da NIT-DICLA-026. Para atender aos requisitos, foram conduzidas análises através de Ensaios de Proficiência e por monitoramento interno da qualidade pelos métodos do Erro Normalizado e de Análise de Variância. Quando identificados desvios dos critérios de aceitação, ações corretivas foram tomadas, visando a melhoria contínua dos sistemas de medição. Os resultados do trabalho demonstraram que a implantação sistemática de métodos estatísticos para o monitoramento da qualidade dos ensaios foi fundamental para o processo de aumento do escopo de acreditação do LAMEF, o qual foi consolidado no início de 2014. / The implementation of Quality Management Systems in laboratory environment is being an increasing necessity, due to customer demand for reliable and traceable test results. The General Coordination for Accreditation of Inmetro (Cgcre/Inmetro) is the Brazilian body responsible for the accreditation of laboratories according to ISO/IEC 17025 standard, and the Physical Metallurgy Laboratory (LAMEF) from the Federal University of Rio Grande do Sul (UFRGS) has been accredited in performance of metallic materials tests since 2010. In 2013, during the process of accreditation scope extension on mechanical testing, there were identified nonconformities to the quality assurance requirements of ISO/IEC 17025 and NIT-DICLA-026. In order to comply with these requirements, studies have been carried out through Proficiency Testing and internal quality monitoring via Normalized Error and Analysis of Variance methods. Corrective actions were taken when deviations from the acceptance criteria were identified, aiming the measurement systems continuous improvement. The results of this work have demonstrated that the systematic implementation of statistical methods for monitoring the testing quality was critical to the process of LAMEF extension of scope, which was consolidated in the early 2014.

The Globalization of English: Its Impact on English Language Education in the Tertiary Education Sector in Taiwan

The overall agenda for the research reported here grew out of semi-structured interviews with senior educational managers from a tertiary educational institution in Taiwan. These managers raised a number of issues, including the changing profile of tertiary students, the changing nature of English curricula, the increasing need for English teaching staff to be adaptable, highly qualified and research-active, and the growing pressure on institutions to introduce English language proficiency benchmarking. Each of these issues can be related to the impact of globalization and, in particular, the impact of the globalization of English, on the education sector. Following a critical review of selected literature on the impact of globalization on the teaching and learning of English, each of these issues, as it affects the tertiary education sector in Taiwan, was explored. Analysis of the Taiwanese national curriculum guidelines for schools, strongly influenced by academics in the tertiary education sector, revealed a number of problems relating to a lack of proficiency benchmarking and a lack of coherence, consistency and transparency in some areas. These problems may be associated with the initial phase of transition from a grammar-based curriculum to a more communicatively-oriented, outcomes-centered one. Problems of a similar type were indicated in responses to questions relating to curriculum matters included in a questionnaire distributed to a sample of teachers of English in the tertiary sector. Among other things revealed by questionnaire responses was the fact that many survey participants had received no training in English teaching. The results of a C-test (one that was initially used in a major European study) taken by a sample of entry-level and exit-level Bachelors degree students indicated a wide variation in proficiency, with individual scores differing by as much as 64 percentage points in the case of exit-level students. Furthermore, there was a difference of almost 10 percentage points between the mean scores of students from two different institutions who had majored in English. These results indicate some of the difficulties that Taiwan faces in attempting to establish graduation proficiency benchmarking. -ii- C-test participants completed a background questionnaire, the responses indicating a generally positive attitude towards English-speaking people, a general willingness to use English in situations where there was the option of not doing so, and a strong tendency towards instrumental motivation. Although one of the factors that appeared to have a positive impact on C-test performance was time spent in an English-speaking country, fewer than 18% of respondents had done so. Although there appears to be considerable anxiety and uncertainty associated with the teaching of English at tertiary level in Taiwan, and some genuine cause for concern, there are also many positive indicators of future success. Teachers and educational managers are aware of the problems they currently face and appear determined to resolve them. Taiwanese academics are increasingly involved in language-related research and increasingly prepared to interrogate their own practices, and Taiwan, unlike some other countries in Asia, is moving towards graduation proficiency benchmarking.

The Development and Validation of a Spanish Elicited imitation Test of Oral Language Proficiency for the Missionary Training Center

The Missionary Training Center (MTC), affiliated with the Church of Jesus Christ of Latter-day Saints, needs a reliable and cost effective way to measure the oral language proficiency of missionaries learning Spanish. The MTC needed to measure incoming missionaries' Spanish language proficiency for training and classroom assignment as well as to provide exit measures of institutional progress. Oral proficiency interviews and semi-direct assessments require highly trained raters, which is costly and time-consuming. The Elicited Imitation (EI) test is a computerized, automated test that measures oral language proficiency by having the participant hear and repeat utterances of varying syllable length in the target language. It is economical, simple to administer, and rate. This dissertation outlined the process of creating and scoring an EI test for the MTC. Item Response Theory (IRT) was used to analyze a large bank of EI items. The best performing 43 items comprise the final version MTC Spanish EI test. Questions about what linguistic features (syllable length, grammatical difficulty) contribute to item difficulty were addressed. Regression analysis showed that syllable length predicted item difficulty, whereas grammar difficulty did not.

[pt] A participação de laboratórios clínicos em programas de ensaio de proficiência é uma importante ferramenta para gestão da qualidade. A maior parte das avaliações de desempenho de laboratórios clínicos por ensaio de proficiência se realiza com base no critério Z-Score, cujo valor designado é o consenso de resultados obtidos pelos participantes da rodada de proficiência, que é fortemente dependente da qualidade dos sistemas analíticos dos participantes. Utilizando uma base de dados fornecida por um provedor de ensaios de proficiência acreditado pelo INMETRO, este trabalho compara o desempenho de critérios para a avaliação da proficiência de laboratórios clínicos para medição de glicose. Consideraram-se não só critérios comumente utilizados, mas também novos critérios propostos que independem do consenso, baseados apenas em parâmetros associados ao valor designado pelo material de referência. As discrepâncias de classificação de desempenho dos laboratórios em função dos critérios de avaliação empregados indicaram inadequações da classificação baseada em critérios com parâmetros afetados pelo consenso entre participantes. O estudo também permitiu caracterizar o desempenho dos sistemas analíticos empregados pelos participantes, evidenciando-se uma discrepância significativa entre seus resultados. Os resultados do presente trabalho possibilitam analisar o impacto dos critérios utilizados no ensaio de proficiência para a adequada avaliação do desempenho dos laboratórios, contribuindo, assim, para a garantia da confiabilidade dos diagnósticos clínicos e condutas terapêuticas. / [en] The participation of clinical laboratories in proficiency testing programs is an important tool for quality management. Most evaluations of clinical laboratory performance by proficiency testing are carried out based on the Z-Score criterion; whose assigned value is the consensus of results obtained by the participants of the round, which is heavily dependent on the quality of analytical systems of participants. Using a database provided by a proficiency testing provider accredited by INMETRO, this paper compares the performance of criteria for the assessment of proficiency of clinical laboratories for measuring glucose. The analysis considered not only commonly used criteria, but also new proposed criteria that are independent of the consensus effect, based only on parameters associated with the assigned value defined by the reference material. The observed discrepancies in the performance rating of laboratories, according to the evaluation criteria employed, allowed the evidencing of inadequacies in the classifications based on criteria using parameters affected by the consensus among participants. The study also characterized the performance of analytical systems employed by the laboratories, evidencing a significant discrepancy between the results its. The present work allowed a better understanding of the impact of criteria applied in proficiency testing programs on the adequacy of evaluation of laboratories performance, thus contributing to ensuring the reliability of clinical diagnostics and therapeutical procedures.

Développement de méthodes de référence pour les biomarqueurs du bilan lipidique : application au contrôle qualité en biologie clinique / Development of reference methods for lipid profile biomarqueurs : application to quality controle in clinical chemistry

En biologie clinique, il est indispensable de disposer de mesures fiables et comparables dans le temps et entre les laboratoires afin de permettre un dépistage et un suivi appropriés des patients. Pour cela, il est indispensable d’établir leur traçabilité métrologique aux unités du système international notamment par des méthodes de référence primaires ou des matériaux de référence certifiés (MRC) d’ordre supérieur. Ces travaux de thèse ont consisté à développer et valider des méthodes de référence pour le cholestérol total, les triglycérides, le HDL-cholestérol et le LDL-cholestérol. Leur valeur ajoutée par rapport à une valeur consensuelle a été démontrée lors d’évaluations externes de la qualité. Elles ont également permis de certifier un MRC qui pourra être utilisé pour le contrôle qualité et/ou l’étalonnage des méthodes de routine. Nous avons montré que le MRC était commutable pour la plupart des méthodes de routine pour les différents biomarqueurs, ce qui a permis de l’utiliser pour évaluer leur justesse. Les méthodes de routine avaient généralement tendance à sous-estimer la concentration en triglycérides (en particulier aux valeurs basses) et à surestimer nettement la concentration de cholestérol total et de LDL-cholestérol (en particulier aux concentrations proches du seuil de décision clinique), ce qui se traduit par une augmentation du nombre de faux-positifs (patients traités à tort). Une approche de correction de non commutabilité a également été proposée afin de permettre l’utilisation de matériaux non commutables pour évaluer la justesse. Pour conclure, ces travaux ont démontré l’importance de disposer de méthodes de référence ainsi que de MRC commutables / Reliable measurements in medical biology are essential for early screening and appropriate follow-up of patients. Ensuring metrological traceability of clinical measurements to higher order reference methods or certified reference materials enables to obtain comparable results over time and between different laboratories that could use different methods to quantify the same biomarker.In this study, reference methods were developed and validated for lipid profile biomarkers (total cholesterol, triglycerides, HDL-C, and LDL-C). Their value added in proficiency testing schemes was demonstrated against consensus mean. They were also used to characterize a certified reference material (CRM) that may be used both as quality control and/or calibrator of field methods. The CRM was shown to be commutable for most field methods and lipid profile biomarkers, which proved it was suitable to assess trueness. Results of our multicenter study showed that field methods tend to underestimate triglycerides (particularly at low concentrations) and overestimate total cholesterol and LDL-C (especially around the clinical threshold), resulting in false positives and significant patient misclassifications. An approach of non-commutanility correction was also presented to allow trueness assessment with non-commutable samples. In conclusion, this work highlights the importance of using reference methods and also commutable CRM to rigorously assess accuracy of field methods used in clinical laboratories

Qualitat en l’anàlisi de drogues d’abús en cabell i en fluid oral

The use of hair and oral fluid for drugs of abuse testing has increased over the last years. For this reason, the assurance that results provided using these matrices are reliable and error-free is needed. The objective of this thesis is to develop tools to assess the quality of results provided by laboratories analysing drugs of abuse in hair and in oral fluid and to evaluate the effect on the quality of results of different actions carried out. For this reason, intercomparison exercises have been organized and some studies have been performed to develop appropriate quality control materials. Regarding the analysis of drugs of abuse in hair, nine different intercomparison exercises have been organized. The evaluation of qualitative and quantitative results reported by laboratories together with the study of the methodology used, has led to know the quality of the results, to identify the sources of error and to know the corrective actions that should be developed. Concerning the quality control material, these exercises have enabled to know the influence on the results of the type of hair used to perform the analysis. Concerning the analysis of drugs of abuse in oral fluid, a method has been developed to identify and quantify 6-monoacetyl morphine, morphine, codeine, cocaine and benzoylecgonine in oral fluid samples. On the other hand, stability studies of the main drugs of abuse in oral fluid have been done to establish the optimal preparation, transport and storage conditions, and finally, two intercomparison exercises have been conducted to know the performance of analytical laboratories when analysing drugs of abuse in oral fluid and to know the stability of some drugs of abuse in two commercial collection devices. / La creixent utilització del cabell i del fluid oral per a l’anàlisi de drogues d’abús, ha portat a la necessitat d’assegurar que els resultats obtinguts utilitzant aquestes matrius són fiables i lliures d’error. L’objectiu d’aquesta tesi és desenvolupar eines que permetin avaluar la qualitat dels resultats dels laboratoris que analitzen drogues d’abús en cabell i en fluid oral i avaluar l’efecte de diferents accions sobre la qualitat final dels resultats. Per això s’han organitzat exercicis interlaboratori i s’han realitzat estudis per desenvolupar material d’assaig adequat. Pel que fa a l’anàlisi de drogues d’abús en cabell, s’han realitzat nou exercicis interlaboratori. A través de l’avaluació dels resultats qualitatius i quantitatius informats pels laboratoris i mitjançant l’estudi de la metodologia emprada, ha estat possible conèixer la qualitat dels resultats obtinguts, detectar les fonts d’error i conèixer quines mesures correctives caldria desenvolupar. Pel que fa al material d’assaig, aquests exercicis han permès conèixer la influència que té, en els resultats, el tipus de cabell del qual es parteix per realitzar l’anàlisi. Pel que fa a l’anàlisi de drogues d’abús en fluid oral, s’ha desenvolupat i validat un mètode analític que ha permès identificar i quantificar 6-monoacetil morfina, morfina, codeïna, cocaïna i benzoïlecgonina en mostres de fluid oral. Per altra banda, s’han realitzat estudis d’estabilitat de les principals drogues en fluid oral que han permès establir les condicions òptimes de preparació, transport i conservació del material d’assaig i, per últim, s’han realitzat dos exercicis interlaboratori que han permès conèixer la qualitat dels resultats analítics obtinguts pels laboratoris i també l’estabilitat de les drogues en dos dispositius de recollida comercials.

Analýza procesu čtení v moderní spisovné arabštině v testových podmínkách / Exploring the Process of Reading in Modern Standard Arabic

(English) The dissertation presents the findings of a mixed method research project aimed at analysing the process of reading in Modern Standard Arabic. Think aloud method represents the main instrument for collecting qualitative data in the form of verbal reports that the students produce while taking a test of reading comprehension. The test is used as an instrument for distinguishing good and poor readers whose reading strategies are analysed using protocol analyses. The resulting codes of reading strategies are then categorized and their specificities are discussed at length.
