Sexuality and marriage : guidelines for the twenty-first century

The research commences by explaining the relevance and importance of marriage in the post-modern world although the post-modern era is known for disregarding pre-modern norms and customs. Marriage as traditionally known in the church is no exception. Communities no longer share the church’s strict rules when it comes to relationships. Church teaching about sex seems contradictory; on the one hand it regards sex as sinful, while on the other hand it preaches that it should be saved for the person you love (Berry 2005:15). Modern society’s expectations are very different from Biblical times. Unlike Biblical times where adolescents got married between the age of fourteen and eighteen, modern day couples rarely get married before they reach their mid-twenties. The implication of this is that the youth should live in abstinence for a decade or more although they are physically mature. The question is whether it is realistic of society to expect long-term couples not to have a sexual relationship. Modern society questions the fact that traditional marriage should be seen as the only moral and lawful option where a permanent love relationship between two people may exist. Throughout history sex was usually portrayed as something wrong, shameful, even sometimes as sin. “After sexual intercourse both man and woman had to bath, and they remained unclean until evening” (Lev. 15:18). The negative view on sexual intimacy continued in the early church, where they taught that sex is meant for procreation and not for enjoyment. Sex was always treated as something that should not be talked about, especially not in church. Although most of these views have been overcome in the last few decades the church is faced with new challenges in the present milieu such as sex before marriage, cohabitation and homosexuality. According to Van Eck (2007a:81) marriage as institution in modern society is in crisis. Additionally the guidance that the church provides on this matter might be in worse crisis, particularly the Church’s silence and lack of guidance regarding cohabiting relationships and pre-marital sex. The revolution of marriage is investigated to understand its development over the last two thousand years. Biblical text is studied in search of guidelines for young adults on the matters of sexuality and marriage. A look into modern day ethics will give us an idea of the challenges the Church face on this subject. The universal importance of marriage, the diversity and complexity of social and cultural circumstances, the needs within society and the church, and the quest for Scriptural moral guidance necessitates this research. This study seeks to investigate whether modern-day non-marital sexual relationships should be accepted in the Church and to suggest guidelines for young adults on the matters of sex and marriage in order to make Christian based meaningful life choices. The purpose of this study is to find Biblical guidelines in an attempt to find a link between spirituality and sexuality. In order to undo the sexuality device s knots, this study demonstrates that sexual desire is a dimension that cannot be reduced to the logic of representation. Rather it is beyond the identity politics and normative macropolitical games of the right. Desire battles insistently between: the individual and the collective; sex and the norm; bliss and love; and also I and the other . These battles form promiscuous zones. Therefore this study demonstrates two characters: the whores/prostitutes and the lovers. An empty and inconvenient sensation is experienced as we are in a zone of non-knowledge and non-recognition. This zone allows the expression of creative potential, mobilized exactly by the emptiness that occurs in the capitalist cultural regime. In the limits of the desire-sex-pleasure game zone, the concepts operate on the dynamics of affections, encounters and concrete experience. It is a game of sensations that desires sensitivity to tensions and paradoxes so that alternative ways for sexualities to vibrate can be reinvented. <p>This study aims to compare the sexual behaviour patterns of male elite student athletes, male student recreational sports (RSP) and male student non-athletes at the University of Botswana in relation to high-risk sexual behaviours. High-risk behaviors in the domain of sexual behaviour were investigated in relation to the determinants of risk behaviour such as multiple partnerships, condom use, and history of sexually transmitted infections, the partners past sex life, current sexual life, casual sexual partners and age of first exual experience. The purpose was to establish which group of the male students is more at risk of HIV infection due to its engagement in high-risk sexual behaviour. Data was collected by means of an anonymous self-report questionnaire. Participants included 235 male students (94 non-athletes, 92 athletes and 50 elite athletes) at the University of Botswana. Selfreport questionnaires were administered, which included items from the 2005 Youth Risk Surveillance System (YRBSS), Student Life Style questionnaire, as well as items from a questionnaire assessing knowledge and practice of safe sex amongst Rhodes University students (Simpson, 1996).</p>

Exploiting enzyme promiscuity for rational design

Enzymes are today well recognized in various industrial applications, being an important component in detergents, and catalysts in the production of agrochemicals, foods, pharmaceuticals, and fine chemicals. Their large use is mainly due to their high selectivity and environmental advantage, compared to traditional catalysts. Tools and techniques in molecular biology offer the possibility to screen the natural sources and engineer new enzyme activities which further increases their usefulness as catalysts, in a broader area. Although enzymes show high substrate and reaction selectivity many enzymes are today known to catalyze other reactions than their natural ones. This is called enzyme promiscuity. It has been suggested that enzyme promiscuity is Nature’s way to create diversity. Small changes in the protein sequence can give the enzyme new reaction specificity. In this thesis I will present how rational design, based on molecular modeling, can be used to explore enzyme promiscuity and to change the enzyme reaction specificity. The first part of this work describes how Candida antarctica lipase B (CALB), by a single point mutation, was mutated to give increased activity for aldol additions, Michael additions and epoxidations. The activities of these reactions were predicted by quantum chemical calculations, which suggested that a single-point mutant of CALB would catalyze these reactions. Hence, the active site of CALB, which consists of a catalytic triad (Ser, His, Asp) and an oxyanion hole, was targeted by site-directed mutagenesis and the nucleophilic serine was mutated for either glycine or alanine. Enzymes were expressed in Pichia pastoris and analyzed for activity of the different reactions. In the case of the aldol additions the best mutant showed a four-fold initial rate over the wild type enzyme, for hexanal. Also Michael additions and epoxidations were successfully catalyzed by this mutant. In the last part of this thesis, rational design of alanine racemase from Geobacillus stearothermophilus was performed in order to alter the enzyme specificity. Active protein was expressed in Escherichia coli and analyzed. The explored reaction was the conversion of alanine to pyruvate and 2-butanone to 2-butylamine. One of the mutants showed increased activity for transamination, compared to the wild type. / QC 20100929

Chemoenzymatic Resolution in Dynamic Systems : Screening, Classification and Asymmetric Synthesis

This  thesis  is  divided  into  four  parts,  all  centered  around  Constitutional Dynamic  Chemistry  (CDC)  and  Dynamic  Kinetic  Resolution  (DKR)  using biocatalysts for selective transformations, and their applications in screening of bioactive compounds, organic synthesis, and enzyme classification.    In  part  one,  an  introduction  to  CDC  and  DKR  is  presented,  illustrating  the basic  concepts,  practical  considerations  and  potential  applications  of  such dynamic systems, thus providing the background information for the studies in the following chapters.   In part two, Dynamic Systemic Resolution (DSR), a concept based on CDC is exemplified.  With  enzyme-catalyzed  transformations  as  external  selection pressure,  optimal  structures  can  be  selected  and  amplified  from  the  system. This  concept  is  expanded  to  various  types  of  dynamic  systems  containing single, double cascade/parallel, and multiple reversible reactions. In addition, the  substrate  selectivity  and  catalytic  promiscuity  of  target  enzymes  are  also investigated.   In   part   three,   DKR   protocols   using   reversible   reactions   for   substrate racemizations  are  illustrated.  Biocatalysts  are  here  employed  for  asymmetric transformations,  resulting  in  efficient  synthetic  pathways  for  enantioenriched organic compounds.   Part  four  demonstrates  two  unique  applications  of  CDC:  one  resulting  in enzyme  classification  by  use  of  pattern  recognition  methodology;  the  other involving  enzyme  self-inhibition  through  in  situ  transformation  of  stealth inhibitors employing the catalytic activity of the target enzyme. / <p>QC 20130614</p>

In 1996 Francois Bloemhof's teenage fiction, Slinger-slinger, was a prize-winner in the beginners' category of the Sanlam competition for fiction for youth. The book was acclaimed because of the open manner in which the author wrote about sensitive themes such as hetero- and homosexuality, promiscuity, divorce, incest, tense family relationships, and drug and alcohol abuse. The main theme of Slinger-slinger is teenage sexual aspects, such as the sexual awakening and identity of teenagers. In 1998 a sensation was caused in the media as upset parents, teachers and other adults protested against the inclusion of Slinger-slinger in the ATKV's Children's Book Award, category 13-15 years of age. Two conflicting opinions about Slingerslinger could be distinguished: one group was of the opinion that the sexual theme and strong language were not suited for teenage fiction, while the other group was of the opinion that teenagers needed to read about realistic themes to prepare them for the real world. No comments were published about the opinions of the supposed readers, namely teenagers. Steenberg (1988:168-172), Van der Westhuizen (1989:5-7) and Van Zyl (1990:17) wrote that teenagers read to fulfil their needs. Teenagers' reading needs are directly connected to their emotional, cognitive and physical developmental stages. The aim of the study The aim of this study is firstly to evaluate Slinger-slinger according to the elements of the reader study model to determine whether it addresses the reading needs and interests of teenage readers; and secondly to ascertain whether teenagers liked Slinger-slinger. Research procedures and methodology A study of the literature was done in order to determine the reading needs and developmental tasks of teenagers, and the role that teenage fiction can play in their development to adulthood. Together with this background study, an evaluation model, containing story elements viewed from a reader study perspective, as well as criteria for the evaluation of the handling of sexual aspects in teenage fiction, was developed to determine whether Slinger-slinger is successful teenage fiction. In the empirical study the above-mentioned evaluation model for teenage fiction were applied to Slinger-slinger. In the second part of the empirical study a questionnaire was compiled to obtain the opinions of teenage readers about the success of Slinger-slinger as a whole, as well as the success of the handling of the sexual aspects. The questionnaire also served to obtain information about the reading needs of teenage readers in regard to realistic teenage fiction and teenage fiction with a sexual theme. As triagens enzimáticas de micro-organismos isolados de água de drenagem de mina de prospecção de cobre permitiram a identificação de hidrolases e monoxigenases, sendo que as cepas de bactérias que apresentaram as melhores atividades oxidativas mostraram-se promissoras para oxidação de diversas classes de sulfetos, em ensaios de biocatálise convencional. A atuação promíscua de lipases foi avaliada frente a oxidação de cetonas cíclicas em meio orgânico, e as interações intermoleculares deste sistema foram estudadas por RMN, onde foi observado características de um sistema organizado em micelas reversas. A partir destas evidências, foi proposto que a condição necessária para a reação enzimática é o confinamento da enzima num ambiente aquoso restrito limitado por uma barreira molecular de ácido graxo. Finalmente a inibição da atividade enzimática de proteína tirosina fosfatases foi avaliada com implementação de uma metodologia fluorimétrica rápida e eficiente. Logo, este trabalho apresentou uma visão ampla no campo da enzimologia com o emprego de biocatalisadores, sejam eles enzimas isoladas ou células íntegras, em estudos relacionados a aspectos químicos, bioquímicos e terapêuticos das enzimas / Abstract: In this thesis the enzymatic versatility was evaluated in microorganisms selecting specific biocatalysts, in studies of intermolecular interactions of complex systems focusing enzyme promiscuity, and last in enzyme inhibition related to the occurrence of various diseases. The enzymatic screening of microorganisms isolated from copper mine water drainage led to the identification of hydrolases and monooxygenases, and bacterial strains with relevant oxidative activities were further investigated in larger scale oxidation of several sulfides. The promiscuous activity of lipases was evaluated in organic medium catalyzing the oxidation of cyclic ketones. The supramolecular interactions involved in these reactions were studied by NMR, revealing the presence of reverse micelles in a highly organized system. Therefore confinement of the enzyme in a restricted aqueous environment limited by a narrow molecular barrier of fatty acids in an organic medium is the necessary condition for this enzymatic reaction to occur. Finally, inhibition of enzymatic activity of protein tyrosine phosphatases was assessed with the implementation of a quick and efficient fluorimetric method.
