As doenças neurodegenerativas (DN), incluindo doença de Alzheimer e doença de Parkinson, estão entre as principais causas de mortalidade e morbidade nos países ocidentais. Não há ainda um tratamento definitivo para estas neuropatias, mas os estudos têm apontado para mecanismos comuns de toxicidade, que incluem disfunção mitocondrial, estresse oxidativo e apoptose. Assim, as mitocôndrias constituem alvos importantes para futuras estratégias de neuroproteção visando tratar, prevenir ou retardar a neurodegeneração. A biodiversidade da fauna brasileira representa uma fonte promissora e ainda pouco explorada de novas moléculas com (i) atividade neuroprotetora e, portanto, potencial para originar novos fármacos para o tratamento destas doenças; ou ainda com (ii) atividade neurotóxica, podendo ser utilizadas como ferramentas de estudo de mecanismos de neurotoxicidade. O presente estudo teve por objetivo investigar o possível potencial neuroprotetor e/ou neurotóxico de peptídeos de baixo peso molecular, obtidos a partir do veneno de diferentes espécies de serpentes do gênero Bothrops por meio da investigação dos efeitos destes compostos na função mitocondrial de cérebro de rato. Duas das frações estudadas (obtidas a partir do veneno da B. atrox e da B. jararaca) apresentaram um interessante potencial protetor contra o intumescimento osmótico mitocondrial e a produção de espécies reativas de oxigênio (ERO), eventos associados à transição de permeabilidade mitocondrial e à morte celular. Por outro lado, outra fração, obtida a partir do veneno da B. pirajai, apresentou potencial neurotóxico. Estes achados podem ser úteis para estudos mecanísticos e também no estabelecimento de futuras estratégias de tratamento das doenças neurodegenerativas, tendo as mitocôndrias como alvos terapêuticos (terapia alvo). / The neurodegenerative diseases, including Alzheimer\'s and Parkinson\'s diseases, are among the main causes of morbidity and mortality in Western countries. A definitive treatment for these neuropathies has not yet been found, but studies have indicated common mechanisms of toxicity, including mitochondrial dysfunction, oxidative stress and apoptosis. Therefore, mitochondria represent important targets for the future neuroprotective strategies aimed to treat, prevent or delay the neurodegeneration. The Brazilian fauna biodiversity represents a promising and under explored source of new molecules with (i) neuroprotective activity and potential to originate new drugs for the treatment of these diseases; or yet, with (ii) neurotoxic activity, representing tools to study neurotoxicity mechanisms. The present study aimed to investigate the possible neuroprotective and/or neurotoxic potential of low-molecular-mass peptides extracted from the venom of different species of Bothrops genus snakes by investigating their effects on rat brain mitochondrial function. Two of the studied fractions (from B. atrox and B. jararaca venoms) presented an interesting protective potential against both the mitochondrial swelling and reactive oxygen species (ROS) production, events associated with mitochondrial permeability transition and cell death. On the other hand, other fraction (from B. pirajai venom) presented a neurotoxic potential. These findings might be useful for mechanistic studies and also for the establishment of future strategies of neurodegenerative diseases treatment, using mitochondria as therapeutic targets (targeted therapy).

Fatores prognósticos por ocorrência de necrose e abscesso no envenenamento por serpente Bothrops jararaca / Prognostic factors for occurrence of necrosis and abscesses in Bothrops jararaca snake poisoningRibeiro, Lindioneza Adriano 20 June 1996 (has links)
No Brasil, ocorrem anualmente mais de 20.000 acidentes ofídicos, sendo mais de 80 por cento por espécies do gênero Bothrops. Envenenamentos por serpentes desse gênero raramente causam óbito mas, muitas vezes, levam a necrose e infecção secundária, que podem determinar a perda de um membro acometido ou de segmento desse. A dose de soro antibotrópico utilizada para o seu tratamento é baseada na gravidade do envenenamento ao exame inicial. Vários fatores têm sido associados a essa gravidade, mas poucos são os dados científicos e nenhuma avaliação foi feita para acidentes comprovadamente causados por B. jararaca. O presente estudo teve como objetivo identificar: as variáveis associadas à ocorrência de necrose; as variáveis asociadas à ocorrência de abscesso; os fatores prognósticos independentes para a ocorrência de necrose; os fatores prognósticos independentes para a ocorrência de abscesso. Foram analisados no presente estudo 779 casos de acidentes por B. jararaca atendidos no Hospital Vital Brazil do Instituto Butantan, no período de 1982 a 1990. Os dados foram levantados a partir de prontuários médicos arquivados nesse hospital. Os casos que apresentaram necrose e os que evoluiram para abscesso, foram comparados no que se refere a uma série de variáveis, respectivamente, com aqueles que não apresentaram necrose e não evoluiram para abscesso. Correlacionaram-se estatisticamente à análise bivariada com necrose e abscesso (p < 0,05) o acidente por serpentes de grande porte (> 60cm), a picada em perna, o uso de torniquete, a presença de dor, edema, equimose, bolha, choque, sangramento fora da região da picada. Acidentes em determinados meses do ano, picadas nos dedos da mão e alteração da coagulação sanguínea só se correlacionaram com necrose (p < 0,05). Mostraram-se fatores prognósticos independentes para necrose à análise multivariada: tamanho da serpente, conforme diferentes faixas de comprimento, 60 - 80cm, 80 - lOOcm, 100 - 140cm; mês de ocorrência; acidentes com pessoas do sexo feminino; picadas no dedo da mão, e na perna; uso de torniquete; ocorrência de sangramento. Mostraram-se fatores prognósticos independentes para abscesso, à análise multi variada: tamanho da serpente, conforme diferentes faixas de comprimento 60-80cm, 80- lOOcm, 100-140cm; mês de ocorrência; acidentes com pessoas do sexo feminino; picada na perna; alteração da coagulação sanguínea; ocorrência de sangramento. Conclui-se que, as variáveis relacionadas com necrose e aquelas relacionadas com abscesso, os fatores prognósticos independentes para necrose e aqueles para abscesso, são os apresentados acima e que o porte de B. jararaca é o fator prognóstico independente mais importante para necrose e, principalmente, para abscesso, que são tanto mais freqüentes quanto maior o comprimento da serpente. / There is an annual incidence of over 20 000 snakebite cases in Brazil; of these over 80 per cent are caused by Bothrops species. Bothropic envenoming commonly causes necrosis and secondary infection which may lead to loss of part or of all the bitten limb. Fortunately fatalities are rare. During treatment, the dose of antivenom administered is based on the clinical severity of the envenoming. Several factors have been shown to be associated with the severity of the envenoming, however data are scarce and no such studies on Bothrops jararaca exist. The objective of this study was to identify the following: the variables associated with necrosis; the variables associated with abscess formation; the independent prognostic factors of necrosis; the independent prognostic factors of abscess formation. The study analysed 779 B. jararaca envenoming cases admitted from the Hospital Vital Brazil of the Instituto Butanatan in the period from 1982 to 1990. Data were taken from medical records in this hospital. Occurrence of necrosis or of abscesses was independently compared, in the light of a series of specific variables, with cases in which, necrosis or abscesses did not occur. Occurrence of necrosis and of abscess (p<0,05) was correlated by bivariable statistical analysis with the following: bites caused by large-sized snakes (>60cm in length); bites on the leg; use of tourniquet; presence of pain, oedema, equimosis, blisters, shock; and systemic bleeding. Occurrence of necrosis (p<0.05) was only correlated with bites on the fingers, impaired blood coagulability, and with accidents grouped according to the month of the year. The following were shown, on multivariate analysis, to be independent prognostic factors for necrosis: size of snake (grouped according to length 60-80cm/ 80- l00cm/ 100-140cm); month in which bite occured; female sex of the victim; bites on fingers and legs; use of tourniquet; and the presence of haemmorrhage. The following were shown, on multivariate analysis, to be independent prognostic indicators for abscess formation: size of snake (grouped according to length 60-80cm/ 80-l00cm/ 100-140cm); month in which bite occured; female sex of the victim; bites on the leg; impaired blood coagulability; and the presence of haemmorrhage. It is concluded that the forementioned are: associated factors of necrosis and abscess formation; and the independent prognostic factors of necrosis; and independent prognostic factors of abscess. The size of B. jararaca responsible for the bite was shown to be the most important independent prognostic factor of necrosis and, even more so, of abscess.

Monte Carlo Simulations of Bowing Effects Using Realistic Fuel Data in Nuclear Fuel AssembliesWestlund, Marcus January 2019 (has links)
Deformations of nuclear fuel assemblies have been observed in nuclear power plants since the mid-90s. Such deformations are generally called bowing effects. Fuel assemblies under high irradiation undergo growth and creep induced by high loading forces and low skeleton stiffness of the assemblies which gives permanent deformations and modifies moderation regions. Hence, giving an unpredicted neutron flux spectrum, power distribution, and isotopic concentrations in the burnt fuel. The aim of this thesis is to study the effects of local fuel bowing in terms of power distribution and isotopic composition changes through simulations of the reactor core. The reactor is simulated with realistic bowing maps and previous deterministically simulated realistic fuel data from a present reactor by deploying the Monte Carlo method using the nuclear reactor code Serpent 2. Two subparts of a full reactor core with fuel from separate fuel cycles are investigated in 2D using burnup. To quantify the impact of the bowing, the change in power distribution and the induced isotopic composition change are calculated by a relative difference between a nominal case and a simulation with perturbed fuel assemblies. The results are presented in colormaps, for visualization. The isotopic composition for U235, U238, Pu239, Nd148, and Cm244 are investigated. Also, statistical uncertainty estimations in the composition of the depleted fuel are done by multiple calculations of the same geometry while changing the seed of random variables in the Monte Carlo calculation. The mean value and the standard deviation in the mass density of U235 and Pu239 are calculated for two pins together with histograms with a normal fit for each case to clarify the mathematical distribution of the calculations. The simulations performed in this thesis have detected clear impacts of the reactor behavior in terms of power distribution and isotopic composition in the burnt fuel introduced by the bowing. Assembly perturbations of about 10 mm may locally introduce a 10 % relative difference in power density and U235 content between the nominal and the bowed case at 15 MWd/kgU burnup. The power and the isotopic composition changes agree with expectations from the bowing maps.

Serpents of Empire : moral encounters in natural history, c.1780-1870Hall, James Robert January 2019 (has links)
This dissertation examines encounters between humans and snakes from the 1780s to the 1860s, principally focusing upon Britain and British India, to reassess the production and circulation of natural historical knowledge. Serpents were at once familiar and ambiguous in nineteenth-century Britain and its empire, present at every level of society through Scripture, works of natural history, and imperial print culture. They appeared across literary genres - in works of art, as dead specimens in museums, and living attractions in shows and menageries - and their material and figurative presence in London was dependent upon British imperial networks. Snakes loomed disproportionately large in the imperial imaginary, where they were entangled in a discourse of difference. The practices of the natural history of snakes were harnessed to personal ambition and colonial exigencies. By analyzing scientific books and papers, newspapers and periodicals, taxidermy and cartoons, travel accounts, and government archives from Britain and India, this study provides a connected account of how snakes were collected, transported, described, experimented with, and used for a variety of ends. Following an animal around, whether as material, textual, or visual representation, reveals a more comprehensive picture of how people engaged with animals in the nineteenth century, not confined by disciplinary or institutional boundaries at a time when these were being constructed. The cultural and emotive power of snakes makes visible the heterogeneous nature of those contributing to the production of natural historical knowledge. This thesis shows how the moral character of snakes was implicated in how they were encountered and understood by a range of actors, from museum naturalists to imperial agents, and Indian snake-charmers to working-class visitors to the zoo. The chapters examine different but overlapping modes of encounter with snakes: collecting, preserving, and presenting them in museum settings; the imbrication of anthropocentric concerns in attempts to classify and anatomize them; the mechanisms and motivations behind attempts to produce authoritative 'useful knowledge' incorporating vivisectional experiments in the Madras Presidency in the late eighteenth century; Orientalist representations of non-European interactions with snakes in nascent print culture; and the emotional economy of educational displays of living snakes in metropolitan Britain, especially with the emergence of new spaces for natural history, notably the first reptile house at the Zoological Gardens in Regent's Park. The approach brings together insights from from history of science, animal history, and new imperial histories to recover an affective dimension of natural history in imperial encounters.

A conventional army is required to meet many needs and face many threats. However, when conventional warfare methods do not make the cut, Special Operations and Special Forces become a necessity. However, there exists few theories that sufficiently explain how Special Forces units around the world accomplishes these Special Operations. One of the few researches who has developed a theory regarding Special Operations is William H. McRaven with his theory of relative superiority. McRaven´s theory proposes that there are six principles which determine the success of a Special Forces unit´s ability to plan, prepare and execute a Special Operation. The aim of the study is to understand McRaven´s theory´s explanatory power in explaining the results of Operation Neptune Spear and Operation Gothic Serpent. This study concludes that Operation Neptune spear attained almost all of the required principles. However, Operation Gothic serpent lacked half of the required principles. In order to create a favourable relation, one must create synergy between the principles in order to achieve relative superiority. The study concludes by illustrating that McRaven´s theory´s explanatory power increased.

An investigation of fuel cycles and material flows for a lead-cooled fast reactor using the Monte Carlo code SerpentMoberg, Kristina January 2012 (has links)
The Monte Carlo code Serpent has been used to model the material flows andisotope compositions for a lead cooled fast reactor. The demonstration sized trainingreactor ELECTRA was chosen for the investigation, and different fuel cycle scenarioswere studied. The scenarios differed in operation length (3 months, 1 year or 5years) and recycling technique (single and double PUREX or GANEX). The simulations gave detailed information on the changes of the isotope composition,activity and decay heat. The analysis of the generated waste also showed that thechoice of recycling method had great impact on the final storage time of the wastefrom the reprocessing. Performing double GANEX recycling, as compared to singlePUREX, reduced the storage time by a factor of about 3500. The results can be used for future work related to even more detailed studies ofmaterial flows and for designing an appropriate safeguards system.

Performance differences in encryption software versus storage devicesOlsson, Robin January 2012 (has links)
This thesis looked at three encryption applications that all use the symmetric encryption algorithms AES, Twofish and Serpent but differ in their implementation and how this difference would illustrate itself in performance benchmarks depending on the type of storage device that they were used on. Three mechanical hard drives and one solid state drive were used in the performance benchmarks which measured a variety of different disk operations across the three encryption applications and their algorithms. From the benchmarks performance charts were produced which showed that DiskCryptor had the best performance when using a solid state drive and that TrueCrypt had the best performance when using mechanical hard drives. By choosing DiskCryptor as the encryption application when using a solid state drive a performance increase of 38.9% compared to BestCrypt and 28.4% compared to TrueCrypt was achieve when using the AES algorithm. It was also shown that Twofish was overall the best performing algorithm. The primary conclusion that can be drawn from this thesis is that it is important to choose the right encryption application depending on the type of storage device used in order to get the best performance possible.

Operation Gothic Serpent och Kilcullens upprorsbekämpningsteori : En teoriprövande fallstudie på Task Force Rangers insatser i Mogadishu 1993Broman, Carl January 2014 (has links)
Den tredje och fjärde oktober 1993 sköt somaliska rebeller ner två amerikanska Black Hawk-helikoptrar i Mogadishu, Somalia. Dels på grund av ett bristande doktrinärt agerande resulterade det i en 14 timmar lång strid om att överhuvudtaget överleva och många skadades eller omkom. Inte långt därefter lämnade FN och USA Somalia och insatsen sågs som ett militärt misslyckande. Insatsen i Somalia har påverkat amerikanarnas syn på utrikespolitik och den visar tydliga exempel på den komplexitet som counterinsurgency-operationer för med sig. Uppsatsen är en teoriprövande fallstudie där David Kilcullens teorier om upprorsbekämpning prövas på Specialförbandsstyrkan Task Force Rangers insats i Mogadishu, Somalia. Uppsatsen syftar inte till att pröva huruvida Kilcullens teorier fungerar eller ej, utan till att pröva hur och i vilken utsträckning Task Force Rangers medel och metoder överensstämmer med vad Kilcullen förespråkar. Resultatet av denna studie visar att förekomsten av Kilcullens teorier i vissa fall överensstämmer men överlag är väldigt få. Bland annat på grund av komplexiteten i det somaliska samhället och dess kultur. Vidare visar arbetet att den största bidragande faktorn till utfallet av insatsen beror på det bristfälliga underrättelsearbetet som gjordes av Task Force Ranger under den berörda tidsperioden.
