Análise da proteína hnRNP K nas linhagens celulares NB4 e NB4-R2 de leucemia promielocítica aguda com ênfase na patogênese e na diferenciação celular pelo ácido all-trans retinoico / Analysis of hnRNPK protein in cell lines NB4 and NB4-R2 of acute promyelocytic leukemia with emphasis in pathogenesis and cell differentiation by all-trans retinoic acid

A ribonucleoproteína heterogênea nuclear K (hnRNP K) e o inibidor endógeno da fosfatase 2A (SET) são superexpressos e propostos como marcadores prognósticos em leucemia mieloide aguda e crônica. O objetivo do estudo foi caracterizar a participação das proteínas hnRNP K e SET na leucemogênese da leucemia promielocítica aguda (LPA), assim como na diferenciação celular induzida pelo ácido all-trans retinóico (ATRA). Os resultados iniciais de qRT-PCR demonstram que os níveis de RNAm de HNRNPK e SET estão aumentados em pacientes ao diagnóstico de LPA em comparação com amostras de indivíduos saudáveis e diminuem após indução e durante a manutenção do tratamento. Os resultados foram validados por Western blot, sugerindo hnRNP K e SET como marcadores diagnóstico e de resposta ao tratamento. O knockdown de hnRNP K e SET foi realizado em células de LPA sensível, NB4, e resistente ao ATRA, NB4-R2, utilizando RNA de interferência. Ambas as proteínas também foram testadas como alvo terapêutico com a utilização de inibidores de hnRNP K (U0126) e SET (OP449 e FTY720). A diminuição de hnRNP K nas células levou ao aumento da diferenciação celular granulocítica em ambas as células, principalmente na presença de ATRA, e portanto, foi capaz de reverter o fenótipo de resistência ao ATRA das células NB4-R2. O knockdown de hnRNP K, assim como o tratamento com U0126, levou a perda de viabilidade dessas células por indução de apoptose acompanhada da clivagem da proteína SET. O knockdown de SET em células LPA confirmou que a indução de apoptose em células com knockdown de hnRNP K ocorreu por clivagem e não pela diminuição da proteína SET nas células. Além disso, demonstrou também que SET prejudica a atuação do ATRA no processo de diferenciação celular. O modelo in vivo utilizando transplante de NB4-R2 em camundongos nude confirmou que o trióxido de arsênico (ATO) combinado a U0126 tem um maior potencial contra a progressão tumoral quando comparado ao tratamento isolado com ATO. FTY720 e OP449 tiveram efeito anti-leucêmico significativo com redução da viabilidade celular. Quando em associação, FTY720 e OP449, apresentaram efeito sinérgico significativo em NB4-R2 (CID<0,7). Concluímos que a superexpressão de hnRNP K e SET contribui para o bloqueio da diferenciação celular em promielócitos e prejudicam a atuação do ATRA no tratamento da LPA e, portanto, hnRNPK em associação com a proteína SET representam alvo terapêutico em potencial para terapia anti-leucêmica da LPA, principalmente para pacientes resistentes ao ATRA / Heterogeneous nuclear ribonucleoprotein K (hnRNP K) and endogenous inhibitor of phosphatase 2A (SET) are overexpressed and proposed as prognostic markers in acute and chronic myeloid leukemia. The study aim was to characterize the hnRNP K and SET proteins involvement in acute promyelocytic leukemia leukemia (APL) leukemogenesis as well as all-trans retinoic acid (ATRA) induced cell differentiation. Initial qRT-PCR results demonstrate that HNRNPK and SET mRNA levels are increased in patients diagnosed with APL compare to samples from healthy donors and decrease after induction and during maintenance of treatment. The results were validated by Western blot, suggesting hnRNP K and SET as diagnostic and response to treatment markers. The knockdown of hnRNP K and SET was performed on sensitive, NB4, and ATRA-resistant, NB4-R2, LPA cells using interfering RNA. Both proteins were also tested as a therapeutic target with a use of hnRNP K (U0126) and SET inhibitors (OP449 and FTY720). The decrease of hnRNP K in cells led to increased granulocyte cell differentiation in both cells, especially in the presence of ATRA, and thus was able to reverse the NB4-R2 cells resistance to ATRA phenotype. The hnRNP K knockdown, as well as the treatment with U0126, had a loss of cell viability by induction of apoptosis accompanied by cleavage of the SET protein. The SET knockdown in APL cells confirmed that an induction of apoptosis in cells with hnRNP K knockdown occurred by cleavage and not by the SET protein decrease in the cells. Furthermore, it has also shown that SET impairs the ATRA\'s performance in the cellular differentiation process. The in vivo model using NB4-R2 transplant in nude mice confirmed that arsenic trioxide (ATO) combined with U0126 has a greater potential against tumor progression compared to the treatment isolated with ATO. FTY720 and OP449 have significant anti-leukemic effect reducing cell viability. When in combination, FTY720 and OP449, they had a significant synergistic effect on NB4-R2 (CDI <0.7). We conclude that overexpression of hnRNP K and SET contributes to block cell differentiation in promyelocytes and impair the performance of ATRA in the treatment of APL and therefore hnRNPK in association with a SET protein represent a potential therapeutic target for anti-leukemic therapy of APL, mainly for patients resistant to ATRA

A Combinatorial Miscellany: Antipodes, Parking Cars, and Descent Set Powers

In this dissertation we first introduce an extension of the notion of parking functions to cars of different sizes. We prove a product formula for the number of such sequences and provide a refinement using a multi-parameter extension of the Abel--Rothe polynomial. Next, we study the incidence Hopf algebra on the noncrossing partition lattice. We demonstrate a bijection between the terms in the canceled chain decomposition of its antipode and noncrossing hypertrees. Thirdly, we analyze the sum of the 𝑟th powers of the descent set statistic on permutations and how many small prime factors occur in these numbers. These results depend upon the base 𝑝 expansion of both the dimension and the power of these statistics. Finally, we inspect the ƒ-vector of the descent polytope DPv, proving a maximization result using an analogue of the boustrophedon transform.

As a capstone, we hope to introduce a new large cardinal notion and give a similar characterization theorem of this new notion. Our new notion of near strong compactness was inspired by the similar notion of near supercompactness, due to Jason Schanker.

Detta examensarbete har utförts vid Saab Dynamics AB(SBD) i Karlskoga med syftet att studera gamla konstruktioner av set-back-generatorer. En set-back-generator (SBG) ska ge momentan energi vid utskjutning av en projektil genom att en magnet rör sig genom en spole. SBG har funnits länge men har bedömts ge för lite energi för att kunna driva elektronik. Men nya typer av magneter har potential att öka energiutbytet väsentligt. SBD har ett antal äldre SBG:er och arbetet har varit att utifrån dessa undersöka om det går att utvinna mer energi genom ett utbyte av magnet. Andra parametrar som är viktiga för en SBG:s funktion har också studerats och testats i olika konfigurationer i hopp om ytterligare förbättringar. Förväntade resultat har sedan analyserats och jämförts med mätresultat. Med detta som grund har rekommendationer vid en ny konstruktion av SBG levererats. / This bachelor thesis has been performed at Saab Dynamics AB (SBD) in Karlskoga, with the purpose to examine old designs of set-back-generators. A set-back-generator (SBG) shall provide instantaneous energy during firing of a projectile by a magnet moving through a coil. Previous designs of SBG:s is not providing enough energy to power electronics. New types of magnets have the potential to increase the energy yield significantly. SBD has a number of older SBG's and the work is based on these to examine the possibility to extract more energy by exchanging magnet. Other important parameters for an SBG's function has also been studied and tested in different configurations in the hope of further improvements. Expected results has then been analyzed and compared with the measurement results. On this basis suggestions and recommendations on a new SBG design has been delivered.

The Relationship between Executive Functions and Metacognitive Strategy Learning and Application

This project examined whether the executive functions of set maintenance and switching, as assessed by neuropsychological testing, were predictors of set maintenance and switching within a more ecologically valid task that used metacognitive strategies during reading comprehension tasks as a framework for evaluation. Gaze times to key words during reading were used as an indirect measure of strategy use. A few significant relationships were found between set maintenance and set switching on the neuropsychological measures and the strategy learning and application tasks. Participants were more likely to switch to appropriate strategies in a situation in which they were given free choice of strategies to use, and in which characteristics of the text pulled for the use of a particular strategy. In contrast, participants were less consistent with expected strategy use when they had just learned a strategy and were asked explicitly to apply it to a text that did not pull for use of a particular strategy. Factors such as visual scanning, motor speed, working memory, and passage comprehension affected the relationship between executive functions and the more ecologically valid task.

Der Ziel dieser Arbeit ist der Hausdorff Konvergenz der Juliamengen zu beweisen, als wir eine Familie von ganzen transzendenten Funktionen, die ein einziges Bakergebiet enthalten, approximieren. Als erstes geben wir eine vollständige dynamische Beschreibung der approximierenden transzendenten Funktionen und zeigen die Existenz von invarianten Gebiete unter der Iterierte. Insbesondere besitzen die approximierenden Funktionen ein Attraktionsgebiet, das gegen das Bakergebiet als Kernel im Sinn von Carathéodory konvergiert. Letztlich beweisen wir Hausdorff Konvergenz auf zwei Wege. Einerseits zeigen wir unter bestimmten Bedingungen der Fatoumenge der Grenzfunktion die Hausdorff Konvergenz der Juliamengen. Anderseits zeigen wir unter verschiedenen Bedingungen der Fatoumenge der Grenzfunktion die Hausdorff Konvergenz der ausgefüllten Juliamengen, die bezüglich der Bakergebiet oder der Attraktionsgebiet definiert sind.

Compact and weakly compact Derivations on l^1(Z_+)

In this thesis, we aim to study derivations from l^1(Z+) to its dual, l^\infty(Z+). We first characterize them as certain closed subspace of l^1(Z+). Then we present a necessary and sufficient condition, due to M. J. Heath, to make a bounded derivation on l^1(Z+) into l^\infty(Z+), a compact linear operator. After that base on the work in [6], we study weakly compact derivations from l^1(Z+) to its dual. We introduce T-sets and TF-sets and then state their relation with weakly compact operators on l^1(Z+). These results are originally due to Y. Choi and M. J. Heath, but we give simpler proofs. Finally, we will study certain classes of derivations from L^1(R+) to L^\infty(R+), and give an elementary proof that they are always mapped into C0(R+).

Promoting generalization of coin value relations with young children via equivalence class formation

Sidman and Tailby (1982) established procedures to analyze the nature of stimulus to stimulus relations established by conditional discriminations. Their research describes specific behavioral tests to determine the establishment of properties that define the relations of equivalence. An equivalence relation requires the demonstration of three conditional relations: reflexivity, symmetry, and transitivity. The equivalence stimulus paradigm provides a method to account for novel responding. The research suggests that equivalence relations provide a more efficient and effective approach to the assessment, analysis, and instruction of skills. The present research examined the effectiveness of the formation of an equivalence class in teaching young children coin value relations. The second aspect of the study was to determine if there was a relationship between equivalence class formation and generalization of the skills established to other settings. Five children, 4- and 5-years old, were selected to participate in the study based on their lack of skills in the area of coin values and purchasing an item with dimes or quarters equaling fifth cents. The experimental task was presented on a Macintosh computer with HyperCard programming. The experimental stimuli consisted of pictures of dimes, quarters, and Hershey candy bars presented in match-to-sample procedures. Two conditional discriminations were taught (if A then B and if B then C.). The formation of an equivalence class was evaluated by if C then A. Generalization across settings was tested after the formation of an equivalence class by having the children purchase a Hershey candy bar with dimes at a play store. A multiple baseline experimentaldesign was used to demonstrate a functional relationship between the formation of an equivalence class and generalization of skills across settings. The present research provides supportive evidence that coin value relations can be taught to young children using equivalence procedures. The study also demonstrated generalization of novel, untaught stimuli across settings, after the formation of an equivalence class. A posttest on generalization across settings was conducted 3 months after the study. Long-term stability of equivalence relations was demonstrated by three of the subjects. / Department of Special Education
