Caracterização e avaliação de amido nativo e modificado de pinhão mediante provas funcionais e térmicas

O pinhão é a semente comestível da Araucaria angustifolia, sendo consumido no sul e sudeste do Brasil principalmente na sua forma in natura. A falta de diversidade em relação ao seu consumo acarreta a sobra da safra e a sua utilização como ração animal, o que diminui o valor de comercialização do produto, prejudicando os ganhos econômicos do produto. A semente do pinhão é composta de 34% de amido e um conteúdo muito baixo de proteínas e lipídios, o que a torna favorável para a obtenção de amido, aditivo amplamente utilizado pela indústria de alimentos. O objetivo deste trabalho foi caracterizar uma nova fonte de amido a partir de semente de Araucária angustifolia, através de um estudo detalhado da metodologia de extração do amido de pinhão, das características físico-químicas, funcionais e reológicas do grânulo e de sua modificação estrutural. Primeiramente foi determinado um protocolo para a extração do amido e realizado o estudo do efeito do tratamento alcalino (NaOH 0,05 mol.l-1 e NaOH 0,1 mol.l-1), em relação ao tratamento com água, na estrutura dos grânulos do amido extraídos. Para tanto foram feitas análises de fluorescência intrínseca, microscopia eletrônica de varredura e de força atômica e difractometria de raios X. A estrutura granular do amido de pinhão foi alterada significativamente quando as soluções alcalinas foram utilizadas durante a extração. O amido de pinhão apresentou estrutura do tipo C. Foram determinadas as características do grânulo de amido nativo, através de análises físico-químicas, MEV, microscopia ótica e sinerese, onde se verificou que o diâmetro médio dos grânulos de amido varia de 7 a 20 μm e as formas geométricas variam entre a oval, a elipsóide truncada e a hemisférica. O amido de pinhão mostrou-se menos propenso à retogradação, comparado ao amido de milho. Foram elaboradas as isotermas de adsorção do amido nativo às temperaturas de 10°, 20°, 30° e 40°C a partir do ajuste dos dados experimentais a diversos modelos de sorção, sendo os modelos de Peleg, seguidos pelos de Chung-Pfost e GAB os que tiveram os maiores coeficientes de correlação. A partir do modelo de GAB foram calculados os valores de umidade de monocamada, entalpia e entropia diferenciais de sorção. Foi realizado também um planejamento experimental a fim de estudar a reação de hidrólise do amido, através da influência das variáveis temperatura (30°a 44°C) e concentração de ácido clorídrico (1,2 a 3,2 mol.l-1) no valor de dextrose equivalente (DE) e na capacidade de formação do gel e de sua termorreversibilidade. Foi observado que sob os maiores valores de temperatura e concentração de ácido empregados, o valor de DE foi maior. No tratamento que resultou no maior valor de DE (3,2 mol.l-1 de HCl e 44°C) foi observada a formação de gel termorreversível, o que indicou a potencialidade do amido modificado como substituto de gordura e agente ligante, em alimentos. A funcionalidade do amido de pinhão foi verificada através das análises de formação, fusão e termorreversibilidade do gel, tendência à retrogradação, estabilidade ao congelamento e descongelamento, poder de inchamento, índice de solubilização, MEV, viscosidade de pasta e propriedades térmicas, como temperatura e entalpia de gelatinização. Os resultados indicaram que: somente os amidos modificados apresentam géis termorreversíveis; os amidos de pinhão nativo e modificados possuem menor tendência à retrogradação do que seus similares de milho; a modificação do amido reduziu a tendência à retrogradação das pastas de amido; o amido de pinhão mostrou menor sinerese sob armazenamento à 5°C e após ciclos de congelamento e descongelamento; a temperatura de armazenamento influi significativamente no poder de inchamento (PI) e no índice de solubilização dos amidos de pinhão e milho nativos; os amidos modificados apresentaram menor PI do que os nativos; a viscosidade máxima do amido de pinhão foi maior à encontrada para o amido de milho, sendo ele menos resistente à agitação mecânica à quente (maior breakdown); a hidrólise ácida reduziu drasticamente a viscosidade das pastas formadas; a temperatura de gelatinização do amido de pinhão é de 47,64°C e a entalpia de gelatinização de 15,23 J.g-1; a hidrólise ácida aumentou a temperatura de gelatinização das espécies testadas. / Pinhão is the edible seed of Araucaria angustifolia, being consumed in the south and southeastern Brazil mainly in the natural form. The lack of diversity in relation to its consumption results in plenty of crop and its use as animal feed, which reduces the value of product marketing and affect the economic gains of the product. The pinhão seed is composed of 34% starch and a very low content of proteins and lipids, making it favorable for obtaining starch that is widely used by the food industry. The objective of this study was to characterize a new source of starch from seeds of Araucaria angustifolia, through a detailed study of the method for starch extraction, the physical-chemical, functional and rheological properties of the granules and their structural modification. To do this, a protocol for the starch extraction and a study of the effect of alkaline treatment (NaOH 0.05 mol.l-1 NaOH and 0.1 mol.l-1) compared to water treatment, was first established. Therefore, intrinsic fluorescence analysis, scanning electron and atomic force microscopy and x-ray diffractometry were carried out. The granular structure of pinhão starch was changed significantly when the alkaline solutions were used during extraction. The pinhão starch had a type C structure. The characteristics of native starch granules, through physical and chemical analysis, SEM, optical microscopy and syneresis were determined which found that the average diameter of starch granules varies from 7 to 20 μm and geometric shapes range from oval, the ellipsoid truncated and hemispheric. The pinhão starch was less prone to retrogradation compared to maize starch. The adsorption isotherms of native starch at temperatures of 10 °, 20 °, 30 ° and 40 ° C were drafting from fitting the experimental data to various sorption models. The Peleg model, followed by Chung-Pfost and GAB model had the highest correlation coefficients. From the GAB model were calculated monolayer moisture, enthalpy and entropy differential sorption. It was also carried out an experimental design to study the reaction of starch hydrolysis, through the influence of temperature (30° to 44°C) and hydrochloric acid concentration (1.2 to 3.2 mol.l- 1) on the dextrose equivalent value (DE) and on the ability of the gel formation and its thermoreversibility. It was observed that under the highest values of temperature and concentration of acid, the value of DE was higher. In larger value of DE (3.2 mol l-1 HCl and 44°C) was observed the formation of thermoreversible gel, which indicated the potential of modified starch as a fat substitute and a binder in food. The functionality of the pinhão starch was verified by analyzing the gel formation and fusion, gel thermoreversibility, tendency to retrogradation, stability to freezing and thawing, swelling power, solubility index, SEM, pasting viscosity and thermal properties such as temperature and enthalpy of gelatinization. The results showed that: only the modified starches exhibit thermoreversible gels, the native and modified pinhão starch have less tendency to retrograde than their counterparts in maize, the modification of starch reduced the tendency to retrogradation of starch pastes, the pinhão starch showed lower syneresis under storage at 5°C and after freezing and thawing, storage temperature affects significantly the swelling power (SP) and the solubility index of pinhão and maize native starches, the modified starches had lower SP of the natives, the maximum viscosity of pinhão starch was higher than that found for maize starch, the pinhão starch is less resistant to hot mechanical stirring (greater breakdown) than for maize starch, the acid hydrolysis reduced the viscosity of the pulp formed, the gelatinization temperature of pinhão starch is 47.64°C and its gelatinization enthalpy is 15.23 Jg-1; acid hydrolysis increased the gelatinization temperature of the species tested.

Substitute family care is a form of care for abandoned or orphaned children when a child is brought up by foster parents in an environment which is similar to a natural family environment. In the Czech Republic, there are various forms of substitute family care, mainly adoption and foster care. Arranging foster care means seeking children who are suitable for placement in foster care and finding correspondent foster parents for those children. There are few different groups of children acceptable for foster care. One of those groups includes children with handicap. Handicap means an obstacle, difficulty, disadvantage or unfavourable situation which ensues from a limited ability or even inability of an individual to assert as well as other people. The aim of this dissertation is to asses foster care situation abroad and to compare foster care for children with handicap in South Bohemia and Pilsner Regions. Based on the aim, two hypothesis were determined. The first one: Variability of different foster care forms in the Czech Republic is not optimal compared to abroad. The second one: Comparison results of data from both surveyed regions will not be fundamentally different. Secondary data analysis method was used for the survey. The data was acquied from available professional literature and foster family files from Foster Care Centre of South Bohemia County Council in České Budějovice and Pilsner County Council in Plzeň. Required data were gathered from foster parents' records and fostered children documentation from 2001 to 2005. The established aim was accomplished. The first hypothesis was confirmed. The second hypothesis was not confirmed. The survey established that some European countries have already introduced further forms or foster care, especially professional foster care. This form of foster care is not available in our country yet but as the interest to become foster parents decreases in the Czech Republic, professional foster care may be the solution of this situation. Furthermore, the survey established that there is hardly any difference among foster parents and fostered children in individual character in both counties. However, there is a distinctive difference in the amount of foster parents who accepted a child to foster care. The same situation arises in the amount of children placed in foster care. Contribution of this dissertation for the health and social section consists in a creation of statistics during the years 2001-2005 and in an estimate of characteristics of foster parents which can assist in searching for new foster parents.

The origin of this case study comes from my career as a substitute in the course in Spanish letters UFSM. Investigated the representations made by graduates of this course on substitute teachers who acted with them throughout their training process, thinking about his involvement at the individual level and teacher training. The empirical field was composed of a group of five graduates who had the peculiarity in the presence graduation almost full this occupational category in the subjects who attended. The instrument for data collection was the semistructured interview. I also collected information about the course and composition of its faculty during the graduation of the study subjects. A case study, qualitative-phenomenological, in which the data have been triangulated. Based on literature review on theories of teacher education and having the background my career as a graduate student and teacher, build a sense of presence and performance of substitutes among this group of graduates. The results indicate that this category was represented as limiting devices for initial training, especially regarding the investigation. The informants represent their substitute teachers as deficient as knowledge of area and without political expression in the institution. However, regard them as pedagogically most articulate. Far from exhausting the subject, this case study is representative of what is lived by the students, without attempting to generalize or a definitive answer. In this sense, the present study is justified to bring the discussion an important aspect of higher education, which is the training of teachers serving in it, contributing to improving the training given to academic courses for undergraduate degrees. / A origem deste estudo de caso advém de minha trajetória como substituto no curso de Letras Espanhol da UFSM. Objetivei pesquisar as representações feitas por graduados do referido curso sobre os professores substitutos que com eles atuaram durante seu processo formativo inicial e pensar sua implicação em nível individual e da formação docente. O campo empírico foi composto por um grupo de cinco egressos, que tiveram como peculiaridade em sua graduação a presença quase que total desta categoria profissional nas disciplinas que cursaram. O instrumento de coleta de dados foi a entrevista semiestruturada. Também coletei informações sobre o curso e sobre a composição de seu corpo docente durante o período de graduação dos sujeitos investigados. Um estudo de caso de cunho qualitativo-fenomenológico, em que os dados obtidos foram triangulados. Baseado na revisão de literatura feita sobre teorias da formação docente e tendo como pano de fundo minha trajetória como aluno de graduação e professor do curso, construi sentido sobre a presença e a atuação dos substitutos junto a este grupo de egressos. Os resultados apontam que esta categoria docente foi representada como limitadora dos dispositivos da formação inicial, principalmente quanto à pesquisa. Os sujeitos investigados representam seus professores substitutos como deficitários quanto ao saber de área e sem expressão política institucional. Entrentanto, os consideram como pedagogicamente mais desenvoltos. Longe de esgotar o tema, este estudo de caso é representativo do vivido pelos egressos, sem pretender a generalização ou uma resposta definitiva. Neste sentido, o presente estudo se justifica por trazer à discussão um aspecto importante do ensino superior, que é o da formação do profissional docente que nele atua, contribuindo para a melhoria da formação inicial dispensada aos acadêmicos de cursos de graduação em licenciaturas.

This master's thesis deals with the present legal regulation of foster care as one of the forms of a substitute care. Foster care is a traditional private law institute with public law elements. Its purpose is to ensure substitute care to children whoes parents, or even custodian are not able to care for them because of various reasons. So foster care is a subsidiary to biological parent's care. On the other hand, based on law, foster care has priority over institutional care. Due to the reformation of the substitute care system, foster care went through lots of changes lately. Primarily it was approached to the extensive amendment of Act no. 359/1999 Coll., on the social and legal protection of children and with the effectiveness since the 1st of January the family law regulation has been incorporated to the Act No. 89/2012 Coll., civil code. The aim of this thesis is to provide a complete picture of the present legal regulation of foster care in the Czech Republic due to conducted legislative changes. Extent and reasons for substituting and switching highly active antiretroviral therapy at the Katutura Intermediate Hospital in Windhoek, Namibia

Magister Public Health - MPH / Background: Namibia is one of the Southern African countries hardest hit by the HIV epidemic, with an estimated one out of every five people infected (MoHSS, 2004). Approximately 80,000 of the infected population currently require antiretroviral treatment (ART). In order to prevent the progression of the HIV infection to AIDS, patients are required to take antiretroviral medicines lifelong. This lifelong use exposes patients to toxicities of these medicines and the only available options of managing the toxicities of ARVs are to treat the toxicity or substitute or switch the offending medicines. Aim: The current study aimed to describe the extent and reasons for substituting and switching HAART at the Katutura Intermediate Hospital in Windhoek, Namibia. Methodology: A descriptive retrospective case series study, in which medical records were reviewed to determine the extent and reasons for substituting and switching HAART was conducted. Random sampling was used to draw a sample of 500 from 3477 adult HAART patients who commenced treatment between 1 January 2002 and 31 December 2006. A prepiloted data collection tool was used to collect the data. The following information was collected: baseline CD4 count, weight, initial ARVs, first and second ARV substitutions, ART switch and the reasons for substituting ARVs or switching ART during the indicated period. Epi Info version 6 was used to analyse frequencies, means and medians of all important variables in the data set. Results: The sample was made up of 500 HAART patients; 60% were females. The median age of the sample was 34 years (Inter-quartile range (IQR) 30 – 40) and the median CD4 cell count was 153 cells/mm3 (IQR 96 – 212) at initiation of therapy. The median time on treatment before first substitution was 28 months (IQR 24 – 34), whereas the median time before second substitution was 10 months (IQR 6 – 15) from the time of the first substitution. The median time before switching was 31 months (IQR 24 - 39). A total of 31% of the study subjects underwent a substitution once, whereas 1.8% underwent a second substitution. Only six (1.2%) patients switched to a second line treatment after the modification of the treatment. The most commonly recorded reason for the first substitution was toxicity (19%). As in other studies, stavudine (D4T), nevirapine (NVP) and efavirenz (EFV) were the ARVs associated with most of the recorded toxicities. High viral load (50%) was the most reported reason for switching. In almost half of the substitution cases the reasons for substitution were not stated, and in a third of the switch cases the reasons for switching were not stated. Conclusion: The rate of substitution at 31% was similar to that found in other resource poor settings, however, the rate of switching (1.2%) was much lower than was found in similar settings. The aim of this thesis is to design a software for elementary, middle and high schools, which will serve to create alternative timetables for staff substitutions. There are similarly oriented programs, but most schools don't use them. These programs contain many flaws, which are also discussed in this work. Creating substitution schedules is complicated, and a lot of conditions must be taken care of. Checking of all the conditions should be the main advantage of the application. Design and implementation of the application is based on requirements by a school, where substitution timetables are produced manually. Great emphasis is placed on user friendliness and ease of use.

Flödescytometrisk analys av DNA-ploiditeten används vid diagnostisering av endometriecancer. DNA-ploidi reflekterar cellcykeln och avgör om tumörens cellpopulationen är diploid eller aneuploid, där aneuploiditet förknippas med sämre prognos. Vid analys av paraffininbäddat vävnadsmaterial används avparaffineringsmedlet xylen, vars toxiska egenskaper försämrar arbetsmiljön på laboratoriet. Den har en stark och obehaglig lukt som kan orsaka illamående och yrsel. Syftet med studien var att undersöka om xylen kan ersättas med xylensubstitutet Tissue Clear, ett isoparaffinskt kolväte som är mindre toxiskt. Studien omfattade paraffininbäddad humanvävnad från endometrioid carcinom (n=20), både diploid (n=15) och aneuploid (n=5) vävnad, som avparaffinerades med xylen respektive Tissue Clear innan DNA-ploidi utfördes. Eventuella skillnader inom de flödescytometriska parametrarna % CV-diploid, % S-fas, % debris och DI-aneuploid undersöktes och vid statistisk analys kunde ingen signifikant skillnad ses på samtliga parametrar. Eftersom analysen utförs sällan i rutin är antalet prover i studien relativt stor, trots att detta kan anses vara en liten kvantitet. Av dessa var endast 25 % av proverna aneuploida. Att en patient uppvisar aneuploiditet är ovanligt och därför ansågs även denna mängd som tillräckligt stor. Studien visar att avparaffinering med Tissue Clear är ekvivalent med xylen och därmed kan Tissue Clear ersätta xylen oavsett om vävnaden är diploid eller aneuploid. / DNA ploidy is used for endometrial cancer diagnosis. It reflects the cell cycle and determines whether the cell population in tumors is diploid or aneuploid. When analyzing paraffin embedded tissues xylene can be used for deparaffinization, whose toxicity impairs the laboratory´s work environment. Its strong and unpleasant smell can cause nausea and dizziness. The aim of this study was to investigate if xylene can be replaced with Tissue Clear, an isoparaffinic hydrocarbon that is less toxic. The study included paraffin embedded human tissues from endometrioid carcinoma (n=20), both diploid (n=15) and aneuploid (n=5), deparaffinized with xylene or Tissue Clear before DNA ploidy was performed. Potential differences between the parameters % CV-diploid, % S-phase, % debris and DI-aneuploid were statistically examined and showed no significant differences. The sample amount in this study might be considered low, though it is relatively high since the analysis is rarely performed routinely. Among these only 25 % were aneuploid. Patients showing aneuploidy is rare and the amount was therefore considered to be sufficient as well. The study shows that deparaffinization with Tissue Clear generates equivalent results as for xylene and can thereby replace xylene regardless if the tissue is diploid or aneuploid.

Empatie a socio-emoční kompetence u dětí v různých formách náhradní péče / Empathy and socio-emotional competencies among various forms of substitute care

This diploma thesis focuses on the level of socio-emotional competencies, the ability of empathy and the prevalence of psychosocial problems amongst children in various forms of substitute care (biological families, institutional care, foster families). The aim of this thesis is to clarify the relations in between the variables and to find possible patterns bound to different kinds of care. The thesis also aims at the effect of the lenght od institutional care and of the strategies of emotional regulation and behaviour in social situations. The sample includes 34 respondents. The data were collected from the children themselves (IDS), from caregivers (EQ-C, SDQ) and from the file documentation (anamnestic questionaire). The results show a significant correlation between the variables. The effect of length of stay in institutional care has not been proven. As for comparisons across different forms of care, differences in empathy rates and the occurrence of psychosocial problems have emerged, but not at the level of socio-emotional skills. The subsequent analysis of strategies of emotional regulation and social behavior has pointed to an interesting tendency regarding the socio-emotional functioning of children in substitute care.
