Effects of nutrient-tannin interactions on intake and germination of woody plant species by ruminants

Woody plant encroachment is one of the major problems worldwide because it affects negatively the herbaceous layer, which provide forage for livestock production. However, the role of ruminants particularly browsers in the dispersal of woody plant seeds still remains a concern for farmers interested in grass production. Seedpods of various woody plant species constitute a crucial part of the diet of herbivores during the dry season because of their high nutritional quality compared to herbaceous material. The interaction of associated diet quality, seed characteristics and animal species among other factors play a pivotal part in the success of livestock faecal seeds dispersion. Furthermore, dispersed seeds that successfully grow into mature woody plants become an important source of protein for herbivores. The use of woody plants as a source forage is known to be limited by plant secondary metabolites (PSMs) such as condensed tannins. The objectives of this study were to determine 1) the effects of condensed tannins and crude protein of Vachellia tortilis and Dichrostachys cinerea pods in seed recovery and germination fed to goats, and 2) the effects of diet mixing on the feed intake of plant species by goats. In the first experiment, a total of 12 female indigenous goats and 12 female Pedi sheep were utilised in this study, with the average body weights of 29.50 kg ± 1.60 (S.E) and 28.70 kg ± 1.60, respectively. Twelve goats were grouped into two groups of six goats per group, one group was fed D. cinerea pods and the other group was fed V. tortilis pods. The group of 12 sheep were divided similarly, the one group was fed D. cinerea pods and the other group was fed V. tortilis pods. Each animal was given V. tortilis and D. cinerea pods at 2.50% of their body weight. All animals were allowed to consume D. cinerea or V. tortilis pods within 24 h, after which the remaining pods were collected and weighed. Faecal collection commenced immediately after the 24 h pods feeding and was carried on until no seeds were discovered in faeces. All faeces extracted from sheep and goats were collected daily in the morning from the faecal bags. In the second experiment, a total of 24 indigenous goats with average body weight of 26.6 kg ± 0.51 were utilised. Goats were arbitrarily selected and grouped into four groups of six goats per group (goats were placed individually in 2 m2 pens). Each group was fed one of the following diets: diet one - Searsia lancea, diet two - S. pyroides, diet three - Euclea crispa and diet four - was a combination of the three plant species (Searsia lancea, S. pyroides and Euclea crispa). Searsia lancea, S. pyroides and E. crispa branches were collected every morning prior to feeding, and were weighed before offering the animals. Refusals were gathered and weighed, and intake was calculated as distinction between weight in and refusals. Plant species foliage were analysed for crude protein, condensed tannin, acid detergent lignin, acid detergent fibre and neutral detergent fibre. During the first experiment, the cumulative percentage seed recovery of V. tortilis from goats (46.00 % ± 1.90) and sheep (52.00 % ± 2.93) was significantly higher than D. cinerea from goats (13 % ± 1.47) and sheep (24.00 % ± 1.16). Germination percentage of D. cinerea seeds that passed through the gastro-intestinal tract of goats (33.12 % ± 2.94) and sheep (36.00 % ± 2.68) was significantly higher than V. tortilis seeds that passed through the gastro-intestinal tract of goats (28.98 % ± 2.68) and sheep (23.04 % ± 2.81). Average D. cinerea (34.56 % ± 1.99) and V. tortilis (26.02 %± 2.10) seeds that went through the gastro-intestinal of goats and sheep had a significantly higher germination rate than the control (i.e. no passage through the gut; D. cinerea = 2.31 % ± 1.55, V. tortilis = 5.07 % ± 2.68). The high mean cumulative percentage seed recovery of V. tortilis (18.80 %) may be attributed to the relatively higher crude protein than D. cinerea (12.20 %). This may encourage animal seed dispersal and germination of woody plant species with relatively high crude protein content. In the second experiment, Searsia lancea contained 8.50 % CP, 21.46 % acid detergent fibre (ADF), 12.50 % ADL and 39.37 % NDF. Searsia pyroides had 9.03 % CP, 27.07 % ADF, 10.89 % ADL and 40.30 % NDF. Euclea crispa had 6.19 % CP, 26.20 % ADF, 16.63 % ADL and 30.02 % NDF. Mixed diet (combination of the three plant species) had 8.96 % CP, 23.72 % ADF, 11.13 % ADL and 38.28 % NDF. Searsia lancea had 2.70 % of CTs while S. pyroides had 5.20 % CT, E. crispa had 6.44 % CT and mixed diet had 7.20 % CT. The mean dry matter intake varied significantly among dietary groups (P < 0.001). Similarly, goats offered a mixed diet consumed more CTs (P < 0.01) than those offered individual forage species. The high mean cumulative percentage seed recovery of V. tortilis may be attributed to the higher crude protein of V. tortilis (18.80 %) than D. cinerea (12.20 %). Higher passage rate may encourage animal seed dispersal and germination of plant species. The results from experiment two support the postulation that animals foraging in mixed diet systems consume more PSMs and achieve higher dry matter intake than animals confined to monocultures or single species feeding systems. Given that woody plant encroachment is already reducing farm-grazing capacities in African savannas and this problem is predicted to double by 2050, strategies that improve herbivore ability to consume woody plants will increase forage availability and inform bush control programmes and policies. Conselho Nacional de Desenvolvimento Científico e Tecnológico / This work aimed at evaluating the quality of particleboards manufactured with different wood proportions (Eucalyptus grandis W. Hill. ex Maiden) and rice husk. The composites were produced in the proportions 0, 20, 40, 60, 80 and 100% of peels of rice with the use of urea-formaldehyde and tannin-formaldehyde adhesives. In order to characterize the quality of the particleboards, physical properties (specific mass; moisture content; water absorption and swelling in thickness after 2 and 24 hours), mechanical resistance (static bending; internal bonding and screw withdrawal) and xylophages resistance (Nasutitermes corniger under alimentary preference assay and decay fungi Gloeophyllum trabeum and Trametes versicolor in laboratory conditions) were evaluated. Results showed that particleboards manufactured with tannin-formaldehyde resins presented a higher quality. The increase of rice husk provided a larger instability and a smaller resistance of the particleboards. Particleboards produced with peels of rice presented a higher resistance to the attack of fungi or xylophages termites. Particleboards glued with tannin-formaldehyde were more resistant to the attack of termites; for the fungi, the difference among adhesives happened when these particleboards were submitted to the attack of Gloeophyllum trabeum, and the particleboards produced with urea-formaldehyde were the most resistant. In the fungi, Trametes versicolor was the fungus that provided the largest mass loss to the particleboards. / Avaliou-se a qualidade de painéis aglomerados produzidos com diferentes proporções de madeira (Eucalyptus grandis W. Hill. ex Maiden) e casca de arroz. Os compósitos foram produzidos nas proporções 0, 20, 40, 60, 80 e 100% de casca de arroz com a utilização dos adesivos ureia-formaldeído e tanino-formaldeído. Para caracterizar a qualidade das chapas, foram avaliadas as propriedades físicas (massa específica; teor de umidade; absorção de água e inchamento em espessura as 2 e 24 horas), a resistência mecânica (flexão estática; ligação interna; e resistência ao arrancamento de parafusos) e a resistência a xilófagos (aos cupins subterrâneos Nasutitermes corniger, em ensaios de preferência alimentar e aos fungos apodrecedores Gloeophyllum trabeum e Trametes versicolor em ensaios de laboratório). Os resultados obtidos indicaram que quando confeccionados utilizando a resina à base de tanino os painéis apresentaram qualidade superior. O acréscimo da casca de arroz proporcionou uma maior instabilidade e uma menor resistência das chapas. Em contrapartida, painéis produzidos com casca de arroz geralmente apresentaram maior resistência ao ataque de fungos apodrecedores ou aos cupins subterrâneos. Painéis colados com tanino-formaldeído foram mais resistentes ao ataque dos térmitas, enquanto que, para os fungos, a diferença entre adesivos ocorreu apenas quando submetidas ao ataque de Gloeophyllum trabeum, sendo as chapas produzidas com ureia-formaldeído as mais resistentes. Dentre os fungos, Trametes versicolor proporcionou uma maior perda de massa aos painéis.

Tanino como macromonômero na síntese de polímeros fenólicos visando a preparação de compósitos reforçados com material de origem vegetal / Tannin as substitute of phenols in the formulation of phenolic resins for the processing of composites reinforced with material from renewable source

No presente trabalho, tanino (macromolécula de origem natural) foi utilizado como substituinte de fenol na formulação de matriz fenólica usada na preparação de compósitos, o que é possível devido à presença de anéis fenólicos sua estrutura. Os compósitos de matriz taninofenólica (50% em massa de tanino) apresentaram propriedades mecânicas superiores aquelas dos compósitos de matriz fenólica, quando reforçados pelo mesmo tipo de fibra, mostrando que a substituição de material obtido em larga escala a partir de fonte fóssil (fenol) é viável e pode ser feita por material obtido de fonte renovável (tanino) sem comprometimento de propriedades. Ainda, avaliou-se para propriedades de compósitos com diferentes tipos de reforços (fibras e cargas) através de caracterizações via análise térmica (TG e DSC), análise dinâmico-mecânica, espectroscopia na região do infravermelho, resistência ao impacto, absorção de água e microscopia eletrônica de varredura. Os ensaios de resistência ao impacto indicaram uma melhora de propriedades mecânicas quando da incorporação de fibras vegetais (juta e coco) nos termorrígidos fenólico e taninofenólico, além de mostrar que as fibras de juta reforçam as matrizes taninofenólicas mais eficientemente que as fibras de coco. As cascas da árvore de Acácia Negra, ricas em taninos, também foram utilizadas como agentes de reforço em compósitos na forma de fibras e cargas, obtidos através de desfibramento e pulverização, respectivamente. A utilização destes reforços, em diferentes proporções de massa, em compósitos de matriz taninofenólica (50% de tanino) levou a diferenças nas propriedades do compósito, com destaque para baixa absorção de água. Provavelmente, a presença de taninos no reforço e na matriz levou a intensas interações na interface fibra/matriz, diminuindo o número de vazios que podem alojar moléculas de água. As fibras de coco foram tratadas por ultra-som, a fim de avaliar a influência deste tratamento nas propriedades da fibra e dos compósitos reforçados por elas. As fibras de coco tratadas e não-tratadas foram caracterizadas quanto à composição química, índice de cristalinidade, ensaio de tração e microscopia eletrônica de varredura. Os resultados obtidos mostraram que o ultra-som é um tratamento promissor, pois modifica a morfologia da superfície das fibras sem levar à degradação química, sendo que a separação dos feixes de fibra que ocorre permite melhor interação fibra/matriz na interface, obtendo-se assim compósitos mais resistentes à água. Outros tratamentos utilizados em trabalhos anteriores a este, como mercerização, diminuem absorção de água dos compósitos por ela reforçados, ao custo de parcial degradação das fibras lignocelulósicas. No presente trabalho, materiais preparados a partir de matéria-prima oriunda de fontes renováveis foram processados, o que vem de encontro aos anseios atuais. Os compósitos obtidos têm potencial para aplicações não estruturais, por exemplo, em partes internas de veículos automotivos. / In the present work, tannin (macromolecule obtained from natural source) was used as substitute of phenol in the formulation of phenolic matrix composites, due to the presence of phenolic rings in its structure. The tanninphenolic matrix composites (50% w/w of tannin) presented mechanical properties better than those of phenolic matrix composites showing that substitution of material obtained in large scale from non-renewable source (phenol) can be done by material obtained from natural source (tannin) without compromising the properties of the composite. The tanninphenolic matrix composites reinforced by different reinforcing agents (fibers and particules) were characterized by different techniques: Izod impact strength, thermogravimetry (TG), differential scanning calorymetry (DSC), infrared spectroscopy (IV), dynamic-mechanical analysis (DMA) and scanning electron microscopy (SEM). The Izod impact strength showed an improvement of mechanical properties due to the incorporation of natural fibres (jute and coir) in the phenolic and tanninphenolic matrices and also the better reinforcement of these matrices by jute fibres, when compared to coir fibres. The barks of Acacia Mimosa (high content of tannin) were also used as reinforcing agents of the tanninphenolic matrices in the forms of fibres and particules. The presence of these reinforcing agents in the matrix led to differences in the properties of the composites, highlighted by its lower water uptake. The presence of tannins in both reinforcing agents and matrix enhanced the fiber/matrix interactions, lowering the voids that increase water uptake. The coir fibres were treated by ultrasound, in order to evaluate the influence of this treatment in the properties of the fibres and, therefore, the composites reinforced with them. Besides chemical composition, all the fibres were characterized by the following techniques: X-ray diffraction, tensile strengh, infrared spectroscopy (IV), and scanning electron microscopy (SEM). The results revealed that the ultrasound is a promising treatment of fibres for the processing of composites, because it modifies the morphology of the surface of fibres without leading them to chemical degradation. The separation of fiber beams allows enhancement of the fiber/matrix interactions, leading to composites with lower water absorption capacity. Other treatments, such as mercerization, for example, improved the impregnation of the fibres by the pre-polymer, leading to composites with better properties, at expenses of partial degradation of lignocellullosic fibres. In the present work, composites were prepared using material obtained from renewable source, according to the purpose of this work. The obtained composites presents potential for non-structural applications, such as, internal panels of cars, for example.

Sorgo em dietas para o Jundiá (Rhamdia quelen) / Sorghum in diets for Silver Catfish (Rhamdia quelen)

The experiment was conducted at the Instituto de Pesquisa em Aquicultura Ambiental - InPAA belonging to Universidade Estadual do Oeste do Paraná, Toledo campus. Six hundred fingerlings with initial average weight and length of 4.49 ± 0.8 grams and 3.42 ± 0.24 inches respectively were distributed in 24 tanks - network with capacity 1m3 of usable volume. Completely randomized design with six treatments and four replications was used with an experimental unit formed by tank with 25 fish. Treatments consisted of six diets with increasing levels of 0; 6; 12; 18; 24; 30% sorghum and decreasing 30; 24; 18; 12; 6; 0% corn.. For 114 days, the fingerlings were fed three times a day (8, 12 and 17h), ad libitum. Survival was lower in the lot of fish fed a diet containing 30% sorghum, differing significantly (p <0.05) diets with 0 and 6% sorghum. The values of the chemical composition of the carcass and total protein were normal for the species and may serve as a basis for comparison with studies involving different culture conditions of silver catfish. It was concluded that sorghum can replace corn by 80% in diets for silver catfish. / O objetivo desse trabalho foi avaliar o efeito da substituição do milho pelo sorgo de baixo tanino em dietas para jundiá (Rhamdia quelen) no desempenho produtivo, sobrevivência, composição centesimal da carcaça, em dois parâmetros hematológicos proteínas totais e leucócitos e no tamanho e densidade das micro vilosidades do peixe. O experimento foi realizado no Instituto de Pesquisa em Aquicultura Ambiental InPAA, pertencente a Universidade Estadual do Oeste do Paraná, Campus Toledo - PR . Foram utilizados 600 alevinos com peso e comprimento inicial médio de 4,49 ± 0,80g e 8,68 ± 0,62 cm respectivamente, distribuídos em 24 tanques rede com capacidade de 1m3 de volume útil. Utilizou-se delineamento experimental inteiramente casualizado com seis tratamentos e quatro repetições, sendo cada unidade experimental formada por um tanque com 25 peixes. Os tratamentos constituíram-se de seis dietas com níveis crescentes de 0; 6; 12; 18; 24 e 30% de sorgo e decrescentes de 30; 24; 18; 12; 6 e 0% de milho. Durante 114 dias os alevinos foram alimentados três vezes ao dia (8; 12 e 17h), ad libitum. A sobrevivência foi menor no lote de peixes alimentados com dieta contendo 30% de sorgo, diferindo estatisticamente (p<0,05) das dietas com 0 e 6 % de sorgo. Os valores da composição química da carcaça e proteínas totais foram normais para a espécie, podendo servir de base para comparação com trabalhos que envolvam situações diferentes de cultivo de Jundiá. Concluiu-se que o sorgo pode substituir o milho em até 80% em dietas para o Jundiá.

Emprego de coagulante à base de tanino em sistema de pós-tratamento de efluente de reator UASB por flotação / Use of tannin-based coagulant in exhaust post-treatment of UASB reactor effluent by flotation

O presente trabalho teve como objetivo principal a avaliação da eficiência de remoção de carga orgânica, fósforo e sólidos em suspensão de sistema de flotação tratando esgoto sanitário pré-tratado em reatores UASB, da ETE Monjolinho da cidade de São Carlos/SP, e coagulado com tanino associado ou não a polímeros sintéticos. Os ensaios com variação de dosagem de coagulante Tanino e polímeros (polímero catiônico, polímero não-iônico e polímero aniônico) foram realizados em escala de laboratório, com o uso do equipamento flotateste. Em todos os ensaios foram fixados os valores de gradiente de velocidade na mistura rápida (800/s) e na floculação (90/s), ao tempos de detenção na mistura rápida (20 s) e na floculação (15 min) e, a pressão de saturação 5 Bar e a razão de recirculação (17%). Durante os ensaios com unidade de flotação em escala de bancada foram avaliados três valores de velocidades de flotação: 12,0 cm/min, 16,0 cm/min e 20,0 cm/min. Os ensaios foram divididos em duas fases: a fase 1 teve como objetivo analisar a resposta do sistema de flotação com o emprego do coagulante Tanino no tratamento do efluente do reator UASB da ETE Monjolinho (fase 1A) e também determinar as duas dosagens do coagulante Tanino mais adequadas para o tratamento do efluente de UASB da ETE Monjolinho para cada polímero testado (fase 1B). As fases 1A e 1B foram realizadas com efluente coletado às 8:00 h e 14:00 h, respectivamente. A fase 2 teve como objetivo a busca da dosagem ótima de tanino dentro de uma faixa mais estreita de valores, assim como a verificação do melhor tipo e dosagem de polímero sintético (não-iônico, aniônico e catiônico) para ser associado ao coagulante tanino. Na fase 1A, verificou-se que a flotação foi efetiva para remoção de cor aparente mesmo para dosagem nula de coagulante Tanino. Quanto à turbidez, verificou-se efeito marcante da adição de dosagem crescente de coagulante no aumento da eficiência de remoção desse parâmetro. Após a obtenção dos resultados da fase 1B, foram escolhidas para a fase 2 as duas menores dosagens de coagulante capazes de promover redução da turbidez em pelo menos 85%. Além destas duas dosagens, foram avaliados na fase 2 outras 3 dosagens compreendidas entre elas. Sendo assim, na fase 2 foram estudadas as doses de 65, 75, 85, 95 e 105 mg/L de coagulante, cada qual associada a seis dosagens de cada um dos polímeros estudados. Entre os tratamentos empregados, pôde-se verificar maior eficácia do polímero não-iônico e do polímero catiônico para o tratamento por flotação, associado ao coagulante tanino. O conjunto de dosagem de coagulante tanino e polímero mais vantajoso, ou seja, menor dosagem de tanino que, associada a polímero, apresentou remoção satisfatória dos parâmetros estudados, foi 65 mg/L de Tanino e 2,0 mg/L de polímero catiônico, atingindo remoções de 95,2% de turbidez (Residual de 3,65 NTU), 82,1% de cor aparente (Residual de 98 mgPT/L), 49,2% de fósforo total (Residual de 3,2 mg/L), 80,7% de DQO (Residual de 25 mg/L) e 87,9 % de SST (Residual de 13 mg/L) nas condições encontradas na época dos ensaios, para o pós-tratamento por flotação de efluente de reator UASB da ETE Monjolinho, São Carlos/SP. / The main objective of this work is the evaluation of the efficiency of removal of organic load, phosphorus and suspended solids in the flotation system treating domestic wastewater pretreated in UASB reactors, of the Monjolinho Wastewater Treatment Plant (WWTP) of Sao Carlos, SP and clotted with tannin alone or combined with synthetic polymers. Tests varying tannin dosage of coagulant and polymer (cationic polymer, nonionic polymer and anionic polymer) were performed on a batch scale, using flotatest. In all the tests were fixed the values of rapid mix (800/s) and flocculation (90/s) velocity gradients, the rapid mix (20 s) and flocculation (15 min) times, saturation pressure (5 Bar) and recycle rate (17%). During the test in the flotation bench scale unity three values of flotation velocities were evaluated: 12.0 cm/min, 16.0 cm/min and 20.0 cm/min. The tests were divided in two phases: the objective of phase 1 was to examine the response of the flotation system with the use of tannin coagulant in the treatment of Monjolinho WWTP UASB effluent (Phase 1A) and also determine the two most suitable dosages of tannin coagulant for the treatment of wastewater from the Monjolinho WWTP UASB tested for each polymer (phase 1B). Phases 1A and 1B were performed with wastewater collected at 8:00 h and 14:00 h, respectively. The objective of phase 2 was to find the optimum dosage of tannin within a narrower range of values, as well as verification of the optimal type and dosage of synthetic polymer (non-ionic, anionic and cationic) to be associated with the tannin coagulant. In phase 1A, it was found that flotation was effective for apparent color removal even to zero coagulant dosage tannin. As for turbidity, there is a marked effect when increasing the coagulant dose, which increased the removal efficiency of this parameter. After obtaining the results of Phase 1B, were chosen for phase 2 two low coagulant dosages which can promote reduction of turbidity by 85%. Besides the two dosages obtained in phase 1B, in phase 2 were evaluated other three between them. Thus, in phase 2 were studied coagulant doses of 65, 75, 85, 95 and 105 mg/L, each one associated with six doses of each one of the studied polymers. Among the treatments employed, it could be seen that the non-ionic polymer and the cationic polymer were the most efficient for treatment by coagulation associated with tannin in flotation. The set of tannin coagulant dosage and polymer which showed satisfactory removal of parameters was 65 mg/L of Tannin with 2.0 mg/L of cationic polymer, reaching 95.2% removal of turbidity, 83% of color apparent, 49.2% of total phosphorus, COD 80.7% and 87.9% of TSS under the conditions found at the time of testing, for post-treatment of Monjolinho WWTP UASB effluent by flotation.

This study presents mountain forest Norway spruce (Picea abies) injured by surface ozone and oxidative stress. Norway spruce is not a sensitive species but it is the most representative tree in our country and this is the reason to pay attention on its injury. Study was connected to the ozone measurement by CHMI in Jizerske mountains. Aim of our study was to find influence of surface ozone on the forest and show correlation between ozone concentrations, altitude and Norway spruce injury. Ozone concentrations are growing with the altitude. Increasing percentage of injury with growing concentrations and altitude was expected. Needles were collected at the altitude 750-1100 meters. Collected needle years were 2006, 2007 and 2009. The 2009 year was not evaluated because no symptoms were found. Injury was observed on the maroscopical, microscopical and biochemical level. Visual injury was assessed by percentage of injured needle surface by chlorosis and necrosis. Microscopical analysis studied injury on cross and longitudinal sections of needles. On the longitudinal sections were visible typically bone-cells. These cells have lost its shape by ozone stress and were narrowed in its width. Bone-cells were best visible near the stomata. Beiträge zum molekularen Verständnis der Aggregation und der Komplexierung von GnRH-Antagonisten und GHRH-Analoga

Die vorliegende Arbeit beschäftigt sich mit der molekularen Struktur der amyloiden Aggregate von Antagonisten des Gonadotropin-Releasing Hormones (GnRH) und Analoga des Growth Hormone-Releasing Hormones (GHRH) sowie deren schrittweiser Bildung und der Inhibierung dieses Prozesses durch verschiedene Polyphenole. Die dabei erhaltenen Erkenntnisse werden genutzt, um pharmazeutisch relevante Depotformulierungen der genannten Peptide zu generieren und deren Freisetzungsverhalten zu untersuchen. Die 3D Strukturen der Peptidmonomere werden basierend auf zweidimensionalen NMR-Experimenten aufgeklärt. Zur Untersuchung der einzelnen Teilschritte der Aggregation dienen zahlreiche strukturanalytische Methoden, u.a. zeitaufgelöste Fluoreszenzspektroskopie (TCSPC), Untersuchungen mit strukturselektiven Sonden sowie TEM-Messungen. Ein Schwerpunkt liegt auf der Sekundärstrukturanalyse durch Bandenformanalyse von ATR-FTIR-Banden, um die Ausbildung der amyloid-typischen β Faltblattstrukturen detailliert zu beschreiben. Aus den Ergebnissen wird für GnRH-Antagonisten ein fibrilläres Aggregatmodell mit paralleler β Faltblattstruktur abgeleitet, dass zwei voneinander unterscheidbare β Faltblattbereiche enthält. Mithilfe einer eigens entwickelten Kongorot-Titrationsmethode wird der amyloid-aggregierte Anteil der Peptide in Gegenwart verschiedener Partnermoleküle untersucht. Dabei zeigt sich u.a., dass Poly-L-Glutamat nur eine der beiden β Faltblattstrukturen schwächen kann, wohingegen eine Gallotanninmischung aus Rhus chinensis die Amyloidbildung vollständig unterdrückt. Durch umfangreiche Arbeiten zur Trennung und Charakterisierung der polyphenolischen Komponenten der Gallotanninmischung werden Untersuchungen zu Struktur-Eigenschaftsbeziehungen ermöglicht. Unter Nutzung dieser Erkenntnisse werden pharmazeutisch relevante Formulierungen aus den Peptiden und Partnermolekülen hergestellt und mit einer speziell entwickelten Dialyse-Liberationsmethode auf ihr Freisetzungsverhalten untersucht. Da der aggregationsinhibierende Effekte der Polyphenole diffusionsbedingt schnell verloren geht, wird schließlich die Synthese von mit Polyphenolen modifizierten Chitosanen und deren erfolgreicher Einsatz zur Komplexierung der Peptide beschrieben, wodurch erstmals eine Formulierung zur Verfügung steht, welche die Aggregation des Peptidwirkstoffes verhindert und gleichsam eine kontrolliert verzögerte Freisetzung ermöglicht. / The present study addresses the molecular structure of the amyloid type aggregates of GnRH antagonists and GHRH analogs, their stepwise formation and the inhibition of this process by various polyphenols. The findings thus obtained are used to prepare pharmaceutically relevant drug delivery systems of the peptides and to study the corresponding release behavior. The 3D structure of the peptide monomers are elucidated by two-dimensional nmr experiments. Numerous structure analytical approaches are applied to obtain further insight into the partial steps of the aggregation process, e.g. TCSPC, experiments with structure selective probes and TEM. Emphasis is placed on the investigation of the secondary structure by band shape analysis of amide bands obtained by ATR-FTIR spectroscopy in order to analyze the formation of amyloid type β sheet structures. Based on the analytical results, a fibrilar aggregate model is described, which contains two distinguishable β sheet structures. Using a newly developed congored titration method, the amyloid content of the peptides is examined in the presence of various additives. Whereas poly-L-glutamate is only capable of weakening one of the two β sheet structures, a crude tannic acid mixture from Rhus chinensis may suppress the aggregate formation completely. Extensive efforts are made to enable experiments targeting the structure-activity relationships of the polyphenolic components of the tannic acid mixture. With this in mind, pharmaceutically relevant formulations of the peptides and several additives are prepared and their release properties investigated using a newly developed dialysis liberation device. Since the aggregation inhibiting effect is lost quite fast due to the diffusive loss of the polyphenolic compounds, furthermore the synthesis of chitosan, which is covalently modified with the polyphenlic components, and its successful application for the generation of drug delivery systems of the peptides is described.
