High Resolution study of NF-kB - DNA Interactions

In this thesis we have attempted to study four basic aspects of DNA-protein interactions: Affinity, specificity, accessibility and kinetics. With NF-kB as our model transcription factor, we wanted to investigate how a particular dimer recognizes a specific binding sequence? How fast are these interactions? And finally, how does the NF-kB interact with it binding site in the chromatin context? Specificity of NF-kB-DNA interactions has recently come into focus after it was shown that these dimers can bind to the sequences which do not fall into the NF-kB general consensus motif. We studied seven such sequences for their specificity for four NF-kB dimers. Our results show that p50 homodimers are least discriminative and can bind specifically to all these sequences. While as, RelA homodimers were highly discriminative and did not bind to most of these nontraditional sequences. We used two different methods to measure binding affinities: traditional gel mobility shift assay (EMSA) and a novel technique called as UV laser footprinting. Our results show that UV laser footprinting is the better method to determine the binding constants.For studying the dynamics of NF-kB-DNA binding, we combined UV laser footprinting with stopped flow device. This combination, not only give us one base pair resolution but also milli-second time resolution. Using p50 homodimers as a model transcription factor, we showed that the binding of this factor follows a two-step mechanism. First step involves the fast recognition of the sequence and second step follows a slower kinetics most likely for the stabilization of the complex. Our experiments suggest that flanking sequences play a role in the recognition and stabilization process of the complex formation.Finally, we also studied the accessibility of nucleosomes to NF-kB. Our in vitro data sheds light on the in vivo requirements for the alterations in chromatin structure necessary for the productive binding of NF-kB. These include either a removal of H2A-H2B dimers from the nucleosome and/or chromatin remodeler induced relocation of the histone octamer.Our data sheds light on the in vivo requirements for the alterations in chromatin structure necessary for the productive binding of NF-kB. We hypothesize that some factors like PU.1 might be able to target the chromatin remodeling/dimer eviction machinery to particular nucleosomes and lead to productive binding of NF-kB.

Since democracy in 1994, the government has promulgated a number of acts to achieve the goal of adequate housing for all. These include the Prevention of Illegal Eviction from and Unlawful Occupation of Land Act (PIE) and the Rental Housing Act (RHA). The problem for the courts is knowing when to apply each act. To reach the goal embodied in the constitutional right of adequate housing for all, the government has invested R18 billion in housing since 1994. Despite this, the need for housing has escalated. The RHA, in which the legislature tried to create a balance between the rights of landlords and tenants, followed. This was done in order to alleviate some of the pressure to ensure access to land, which rests solely on the shoulders of the government. The legislature tried to create a sphere into which private investors would want to invest their money. A number of recent cases dealing with tenants who defaulted on their rentals and the landlord's capacity to effect eviction raised awareness about the existing inadequacies of the law in this particular field. In a Supreme Court of Appeal ruling, the court found that when a landlord wants to evict a defaulting tenant the time-consuming and costly procedure of PIE should be used. The assumption underlying this study is that PIE should not be applicable in cases of evicting a defaulting tenant. The rights and duties of the various parties involved in rental housing therefore need to be examined. The main aim is, however, to ascertain which procedure should be employed when obtaining an eviction order against a party holding over and what the effects are when the most appropriate eviction procedure is not used. A well-regulated relationship would ensure the best balance of interest for the landlord, tenant and the government by creating a market in which a landlord could make money out of letting and more tenants could obtain adequate housing through renting. A further assumption is that the rei vindicatio should be used when having a defaulting tenant evicted. It offers an alternative procedure that does not undermine the objectives of the housing legislation. / AFRIKAANSE OPSOMMING: Suid-Afrika ervaar tans 'n probleem met die verskaffing van behuising vir almal. Sedert die land se verwerwing van demokrasie in 1994 het die wetgewer 'n hele reeks wette aangeneem om die probleem op te los, ondermeer die Wet op Huurbehuising en die Wet op die Voorkoming van Onwettige Uitsetting en Onregmatige Okkupasie van Grond (hierna verwys as PIE). Die howe ondervind soms probleme wanneer daar bepaal moet word wanneer 'n spesifieke wet van toepassing behoort te wees en wanneer. Ten spyte van die R18 miljard wat die regering reeds bestee het aan armes sonder huise, het die getal mense wat sonder geskikte behuising woon gegroei. Die wetgewer het deur die promulgasie van die Wet op Huurbehuising gepoog om 'n mark te skep waarin daar behuising verskaf sal word in die vorm van huurbehuising. Terselfdertyd sal die privaatsektor baie nodige geld in die huurmark kan investeer. Onlangse regsspraak in die verband dui daarop dat daar nog baie leemtes bestaan veral met verwysing na uitsetting. Na 'n resente Appelhof beslissing sal die verhuurder van die meer tydrowende en duurder prosedures in PIE gebruik moet maak om 'n persoon uitgesit te kry. Die onderliggende aanname is dat PIE nie van toepassing behoort te wees wanneer 'n verhuurder 'n huurder wat agterstallig is met die huur wil uitsit nie. Die regte van beide huurder en verhuurder word gevolglik bestudeer. Die hoof-oogmerk van die studie is egter om vas te stel watter uitsettingsprosedure die beste sal wees en wat die gevolge sal wees indien die prosedure nie gebruik word nie. 'n Goed gereguleerde huurmark sal sorg dat huurders genoegsame beskerming geniet, dat die verhuurder geld sal kan maak uit die huurmark en dat die regering se druk tot 'n mate verlig word. 'n Verdere aanname is dat die prosedure vir die rei vindicatio die korrekte prosedure is om te gebruik om 'n huurder wat versuim om sy/haar huur te betaal uit te sit. Die rei vindicatio word gevolglik bestudeer en daar word getoon dat die prosedure aansienlik van die van PIE verskil. Dit bied 'n alternatief en is nie van so aard dat dit die behuisings wetgewing se oogmerke belemmer nie.

Management of official accommodation in the Department of Roads and Public Works in Port Elizabeth

The study investigates management of official accommodation in the Department of Roads and Public Works. Government immovable asset plays an essential role in ser-vice delivery. The official accommodation policies were not implemented as intendant and as a result, the former government employees are illegal occupants in government properties. The implementation of legislation and departmental policy for official ac-commodation are problematic when illegal occupants occupied government immova-ble assets without lease agreement. The present of illegal occupants has a negative impact to officials who qualify to occupy government properties in terms of the depart-mental policy for service delivery purposes. The study proposed to provide literature search using books, legislation, policies, and personal interviews among others. The research methodology employed in this study was described, followed by the legislative framework employed in this study. Some of the findings on a legislative framework for official accommodation in the department of Roads and Public Works in the department of Roads and Public Works include gaps at the implementation of departmental policy on Allocation of Official Housing in rela-tion to the lease of state owned properties. Recommendations as a result of descriptive literature search are presented and how the existing problem of implementation of official accommodation policies can be changed to enhance service delivery and to provide accommodation to officials who qualify to occupy government immovable assets. The recommendations will enable the Department of Roads and Public Works as custodian of immovable assets, to fulfil its Constitutional mandate to provide official accommodation to their officials and cli-ents departments who qualify in terms of the policy for service delivery purposes.

Domácí násilí jako sociální problém okresu České Budějovice / Domestic violence as a social problem in České Budějovice district

Violence at home, a.k.a. home violence, is a spiraling cycle of brutality committed in private between closely related persons. Such violence can by physical, psychological, sexual, economic or social, with the roles of the attacker and the victim clearly defined and never changing. Anyone can be exposed to home violence, irrespective of her or his social position, education, job, sex, age, religion, or the type of partnership that people live in. Advanced societies have developed a tendency to tackle the issue, and the awareness of the problem has improved. In our country this favorable development is driven primarily by the newly enacted regulations and the keener interest stirred up in the public. The thesis aimed to investigate the general public awareness of home violence within the District of České Budějovice. As followed from the survey, the public living in the District is well aware of the issue. The results showed that as much as 69% of respondents knew about home violence, while the remaining 31% were ignorant of the problem. Hypothesis: Females are better informed about home violence than males - confirmed by comparison between what the women and the men knew about the issue. The difference encountered, however, was just minor. Hypothesis: People living in the District of České Budějovice are not knowledgeable about changes newly passed in Act 135/200 to solve the problem of violence at home - not confirmed. The majority of respondents heard of the Act and were able to specify at least the essential changes it enacted. The change the respondents mentioned most often was the 10 day period of flat-eviction order. It comes as a matter of interest that males were better aware of the Act and could better quote the changes it introduced. Hypothesis: The public accepts as true that home violence is confined to just problem-ridden families - not confirmed. Eighty seven percent of females and 81% of males were convinced that home violence was not limited to only such families. Merely 13% of females and 19% of males believed the home violence was a matter of the problem-ridden families. The survey results showed that the public living in the District of České Budějovice had dropped this myth, i.e. they had not seen the hypothesis as truthful. The thesis can serve as a source of information for the lay public, as a piece of instruction material for the students of the University of South Bohemia, or it could also be given to the Intervention Center to be distributed among the victims of home violence.

This research will focus on the influence of legislation (as indicated in this research) on the investment decision in residential income-producing property. Assumptions, as recorded in the hypothesis of this study, indicate that the legislation had a changing influence on the investment decision in residential income-producing property in comparison to the time period prior to the promulgation of the legislation. The research methodology will be based on a comparative analysis of the current legislation and the proposed Draft Amendment Bill. This analysis will be tested by means of a case study analysis incorporating a phenomenological study based on written data. The problems, sub-problems and hypothesis will be addressed and tested in this research in conjunction with the prescribed research methodology. This research is concluded by means of a synopsis and recommendations.

Carmen Tomasa Chávez Benavides, en adelante la demandante, presentó una demanda de Desalojo por Vencimiento de Contrato en contra de la Asociación de Comerciantes “Feria Jerusalén Yanahuara” solicitando que los demandados desocupen el inmueble ubicado en la Calle Jerusalén N°108, esquina Av. Ejército N°313 del Distrito de Yanahuara, Arequipa. Fundamentos de hecho: La Asociación señaló que el demandado se encuentra ocupando su propiedad, debidamente inscrita en la partida electrónica N°01151967 del Registro de Predios de la Zona Registral XII Sede Arequipa, bajo los siguientes fundamentos: Con fecha 08 de mayo de 2012, la Sra. Carmen Tomasa Chávez Benavides adquirió el inmueble ubicado en el distrito de Yanahuara a mérito de una Escritura Pública de Donación, otorgada por su anterior propietario, el Sr. Jesús Oscar Gonzalo Torres Velásquez, la misma que está inscrita en la partida referida. Al adquirir la propiedad, el donante Sr. Jesús Oscar Gonzalo Torres Velásquez, informó a la donataria que el inmueble estaba alquilado a La Asociación de Comerciantes “Feria Jerusalén Yanahuara”, mediante contrato de arrendamiento de fecha primero de setiembre de 2011, por un período de un año, es decir hasta el 31 de agosto de 2012. From the filing of an application to evict to the final execution of an eviction order is a long and complicated process. This raises questions about how the actors involved experience and act during this process. The general aim of the study was to describe and analyse this process from the respective perspectives of the landlords, the social services and the individuals (families) facing eviction in three municipalities in the county of Stockholm. The issues discussed are how the landlords and the social welfare offices are organised, their work procedures, respective scopes of action and resources, and how individuals facing eviction cope with their life situation. The material for the study consists of qualitative interviews with nine handling officers at six landlord offices, fifteen social workers at three social welfare offices, twelve individuals who were facing eviction and fifteen observations of conversations between social workers and individuals who were facing eviction. The analytic framework used consists of perspectives and concepts derived from organisation theory – Human Service Organizations and street-level bureaucrats – and theories on coping. A main result of the study is that the work procedures of the professionals are standardised in many ways and are characterised by similar procedures for classifying and categorising the individuals facing eviction, irrespective of how the three municipalities have organised their work. Another main result is that the primarily interests of the individuals facing eviction are to meet what they experience to be their children’s needs, and to avoid eviction. Further, the individuals experience as stressful their life situation marked by the threat of eviction in combination with reduced financial resources that make it difficult for them to meet their children’s needs. The study has identified two types of strategies that individuals facing eviction use in order to cope with their life situation and to achieve their interests. These are emotion-focused strategies and problem-focused strategies, which are described and discussed.

The purpose of this thesis is to investigate how children are affected by eviction and structural homelessness in Sweden, and also to investigate what rights children have towards the Swedish state and Swedish municipalities under the Convention on the Right of the Child. To achieve the purpose, the author has used rapports of evicted and homeless children in Sweden, and analyzed the Convention on the Rights of the Child. The author did this to see what obligations the Swedish state and the Swedish municipalities have towards the children. Documents from Malmo and Gothenburg municipality are also used to see how the implementation of the Convention on the Rights of the Child looks like at the local level. A comparison between the municipalities was made to see if there are similarities or differences between the municipalities implementation of the convention. An investigation was also done to see whether the municipalities comply the claims of the convention.
