Development and Characterization of Gel-Like Structures from Aquatic Biomass for Food Applications / Obtención y caracterización de estructuras tipo gel a partir de biomasa acuática para aplicaciones alimentariasFontes Candia, Cynthia 28 July 2022 (has links)
Tesis por compendio / [ES] El objetivo de esta tesis doctoral ha sido el diseño y caracterización estructural de estructuras tipo gel basadas de polisacáridos extraídos de biomasa acuática con interés para aplicaciones relacionadas con la alimentación. Las propiedades de los polisacáridos extraídos de algas y plantas acuáticas son adecuadas para producir diferentes estructuras tipo gel basadas en la formación de redes reticuladas, como hidrogeles, aerogeles y emulsion-gels.
En la primera parte de esta tesis se investigaron los diferentes mecanismos de gelificación de polisacáridos sulfatados, así como los parámetros que afectan a la estructura y las propiedades funcionales de los hidrogeles obtenidos. En base a los resultados, se evaluó la potencial aplicación de los hidrogeles y aerogeles de agar y k-carragenato para encapsular una proteína alimentaria modelo como la caseína, explorando así el efecto protector contra la hidrólisis enzimática tras digestiones gastrointestinales simuladas.
En la segunda parte de esta tesis, se desarrollaron estructuras de aerogeles mediante la valorización de una fuente de biomasa residual infrautilizada, como es el Arundo donax. Con esta biomasa se generaron fracciones celulósicas con diferentes grados de purificación y extractos bioactivos solubles en agua, que posteriormente se utilizaron para producir aerogeles bioactivos híbridos. La estructura altamente porosa y la elevada capacidad de sorción de los aerogeles los convierten en excelentes candidatos para la sustitución de las almohadillas absorbentes para mantener la calidad de los productos cárnicos envasados.
Los emulsion-gels son reconocidos por su gran potencial como ingredientes funcionales en la industria alimentaria como modificadores de textura y como sustitutos de grasas sólidas. Además, pueden utilizarse como vehículo para la liberación controlada de compuestos bioactivos liposolubles. Así, en la última parte de esta tesis, se investigó la naturaleza de las interacciones entre los componentes en las formulaciones de emulsion-gels basadas en polisacáridos y se relacionó con su estructura y comportamiento mecánico y reológico. Después de estudiar el mecanismo de gelificación de los emulsion-gels de carragenato, estos sistemas se adaptaron y utilizaron para dos diferentes aplicaciones relevantes para los sectores de alimentación y biomedicina. En primer lugar, se produjeron y evaluaron estructuras gelificadas a partir de emulsion-gels de agar y k-carragenato y aerogeles cargados de aceite como encapsulantes de un bioactivo lipofílico como la curcumina. Los resultados mostraron que el tipo de polisacárido y el estado físico de las redes del gel tenían un impacto en la estructura de los productos de la digestión.
Por otro lado, se evaluó el potencial de los emulsion-gels basados en polisacáridos sulfatados (k-carragenato y agar) para la producción de un material capaz de simular tejido graso. Los resultados evidencian que los emulsions-gels de agar son adecuadas para producir materiales que simulan las propiedades dieléctricas para imitar tejidos de bajo y alto contenido en agua. / [CA] L'objectiu d'aquesta tesi doctoral ha sigut, el disseny i caracterització estructural d'estructures tipus gel, basades en polisacàrids extrets de biomassa aquàtica amb interès per a aplicacions relacionades amb l'alimentació. Les propietats dels polisacàrids extrets d'algues i plantes aquàtiques, són adequades per a produir diferents estructures tipus gel basades en la formació de xarxes reticulades, com a hidrogels, aerogels i emulsió-gels.
En la primera part d'aquesta tesi es van investigar els diferents mecanismes de gelificació de polisacàrids sulfatats, així com els paràmetres que afecten l'estructura i les propietats funcionals dels hidrogels obtinguts. Sobre la base dels resultats, es va avaluar la potencial aplicació dels hidrogels i aerogels d'agar i k-carraguenina per a encapsular una proteïna alimentària model com la caseïna, explorant així l'efecte protector contra la hidròlisi enzimàtica després de digestions gastrointestinals simulades.
A la segona part d'aquesta tesi, es van desenvolupar estructures d' aerogels mitjançant la valorització d'una font de biomassa residual infrautilitzada, com és el Arundo donax. Amb aquesta biomassa es van generar fraccions cel·lulòsiques amb diferents graus de purificació i extractes bioactius solubles en aigua, que posteriorment es van utilitzar per a produir aerogels bioactius híbrids. L'estructura altament porosa i l'elevada capacitat de sorció dels aerogels els converteixen en excel·lents candidats per a la substitució dels coixinets absorbents per a mantenir la qualitat dels productes carnis envasats.

A més, poden utilitzar-se com a vehicle per a l'alliberament controlat de compostos bioactius liposolubles. Així, a l'última part d'aquesta tesi, es va investigar la naturalesa de les interaccions entre els components dins les formulacions d' emulsió-gels basades en polisacàrids i es va relacionar amb la seua estructura i comportament mecànic i reològic. Després d'estudiar el mecanisme de gelificació dels emulsió-gels de carraguenina, aquests sistemes es van adaptar i es van utilitzar per a dues aplicacions diferents, rellevants per als sectors de l' alimentació i la biomedicina. En primer lloc, es van produir i van avaluar estructures gelificades a partir de emulsió-gels d'agar i k-carraguenina i aerogels carregats d'oli com a encapsulants d'un bioactiu lipofílic com la curcumina. Els resultats van mostrar que el tipus de polisacàrid i l'estat físic de les xarxes del gel tenien un impacte en l'estructura dels productes de la digestió.
D'altra banda, es va avaluar el potencial dels emulsió-gels basats en polisacàrids sulfatats (k-carraguenina i agar) per a la producció d'un material capaç de simular teixit gras. Els resultats evidencien que els emulsió-gels d'agar són adequats per a produir materials que simulen les propietats dielèctriques podentimitar teixits de baix i alt contingut en aigua. / [EN] The aim of this doctoral thesis was to design and characterize the structure of gel-like structures based on polysaccharides extracted from aquatic biomass, with interest for food-related applications. The properties of polysaccharides extracted from seaweeds and aquatic plants make them suitable to produce a range of gel-like structures based on the formation of interconnected networks, such as hydrogels, aerogels and emulsion-gels.
In the first part of this thesis, the different gelation mechanism of sulphated polysaccharides and the parameters affecting the structure and functional properties of the obtained hydrogels were investigated. Based on the results, the potential application of agar and k-carrageenan hydrogels and aerogels to encapsulate a model food protein such as casein was evaluated, thus exploring the protective effect against the enzymatic hydrolysis upon simulated gastrointestinal digestions.
In the second part of this thesis, aerogel structures were developed by valorising an underutilized waste biomass source such as Arundo donax. This biomass was used to generate cellulosic fractions with different purification degrees and water-soluble bioactive extracts, which were subsequently used to produce hybrid bioactive aerogels. The highly porous structure and high sorption capacity of aerogels make them excellent candidates for the replacement of absorbent pads to maintain the quality of packaged meat products.
Emulsion-gels are recognized for their great potential as functional ingredients in the food industry to modify texture and for solid fat replacement. Moreover, they can be used as a delivery vehicle for the controlled release of fat-soluble bioactive compounds. Thus, in the last part, the nature of interactions between the components in polysaccharide-based emulsion-gel formulations was investigated and related to their structure and mechanical and rheological behavior. After studying the gelation mechanism of carrageenan emulsion-gels, these systems were adapted and used for two different applications relevant to the food and biomedicine sectors. Firstly, oil-filled gel-like structures from agar and k-carrageenan emulsion-gels and oil-filled aerogels were produced and evaluated as carriers of a lipophilic bioactive such as curcumin. The results showed that the polysaccharide type and the physical state of the gel network had an impact on the structure of the digestion products.
On the other hand, the potential of emulsion-gels based on sulphated polysaccharides (k-carrageenan and agar) for the production of tissue mimicking phantoms was evaluated. Humans and Seagrasses in East Africa : A social-ecological systems approachde la Torre-Castro, Maricela January 2006 (has links)
<p>The present study is one of the first attempts to analyze the societal importance of seagrasses (marine flowering plants) from a Natural Resource Management perspective, using a social-ecological systems (SES) approach. The interdisciplinary study takes place in East Africa (Western Indian Ocean, WIO) and includes in-depth studies in Chwaka Bay, Zanzibar, Tanzania. Natural and social sciences methods were used. The results are presented in six articles, showing that seagrass ecosystems are rich in seagrass species (13) and form an important part of the SES within the tropical seascape of the WIO. Seagrasses provide livelihoods opportunities and basic animal protein, in from of seagrass associated fish e.g. Siganidae and Scaridae. Research, management and education initiatives are, however, nearly non-existent. In Chwaka Bay, the goods and ecosystem services associated with the meadows and also appreciated by locals were fishing and collection grounds as well as substrate for seaweed cultivation. Seagrasses are used as medicines and fertilizers and associated with different beliefs and values. Dema (basket trap) fishery showed clear links to seagrass beds and provided the highest gross income per capita of all economic activities. All showing that the meadows provide social-ecological resilience. Drag-net fishery seems to damage the meadows. Two ecological studies show that artisanal seaweed farming of red algae, mainly done by women and pictured as sustainable in the WIO, has a thinning effect on seagrass beds, reduces associated macrofauna, affects sediments, changes fish catch composition and reduces diversity. Furthermore, it has a negative effect on i.a. women’s health. The two last papers are institutional analyses of the human-seagrass relationship. A broad approach was used to analyze regulative, normative and cultural-cognitive institutions. Cooperation and conflict take place between different institutions, interacting with their slow or fast moving characteristics, and are thus fundamental in directing the system into sustainable/unsustainable paths. Ecological knowledge was heterogeneous and situated. Due to the abundance of resources and high internal control, the SES seems to be entangled in a rigidity trap with the risk of falling into a poverty trap. Regulations were found insufficient to understand SES dynamics. “Well” designed organizational structures for management were found insufficient for “good” institutional performance. The dynamics between individuals embedded in different social and cultural structures showed to be crucial. Bwana Dikos, monitoring officials, placed in villages or landing sites in Zanzibar experienced four dilemmas – kinship, loyalty, poverty and control – which decrease efficiency and affect resilience. Mismatches between institutions themselves, and between institutions and cognitive capacities were identified. Some important practical implications are the need to include seagrass meadows in management and educational plans, addressing a seascape perspective, livelihood diversification, subsistence value, impacts, social-ecological resilience, and a broad institutional approach.</p>

Marine Seaweed Invasions : the Ecology of Introduced <i>Fucus evanescens</i>Wikström, Sofia A. January 2004 (has links)
<p>Biological invasions are an important issue of global change and an increased understanding of invasion processes is of crucial importance for both conservation managers and international trade. In this thesis, I have studied the invasion of the brown seaweed <i>Fucus evanescens</i>, to investigate the fate and effect of a perennial, habitat-forming seaweed introduced to a coastal ecosystem. A long-term study of the spread of <i>F. evanescens</i> in Öresund (southern Sweden) showed that the species was able to expand its range quickly during the first 20 years after the introduction, but that the expansion has been slow during the subsequent 30 years. Both in Öresund and in Skagerrak, the species is largely restricted to sites where native fucoids are scarce. Laboratory experiments showed that the restricted spread of <i>F. evanescens</i> cannot be explained by the investigated abiotic factors (wave exposure and salinity), although salinity restricts the species from spreading into the Baltic Sea. Neither did I find evidence for that herbivores or epibiota provide biotic resistance to the invader. On the contrary, <i>F. evanescens</i> was less consumed by native herbivores, both compared to the native fucoids and to <i>F. evanescens</i> populations in its native range, and little overgrown by epiphytes. Instead, the restricted spread may be due to competition from native seaweeds, probably by pre-occupation of space, and the establishment has probably been facilitated by disturbance. </p><p>The studies provided little support for a general enemy release in introduced seaweeds. The low herbivore consumption of <i>F. evanescens</i> in Sweden could not be explained by release from specialist herbivores. Instead, high levels of chemical anti-herbivore defence metabolites (phlorotannins) could explain the pattern of herbivore preference for different fucoids. Likewise, the low epibiotic colonisation of <i>F. evanescens </i>plants could be explained by high resistance to epibiotic survival. This shows that colonisation of invading seaweeds by native herbivores and epibionts depends on properties of the invading species. The large differences between fucoid species in their quality as food and habitat for epibionts and herbivores imply that invasions of such habitat-forming species may have a considerable effect on a number of other species in shallow coastal areas. However, since <i>F. evanescens</i> did not exclude other fucoids in its new range, its effect on the recipient biota is probably small.</p>

Potencial fotoprotetor de extratos e substâncias isoladas de fungos endofíticos da alga marinha vermelha Bostrychia radicans e de algas originárias da Antártica / Photoprotective potential of extracts and isolated compounds from endophytic fungi of red marine alga Bostrychia radicans and algae originating from AntarcticaTavares, Renata Spagolla Napoleão 14 April 2016 (has links)
A necessidade de proteger a pele contra os raios ultravioleta (UV) é imprescindível, tendo em vista os efeitos deletérios gerados pelos mesmos. Compostos convencionais com atividade fotoprotetora sofrem interações, instabilidade e em sua maioria protegem contra os raios UVB. Poucos compostos apresentam proteção contra os raios UVA. No ambiente marinho, como nas algas marinhas, adversidades do ambiente relacionadas, principalmente, a exposição solar, aumentam as defesas naturais contra os raios UV por meio da produção de metabólitos secundários que podem absorver/refletir os raios solares, ou agir como antioxidantes. Tais moléculas podem ser produzidas pelas próprias algas ou pelos fungos endofíticos associados a elas. Assim, o presente estudo tem como objetivo investigar o potencial fotoprotetor de extratos, frações e substâncias isoladas dos fungos endofíticos Xylaria sp e Annulohypoxylon stygium, associados à alga vermelha Bostrychia radicans. Bem como de quatro espécies de algas da Antártica, Palmaria decipiens, Monostroma hariotii, Desmarestia anceps e a Gigartina skottsbergii. As algas da Antártica foram disponibilizadas pelo grupo do Prof. Dr. Pio Colepicolo Neto do IQ-USP e o cultivo dos fungos, a extração e o isolamento foram desenvolvidos em parceria com o Laboratório de Química Orgânica do Ambiente Marinho - NPPNS da FCFRP-USP. Os extratos e frações obtidos foram submetidos à análise dos espectros de absorção no UV e à fotodegradação. A seguir, os extratos e frações mais promissoras foram submetidos à avaliação da fototoxicidade em cultura de fibroblastos 3T3 para a determinação da viabilidade celular na presença e ausência da radiação, de acordo com o protocolo OECD TG 432. O fungo A. stygium apresentou frações com absorção no UVB, duas subfrações se mostraram não cito/fototóxicas e são provenientes de frações fotoestáveis. Destas subfrações, duas substâncias, ambas inéditas, foram isoladas e elucidadas por RMN H1, 1D, 2D, IV, ESI, e estas foram consideradas potenciais ativos para fotoproteção no UVB. O fungo Xylaria sp apresentou frações com absorção na região do UV, porém apenas uma, com absorção no UVB se mostrou fotoestável. Esta mesma fração foi considerada cito e fototóxica. Não foi possível identificar as duas substâncias isoladas pela baixa massa. Entretanto, um metabólito já isolado dessa linhagem anteriormente, o ácido gentísico, foi considerado não fototóxico, devendo ser melhor investigado quanto ao seu potencial como filtro biológico. Quanto às macroalgas antárticas, três espécies apresentaram absorção no UV. Apesar de a maioria dos extratos terem sido considerados fotoinstáveis, o extrato da alga D. anceps, de maior rendimento, apresentou frações com ampla absorção no UVA/VIS, no entanto elevada citotoxicidade. Desta fração foi isolada e identificada a fucoxantina (RMN H1 1D e 2D, ESI), um carotenoide marinho que apresentou potencial fototóxico no modelo monocamadas, mas não foi considerada citotóxica. Sendo assim, este pode ser considerado um promissor candidato a ativo cosmético, pelas suas propriedades antioxidantes e de filtro biológico, pois devido a sua alta massa molecular, 658.90 g/mol, pode-se inferir baixa permeação nas camadas viáveis da pele e ausência de fototoxicidade in vivo. Nesse sentido, novos estudos em modelo de pele 3D deverão ser realizados a fim de se comprovar a segurança de uso tópico da fucoxantina. As espécies também devem ser investigadas para outras atividades biológicas uma vez que este material pertence a uma região pouco estudada e estas podem apresentar potencial para os mais diversos empregos farmacológicos ou cosméticos. / The need to protect the skin against ultraviolet (UV) radiation is essential, due to the deleterious effects caused by them. Conventional UV-filters undergo chemical interactions, instability and mostly protect against UVB rays. Few compounds have protection against UVA rays. In the marine environment, especially in marine algae, adversities, mainly related to sun exposure, increases the natural defenses against UV radiation through the production of secondary metabolites that can absorb/reflect the UV radiation, or even act as antioxidants. Such molecules may be produced by algae themselves or by the endophytic fungi associated with them. The present study aims to investigate the potential of new sunscreen extracts, fractions and isolated compounds from endophytic fungi Xylaria sp and Annulohypoxylon stygium, associated with red algae Bostrychia radicans, and four species of algae from Antarctica: Palmaria decipiens, Monostroma hariotii, Desmarestia anceps and Gigartina skottsbergii. The algae material from Antarctica were given by the group of Prof. Dr. Pio Colepicolo Neto, IQ-USP, and the fungi cultivation, extractions and the compounds isolation were obtained in partnership with the Laboratory of Organic Chemistry of the Marine Environment - NPPNS of FCFRP-USP. The extracts and fractions obtained were submitted to analysis of the absorption spectra in the UV and to photodegradation. After that, the most promising extracts and fractions were submitted to the assessment of phototoxicity in 3T3 fibroblasts in culture for determination of cell viability in the presence and absence of radiation, in accordance with the OECD TG 432 protocol. The fungus A. stygium showed fractions with UVB absorption, from there were isolated two novel compounds and had the structure elucidated (RMN H1 1D/2D; IV; ESI). These compounds were considered potential ingredients for photoprotection on the UVB range. The fungus Xylaria sp. showed fractions with absorption on the UV, but only one of them, with UVB absorption, was considered photostable. The same fraction was also considered cyto/ phototoxic. It was not possible identify two of the compounds isolated due its low weight. Therefore, the gentisic acid, a metabolite already isolated from this fungus, was not considered phototoxic, what suggests that it should be better investigated as a potential biological UV filter. Regarding the seaweeds, three species have shown absorption in UV region. Most of the extracts were considered photounstable, but the D. anceps extract, of highest yield, showed fractions with large UVA/VIS absorption, but higher cytotoxic potential. Fucoxanthin, a marine carotenoid, was isolated from this fraction and identified by NMR H1 1D/2D, ESI. This compound showed phototoxic potential in a monolayer model, but it was not considered cytotoxic. Therefore, fucoxanthin is a promising candidate to a cosmetic ingredient, once it has antioxidant and biological UV filter properties and its high molecular weight, 658.90 g/mol, suggests low skin permeation into viable epidermis and absence of in vivo phototoxicity. Thus, more studies using 3D skin model should be performed in order to prove the safety of the fucoxanthin topical use. The species should be also investigated for other biological activities once this material belongs to a poorly studied region and can show potential for many of the pharmacological/cosmetics activities.

Notre travail est une recherche originale qui apporte à la géographie des littoraux et de la mer un nouvel exemple d’étude. Nous proposons d’analyser les dynamiques spatiales et économiques d’une aquaculture singulière : l’algoculture. La culture des algues est un domaine significatif de la géographie littorale comme de la géographie des ressources alimentaires et énergétiques. Nous avons identifié à plusieurs échelles les territoires productifs et aménagés de l’algoculture des macro-algues comme des micro-algues. Pour mener ce projet de recherche, nous avons choisi trois pays : le territoire chinois qui est le premier producteur de macro-algues, le territoire français qui adapte sa production traditionnelle à une nouvelle demande commerciale et industrielle et le territoire des États-Unis qui connaît le plus d’innovations dans le domaine des micro-algues. Cette thèse a une démarche transversale et prospective et s’appuie sur plusieurs méthodes de recherche : relevés de terrain sur des sites de production, entretiens avec les acteurs, chercheurs, distributeurs, recherche bibliographique à la croisée des sciences biologiques et humaines. Cette étude des cultures, des territoires et des enjeux de l’algoculture progresse en trois étapes : tout d’abord la présentation de l’algoculture (macro et micro-algue) dans sa production globale et régionale avec les modes de production et les valorisations, puis l’analyse des filières algacoles en montrant leurs aménagements locaux et l’investissement des acteurs territoriaux dans les territoires de référence ; enfin les algues sont une ressource qui participent à des enjeux énergétiques, environnementaux et alimentaires du XXIe siècle. / Our work is an original research that brings a new study example to the geography of sea and coastlines. We propose to describe and analyze the spatial and economic dynamics of a singular type of aquaculture: seaweed farming. Seaweed farming is a significant research area in coastal geography as well as in the geography of food and energy. We sought to identify the different scales of territories organized for the production of both macro- and microalgae. To conduct this research, we chose three study countries : China, the largest producer of macroalgae, France, where traditional production is being adapted to a new commercial and industrial demand, and finally the United States, where most innovations in the field of microalgae are made. This thesis has a cross- and forward-looking approach and is based on different research methods: field surveys on production sites, interviews with stakeholders, researchers, and distributors, bibliographic research at the intersection of the biological and social sciences. The study of crops, territories and issues of seaweed farming progresses along a three-step analysis: first, the presentation of algae farming (macro and microalgae) in its global and regional production with the production methods, second the analysis of the algae value chains, showing the development of local equipment and installations, and the investment of authorities in the territories of reference. Finally, we explore algae as a resource involved in energy, environmental and food issues of the twenty-first century.

A natureza sempre constituiu uma fonte extremamente rica de compostos e substâncias utilizados pelo homem para diferentes fins, principalmente na área médica, pois os produtos naturais abrangem uma variedade de agentes terapêuticos para diversas doenças, como moléstias infecciosas, câncer, desordens lipídicas e imunológicas. Com a necessidade de se encontrar novos fármacos, tanto para doenças bem definidas, quanto para novas doenças, a pesquisa de substâncias com atividades biológicas torna-se fundamental. Fontes exóticas como o ambiente marinho e, recentemente o universo dos micro-organismos, ganharam grande interesse no meio científico devido ao fato de terem sido pouco conhecidas e exploradas. Nesse contexto, o objetivo desse estudo foi avaliar se diferentes frações da macroalga marinha Bostrychia tenella e extratos de micro-organismos endofíticos isolados dessa mesma espécie possuíam atividade imunomoduladora sobre o sistema complemento (SC) humano, que é reconhecido como um dos maiores efetores do processo inflamatório. Iniciado por três vias distintas (clássica, alternativa e das lectinas), ele leva à formação de produtos de clivagem e complexos de ativação que exercem diferentes funções biológicas importantes, como quimiotaxia, ativação de fagócitos, desgranulação de mastócitos, citólise, ativação celular e apoptose. Sendo assim, este sistema desempenha papel importante em várias reações inflamatórias, bem como em diferentes mecanismos patogênicos de dano tecidual. Através do ensaio de atividade hemolítica, 10 das 20 amostras iniciais foram selecionadas por modularem esta atividade do SC no pool de soro humano normal (SHN). As 10 amostras (dentre frações, subfrações e extratos de B. tenella e seus micro-organismos endofíticos) foram capazes de induzir a geração de fragmentos de C3, componente central do SC, observados tanto em técnicas imunoeletroforéticas, quanto em Western blotting. Também, observou-se a formação do complexo de ataque à membrana (MAC) em ensaio imunoenzimático, indicando que houve ativação total do SC. Através dos resultados obtidos no Western blotting para fragmentos de C4 e Fator B foi possível identificar quais vias do SC são ativadas, quando correlacionados com os resultados dos ensaios hemolíticos para via clássica/das lectinas (VC/VL) e via alternativa (VA). Desse modo, foi possível concluir que as 10 amostras selecionadas possuem efeito imunomodulador sobre o SC humano por serem capazes de ativá-lo, seja pela VC/VL ou pela VA. Conclui-se, também, que o ensaio hemolítico cinético constitui uma importante ferramenta de triagem, assim como o Western blotting do SHN para detecção de fragmentos do SC. / Nature has always provided an extremely rich source of compounds and substances used by man for various purposes, mainly in the medical field because natural products include a variety of therapeutic agents for various diseases such as infectious diseases, cancer, lipid and immune disorders. With the need to find new drugs for either well defined diseases as for new ones, the search for substances with biological activities becomes essential. Exotic sources such as marine environment, and recently the world of micro-organisms have gained great interest in the scientific community due to the fact that they were little known and explored. In this context, the objective of this study was to assess whether different fractions of marine macroalgae Bostrychia tenella and extracts of endophytic micro-organisms isolated from this species possess immunomodulatory activity on the human complement system (CS), which is recognized as one of the major effectors of the inflammatory process. Started by three distinct pathways (classical, alternative and lectin), it leads to the formation of cleavage products and activation complexes which have various important biological functions such as chemotaxis, activation of phagocytes, mast cell degranulation, cytolysis, cellular activation and apoptosis. Thereby, this system plays an important role in various inflammatory reactions, as well as various pathogenic mechanisms of tissue damage. Through the hemolytic activity assays, 10 of the initial 20 samples were selected because they were able to modulate this activity of the CS in the pool of normal human serum (NHS). The 10 samples (among fractions, subfractions and extracts of B. tenella and its endophytic micro-organisms) were able to induce the generation of fragments from C3, a central component of the CS, which was observed both in immuno electrophoresis technique and Western blotting. Also, we observed the formation of the membrane attack complex (MAC) in enzyme immunoassay, indicating that there was total activation of the CS. By the results obtained in Western blotting for fragments of C4 and Factor B was possible to identify which pathways are activated in CS, when correlated with the results of hemolytic assays for the classical/lectin (VC/VL) and alternative pathway (VA). Thus, we conclude that the 10 selected samples have immunomodulatory effects in human CS because they were able to activate it either by VC/VL or VA. It follows also that the hemolytic assay by the kinetic method is an important tool for screening, as well as Western blotting of NHS to detect fragments of the CS.

A extração de Carne Mecanicamente Separada de tilápia tem se destacado como um processo atraente pela possibilidade de maior recuperação da carne após a filetagem. Porém, a separação mecânica aumenta a superfície de exposição, levando à incorporação de oxigênio e consequentemente ao \"off flavor\" devido à rancidez, tornando necessário o uso de aditivos para sua conservação. A tendência é utilizar produtos naturais como alternativas aos aditivos químicos. Entre os produtos naturais, as algas marinhas apresentam, em sua composição, metabólitos secundários com alta atividade antioxidante e antimicrobiana. Esta pesquisa teve como objetivo avaliar a composição química e a atividade biológica de quatro algas marinhas e seus efeitos, quando aplicados em Minced de tilápia. As algas Nori, Kombu, Hijiki e Wakame foram extraídas por 2 e 7 dias, em temperatura ambiente, com etanol 60, 80 e 100%. O teor de compostos fenólicos, a atividade antioxidante e a antimicrobiana in vitro foram determinados. A atividade antioxidante por métodos acelerados, Rancimat e Oxipres, também foi avaliada. As algas bioativas tiveram seu perfil químico elucidado por cromatografia líquida e gasosa. Essas algas foram aplicadas em Minced de tilápia, que, em seguida, foi armazenado a -18ºC por 180 dias. No Minced foram analisadas a composição centesimal, características de frescor por pH, BNVT e TBARS; cor instrumental, cor e aroma (de ranço) pela análise sensorial, padrões microbiológicos e composição de ácidos graxos, nos tempos 0, 60, 120 e 180 dias de armazenamento. Observou-se que a extração de 2 dias, com etanol 60%, foi a mais eficiente para extrair os compostos fenólicos. As algas Nori e Hijiki apresentaram os maiores teores desses compostos e, consequentemente, maior atividade antioxidante in vitro. Os extratos de algas não apresentaram atividade antimicrobiana contra Salmonella Enteritidis, Escherichia coli e Staphylococcus aureus. Para Klebsiella pneumoniae e Listeria monocytogenes, os extratos mais eficientes foram os extraídos com etanol 100%. As algas Nori e Hijiki foram selecionadas como as mais bioativas e submetidas aos testes de oxidação acelerada. Quando aplicadas em óleo de soja, no Rancimat, e em Minced de tilápia, no Oxipres, estas algas demonstraram efetiva atividade antioxidante. Pela cromatografia, os principais compostos identificados foram, na Nori, os ácidos clorogênico, vanílico e caféico, além dos aminoácidos alanina, glicina, valina e prolina; bem como, glicose e sorbitol. Na alga Hijiki foram detectados os ácidos clorogênico, caféico e cinâmico; alanina e prolina, bem como, xilitol e ribitol, frutose e ácido linoléico. No teste de armazenamento congelado do Minced, por 180 dias, a aplicação dos extratos de Nori e Hijiki não infuenciou na composição centesimal e pH. As algas mostraram-se eficientes no controle da produção das BNVT. No entanto, por TBARS, apesar de estarem dentro dos limites aceitáveis, as algas apresentaram comportamento pró-oxidante. Sob os aspectos microbiológicos, os minceds atenderam à legislação vigente. Sensorialmente, os provadores não detectaram diferença no \"aroma de ranço\", apenas mínimas diferenças na cor do produto. Pode-se concluir que o Minced de tilápia, adicionado de algas marinhas, manteve-se dentro dos padrões de qualidade durante o armazenamento congelado. Ressalta-se, ainda, a diferença na resposta antioxidante de acordo com o método utilizado / The extraction of minced tilapia has emerged as an attractive process due to the possibility of greater recovery of the tissue after filleting. However, mechanical separation increases the surface exposure, leading to incorporation of oxygen and consequently to the \"off-flavor\" due to rancidity, making necessary the use of additives for its preservation. The tendency is to use natural products as alternatives to chemical additives. Among the natural products, seaweeds present in its composition secondary metabolites with high antioxidant and antimicrobial activities. This research aimed to evaluate the chemical composition and biological activities of four seaweeds varieties and their effects when applied to the tilapia Minced. Nori, Kombu, Hijiki and Wakame seaweeds were extracted by 2 and 7 days, at room temperature, with ethanol 60, 80 and 100%. The phenolic content, antioxidant and antimicrobial activities in vitro were determined. The antioxidant activity by accelerated methods, Oxipres and Rancimat, were also evaluated. Seaweeds bioactive profiles were elucidated by liquid and gas chromatography. These seaweeds were applied to the tilapia Minced, that was stored at -18 °C for 180 days. The Minced were analyzed as to their composition, freshness characteristics by pH, TBARS and TVB-N, instrumental color, color and rancidity aroma by sensory analysis, microbiological standards, and fatty acid composition, at 0, 60, 120 and 180 days of storage. It was observed that the extraction after two days, with 60% ethanol, was the most efficient way to extract the phenolic compounds. Hijiki and Nori showed the highest levels of these compounds and therefore higher antioxidant activity in vitro. The extracts showed no antimicrobial activity against Salmonella Enteritidis, Escherichia coli and Staphylococcus aureus. For Klebsiella pneumoniae and Listeria monocytogenes, more efficient extracts were extracted with ethanol 100%. Hijiki and Nori seaweeds were selected as the most bioactive and submitted to accelerated oxidation tests. When applied in soy oil in the Rancimat, and tilapia Minced, in the Oxipres, these seaweeds have demonstrated effective antioxidant activity. By chromatography, the most important compounds identified were, in Nori, chlorogenic, caffeic and vanillic acids, besides the amino acids alanine, glycine, proline and valine, glucose and sorbitol. In the Hijiki seaweed, it was detected chlorogenic, caffeic and cinnamic acids, alanine and proline, as well as, ribitol and xylitol, fructose, and linoleic acid. In the test of frozen Minced storage, for 180 days, the application of Nori and Hijiki extracts did not modify neither the composition nor the pH. Seaweeds were effective in controlling the TVB-N production, but, for TBARS, although they were within acceptable limits, the seaweeds showed pro-oxidant activities. As to the microbiological aspects, the minceds complied with legal requirements. Sensorially, the tasters detected no difference in the \"rancid aroma\", only small differences in the product color. It can be concluded that the Minced tilapia, seaweed added, remained within the standards of quality during frozen storage. It should be noted, moreover, the difference in the antioxidant response, according to the method used.

Anthropogenic Disturbances and Shifts in Tropical Seagrass EcosystemsEklöf, Johan S. January 2008 (has links)
<p>Seagrasses constitute the basis for diverse and productive ecosystems worldwide. In East Africa, they provide important ecosystem services (e.g. fisheries) but are potentially threatened by increasing resource use and lack of enforced management regulations. The major aim of this PhD thesis was to investigate effects of anthropogenic distur-bances, primarily seaweed farming and coastal fishery, in East African seagrass beds. Seaweed farming, often depicted as a sustainable form of aquaculture, had short- and long-term effects on seagrass growth and abundance that cascaded up through the food web to the level of fishery catches. The coastal fishery, a major subsistence activity in the region, can by removing urchin predators indirectly increase densities of the sea urchin <i>Tripneustes gratilla</i>, which has overgrazed seagrasses in several areas. A study using simulated grazing showed that high magnitude leaf removal – typical of grazing urchins – affected seagrasses more than low magnitude removal, typical of fish grazing. Different responses in two co-occurring seagrass species furthermore indicate that high seagrass diversity in tropical seagrass beds could buffer overgrazing effects in the long run. Finally, a literature synthesis suggests that anthropogenic disturbances could drive shifts in seagrass ecosystems to an array of alternative regimes dominated by other or-ganisms (macroalgae, bivalves, burrowing shrimp, polychaetes, etc.). The formation of novel feedback mechanisms makes these regimes resilient to disturbances like seagrass recovery and transplantation projects. Overall, this suggests that resource use activities linked to seagrasses can have large-scale implications if the scale exceeds critical levels. This emphasizes the need for holistic and adaptive management at the seascape level, specifically involving improved techniques for seaweed farming and fisheries, protection of keystone species, and ecosystem-based management approaches.</p>

Marine Seaweed Invasions : the Ecology of Introduced Fucus evanescensWikström, Sofia A. January 2004 (has links)
Biological invasions are an important issue of global change and an increased understanding of invasion processes is of crucial importance for both conservation managers and international trade. In this thesis, I have studied the invasion of the brown seaweed Fucus evanescens, to investigate the fate and effect of a perennial, habitat-forming seaweed introduced to a coastal ecosystem. A long-term study of the spread of F. evanescens in Öresund (southern Sweden) showed that the species was able to expand its range quickly during the first 20 years after the introduction, but that the expansion has been slow during the subsequent 30 years. Both in Öresund and in Skagerrak, the species is largely restricted to sites where native fucoids are scarce. Laboratory experiments showed that the restricted spread of F. evanescens cannot be explained by the investigated abiotic factors (wave exposure and salinity), although salinity restricts the species from spreading into the Baltic Sea. Neither did I find evidence for that herbivores or epibiota provide biotic resistance to the invader. On the contrary, F. evanescens was less consumed by native herbivores, both compared to the native fucoids and to F. evanescens populations in its native range, and little overgrown by epiphytes. Instead, the restricted spread may be due to competition from native seaweeds, probably by pre-occupation of space, and the establishment has probably been facilitated by disturbance. The studies provided little support for a general enemy release in introduced seaweeds. The low herbivore consumption of F. evanescens in Sweden could not be explained by release from specialist herbivores. Instead, high levels of chemical anti-herbivore defence metabolites (phlorotannins) could explain the pattern of herbivore preference for different fucoids. Likewise, the low epibiotic colonisation of F. evanescens plants could be explained by high resistance to epibiotic survival. This shows that colonisation of invading seaweeds by native herbivores and epibionts depends on properties of the invading species. The large differences between fucoid species in their quality as food and habitat for epibionts and herbivores imply that invasions of such habitat-forming species may have a considerable effect on a number of other species in shallow coastal areas. However, since F. evanescens did not exclude other fucoids in its new range, its effect on the recipient biota is probably small.

Humans and Seagrasses in East Africa : A social-ecological systems approachde la Torre-Castro, Maricela January 2006 (has links)
The present study is one of the first attempts to analyze the societal importance of seagrasses (marine flowering plants) from a Natural Resource Management perspective, using a social-ecological systems (SES) approach. The interdisciplinary study takes place in East Africa (Western Indian Ocean, WIO) and includes in-depth studies in Chwaka Bay, Zanzibar, Tanzania. Natural and social sciences methods were used. The results are presented in six articles, showing that seagrass ecosystems are rich in seagrass species (13) and form an important part of the SES within the tropical seascape of the WIO. Seagrasses provide livelihoods opportunities and basic animal protein, in from of seagrass associated fish e.g. Siganidae and Scaridae. Research, management and education initiatives are, however, nearly non-existent. In Chwaka Bay, the goods and ecosystem services associated with the meadows and also appreciated by locals were fishing and collection grounds as well as substrate for seaweed cultivation. Seagrasses are used as medicines and fertilizers and associated with different beliefs and values. Dema (basket trap) fishery showed clear links to seagrass beds and provided the highest gross income per capita of all economic activities. All showing that the meadows provide social-ecological resilience. Drag-net fishery seems to damage the meadows. Two ecological studies show that artisanal seaweed farming of red algae, mainly done by women and pictured as sustainable in the WIO, has a thinning effect on seagrass beds, reduces associated macrofauna, affects sediments, changes fish catch composition and reduces diversity. Furthermore, it has a negative effect on i.a. women’s health. The two last papers are institutional analyses of the human-seagrass relationship. A broad approach was used to analyze regulative, normative and cultural-cognitive institutions. Cooperation and conflict take place between different institutions, interacting with their slow or fast moving characteristics, and are thus fundamental in directing the system into sustainable/unsustainable paths. Ecological knowledge was heterogeneous and situated. Due to the abundance of resources and high internal control, the SES seems to be entangled in a rigidity trap with the risk of falling into a poverty trap. Regulations were found insufficient to understand SES dynamics. “Well” designed organizational structures for management were found insufficient for “good” institutional performance. The dynamics between individuals embedded in different social and cultural structures showed to be crucial. Bwana Dikos, monitoring officials, placed in villages or landing sites in Zanzibar experienced four dilemmas – kinship, loyalty, poverty and control – which decrease efficiency and affect resilience. Mismatches between institutions themselves, and between institutions and cognitive capacities were identified. Some important practical implications are the need to include seagrass meadows in management and educational plans, addressing a seascape perspective, livelihood diversification, subsistence value, impacts, social-ecological resilience, and a broad institutional approach.
