Quorum Sensing and Candida Albicans

Candida albicans is one of the most commonly identified nosocomially acquired pathogens. This organism has a number of virulence traits including production of degrading enzymes, the ability to undergo phenotypic switching, and can rapidly undergo morphogenic switch from a blastospore (yeast) phase to that of a hyphal state. Interest in C. albicans morphogenic regulation has been the focus of a large number of studies, which have characterised transcriptional modulators of these morphologies. Recently, C. albicans has been shown to regulate its morphogenic shift through changes in cell density. It was observed that C. albicans inoculated at cell densities below 106 cells ml -1 under conditions which favour hyphal morphogenesis (pH 7.5, 37°C), will germinate to form hyphae. However, if cells densities are greater than 106 cells ml-1, little germination will occur and cells will maintain yeast morphology. The basis for this cell-density-dependent control of morphogenesis is similar to that which is seen with bacterial cells regulating their activities via quorum sensing (QS). A number of molecules have been identified which affect the ability of C. albicans to undergo the yeast-to-hyphal shift, and three compounds have been demonstrated to be quorum-sensing molecules. The scope of this review is to bring to light what is now understood about QS in C. albicans and address the roles of these molecules in relation to virulence in the host and potential roles in cross-kingdom interactions.

Tyrosol has been formerly confirmed as a substance which shortens the lag phase of Candida albicans cell cycle and stimulates its conversion to the hyphal form, farnesol stimulates blastospores. This work covers the effect of tyrosol combined with two antimycotics, fungistatic fluconazole and fungicidal amphotericine B. Candida albicans strains used in the experiment were mostly isolated from samples taken from patients suffering from variously located Candida albicans infections and four laboratory standard strains were also used. The method used for the experiment was a modified broth dilution method using combinations of both tyrosol and antimycotics various concentrations, which were prepared by two-fold dilution. Using amphotericine B, 28 strains did not show any difference in MIC compared with the control sample, 6 strains showed mildly higher MIC and 3 strains shower mildly lower MIC. Candida lusitaniae est une levure pathogène opportuniste émergente, responsable d’infections sévères chez les sujets immunodéprimés, et constitue un important modèle d’étude fonctionnelle de gènes par génétique inverse. Cette levure est souvent associée à des résistances aux antifongiques, d’où la nécessité de mieux comprendre l’interaction entre l’hôte et le pathogène, afin d’identifier de nouvelles cibles thérapeutiques potentielles. Les molécules signal sont décrites comme modulatrices de cette interaction. Au cours de ce travail, nous avons créé un mutant dpp3Δ, inactivé pour une pyrophosphate phosphatase, chez C. lusitaniae. La mutation a pour conséquence de modifier le taux secrété de PEA/tyrosol, des alcools aromatiques récemment décrits comme molécules signal chez les espèces Candida. Les levures du mutant dpp3Δ présentent aussi un défaut de pseudo-filamentation et de reproduction sexuée. Comparé à la souche sauvage, le mutant dpp3Δ module différemment la réponse macrophagique, notamment la production de NO et ROS, ainsi que la sécrétion de TNF-α et d’IL-10. Curieusement, la sécrétion d’IL-10 est modulée par les levures même sans contact physique avec les macrophages. Par ailleurs, nous avons observé des effets du traitement des levures par le PEA ou le tyrosol sur la modification des réponses immunes macrophagiques. Enfin, nous avons montré que le gène DPP3 chez C. lusitaniae était un facteur de virulence essentiel dans des modèles murins d’infection, et qu’il augmentait le taux de TNF-α secrété et la colonisation des cerveaux. / C. lusitaniae, an emerging Candida species often associated with antifungal resistance, is an important model to study gene function by reverse genetics. Understanding the host-pathogen interaction helps identifying new virulence factors and potentially new antifungal targets. Signaling molecules are often reported as modulators of this interaction. Here we created a dpp3Δ knocked-out mutant, lacking a pyrophosphate phosphatase, in C. lusitaniae. This mutation modified the ratio of secreted PEA/tyrosol, two newly reported signaling molecules in Candida species. Colony morphology of yeast cells was also altered as yeasts had a defect in pseudo-filamentation, and mating capacity was severely reduced. Compared to the wild-type strain, the dpp3Δ knocked-out mutant differently affected NO and ROS production by macrophages as well as TNF-α and IL-10 secretion. Interestingly IL-10 secretion was found to be modulated by C. lusitaniae without the need of a physical contact with the phagocytes. Moreover we elucidated the effects of PEA and tyrosol on yeast cells leading to modulations in macrophage immune responses. At last, the DPP3 gene was found to be an essential pathogenicity factor in mice models, leading to an increase of TNF-α secretion and brain colonization.

Développement de matériaux à base d'argiles pour l'oxydation catalytique de polluants organiques par des réactions de type Fenton / Development of materials based on clays for the catalytic oxidation of organic pollutants by Fenton-type reactions

Dans ce travail, nous nous sommes intéressés à la synthèse et à la caractérisation de matériaux à base d'argiles naturelles en provenance d'Algérie et de Tunisie, pouvant être utilisés en tant que catalyseurs dans l'oxydation de polluants organiques par les réactions Fenton et photo-Fenton. Nous avons également optimisé les paramètres réactionnels (quantité de catalyseur, concentration en polluant...) dans le but d'améliorer les performances catalytiques de ces derniers. En effet, l'oxydation de polluants organiques récalcitrants par des réactions de type Fenton est considérée comme l'une des techniques les plus performantes parmi les procédés d'oxydation avancée (POA). Elle permet d'aboutir dans certains cas au stade ultime de l'oxydation qui est la minéralisation (transformation en CO₂ et H₂O). La caractérisation des argiles à l'état brut par différentes techniques physico-chimiques montre qu'en plus des matériaux argileux, il existe quelques oxydes métalliques dans toutes les argiles étudiées, avec des proportions plus ou moins variées. En particulier, la smectite a montré une contenance assez importante en fer. Les argiles synthétisés montrent une très bonne performance catalytique en un minimum de temps (2 heures de réaction). Le catalyseur à base de smectite calciné à 450°C et tamisé donne le meilleur rendement catalytique. Cette activité peut être expliquée par la forte teneur en fer ainsi qu'à la combinaison entre le choix de la taille des particules et la température de calcination de la smectite. La caractérisation de ce catalyseur a montré que ce matériau mésoporeux contenait une quantité de fer (III) stabilisée dans la structure par le fait de la calcination. / In this study, we are interested to the synthesis and charaterization of materials based on natural clays from Algeria and Tunisia, usable as catalysts in the oxidation of organic pollutants by the Fenton's and photo-Fenton's reaction. We have also optimized the reaction parameters (amount of catalyst, concentration of pollutant...) in order to improve the catalytic performance of these catalysts. Indeed, the oxidation of recalcitrant organic pollutants by Fenton-type reactions is regarded as one of the most effective method amongst the advanced oxidation process (AOPs). It can lead in some cases to the total mineralization of pollutants (conversion into CO₂ and H₂O). The caracterization of the raw clays various physicochemical methods shows that, in addiction to clay minerals, there are some metallic oxides in all studied clays, with varied proportions. Especially, smectite showed a fairly significant iron capacity. The synthesized clays show a good catalytic performance in minimum of time (2 hours of reaction). The smectite catalyst, sieved and calcined at 450°C gives the best performance. This activity can be explained by the high content of iron as well as the combination of the choice of particle size and the calcination temperature of the smectite. The caracterization of this catalyst, showed that this mesoporous material contains an amount of iron (III) stabilized in the structure by the fact of calcination.

Previous research of colonies formed by yeast Saccharomyces cerevisiae growing on glycerol agar medium revealed two major cell types of U and L cells that are formed within these colonies. This colonial cell differentiation seem to be caused by communication among yeast cells as well as whole colonies and affected by changes in the environment (for example changes in nutrients). Studies of U and L cells showed that U cells are more resistant against biological, chemical and physical stresses than L cells. The aim of this thesis was to isolate U and L cell types and investigate their resistance against selected low molecular weight chemical substances produced in Ehrlich pathway. Ehrlich pathway was discovered in 1907 and was classified as amino acid catabolic pathway in yeast S. cerevisiae. The low molecular intermediates are formed in Ehrlich pathway which are called fusel (original name from German) alcohols and acids. These chemical substances are widely used in food industry and cosmetics especially because of their aroma. Several studies provided indications that these chemical substances may affect development of colonies and important yeast functions such as switching to the pseudohypfal growth of S. cerevisiae cells. Molecular mechanisms involved in the protective effect of Mediterranean diet and olive oil consumption in humans

The scope of the present work was to investigate whether the protective role of the traditional Mediterranean diet (TMD), and virgin olive oil (VOO) rich in phenolic compounds (PC), towards cardiovascular disease can be mediated through gene expression changes. Two trials were performed to assess the in vivo nutrigenomic effects of TMD and VOO in healthy volunteers. The results point out: a) significant gene expression changes of those genes related with cardiovascular-risk processes after VOO ingestion; b) a down-regulation in the expression of atherosclerosis-related genes after a 3-month intervention with a TMD; and c) an olive oil PC health-protective nutrigenomic effect within the frame of the TMD. Changes in gene expression were concomitant with decreases in oxidative damage and systemic inflammation markers. Data from our studies provide further evidence to recommend both the TMD and the VOO as a useful tool for the prevention of atherosclerosis. / El objetivo de este estudio es investigar si el papel protector de la dieta Mediterránea tradicional (TMD) y del aceite de oliva virgen (VOO), rico en compuestos fenólicos (PC), puede ser mediado a través de cambios en la expresión génica. Se realizaron dos ensayos clínicos para evaluar los efectos nutrigenómicos de la TMD y del VOO, in vivo, en voluntarios sanos. Los resultados mostraron a) cambios en la expresión génica de genes relacionados con el riesgo cardiovascular tras la ingestión del aceite virgen de oliva, b) una infra-expresión en la expresión de genes relacionados con el proceso aterosclerótico tras una intervención con TMD de 3 meses y c) que los compuestos fenólicos del aceite de oliva ejercen un efecto nutrigenómico protector en el marco de la TMD. Los cambios en la expresión génica fueron coherentes.

The present Doctoral Thesis focuses on the investigation, characterization and influence of polyacrylic materials in different scientific and technological disciplines via a detailed computer simulation using the Molecular Dynamics (MD) technique, in conjunction with the very accurate, all-atom Dreiding force-field. The main research concepts and objectives are discussed and analyzed in three separate parts. In the first part, atomistic configurations of two model pressure-sensitive acrylic adhesives (PSAs), the atactic homopolymer poly(n-BA) [poly(n-butyl acrylate)] and the atactic copolymer poly(n-BA-co-AA) [poly(n-butyl acrylate-co-acrylic acid)] in the bulk phase or confined between two selected substrates, glassy silica (SiO2) and metallic α-ferrite (α-Fe), were built and simulated by MD in the NPT statistical ensemble. First, an equilibration cycle consisting of temperature annealings and coolings was followed, in order to generate well-equilibrated configurations of the PSA systems. Detailed results from the atomistic simulations are presented concerning their volumetric behavior, glass transition temperature, conformational, structural, viscoelastic and dynamic properties. Particular emphasis was given to the analysis and characterization of the hydrogen bonds that form in the poly(n-BA-co-AA) system. By analyzing the MD trajectories, poly(n-BA-co-AA) was found to exhibit a higher density than poly(n-BA) by about 7% at all temperatures, to be characterized by smaller-size chains for a given molecular weight (MW), to exhibit significantly slower terminal and segmental dynamics properties, and to be characterized by a glass transition temperature that was approximately 40% higher than that of poly(n-BA). We also examined the type and degree of adsorption of the two acrylic systems on the selected substrates by analyzing the MD results for the local mass density as a function of distance from the solid plane and the distribution of adsorbed chain segments in train, loop, and tail conformations, and by computing the work of adhesion at the two substrates. The results revealed a stronger adsorption for both acrylics on the SiO2 surface due to highly attractive interactions between polymer molecules and substrate atoms, and as a consequence a higher value for the work of adhesion compared to that on the α-Fe surface. Furthermore, we have developed a generalized non-equilibrium molecular dynamics (NEMD) algorithm to simulate the mechanical response of the two adhesives under a uniaxial stretching deformation. In the second part of the Thesis, results have been obtained from a hierarchical simulation methodology that led to the prediction of the thermodynamic, conformational, structural, dynamic and mechanical properties of two polymer nanocomposites based on syndiotactic poly(methyl methacrylate) or sPMMA. The first was reinforced with uniformly dispersed graphene sheets and the second with fullerene particles. How graphene functionalization affects the elastic constants of the resulting nanocomposite has also been examined. The phase behavior of the nanocomposite (in particular as we varied the relative size between the sPMMA chains and the diameter of fullerene molecules) has also been studied as a function of fullerene volume fraction. The simulation strategy entailed three steps: 1) Generation of an initial structure, which was then subjected to potential energy minimization and detailed molecular dynamics (MD) simulations at T = 500K and P = 1atm to obtain well relaxed melt configurations of the nanocomposite. 2) Gradual cooling of selected configurations down to room temperature to obtain a good number of structures representative of the glassy phase of the polymer nanocomposite. 3) Molecular mechanics (MM) calculations of its mechanical properties following the method originally proposed by Theodorou and Suter. By analyzing the results under constant temperature and pressure, all nanocomposite systems were found to exhibit slower terminal and segmental relaxation dynamics than the pure polymer matrices. The addition of a small fraction of graphene sheets led in all cases to the enhancement of the elastic constants; this was significantly more pronounced in the case of functionalized graphene sheets. We further mention that, for all polymer/fullerene nanocomposites addressed here, no phase separation or variation of polymer chain dimensions was observed as a function of fullerene size and/or fullerene volume fraction. In the third part of the Thesis, and motivated by the use of acrylic polymers for the design of membranes with aligned carbon nanotubes (CNTs) for several separation technologies (such as water desalination and wastewater treatment), we report results from a detailed computer simulation study for the nano-sorption and mobility of four different small molecules (water, tyrosol, vanillic acid, and p-coumaric acid) inside smooth single-wall CNTs (SWCNTs). Most of the results have been obtained with the molecular dynamics (MD) method, but especially for the most narrow of the CNTs considered, the results for water molecule were further confirmed through an additional Grand Canonical (μVT) Monte Carlo (GCMC) simulation using a value for the water chemical potential μ pre-computed with the particle deletion method. Issues addressed in the Thesis include molecular packing and ordering inside the nanotube for the four molecules, average number of sorbed molecules per unit length of the tube, and mean residence time and effective axial diffusivities, all as a function of tube diameter and tube length. In all cases, a strong dependence of the results on carbon nanotube diameter was observed, especially in the way the different molecules are packed and organized inside the CNT. For water for which predictions of properties such as local structure and packing were computed with both methods (MD and GCMC), the two sets of results were found to be fully self-consistent for all types of SWCNTs considered. Water diffusivity inside the CNT (although, strongly dependent on the CNT diameter) was computed with two different methods, both of which gave identical results. For large enough CNT diameters (larger than about 13 Å), this was found to be higher than the corresponding experimental value in the bulk by about 55%. Surprisingly enough, for the rest of the (phenolic) molecules simulated in this Thesis, the simulations revealed no signs of mobility inside nanotubes with a diameter smaller than the (20, 20) tube. This has been attributed to strong phenyl-phenyl attractive interactions, also to favorable interactions of these molecules with the CNT walls, which cause them to form highly ordered, very stable structures inside the nanotube, especially under strong confinement. The interaction, in particular, of the methyl group (present in tyrosol, vanillic acid, and p-coumaric acid) with the CNT walls seems to play a key role in all these compounds causing them to remain practically immobile inside nanotubes characterized by diameters smaller than about 26 Å. It was only for larger-diameter CNTs that tyrosol, vanillic acid, and p-coumaric acid were observed to demonstrate appreciable mobility. / Η παρούσα Διδακτορική Διατριβή εστιάζει στη μελέτη της σχέσης μεταξύ δομής και μακροσκοπικών φυσικών ιδιοτήτων υλικών από πολυακρυλικά μέσω μίας λεπτομερούς προσομοίωσης στον υπολογιστή με τη μέθοδο της Μοριακής Δυναμικής (ΜΔ), σε συνδυασμό με ένα πολύ επακριβές πεδίο δυνάμεων (το Dreiding) σε ατομιστική λεπτομέρεια. Οι κύριες ερευνητικές έννοιες καθώς και οι στόχοι συζητιούνται και αναλύονται σε τρία ξεχωριστά μέρη.
