Grain and artificial stimulation of the rumen change the abundance and diversity of methanogens and their association with ciliates

[Truncated abstract] In Australia, there is pressure to reduce the amount of methane produced by ruminant livestock because they are the single largest source of methane emitted from anthropogenic sources, accounting for 70.7% of agricultural methane emissions. In addition, methane production represents a loss of gross energy intake to the animal. The organisms that are responsible for methane production in the animal gut are a distinct group of Archaea called methanogens. Methanogens occupy three different niches within the rumen. Some live freely in the rumen digesta (planktonic), others are attached to the outer surface of the rumen ciliates (ectosymbiotic), and some reside within the ciliates (endosymbiotic). The types and number of methanogens, as well as rumen ciliates and their symbiotic interactions, influence the amount of methane produced from the rumen. These factors in turn are affected by many factors, including diet and ruminal retention time. In this thesis, I tested the general hypothesis that increasing the amount of grain in the diet and reducing the retention time would affect the abundance and diversity of methanogens in their different niches, including their association with ruminal ciliates. Twenty-four fistulated sheep were used in a complete factorial design with the sheep randomly divided into four groups. ... The change in DGGE banding patterns and Shannon indices when sheep were fed grain indicated that the types of methanogens changed when sheep were fed low and high grain diets, but their diversity did not. In contrast, the diversity of rumen ciliates decreased when sheep were fed a high grain diet. A total of 18 bands from the DGGE analysis of the ciliates were sequenced. All except one, which was 98% similar to Cycloposthium sp. not found previously in the rumen, matched the sequences for previously identified rumen ciliates. Some of the rumen ciliates identified were not present in sheep fed the high grain diet. On a high grain diet, methanogens associate endosymbiotically with rumen ciliates to get better access to hydrogen. It appears that the association between methanogens and rumen ciliates is dictated by the availability of hydrogen in the rumen and not the generic composition of the ciliate population. Furthermore, endosymbiotic methanogens appear to produce less methane than methanogens in other niches. The pot scrubbers did not change ruminal retention time but they did reduce the acetate/propionate measurements observed in sheep on the high grain treatment. The reason why pot scrubbers had this effect remains unknown, but it is interesting to consider that some physical interaction has occurred between the pot scrubbers, the grain and the sheep that has improved the fermentation parameters in sheep fed a high grain diet. The results from this study have advanced our understanding of the interaction between methanogens and ruminal ciliates, and methanogenesis in the rumen in response to dietary changes and mechanical challenges. Extending this work to look more specifically at the species of methanogens that are most closely linked to high methane production and how they interact with the ruminal ciliates will be critical for manipulating enteric greenhouse gas emissions.

Another concern is the correct disposal of agroindustrial residues. The wet brewery grain is considered as polluting as animal wastes. Thus, such wastes must have correct destination, so their use to feed animal is an option. The wet brewery grain has been recorded as an important substitute for fodder and/or grain in ruminants diet. Thus, this study aimed at evaluating the potential for methane production in cattle fed diets containing different levels of wet brewery grain inclusion, to replace bulky diet, with 0, 20, 25 and 30% rates in dry matter percentage. Four castrated steers, with rumen cannula were studied and distributed in a Latin square design (4x4) in four experimental periods of 21 days each (14 days of adjustment and 7 days for data collection). During the last week of each period, the animals manure and ruminal fluid were collected, whose anaerobic digestion essays were analyzed, during the period of 120 days. So, the following parameters were evaluated: removal of total and volatile solids, removal of soluble chemical oxygen demand, cumulative and daily production of methane, volatile fatty acids production (lactic, acetic, propionic and butyric), acidity/alkalinity ratio and pH of reactors. In order to record ruminal fluid, 96-hour tests were performed in the laboratory to observe the influence of wet brewery grain inclusion on methane and volatile fatty acids production inside the rumen. The statistical evaluation of the process was obtained with curves of cumulative production of methane that were adjusted according to Gompertz modified model. Tukey test was used to compare averages with 5% significance. According to these data, it is concluded that wet brewery grain inclusion influenced on methane production for these animals. There was effect of treatment and period for cumulative production of methane (L). The treatment with the largest inclusion of wet brewery residue showed the highest potential of methane production in wastes. The wet brewery residue inclusion did not affect the production of volatile fatty acids inside rumen. The cumulative methane production (mL) of ruminal fluid was influenced by the inclusion of wet brewery residue on animals diet. Nevertheless, the lowest production occurred with an increasing supply of wet brewery residue. The Gompertz modified model provided a good adjustment to the methane production data. Consequently, wet brewery residue can be considered an alternative diet to replace corn silage up to 30% (DM), since it reduces methane production inside the rumen (environmental benefit) and increases methane production from residues (energy and economic benefit). / A pecuária, sobretudo a bovina, tem sido considerada um dos principais setores responsáveis pelo aquecimento global, devido à emissão de metano ruminal e, em menor proporção, à emissão de metano pelos dejetos. Outra preocupação é a correta destinação dos resíduos agroindustriais. O resíduo úmido de cervejaria é considerado tão poluidor quanto os dejetos animais. Desta forma, é importante que este subproduto tenha destino correto e uma opção é o uso na alimentação animal. O resíduo úmido de cervejaria tem se mostrado um importante substituto para forragens e/ou grãos na dieta de ruminantes. Portanto, o objetivo deste trabalho foi avaliar o potencial de produção de metano de bovinos alimentados com dietas que continham diferentes níveis de inclusão de resíduo úmido de cervejaria (RUC), em substituição ao volumoso da dieta, cujas doses foram: 0, 20, 25 e 30% em porcentagem de massa seca. Foram utilizados quatro bovinos machos, castrados, portadores de cânula ruminal, distribuídos em delineamento quadrado latino (4x4), em quatro períodos experimentais de 21 dias cada (14 dias de adaptação e 7 dias de coleta de dados). Na última semana de cada período, coletaram-se dejetos e líquido ruminal dos animais. Com os dejetos coletados, realizaram-se ensaios de biodigestão anaeróbia, no período de 120 dias, nos quais foram avaliados a remoção de sólidos totais e voláteis, a remoção da demanda química de oxigênio solúvel, a produção diária e acumulada de gás metano, a produção de ácidos graxos voláteis (lático, acético, propiônico e butírico), a relação acidez/alcalinidade e o pH dos reatores. Em relação ao líquido ruminal, foram realizados testes de 96 horas, em laboratório, para verificar a influência da inclusão de RUC na produção de gás metano e ácidos graxos voláteis no rúmen. Para a avaliação estatística do processo, utilizaram-se curvas de produção acumulada de metano, ajustadas segundo modelo de Gompertz modificado. O teste de Tukey foi adotado para comparação das médias, com nível de significância de 5%. Com os dados obtidos, conclui-se que a inclusão de resíduo úmido de cervejaria teve influência na produção de metano nesses animais; houve efeito de tratamento e período para a produção acumulada de metano (L). O tratamento com maior inclusão de resíduo úmido de cervejaria apresentou maior potencial de produção de metano nos dejetos; a inclusão de RUC não alterou o perfil de produção de ácidos graxos voláteis no rúmen; a produção acumulada de metano (mL) do líquido ruminal foi influenciada pela inclusão de resíduo úmido de cervejaria à dieta. Todavia, a menor produção ocorreu com o aumento do fornecimento de resíduo úmido de cervejaria. O modelo de Gompertz modificado apresentou bom ajuste aos dados de produção de gás metano. Logo, o resíduo úmido de cervejaria pode ser considerado alimento alternativo para substituir a silagem de milho, em até 30% (MS), uma vez que diminui a produção de metano no rúmen (benefício ambiental) e aumenta a produção de metano nos dejetos (benefício energético e econômico).

Mise en oeuvre du procédé d'électrocoagulation pour le traitement des eaux usées et pour la séparation et la purification de milieux biologiques / The application of electrocoagulation process for wastewater treatment and for the separation and purification of biological media

L'électrocoagulation (EC) est une méthode électrochimique non spécifique couramment utilisée pour le traitement de l'eau et des eaux usées. Dans ce travail, l’EC est d'abord étudiée comme une technique classique de traitement des eaux usées dédiée à l'élimination des protéines de lactosérum de l'eau pour laquelle les mécanismes d'élimination sont expliqués et un modèle est développé. Ensuite, l'utilisation de l’EC est étendue à la séparation et à la purification d’acides gras volatils issus de la fermentation acidogénique. Dans cette deuxième étude, les effets des paramètres opératoires sur l'efficacité et le coût de l’EC sont discutés. En outre, l’EC est utilisée pour la récolte de deux espèces de microalgues de leur milieu de culture. En ce qui concerne la récolte de Chlamydomonas reinhardtii, la méthodologie de la surface de réponse est utilisée et deux modèles permettant de prédire respectivement l'efficacité de la récupération et le coût opératoire sont développés. La récolte d’une autre espèce de microalgues, Chlorella vulgaris, est étudiée en utilisant l’EC respectivement en mode discontinu et continu. En mode discontinu, les effets des principaux paramètres de fonctionnement sur l'efficacité du processus sont expliqués et les mécanismes de récupération sont discutés. Dans l'étude en mode continu, la méthodologie de la surface de réponse est utilisée et un modèle permettant de prédire l’efficacité de récupération des microalgues est développé. Enfin, la comparaison des performances de l'EC en mode continu avec et sans échange de polarité aux performances de l'EC en mode discontinu est effectuée. / Electrocoagulation (EC) is a non-specific electrochemical method usually used for water and wastewater treatment. In this work, EC is firstly investigated as a conventional wastewater treatment technique for the removal of whey proteins from water, where the mechanisms of removal are explained and a model on whey proteins elimination is developed. Then, EC use is extended for the separation and purification of volatile fatty acids issued from acidogenic fermentation. In this second study, the effects of operating parameters on EC efficiency and cost are discussed. Moreover, EC is used for the harvesting of two microalgae species from their culture medium. In the study that concerns recovering Chlamydomonas reinhardtii, response surface methodology (RSM) is employed and two models for predicting recovery efficiency and operating cost are developed. The harvesting of the other microalgae species Chlorella vulgaris is studied using EC in the batch and continuous modes. In the batch mode, the effects of the main operating parameters on the process effectiveness are explained along with discussing the mechanisms of recovery. In the continuous mode study, response surface methodology (RSM) is applied and a model for predicting microalgae recovery is developed. Finally, comparison of EC performance in continuous mode with and without polarity exchange (PE) to EC performance in batch mode is carried out.

Life cycle assessment of DHA produced by microalgae using food waste : Assessing global warming, fossil energy use and effects on biodiversity

Biodiversity is a key component for life on Earth since it contributes to clean water, fresh air and food security. Today, fatty fish farmed in aquaculture is the main Omega 3 source consumed by humans, including the essential fatty acid docosahexaenoic acid (DHA). DHA origin from plants and accumulate in fish via the marine food web. Therefore, DHA in the form of fish oil is often added to fish feed used in aquaculture. This process is dependent on fossil energy and marine raw materials, which infer increased global warming, damage to ecosystem and ultimately loss of biodiversity. In order to reduce the environmental impact, the essential fatty acid could instead be derived from the marine primary producer of DHA: microalgae. In this thesis, a life cycle approach was used to assess global warming, use of fossil fuels and Ecosystem damage when DHA is produced by the microalgae Crypthecodinium Cohnii. The environmental impact was modelled using SimaPro 9 and assessed with CML-IA and ReCiPe Endpoint. In this model, volatile fatty acids derived from dark fermentation of food waste was used as feedstock to the algae. The studied systems consisted of two parallel scenarios, one conventional food waste-to-biogas with DHA from fish oil and one conceptual food waste-to-DHA with DHA from algae oil. The aim was to evaluate the future potential of DHA produced from algae, by assessing and comparing environmental impact to DHA produced from Peruvian anchovy. For every ton DHA produced by microalgae the assessed impact was -1.9E+02 tonCO2e, -1.9 TJ and 9.7E-04 species.yr. DHA produced by microalgae using VFA from food waste was shown to mitigate global warming and reduce use of fossil fuels. The most important conclusion show that DHA from algae infer 37% lower biodiversity loss in comparison to DHA from Peruvian anchovy. Thus, DHA from microalgae could reduce dependency on marine raw material and decrease biodiversity loss. / Biodiversitet är en nyckelkomponent för liv på jorden eftersom det bidrar till rent vatten, frisk luft och säker livsmedelsproduktion. Idag är fet fisk odlad i vattenbruk den viktigaste källan till Omega 3 som konsumeras av människor, inklusive den essentiella fettsyran dokosahexaensyra (DHA). Då DHA härstammar från växter och ackumuleras i fisk via den marina näringskedjan, tillsätts DHA ofta till fiskfoder i form av fiskolja. Denna process är beroende av fossil energi och marina råmaterial, som leder till ökad global uppvärmning, skadar naturliga ekosystem och orsakar förlust av biologisk mångfald. För att minska miljöpåverkan skulle den essentiella fettsyran istället kunna produceras från den marina primärproducenten av DHA: mikroalger. I detta examensarbete användes livscykelanalys för att utvärdera miljöpåverkan med avseende på global uppvärmning, användning av fossila bränslen och påverkan på biodiversitet, då DHA produceras av mikroalgen Crypthecodinium Cohnii. Flyktiga fettsyror, VFA, som bildas vid mörk fermentering av matavfall användes som råmaterial till algerna. De studerade systemen bestod av två parallella scenarier, en konventionell matavfall-till-biogas med DHA från fiskolja och en konceptuell matavfall-till-DHA med DHA från algolja. Systemet modellerades i SimaPro 9 och miljöpåverkan beräknades med CML-IA och ReCiPe Endpoint. Syftet var att utvärdera DHA som produceras från alger genom att beräkna miljöpåverkan och jämföra med DHA producerad från peruansk ansjovis. För varje ton DHA producerat av mikroalger var påverkan -1.9E+02 tonCO2e, -1.9 TJ och 9.7E-04 arter per år. DHA producerad av mikroalger där VFA från matavfall använts som näring, visade sig minska den globala uppvärmningen, reducera användningen av fossila bränslen och innebar 37% lägre förlust av biologisk mångfald jämfört med DHA producerad från peruansk ansjovis. Denna studie visade därmed att DHA från mikroalger kunde minska beroendet av marina råmaterial och minska förlusten av biologisk mångfald.

High-loaded thermophilic anaerobic digestion of mixed sewage sludge : A pilot study / Högbelastad termofil rötning av blandat avloppsslam : En pilotstudie

Municipal wastewater treatment plants (WWTP) are important infrastructural components in a society and also important for sustainability. In a WWTP the most common treatment configuration is mechanical, biological, and chemical treatment of the wastewater. The treatments reduce nutrients and organic matter before the water is transferred to the recipient. During wastewater treatment, sludge is produced and then stabilized in an anaerobic digestion (AD) process.  The Käppala Association operates the third largest WWTP in Sweden. In the future the Käppala plant is expecting an increased number of connected households and also stricter sludge hygienization regulations. This implies that current strategies for the WWTP have to be developed. One idea has been to use a thermophilic AD process instead of mesophilic AD.Thermophilic AD has previously been shown to have higher capacities, lower hydraulic retention times (HRT) and increased pathogenic destruction compared to their mesophilic counterpart. A common negative aspect for a thermophilic process has been process instability.  In this study a 5 m3 pilot plant rented from Research Institutes of Sweden (RISE) was used to evaluate maximal organic loading rate (OLR) for a stable thermophilic AD process using mixed sludge as substrate. Four HRT were chosen, and each HRT was maintained for 3 retention times. Laboratory analyzes of the raw and digested sludge and on-line monitoring were performed regularly to collect information about process stability and efficiency. The pilot plant was controlled through a surveillance system where operating parameters were introduced. The main objectives of this study were to investigate how an increase of OLR affected pH, alkalinity, and volatile fatty acid (VFA) content and also how the alkalinity and VFA affected the process stability. Gas composition, gas production, degree of digestion (DD) and foaming were also investigated throughout this study. Results showed that VFA initially increased, and alkalinity decreased when every OLR increase occurred. The VFA and alkalinity returned to lower values after the process was given time to recover during constant OLR. The OLR increase caused slight variations in process pH but not enough for process failure. Increased OLR did not seem to have a large impact on the DD since it seemed to remain between 40-50% throughout the entire study. Composition monitoring showed an increased hydrogen sulfide content in the gas as a consequence of increased OLR. An OLR increase also lead to a volumetric gas production (VGP) and volumetric methane production (VMP) increase while efficiency of the AD process was seen to decrease when evaluating specific gas production (SGP) and specific methane production (SMP). The conclusion of the study was that a stable thermophilic AD process using mixed sludge as substrate could be operated with an OLR of 6.55 ± 0.06 kg VS m-3d-1and a HRT of 7 days. Maximal OLR for a stable thermophilic AD process was never achieved due to the process remaining relatively stable the entire experimental period. The process recovered from signs of instability during all HRT-transition times indicating it can maintain all investigated OLR. / Kommunala avloppsreningsverk är en viktig infrastruktur i ett samhälle och även viktigt för hållbarheten. I ett avloppsreningsverk renas spillvatten genom mekaniska, biologiska och kemiska behandlingar processteg. Behandlingarna minskar näringsämnen och organiskt material innan vattnet rinner ut i recipienten. Vid rening av avloppsvatten erhålls slam. Slammet kan användas som substrat för en anaerobisk nedbrytningsprocess. Käppalaförbundet driver det tredje största reningsverket i Sverige. I framtiden förväntas antal anslutna hushåll till reningsverket att öka samt strängare slamhanteringskrav appliceras. Detta innebär att nuvarande slamstrategi måste utvecklas. En idé är att tillämpa en termofilrötnings process. En termofil rötningsprocess har visat sig ha högre kapacitet, lägre uppehållstider och ökad patogen reduktion i jämförelse med en mesofil rötning. En nackdel med termofil rötning är den försämrade processtabiliteten.  I denna studie har en 5 m3 pilotanläggning inhyrd från RISE använts för att utvärdera den maximala organiska belastningen för en stabil termofil rötningsprocess med blandslam som substrat. Fyra uppehållstider valdes och varje uppehållstid hölls tre gånger. Regelbundna analyser av rå och rötslam utfördes för att erhålla information om process stabilitet och effektivitet. Pilotanläggningen styrdes genom ett människa-maskingränssnitt där driftparametrar matades in. Huvudmålen för studien var att se hur en ökning av organisk belastning påverkade pH, alkaliniteten och halt av flyktiga fettsyror samt att undersöka alkalinitetens och halten av flyktiga fettsyrors påverkan på processtabiliteten. Påverkan av en ökad organisk belastning på gas sammansättning, gasproduktion, rötningsgrad har även undersökts. Resultaten visade att flyktiga fettsyror initialt ökade och alkaliniteten minskade vid en belastnings ökning. Flyktiga syror och alkalinitet återgick till mer stabila värden efter processen fick återhämta sig vid en konstant belastning. Belastningsökningen orsakade små variationer i pH värdet men inte tillräckligt för att orsaka processkollaps. En ökad belastning verkade inte ha någon större påverkan på rötningsgraden då den låg runt 40–50% hela rötningsperioden. Skumutvecklingen visade sig inte heller vara ett problem. Resultaten visade även att väte sulfid ökade i gasen när en belastningsökning gjordes. Den volymetriska gas och metan produktionen ökade vid en ökad OLR medan specifika gas och metan produktionen minskade. Slutsatsen var att en stabil termofil rötningsprocess av blandslam kan drivas med en organisk belastning på 6.55 ± 0.06 kg VS m-3d-1 och en uppehållstid på 7 dagar. Maximal belastning för stabil termofil rötningsprocess uppnåddes dock ej då processen förblev relativt stabil under alla undersökta uppehållstider. Processen återhämtade sig även efter tecken på instabilitet vilket indikerar att AD processen kan behålla stabilitet vid de undersökta belastningar.

Production of filamentous fungal biomass on waste-derived volatile fatty acids for ruminant feed supplementation and it's in vitro digestion analysis

Single cell proteins such as that of edible filamentous fungal biomass are considered as a promising sustainable source of animal feed supplementation. Filamentous fungi can be cultivated on different organic substrates including volatile fatty acids (VFAs) such as acetic, propionic, and butyric acids. These VFAs can be generated through the famous waste valorisation approach of anaerobic digestion (AD) as intermediate metabolites. This project investigates a sustainable approach for the production of animal feed supplementation through cultivation of fungal biomass on waste derived VFAs along with the in vitro analysis of fungal biomass digestibility as ruminant feed. In this regard, optimum conditions for the production of Aspergillus oryzae biomass on different VFAs effluents derived from anaerobic digestion process of food waste plus chicken manure (FWCKM) and potato protein liquor (PPL) at different pH, nitrogen sources, and feed mixture was studied. Accordingly, analyses showed that PPL has the highest biomass yield with 0.4 (g biomass/g consumed VFAs) based on the volatile solids (VS) by adjusting pH to 6.2. Furthermore, the digestibility of the produced fungal biomass is analysed by using three different in vitro digestion methods including Tilley and Terry (TT) method, Gas Production Method (GPM), and Nylon Bag Method (NBM) and the results are compared with the conventional feed (silage and rapeseed meal). Results obtained from different digestibility methods illustrate that different A. oryzae fungal biomass had approximately 10-15 % higher dry matter digestibility fraction compared to silage and rapeseed meal (reference feeds). Hence, these results revealed that A. oryzae fungal biomass can grow on VFAs effluents and produce protein-rich fungal biomass while this biomass has better digestibility compared to conventional feeds and confirmed the initial hypothesis of the study.

Effect of substrate types on fermentation of mixed cultures to produce volatile fatty acids / Effekt av substrattyper för fermentering av blandade kulturer för att producera flyktiga fettsyror

Innovativa teknologier, produkter och processer krävs på grund av marknadens ökande behov av bioprodukter med låga koldioxidavtryck. Idén att producera flyktiga fettsyror via biologiska processer, främst genom anaerob fermentering med blandkulturer, har återupplivats på grund av den omfattande exploateringen av oljeresurser. Målet för denna studie var att utvärdera kvaliteten och livskraften för produktion av flyktiga fettsyror från enkla substrat. Flyktiga fettsyror-generering undersöktes i ett satsvis syrafermenteringssystem för tre enkla substrat (olja, stärkelse, och gelatin). Vi visade att ett initialt surt pH var fördelaktigt för acidogenes och den totala genereringen av flyktiga fettsyror för alla tre substraten. Den högsta produktionseffektiviteten av flyktiga fettsyror uppnåddes och flyktiga fettsyror genomgick evolutionär utveckling med blandad kulturfermentering som kördes vid ett initialt surt pH på 5,0. 7933,08 mg COD/L smörsyra producerades från stärkelse vid pH 3,87 på dag 15, vilket var den högsta nivån. Smörsyra och ättiksyra var de dominerande produkterna i det initiala sura pH-värdet 5. Alla tre substraten i batch-reaktorer visade högt produktionsutbyte av ättiksyra och smörsyra. Vid lågt initialt pH var propionsyra den vanligaste syratypen i olja. / Innovative technology, products, and processes are required because of the market's rising need for bioproducts with low carbon footprints. The concept of producing volatile fatty acids (VFAs) via biological processes, primarily through anaerobic mixed culture fermentation, has been revived because of the extensive exploitation of oil resources. The goal of this study was to evaluate the quantity and efficiency of VFA production from simple substrates. The generation of VFA was examined using a batch type acid fermentation system for three simple substrates (oil, starch, and gelatin). We showed that an initial acidic pH was advantageous for acidogenesis and the total generation of VFAs for all three substrates. The VFA production efficiency was achieved and underwent evolutionary development with mixed culture fermentation (MCF)  tests running at an initial acidic pH of 5.0. 7933.08 mgCOD/L butyric acid were produced from starch at pH 3.87 at day 15, which was the highest level. Butyric and acetic acids were the predominant products in the initial acidic pH 5. All the three substrates in reactors showed high VFA production yield of acetic acid and butyric acid. At low initial pH, propionic acid was the most prevalent acid type in oil.

In conventional wastewater treatment plants, methanol, ethanol, and acetate are used as carbon source for the denitrification process in the biological nutrient removal. However, growing concern regarding economical costs and carbon footprints from the fossil-based production of these chemicals have forced the companies to look for other alternatives. VFAs have shown a great potential in replacing the conventionally used carbon sources. If implemented this will result in lower chemical cost and a drastic decrease in carbon footprint as well as striving WWTPs towards sustainable development. In this work denitrification has been analysed using different variations of VFAs such as fermented potato protein liquor, food waste and chicken manure VFA. This was done using a basic laboratory setup of a denitrification reactor which used basic stirring agitation and nitrogen purging to ensure anoxic conditions. Nutrients and excess sCOD were added to ensure the highest denitrification rates. The denitrifying biomass was collected at Gryaab AB in the form of k1-carriers making this process a MBBR. The most influential characteristic of the VFAs is the distribution of the acids in the VFA effluent. Butyric acid along with caproic acid showed the best potential for efficient denitrification. The possibility of concentration of VFA effluent showed a high potential when using a nanofiltration system. A C/N ratio of 4.5 conventionally used when methanol is added showed to be the most optimal condition for VFA addition. The combination of VFAs together with conventional used carbon sources showed the best potential in denitrification efficiency proving to be as good or even better than pure synthetic ones. VFAs effluents showed the best potential in removing the intermediate nitrite from the wastewater at high rates. Overall, VFAs shown a great potential for replacing conventionally used carbon sources, demonstrating the potential of substitution, which if implemented will result in lower carbon footprint and a strive towards sustainable development.

Pilotstudie - rötning med slamrecirkulering på Käppalaverket / Pilot study - anaerobic digestion of sludge from Käppalaverket

Kommunala avloppsreningsverk renar avloppsvatten och avlägsnar näringsämnen innan vattnet släpps ut i den omgivande miljön. Under reningen avskiljs partiklar som bildar råslam. Genom att bryta ner råslammet anaerobt (syrefritt) via rötning kan användbara produkter, såsom biogas till bussar, erhållas. Rötning innebär att organiskt material bryts ner under kontrollerade former. Sveriges tredje största kommunala avloppsreningsverk är Käppalaverket i Stockholm. Framtidsprognosen för verket visar att belastningen kommer öka, samtidigt förväntas kraven på slamhantering bli strängare.  Syftet med denna studie var att utreda en metod för att öka Käppalaverkets slambehandlingskapacitet. Idag rötas slammet konventionellt med mesofil rötning. Ett förslag för att öka kapaciteten är att förtjocka och därefter recirkulera slammet in i samma rötningskammare. Möjliga fördelar med detta kan vara ökad biogasproduktion samt möjlighet till högre belastning i rötkamrarna.  Till experimentet nyttjades en pilotanläggning bestående av två rötkammrar på 7 dm3. Rötkamrarna användes för att studera konventionell mesofil rötning (i studien benämnd BR01), jämfört med mesofil rötning med recirkulering (i studien benämnd BR02). Råslammet och rötslammet analyserades löpande (på dag- och veckobasis) med avseende på parametrarna: biogasproduktion och gassammansättning, torrsubstanshalt, halt av flyktiga syror, pH, alkalinitet, organisk belastning, andel mineraliserat kväve, utrötningsgrad samt reduktion av makromolekyler. Uppehållstiden (HRT) för slammet i rötkamrarna var inledningsvis 14 dagar. HRT sänktes stegvis  under studien (från 14 till 11 dagar och därefter till 9 dagar) genom att inflödet av råslam ökades. Därmed pressades BR01 och BR02 i syfte att utröna vid vilken belastning rötningsprocessen förändrades från stabil till instabil. Resultaten visade att under 11-dagars perioden var processen i BR01 stabilare än i BR02. Förhöjda halter av flyktiga syror uppmättes i BR02 under denna period. Under 9-dagars perioden med högre belastning observerades att processen i BR01 var instabil. Processen i BR02 var stabilare under samma period och ansågs därför prestera bättre under högre belastning. Statistiska t-tester visade att flertalet reaktorparametrar (i en och samma reaktor) genomgick säkerställda förändringar från experimentets start till slut. / Municipal wastewater treatment plants (WWTPs) purifies wastewater from nutrients before releasing it to the recipient. Particles that are separated from the water creates a sewage sludge. Anaerobic (without oxygen) digestion is mainly used as a stabilization method for the sludge. Digestion of the sludge produces energy-rich biogas and a nutrient-rich digestate which can be used as a fertilizer. The third largest municipal WWTP in Sweden is Käppala WWTP, situated in Stockholm. In the future, the amount of incoming wastewater is expected to increase. Simultaneously, regulations regarding sludge management are expected to be stricter. The main purpose of this study was to investigate a method to increase the sludge load to the digesters at Käppala WWTP. Today the sludge is digested conventionally with mesophilic anaerobic digestion. One suggestion is to recirculate thickened sludge into the same digestion reactor. This method has been shown to give an increased biogas production as well as improved digester performance.   During the experiment a pilot-digester consisting of two anaerobic reactors (each with a volume of 7 dm3) were used. These reactors were used to study conventional mesophilic digestion (BR01 in this study) and mesophilic digestion with sludge-recirculation (BR02 in this study). Several parameters of the sludge (raw and digested) were analysed on daily and weekly basis: biogas production and gas composition, dry matter content, volatile fatty acids, pH, alkalinity, organic loading, mineralised nitrogen, degree of digestion and reduction of macromolecules. The hydraulic retention time (HRT) of the sludge was initially 14 days. HRT was lowered stepwise (from 14 to 11 and finally down to 9 days) by increasing the inflow of raw sludge. This way the two reactors were pushed to see at what point the digestion process became unstable. The results showed that during the 11-days HRT period BR01 was more stable compared to BR02. Heightened VFA-content was measured in BR02 during this period. During the 9-days HRT period the process in BR01 was deemed unstable. The process in BR02 was more stable during the same period and was considered to have a better performance during high sludge inflow. Statistical t-tests showed that several digestion-parameters (compared for the same reactor) underwent statistically significant changes from the start to the end of the experiment.
