Microbial Activity ̶ Indicators & Drivers

Biota des Kohlenstoffkreislaufs spielen bei der Speicherung und Verteilung des Kohlen-stoffs im Bodennahrungsnetz eine herausragende Rolle. Wobei der Abbau organischer Bodensubstanz stark von deren Verfügbarkeit für Mikroorganismen und Enzyme abhängt. Rhizosphäre und Detritusphäre verfügen über außerordentlich große Mengen an organischem Kohlenstoff. Dies macht diese Biosphären zu Hot-Spots mikrobieller Aktivität. Während der letzten Jahrzehnte stieg das Interesse an der Forschung zu mikrobieller Aktivität im Boden. Dennoch herrscht noch immer kein umfassendes Verständinis von Indikatoren und Triebkräften mikrobieller Aktivität in der Rhizosphäre und Detritusphäre. Dies ist nicht zuletzt der Tatsache geschuldet, dass es mehr als einer einzigen Methode zur Bestimmung dieser Faktoren bedarf. Aus diesem Grund setzt sich diese Arbeit zum Ziel, die mikrobielle Aktivität, ihre Indikatoren und Treiber zu explorieren. Folglich wurden diverse Indikatoren mikrobieller Aktivität, wie mikrobielle Respiration, mikrobielle Biomasse und Enzymkinetik, an Proben aus einem Feldexperiment mit diffe-rierendem Substrat-Eintrag (Mais-Rhizodeposite vs. Mais-Streu) ermittelt. Tiefengradienten der Indikatoren mikrobieller Aktivität dienten der Beurteilung der Auswirkungen sinkender Substratqualität und -quantität in Rhizo- und Detritusphäre mit zunehmender Bodentiefe. Besonderer Fokus lag dabei auf den Indizes unterschiedlicher Enzyme und deren Aktivität (z. B. spezifische Aktivität, katalytische Effizienz und Verhältnisse zwischen Enzymen des Kohlenstoff- und Stickstoff-Kreislaufs) im Tiefenprofil des Bodens. Neben dieser Feldstudie wurde das Verhältinis von RNA zu dsDNA als Indikator mikrobieller Aktivität entlang eines klimatischen Grandienten ermittelt, um den metabolischen Status innerhalb unterschiedlicher Bodentypen zu bestimmen. Die Feldstudien wurden um Experimente in Labor und Gewächshaus ergänzt, in denen mittels Isoto-penmarkierungsverfahren die Effekte von Wurzelhaaren und Protisten auf die mikrobielle Aktivität beleuchtet wurden. Diese Methode ermöglichte ein Nachvollziehen des C- und N-Flusses und trug damit zum Verständnis der Verflechtungen der Organismen im terrestrischen Boden-Nahrungsnetz bei. Die räumliche Verteilung der Enzymaktivität in der Rhizosphäre wurde anhand der Boden-Zymographie in situ untersucht. Durch unterschiedliche Substratverfügbarkeit wurde ein Wandel der funktionellen Eigenschaften der Mikroorganismengemeinschaften und des enzymatischen Systems induziert. Speziell der durchwurzelte Oberbodenhorizont zeigte einen Anstieg der mikro-biellen Aktivität im Vergleich zum Boden mit Streueintrag und der Kontrolle. Rhizodeposite sind eine grundlegende Kohlenstoff- und Energiequelle für Bodenmikroorganismen und stimulieren deren Wachstum und Aktivität. Die Präsenz von Rhizodepositen in Hot-Spots macht diese zu bevorzugten Habitaten für Mikroorganismen. Die Mehrzahl der Indikatoren für mikrobielle Aktivität wurde ausschließlich im Oberboden durch den Substrateintrag beeinflusst. Darunter auch die katalytische Effizienz, die – ungeachtet des Substrateintrags – von Oberboden (< 40 cm) zu Unterboden (> 40 cm) um das 2- bis 20-fache abnahm. Dies ließ auf die Relevanz der mit der Tiefe abnehmenden Menge und Qualität der Substrate im Boden als einflussnehmenden Faktor auf die mikrobielle Aktivität schließen. Das Verhältnis von RNA zu dsDNA spiegelte den metabolischen Status der mikrobiellen Organismengesellschaften in den meisten der beprobten Böden wider. Wohingegen das RNA:dsDNA Verhältnis dieser Indikatoreigenschaft widersprach, lagen erhöhte Tongehalte vor, die nach der Extraktion zu Ungenauigkeiten bei der Bestimmung der RNA-Quantität führten. Protozoen wird beim Vorgang des Kohlenstoffflusses von Bakterien zu Organismen höherer Trophieebenen eine bedeutende Rolle zugesprochen, was ebenfalls ihren Einfluss auf die mikrobielle Aktivität im Boden unterstreicht. Um diesen Effekten, im Speziellen jenen der Acanthoamoebe auf den Kohlenstoff- und Stickstofffluss, sowie die Indikatoren mikrobieller Aktivität in der Rhizo- und Detritusphäre nachzugehen, wurde ein dreifaches Isotopenmarkierungs-Experiment durchgeführt. Es ergab, dass Kohlenstoffflüsse und Enzymaktivitäten sowohl von Substrateintrag als auch Substratqualität in Rhizo- wie Detritusphäre sowie deren faunistischer Besiedlung abhängen. Daraus erschloss sich, dass die Besiedlung mit Acanthamoeben als potenzielle Triebkraft mikrobieller Aktivität, besonders innerhalb der Rhizosphäre, gedeutet werden kann. Um den Einfluss von Wurzelhaaren auf die mikrobielle Aktivität und den Priming Effekt in der Rhizosphäre einzuschätzen, wurde ein Experiment im Gewächshaus mit kontinuierlicher Markierung von Boden mit Pflanzenbewuchs und einer Kontrolle ohne Bewuchs mit 13C-Isotopen durchgeführt. Wurzelhaare zeigten sich darin als Initiatoren eines positiven Rhizosphären-Priming Effektes während der Wachstumsphase, wohingegen der Abbau organischer Bodensubstanz in den Kontrollen gehemmt war. Im Falle der positiven Initialwirkung der Wurzelhaare stiegen zudem die Enzymaktivitäten von Chitinase und ß-Xylosidase an, was auf eine Zersetzung stabiler, organischer Bodensubstanz hinwies. Damit konnte ein deutlicher Effekt von Wurzelhaaren auf die mikrobielle Aktivität im Boden während der Phase des Pflanzenwachtums nachgewiesen werden. Somit vermittelt diese Arbeit ein weiterführendes Verständnis der auf mikrobielle Aktivität im Boden einwirkenden Faktoren und stellt eine Auswahl von Indikatoren zur Charakterisierung dieser Aktivität vor, die sowohl auf der Landschaftsebene als auch in der prozessorientierten Forschung im Wurzelraum Anwendung finden kann.

IDENTIFICATION OF PEPTIDASES IN HIGHLY-PATHOGENIC VERSUS WEAKLY-PATHOGENIC NAEGLERIA FOWLERI AMEBAE

Naegleria fowleri, a free-living ameba, is the causative agent of Primary Amebic Meningoencephalitis. Highly-pathogenic mouse-passaged amebae (Mp) and weakly-pathogenic axenically-grown (Ax) N. fowleri were examined for peptidase activity. Zymography and azocasein peptidase activity assays demonstrated that Mp and Ax N. fowleri exhibited a similar peptidase pattern. Prominent for whole cell lysates, membranes and conditioned medium from Mp and Ax amebae were the presence of an activity band of approximately 58kDa and 100 kDa bands susceptible to the action of cysteine and metallopeptidase inhibitors, respectively. Further roles of the peptidases during the invasion process were examined by in vitro invasion assays in the presence of inhibitors and Cysteine and metallopeptidase inhibitors were found to greatly reduce invasion through the ECM. This study establishes a functional linkage of the expressed peptidases to the invasion process, and these peptidases may serve as a candidate target for therapeutic management of N. fowleri infection.

O diagnóstico da tromboembolia pulmonar aguda (EPA) e a avaliação da gravidade desta condição é desafiador. Enquanto as concentrações de troponina I cardíaca (TI) já estão bem estabelecidas quanto ao risco de estratificação, não há estudos prévios que tenham examinado se há alguma relação linear entre as concentrações de TI cardíaca e a gravidade da EPA. Além disso, as metaloproteinases (MMPs) da matriz extracelular estão envolvidas na fisiopatologia da EPA. Entretanto, é desconhecido se o aumento da atividade gelatinolítica das MMPs após a EPA reflete a gravidade desta condição. Nós examinamos se as concentrações circulantes destes biomarcadores aumentam em proporção à gravidade da EPA experimental induzida em cães anestesiados. A EPA foi induzida com coágulos de sangue autólogo (salina, 1, 3 ou 5 mL/Kg) injetados no átrio direito. As avaliações hemodinâmicas foram realizadas no momento basal e 120 minutos após a EPA. Da mesma forma, foram realizadas as quantificações de troponina I no soro e a zimografia das MMPs 2 e 9 no plasma. Nossos resultados sugerem não haver aumento significativo da atividade gelatinolítica da pró-MMP-2 no plasma após a EPA, enquanto que a atividade da pró-MMP-9 aumenta em 80% apenas no grupo que recebeu 5 mL/Kg de coágulos. A TI cardíaca no soro e a atividade da pró-MMP-9 no plasma tiveram uma correlação positiva com o índice de resistência vascular pulmonar (p=0,007 e rs=0,833 para a TI, e p=0,034 e rs=0,684 para a pró-MMP-9) e com a pressão média na artéria pulmonar (p=0,005 e rs=0,610 para a TI, e p=0,022 e rs=0,720 para a pró-MMP-9). Concluímos que a TI cardíaca e a pró-MMP-9 circulantes aumentam em proporção à gravidade da EPA, embora o aumento da pró-MMP-9 não seja muito evidente em graus menos severos da EPA. Estes achados podem ser relevantes para a clínica da EPA. / Making the diagnosis of acute pulmonary thromboembolism (APT) and assessing its severity is very challenging. While cardiac troponin I (CTI) levels are promising in risk stratification, no previous study has examined whether there is a linear relation between CTI levels and the severity of APT. Moreover, matrix metalloproteinases (MMPs) are involved in the pathophysiology of APT. However, it is unknown whether the increases in MMP levels after APT reflect the severity of this condition. We examined whether the circulating levels of these biomarkers increase in proportion to the severity of experimental APT induced in anesthetized dogs. APT was induced with autologous blood clots (saline, 1, 3, or 5 mL/kg) injected into the right atrium. Hemodynamic evaluations were carried out for 120 min. Gelatin zymography of MMP-2 and MMP-9 from plasma samples were performed and serum CTI levels were determined at baseline and 120 min after APT. Our results sugest that while no significant increases in pro-MMP-2 levels were found after APT, pro-MMP-9 levels increased by 80% only after 5 mL/kg of clot embolization. Serum CTI and plasma pro-MMP-9 levels correlated positively with pulmonary vascular resistance (p=0.007 and rs=0.833 for troponin I, and p=0.034 and rs=0.684 for pro-MMP-9) and with pulmonary artery pressure (p=0.005 and rs=0.610 for troponin I, and p=0.022 and rs=0.720 for pro-MMP-9). We conclude that circulating CTI and pro-MMP-9 increase in proportion to the severity of APT, although the increases in plasma pro-MMP-9 are less clear with less severe APT. These findings may be relevant for clinical APT.

Efeito do uso de primers bioativos na interface dentina-adesivo / Effect of bioactive primers on dentin--adhesive interface

A longevidade das restaurações com compósitos está diretamente relacionada ao sucesso do tratamento estético. A vida útil das restaurações pode ser influenciada por vários fatores como o material empregado, tempo de uso clínico e a degradação da matriz dentinária por metaloproteinases da matriz (MMPs), que podem afetar a integridade da interface adesiva, fazendo com que a união dentina-­adesivo seja comprometida. Assim, buscando melhores resultados na durabilidade das restaurações adesivas, o objetivo deste estudo foi avaliar o efeito de substâncias inibidoras de MMPs (soluções de clorexidina -­ CHX a 0,2% e doxiciclina a 3%) e agente de ligação cruzada (extrato de semente de uva -­ grape seed extract - GSE a 15%) na Resistência de União (RU) de restaurações de resina composta submetidas à ciclagem mecânica. Cavidades de Classe I foram preparadas na área central da superfície exposta de 48 molares humanos hígidos, posteriormente separados em 4 grupos (n=12) de acordo com o tratamento de superfície recebido após o condicionamento de ácido: Grupo I -­ Controle (Sistema Adesivo -­ Adper Single Bond Plus -­ 3M ESPE); Grupo II -­ CHX 0,2% + Sistema Adesivo; Grupo III -­ GSE 15% + Sistema Adesivo e Grupo IV -­ Doxiciclina 3% + Sistema Adesivo. Após os procedimentos restauradores (Filtek Supreme Ultra -­ 3M ESPE), as amostras foram armazenadas em água destilada durante 24 horas a 37°C e subdividas em dois subgrupos: A -­ Controle (armazenado em saliva artificial) e B -­ Submetido à ciclagem mecânica (Coil Cycler, Proto-­tech, forca de 50 N, frequência de 1,5 Hz em 1 x 106 ciclos de aplicação). Após ciclagem mecânica, os corpos de prova foram cortados em palitos (0,8 x 0,8 mm; n = 5 por dente) e submetidos ao teste de microtração (Bisco) a velocidade de 1 mm/min. A atividade colagenolítica da MMP-­2 sob a ação dos primers bioativos foi analisada utilizando o kit EnzChek Gelatinase/Colagenase Assay (D-­12054, Molecular Probes, Eugene, OR, EUA). A atividade gelatinolítica das MMPs-­2 e -­9 após incubação com os primers bioativos foi avaliada através de zimografia convencional. Os resultados obtidos das duas análises foram apresentados na forma de porcentagem de inibição. Zimografia in situ foi realizada utilizando gelatina de fluorescência conjugada (Kit EnzChek Gelatinase/Collagenase Assay) como substrato de MMPs e avaliada com aumento de 40x através de microscopia de fluorescência. Posteriormente, os fótons fluorescentes foram analisados em software (ImageJ). Obtidos os resultados, os dados foram submetidos à análise estatística (2way ANOVA, Tukey, p>0,05) e demonstraram que os primers bioativos não exerceram influência significante sobre a RU dentina-­adesivo, independente da ação da ciclagem mecânica. A atividade colagenolítica, segundo zimografia convencional, demonstrou que os primers bioativos foram capazes de inibir as MMPs, sendo a CHX menos eficiente. Na análise de zimografia in situ, os primers bioativos diminuíram a atividade das MMPs localizadas na interface adesiva, com aumento da atividade gelatinolítica do grupo Controle sendo maior quando submetidos à ciclagem mecânica, efeito não encontrados no grupos tratados com primers bioativos. Concluiu-­se que, apesar de não apresentarem efeitos sobre a RU da interface adesiva, os primers bioativos apresentam ação sobre a degradação da camada híbrida. / The durability of composite resin restorations is one of the main points related to the success in aesthetic treatments. The longevity of adhesive restorations can be affected by various factors such as the materials, the clinical time of use and the dentin matrix degradation mediated by matrix metalloproteinases (MMPs). MMPs can affect the adhesive interface, compromising the bonding between dentin and adhesive. Thus, searching for better results on longevity of adhesive restorations, the aim of this study was to evaluate the effect of MMPs inhibitors (chlorhexidine -­ CHX 0.2% and doxycycline 3% ) and crosslinking agent (grape seed extract -­ GSE 15%) on resin composite bond strength (BS) of restorations submitted to load cycling. Class I cavities were performed in the central area of the exposed surface of 48 sound human molars. Then, it was divided into 4 groups (n=12) according to the surface treatment received after the acid etching: Group I -­ Control (Adhesive System -­ Adper Single Bond Plus -­ 3M ESPE); Group II -­ CHX 0.2% + Adhesive System; Group III -­ Doxycycline 3% + Adhesive System e Group IV -­ GSE 15% + Adhesive System. After restorative procedures (Filtek Supreme Ultra -­ 3M ESPE), the samples were stored in distilled water for 24 hours at 37°C and then subdivided into two subgroups (n=6): A -­ Control (stored in artificial saliva) and B -­ Submitted to load cycling (Coil Cycler, Proto-­tech, 50 N, 1.5 Hz, 1 x 106 cycles). After load cycling, the specimens were cut into sticks (0.8 x 0.8 mm; n = 5 per teeth) and submitted to the microtensile test (Bisco) at 1 mm/min. The collagenolytic activity of MMP-­2 under action of bioactive primers was analyzed using EnzChek Gelatinase/Colagenase Assay Kit (D-­12054, Molecular Probes, Eugene, OR, EUA). The gelatinolytic activity of MMPs-­2 and -­9 after incubation with the bioactive primers was evaluated by gelatin zymography. The results of these two analyzes were presented by percentage of inhibition. In situ zymography was performed using fluorescence conjugated gelatin (EnzChek Gelatinase/Collagenase Assay Kit) as MMP substrate and evaluated using 40x magnification under fluorescence microscopy. Then the fluorescent photons were analyzed with an appropriate software (ImageJ). The results were subjected to statistical analysis (2-­way ANOVA, Tukey, p<0.05) and the bioactive primers showed no significant influence on the dentin-­adhesive BS, regardless the load cycling action. According to gelatin zymography, the collagenolytic activity demonstrated that the bioactive primers were able to inhibit MMPs activity and CHX was less efficient. In situ zymography analysis, showed that the bioactive primers decreased the activity of MMPs located in adhesive interface. The data also demonstrated increased gelatinolytic activity on Control group, being higher when submitted to load cycling, and which effect was not found in the groups treated with bioactive primers. It could be concluded that although the bioactive primers did not influenced the BS of adhesive interface, they demonstrated positive results on hybrid layer degradation.

Investigation into the proteolytic activity in chronic wound fluid and development of a remediation strategy

Chronic ulcers are an important and costly medical issue, causing their sufferers a large amount of pain, immobility and decreased quality of life. The common pathology in these chronic wounds is often characterised by excessive proteolytic activity, leading to the degradation of both the extracellular matrix, as well as key factors critical to the ulcer's ability to heal. As matrix metalloproteinases (MMPs), a large family of zinc-dependent endopeptidases, have been shown to have increased activity in chronic wound fluid (CWF), it was hypothesised that this specific proteolytic activity was directly related to an ulcer's chronic nature. Although previous studies have identified elevated proteases in CWF, many have reported contradictory results and therefore the precise levels and species of MMPs in CWF are poorly understood. The studies reported herein demonstrate that MMP activity is significantly elevated in CWF compared with acute wound fluid (AWF). In particular, these studies demonstrate that this proteolytic activity can be specifically attributed to MMPs and not another class of proteases present in wound healing. Furthermore, it is shown that MMP-9 is the predominant protease responsible for matrix degradation by CWF and is an indicator of the clinical status of the wound itself. Moreover, MMP-9 can be inhibited with the bisphosphonate alendronate, in the form of a sodium salt, a functionalised analogue, and also tethered to a synthetic biocompatible hydrogel compromised of aqueous poly (2-hydroxy methacrylate) PHEMA synthesised in the presence of poly(ethylene glycol) (PEG). Together, these results highlight the potential use of a tethered MMP inhibitor as an improved ulcer treatment to inhibit protease activity in the wound fluid, while still allowing MMPs to remain active in the wound bed where they perform vital roles in the activation of growth-promoting agents and immune system regulation.

Salvador, Bahia, Brasil / As doenças cardiovasculares são a maior causa de morbimortalidade nos países emergentes, consistindo em doença arterial central, cerebral e periférica, em especial, a coronária e a carotídea. Dentre os diversos fatores e marcadores de risco que atuam no desenvolvimento e no agravo dessas doenças, encontram-se os relacionados com a aterosclerose. Este estudo avaliou a concentração sérica e a atividade de metaloproteinases de matriz (MMP-1, 2, 8, 9 e 12), a incorporação de colesterol livre na HDL, a atividade da paraoxonase e associação com outros fatores em pacientes com doença carotídea aterosclerótica. Métodos. Este estudo de corte transversal avaliou, em população de 65 indivíduos portadores (32) e não (34) de DCA (doença carotídea aterosclerótica), determinada por ecografia vascular com doppler, ambos os sexos, idade média de 51,1 (± 6,7) anos, a existência de diferenças entre de fatores associados à aterogênese, sua relação com o remodelamento do endotélio vascular e transporte reverso do colesterol. Foram padronizados e utilizados métodos com marcadores isotópicos, ELISA e zimografia por SDS-PAGE para determinação, identificação e verificação da atividade proteolítica. Resultados. Foi constatado que as amostras de soro dos pacientes portadores de DCA incorporaram menos colesterol livre triciado (CL-3H) quando comparados aos pacientes NDCA (não portadores de DCA) e, também, que os índices e demais parâmetros de avaliação do metabolismo lipídico corroboram o fenômeno (HDL-C, r=-0,330; p=0,0606, Pearson), mostrando uma tendência (HDL-C/apoA, r=0,511; p=0,0254) e relação positiva no grupo NDCA. Foram observadas correlações positivas entre MMP-1 e HDL-C (r=0,6175; p=0,002, Pearson) e MMP-1 com MMP-9 (r=0,5109; p=0,0215; Pearson) e MMP-1 com CMV-IgG (r=0,619; p=0,0012; Pearson) e correlação negativa entre MMP-9 e apoB no grupo DCA. Por ELISA foram evidenciadas, no grupo DCA, menores densidades ópticas (DO) de MMP-2 e MMP-9. Contudo, essas mesmas MMP (2 e 9) apresentaram maior atividade proteolítica no zimograma do grupo DCA. Conclusão. A menor incorporação de CL-3H no grupo DCA indica deficiência no transporte reverso do colesterol, essa alteração prejudica os mecanismos de proteção contra a aterogênese. Aliado a isso, observou-se redução das mensurações séricas das MMP no grupo DCA, a qual pressupõe a participação dessas MMP no remodelamento vascular, principalmente, nas placas e/ou lesões ateroscleróticas. As MMP e outros marcadores avaliados nos DCA mostraram que o evento não tem participação somente de moléculas relacionadas ao transporte lipídico, mas também, aquelas relacionadas à infecção por CMV (citomegalovirus), evidenciadas pela presença de correlação positiva entre MMP-1 e CMV-IgG. A menor incorporação de CL-3H, baixa DO de MMP-2 e 9, maior atividade proteolítica no zimograma e a MMP-1 associada à infecção por CMV são fatores a serem avaliados para utilização na predição de risco cardiovascular em pacientes com DCA. As menores DO de MMP-2 e 9 nos DCA em contraposição à maior atividade dessas proteinases suscita a participação de fatores reguladores (e.g. TIMP) da atividade dessas proteinases, atuando de forma distinta daquela dos NDCA. / The cardiovascular diseases are the biggest cause of morbimortality in evidence in the emerging countries; they consist into central, cerebral and peripheral arterial diseases, mainly coronary and carotidal. Among the several risk factors and markers that contribute to the development and severity of those diseases, there are that related to atherosclerosis. This study evaluates the serum concentration and activity of matrix metaloproteinases (MMP-1, 2, 8, 9 e 12), the paraoxonase activity and the association with others factors in patients with carotidal atherosclerotic disease (DCA). Methods. It was done a transversal study to evaluate, in a population of 65 individuals with (31) and without DCA (34), indicated by vascular echography with doppler, both genders, average age 51.1 (± 6.7) years, the occurrence of differences between clinical laboratory markers and their relationship with vascular endothelium remolding and reverse cholesterol transport. It was standardized and executed methods with isotopic markers, ELISA and SDS-PAGE zymography to the determination, identification and verification of proteolytic activity. Results. From the incorporation, it was verified that the DCA patients’ serum samples assume little free cholesterol when compares to NDCA (no DCA carrier patients). Also, regarding the data of CL-3H incorporation, it was verified that the index and parameters of lipid metabolism evaluation corroborate the event (HDL-C, r=-0.330; p=0.0606, Pearson), which shows a tendency (HDL-C/apoA, r=0.511; p=0.0254) and positive relationship in the NDCA group. Besides, positive correlations between MMP-1 and HDL-C (r=0.6175; p=0.002, Pearson) and MMP-1 with MMP-9 (r=0.5109; p=0.0215; Pearson) and MMP-1 with CMV-IgG (r=0.619; p=0.0012; Pearson) and negative correlation between MMP-9 and apoB in the DCA group were observed. The ELISA demonstrates lower MMP-2 and MMP-9 optic density (DO) in DCA group. Otherwise, these same MMP (2 and 9) shows bigger digestive activity at zymogram assay in the DCA group when compared to NDCA. Conclusion. The lower incorporation of CL-3H in the DCA group drives to reverse cholesterol transport deficiency, these alterations disables the protection mechanisms against the atherosclerosis. Join to this, it was observed the serum measures reduction of MMP in DCA group that presume the participation of these MMP in the vascular remolding process, mainly on the plates or/and atherosclerotic lesions. The MMP and others markers evaluated in DCA reveal that the event does not have solely participation of molecules related to lipid transport, but also that ones related with infection with CMV (cytomegalovirus) exposed by positive correlation between MMP-1 and CMV-IgG marker. Thus, the small CL-3H incorporation, the lowest MMP-2 and 9 DO, biggest digestion of the polyacrylamide gel copolymerized with gelatin and MMP-1 when associated to CMV infection are markers that must be evaluated to predict the cardiovascular risk in DCA patients. The lower DO of MMP-2 and 9 in DCA group in opposition to the bigger activity of these proteinases give raise to participation of regulatory factors (e.g. TIMP) that run in a distinct way from NDCA group.

Efeito do uso de primers bioativos na interface dentina-adesivo / Effect of bioactive primers on dentin--adhesive interface

A longevidade das restaurações com compósitos está diretamente relacionada ao sucesso do tratamento estético. A vida útil das restaurações pode ser influenciada por vários fatores como o material empregado, tempo de uso clínico e a degradação da matriz dentinária por metaloproteinases da matriz (MMPs), que podem afetar a integridade da interface adesiva, fazendo com que a união dentina-­adesivo seja comprometida. Assim, buscando melhores resultados na durabilidade das restaurações adesivas, o objetivo deste estudo foi avaliar o efeito de substâncias inibidoras de MMPs (soluções de clorexidina -­ CHX a 0,2% e doxiciclina a 3%) e agente de ligação cruzada (extrato de semente de uva -­ grape seed extract - GSE a 15%) na Resistência de União (RU) de restaurações de resina composta submetidas à ciclagem mecânica. Cavidades de Classe I foram preparadas na área central da superfície exposta de 48 molares humanos hígidos, posteriormente separados em 4 grupos (n=12) de acordo com o tratamento de superfície recebido após o condicionamento de ácido: Grupo I -­ Controle (Sistema Adesivo -­ Adper Single Bond Plus -­ 3M ESPE); Grupo II -­ CHX 0,2% + Sistema Adesivo; Grupo III -­ GSE 15% + Sistema Adesivo e Grupo IV -­ Doxiciclina 3% + Sistema Adesivo. Após os procedimentos restauradores (Filtek Supreme Ultra -­ 3M ESPE), as amostras foram armazenadas em água destilada durante 24 horas a 37°C e subdividas em dois subgrupos: A -­ Controle (armazenado em saliva artificial) e B -­ Submetido à ciclagem mecânica (Coil Cycler, Proto-­tech, forca de 50 N, frequência de 1,5 Hz em 1 x 106 ciclos de aplicação). Após ciclagem mecânica, os corpos de prova foram cortados em palitos (0,8 x 0,8 mm; n = 5 por dente) e submetidos ao teste de microtração (Bisco) a velocidade de 1 mm/min. A atividade colagenolítica da MMP-­2 sob a ação dos primers bioativos foi analisada utilizando o kit EnzChek Gelatinase/Colagenase Assay (D-­12054, Molecular Probes, Eugene, OR, EUA). A atividade gelatinolítica das MMPs-­2 e -­9 após incubação com os primers bioativos foi avaliada através de zimografia convencional. Os resultados obtidos das duas análises foram apresentados na forma de porcentagem de inibição. Zimografia in situ foi realizada utilizando gelatina de fluorescência conjugada (Kit EnzChek Gelatinase/Collagenase Assay) como substrato de MMPs e avaliada com aumento de 40x através de microscopia de fluorescência. Posteriormente, os fótons fluorescentes foram analisados em software (ImageJ). Obtidos os resultados, os dados foram submetidos à análise estatística (2way ANOVA, Tukey, p>0,05) e demonstraram que os primers bioativos não exerceram influência significante sobre a RU dentina-­adesivo, independente da ação da ciclagem mecânica. A atividade colagenolítica, segundo zimografia convencional, demonstrou que os primers bioativos foram capazes de inibir as MMPs, sendo a CHX menos eficiente. Na análise de zimografia in situ, os primers bioativos diminuíram a atividade das MMPs localizadas na interface adesiva, com aumento da atividade gelatinolítica do grupo Controle sendo maior quando submetidos à ciclagem mecânica, efeito não encontrados no grupos tratados com primers bioativos. Concluiu-­se que, apesar de não apresentarem efeitos sobre a RU da interface adesiva, os primers bioativos apresentam ação sobre a degradação da camada híbrida. / The durability of composite resin restorations is one of the main points related to the success in aesthetic treatments. The longevity of adhesive restorations can be affected by various factors such as the materials, the clinical time of use and the dentin matrix degradation mediated by matrix metalloproteinases (MMPs). MMPs can affect the adhesive interface, compromising the bonding between dentin and adhesive. Thus, searching for better results on longevity of adhesive restorations, the aim of this study was to evaluate the effect of MMPs inhibitors (chlorhexidine -­ CHX 0.2% and doxycycline 3% ) and crosslinking agent (grape seed extract -­ GSE 15%) on resin composite bond strength (BS) of restorations submitted to load cycling. Class I cavities were performed in the central area of the exposed surface of 48 sound human molars. Then, it was divided into 4 groups (n=12) according to the surface treatment received after the acid etching: Group I -­ Control (Adhesive System -­ Adper Single Bond Plus -­ 3M ESPE); Group II -­ CHX 0.2% + Adhesive System; Group III -­ Doxycycline 3% + Adhesive System e Group IV -­ GSE 15% + Adhesive System. After restorative procedures (Filtek Supreme Ultra -­ 3M ESPE), the samples were stored in distilled water for 24 hours at 37°C and then subdivided into two subgroups (n=6): A -­ Control (stored in artificial saliva) and B -­ Submitted to load cycling (Coil Cycler, Proto-­tech, 50 N, 1.5 Hz, 1 x 106 cycles). After load cycling, the specimens were cut into sticks (0.8 x 0.8 mm; n = 5 per teeth) and submitted to the microtensile test (Bisco) at 1 mm/min. The collagenolytic activity of MMP-­2 under action of bioactive primers was analyzed using EnzChek Gelatinase/Colagenase Assay Kit (D-­12054, Molecular Probes, Eugene, OR, EUA). The gelatinolytic activity of MMPs-­2 and -­9 after incubation with the bioactive primers was evaluated by gelatin zymography. The results of these two analyzes were presented by percentage of inhibition. In situ zymography was performed using fluorescence conjugated gelatin (EnzChek Gelatinase/Collagenase Assay Kit) as MMP substrate and evaluated using 40x magnification under fluorescence microscopy. Then the fluorescent photons were analyzed with an appropriate software (ImageJ). The results were subjected to statistical analysis (2-­way ANOVA, Tukey, p<0.05) and the bioactive primers showed no significant influence on the dentin-­adhesive BS, regardless the load cycling action. According to gelatin zymography, the collagenolytic activity demonstrated that the bioactive primers were able to inhibit MMPs activity and CHX was less efficient. In situ zymography analysis, showed that the bioactive primers decreased the activity of MMPs located in adhesive interface. The data also demonstrated increased gelatinolytic activity on Control group, being higher when submitted to load cycling, and which effect was not found in the groups treated with bioactive primers. It could be concluded that although the bioactive primers did not influenced the BS of adhesive interface, they demonstrated positive results on hybrid layer degradation.

Compuseram-se quatro grupos de vinte e quatro animais, que foram submetidos à queimadura química corneal com NaOH 1N. Os tratamentos foram realizados a intervalos de 6 horas e as córneas fotografadas, nos mesmos períodos, após tingimento com fluoresceína, até a reepitelização. Doze animais de cada grupo foram submetidos à eutanásia humanitaria, 20 horas após ulcerações. O mesmo procedimento foi repetido às 42 horas para os outros doze animais de cada grupo. As córneas foram processadas para quantificação de microvilosidades do epitelio corneal por microscopia eletrônica de varredura e para quantificação da MMP-2 e -9, por zimografia. O tempo médio de epitelização em horas foi de 28,00±12,82 (SC), 29,00±11,01 (NAC), 35,00±7,01 (EDTA) e de 35,00±7,01 (GC), sem diferença signficativa (p=0,46) entre os grupos. Não se observou diferença quanto ao número de MEC entre os grupos (p>0,05). Às 20 e às 42 horas, a forma latente da MMP-2 foi significativamente mais elevada nos grupos NAC e SC (p<0,001), e às 42 horas observou-se maior concentração em EDTA, relativamente ao GC (p<0,001). Às 42 horas, a concentração da MMP-2, em sua forma ativa, foi significativamente maior, no grupo SC, relativamente aos demais grupos (p<0,001). A MMP-9 só se expressou em sua forma latente, sendo sua concentração significativamente mais elevada, no grupo NAC, às 42 horas (p<0,001). Concluiu-se, que das substâncias testadas, só o sulfato de condroitina acelerou a epitelização corneal, mas nenhuma delas apresentou efeitos protetores às microvilosidades do epitélio corneal... / This study aimed evaluate the effects of topical 3% chondroitin sulfate (SC), 10% N-acetylcysteine (NAC), 1% EDTA (EDTA) and 0,9% NaCl on corneal epithelial viability and the expression of matrix metalloproteinase-2 and -9 (MMP-2 and -9), in alkali burned rat corneas. Ninety eight healthy rats were divided into four groups and animals were submitted to corneal burn with NaOH 1N. All treatments occurred every 6 hours, and the corneas were photographed at the same time points until lesions were fluorescein negative. Twelve animals of each group were euthanized 24 and 42 hours after burn, and corneas were processed for corneal epithelial microvilli quantification (CEM) by scanning electron microscopy and MMP-2 and -9 evaluation by zimography. Images were quantified by Image J. Average corneal wound healing rate was 28,00±12,82 (SC), 29,00±11,01 (NAC), 35,00±7,01 (EDTA) and 35,00±7,01 hours in controls (GC) (p=0,46). CEM count did not change significantly among groups (p>0,05). At 20 and 42 hours, MMP-2 latent form quantity was significantly increased in NAC and SC groups (p<0.001); at 42 hours, MMP-2 latent form was significantly elevated in EDTA, in comparison to GC (p<0,001). At 42 hours, MMP-2 active form was significantly increased only in SC (p<0,001). MMP-9 was expressed only in its latent form, being significantly higher in NAC, at 42 hours (p<0,001). This study indicated that only 3% chondroitin sulfate accelerated corneal wound healing. 10% N-acetylcysteine, 1% EDTA and 3% chondroitin sulfate did not protect the corneal epithelial microvilli.
