Desenvolvimento de queijo Minas frescal probiótico com \'Lactobacillus acidophilus\' e \'Bidifobacterium lactis\' isolados e em co-cultura / Probiotic minas fresh cheese with Lactobacillus acidophilus and Bidifobacterium lactis added as individual cultures or in co-culture

O interesse do consumidor por produtos alimentícios saudáveis e até capazes de prevenir doenças, como alimentos funcionais probióticos, tem aumentado visivelmente nos últimos anos. A possibilidade de se desenvolver produtos lácteos de grande aceitabilidade nacional como o queijo Minas frescal com microrganismos probióticos tem futuro promissor. O presente trabalho teve como objetivo verificar a influência do emprego de culturas probióticas compostas pelos microrganismos Lactobacillus acidophilus e Bifidobacterium lactis isolados e em co-cultura na tecnologia de fabricação de queijo Minas frescal sobre as características do produto ao longo de seu armazenamento refrigerado. Os queijos foram preparados com acidificação com ácido lático e 4 tratamentos (em quadruplicata) foram empregados para se avaliar a ação dos microrganismos probióticos: T1 (controle), T2 (adição de L. acidophilus), T3 (adição de B. lactis) e T4 (adição de L. acidophilus + B. lactis). Avaliou-se o comportamento de L. acidophilus, B. lactis, bactérias láticas totais, Staphylococcus spp., coliformes totais e E. coli. Paralelamente, foi feita a determinação do perfil de textura dos queijos através de teste de dupla compressão de amostras cilíndricas, em analisador de textura TA-XT2 (Stable Micro System) e determinação de umidade, pH, acidez titulável e atividade de água. As análises foram realizadas no dia de processamento (dia 1) e após 7, 14 e 21 dias de armazenamento a 5 C. As análises microbiológicas também foram realizadas no leite e no queijo em processamento. A comparação entre os queijos foi estudada também através de análise sensorial, empregando-se teste de aceitação/preferência. Os queijos T4 revelaram-se mais apropriados quanto à multiplicação das bactérias probióticas. Com relação a multiplicação de coliformes, Staphylococcus spp. e bactérias láticas totais, não foi observada diferença significativa (p>0,05) entre os tratamentos. Houve, porém, aumento significativo ao longo do armazenamento para Staphylococcus spp. nos queijos T3 e para coliformes totais nos queijos T1 (p<0,05). E. coli não foi detectado em nenhum dos queijos. A atividade de água não atuou como obstáculo no desenvolvimento de microrganismos, ficando sempre acima de 0,97 em todos os queijos. O pH e a acidez mostraram-se estatisticamente iguais (p>0,05) em todos os 4 queijos. Houve, porém, queda estatisticamente significativa do pH médio e aumento significativo da acidez (p<0,05) nos queijos T1 e T2. Nos parâmetros de textura, observou-se aumento significativo da dureza que é o parâmetro mais importante, na primeira semana para todos os queijos (p<0,05) e nas datas seguintes, tendência à estabilidade exceto para T3 que mostrou novos aumentos significativos (p<0,05) em 7 e 14 dias. A coesividade, a elasticidade e a adesividade mostraram-se estatisticamente iguais (p>0,05) em todas as datas de amostragem em todos os queijos. Na avaliação sensorial dos queijos, não foi observada aceitabilidade diferenciada para nenhum dos queijos. Concluiu-se que o queijo frescal probiótico tipo Minas frescal mostrou-se com propriedades sensoriais, de textura e microbiológicas adequadas e apropriado como veículo para L. acidophilus e B. lactis, porém com melhor desenvolvimento destes quando em co-cultura (T4), associado ao fato deste queijo ter se comportado da forma mais estável dentre os queijos estudados, quando avaliados todos os pontos de variação significativa ao longo do armazenamento. / Functional foods consuming habits have been increasing over the years, particularly in the case of probiotic dairy food. The possibility of developing new probiotic dairy products, like the popular Brazilian Minas fresh cheese, is very promising. The objective of the present work was to verify the influence of the probiotic cultures containing Lactobacillus acidophilus and Bifidobacterium lactis, added individually and in co-culture during fresh Minas cheese production, over the microbiological, textural and physico-chemical behavior of the product during refrigerated storage. Four treatments were prepared (four times each), all of them were made through direct acidification with lactic acid: T1 (control), T2 (addition of L. acidophilus), T3 (addition of B. lactis) e T4 (addition of L. acidophilus + B. lactis). Microbiological counts of L. acidophilus, Bifidobacterium spp., total lactic acid bacteria (LAB), coliforms, Staphylococcus spp. and E. coli and analysis of pH, acidity, moisture content and water activity proceeded after 1, 7, 14 and 21 days of storage at 5 C. Microbiological analysis was also conducted in milk and during cheese manufacturing process. Additionally, texture properties of cheeses were evaluated during storage, employing a TA-XT2 Texture Analyser (Stable Micro Systems, Haslemere, England), using a two-bite compression of cylindrical samples. Parameters measured consisted of hardness, elasticity, cohesiveness, chewiness, gumminess and adhesiveness. Comparison of cheeses was also conducted through sensory evaluation after 7 days of storage, using Preference- Ranking test. Cheeses T4 revealed to be more appropriate for growth of both probiotic bacteria, maintaining populations of 6 log CFU/g or above. No significant differences (p>0.05) between treatments were observed in relation to coliforms, Staphylococcus spp. and LAB, though Staphylococcus spp. and coliforms increased significantly during storage of cheeses T3 and T1, respectively (p<0.05). E. coli was not detected in any of the cheese and milk samples. Water activity values stayed above 0.970 for all samples, therefore proper for microbial growth. Mean pH and acidity did not differ significantly among treatments (p>0.05). Nevertheless, cheeses T1 and T2 revealed a significant decrease in pH (6.2 to 5.9 and 6.7 to 6.1, respectively) and increase in acidity from day 1 to day 21 of storage (p<0.05). As for the texture profile of cheeses, a significant increase in hardness (p<0.05) was detected for all cheeses studied in the first week of storage, followed by a tendency for stability during the subsequent storage period, except for cheeses T3, for which a significant increase in hardness was also detected from day 7 to day 14 of storage. Cohesiveness, springiness and dhesiveness showed to be statistically equal (p>0.05) for all cheeses and for all sampling days. No significant differences were detected through sensory evaluation between the four kinds of cheeses studied (X0<c2). Results indicated that probiotic fresh Minas cheese presented adequate sensorial, textural and microbiological properties. Therefore, Minas cheese is a suitable vehicle for delivery of L. acidophilus and B. lactis, especially when they are in a co-cultural system, condition which resulted in a slightly better survival rate of both probiotic strains and in a more stable product during refrigerated storage.

The probiotic used was composed of Lactobacillus acidophillus, Bifidobacterium bifidum and Streptococcus faecium. The characteristics evaluated were weight gain, final weight, average feed intake, feed conversion, viability 1-40 days of age. At 21 and 35 days of age were measured blood parameters (calcium, phosphorus, uric acid and total protein). At 40 days was carried out the slaughter of birds to assess the yield of carcass, breast, drumstick, thigh, wing and abdominal fat percentage. Within the analysis of economic viability was calculated the cost of the experimental diets, the index of economic efficiency and cost index. In the initial phase and growth was not found significant results for the performance variables. In the total periods of 1-40 days of age for the variable feed no significant effect (P <0.05) for levels of inclusion. Carcass yield was influenced (P <0.05) by food type regardless of the levels of inclusion of inulin. For the cuts, only the thigh showed a significant difference (P <0.05) for the type of diet. Was observed in abdominal fat interaction (P <0.05) between type of diet (plant and animal) and the levels of inclusion of inulin. For the blood parameters for 21 days was significantly different (P <0.05) between levels of inulin inclusion for the variables P and total protein, and 35 was again a significant difference (P <0.05) for phosphorus . The other variables were not affected with the use of inulin. The use of 2.5 g / kg inulin provides better feed for broilers at 1-40 days old. Inclusion of inulin reduces abdominal fat independent mind the type of diet. The use of feed containing animal ingredients provides improved higher carcass yield and higher thigh yield. The use of inulin affected the phosphorus stock you live at 21 and 35 days old. The use of inulin in diets of broiler feed cost rises mind independent of the use of ingredients of animal or vegetable / Objetivou-se com este trabalho avaliar o efeito de diferentes níveis de inclusão de inulina (extrato de raiz chicória) em rações contendo ingredientes de origem vegetal e animal, sobre o desempenho, as características de carcaça (cortes nobres e gordura abdominal) e os parâmetros sanguíneos, para frangos de corte de 1 a 40 dias de idade. Foram utilizados 1056 pintos de um dia de idade, alojados sobre cama reutilizada, em um delineamento experimental inteiramente casualizado, com oito tratamentos e seis repetições e 22 aves por unidade experimental, os quais foram arranjados em um esquema fatorial 2 x 4, onde o primeiro fator foi a utilização de duas dietas (vegetal e animal) e o segundo fator representado por quatro níveis de inclusão de inulina (0,00; 0,25; 0,50; 0,75%). O probiótico utilizado era composto por Lactobacillus acidophillus; Streptococcus faecium e Bifidobacterium bifidum. As características avaliadas foram ganho de peso, peso final, consumo médio de ração, conversão alimentar, viabilidade de 1 a 40 dias de idade. Aos 21 e 35 dias de idade foram mensurados os parâmetros sanguíneos (cálcio, fósforo, ácido úrico e proteínas totais). Aos 40 dias foi realizado o abate das aves para avaliar o rendimento de carcaça, peito, coxa, sobrecoxa, asa e percentual de gordura abdominal. Dentro da análise da viabilidade econômica foram calculados os custos das dietas experimentais, o índice de eficiência econômica e o índice de custo. Na fase inicial e de crescimento não foi encontrado resultado significativo para as variáveis de desempenho. No período total de criação de 1 a 40 dias de idade, para a variável conversão alimentar houve efeito significativo (P<0,05) para os níveis de inclusão. O rendimento de carcaça foi influenciado (P<0,05) pelo tipo de ração independente dos níveis de inclusão de inulina. Para os cortes, somente a coxa apresentou diferença significativa (P<0,05) para o tipo de ração. Na gordura abdominal foi observado interação (P<0,05) entre o tipo de ração (vegetal e animal) e os níveis de inclusão de inulina. Para os parâmetros sanguíneos aos 21 dias foi observada diferença significativa (P<0,05) entre os níveis de inclusão inulina para as variáveis fósforo e proteínas totais, e aos 35 novamente foi observada diferença significativa (P<0,05) para o fósforo. As demais variáveis não foram afetadas com a utilização da inulina. A utilização de 2,5g/kg de inulina proporciona melhor conversão alimentar para aves de 1 a 40 dias de idade. A inclusão de inulina reduz a deposição de gordura abdominal independentemente do tipo de ração. A utilização de ração com ingredientes de origem animal proporciona melhor rendimento de carcaça e maior rendimento de coxa. A utilização de inulina afetou os níveis de fósforo circulante aos 21 e 35 dias de idade. A utilização de inulina em rações de frango de corte eleva o custo da ração independente mente da utilização de ingredientes de origem animal ou vegetal

Colonização oral experimental por Candida albicans em camundongos imunossuprimidos e tratados com Lactobacillus acidophilus e Lactobacillus rhamnosus / Experimental oral colonization by Candida albicans in immunosuppressed mice treated with Lactobacillus acidophilus and Lactobacillus rhamnosus

Bactérias probióticas, como Lactobacillus sp, são conhecidas como inibidoras do crescimento de microrganismos patogênicos, têm a capacidade de modificar o equilíbrio microbiológico do hospedeiro e reduzir o crescimento de patógenos, como o microoganismo Candida albicans. Para avaliar a colonização oral experimental e o seu tratamento com probióticos, 152 camundongos DBA/2 imunossuprimidos foram inoculados oralmente com uma suspensão (108 células viáveis) de C. albicans. Os animais foram divididos em 4 grupos: controle positivo (sem tratamento), tratados oralmente com nistatina, com Lactobacillus acidophilus e Lactobacillus rhamnosus. No grupo que recebeu nistatina, o tratamento foi iniciado um dia após a inoculação por Candida, já nos grupos que receberam as bactérias probióticas, os tratamentos foram iniciados 14 dias antes da inoculação. Tratamentos com nistatina e probióticos foram diários e duraram 13 dias. As avaliações foram realizadas 1, 3, 5, 7, 9, 11 e 13 dias após a inoculação inicial e feitas através de análises microbiológicas da mucosa oral dos animais. A colonização por C. albicans iniciou-se um dia após a inoculação, sendo o aumento das unidades formadoras de colônia progressivo e significante até o sétimo dia. Após este período, observou-se uma redução significativa no isolamento de leveduras. Todos os tratamentos probióticos reduziram significativamente a colonização de C. albicans na mucosa oral dos animais, comparada com a do grupo de animais não tratados. No grupo tratado com L. rhamnosus, a redução da colonização de levedura foi significativamente maior comparado ao grupo nistatina. Concluiu-se que o modelo animal DBA/2 imunossuprimidos é um bom modelo experimental para o estudo da colonização oral e o tratamento com probióticos, no presente modelo, pode ser uma alternativa eficaz para a redução da Candida na cavidade oral. / To evaluate experimental oral candidiasis and the treatment using probiotics, 152 DBA/2 mice after being immunosuppressed were orally inoculated with a suspension of C. albicans containing 108 viable cells of yeast. The animals were devided into four groups: positive control (untreated), trated oraly with nystatin, with Lactobacillus acidophilus and with Lactobacillus rhamnosus. In the group that received nystatin, the treatment was initiated one day after Candida inoculation, and in the groups that received the probiotic bacteria, the treatment began fourteen days before inoculation. Treatments with nystatin and probiotics were daily and lasted 13 days. Evaluations were performed at 1, 3, 5, 7, 9, 11 and 13 days (after the initial inoculation) and made by microbiological analysis of the oral mucosa of these animals. The colonization of C. albicans in the oral mucosa animals began one day after the initial challenge and it was progressive and significant until the seventh day, when there was a significant reduction in the isolation of yeast. All treatments with probiotic bacteria significantly reduced the colonization of C. albicans in oral mucosa of the animals, compared to the untreated animal group. In group treated with L. rhamnosus the reduction of colonization of yeast was significantly higher compared to the group receiving nystatin. Based on the findings of this study we suggest that animal model DBA/2 immunosuppressed is a good model for experimental oral candidiasis and the treatment with probiotics in this model may be an effective alternative to the treatment of oral candidiasis.

DESENVOLVIMENTO DE CREAM CHEESE SIMBIÓTICO: CARACTERIZAÇÃO E PERFIL LIPÍDICO COM ÊNFASE EM ÁCIDO LINOLÉICO CONJUGADO / DEVELOPMENT OF SYMBIOTIC CREAM CHEESE: CHARACTERIZATION AND LIPID PROFILE WITH EMPHASIS ON CONJUGATED LINOLEIC ACID

Fresh cheeses are propitious to addition of probiotics due to their suitable pH, acidity and moisture. The objective of this study was evaluate the potential of cream cheese as a vehicle of probiotics, the physical-chemical, microbiological and sensory characteristics and the lipid profile of this product when additioned of prebiotic and probiotics. Treatments of cream cheeses were produced with addition of different levels of probiotics (Bifidobacterium animalis and Lactobacillus acidophilus) and prebiotic (inulin) in amounts according to Central Composite Design, totalizing 12 treatments, with 4 fatorials, 4 axial, 3 repetitions on central point and 1 control. The characteristics were evaluated in all treatments during storage for 45 days at 8°C. No significant changes were observed on physical-chemical characteristics of fat, protein and ash in comparison with control, while moisture was altered by prebiotic addition. The surface response was done to verify the influence of different levels of inulin and probiotic on pH and tritable acidity during storage; only linear and quadratic terms of probiotic level were significant (p<0,05). The pH and tritable acidity affect the viability of cultures; starter culture (S. termophilus) exhibited high viability during storage. Bifidobacterium animalis showed counts above 106 UFC g-1 during all period, assuring the probiotic potential in all treatments. The counts of Lactobacillus acidophilus reduced along storage, probably by competition with starter culture. Assessment of sensory attributes (global appearance, color, aroma, texture, acidity and taste) was evaluated using test with hedonic scale of 7 levels and ordering test. No significant difference was found by Tukey s test (p>0,05) on assessment using hedonic scale, neither on the ordering test. The linear, quadratic and interaction terms of multiple linear regression were not significant (p>0,05) for any sensory attribute; only the intercept of each equation was significant (p<0,01). Lipid profile was not significantly changed by the different levels of probiotic, with amounts of saturated fatty acids, monounsaturated, polyunsaturated and trans fatty acids similar to that of the milk used on cheese manufacture and control. The different levels of probiotic no altered the concentration of conjugated linoleic acid (CLA) in comparison with control, probably due to the low addition of probiotic, the reduced viability of Lactobacillus acidophilus during storage and the short shelf-life of cream cheese, with small time to produce CLA. In conclusion, cream cheese with addition of prebiotic and probiotic on levels of this study showed functional potential; nevertheless, the amount of probiotic used does not increase the concentration of CLA compared to the traditional product. / Derivados lácteos compõem a maior parte dos alimentos funcionais disponíveis no mercado. Queijos frescos são propícios à adição de probióticos por seu pH, acidez e umidade favoráveis. O objetivo deste estudo foi avaliar o potencial do cream cheese em atuar como condutor de probióticos, as características físico-químicas, microbiológicas e sensoriais e o perfil lipídico do produto quando adicionado de prebiótico e probióticos. As formulações de cream cheese foram elaboradas com adição de diferentes concentrações de probiótico (Bifidobacterium animalis e Lactobacillus acidophilus) e de prebiótico (inulina), em quantidades adotadas de acordo com o Delineamento Central Composto Rotacional, totalizando 12 ensaios, sendo 4 fatoriais, 4 axiais, 3 repetições no ponto central e 1 tratamento-controle. As características foram avaliadas durante o armazenamento do produto por 45 dias a 8°C. Não foi observada mudança significativa nas características físico-químicas quanto a gordura, proteína e cinzas em comparação à formulação-controle, enquanto o teor de umidade foi alterado pela adição do prebiótico. A superfície de resposta foi elaborada para verificar a influência das diferentes concentrações de inulina e probióticos sobre o pH e acidez (expressa em ácido láctico) das formulações durante o armazenamento, tendo somente os termos linear e quadrático da concentração de probióticos apresentado significância (p<0,05). O pH e acidez titulável das formulações influenciaram a viabilidade das culturas, tendo a cultura starter (S.termophilus) alta viabilidade durante todo o armazenamento. Bifidobacterium animalis apresentou contagens acima de 106 UFC/g durante todo o período, garantindo a potencialidade probiótica de todas as formulações. As contagens de Lactobacillus acidophilus declinaram ao longo do armazenamento, provavelmente por competição com a cultura starter. A avaliação dos atributos sensoriais (aparência global, cor, aroma, textura, acidez e sabor) foi realizada através de teste com escala hedônica de 7 níveis e teste de ordenação. Não foi encontrada diferença estatística pelo Teste de Tukey (p>0,05) na avaliação por escala hedônica, tampouco preferência significativa no teste de ordenação. Os termos linear, quadrático e de interação da regressão linear múltipla não foram significativos (p>0,05) para nenhum dos parâmetros sensoriais avaliados; somente o intercepto da equação de cada atributo apresentou significância estatística (p<0,01). O perfil lipídico das formulações não foi significativamente alterado pela adição de diferentes concentrações de probiótico, sendo as quantidades de ácidos graxos saturados, monoinsaturados, poliinsaturados e trans semelhantes aos do leite usado como matériaprima e à formulação-controle. As diferentes concentrações de probiótico igualmente não alteraram a concentração de ácido linoléico conjugado (CLA) em relação ao controle, provavelmente pela reduzida concentração de cultura usada, baixa viabilidade de Lactobacillus acidophilus durante ao armazenamento e ao fato de cream cheese ter vida-de-prateleira breve, com curto espaço de tempo para formação do CLA. Conclui-se que o cream cheese adicionado das concentrações estudadas de pre- e probiótico apresenta potencial funcional; entretanto, a quantidade de probióticos não aumentou o teor de CLA em relação às quantidades do produto tradicional.

Objetivo: Evaluar la actividad antibacteriana de tres ionómeros de vidrio (CIVc) Ion Z®, Ketac Molar Easymix® y Fuji IX GP® sobre Streptococcus mutans (ATCC 35668) y Lactobacillus acidophilus (ATCC 4366). Materiales y métodos: El presente estudio fue experimental in vitro. Se realizó mediante el método de contacto directo y densidad óptica con una microplaca, donde se utilizó seis pocillos por cada ionómero de vidrio. Posteriormente, se adicionaron 150 ul de Streptococcus mutans y Lactobacillus acidophilus de manera independiente en agar BHI. Las muestras fueron incubadas a 37 °C en condiciones de anaerobiosis controlada. En el caso de la viabilidad bacteriana se utilizó el ensayo de MTT. Resultados: Se encontró que el efecto antibacterianode Ketac Molar Easymix® frente a la cepa de S. mutans tuvo 0.06 ± 0.01, mientras que Ion Z® 0.16 ± 0.03 ante la cepa de L. acidophilus, presentando diferencias estadísticamente significativas. Con respecto a la viabilidad bacteriana de S. mutans frente al Ketac Molar Easymix® obtuvo 0.92 ± 0.02, lo que corresponde a 29 % de bacterias viables, mientras que, L. acidophilus frente al Ion Z® tuvo 0.31± 16, que representa 19% de bacterias viables. Conclusiones: Los tres CIVs mostraron actividad antibacteriana, en el caso de Ketac Molar Easymix® demostró mayor efecto antibacteriano ante S. mutans, por otro lado, se evidenció que Ion Z® fue más eficaz frente L. acidophilus. / Objective: To evaluate the antibacterial activity of three glass ionomers cements (Ion Z®, Ketac Molar Easymix® and Fuji IX GP®) on Streptococcus mutans (ATCC 35668) and Lactobacillus acidophilus (ATCC 4366). It was made out using the direct contact method and optical density with a microplate, using six wells for each glass ionomer cement. Later, 150 ul of Streptococcus mutans and Lactobacillus acidophilus were independently added on BHI agar. The samples were incubated at 37 °C under conditions of controlled anaerobiosis. In the case of bacterial viability, the MTT assay was used. Results: It was found that the antibacterial effect of Ketac Molar Easymix® against the strain of Streptococcus mutans had 0.06 ± 0.01, while in Z® it was 0.16 ± 0.03 before the strain of Lactobacillus acidophilus, presenting statistically significant differences. Regarding the bacterial viability of Streptococcus mutans against Ketac Molar Easymix® obtained 0.92 ± 0.02, what corresponds to 29% of viable bacteria, whereas Lactobacillus acidophilus against Ion Z® had 0.31± 16, which represents 19% of viable bacteria. Conclusions: The three glass ionomers cements showed antibacterial activity. Ketac Molar Easymix® demonstrated greater antibacterial effect against Streptococcus mutans; on the other hand, it was evidenced that Ion Z® was more effective against Lactobacillus acidophilus. / Tesis

As formulações de bebidas fermentadas BLF e BLFC apresentaram diferenças significativas (p < 0,05) em relação às amostras não fermentadas BL e BLC nos parâmetros glicídeos redutores em lactose e em sacarose, pH e índice de proteólise, com menores médias absolutas e acidez com maiores médias em todos os tempos estudados. Entretanto, a amostra controle (BL) apresentou contagens... / The goal of this work was to develop a strawberry flavored dairy beverages carbonated and fermented with probiotic bacteria. Four formulas of dairy beverages were elaborated: Control (BL); Fermented (BLF); Carbonated (BLC) and Carbonated Fermented (BLFC). In samples submitted for carbonation, a carbonator was used for the carbon dioxide (CO2) gas injection dissolved in drinking water, resulting in a product with a ratio of 2:1 (volume of gas volume of dairy beverages) and the cultivation consisting of lactic bacteria Lactobacillus acidophilus-LA-5®, Bifidobacterium BB-12® and Streptococcus thermophilus was employed on the fermented samples. The samples were characterized by physical and chemical parameters (chemical composition, proteolysis index and carbonation proportion), microbiological parameters (total coliform counts, E. coli, yeasts and molds and viability of the lactic bacteria) and sensory aspects through acceptance testing and purchase intention, over the course of 28 days of refrigerated storage of the dairy beverages. The formulations of the BLF and BLFC fermented drink presented significant differences (p < 0,05) in relation to the non-fermented samples BL and BLC in glucides reducer of lactose and sucrose parameters, pH and proteolysis index, with lower absolute averages and acidity with higher averages at all studied times. Isolation and Biochemical Characterization of Acid- and Bile- Tolerant Strains of Lactobacillus Acidophilus and Bifidobacterium Bifidum

Lactic acid bacteria have been reported to be used as a health adjunct in food for u many years. However, these health benefits have not been proven. and how these bacteria pass through the digestion process and remain viable in the human intestinal tract is still not clear. The aim of this work was to isolate mutants from Lactobacillus acidophilus or Bifidobacterium bifidum that could tolerate the conditions of the digestion process (low pH and bile conduction) and to characterize these isolated mutants. Acid- and bile-tolerant mutants of L. acidophilus were isolated from parental strains successfully using natural selection techniques. These mutants survived and grew at conditions of pH 3.5 with 0.2% mixed bile salts added. After the selection, phenotypic characterization was identified to further clarify desirable traits for use as probiotic adjuncts in foods. These phenotypic characteristics included protease, aminopeptidase, ß-galactosidase, and bile salt hydrolase activity. Based on different protease, aminopeptidase, and ß-galactosidase activity, selected acid- and bile-tolerant mutants contained different growth characteristics compared with their parents. All the isolates tested showed different bile salt hydrolase activity, and this activity was not strain and medium dependent. Plasmid profiles and fatty acid analysis were conducted to provide more information of these acid/bile tolerant isolates and whether or not they were mutants from their parent strains rather than only adapted variants. Results showed the acid-/bile-tolerant isolates contained different plasmid profiles and cell wall fatty acids compared with their parents, which indicated these isolates were mutants. Protein expression by two-dimensional gel electrophoresis showed different protein expression patterns between acid- and bile-tolerant mutants and their parents. fm1her suggesting these isolates were mutants. We observed the protein production in parent strains decreased as the pH decreased. and protein expression in mutants remained the same as pH decreased. Two of the proposed health benefits of probiotic bacteria are anticholesterol activity and antimicrobial activity. These were evaluated using selected acid- and bile-tolerant mutants. Results showed no decrease of cholesterol in the test medium during bacterial growth. The observed antimicrobial activity was due to the presence of active cells. and this may relate to the acid production during cell growth and not to the production of antimicrobial substances. We concluded that the acid-/bile-tolerant isolates were mutants, and they survived and grew better in harsh environments compared with their parent strains. These mutants may be useful as a food adjunct in the future, but further study is needed to establish their use and possible probiotic benefits.

Studies on enhancing the viability and survival of probiotic bacteria in dairy foods through strain selection and microencapsulation

In this study, strains of probiotic bacteria have been selected for tolerance to low pH, bile, sucrose, oxygen in media and low storage temperatures. Lactobacillus acidophilus 2401 and Bifidobacterium infantis 1912 were selected as strains able to survive in these conditions. These two strains were then offered further protection from the adverse conditions of food processing and storage by microencapsulation in a calcium alginate and starch gel matrix. Encapsulation in calcium alginate increases survival in yoghurt. In cheddar cheese the free L. acidophilus 2401 and B. infantis 1912 cells survived better than the encapsulated cells, probably due to the dense nature of the cheddar cheese matrix combined with the encapsulation restricting the flow of the nutrients and metabolites between the outside environment and the cells. In ice cream survival was high, probably due to the high fat and solids nature of the ice cream combined with the low storage temperature. The trial results of the laboratory scale production was consistent with the survival results for yoghurt and cheddar cheese. Incorporation of encapsulated probiotic bacteria into ice cream and cheddar cheese was acceptable by sensory standards and largely unnoticeable in comparison with the same foods without capsules. The capsules were visible and able to be felt on the tongue when eaten in yoghurt causing the product to be disliked by the panellists. / Master of Science (Hons)

Probiotics are viable microorganisms that show beneficial effects on the health of the host by improving their intestinal microflora. The microorganisms applied as probiotics mainly include Lactobacillus and Bifidobacterium species. Probiotics can inhibit the bacterial pathogens, reduce serum cholesterol levels, improve lactose tolerance and stimulate the immune response. They also have other properties such as / tolerance to acid and bile salts, adherence to gastrointestinal cells for colonization, resistance to antibiotics and &beta / -galactosidase acitivity. The properties of probiotic products are determined by the characteristics of the microorganisms they contain. For that reason, isolation and characterization of new strains having probiotic properties is an important issue. New strains are generally isolated from their natural habitats which are fermented dairy products such as kefir. In order to exert beneficial health affects in the digestive system, commercial probiotic products should contain adequate numbers of viable cells. Probiotic microorganisms should protect their viability during their shelf storage. Therefore, the viability of probiotics is especially important for food manufacturers that search for new probiotic strains with good survival and stability properties upon storage. In this study, probiotic microorganisms were isolated from traditional kefir grains known as a &lsquo / complex probiotic&rsquo / . The isolates were firstly identified using biochemical tests, then the putative species belonging to &lsquo / Lactobacillus acidophilus group&rsquo / were identified with 16S rRNA gene sequencing. Analysis of sequencing resulted in differentiation of &ldquo / L. acidophilus group&rdquo / organisms, namely L. amylovorus and L. acidophilus. Moreover, typing of commercial and traditional L. acidophilus strains and L. amylovorus strains were performed with RAPD-PCR by using primer M13. While several L. acidophilus strains showed different RAPD fingerprints most of the L. acidophilus and L. amylovorus strains could not be differentiated due to high similarity of their RAPD fingerprints. Following identification, survival of these isolates in probiotic yogurt preparations were investigated and compared to the survival of commercial probiotics. Consequently, although the survival of kefir grain isolates were less than commercial probiotics, they sustained the minimum recommended level for probiotics (106 cfu/ml) during cold storage. Such level of survival makes them considerably good candidates to be used as commercial probiotic cultures.

The effect of two Lactobacillus strains and an acidophilic fungus on production and immune responses of broiler chickens /

Three potential probiotics, Lactobacillus acidophilus, Lactobacillus casei and the fungus Scytalidium acidophilum were prepared in different ways. They were evaluated for their effects on performance and immune responses in broiler chickens in two 6-week trials. In the first trial, probiotics were orally administered at the ages of day 1 and day 8. There were no differences between live and disrupted cells. However, cobalt-containing cells had improved effects numerously in comparison with non cobalt-containing cells. As a result, these three disrupted probiotics, each in two different concentrations, were sprayed onto a mash basal feed before pelleting in the second trial. Improved feed intake (up to 6%) and body weight gain (up to 7%) were observed in all probiotic treatments. The anti-KLH IgA and IgG titers of probiotic-treated chickens were not significantly different from the negative controls in both trials. In summary, disrupted and cobalt-containing probiotics were proved to enhance the performance of broiler chickens and are promising for commercial application. In addition, a new fungal mycelium, Scytalidium acidophilum, was suggested to be a good probiotic to improve the growth of broilers.
