Florian, Stephanie 16 October 2007 (has links)

Life History Studies of Two Digenetic Trematodes, Bolbophorus Damnificus and an Unknown Clinostomoid Species, that Infect Channel Catfish Ictalurus Punctatus

Doffitt, Cynthia Michelle 09 December 2011 (has links)
The commercial production of channel catfish (Ictalurus punctatus) is major industry in Mississippi. Infections of channel catfish with the digenetic trematode Bolbophorus damnificus have often been associated with heavy economic losses in the industry. To efficiently control transmission of this trematode, the avian hosts need to be identified. In the first study, two American white pelicans, two double-crested cormorants, two great blue herons, and two great egrets were fed channel catfish infected with B. damnificus metacercariae. The presence of Bolbophorus damnificus ova in pelican feces at three days post infection (dpi) indicated the pelicans had patent infections. Mature B. damnificus were recovered from the intestines of both pelicans at 21 dpi. No B. damnificus infections were observed in the other bird species. In a second study, 33 American white pelicans, 34 double-crested cormorants, 35 great blue herons, and 32 great egrets were collected in the Mississippi Delta. The prevalence of B. damnificus in the American white pelican was 93.9%, with an average of 158 B. damnificus found per bird (range 0-681). Bolbophorus damnificus was not found in any of the other bird species. The results of these two studies confirm that the AWPE is the only proven natural host for B. damnificus. In a third study, two previously undescribed cercariae were found infecting rams-horn snails in commercial catfish ponds. In challenge studies, channel catfish were exposed to both cercariae types. Only one type of cercariae (type I) was infective to channel catfish. The first evidence of type I metacercariae was seen histologically at 14 dpi and grossly at 21 dpi. Development continued until 120 dpi, when both gross examination and histology suggested that the metacercariae were mature. The type I metacercariae appeared to cause little host damage. Molecular analysis of the 18S rRNA gene region indicated that the type I cercariae and metacercariae may be a species of Clinostomum. The data generated in these three studies provides additional information that can be used in the development of efficacious management schemes to control digenetic trematodes infecting commercial catfish.

D'Aloia, Maria 25 May 2007
Onset of flowering is a major transition in the plant life cycle and is controlled by environmental factors including photoperiod, light quality and temperature. Prevalence of controlling factors depends on species, hence physiological models were selected for their strong requirement for one or another environmental cue. Among Brassicaceae, Sinapis alba was intensively studied for its strong response to photoperiod while molecular-genetic analyses of Arabidopsis thaliana disclosed complex interactions between pathways inducing flowering in response to photoperiod and other environmental cues, such as vernalization. We were therefore interested in studying the vernalization process in S. alba and its interactions with the previously characterized floral response to long days (LDs). Two-week old seedlings grown in non-inductive short days (SDs) were vernalized at 7°C for increasing durations and a strong promotive effect of vernalization was observed. In contrast to the common view of vernalization as a preliminary step bringing the competence to flower, we observed that vernalization had a direct inductive effect on flowering: floral buds were initiated during cold-exposure. Floral integrator genes SaMADSA (homologous to SUPPRESSOR OF OVEREXPRESSION OF CO 1) and SaLEAFY were up-regulated in the shoot apex after 3-4 weeks of vernalization. To monitor the vernalization process at the molecular level, we isolated SaFLC which, based on sequence analysis, expression patterns and complementation test, appeared as orthologous to FLOWERING LOCUS C. Down-regulation of SaFLC by vernalization was fast since transcript level was already very low after one week of vernalization, but stability of the repression required longer exposure to cold. To test the physiological significance of these observations, we studied the floral response to 16-h LDs after unstable and stable repression of SaFLC. We observed that one week of vernalization which was sufficient for SaFLC repression but not for maintenance of the silenced state - increased the flowering response of S. alba to LDs when the LDs just followed the cold treatment. This effect was lost after two weeks post-vernalization. In contrast, the promotive effect of longer vernalization on flowering response to LDs was maintained post-vernalization. These results suggested that vernalization not only works when plants experience long exposure to cold in winter: short cold periods might stimulate flowering of LD-plants if occurring when photoperiod is increasing, i.e. in spring.

Interactions Among Rodents, Owls, Food Resources and Habitat Structure in a Malaysian Oil Palm Agroecosystem

Chong Leong Puan Unknown Date (has links)
Rodents often colonise agricultural lands and become pests that cause economic loss and crop damage. Eradicating or regulating pest numbers has economic and environmental costs and has always been of great interest to farmers and nature conservationists. To reduce rodent numbers poison baits are used more often than biological control. However, poisoning has adverse environmental impacts and ignores the underlying biological factors that influence pest abundance. This study examined the interactions of the common rat species of oil palm plantations, namely Rattus rattus diardii, R. argentiventer and R. tiomanicus, with their food resource, habitat structure and introduced predators in an attempt to provide a better understanding and approach for their control. To investigate the interrelationships among rodents, owls and oil palms, rodent trapping was conducted simultaneously with pellet collection and assessment of the breeding of owls over six study plots and seven trapping intervals in one oil palm plantation. Trapping records suggested that relative abundances of rodent species differed in plots with palms of differing ages. There were more R. r. diardii as a proportion of the total captures in older palms (seven years old) while R. argentiventer was the most common rat captured in younger palms (three years old). The abundance of R. tiomanicus remained low throughout trapping sessions for palms of all ages. A numerical response of rats to fruit availability in older palms was demonstrated by a positive and significant correlation between the numbers of fruit bunches present and the total number of rats captured. This relationship was observed in both older and younger palms only for R. argentiventer but not R. r. diardii or R. tiomanicus in either age of palms. This suggests that the competitiveness of R. argentiventer may be higher than that of the other two species providing that there were no factors other than food availability that limit the numbers of other two species. However, the levels of damage to palm fruit were significantly correlated with the relative abundance of R. argentiventer only in younger palms, not in older ones where R. r. diardii were more abundant. The more terrestrial nature of R. argentiventer may have restricted their acquisition of food when the palms became taller. In addition to differences in feeding niche, habitat heterogeneity may also be important in determining the rat species compositions at different ages of palms. Vegetation cover may act as a refuge for rats; the overall occurrence of rats, and especially R. argentiventer, was positively correlated with vegetation cover and height, and even certain vegetation associations. This study supported the continued use of the barn owl Tyto alba javanica for biological control of rodent pests in the palm oil plantations. The numbers of different rat species consumed by owls, as measured by pellet numbers, were proportional to prey captures with R. r. diardii dominating the prey items. Weights and sexes of rats, based upon sizes of bones recovered from owl pellets, indicated that the birds did not preferentially prey on any size or sex classes of rats. Although there was no differential predation by the owls, a functional, and possibly a numerical, response of the birds to changes in rat numbers was demonstrated. A functional response of barn owls to prey abundance was evidenced by a significant positive relationship between the relative abundance of rats captured and numbers of pellets collected. Some form of numerical response of barn owls was suggested by higher breeding records when rat abundances were significantly higher. Since both functional and numerical responses are important determinants of whether predators are likely to be able to regulate prey numbers, the role of barn owls as a practical biological control agent in oil palm plantations was supported. This study suggested that the regulation of rodent pests in oil palm plantations should not be limited to chemical measures but can be complemented by other biological factors including interspecific interactions, manipulation of the availability and density of food and habitat structure, in addition to predation by barn owls. The findings suggested that an integrated approach to rodent control, considering all biological factors that influence rat numbers, should be properly applied if an environmentally friendly and possibly cost effective approach is to be applied for the palm oil industry.

Anestesia com óleo essencial de Lippia alba seguida de atordoamento elétrico ou hipotermia: impacto sobre indicadores de estresse e qualidade da carne de Jundiá (Rhamdia quelen) / Anesthesia with essential oil of Lippia alba followed by electrical stunning or hypothermia: impact on indicators of stress and meat quality of silver catfish (Rhamdia quelen)

Veit, Juliana Cristina 15 April 2016 (has links)
This study aimed to optimize the electrical stunning for silver catfish and to evaluate whether the anesthesia with essential oil (EO) of L. alba before electrical stunning or hypothermia, used in pre-slaughter stunning stage, would reduce stress and influence on stability and sensory characteristics of fish produtcts. The effect of electrical frequencies between 50 and 1000 Hz associated to 128 or 256 V m-1 electric field strength and duration of exposure to the current between 1 and 60 seconds were evaluated. The electrical frequencies of 100, 300 and 500 Hz, combined with electric field strength of 128 V m-1 during 5 seconds were the most appropriate electrical combinations for pre-slaughter stunning of silver catfish as they yielded longer apparent stunning, lower levels of glucose and lactate and smaller changes in flesh characteristics compared to the others. Anesthesia with EO (300 mg L-1) did not reduce the biochemical markers of stress in silver catfish. However, compared to non-anesthetized fish, anesthesia did delay the loss of freshness and deterioration of chilled fish as it delayed the degradation of ADP to AMP and the generation of inosine, and contributed to reduce the sensory demerit scores for overall and gills odor, belly firmness and whole TFRU scheme, especially when associated with the electrical stunning. Furthermore, non-anesthetized fish were unacceptable for consumption on the 23rd day of chilled storage, whereas anesthetized fish become unacceptable only on the 33rd day. The EO also showed antimicrobial activity against Enterobacteriaceae species, however, when used before hypothermia the EO anesthesia did not prevent the post mortem oxidative changes in the frozen fillets, and potentialized lipid oxidation of frozen fillets when associated with electrical stunning. Hypothermia stunning did modify post mortem changes of fish stored in ice, however fillet from fish submitted to hypothermia had greater oxidation of sulfhydryl groups and protein carbonylation with consequent reduction in lightness and increase in yellowness along frozen storage, which may be responsible for the greater total color difference and increased elasticity of fillets over 18 months of frozen storage compared to electrical stunning. In contrast, electrical stunning resulted in faster onset of rigor mortis than hypothermia but did not accelerate the resolution of rigor mortis or the degradation of ATP in whole fish stored in ice. In addition, electrical stunning favored the generation of hydroperoxides in fillets by the end of frozen storage, as well as more intense red color at the beginning of storage. Thus, electrical stunning with 128 V m-1 electric field strength combined with 300 Hz frequency during 5 seconds, can be used in the pre-slaughter stunning of silver catfish as it was less stressful for fish than hypothermia and did not have any negative effect on the post mortem stability of chilled fish or frozen fillets compared to hypothermia. In addition, prior anesthesia with 300 mg L-1 of EO of L. alba may also be used in this step of the fish processing, especially for whole fish stored in ice.

Functional Characterization of PtaRHE1, a gene that encodes a RING-H2 type protein in poplar / Caractérisation fonctionnelle de PtaRHE1, un gène qui code pour une protéine de type RING-H2 chez le peuplier

Mukoko Bopopi, Johnny 14 January 2011 (has links)
PtaRHE1 is a poplar (Populus tremula x P. alba) gene encoding a REALLY INTERESTING NEW GENE (RING) domain-containing protein. RING proteins are largely represented in plants and play important roles in the regulation of many developmental processes as well as in plant-environment interactions. In this thesis, we present a functional characterization of PtaRHE1. To gain further insight into the role of this gene, molecular and genetic alteration approaches were used. The results of in vitro ubiquitination assays indicate that PtaRHE1 protein is a functional E3 ligase and this activity was shown to be specific with the human UbCH5a, among the tested ubiquitin-conjugating enzymes. Histochemical GUS stainings showed that the PtaRHE1 promoter is induced by plant pathogens and by elicitors such as salicylic acid and cellulase and is also developmentally regulated. In silico predictions and the transient expression of PtaRHE1-GFP fusion protein in N. tabacum epidermal cells revealed that PtaRHE1 is localized both in the plasma membrane and in the nucleus. The localization of expression of PtaRHE1 in poplar stem by in situ hybridization indicated that PtaRHE1 transcripts are localized within the cambial zone mainly in ray cells, suggesting a role of this gene in vascular tissue development and/or functioning. The overexpression of PtaRHE1 in tobacco resulted in a pleiotropic phenotype characterized by a curling of leaves, the formation of necrotic lesions on leaf blades, growth retardation as well as a delay in flower transition. Plant genes expression responses to PtaRHE1 overexpression provided evidence for the up-regulation of defence and/or programmed cell death (PCD) related genes. Moreover, genes coding for WRKY transcription factors as well as for MAPK, such as WIPK, were also found to be induced in the transgenic lines as compared to the wild type (WT). Taken together, our results suggest that the E3 ligase PtaRHE1 plays a role in the signal transduction pathways leading to defence responses against biotic and abiotic stresses. Identification of PtaRHE1 target(s) is required in order to fully assess the role of this E3 ligase in the ubiquitination-mediated regulation of defence response./<p>RÉSUMÉ<p><p><p>PtaRHE1 est un gène qui code pour une protéine possédant un domaine RING (REALLY INTERESTING NEW GENE) chez le peuplier (Populus tremula x P. alba).

Blotta, Rosa Maria January 2011 (has links)
As amostras foram submetidas a análise imuno-histoquímica utilizando anticorpos policlonais anticolágeno tipos I e III. / Differences observed in the midline of the abdominal wall in patients undergoing abdominoplasty and evidence from a number of studies showing the importance of collagen to aponeurotic structures and tensile strength have raised interest in investigating the rates of type I and type III collagen in this anatomic region. The aim of this study was to assess the amount of type I and type III collagen in the linea alba of women with and without diastasis recti and to determine collagen type I/III ratio by comparing these two groups. This is a case-control study nested within a surgical cohort of 18women with diastasis recti and a control group with 18 women without diastasis recti. Samples were collected from de midline of the abdominal wall three centimeters above and two centimeters bellow the umbilical scar. The samples were analyzed by immunohistochemistry using polyclonal antibodies to collagen type I and type III. The amount of collagen type I and type III was smaller in women with diastasis recti than in those without this condition in samples collected from the linea alba above and bellow the umbilical scar (P<0,001). Collagen type I/III ratio was lower in womem with diastasis recti in the samples collected above de umbilical scar (P<0,001). However, there was no statistically significant difference between groups with and without distasis recti in the samples collected bellow de umbilical scar (P = 0,110). The lower amount of collagen type I and III observed in the midline of the abdominal wall could be a factor in diastasis recti.

Made available in DSpace on 2015-12-06T23:46:09Z (GMT). Conclusao: A sutura triangular produziu encurtamento do eixo vertical do plano musculo-aponeurotico da parede abdominal em longo prazo / Background: Diastasis recti secondary to pregnancy is multidirectional. Plication of the anterior rectus sheath is the most widely used technique for correction of this condition. However, it would be desirable to simultaneously perform the transverse and longitudinal repair of the defect. The aim of this study was to assess changes in the length of the musculoaponeurotic layer after diastasis recti repair using triangular sutures. Methods: Thirty-one women with Nahas’ type III/A deformity were divided into two groups: the triangular suture (TS) group and the continuous suture (CS) group. All patients underwent conventional abdominoplasty and diastasis recti repair with medial longitudinal plication performed between two metal clips. The two types of suture were used in both groups. In the TS group, after a continuous suture was performed and removed, triangular sutures were used and maintained in place. In the CS group, the order of suture placement was reversed. The distance between clips was measured before and immediately after suturing, and at 3 weeks and 6 months postoperatively using plain abdominal radiographs. Statistical analysis was conducted with the Friedman analysis of variance and Wilcoxon test. Results: The use of triangular sutures significantly reduced the length of the aponeurosis compared with both the intraoperative Abstract | 87 situation without suture (P < 0.001) and the use of continuous suture (intraoperatively and 6 months after surgery; P < 0.001). Conclusion: The repair of diastasis recti using triangular sutures resulted in vertical shortening of musculoaponeurotic layer immediately after the procedure and in the long term. / BV UNIFESP: Teses e dissertações
