A reading enrichment program for gifted adolescent / Programa de enriquecimiento en lectura para adolescentes talentosos

This descriptive research examines the performance on reading comprehension of four gifted adolescents participating in a reading enrichment program. This performance is related to the variables associated to the gifted performance following the model developed by Mönks: motivation, creativity, high ability, school, peers and family. Results showed that school does not satisfy the cognitive needs of these students and it does not offer the required social support either. The family provides the main social support. The motivation, creativity, the high level of language and concentration of the participants facilitate learning. The reading program is described by the participants as motivating due to the subject-matters and methodology; furthermore, due to the opportunity to deal with peers who have a similar cognitive level. / Esta investigación de nivel descriptivo examina el desempeño en comprensión de lectura de cuatro adolescentes talentosos durante un programa de enriquecimiento en lectura. Este desempeño es relacionado con las variables asociadas a la conducta talentosa, de acuerdo al modelo de Mönks: motivación, creatividad, alta capacidad, escuela, pares y familia. Se encontró que la escuela no satisface las demandas cognitivas ni brinda el soporte social esperado. La familia brinda el soporte social principal. La motivación, la creatividad, el alto nivel de lenguaje y de concentración de los participantes facilitan el aprendizaje. El programa de lectura es descrito por los participantes como motivador debido a los temas y la metodología; además, por la oportunidad del intercambio con pares del mismo nivel cognitivo.

Les situations péri-contractuelles se caractérisent par le fait qu'une tierce personne a réalisé des prestations au profit d'une personne publique en l'absence de contrat valide. Le prestataire ne peut réclamer le paiement du service rendu à la collectivité en invoquant les obligations résultant de l'accord qu'il a passé avec l'Administration. Pour sa part, la personne publique ne bénéficie d'aucune garantie contractuelle, ou post-contractuelle, au titre des malfaçons affectant l'ouvrage livré. Afin de garantir les divers intérêts en présence, le juge administratif a progressivement élaboré une solution juridique équitable, qui permet de régler le déséquilibre entre les situations financières des parties, en combinant les fondements de responsabilité, soit quasi-contractuelle, soit quasi-délictuelle. L'indemnité d'enrichissement sans cause permettra ainsi de couvrir l'ensemble des dépenses que le prestataire a utilement exposées au profit de la personne publique et une indemnisation complémentaire pourra lui être accordée afin de compenser le manque à gagner qu'il a subi du fait de l'inexistence d'un contrat. De plus les dommages résultant pour la personne publique, de la livraison d'un ouvrage non conforme à sa destination, pour avoir été construit en méconnaissance des règles de l'art, ouvrent droit à réparation sur le fondement de la responsabilité quasi-délictuelle des entreprises / The situations peri-contract are characterized by the fact that a third person realizes services for the benefit of a public person in the absence of valid contract. The person receiving benefits cannot demand the payment of the service provided in the community by calling upon the obligations resulting from an agreement spent with the Administration. The public person does'nt benefit from any contractual guarantee, or post-contractual, in conformance for the faults affecting the delivered work. To guarantee the diverse interests in presence, the administrative judge gradually developes a legal solution which enables to adjust the imbalance between the financial situations of the parts by combining the foundations of quasi-contractual or quasi-delictual responsibility. The compensation of enrichment without cause will in that way allows to cover the total expense that the person receiving benefits usefully exposed for the benefit of the public person and a complementary compensation can be granted to him to compensate for the loss of income which he suffered because of the absence of contract. More the damage resulting for the public person, of the delivery of a work not in compliance with its destination, to have been built in misunderstanding of the rules of the art, opens straight ahead to repair on the foundation of the quasi-delictual responsibility of companies

Método dos elementos finitos generalizados para análise de estruturas em cascas de revolução / Generalized finite element method to analysis of structures in revolution shell

O presente trabalho está inserido no campo de estudo das cascas axissimétricas, tendo como objetivo a análise de seu comportamento estrutural mediante o desenvolvimento e aplicação de uma ferramenta numérica baseada no método dos elementos finitos generalizados. A utilização desse recurso é uma alternativa eficaz e difere do método dos elementos finitos convencional pela possibilidade de enriquecimento nodal das funções de aproximação. Como resultado pode-se dispensar o uso de redes muito refinadas. Com o intuito de evidenciar as vantagens do método adotado são apresentados exemplos comparando-se as soluções numéricas obtidas com soluções analíticas ou numéricas geradas com o do método dos elementos finitos convencional. Os resultados obtidos com um pequeno número de elementos finitos e com enriquecimento por funções polinomiais, mostraram-se convergentes já nos primeiros graus de enriquecimento. Desenvolve-se uma análise complementar de convergência baseada em estimativa de erro, mostrando que a metodologia adotada pode proporcionar melhores taxas de convergência em relação ao refino h quando predomina a regularidade da solução. A mesma análise aponta que a combinação dos refinos h e p pode levar a resultados mais precisos, com elevadas taxas de convergência, quando a solução (particularmente suas derivadas) apresentar regularidade menor. / The present dissertation is inserted in the field of study of the axisymmetric shells. The objective is to analyze the structural behavior by means of the development and application of a numerical tool based on the generalized finite element method. The use of this resource is an efficient alternative to the conventional finite element method for the possibility of nodal enrichment of the approach functions. Therefore one can avoid the use of very fine nets. In addition, in order to evidence the advantages of the adopted method, there are shown examples comparing the numerical solutions with analytical or numerical values generated with the conventional finite element method. The results obtained with a small number of elements, including enrichment by polynomial functions, had revealed convergence in the first degrees of enrichment. It is developed a complementary convergence analysis based on estimate of error, showing that the adopted methodology can provide better convergence ratios in relation to the h-refinement, in the cases where the regularity of the solution predominates. The same analysis shows that the combination of the refinement in its versions h and p can give more accurate results, by increasing convergence, when the solution (particularly its derivatives) presents lower regularity.

O presente trabalho trata da aplicação do Método de Resíduos Ponderados, mais especificamente do Método dos Mínimos Quadrados, na obtenção de soluções aproximadas de problemas de estruturas em casca, em especial os reservatórios cilíndricos submetidos a carregamento hidrostático em regime de comportamento linear. Os meios empregados para a obtenção das soluções aproximadas referem-se à adoção de bases aproximativas lineares, polinomiais, além da possibilidade de enriquecimento da aproximação mediante a adição de funções com características similares à própria solução exata. Uma outra alternativa utilizada refere-se à aplicação do Método dos Mínimos Quadrados com divisão do domínio de integração. Tais procedimentos podem ser úteis na análise de estruturas por evitar, de modo significativo, a elevação do esforço computacional, mediante a utilização de uma base aproximativa que corresponda às características requeridas pela solução analítica do problema. / The present dissertation deals with the application of the Weighed Residual Method to analysis of cilindrical shells, more specifically of the Least Squared Method, in the attainment of approach solutions of shells structural problems, in special the cylindrical reservoirs submitted the hydrostatic shipment in regimen of linear behavior. The half employees for obtention of the approach solutions refer to adotion of linear, polynomial approaches bases, beyond the possibility of enrichment of the approach by means of the addition of functions with similar characteristics to the proper accurate solution.One another used alternative mentions the application to Least Squared Method with division of the integration domain. Such procedures can be useful in the analysis of structures for preventing, in significant way, the rise of the computational effort, by means of the use of a aproximativa base that correspond to the characteristics required for the analytical solution of the problem.

Exercício físico e estimulação ambiental como estratégias terapêuticas após a hipoperfusão cerebral crônica em ratos Wistar : aspectos neurais, gliais e comportamentais

O objetivo geral deste estudo foi verificar os efeitos terapêuticos do enriquecimento ambiental e do exercício físico forçado sobre as conseqüências funcionais e cognitivas da Hipoperfusão Encefálica Crônica (HEC) em ratos Wistar. Além disso, padronizamos protocolo alternativo de HEC, a fim de aumentar a taxa de sobrevivência característica deste modelo, juntamente com a análise comportamental e imunohistoquímica do sétimo ao sexto mês após a cirugia, analisando o comportamento de neurônios e astrócitos no hipocampo. Os resultados mostraram que o protocolo modificado, ou seja, com a oclusão das carótidas realizadas com uma semana de intervalo, apresentam resultados comportamentais e morfológicos similares aos do protocolo padrão; todavia, a taxa de sobrevida dos animais no protocolo modificado é significativamente maior quando comparados ao padrão. Além disso, em ambos os protocolos, os animais hipoperfundidos apresentam um déficit cognitivo no Teste Water Maze três meses após a cirurgia, mas os grupos isquêmicos não apresentaram diferenças morfológicas quando comparados ao seus controles em relação a volume total Hipocampal e área Estriatal. Ao analisar o tempo pós- lesão do déficit cognitivo, verificamos que a HEC causa danos comportamentais a longo prazo, chegando a seis meses após o evento isquêmico. Enquanto a cognição é afetada na fase crônica, as células neuronais e astrogliais são acometidas nas fases mais agudas deste modelo, começando a aparecer morte neuronal (através da quantificação de NeuN) e astrogliose reativa (através da quantificação do imunoconteúdo de GFAP) já nos primeiros dias, permanecendo até aproximadamente 3 meses após a lesão. Após o dano estabelecido, testamos estratégias terapêuticas com o objetivo de amenizar principalmente o dano cognitivo apresentado pelos animais a partir dos três meses após cirurgia. Inicialmente foi analisado o efeito pré e pós - isquemia do exercício físico forçado através de uma esteira para ratos na memória espacial e de trabalho, através do Water Maze e bioquimicamente no estado oxidativo cerebral. Posteriormente, os mesmos parâmetros foram testados, só que com a intervenção do enriquecimnto ambiental tanto pré quanto pós-isquemia também. O modelo de hipoperfusão cerebral crônica causa como citado anteriormente, um déficit bastante importante na memória e no apredizado. Somado a isso, em relação ao estresse oxidativo, a quantidade de radicais livres e o estado de lipoperoxidação celular encontra-se bastante aumentados no hipocampo de animais submetidos a isquemia, assim como as enzimas antioxidantes. O protocolo de atividade física regular forçada (3 vezes por semana, durante 20 minutos num período de 3 meses), sobretudo nos grupos pré e pré+pós-isquemia, preveniu os efeitos cognitivos e bioquímicos da HEC. Tal reversão também ocorreu com o uso do enriquecimento ambiental realizado 3 vezes por semana, 1 hora durante 3 meses em todos os grupos estimulados. / The aim of this study was to verify the experimental therapeutic effects of environmental enrichment and forced physical exercise on the consequences of cognitive and functional chronic cerebral hypoperfusion (CCH) in Wistar rats. To do that, a CCH alternative protocol was standardized, in order to increase the survival rate of this model, together with the behavioral and immunohistochemistry analysis done up to the sixth month after the surgery, analyzing hippocampus neurons and astrocytes. Results showed that the modified protocol, with occlusion of both carotid arteries performed with a one-week interval, presents morphological and behavioral results similar to the standard protocol; however, the rate of survival after the modified protocol was significantly greater when compared to the standard one. In addition, both protocols produced ischemic animals with cognitive deficits in the Water Maze Task, three months after the surgery, however with no gross morphological lesion, as assessed by hippocampal volume and estriatal area. The cognitive deficit in CCH rats is long lasting, reaching six months after the ischemic event. While the cognition is affected in the chronic phase of hypoperfusion, the neuronal and astrogliais cells are affected in acute phases of this model; neuronal death (through quantification of NeuN) and reactive astrogliosis (through quantification of imunocontent of GFAP) are already present in the first days and remain until approximately 3 months after the lesion. We tested therapeutic strategies with the aim of alleviating mainly the cognitive damage presented by animals, as from three months after surgery. Initially, the effects of preand post-ischemia forced physical exercise in spatial and working memory, through the Water Maze Tak and oxidative stress parameters were analyzed. Subsequently, we tested the effects of environmental enrichment, both pre- and post-ischemia. The model of chronic cerebral hypoperfusion as previously mentioned, causes an important deficit in spatial learning and memory; the free radicals content and cellullar lipid peroxidation is also substantially increased in the hippocampus of animals submitted to hypoperfusion, as well as the antioxidant enzymes. Forced regular physical activity protocol (20-min, 3 times per week during 12 weeks – moderate), especially in groups pre and pre+post-ischemia, warned cognitive and biochemical preventive effects in CCH rats; the same occurred with the use of enrichment environmental performed 1- hr, 3 times per week during 12 weeks in all groups stimulated.

The environmental impact caused by oil, sewage, industrial, mining, leaching of soil and urban surfaces in the Brazilian coast has generated effluents mixed, with the metal trace a specific problem because they are deposited in marine sediments and present toxicity at the same time, persistence and bioaccumulation. Sediments act as operating compartment corded an essential role in the redistribution of marine biota, because that guard the historical record of contamination of water body. Oil exploration extends to offshore environment, occupying a prominent position in the energy matrix, responsible for most of the national supply. The oil and gas activities may include a significant source of pollution of the planet. The aim of this study was to observe the sediment profiles of the continental shelf off Sergipe and Alagoas southern state, checking for a variation of the concentration of trace metals according to depth. Three cores (PCM-9 (A), PCM-9 (B), Est.2 (A)) were performed to obtain a vertical profile of sediment samples obtained at various depths. In this study, drew up the metals Co, Cr, Fe, Li, Mn, Ni, Pb, Zn, Al and Cu total and partial, were determined by atomic absorption spectrometric (AAS). In the extraction part, the result of the recovery was above 90%, ranging from 92% to 111.1%. In the extraction result of the total recovery ranged from 77.6 to 99%. These values were obtained in the studied parameters, indicating good accuracy of the methodology. The values of RSD are below 10%, expressing the accuracy of methodology. Showed that the concentration distribution of trace metals in sediment profiles vary with depth. Colors in PCM-9 (A), PCM-9 (B), increased concentrations of metals and the core Est.2 (A) decreased gradually from base to surface. The iron was approved as a geochemical normalizer better. The values of the calculated enrichment factor shown to be low, considering the low enriched, except Pb, Co, Mn and Al that have moderate values. The geochemical background values were estimated in three different procedures, which can generate unrealistic characterizations of the state of pollution. The values of trace metals are below the sediment quality guidelines, which leads to the adverse effect on aquatic biota is unlikely. / O impacto ambiental causado por óleo, esgotamento sanitário, industrial,mineração, lixiviação de solos e superfícies urbanas, na costa brasileira temgerado efluentes variados, sendo os metais traço um problema particular,pois se depositam nos sedimentos marinho e apresentam ao mesmo tempo toxicidade, persistência e bioacumulação. Os sedimentos funcionam comocompartimento operacional que exerce papel essencial na redistribuição àbiota marinha, visto que guardam registro histórico da contaminação decorpo d água. A exploração de petróleo se estende ao ambiente offshore, ocupando uma posição de destaque na matriz energética, responsável pela maior parte do suprimento nacional. As atividades petrolíferas podem consistir uma fonte poluidora significativa do planeta. O objetivo desse estudo foi observar os perfis sedimentares da plataforma continental de Sergipe e sul de estado de Alagoas, verificando se há uma variação da concentração dos metais traço de acordo com a profundidade. Três cores (PCM-9(A), PCM-9(B), Est.2(A)) foram realizadas para obter um perfil vertical do sedimento, obtendo amostras a várias profundidades. Nesse estudo, extraiu-se os metais Co, Cr, Fe, Li, Mn, Ni, Pb, Zn, Al e Cu totais e parciais e foram determinados através da espectrometria de absorção atômica. Na extração parcial, o resultado da recuperação foi acima de 90 %, variam de 92% a 111,1%. Já na extração total o resultado da recuperação variou de 77,6 a 99%. Esses valores foram obtidos nos elementos estudados, indicando boa exatidão da metodologia. Os valores do desvio padrão relativo estão abaixo de 10%, expressando a precisão da metodologia. Mostrou-se que a distribuição da concentração dos metais traço nos perfis sedimentares variam com a profundidade. Nos cores PCM- 9(A), PCM-9 (B), as concentrações dos metais aumentaram e no core Est.2 (A) diminuiu gradativamente da base até a superfície. O ferro foi aprovado como melhor normalizador geoquímico. Os valores do fator de enriquecimento calculado mostraram-se baixos, considerando-se pouco enriquecida, exceto o Pb, Co, Mn e Al que apresentam valores moderados. Os valores de background geoquímico estimado foram diferentes nos três procedimentos utilizados. Os valores dos metais traços estão abaixo dos valores guia de qualidade dos sedimentos, o que leva o efeito adverso a biota aquática ser pouco provável.

Desenvolvimento de metodologias utilizadas nas áreas de salvaguardas e forense nuclear baseadas na técnica LA-HR-ICP-MS / Development of methodologies used in Nuclear Safeguards and Nuclear Forensic based on LA-HR-ICP-MS technique

A amostragem ambiental por esfregaço ou swipe samples é uma metodologia que vem sendo empregada rotineiramente pela Agência Internacional de Energia Atômica (AIEA) para verificar se os Estados signatários do Acordo de Salvaguardas estão realizando atividades nucleares não declaradas. A análise dessas amostras ambientais é complementar aos procedimentos de salvaguardas convencionais para a verificação das informações cedidas pelos Estados. Neste trabalho, foi descrita uma metodologia que visa a fortalecer os processos de salvaguarda nuclear e da ciência forense nuclear no país. A proposta é estudar e avaliar a técnica de ablação à laser acoplada a um espectrômetro de massa de alta resolução com fonte de plasma indutivamente acoplado (LA-HR-ICP-MS Laser Ablation High Resolution Inductively Coupled Plasma Mass Spectrometry), como técnica alternativa para a análise das amostras de esfregaço. Adicionalmente, foram empregadas outras técnicas complementares (atividade radioativa, microscopia eletrônica de varredura e espectroscopia de energia dispersiva) com o intuito de garantir maior segurança ao processo de inspeção de salvaguardas e investigações forenses nucleares. A precisão, medida através do desvio padrão relativo (DPR),dos resultados obtidos para as razões isotópicas 234U/238U, 235U/238U e 236U/238U do padrão analisado (CRM -125A) para a medida do fator de discriminação de massa foram, respectivamente, 1,3 %, 0,2 % e 7,6 %. Já as incertezas percentuais (u %), que também contemplam o DPR das medições, variaram de 3,5 % a 29,8 % para as medições da razão isotópica 235U/238U e de 16,6 % a 42,9 % para a razão isotópica 234U/238U. Esses valores mostraram-se compatíveis com outros estudos que utilizaram a mesma técnica para análise de amostras reais coletadas em uma instalação nuclear. As amostras coletadas apresentaram nível de enriquecimento que variou de (2,3 ± 0,7) % (amostra 3 corredor de acesso à instalação) a (17,3 ± 2,8) % (amostra 18 bancada de recuperação de UF4). A partir da coleta de amostras nesses diversos pontos, desde a entrada da instalação até a bancada em que se manuseia o urânio enriquecido, foi possível detectar diversos níveis de enriquecimento no isótopo 235U. / Environmental sampling performed by means of swipe samples is a methodology frequently employed by International Atomic Energy Agency (IAEA) to verify if the signatory States of the Safeguards Agreements are conducing unauthorized activities. Swipe samples analysis is complementary to the Safeguards ordinary procedures used to verify the information given by the States. In this work it was described a methodology intending to strengthen the nuclear safeguards and nuclear forensics procedures. The proposal is to study and evaluate the laser ablation high resolution inductively coupled plasma mass spectrometry (LA-HR-ICP-MS) technique as an alternative to analyze the real-life swipe samples. The precision achieved through the standard(CRM -125A)measurements, represented by the relative standard deviation (RSD), was respectively 1.3 %, 0.2 % e 7.6 %for the 234U/238U, 235U/238U e 236U/238Uisotopes ratios. The percent uncertainties (u %), which covers the RSD, ranged from 3.5 % to 29.8 % to the 235U/238U measurements and from 16.6 % to 42.9 % to the 234U/238U isotope ratio. These results were compatible with former studies performed by the LA-HR-ICP-MS that analyzed real-life swipe samples collected at a nuclear facility. Swipe samples collected from several points of the nuclear facility presented enrichment level ranging from (2.3 ± 0.7) % (sample 3) to (17.3 ± 2.8) % (sample18). They also allowed detecting different enrichment levels within the facility.

The Effects of a Home-Based, Audio Cassette Marriage Enrichment Course on Marital Communication and Marital Adjustment

This study investigated the effects of a home-based, audio cassette marriage enrichment course on marital communication and marital adjustment. The marriage enrichment course evaluated in this study consisted of two audio cassette tapes, each containing two sessions of approximately 45 minutes in length, and one work booklet. The course contained exercises emphasizing the development of communication skills, encouragement of self-disclosure, learning of empathy skills, and the setting of personal and mutual goals. The unique aspects of the course were the home-based setting in which the couples completed the program, and the self-enclosed audio cassette nature of the course.

Enrichissement d'une communauté microbienne anaérobie oxydante du méthane à partir de sédiments marins : évaluation des performances en bioréacteurs / Performance assessment and enrichment of anaerobic methane oxidizing microbial communities from marine sediments in bioreactors

L'oxydation anaérobie du méthane (AOM) couplé à la réduction du sulfate (AOM-SR) est un processus biologique médié par méthanotrophes anaérobie (ANME) et de bactéries sulfato-réductrices. La communauté scientifique s'inquiète de AOM, en raison de sa pertinence dans la régulation du cycle global du carbone et de la potentielle application biotechnologique pour le traitement de sulfate riches eaux usées.Pour améliorer les connaissances récentes sur les conditions de distribution et d'enrichissement ANME, cette recherche a étudié AOM-SR avec les objectifs suivants: (i) caractériser les communautés microbiennes responsables de AOM dans les sédiments marins, (ii) de les enrichir dans les bioréacteurs avec différentes configurations, à savoir bioréacteur à membrane (MBR), filtre biotrickling (BTF) et bioréacteur à haute pression (HPB), et (iii) d'évaluer l'activité de l'ANME et le processus AOM dans différentes conditions de pression et de température.Les microbes habitant peu profonde dans les sédiments de Marine lac Grevelingen (Pays-Bas) ont été caractérisés et leur capacité de faire AOM-SR a été évaluée. Un test d'activité a été réalisée en discontinu pour 250 jours, AOM-SR est mise en évidence par la production de sulfure et de la prise concomitante de sulfate et de méthane dans des rapports équimolaires et il a été atteint 5 µmoles par gramme de poids par jour de taux de réduction du sulfate. L'analyse des séquences de gènes 16SrRNA a montré la présence de méthanotrophes anaérobie ANME-3 dans les sédiments marins du lac Grevelingen.Deux configurations de bioréacteurs, à savoir MBR et BTF ont été opérés dans des conditions ambiantes pendant 726 jours et 380 jours, respectivement, pour enrichir les micro-organismes de Ginsburg Mud Volcano performantes AOM. Les réacteurs sont exploités en mode fed-batch pour la phase liquide avec un apport continu de méthane. Dans le MBR, une membrane d'ultrafiltration externe a été utilisée pour retenir la biomasse, alors que, dans la BTF, la rétention de biomasse a été accomplie par la fixation de la biomasse sur le matériau d'emballage. AOM-SR a été enregistrée seulement après ~ 200 jours dans les deux configurations de bioréacteurs. L'opération du BTF a montré l'enrichissement de l'ANME dans le biofilm par la méthode Illumina Miseq, en particulier ANME-1 (40%) et ANME-2 (10%). Dans le MBR, les agrégats d'ANME-2 et Desulfosarcina ont été visualisées par CARD-FISH. La production d'acétate a été observée dans le MBR, ce qui indique que l'acétate était un possible intermédiaire d'AOM. Bien que les deux configurations de bioréacteurs ont montré de bonnes performances, le taux de réduction du sulfate était légèrement plus élevée et plus rapide dans la BTF (1,3 mM par jour âpres 280 jours) que le MBR (0,5 mM par jour jour âpres 380 jours).Afin de simuler les conditions de suintement froid et de différencier l'impact des conditions environnementales sur AOM, les sédiments fortement enrichi avec le clade ANME-2a ont été incubées dans HPB à différentes températures (4, 15 et 25 °C à 100 bars) et pressions (20, 100, 200 et 300 bar à 15 °C). L'incubation à une pression de 100 bar et 15 ° C a été observé comme la condition la plus appropriée pour la phylotype ANME-2a, qui est similaire aux conditions in situ (Capitaine Aryutinov Mud Volcano, Golfe de Cadix). L'incubation de ce sédiment aux conditions in situ pourrait être une option privilégiée pour obtenir une activité AOM-SR plus élevée.Dans cette thèse, il a été démontré expérimentalement que la rétention de la biomasse et l'approvisionnement continu de méthane peuvent favoriser la croissance de la lente communauté microbienne qui oxyde le méthane en anaérobiose dans des bioréacteurs, même dans des conditions ambiantes. Par conséquent, la localisation des habitats de ANME dans des environnements peu profonds et l'enrichissant dans des conditions ambiantes peut être avantageuse pour les futures applications de la biotechnologie environnementale / Anaerobic oxidation of methane (AOM) coupled to sulfate reduction (AOM-SR) is a biological process mediated by anaerobic methanotrophs (ANME) and sulfate reducing bacteria. Due to its relevance in regulating the global carbon cycle and potential biotechnological application for treating sulfate-rich wastewater, AOM-SR has drawn attention from the scientific community. However, the detailed knowledge on ANME community, its physiology and metabolic pathway are scarcely available, presumably due to the lack of either pure cultures or the difficulty to enrich the biomass. To enhance the recent knowledge on ANME distribution and enrichment conditions, this research investigated AOM-SR with the following objectives: (i) characterize the microbial communities responsible for AOM in marine sediment, (ii) enrich ANME in different bioreactor configurations, i.e. membrane bioreactor (MBR), biotrickling filter (BTF) and high pressure bioreactor (HPB), and (iii) assess the AOM-SR activity under different pressure and temperature conditions.The microbes inhabiting coastal sediments from Marine Lake Grevelingen (the Netherlands) was characterized and the ability of the microorganisms to carry out AOM-SR was assessed. By performing batch activity tests for over 250 days, AOM-SR was evidenced by sulfide production and the concomitant consumption of sulfate and methane at approximately equimolar ratios and a sulfate reduction rate of 5 µmol sulfate per gram dry weight per day was attained. Sequence analysis of 16S rRNA genes showed the presence of ANME-3 in the Marine Lake Grevelingen sediment.Two bioreactor configurations, i.e. MBR and BTF were operated under ambient conditions for 726 days and 380 days, respectively, to enrich the microorganisms from Ginsburg Mud Volcano performing AOM. The reactors were operated in fed-batch mode for the liquid phase with a continuous supply of gaseous methane. In the MBR, an external ultra-filtration membrane was used to retain the biomass, whereas, in the BTF, biomass retention was achieved via biomass attachment to the packing material. AOM-SR was recorded only after ~ 200 days in both bioreactor configurations. The BTF operation showed the enrichment of ANME in the biofilm by Illumina Miseq method, especially ANME-1 (40%) and ANME-2 (10%). Interestingly, in the MBR, aggregates of ANME-2 and Desulfosarcina were visualized by CARD-FISH. Acetate production was observed in the MBR, indicating that acetate was a possible intermediate of AOM. Although both bioreactor configurations showed good performance and resilience capacities for AOM enrichment, the sulfate reduction rate was slightly higher and faster in the BTF (1.3 mM day-1 at day 280) than the MBR (0.5 mM day-1 at day 380).In order to simulate cold seep conditions and differentiate the impact of environmental conditions on AOM activities, sediment highly enriched with the ANME-2a clade was incubated in HPB at different temperature (4, 15 and 25 oC at 100 bar) and pressure (20, 100, 200 and 300 bar at 15 oC) conditions. The incubation at 100 bar pressure and 15 oC was observed to be the most suitable condition for the ANME-2a phylotype, which is similar to in-situ conditions where the biomass was sampled, i.e. Captain Aryutinov Mud Volcano, Gulf of Cadiz. The incubations at 200 and 300 bar pressures showed the depletion in activities after 30 days of incubation. Incubation of AOM hosting sediment at in-situ condition could be a preferred option for achieving high AOM activities and sulfate reduction rates.In this thesis, it has been experimentally demonstrated that biomass retention and the continuous supply of methane can favor the growth of the slow growing anaerobic methane oxidizing community in bioreactors even under ambient conditions. Therefore, locating ANME habitats in shallow environments and enriching them at ambient conditions can be advantageous for future environmental biotechnology applications

Etude du potentiel de valorisation énergétique ou matière de composés organiques extraits de lixiviats de déchets ménagers / Energetic valorization and soil enrichment with organic compounds extracted from a landfill leachate

Les contraintes techniques et économiques induites par la gestion des lixiviats de déchets ménagers et assimilés (LDMA) pourraient être minimisées en considérant le LDMA non plus comme un déchet mais comme un « co-produit » partiellement valorisable. Parmi la diversité des constituants des LDMA, la matière organique dissoute (MOD) présente 2 intérêts majeurs : une forte charge et des processus de transformations comparables à ceux de la matière organique naturelle (MON). Ce travail de thèse s’attache donc à extraire cette MOD des LDMA selon son caractère hydrophobe et à identifier des axes de valorisation. Seules les fractions de type hydrophobe (HPO*) et transphilique (TPH*), de par leurs caractéristiques et leur réactivité, ont été considérées dans cette étude et 2 voies de valorisation ont été testées : une valorisation énergétique et une valorisation matière par amendement sur un sol en déficit de carbone organique (Corg).La caractérisation des fractions de MOD anthropiques et leur comparaison à des fractions de MON a confirmé l’intérêt majeur de leur valorisation. En effet, elles sont (i) physiquement et chimiquement comparables à la MON, (ii) quantifiables à des concentrations jusqu’à plus de 1000 fois supérieures à la MON permettant ainsi un potentiel d’extraction conséquent en comparaison aux matrices environnementales telles que les eaux de surface, et (iii) plus facilement biodégradables que la MON, tout en ne présentant aucun caractère phytotoxique avéré.Un test BMP optimisé et adapté au LDMA a été développé via un plan d’expérience, puis utilisé pour mettre en évidence l’impact du caractère hydrophobe de la MOD du LDMA sur la digestion anaérobie, et notamment la production de méthane. Selon les résultats obtenus, les molécules organiques de type transphilique favorisent la production de biogaz. De plus, la fraction hydrophile, faiblement aromatique, diminue la teneur en méthane du biogaz, contrairement à la fraction hydrophobe.Enfin, l’amendement de fractions organiques extraites de LDMA présente un réel intérêt dans le cas d’un sol initialement pauvre en Corg. La fraction HPO* permet un apport de Corg plus important comparé à la fraction TPH*, tandis que les composés transphiliques favorisent l’activité biologique du sol.Ce travail de thèse a ainsi mis en évidence l’intérêt d’extraire les fractions hydrophobe (HPO*) et transphilique (TPH*) de la MOD de LDMA afin de les engager dans des processus de valorisation énergétique et agronomique. / The management of municipal solid waste leachates (MSWL) is technically and economically restrictive. It thus should be interesting to consider this effluent no more as a waste but rather as a partially valorizable “co-product”. The dissolved organic matter (DOM) is one of the major components of MSWL and presents a twofold interest: a high load and comparable processes of transformation to those of natural organic matter (NOM).This study thus aimed at extracting the DOM from MWSL according to its hydrophobic character and identifying ways of valorization. Only hydrophobic (HPO*) and transphilic (TPH*) fractions were considered because of their characteristics and reactivity. Two different ways of valorization were investigated: energetic valorization and enrichment of a soil with low content in organic carbon (Corg).The characterization of HPO* and TPH* fractions and their comparison with fractions of NOM confirmed their high valorizable potential. Indeed, they are physically and chemically comparable to NOM, they can be quantified at concentrations up to 1000 times higher than NOM (high extraction potential when compared to environmental matrices such as surface water), and they are more easily biodegradable than NOM without any phytotoxic character.An optimized BMP test was first developed by using an experimental design to adapt the experimental conditions to the digestion of MSWL. It was then applied in different conditions to highlight the impact of the hydrophobic character of the DOM of MSWL on the anaerobic digestion, and mainly on methane production. According to the results, the transphilic-like organic molecules improve the biogas production. Besides, the hydrophilic-like compounds with low aromatic character decrease the methane content of the produced biogas contrary to the hydrophobic fraction.Finally, organic fractions extracted from MSWL were relevant to enrich soil with low content in organic carbon. The HPO* fraction better improves the Corg content when compared to the TPH* fraction whereas transphilic-like molecules enhances the soil biological activity.With the results of this research study, the HPO* and TPH* fractions extracted from a MSWL can thus be considered both to produce biogas and particularly methane and to be amended on soils. Their extraction can thus be economically viable and their treatment no more highly costly.
