Atualmente, os filmes finos de óxido de estanho dopado com índio (ITO) têm encontrado grande aplicação no mercado. Entretanto, devido à pouca disponibilidade do índio na natureza e aos altos custos envolvidos na sua aquisição, elementos alternativos estão sendo estudados para sua substituição. Nesse contexto, destaca-se o óxido de zinco dopado com alumínio (AZO) como um promissor substituto, devido às características de elevada transmissividade, baixa resistividade e band gap da ordem de 3,37 eV, que permitem sua aplicação na síntese de filmes finos semicondutores. Com base no exposto, neste trabalho, foi projetado e desenvolvido o protótipo de uma fonte amplificadora de potência (FAP) de corrente alternada (AC) em baixa frequência, operando entre 15 a 40 kHz, responsável por iniciar e sustentar o campo elétrico utilizado para a geração do plasma. Esta FAP foi utilizada para a deposição de filmes finos de (AZO) por meio da técnica de magnetron sputtering. A análise das características morfológicas, ópticas e elétricas dos filmes de AZO produzidos neste estudo resultaram em uma transmitância superior a 80%, energia de band gap de 3,82 eV, e resistividade de 1,46.10-3 .cm, permitindo concluir que o filme produzido se comporta como um TCO (óxido transparente condutivo). A comparação desses resultados com trabalhos disponíveis na literatura, permite concluir que a fonte amplificadora de potência desenvolvida nesta dissertação possibilita a obtenção de filmes finos de AZO com condutividade e transparência superiores àqueles produzidos com fontes operando em radiofrequência, técnica atualmente disponível e amplamente utilizada no mercado. / The development of thin films deposition techniques allows to increase value and give new features to the materials. Currently, indium doped zinc oxide (ITO) is widely used in the market. However, due to the low availability of the indium in the nature and the high costs involved on its acquisition, alternative elements are being studied for its replacement. Aluminum doped zinc oxide (AZO) stands out as a promising substitute, mainly because of its characteristics, such as high transmissivity, low resistivity and band gap value of 3.37 eV. That allow the application of AZO in the synthesis of thin films semiconductors. In this work, it was developed a prototype of a plasma power source amplifier (FAP) to operate in alternating current (AC) and low frequency (15 - 40 kHz), responsible for initiating and sustaining the electric field used for plasma generation. This FAP was used to deposit AZO thin films by the technique of magnetron sputtering. The analysis of the morphological, optical and electrical characteristics of the AZO films produced in this study resulted in more than 80% transmittance, band gap energy value of 3,82eV, and resistivity of 1,46.10-3 .cm. The thin films synthetized was classified as transparent conductive oxide (TCO). The comparison of these results with the characteristics of similar films avaiable in the bibliography, allows to conclude that the power amplifier source developed in this dissertation makes it possible to obtain thin films of AZO with conductivity and transparency superior to those produced with RF magnetron sputtering, technique currently available and widely used in the market.

Para este fim, neste estudo explorou-se a seleção de uma substância capaz de marcar e ser biocida contra biofilmes fúngicos em superfícies de aço inox, a partir de nove candidatos benzimidazóis fluorescentes, com códigos NB1 a NB9 e oito sais imidazólicos de corantes azóicos, denominados C4MImErioCr, C10MImMO, C16MImMO, (C10)2MImMO, C4MImMO, C10MImORANGEII, C16MImORANGEII e (C10)2MImORANGEII. Desenvolveu-se para este fim um roteiro metodológico para determinar quais destas substancias são capazes de marcar e eliminar biofilmes de forma eficaz e segura. Os métodos utilizados para avaliar as substâncias foram (1) a Concentração Mínima Inibitória (MIC) conforme protocolo do CLSI M27-A3, (2) microscopias verificando capacidade das substâncias em marcar células, (3) ensaios com deposição sobre superfície do corpo de prova (placas de aço inox) com biofilme, (4) verificação da atividade biocida sobre biofilmes utilizando microscopias e (5) ensaios de citotoxicidade. Essas substâncias foram testadas frente a nove cepas de Candida spp., incluindo C. tropicalis, C. albicans e C. parapsilosis Na avaliação das substâncias, SI de corantes azóicos inibiram o crescimento celular de fungos, já o benzimidazol fluorescente NB7 apresentou atividades simultâneas de detecção e ação biocida sobre o biofilme. Todas as cepas testadas foram sensíveis a essa substância. Além disso, os biofilmes formados pelas cepas ATCC 18804 (C. albicans,) ATCC 22019 (C. parapsilosis) e ATCC 750 (C. tropicalis) na superfície de aço inox 304 sofreram ação biocida, quando expostas por 15 segundos a NB7, sendo um potencial sanitizante. / Biofilms provide an environment capable of protecting microbial cells from damage by chemical and physical factors of the immune system, and hinder the penetration of various antimicrobial agents, promoting nosocomial infections related to catheters, increasing mortality and morbidity of patients. Therefore, it is important to ensure proper hygiene to prevent contamination and infections in hospital environments. For this purpose, this study explored the identification of a substance that both detects and have biocide activity against fungal biofilms on stainless steel surfaces. Both nine fluorescent benzimidazole substances, coded NB1 to NB9 and eight imidazolium salts of azo dyes, named denominados C4MImErioCr, C10MImMO, C16MImMO, (C10)2MImMO, C4MImMO, C10MImORANGEII, C16MImORANGEII e (C10)2MImORANGEII were tested as candidates. These substances were tested applying a methodology developed to determine if a substance is able detecting and have biocide activity against fungal biofilms. Overall, this study involved the following methods: (1) Minimum Inhibitory concentration test following the CLSI protocol (M27-A3; the substances were tested against nine fungal strains, including C. tropicalis, C. albicans and C. parapsilosis.), (2) microscopy to determine the marker capacity, (3) spraying tests of the substances on surfaces (stainless steel) with fungal biofilms, (4) tests to verify the capability of the substances to both stain and were biocide against fungal biofilms, applying microscopic techniques and (5) cytotoxicity tests Within the set of seventeen substances, benzimidazole derivative NB7 was identified with the desired capabilities, staining and biocide activity against fungal biofilms at the same time. Les disques optiques numériques sont utilisés par de nombreux organismes, publics ou privés, pour l’archivage de données. Malgré les espoirs fondés dans l’utilisation de ces supports, des pertes d’information ont été observées, parfois après seulement quelques années d’archivage. Cette évolution a été attribuée au vieillissement des disques, sans préjuger des constituants impliqués. Ce travail de thèse visait à identifier les modifications chimiques et physiques résultant de l’application de différentes contraintes (photochimique, thermique ethydrolytique) sur des constituants de disques optiques : substrats de CD-R en polycarbonate, couches enregistrables de types « phtalocyanine » et « azoïque » et vernis de protection. L’étude de ce dernier constituant a permis d’élargir les recherches au cas du Blu-ray Disc. Ces évolutions physico-chimiques ont ensuite été corrélées à des variations des paramètres analogiques et numériques de disques soumis aux mêmes contraintes. L’objectif ultime de ce travail était de mettre en place une méthodologie générale permettant de comprendre l’origine de la perte d’information de disques optiques numériques exposés à des contraintes. / Optical discs are often used by many private or public organizations to archive essential data. For a long time these media were supposed to be reliable. However, a loss of the stored data was highlighted, sometimes only after a few years of storage. This degradation was attributed to the discs ageing, without any identification of the materials involved in this ageing. This work aimed to determine the chemical and physical modifications resulting from the application of different stresses (photochemical, thermal and hydrolytic) on the materials used in optical discs : the CD-R polycarbonate substrates, the recording layers based on phthalocyanines and azo compounds, and the protective layers. The study of this last component allowed us to extend the researches to the case of the Blu-ray Disc. These physico-chemical modifications were then correlated to variations in digital and analog parameters of discs submitted to the same stresses. The ultimate objective of this work was to propose a general method that could help to understand the origin of the loss of information of optical discs submitted to stresses.

Estudo das propriedades de armazenamento óptico e grades de relevo em poliuretano derivado de azo-benzeno / Information optically storage and relief gratings study in material derivative of azo-benzene polyrethane

Polímeros contendo grupos azo-benzênicos têm sido estudados devido a sua capacidade para óptica não linear, formação de superfícies de relevo e de birrefringência fotoinduzida. O fator que possibilita essas aplicações é a propriedade de fotoisomerização reversível trans-cis-trans do grupo azo que produz a sua reorientação molecular. Neste trabalho foram produzidos filmes casting do poliuretano MDI-DR19 derivado de azo-benzeno. Estudou-se o crescimento e decaimento da birrefringência para diferentes valores de potência da luz de excitação da fotoisomerização. Os resultados experimentais do crescimento e decaimento foram analisados usando duas exponenciais e eles mostraram que o polímero MDI-DR19 apresenta boas características para utilização em armazenamento óptico pois cerca de 65% da birrefringência induzida no material permanece por longos intervalos de tempo. Ilustramos o processo de armazenamento de informação gravando uma imagem de birrefringência num filme de MDI-DR19. Foram também produzidas grades de relevo em filmes de MDI-DR19 utilizando franjas de interferência produzidas com luz de polarização P e S. Essas grades apresentaram alta uniformidade e características compatíveis com as mostradas na literatura. / Polymers containing azobenzenic groups have been studied due to their applications on non-linear optics, surface relief gratings (SRG) and photoinduced anisotropy for optical storage. The reversal photoisomerization trans-cis-trans property of the azo group induces a molecular reorientation. In this work, cast films of na azobenzene derivative, the polyurethane MDI-DR19, were studied. The build-up and decay of the birefringence were studied as a function of the photoisomerization exciting light power. The data of the build-up and decay of the birefringence were fitted with a bi-exponential function, which shows the stability of the MDI-DR19 for application in optical storage, since 65% of the induced birefringence remains after a long time. The optical storage is illustrated by recording a birefringence image in a film of MDI-DR19. It was also performed a SRG over films of MDI-DR19 applying an interference finger pattern with a P and S polarized light. The light induced gratings shows good uniformity and characteristics compatible with those presented in the literature.

Removal of Congo red dye from aqueous solution using a clay based nanocomposite

MENVSC / Department of Ecology and Resource Management / In this study, the efficacy of bentonite clay, pre-treated magnesite and their nanocomposite on the removal of Congo red dye from aqueous solution was explored. Batch experimental approach was a technique used to fulfil the goals of this study. A number of operational parameters were optimised, and they include effects of shaking time, adsorbent dosage, initial CR dye concentration, initial solution pH and temperature. Findings of the study revealed that the optimum conditions that are suitable for the removal of CR dye are 20 minutes, 0.5 g of dosage, 120 mg/L, 250 rpm, and pH = 7. This has achieved > 99% removal efficacy of CR dye for the nanocomposite and reduced it to below the South African National Standard (SANS) 241 water quality specifications. Furthermore, kinetic studies revealed that bentonite clay, pre-treated magnesite, and their nanocomposite fitted very well to pseudo-second-order kinetics than pseudo-first-order kinetics. The regression analysis was observed to be 1, 0.9, and 0.9 for bentonite clay, pre-treated magnesite, and their nanocomposite respectively. Adsorption isotherms indicated that CR removal by bentonite clay, pre-treated magnesite, and their nanocomposite fitted well to Langmuir adsorption isotherm than the Freundlich adsorption isotherm hence indicating mono-layer adsorption. Thermodynamic values for CR removal were observed to be: ΔH0 (kJ mol-1) = 43.86, 30.67, and 24.88 for bentonite clay, pre-treated magnesite, and their nanocomposite respectively. This indicates that the reaction is endothermic. The positive ΔS0 (kJ mol-1 K-1) values for bentonite clay and 25 °C for pre-treated magnesite confirms that there is an increase in the degree of randomness at solid/solution interface during the removal of CR ions from aqueous solution. The negative values of ΔG0 (kJ mol-1) for 40 – 70 °C on bentonite and the entire range for the nanocomposite suggest the spontaneity and feasibility of CR adsorption whereas the positive ΔG0 (kJ mol-1) for bentonite clay suggest a non-spontaneous nature of adsorption. As such, pre-treated magnesite/bentonite clay nanocomposite demonstrated superior adsorption capacity in relation to individual materials and other materials reported in literature. / NRF

The degradation of wool keratin under conditions relevant to those of wool dyeing was investigated using the techniques of gel permeation chromatography (GPC), ion exchange gel chromatography, and amino acid analysis. Physical testing of the treated and untreated wool was also carried out to determine the physical changes occurring, parameters used being percentage elongation at the break, and the breaking strain of the fibre. Samples of wool keratin were immersed in various aqueous solutions at 1000C for 24 hours and the filtered, aqueous, oxidised extracts were analysed* The solutions used varied only in the dye, or dye intermediate present in the treatment solution. All treatment baths contained 10% owf 1.02 x 10 -2 MSulphuric VI acid; 10%owf 7.04x 10 -3 MSodium sulphate VI ; A 100 :1 liquor ratio was used in each case. Some of the dye intermediates showed a marked catalytic effect, particularly in their effect on breaking strain, a decrease of 40% in some cases. The GPC profiles of the extracted proteins were examined in detail and compared against previous workers' results. An explanation of the behaviour of the dyes and intermediates was proposed. The amino acid composition data of the extracted and fractionated proteins were compared against various morphological components extracted by other workers, as was the total gelatin obtained from each treatment. / Science Research Council

Decolorization of an azo and anthraquinone textile dye by a mixture of living and non-living Trametes versicolor fungus

Wastewater from the textile industry is difficult to treat effectively due to the prevalent use and wide variety of synthetic dyes that are resistant to conventional treatment methods. White-rot fungi, such as Trametes versicolor, have been found to be effective in decolorizing many of these synthetic dyes and current research is focusing on their application to wastewater treatment. Although numerous studies have been conducted on the ability of both living and nonliving Trametes versicolor to separately decolorize textile dyes, no studies were found to have investigated the use of a mixture of live and dead fungus for decolorization. This study explored potential interactions between live and dead, autoclaved Trametes versicolor biomass in a mixed system by utilizing a series of batch tests with two structurally different synthetic textile dyes. Samples were analyzed by spectrophotometer and compared with controls to determine the effect of any interactions on decolorization. The results of this study indicate that an interaction between living and nonliving biomass occurred that affected the specific dye removal for both Reactive Blue 19, an anthraquinone textile dye, and Reactive Orange 16, an azo textile dye. This interaction was seen to improve the specific dye removal during the first 10-46 hours of experimentation but then diminish the specific dye removal after this period. This effect could be due to hydrophobins, which are surface-active proteins excreted by live fungi that may alter hydrophobicity. Additionally, the presence of adsorptive dead biomass could affect dye contact with degrading enzymes released from the live fungus. By expanding current knowledge of the interactions that take place in a fungal bioreactor and their effect on textile dye decolorization, this research aims to inspire more effective and less costly bioreactor designs for the treatment of textile wastewater.

Synthesis of Catalytic Membrane Surface Composites for Remediating Azo Dyes in Solution

In the past 30 years zero-valent iron (ZVI) has become an increasingly popular reducing agent technology for remediating environmental contaminants prone to chemical degradation. Azo dyes and chlorinated organic compounds (COCs) are two classes of such contaminants, both of which include toxic compounds with known carcinogenic potential. ZVI has been successfully applied to the surfaces of permeable reactive barriers, as well as grown into nanoscale particles (nZVI) and applied in-situ to chemically reduce these contaminants into more environmentally benign compounds. However, the reactivity of ZVI and nZVI in these technologies is limited by their finite supply of electrons for facilitating chemical reduction, and the tendency of nZVI particles to homo-aggregate in solution and form colloids with reduced surface area to volume ratio, and thus reduced reactivity. The goal of this project was to combine reactive nanoparticle and membrane technologies to create an electro-catalytic permeable reactive barrier that overcomes the weaknesses of nZVI for the enhanced electrochemical filtration of azo dyes in solution. Specifically, nZVI was successfully grown and stabilized in a network of functionalized carbon nanotubes (CNTs) and deposited into an electrically conductive thin film on the surface of a polymeric microfiltration support membrane. Under a cathodic applied voltage, this thin film facilitated the direct reduction of the methyl orange (MO) azo dye in solution, and regenerated nZVI reactivity for enhanced electro-catalytic operation. The electro-catalytic performance of these nZVI-CNT membrane surface composites to remove MO was validated, modelled, and optimized in a batch system, as well as tested in a dead-end continuous flow cell system. In the batch experiments, systems with nZVI and a -2 V applied potential demonstrated synergistic enhancement of MO removal, which indicated the regeneration of nZVI reactivity and allowed for the complete removal of 0.25 mM MO batches within 2-3 hours. Partial least squares regression (PLSR) modelling was used to determine the impact of each experimental parameter in the batch system and provided the means for an optimization leading to maximized MO removal. Finally, tests in a continuous system yielded rates of MO removal 1.6 times greater than those of the batch system in a single pass, and demonstrated ~87% molar removal of MO at fluxes of approximately 422 lmh. Degradação anaeróbia de efluente têxtil simulado com corante azo Direct Black 22 na presença de íons sulfato em reator anaeróbio de leito estruturado com fluxo ascendente / Anaerobic biodegradation of simulated textile effluent with azo dye Direct Black 22 and sulphate ions in anaerobic down-flow fixed structured bed reactor

Avaliou-se o emprego de sistema biológico de tratamento em reator anaeróbio de leito estruturado em fluxo ascendente (RAnLE) para remoção do corante Direct Black 22 (DB22) e íons sulfato (65 e 200 mg.L-1, respectivamente), principais constituintes de efluente têxtil de indústrias de jeans da região de Caruaru, PE. Etanol foi utilizado como doador de elétrons na concentração de 1000 mg.DQO.L-1. As médias de remoção de DQO, sulfato e corante foram respectivamente 80,28 ± 6,62%, 78,66 ± 4,04% e 68 ± 5%. Cerca de 3% do sulfeto produzido foi identificado em solução, 10% esteve presente na forma gasosa e aproximadamente 87% precipitou em forma de cristais metálicos inorgânicos ou em enxofre elementar por exposição ao ar atmosférico ou por ação redutora do corante DB22, contribuindo para remoção de cor na transformação do corante em aminas aromáticas. Os ácidos graxos presentes no efluente tratado totalizaram 121 ± 34 mg.L-1, com predominância de ácido acético (95 ± 17 mg.L-1), sem prejuízo da capacidade tamponante do sistema, que apresentou aumento na alcalinidade pela ocorrência da sulfetogênese e pela característica básica das aminas aromáticas, produzidas por meio da quebra da ligação azo do corante, revelando absorção em diferentes comprimentos de onda na região do ultravioleta, denotando a produção de diferentes estruturas aromáticas. O efluente têxtil simulado foi mais tóxico do que seu correspondente tratado para ambas as situações (crônico e agudo) nos testes de toxicidade com Ceriodaphnia dúbia (crônica) e Daphnia magna (aguda), atestando o potencial dos reatores anaeróbios na destoxificação do corante azo Direct Black 22 para as espécies abordadas. Os testes de citotoxicidade in vitro empregando células McCoy (linhagem contínua de fibroblastos) em exposição ao efluente têxtil sintético tratado no RAnLE apresentaram um comportamento dose-dependente. O efluente diluído 3 vezes inibiu 50% das células. Por outro lado, não foi possível verificar um comportamento dose-dependente considerando as diferentes concentrações de DB22. Entretanto, houve diferença estatística significativa na viabilidade celular para as concentrações de DB22 de 0,016, 0,065, 0,2 e 0,26 mg.mL-1. Os resultados demonstram a necessidade do uso de diferentes testes de toxicidade a vários organismos, de forma a complementar os resultados obtidos pelos testes convencionais. Eletromicrografias da biomassa revelaram a presença de arqueias metanogênicas, demonstrando a capacidade de coexistência de comunidades sulfetogênicas e metanogênicas, dado o valor observado de DQO/sulfato de 5. O sequenciamento metagenômico em apontou grande abundância relativa de bactérias redutoras de sulfato (BRS), superior a 50%. Os bioensaios em batelada com o lodo de fundo e o lodo em suspensão do RAnLE apontaram que a biomassa aderida foi mais apta a suportar a carga tóxica proveniente do corante e das aminas aromáticas. A estrutura do RAnLE permitiu o estabelecimento de biomassa aderida e biomassa suspensa, concorrendo ambas para melhor desempenho do sistema. / The present work assessed the employment of an anaerobic up-flow fixed-structured bed (AUFFSB) reactor, which aimed to remove the main constituents of a simulated textile effluent with azo dye Direct Black 22 (DB22) and sulphate ions (65 and 200 mg.L-1, respectively), main constituents in residual waters generated by denim industry in Caruaru, PE. Ethanol was employed as electron donor (1000 mg.COD.L-1). Medium removals for COD, sulphate and dye concentration were respectively 80.28 ± 6.62%, 78.66 ± 4.04% and 68 ± 5%. Circa 3% of produced sulphide was identified in the effluent, 10% in gaseous state and approximately 87% precipitated either as metallic inorganic crystals, as elemental sulphur due to exposition to atmosphere, or due to the azo bound reduction of DB22, which contributed to colour removal in the generation of aromatic amines from the azo dye. The volatile fatty acids totalized 121 ± 34 mg.L-1 while acetic acid was the predominant one (95 ± 17 mg.L-1). Nevertheless, such concentrations did not impair the buffering capacity of the system, which had increased alkalinity due to the occurrence of sulphidogenesis and of the alkaline characteristic of the aromatic amines, which were produced from azo bound cleavage, revealing absorption in different wavelengths in the UV zone, inferring that different aromatic structures were formed. As for the toxicity, the simulated textile effluent was more toxic before treatment than after treatment for both bioessays employing Ceriodaphnia dubia and Daphnia magna, (chronic and acute toxicity), attesting the potential of anaerobic reactors to detoxify azo dye DB22. The in vitro cytotoxicity tests using McCoy cells (fibroblasts of a continuous lineage) exposed to simulated textile effluent treated on the anaerobic bioreactor presented a dose-dependent behaviour. The effluent diluted 3 times showed inhibition to 50% of the cells. On the other hand, it was not possible to verify a dose-dependent behaviour considering the different concentrations of DB22. However, there was significant statistical difference on cell viability for DB22 concentrations of 0.016, 0.065, 0.2 and 0.26 mg.mL-1. The results pointed to the need of diverse toxicity tests to various organisms in order to complement the results obtained by conventional used ones. Micrographs revealed the presence of methanogenic archeas, denoting the possibility of coexistence between sulphidogenic and methanogenic communities, considering the observed COD/sulphate ration of 5. Metadata obtained from metagenomic sequencing revealed high relative abundance of sulphate-reducing bacteria (SRB), which was superior to 50%. Batch bioessays showed that biomass in supporting material is more capable of enduring the toxic load from DB22 and aromatic amines. The AUFFSB structure enabled that suspended biomass developed along with fixed biomass, thus both collaborated for a better system performance.
