To evaluate interaction of water alkalinity and stocking density on body ionic levels, survival, growth and behavior of silver catfish, Rhamdia quelen, juveniles, a short-term experiment (96 h) analyzed the effect of three alkalinity levels (30, 80 and 130 mg/L CaCO3) and four stocking densities (4, 8, 16, and 24 g/L). In addition, a growth experiment of 30 days verified the effect of three stocking densities (4, 8, 16 g/L) and the same alkalinity levels. Survival and body ionic levels were not significantly affected in the short-term experiment, but after 30 days survival was significantly reduced in the group maintained at the stocking density of 16 g/L and 30 mg/L CaCO3. However, this treatment provided higher weight and specific growth rate compared to the treatment with the highest alkalinity and the same stocking density. The time of capture of the food, in both experiments, was faster in the treatments of high stocking density. In the short-term experiment fish remained mainly in the bottom, and the reduction of stocking density increased swimming speed. The color of the animals was significantly clearer in the lowest densities of the growth experiment. Agonistic side behaviors prevailed in the lowest alkalinities of the stocking density of 8 g/L, and cannibalism occurred only in the stocking density of 8 g/L and alkalinity of 130 mg/L CaCO3 of the growth experiment. The stocking density of 16 g/L and alkalinity of 80 mg/L CaCO3 seems to be the most appropriate for silver catfish juveniles growth in these experimental conditions. / Com o objetivo de avaliar a interação da alcalinidade da água com diferentes densidades de estocagem sobre os níveis iônicos corporais, sobrevivência, crescimento e comportamento de juvenis de jundiá, Rhamdia quelen, delineou-se um experimento de curta duração (96 h) com três níveis de alcalinidade (30, 80 e 130 mg/L CaCO3) e quatro densidades de estocagem (4, 8, 16 e 24 g/L). Além disso, em um experimento de crescimento (30 dias) testou-se três densidades de estocagem (4, 8, 16 g/L) e as mesmas alcalinidades. A sobrevivência e os níveis iônicos corporais não foram afetados no experimento de curta duração, mas após 30 dias a sobrevivência foi reduzida no tratamento de maior densidade e menor alcalinidade. Este mesmo tratamento foi o que obteve melhores resultados de peso e taxa de crescimento específico em relação à maior alcalinidade da mesma densidade. O tempo de captura do alimento, em ambos experimentos, foi mais curto nos tratamentos de maior densidade. No experimento de curta duração os peixes se concentravam mais no fundo à medida que se diminuía a densidade de estocagem e com o aumento desta aumentou a velocidade de natação. A coloração dos animais foi significativamente mais clara nas menores densidades do experimento de crescimento. Confrontos laterais predominaram nas menores alcalinidades da densidade de 8 g/L, canibalismos ocorreram apenas na densidade de 8 g/L e alcalinidade de 130 mg/L CaCO3 do experimento de crescimento. A utilização da densidade de estocagem de 16 g/L e alcalinidade de 80 mg/L CaCO3 é a mais adequada para o cultivo de juvenis de jundiá nestas condições experimentais.

Hidratação com bicarbonato de sódio na prevenção de nefropatia induzida por contraste : estudo clínico multicêntrico

Introdução: Nefropatia induzida por contraste (NIC) está associada a piores desfechos intra-hospitalar e a longo prazo. Estudos recentes sugerem que hidratação com bicarbonato de sódio possa ser útil na sua prevenção; no entanto, essa medida de prevenção de NIC não foi, ainda, avaliada em pacientes diabéticos. Método: Subanálise de um estudo multicêntrico envolvendo 301 pacientes com creatinina sérica ≥ 1,2 mg/dl ou depuração de creatinina endógena (DCE) < 50 ml/min submetidos a cineangiocoronariografia ou angioplastia coronária, randomizados para receber hidratação com bicarbonato de sódio ou solução salina (soro fisiológico – SF) a 0,9%. Todos os procedimentos foram realizados com contraste iônico de baixa osmolaridade. Os desfechos avaliados foram incidência de NIC (definida como aumento de 0,5 mg/dl) e variação da creatinina e da DCE em 48 horas após o procedimento. Da totalidade de pacientes, 87 tinham diabetes melito e foram incluídos nesse subestudo. Resultados: Não houve diferença entre os grupos em relação a características demográficas, volume de contraste e níveis basais de creatinina e DCE. Entre os pacientes avaliados, 8 desenvolveram NIC: 4 (9,8%) pacientes no grupo bicarbonato e 4 (8,9%) no grupo SF 0,9% (p = 0,9). A variação da creatinina sérica e da DCE foi similar entre os grupos. Conclusão: Hidratação com bicarbonato de sódio não demonstrou benefício em reduzir a incidência de NIC em pacientes diabéticos submetidos a cateterismo cardíaco ou angioplastia coronária em comparação à hidratação com SF 0,9%. / Background: Contrast-induced nephropathy (CIN) is associated with worse clinical outcomes both at short and long-term follow-up. Recent evidence indicates that intravenous hydration with sodium-bicarbonate may reduce the incidence of CIN. However, this strategy has not been reported in diabetic patients. Methods: Sub-analysis of a multicenter study involving 301 patients with serum creatinine ≥ 1,2 mg/dL or creatinine clearance < 50 mL/min submitted to coronary angiography or percutaneous coronary intervention and randomized to intravenous hydration with sodium-bicarbonate or normal saline. All patients received low-osmolar contrast media. We assessed the incidence of CIN (defined as creatinine increase ≥ 0,5 mg/dL), and the average change in creatinine and creatinine clearance 48 hours after the procedure. A total of 87 diabetic patients were analyzed. Results: There was no difference between groups regarding baseline characteristics, contrast volume used, baseline creatinine levels and creatinine clearance. Eight patients presented CIN: 4 (9.8%) in the bicarbonate group and 4 (8.9%) in the saline group (p = 0.9). The average change in serum creatinine and creatinine clearance were similar between groups. Conclusion: Intravenous hydration with sodium-bicarbonate did not reduce the incidence of CIN in diabetic patients undergoing coronary angiography or percutaneous coronary intervention as compared to hydration with normal saline.

Essas tecnologias foram aplicadas com o intuito de melhorar a estabilidade desse material, uma vez que o mesmo se decompõe na presença de umidade/ou temperaturas mais elevadas. Uma mistura lipídica composta por gordura de palma totalmente hidrogenada (GPTH) e óleo de palma refinado (OP) foi empregada como material de parede/ligante que atendeu ao requisito de ponto de fusão de 55 ºC, com a proporção de 57,5 e 42,5 %, respectivamente. No processo de spray cooling, foi realizado um Delineamento Composto Central Rotacional (DCCR), com 11 ensaios, para cada um dos três diâmetros de abertura de bico atomizador estudados (??igual a 1,0; 1,2 e 1,5 mm), totalizando 33 experimentos, a fim de se avaliar o efeito da temperatura de entrada da dispersão (Te) (65 a 85 ºC) e da pressão de atomização (Pat) (1,0 a 2,0 kgf/cm2) sobre as seguintes variáveis dependentes: (i) eficiência de microencapsulação (EM, % de CO2), (ii), quantidade de bicarbonato de sódio superficial (BSS, % de CO2) e (iii) rendimento (R, em %). As micropartículas apresentaram formato esférico e estrutura de superfície irregular para os ensaios realizados. A análise estatística dos resultados mostrou que para ??= 1,0 mm, um aumento nos níveis da Te e da Pat ocasionou um decréscimo da EM; para o ??= 1,2 mm, o aumento da Te ocasionou uma melhoria nessa resposta, entretanto, a Pat quando aumentada acarretou na sua diminuição. Para o ??= 1,5 mm, essas duas variáveis apresentaram menores efeitos, onde o aumento da Pat favoreceu a EM e o aumento da Te, a sua diminuição. A Te exerceu efeito positivo no R para o ??= 1,0 mm, sem influência nessa resposta para os diâmetros 1,2 e 1,5 mm, enquanto que a Pat teve efeito positivo para ??igual a 1,0 e 1,5 mm (termo linear) e negativo para o termo quadrático deste último. Pela combinação das respostas obtidas, verificou-se que o ensaio 4, diâmetro de bico atomizador (??= 1,5 mm), altas temperatura de entrada e de pressão de atomização (Te = 82 ºC e Pat = 1,85 kgf/cm2) apresentou o melhor desempenho (eficiência 3,18 %; quantidade de bicarbonato de sódio superficial 0,17 % e rendimento 80,0 %) quando comparado com os outros diâmetros de abertura. No processamento em leito vibrofluidizado, foi realizado um DCCR, totalizando 17 ensaios, com o objetivo de avaliar o efeito das variáveis: (i) pressão de atomização (5,0 a 15,0 Psig), (ii) frequência de vibração (186 a 354 rpm) e (iii) amplitude de vibração (0,40 a 2,00 cm), sobre as variáveis dependentes (i): rendimento R (%), (ii) eficiência da aglomeração Eagl (% CO2), para as frações granulométricas de 300, 600 e 840 µm, (iii) crescimento de partícula C3,3 (%) e (iv) índice de torrões Itor (%). O material aglomerado apresentou ampla distribuição de tamanho e a nucleação dos aglomerados ocorreu pelo mecanismo de imersão. Os aglomerados apresentaram Eagl diferentes para as frações avaliadas, sendo que a maior eficiência foi observada para a fração 840 µm, seguida das frações 600 e 300 µm. Contudo, através da comparação entre a Eagl e o C3,3, observou-se que a proteção não depende somente do tamanho da partícula, mas mais importante que isso é o mecanismo de formação do aglomerado, demonstrando a complexidade do processo. Com relação à proteção do material (avaliada pela Eagl), o efeito da pressão, da frequência e da amplitude foi positivo para as frações de 300 µm e 600 e 840 µm (termos quadráticos). Pela combinação das respostas, o ensaio que apresentou melhor desempenho no leito vibrofluidizado foi a de fração 840 µm, cujos parâmetros estavam fixados em baixos valores de pressão e frequência e alta amplitude. Os resultados demonstraram que ocorreu a proteção do bicarbonato de sódio contra a liberação de CO2 para ambos os processos estudados. As técnicas demonstraram grande potencial na produção desses materiais (bicarbonato de sódio microencapsulado e bicarbonato de sódio aglomerado), que se mostram viáveis para serem utilizados na formulação de um novo fermento químico, constituído pelo bicarbonato de sódio microencapsulado ou aglomerado e pelo componente ácido, sem a necessidade do agente inerte (amido) como separador físico / Abstract: This work had the objective of developing: (i) lipid microparticles containing sodium bicarbonate obtained by spray cooling process, and (ii) sodium bicarbonate agglomeration, obtained by melt agglomeration process in a vibrofluidized bed. These technologies were applied with the objective of improving the material stability, since it decomposes in the presence of humidity and/or higher temperatures. A lipid mixture composed of 57.5 % fully hydrogenated palm fat and 42.5 % refined palm oil, whose melting point is 55 ºC, was used. In the spray cooling process, a Central Composite Rotatable Design (CCRD) was applied, with 11 trials for each of the three nozzle atomizer diameters (?) studied (??= 1.0, 1.2 and 1.5 mm). Thirty three experiments were conducted with the objective of evaluating inlet temperature on the dispersion (65 to 85 ºC) and pressure atomization (1.0 to 2.0 kgf/cm2) on the following dependent variables: (i) microencapsulation efficiency (% CO2), (ii) amount of sodium bicarbonate on the surface (% CO2) and (iii) yield (%). The microparticles presented spherical shape and irregular surface. Statistical analysis showed that for ??= 1.0 mm and increases in levels of temperature on the dispersion and pressure atomization, the microencapsulation efficiency dropped. For ??= 1.2 mm, an increase in temperature on the dispersion led to an improvement in this response, however, when pressure atomization was increased, the microencapsulation efficiency fell. For ??= 1.5 mm, these two variables presented smaller effects, on the other hand, an increase in pressure atomization favored the microencapsulation efficiency. Increasing temperature on the dispersion, reduced the microencapsulation efficiency. Temperature on the dispersion presented a positive effect on yield for ??= 1.0 mm (this had no effect on the response for 1.2 and 1.5 mm diameters), while pressure atomization had a positive effect for d = 1.0 and 1.5 mm (linear term), its quadratic term had a negative effect on yield. By combining of dependent variables, it was found for ??= 1.5 mm, test 4, higher temperature on the dispersion and pressure atomization, presented the best performance (3.18 % higher efficiency, 0.17 % lower amount of surface sodium bicarbonate pressure and 80.0 % yield) when compared with the other diameters. In the vibrofluidized bed process, a CCRD was conducted, totaling 17 trials, to assess the effect of these independent variables: (i) atomization pressure (5.0 to 15.0 Psig), (ii) vibration frequency (186 to 354 rpm) and (iii) vibration amplitude (0.40 to 2.00 cm) on the dependent variables: (i) yield (%), (ii) agglomeration efficiency (% CO2), for the particle size fractions of 300, 600 and 840 µm (iii) particle growth (%) and (iv) lump. The agglomerated material presented ample size distribution and the nucleation mechanism occurred by immersion. The agglomeration efficiency presented different values for each fractions tested (300 µm <600 µm <840 µm). However, a comparison of agglomeration efficiency and particle growth, demonstrated that protection depends not only on particle size, but more on the formation mechanism of agglomeration particles, demonstrating its complexity. With regard to the protection of the material (assessed by agglomeration efficiency), the effect of pressure was positive for 300, 600 µm, and 840 µm fractions (quadratic terms), while the frequency had a positive effect for the three fractions. With respect to amplitude, the effect was positive on the larger particle size fractions. Considering all answers, the test that produced the best performance in the vibrofluidized bed was 840 µm fraction, whose parameters were set at low pressure and frequency values and high amplitude. The results showed that there was protection of sodium bicarbonate against the release of CO2 for both processes studied. The techniques demonstrated great potential in the production of these materials (microencapsulated sodium bicarbonate and agglomerated sodium bicarbonate particles), thus demonstrating its viability for use in the formulation of a new baking powder. Evaluation of Anion Transporters as Potential Target Sites for Insect and Nematode Control: Toxicological, Electrophysiological, and Molecular Studies

In this study, four anion transporter (AT) blockers, DIDS (4, 4′-diisothiocyanatostilbene-2, 2′-disulfonic acid), 9-AC (anthracene-9-carboxylic acid), NPPB (5-nitro-2-(3-phenylpropylamino) benzoic acid), and IAA-94 (indanyloxy acetic acid) were selected to evaluate ATs as potential target sites for insect and nematode control. All the AT blockers showed slowly developing toxicity against second-stage larvae of <i>Meloidogyne incognita</i> (Kofoid and White 1919) Chitwood 1949 and adults of <i>Caenorhabditis elegans</i> Maupas 1900 but not against third-stage larvae of <i>Heterorhabditis bacteriophora</i> Poinar 1975 even at 200 ppm. Symptoms of AT blocker toxicity observed in <i>C. elegans</i> adults were increased pharyngeal muscle contractions and decreased locomotion. Exposure of <i>C. elegans</i> as fourth-stage larvae to double-stranded RNA (dsRNA) of <i>ceclc-1</i> and <i>ceclc-2</i> (VGCC genes coding for CeClC-1 and CeClC-2, respectively) either alone or together for 24 h decreased their expression in F1 progeny in a time-dependent manner. Reduction in expression of <i>ceclc-2</i> alone or together with <i>ceclc-1</i> significantly increased pharyngeal contractions and decreased locomotion in significantly higher percentage of F1 progeny. The above findings suggested AT blockers nematicidal activity primarily comes from inhibition of CeClC-2 channels, while inhibition of CeClC-1 channels may enhance this activity. All the AT blockers showed slowly developing toxicity against adults of a susceptible strain (Oregon-R) of <i>Drosophila melanogaster</i> Meigen 1830, while DIDS, was equally toxic to dieldrin-resistant rdl flies. All AT blockers, except 9-AC, at 100 µM showed significant excitatory effect on desheathed central nervous system (CNS) of third-instar larvae of <i>Drosophila</i>, while DIDS showed a modest excitatory effect on ascending peripheral nerves. Feeding adult flies on 10% sugar solution mixed with 100 ppm of DIDS for 6 h decreased the midgut pH by 2 units approximately. All the AT blockers inhibited the growth of larvae (in weight), increased the developmental time, and decreased survival when <i>Ostrinia nubilalis</i> (Hübner 1796) second-instar larvae were fed for seven days. All the AT blockers decreased the midgut alkalinity and inhibited chloride ion transport from midgut lumen into epithelia in fifth-instar larvae when fed for 3 h on treated diet. Positive correlations observed among growth, midgut alkalinity, and midgut chloride transport in AT blocker-fed larvae suggested that inhibition of chloride/bicarbonate exchangers by AT blockers may have contributed to midgut alkalinity decrease affecting the digestion and resulting in observed lethal and sublethal effects. / Ph. The effect of acid buf and combinations of acid buf and socium bicarbonate in dairy cow diets on production response and rumen parameters

Thesis (MScAgric (Animal Sciences))--University of Stellenbosch, 2009. / ENGLISH ABSTRACT: The effect of Acid Buf and combinations of Acid Buf and Sodium bicarbonate in dairy cow diets on production response and rumen parameters The objective of this study was to determine the effect of Acid Buf alone, or in combination with sodium bicarbonate, on milk production, milk composition and selected rumen parameters. A high concentrate TMR, formulated to be potentially acidotic, was used to construct four dietary treatments in which Acid Buf (AB), the skeletal remains of the seaweed Lithothamnium calcareum, was used alone or in combination with sodium bicarbonate (BC). The diets contained 3.5 g/kg of AB (Treatment 1) or 1.75 g/kg of AB and 1.75 g/kg of BC (Treatment 2) or 3.5 g/kg of AB and 3.5 g/kg of BC (Treatment 3) or 3.5 g/kg of AB and 5.2 g/kg of BC (Treatment 4). The response to treatment was determined using eight ruminally cannulated lactating Holstein cows randomly allocated to treatments according to a 4 x 4 (n=2) Latin square design, with four treatments and four periods. The total experimental period was 100 days in which every cow received each diet for a period of 18 days prior to a data collection period of 7 days. Rumen pH was monitored continuously over 2 days using a portable data logging system and in-dwelling electrodes. Samples of rumen fluid were collected for volatile fatty acid (VFA) analyses. During each data collection period, milk production was recorded twice daily for 7 days, whereas milk was sampled twice daily for five consecutive days for component analysis. Cows were fed ad libitum and dry matter intake was recorded individually. Treatment had no significant effect on milk production, milk composition or feed intake. Ruminal pH profiles of all the treatments indicated that the diets were well buffered. Average pH over 24 hours was 6.0, 6.1, 6.1 and 6.2 for Treatments 1, 2, 3 and 4, respectively. The pH did not go below 5.8 for any of the treatments and increasing levels of sodium bicarbonate increased the diurnal profile such that at the highest level (Treatment 4), the pH profile ranged from 6.1 to 6.5. Although not significant, Treatment 1 (Acid Buf alone) numerically resulted in the highest milk output without compromising milk quality. It is proposed that high rumen pH may impact negatively on milk output by increasing the acetate:propionate ratio to the detriment of rumen efficiency. Treatment had no significant effect on total VFA concentration, but there seemed to be a tendency for increased total VFA concentration as the level of sodium bicarbonate increased. The acetate:propionate ratio of Treatment 1 (2.91) was significantly lower than Treatment 4 (3.65) (P<0.05). The use of buffers which react to increasing acidity in the rumen may therefore provide an efficient, safe solution to rumen acidosis. Rumen pH profiele van al die behandelings het aangedui dat die diëte goed gebuffer was. Die gemiddelde pH oor 24 uur was 6.0, 6.1, 6.1 en 6.2 vir Behandeling 1, 2, 3 en 4, onderskeidelik. Die pH het nie onder 5.8 gedaal vir enige van die behandelings nie en die toenemende vlakke van natriumbikarbonaat het ‘n verhoging in die daaglikse pH-profiele tot gevolg gehad. Hoewel nie betekenisvol nie, het Behandeling 1 (slegs Acid Buf) numeries die hoogste melkproduksie getoon, sonder dat melkkwaliteit beïnvloed is. Dit is voorgestel dat te hoë rumen pH ‘n nadelige impak op melkproduksie kan hê, as gevolg van ‘n verhoogde asetaat:propionaat verhouding wat rumen doeltreffendheid benadeel. Behandelings het geen betekenisvolle invloed op totale vlugtige vetsuurkonsentrasies gehad nie, maar daar was ‘n neiging vir hoër totale vlugtige vetsuurkonsentrasies namate die vlakke van natriumbikarbonaat verhoog het. Die asetaat:propionaat verhouding van Behandeling 1 (2.91) was betekenisvol laer as dié van Behandeling 4 (3.65). Die gebruik van buffers wat reageer op toenemende suurheidsgraad kan dus aangewend word as ‘n doeltreffende en veilige oplossing vir rumen asidose. Die studie het bevestig dat ‘n daaglikse inname van 80 g Acid Buf deur koeie wat ‘n hoë kragvoerdieet ontvang, hoë melkproduksie ondersteun sonder dat melksamestelling nadelig beïnvloed word.

Role of Melatonin, Neuropeptide S and Short Chain Fatty Acids in Regulation of Duodenal Mucosal Barrier Function and Motility

The duodenal epithelium is regularly exposed to HCl, digestive enzymes, bacteria and toxins, and sometimes also to ethanol and drugs. The imbalance of aggressive factors in the intestinal lumen and mucosal barrier function increases the risk of tissue injury and inflammation. The key components of the duodenal barrier function include mucosal permeability, bicarbonate transport and the secretion or absorption of fluids. This thesis aims to elucidate the role of melatonin, neuropeptide S (NPS) and short chain fatty acids (SCFAs) in the regulation of intestinal mucosal barrier function and motility in the anesthetized rat in vivo and in tissues of human origin in vitro. Melatonin was found to reduce ethanol-induced increases in paracellular permeability and motility by a neural pathway within the enteric nervous system involving nicotinic receptors. In response to luminal exposure of ethanol, signs of mild mucosal edema and beginning of desquamation were observed in a few villi only, an effect that was not influenced by melatonin. Melatonin did not modify increases in paracellular permeability in response to luminal acid. NPS decreased basal and ethanol-induced increases in duodenal motility as well as bethanechol stimulated colonic motility in a dose-dependent manner. Furthermore, NPS was shown to inhibit basal duodenal bicarbonate secretion, stimulate mucosal fluid absorption and increase mucosal paracellular permeability. In response to luminal exposure of acid, NPS increased bicarbonate secretion and mucosal paracellular permeability. All effects induced by the administration of NPS were dependent on nitrergic pathways. In rats, administration of NPS increased the tissue protein levels of the inflammatory biomarkers IL-1β and CXCL1. Immunohistochemistry showed that NPS was localized at myenteric nerve cell bodies and fibers, while NPSR1 and nNOS were only confined to the myenteric nerve cell bodies. Perfusing the duodenal segment with the SCFAs acetate or propionate reduced the duodenal mucosal paracellular permeability, decreased transepithelial net fluid secretion and increased bicarbonate secretion. An i.v. infusion of SCFAs reduces mucosal paracellular permeability without any effects on mucosal net fluid flux. However, it significantly decreased bicarbonate secretion. Luminal SCFAs changed the duodenal motility pattern from fasting to feeding motility while i.v. SCFAs was without effect on motility. The systemic administration of glucagon-like peptide-2 (GLP-2) induced increases in mucosal bicarbonate secretion and fluid absorption. An i.v. GLP-2 infusion during a luminal perfusion of SCFAs significantly reduced the duodenal motility. In conclusion, the results in the present thesis show that melatonin, NPS and SCFAs influence the neurohumoral regulation of intestinal mucosal barrier function and motility. Nós determinamos a influência da alcalose e acidose metabólica induzida sobre o ritmo de prova, gasto energético e desempenho durante um teste contrarrelógio de ciclismo de 4 km (TCR). Após um estudo preliminar (N=7) determinando as doses ótimas de cloreto de amônio (NH4Cl), bicarbonato de sódio (NaHCO3) e placebo (carbonato de cálcio, CaCO3), 11 ciclistas amadores realizaram um TCR de 4 km 100 min após ingerirem 0,15 g·kg-1 de NH4Cl, 0,3 g·kg-1 de NaHCO3 ou 0,15 g·kg-1 de CaCO3 (desenho de medidas repetidas, contrabalançado e duplo-cego). Comparado com placebo, a ingestão de NaHCO3 resultou em uma alcalose sanguínea pré-exercício (pH: + 0,06 ± 0,04, [HCO3-]: + 4,7 ± 2,6 mmol?L-1, excess de base: + 4,6 ± 2,4 mmol?L-1, P<0,05), enquanto que o NH4Cl resultou em uma acidose sanguínea (pH: - 0,05 ± 0,04, [HCO3-]: - 5,5 ± 2,9 mmol?L-1, excesso de base: - 5,3 ± 2,5 mmol?L-1, P<0.05), com essas diferenças permanecendo durante todo o teste. Apesar de nenhum desconforto gastrointestinal severo, os participantes adotaram um ritmo de prova significativamente mais conservador em NH4Cl comparado com placebo e NaHCO3, que foi acompanhado por uma redução na taxa de gasto energético anaeróbio (~ -14 %, P<0,05) e menor trabalho anaeróbio total (~ -13 %, P<0,05). Consequentemente, o desempenho foi prejudicado em NH4Cl (~ -7 %) comparado com as demais condições (P<0,05). After a preliminary study (N=7) determining the optimal ammonium chloride (NH4Cl), sodium bicarbonate (NaHCO3), and placebo (calcium carbonate, CaCO3) doses, 11 amateur cyclists performed a 4-km TT 100 min after ingesting either 0.15 g·kg-1 of body mass of NH4Cl, 0.3 g·kg-1 of NaHCO3 or 0.15 g·kg-1 of CaCO3 (double-blind, counterbalanced and repeated-measures design). Compared with placebo, the NaHCO3 ingestion resulted in a pre exercise blood alkalosis (pH: +0.06 ± 0.04, [HCO3-]: +4.7 ± 2.6 mmol?L-1, base excess: +4.6 ± 2.4 mmol?L-1, P<0.05), while NH4Cl resulted in a blood acidosis (pH: -0.05 ± 0.04, [HCO3-]: -5.5 ± 2.9 mmol?L-1, base excess: -5.3 ± 2.5 mmol?L-1, P<0.05), with these differences remaining throughout the trial. Despite no gastrointestinal distress, participants adopted a significant more conservative pacing in NH4Cl compared with placebo and NaHCO3, which was accompanied by a reduced anaerobic energy expenditure rate (~ -14 %, P<0.05) and lower total anaerobic work (~ -13 %, P<0.05). Consequently, the performance was less in NH4Cl (~ -7 %) compared with other conditions (P<0.05). Plasma lactate, VCO2 and PetCO2 were lower and the VE/VCO2 higher during the TT in acidosis than in placebo and alkalosis (P<0.05). Efeito in situ de uma solução de bicarbonato de sódio em esmalte dental após simulação de desafios erosivos endógenos / In situ effect of a sodium bicarbonate solution on enamel after simulation of endogenous dental erosion.

Uma das medidas recomendadas para o controle da progressão de lesões de erosão dental de origem intrínseca é o enxágüe bucal com solução de bicarbonato de sódio, visando à neutralização de ácidos gástricos presentes na boca. Contudo, a prescrição de tal solução faz-se em bases empíricas. Assim, este estudo avaliou, através de um modelo in situ, o efeito de uma solução de bicarbonato de sódio no controle da erosão dental causada por desafios ácidos endógenos. Este estudo caracterizou-se por um experimento do tipo cross-over 2x2, composto por duas fases experimentais de 5 dias cada. Após um período de lead-in de 2 dias, quatorze voluntários utilizaram dispositivos intrabucais contendo, em cada lado (direito e esquerdo), um fragmento de esmalte bovino polido, esterilizado e delimitado de modo a obter áreas hígidas de referência. Duas vezes ao dia, um dos espécimes foi exposto, ex-vivo, à solução de ácido clorídrico (0,01 M, pH = 2) por 2 min. Imediatamente após, os aparelhos foram reinseridos na boca e os voluntários realizaram enxágüe bucal com solução de bicarbonato de sódio ou água deionizada por 1 min. Após um período de wash-out de 2 dias, os voluntários iniciaram a segunda fase empregando a substância alternativa para o enxágüe bucal. Ao término das duas etapas, foram avaliadas a profundidade de desgaste através de perfilometria ótica e a microdureza Knoop. ANOVA e o teste de Tukey demonstraram que o desgaste do esmalte foi significativamente reduzido quando se empregou a solução de bicarbonato de sódio para o enxágüe bucal. Para a microdureza, ANOVA demonstrou apenas presença de diferença significativa em função da condição do substrato (p < 0,0001). Os espécimes erodidos apresentaram valores de microdureza significativamente menores do que o grupo não submetido ao desafio ácido. O enxágüe bucal com solução de bicarbonato de sódio controlou a erosão no esmalte submetido a simulações de episódios erosivos endógenos. / One of the recommended measures to control dental erosion of intrinsic origin is the oral rinse with sodium bicarbonate solution, which has been suggested to neutralize the gastric acid regurgitated into the oral cavity. However, the efficacy of this prescription has not yet been proved. Therefore, the aim of this study was to evaluate in situ the effect of a sodium bicarbonate rinse in the control of dental erosion in enamel exposed to gastric acid-like solution. This study comprised two experimental legs. After a 2-day lead-in period, fourteen volunteers wore palatal devices containing two polished, sterilized enamel slabs. Throughout the experimental period of 5-d, one of the two specimens was exposed twice daily extra-orally to hydrochloric acid solution (0,01 M, pH=2) for 2 min. Immediately after, devices were reinserted in the mouth and volunteers rinsed their oral cavity with a sodium bicarbonate solution or deionized water for 60s. In the second leg of this study, after a 2-day washout period, appliances were refilled with a new set of enamel specimens and participants were crossed over to receive the alternate rinse. The wear depth accessed using optical profilometry and Knoop surface microhardness were evaluated. Analysis of Variance (ANOVA) and Tukey\'s test demonstrated that wear of enamel was significantly reduced when a sodium bicarbonate solution was used to rinse the oral cavity following erosive episodes. For microhardness, ANOVA demonstrated only significant differences in function of the enamel condition (p < 0,0001). The eroded specimens presented microhardness values significantly lower than the group not submitted to the acid challenge. Sodium bicarbonate rinse controlled erosion of enamel caused by gastric acid-like solution.

Efeito de antioxidantes na resistência adesiva à microtração do esmalte bovino clareado após 7 dias e 6 meses de armazenamento / Effect of antioxidants on bleached enamel microtensile bond strength after 7 days and 6 months storage

Este estudo in vitro avaliou o efeito do bicarbonato de sódio 10% (BS) e do hidrogel de ascorbato de sódio 10% (AS) na resistência adesiva à microtração de uma resina composta unida à superfície de esmalte bovino clareado com peróxido de hidrogênio 35% (PH). Cento e vinte superfícies de esmalte vestibular com dimensões de 4mmX4mm de incisivos bovinos foram planificadas e separadas em cinco grupos: C - não clareado e restaurado; OI - clareado com PH e restaurado imediatamente; O7 - clareado com PH, armazenado por 7 dias em água destilada e restaurado; OBS - clareado com PH, BS 10% durante 5 min e restaurado; OAS - clareado com PH, AS 10% durante 15 min e restaurado. O gel de PH (Total Blanc Office, Nova DFL) foi aplicado duas vezes de 20 minutos em cada espécime e as restaurações foram confeccionadas com sistema adesivo restaurador (Natural Bond DE + Natural Look, Nova DFL). Após uma semana, os espécimes foram seccionados para obtenção de palitos com 1,00 mm2 de área média e submetidos ao teste de microtração em máquina de teste universal a 0,5mm/min, imediatamente e após 6 meses. As falhas foram avaliadas em estereomicroscópio digital (200x) e classificadas em: adesiva, coesiva em esmalte, coesiva em resina e mista. Os dados foram analisados por ANOVA a dois critérios e teste de Tukey para comparações múltiplas (p<0,05). Os valores médios de resistência adesiva (MPa ± DP; imediatamente / 6 meses) foram: C (24,22 ± 7,74 Ba / 20,13 ± 6,43 BCa); OI (18,29 ± 5,88 Ca / 25,85 ± 5,23 Ab); O7 (40,88 ± 7,95 Aa / 24,61 ± 7,83 ABb); OBS (19,95 ± 5,67 BCa / 15,94 ± 5,57 CDa); OAS (24,43 ± 6,43 Ba / 13,69 ± 5,58 Db). Das estratégias antioxidantes avaliadas após 7 dias, os grupos OAS e OBS não diferiram estatisticamente do grupo C, nem OBS diferiu estatisticamente do grupo OI. Nesse período de avaliação, somente o grupo O7 apresentou diferença significativa em relação aos demais grupos. Após 6 meses de envelhecimento, apenas O7 e OBS apresentaram desempenho semelhante estatisticamente ao grupo C. O percentual de falhas (7 dias / 6 meses) foi: adesiva (65,26% / 67,06%); coesiva em esmalte (14,74% / 24,71%); coesiva em resina (1,58% / 2,35%); mista (18,42% / 5,88%). Em função dos resultados apresentados, a estratégia mais segura é aguardar 7 dias após o clareamento para a realização de restaurações adesivas. Quando não for possível aguardar este tempo, a aplicação de bicarbonato de sódio 10% por 5 minutos sobre o esmalte clareado deve ser feita. / This \"in vitro\" study evaluated the effect of 10% sodium bicarbonate (SB) and 10% sodium ascorbate (SA) hydrogel on the composite resin microtensile bond strength of bleached bovine enamel with 35% hydrogen peroxide (HP). One hundred and twenty buccal enamel surfaces with dimensions of 4mmX4mm of bovine incisors were flattened and separated into five groups: C - not bleached and restored, OI - bleached with HP and immediately restored; O7 - bleached with HP, stored for 7 days in distilled water and restored; OBS - bleached with HP, 10% SB for 5 min and restored; OAS - bleached with HP, 10% SA for 15 min and restored. HP gel (Total Blanc Office, Nova DFL) was applied twice for 20 minutes on each specimen and the restorations were performed with an adhesive restorative system (Natural Bond DE + Natural Look, Nova DFL). After a week, the specimens were sectioned into sticks with a 1.00 mm2 average area and submitted to a microtensile test in a universal testing machine at 0.5 mm/min, immediately and after 6 months. The failures were evaluated in a digital stereoscope (200x) and classified as: adhesive, cohesive in enamel, cohesive in resin and mixed. Data was analyzed by two-way ANOVA and Tukey\'s test for multiple comparisons (p <0.05). The average values of the bond strength (MPa ± SD; immediately / 6 months) were: C (24.22 ± 7.74 Ba / 20.13 ± 6.43 BCa) OI (18.29 ± 5.88 Ca / 25.85 ± 5.23 Ab); O7 (40.88 ± 7.95 Aa / 24.61 ± 7.83 ABb), OBS (19.95 ± 5.67 BCa / 15.94 ± 5.57 CDa), OAS (24.43 ± 6.43 Ba / 13.69 ± 5.58 Db). Among the antioxidant strategies evaluated after 7 days, the OAS and OBS groups did not differ from group C and neither OBS group differed from the OI group. In the same evaluation period, only the O7 group presented a significant difference compared to the other groups. After 6 months of aging, only the O7 and OBS groups performed statistically similar to group C. The percentage of failures (7 days / 6 months) was: adhesive (65.26% / 67.06%); cohesive in enamel (14.74% / 24.71%); cohesive in resin (1.58% / 2.35%); mixed (18.42% / 5.88%). According to the results presented, the safety strategy is waiting 7 days after bleaching for the adhesive restorative procedures. When this waiting period is not possible, 10% sodium bicarbonate should be applied for 5 minutes on the bleached enamel.
