Para o estudo da histopatologia, composto por 12 tratamentos (dois porta-enxertos de tomateiro, ‘Magnet’ e ‘Helper M’; duas espécies de nematoides das galhas, M. mayaguensis e M. javanica; e três épocas de coleta, 10 DAI, 17 DAI e 24 DAI), com três repetições. As plantas foram inoculadas com 500 juvenis infectivos (J2) de M. mayaguensis ou M. javanica. As avaliações foram realizadas em cinco épocas de coleta. Em cada coleta, cinco plantas foram retiradas ao acaso e suas raízes separadas em duas porções. Uma das partes foi submetida à coloração com Fucsina Ácida e dissecada sob microscópio estereoscópico para a contagem dos diferentes estádios de desenvolvimento dos nematoides. Os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey a 5% de probabilidade. Na outra porção do sistema radicular, foram feitos cortes histológicos transversais com 8 μm de espessura, corados com Floxina B seguido de Azul de Toluidina 0,05%, para determinação dos números e áreas das células gigantes e dos sítios de alimentação presentes no cilindro vascular da raiz. Dichomeris famulata Meyrick, 1914 (Lepidoptera: Gelechiidae) é uma nova praga da espiga de milho no Brasil, sendo importante em áreas de produção de sementes porque os grãos atacados pelas lagartas não germinam. O objetivo deste trabalho foi estudar a sua biologia, descrever os danos causados pela praga e verificar o efeito de alguns tratamentos no controle dos adultos, por ingestão, em condições de laboratório (25 ± 2°C, UR de 65 ± 10% e fotofase de 14 horas). Os inseticidas foram fornecidos às mariposas com e sem isca a base de mel a 10%. Os inseticidas utilizados e suas respectivas doses foram: cloridrato de cartape (0,5 g i.a. L-1 calda), metomil (0,258 g i.a. L-1 calda), acetamiprido (0,2 g i.a. L-1calda), espinosade (0,24 g i.a. L-1 calda) e pirimifós-metílico (0,5 g i.a. L-1 calda), além da testemunha. O ciclo biológico (ovoadulto) foi de 35,2 dias. O período de incubação foi de 4,1 dias. A duração média da fase larval foi de 21,1 dias, sendo observados cinco ínstares larvais. A fase pupal durou 8,6 dias para os machos e 8,1 dias para as fêmeas. O peso de pupa de machos e fêmeas foi de 12,4 e 11,3 mg, respectivamente. As fêmeas colocaram, em média, 118 ovos, apresentando um período de préoviposição de 10,7 dias e de oviposição de 14,0 dias. A longevidade média de machos e fêmeas foi de 37,02 e 44,16 dias, respectivamente, e a razão sexual de 0,48. As iscas contendo cloridrato de cartape e metomil causaram 97,92 e 83,33% de mortalidade de adultos, respectivamente 24 horas após a exposição dos tratamentos, sendo promissores no controle desta praga e podendo ser utilizados alternadamente, caso necessário. Cloridrato de cartape sem atrativo alimentar causou mortalidade de 91,67% de adultos. As lagartas danificam os estilo-estigmas e os grãos em formação ou em estado leitoso por meio de pequenos orifícios de entrada, prejudicando o endosperma e principalmente a região do embrião, inutilizando-os para sementes. A visualização dos danos é difícil, sendo que esses passam despercebidos. Os resultados obtidos neste trabalho fornecem subsídios para o estabelecimento de estratégias de manejo do inseto, especialmente em áreas de produção de sementes. / Dichomeris famulata Meyrick, 1914 (Lepidoptera: Gelechiidae) is a new pest of corn in Brazil, and is important in seed fields. The work goals were to study the biology of this pest, describe the damages caused by this pest and evaluate the effect of some insecticides, with and without bait, to control adults by ingestion, under laboratory conditions (25 ± 2°C, 65 ± 10% of RH and 14-hours of photophase). Pesticides were provided to moth with and without honeybee 10%. The insecticides used and respective doses were: cartap (0.5 g a.i. L-1), methomyl (0.258 g a.i. L-1), acetamiprid (0.2 g a.i. L-1), spinosad (0.24 g a.i. L-1), pirimiphos-methyl (0.5 g a.i. L-1) and a control treatment. The biological cycle (egg-adult) was 35.2 days. The incubation period was 4.1 days. The mean time larval was 21.1 days and 5 instars were observed. The pupal period was 8.6 days for male and 8.1 days for female insects. The pupae weight was 12.4 and 11.3 mg for males and females, respectively. The females laid an average of 118 eggs with a preoviposition period of 10.7 days and an oviposition time of 14.0 days. The mean longevity of males and females was 37.02 and 44.16 days, respectively, and sex ratio was 0.48. The baits with cartap and methomyl caused 97.92 and 83.33% of adults mortality, respectively, 24 hours after exposure to the treatments. This is a promising way to control this pest and it may be used in rotation. Cartap without attractive caused 91.67% of adults mortality. The caterpillars damage the silks and beginning of grain development or grains at the milk stage through little entry orifices, damaging the endosperm and, more importantly, the embryo, which disenables them for seed. Therefore, it is difficult to visualize the damage, which is unnoticed without an accurate examination. These results can support the establishment of strategies to manage this new corn ear pest, especially in seeds field.

Observaram-se diferenças significativas entre as médias dos períodos de préoviposição das duas espécies, em todas as temperaturas, excetuando-se entre as obtidas à 25º C. O número médio de ovos/fêmea, colocados nas temperaturas de 21 e 25º C, foram de 13,6 e 18,8; e, de 24,5 e 22,5, respectivamente, para P. nasuta e C. stephanoderis. Levantamentos de campo efetuados no período de 2001 a 2003, constataram a presença desta última espécie em dezoito municípios do Espírito Santo e em seis do estado de São Paulo. Pesticides were provided to moth with and without honeybee 10%. The insecticides used and respective doses were: cartap (0.5 g a.i. L-1), methomyl (0.258 g a.i. L-1), acetamiprid (0.2 g a.i. L-1), spinosad (0.24 g a.i. L-1), pirimiphos-methyl (0.5 g a.i. L-1) and a control treatment. The biological cycle (egg-adult) was 35.2 days. The incubation period was 4.1 days. The mean time larval was 21.1 days and 5 instars were observed. The pupal period was 8.6 days for male and 8.1 days for female insects. The pupae weight was 12.4 and 11.3 mg for males and females, respectively. The females laid an average of 118 eggs with a preoviposition period of 10.7 days and an oviposition time of 14.0 days. The mean longevity of males and females was 37.02 and 44.16 days, respectively, and sex ratio was 0.48. The baits with cartap and methomyl caused 97.92 and 83.33% of adults mortality, respectively, 24 hours after exposure to the treatments. This is a promising way to control this pest and it may be used in rotation. Cartap without attractive caused 91.67% of adults mortality. The caterpillars damage the silks and beginning of grain development or grains at the milk stage through little entry orifices, damaging the endosperm and, more importantly, the embryo, which disenables them for seed. Therefore, it is difficult to visualize the damage, which is unnoticed without an accurate examination. These results can support the establishment of strategies to manage this new corn ear pest, especially in seeds field.

It was first described in 1979 in the State of Maryland, in the vicinity of the Potomac River, United States. It is a disease of great importance in the creation of horses in the southern Brazil, especially the creole s breed, presenting the diarrhea as the main symptom, which can deteriorate and lead to death or complications such as laminitis. Contamination of animals possibly occurs by the oral route, being related to digenetic trematodes of aquatic environments, which in turn require more than one host to complete its life cycle. The location of the horses in overflowed regions provides an increase in the ingestion of aquatic vectors, making higher the chances of contracting the infection, especially in the months of higher temperatures, when the EME presents its characteristic seasonal peaks of occurrence. The objectives of this study were to investigate the species of Heleobia, the first intermediate host of N. risitcii in an endemic area for EME (rural property in Arroio Grande), in order to estimate its rate of parasite contamination. We evaluated the effect of salinity on the survival of mollusk of Heleobia piscium specie, as well as its reproduction and emission frequency of cercariae. Through the cultivation of trematodes in membrane chorioallantoic of chicken eggs, we sought the adult form of the parasite housed in gastropods. The snails found during the collections were classified as Heleobia piscium, noting that 6,24%, among 962 dissected gastropods presented themselves parasitized. Cercarial output occurred in an environment with variable salinity to 12 , which indicates partially the adaptability of the species in estuarine regions. There was no satisfactory results regarding the cultivation of trematodes in chorioallantoic membrane, possibly due to the high specificity of the technique required. Several parameters have been analyzed and the outcomes represent a starting point for the establishment of a specific methodology for obtaining the adult trematode. The results presented are important for the elucidation of the biological cycle of the parasite analysed in this study, contributing to highlight the epidemiology of EME, serving as the basis for future researches that aim to minimize its impact on the creation of horses in Rio Grande do Sul. / A Neorickettsia risticii é um parasito intracelular obrigatório de leucócitos e agente etiológico da Ehrlichiose Monocítica Equina (EME), conhecida como a Febre do Cavalo de Potomac ( Potomac Horse Fever ). Foi descrita pela primeira vez em 1979 no Estado de Maryland, nos arredores do rio Potomac (EUA). É uma enfermidade de grande importância na criação de cavalos no sul do Brasil, especialmente da raça crioula, apresentando a diarréia como principal sintoma, podendo agravar-se, ocasionando a morte. A contaminação dos animais ocorre possivelmente por via oral, havendo relação com trematódeos digenéticos de ambientes aquáticos que, por sua vez, necessitam de mais de um hospedeiro para completar seu ciclo de vida. Os cavalos presentes em regiões alagadiças podem ingerir vetores aquáticos, o que aumenta as chances de infecção, principalmente nos meses de temperaturas mais elevadas, quando a EME apresenta seus característicos picos sazonais de ocorrência. Os objetivos deste estudo foram investigar as espécies de gastrópodes do gênero Heleobia, primeiro hospedeiro intermediário da N. risitcii, em área endêmica à EME (Propriedade Rural em Arroio Grande), estimar a taxa de contaminação parasitária e o efeito da salinidade na sobrevivência dos moluscos da espécie Heleobia piscium, bem como, sua reprodução e frequência de emissão de cercárias. Através do cultivo de trematódeos em membrana corioalantóide de ovos de galinha, buscou-se a forma adulta do parasita albergado nos gastrópodes. Os moluscos encontrados durante as coletas foram classificados como Heleobia piscium, constatando-se que 6,24%, dentre os 962 gastrópodes dissecados apresentaram-se parasitados. Ocorreu emissão de cercárias em local com salinidade variável até 12 , o que indica parcialmente a adaptabilidade da espécie a ambientes estuarinos. Não houve resultados satisfatórios quanto ao cultivo de trematódeos em Membrana corioalantóide, possivelmente devido à alta especificidade da técnica requerida. O conhecimento resultante representa um ponto de partida para o estabelecimento de uma metodologia específica quanto à obtenção do trematódeo adulto. Os resultados apresentados são importantes para a elucidação do ciclo biológico do parasita em estudo, contribuindo com maiores esclarecimentos sobre a epidemiologia da EME, servindo como base para futuras pesquisas que visem minimizar seu impacto na criação de cavalos no Rio Grande do Sul.

Bioécologie et diversité génétique d'Orthops palus (Heteroptera, Miridae), ravageur du manguier à La Réunion / No English title available

La culture du manguier à La Réunion est confrontée à un cortège de bioagresseurs, dont la punaise Orthops palus (Heteroptera, Miridae). Cette espèce est uniquement signalée en Ouganda et à La Réunion mais n'est considérée comme ravageur du manguier qu'à La Réunion. Peu d'études ont donc été consacrées à cet insecte, ce qui est à l'origine du manque de connaissance sur ce ravageur. L'objectif général de cette thèse est l'acquisition de connaissances sur O. palus. Il se décline en plusieurs objectifs spécifiques. Il s'agit, dans un premier temps, de caractériser la diversité de mirides présentes dans les vergers de manguiers de La Réunion. Dans un deuxième temps, l'objectif est de caractériser le cycle biologique d'O. palus et les interactions insecte-plantes. Enfin, dans un troisième temps, le dernier objectif consiste à étudier la distribution et la diversité génétique de l'espèce à La Réunion et dans la zone sud-ouest de l'océan Indien (SOOI). Les principaux résultats de la thèse sont les suivants. La miridofaune des vergers de manguiers a été étudiée et, parmi les 13 espèces de mirides recensées, O. palus est l'espèce la plus abondante sur les inflorescences du manguier lors de sa floraison. Pour identifier O. palus au laboratoire et la reconnaître sur le terrain, trois outils originaux ont été construits (une clé d'identification, des séquences du Cytochrome c Oxydase I et une fiche de reconnaissance sur le terrain). Un élevage d'O. palus a été mis au point ; il a permis de caractériser son cycle biologique et de mesurer les durées de développement des différents stades. Parallèlement, l'inventaire in situ des plantes hôtes d'O. palus (15 espèces à La Réunion) a montré le caractère polyphage de espèce. Les études sur la diversité et la structuration génétique d'O. palus dans les îles du SOOI ont apporté des résultats importants. À La Réunion, la population d'O. palus est structurée en deux clusters, mais les facteurs structurant n'ont pu être totalement identifiés. À Maurice, une seule population est recensée et demeure différente de celles de La Réunion. Aucune race d'hôte n'a été mise en évidence, ni à La Réunion, ni à Maurice. Des flux de populations, dans les deux sens, ont été mis en évidence entre ces deux îles. Enfin, l'analyse de la diversité de l'ADN mitochondrial sur des individus prélevés sur quatre îles du SOOI (La Réunion, Maurice, Mayotte, Grande Comore) montre la présence d'au moins sept haplotypes dont la distribution confirme les échanges inter-îles. La dynamique spatiale et temporelle de l'insecte en relation avec sa stratégie de passage de l'année ainsi que les flux de populations à l'échelle du SOOI font l'objet de discussions transversales. Les résultats montrent qu'au-delà d'être une punaise s'attaquant uniquement au manguier, comme son nom le suggérait jusqu'à présent, O. palus est surtout une "Punaise des fleurs", susceptible de se maintenir toute l'année en se déplaçant, selon la disponibilité des ressources alimentaires, de plante en fleur à plante en fleur. Parmi les plantes participant à sa dynamique annuelle, plusieurs sont des plantes d'intérêt économique (letchi, jujubier, avocatier). Diverses perspectives de recherche sont proposées pour compléter les connaissances déjà acquises sur O. palus, notamment la compréhension des processus de dispersion des populations à l'échelle de l'agroécosystème, la vérification de la présence d'O. palus dans d'autres territoires du SOOI et l'étude de sa diversité génétique dans cette même région. Enfin, à la lumière des résultats acquis dans la thèse, des propositions de gestion agroécologique des populations d'O. palus à l'échelle locale, ainsi que des recommandations sur les mesures de précaution à prendre pour éviter les flux de populations entre les pays de l'océan Indien sont faites. / Mango crops in Reunion Island are threatened by a series of pests, including the bug Orthops palus (Heteroptera, Miridae). This species has only been reported in Uganda and Reunion Island but is considered pest of mango only in Reunion Island. Few studies have therefore been devoted to this bug, which explains the lack of knowledge concerning this pest. The overall aim of my thesis was to acquire knowledge about O. palus, and several specific objectives were pursued. The first was to characterize mirid diversity in the mango orchards of Reunion Island. The second was to characterize the life cycle of O. palus and insect-plant interactions. The third objective was to study the distribution and genetic diversity of the species in Reunion Island and on other islands in the south-west Indian Ocean (SWIO).The main results of these studies are: among the 13 species identified in the community of mirids in mango orchards, O. palus was the most abundant on the mango inflorescences when in flower. Three original tools were built to identify O. palus in the laboratory and to recognize the species in the field: an identification key, COI sequences, and a field recognition card). A method was developed to rear O. palus, which enabled the characterization of its life cycle and the length of the development stages. Meanwhile, the inventory of in situ host plants (15 species in Reunion Island) showed that O. palus is polyphagous. Studies of the diversity and genetic structure of O. palus in SWIO islands also provided significant results. In Reunion Island, the population is structured in two clusters, although the structural factors could not be fully identified. In Mauritius, one cluster was identified and was shown to be different from the clusters in Reunion Island. No host race was found either in Reunion Island or in Mauritius. Population movements in both directions between the two islands were highlighted. Finally, the analysis of the mitochondrial DNA diversity of individuals sampled on four SWIO islands (Reunion, Mauritius, Mayotte, Grande Comore) revealed the presence of at least seven haplotypes whose distribution confirmed exchanges between islands.The dynamics of O. palus in space and over time are discussed in connection with its survival strategy over the course of the year and with population flows across SWIO. The results showed that beyond being a bug that attacks only the mango tree, as the name strongly suggests, O. palus is rather a "flower bug" likely to maintain populations throughout the year by moving from one flowering plant to another according to food availability. Among the plants involved in the annual dynamics of the insect, several species beside mango are of economic interest (lychee, jujube, avocado). Several research topics are proposed to complete the knowledge already acquired on O. palus, including understanding of population dispersion processes at the scale of the agro-ecosystem, and evaluating the presence and the genetic diversity of O. palus in other SWOI territories. Finally, agroecological practices for the local management of O. palus populations are proposed based on the results of the studies that comprise the present dissertation. Recommendations are made for precautionary measures to prevent the flow of O. palus populations between countries located in the Indian Ocean.

Some coccinellids only complete their development and produce viable eggs and progenies when they consume their preferred prey. However, when this prey is scarce or in presence of a prey of low quality, certain coccinellids feed on alternative food resources, such as extra floral nectar and pollen, to guarantee their survivorship. Thus, the absence of these floral resources limits the occurrence and abundance of ladybeetles in the agroecosystems. Due to these characteristics, the coccinellids have great potential for being managed by the three strategies of biological control: classic, augmentative and conservation. However, to use the coccinellids as biological control agent, more information about their biology and ecology are still necessary. In this context, the present work was conducted with the general objective of producing information about ecological and biological aspects of the Coccinellidae. These approaches are in the Chapters I and II, respectively. In the Chapter I, the studies aimed to determine if Anethum graveolens (dill), Coriandrum sativum (coriander) and Foeniculum vulgare (sweet fennel) favor the abundance of Coccinellidae; to evaluate the potential of these aromatic plants to provide vital resources for these predators; to determine the species diversity of these insects attracted by these plants; and to characterize their community structure. The experiment was carried out in an integrated crop-livestock organic production area (Seropedica/RJ, Brazil) in a completely randomized design with three treatments (aromatic plants) and three replicates. From September 14th, 2007 to January 21st, 2008, samplings of adults and immature stages of ladybeetles were realized. Besides an unidentified species of Chilocorini, Coleomegilla maculata, Coleomegilla quadrifasciata, Cycloneda sanguinea, Eriopis connexa, Harmonia axyridis, Olla v-nigrum and Hippodamia convergens were collected. The dill provided significant increase in the abundance of coccinellids in relation to the coriander and sweet fennel. These three aromatic vegetal species were used as ovipositon sites and shelter for immature forms and adults of ladybeetles. The dill and sweet fennel were also used as mating sites and proved food resources (pollen and/or alternative prey). The principal species visiting these aromatic plants were C. sanguinea, H. convergens, and E. connexa (more frequent, constant and dominant). In the Chapter II, the studies aimed to determine biology aspects of Coleomegilla maculata and Eriopis connexa feed on two diets: ultraviolet-unviable and frozen eggs of Anagasta kuehniella (Lepidoptera: Pyralidae) and the alive aphids of Lipaphis erysimi (Hemiptera: Aphididae) under laboratory condition (25 ? 1?C, relative humidity of 70 ? 10%, and photophase of 12 hours). The experimental design was complete randomized in a 2x2 split-plot arrangement (two ladybeetle species and two diets). In general, the biological characteristics of C. maculata and E. connexa were similar, when their larvae and adults fed on the two diets. Both diets were suitable for C. maculata and E. connexa, keeping their development and oviposition for a generation, and the eggs of A. kuehniella still maintained the rearing of these ladybeetles for one more generation. / Entre os inimigos naturais de pragas agr?colas, destacam-se os Coccinellidae predadores. Alguns coccinel?deos s? completam seu desenvolvimento e produzem ovos e prog?nie vi?vel quando consomem sua presa preferencial. Todavia, quando esta presa est? escassa ou na presen?a de uma presa de qualidade inferior, certos coccinel?deos alimentam-se de alimentos alternativos, tais como n?ctar extrafloral e p?len, para garantir sua sobreviv?ncia. Desse modo, a aus?ncia desses recursos florais limita a ocorr?ncia e abund?ncia de joaninhas nos agroecossistemas. Devido essas caracter?sticas, os coccinel?deos apresentam grande potencial para serem manejados por meio do controle biol?gico cl?ssico, aumentativo e conservativo. Todavia, para usar os coccinel?deos como agente de controle biol?gico, mais informa??es sobre sua biologia e ecologia s?o ainda necess?rias. Neste contexto, o presente trabalho foi realizado com o objetivo geral de gerar informa??es sobre aspectos ecol?gicos e biol?gicos dos Coccinellidae, sendo esses temas abordados nos Cap?tulos I e II, respectivamente. No cap?tulo I, os estudos foram conduzidos com os seguintes objetivos: determinar se Anethum graveolens (endro), Coriandrum sativum (coentro) e Foeniculum vulgare (erva-doce) favorecem a abund?ncia de Coccinellidae; avaliar o potencial dessas esp?cies arom?ticas como provedoras de recursos vitais para esses predadores; determinar a diversidade de esp?cies desses insetos atra?dos por essas plantas; e caracterizar a estrutura de sua comunidade. O experimento foi conduzido em ?rea de produ??o org?nica integrada animalvegetal (Serop?dica/RJ), em delineamento de blocos casualizados, com tr?s tratamentos (esp?cies arom?ticas) e tr?s repeti??es. De 14 de setembro de 2007 a 21 de janeiro de 2008, foram realizadas amostragens de adultos e formas imaturas de joaninhas. Al?m de uma esp?cie n?o identificada da tribo Chilocorini, coletou-se Coleomegilla maculata, Coleomegilla quadrifasciata, Cycloneda sanguinea, Eriopis connexa, Harmonia axyridis, Olla v-nigrum e Hippodamia convergens. O endro proporcionou aumento significativo na abund?ncia de coccinel?deos em compara??o ao coentro e ? erva-doce. Essas tr?s esp?cies arom?ticas foram usadas como s?tios de oviposi??o e abrigo para formas imaturas e adultas de joaninhas. O endro e a erva-doce tamb?m foram utilizados como s?tios de acasalamento e de alimenta??o pelas joaninhas. As principais esp?cies visitantes das arom?ticas foram C. sanguinea, H. convergens e E. connexa (mais freq?entes, constantes e dominantes). No cap?tulo II, os estudos foram conduzidos com os seguintes objetivos: 1) determinar aspectos biol?gicos de C. maculata e E. connexa alimentadas com duas dietas: ovos de Anagasta kuehniella (Lepidoptera: Pyralidae) inviabilizados com ultravioleta e congelados e pulg?es vivos de Lipaphis erysimi (Hemiptera: Aphididae), em condi??es de laborat?rio (25?1?C, 70?10% UR e fotofase de 12 horas). O delineamento foi inteiramente casualizado em arranjo fatorial 2 x 2 (duas esp?cies de joaninhas e duas dietas). No geral, as caracter?sticas biol?gicas de C. maculata e E. connexa foram bem semelhantes, quando suas larvas e adultos se alimentaram das duas dietas. Ambas as dietas foram adequadas para C. maculata e E. connexa, assegurando seu desenvolvimento e oviposi??o por uma gera??o e os ovos de A. kuehniella permitiram ainda manter a cria??o dessas joaninhas por mais uma gera??o.
