Logiques sociales de recours aux antidépresseurs : le cas de la rupture conjugale

La dépression et l’antidépresseur, phénomènes plus vastes, plus complexes que l’ordre biomédical ne l’entend, répondent à des logiques qui dépassent les seules considérations organiques, corporelles. Certaines recherches montrent d’ailleurs que ces deux entités sont autonomes l'une de l'autre, signifiant que les antidépresseurs sont prescrits pour des raisons indépendantes de la prévalence de problèmes de santé mentale. C’est dans cette optique que, dans ce mémoire, nous examinons les raisons pour lesquelles les individus ont recours aux antidépresseurs suite à une rupture conjugale. C’est à l’aide des récits de huit femmes ayant pris des antidépresseurs suite à une rupture conjugale qu’on a donc investigué les rouages du recours à ces médicaments. À la lumière des résultats, on observe effectivement que la prise d’antidépresseurs semble répondre à une logique qui soit extra–médicale. D’abord par le constat que seules trois d’entres ces huit femmes ont justifié leur prise d’antidépresseurs par la réception d’un diagnostic formel de dépression. Ensuite, par la découverte qu’elle se déploie essentiellement selon une double symbolique au centre duquel prime le lien social. En effet, on a recours aux antidépresseurs suite à une rupture conjugale parce qu’ils sont à la fois l’incarnation d’un lien de confiance avec un être semblable à soi, empathique, le médecin, et le symbole d’un retour à la vie dite ‘normale’ où les manières d’être et d’interagir sont conformes à celles du groupe auquel on s’identifie. On croit que l’importance accordée à la confiance au médecin et à la normalisation quant au recours à l’antidépresseur témoignent du resserrement des liens d’identification propre à l’individualisme contemporain lesquels, en opposition aux grands récits de jadis, sont désormais axés sur une intimité de valeurs et d’objectifs entre semblables. / Depression and antidepressants are far more complex phenomenons than the biomedical order pretends. Some research show that these two entities are independant from one another, meaning that antidepressants are prescribed for reasons independant of mental health problems. In this master’s thesis we examine why individuals have recourse to antidepressants following a relationship break-up; the relationship (marital or not) break-up being a critical situation associated with the prevalence of depression. Eight women who took antidepressants following a relationship break-up were interviewed for this study. The findings show that the recourse to antidepressants appears to respond to a logic that is non-medical. First, by the fact that only three of the eight women interviewed justified their recourse to antidepressants by receiving a diagnosis of depression. Second, by the discovery that these women relied on antidepressants because they are the embodiment of a relationship of trust with someone familiar to themselves, the doctor, and the symbol of a return to a ‘normal’ life.

Étude de protection conférée par vaccination ADN dans un modèle murin d’hépatite auto-immune

La vaccination ADN à l’aide de plasmides codant pour des autoantigènes s’est avérée efficace dans la protection contre plusieurs maladies auto-immunes. Le but de ce mémoire était dans un premier temps d’établir si un protocole de vaccination ADN composé de 3 injections de pCMV-CTLA-4-NP et de pVR-IL-12 à deux semaines d’intervalle avait un effet protecteur contre le développement d’une hépatite auto-immune chez la souris TTR-NP, un modèle murin transgénique de la maladie et précédemment développé au laboratoire. Dans un deuxième temps, le but était d’élucider, le cas échéant, les mécanismes sous-tendant la protection conférée par la vaccination ADN. Les hypothèses initiales étaient qu’une protection allait effectivement être conférée par la vaccination ADN et que celle-ci pouvait être attribuable à une déviation de la réponse typiquement Th1 de la maladie vers une réponse Th2, à un épuisement des cellules immunitaires et/ou à l’activation et à l’induction de prolifération de cellules régulatrices. Les résultats montrent que la vaccination ADN induit une protection transitoire contre le développement d’infiltrations lymphocytaires au foie. Cette protection se ferait via un épuisement des cellules CD4+, CD8+ et CD19+ se retrouvant à la rate et exprimant PD 1 dans une plus forte proportion à 3 mois, et ne serait médiée ni par les lymphocytes T régulateurs CD4+CD25+FoxP3+, ni par les cellules CD8+FoxP3+. Une déviation de la réponse Th1 vers une réponse Th2 demeure une explication supplémentaire plausible à la protection conférée mais nécessiterait une caractérisation en situation plus physiologique avant de pouvoir inférer sur son implication réelle. La vaccination ADN n’influe ni sur la présence d’autoanticorps, ni sur les niveaux d’alanine aminotransférase, deux marqueurs de la maladie. / DNA immunizations were proven effective in a range of autoimmune diseases. The first goal of this master’s thesis was thus to evaluate if a protocol of 3 intramuscular plasmidic injections of pCMV-CTLA4-NP and pVR-IL-12, at 2 weeks apart, would protect TTR-NP mice, a transgenic murine model previously developed in the laboratory, against the development of an autoimmune hepatitis. The second goal was, if the protection was indeed conferred by DNA vaccination, to elucidate the mechanisms underlying this effect. The initial hypotheses were that DNA vaccination would indeed elicit a protection against autoimmune hepatitis, and that this could be attributed to a skew from the typical Th1 response to a more tolerogenic Th2 response, to the exhaustion of the immune cells and/or to the activation and the proliferation of regulatory T cells. Results show that DNA vaccination induces a transient protection against lymphocytic infiltrates in the liver. This protection is thought to be caused by the exhaustion of the CD4+, CD8+ and CD19+ cells found in the spleen and expressing PD-1 in greater proportion at 3 months, and not to be mediated by CD4+CD25+FoxP3+ or CD8+FoxP3+ regulatory T cells. An immunomodulation from a Th1 to a Th2 response could still be a plausible explanation by the protection conferred by the DNA vaccination, but additional experimentations in a more physiological setting would be necessary to infer on its real implication. DNA vaccination had no effect on autoantibodies or on alanine aminotransferase levels, which are two biological markers of the disease.

Economic viability of a floating gas-to-liquids (GTL) plant / Michael Etim Bassey

Today, a large proportion of the world's plenteous offshore natural gas resource are stranded, flared or re-injected due to constraints pertaining to its utilisation. The major constraint in the utilisation of this resource is linked to its properties, which makes it difficult to transport or store. Although the resource presents an excellent opportunity for the Gas-to-Liquid (GTL) technology (process for converting natural gas into high energy liquid fuels with qualities that surpass the most stringent current and future clean-fuel requirements), the further processing of this resource is still impeded by high cost of transportation. However, it is believed that the emerging Floating GTL concept could offer superb opportunities to bring such offshore stranded natural gas reserves to markets by converting the gas into high quality liquid fuels, at the production sites, before it is transported using conventional oil tankers or vessels. But the question is: can this venture be profitable or economically viable? In response, an Economic Model (the EV Model) to review the economic viability of the Floating GTL option was developed. Analyses on technical and economical aspects of the floating GTL application offshore are presented with case studies on Syntroleum's and Statoil's floating GTL designs. Profitability analyses were conducted using the EV model to evaluate economic parameters such as Net Present Value (NPV), Internal Rate of Return (IRR), Discounted PayBack Period (DPBP), Profitability index (PI), Break-Even Analysis (BEA) and Scale Economies for some assumed case scenarios involving both designs. In addition, sensitivity analyses were also carried out to find the most sensitive parameters which affect the viability of the floating GTL option. The economic analyses revealed that, a modest feedstock cost (~0 - $3/MSCF), high crude oil price (that stays above $30 per barrel) and reduction trend in capital expenditure (for stand alone Floating GTL plant) up to $20,00O/BPD or lower in the next few years, will open windows for the floating GTL concept. Finally, the energy policy needed to achieve the capitalisation of the plenteous offshore stranded gas resource via floating GTL is also discussed. / Thesis (M.Ing. Oesophageal Cancer – Novel Targets for Therapy : With focus on Hsp90, EGFR, LRIG, microtubule and telomerase

Oesophageal cancer is a malignant and aggressive disease with very poor survival. The aim of this thesis was to evaluate novel therapeutic targets in oesophageal cancer. In paper I, Hsp90 was expressed in all 81 oesophageal cancer tissues and also in nine oesophageal cancer cell lines. A specific Hsp90 inhibitor, 17-AAG, could efficiently inhibit cell proliferation, cell survival and sensitise oesophageal cancer cells to gamma photon irradiation. By inhibition of Hsp90 using 17-AAG, EGFR- and IGF-1R-mediated signalling was downregulated. In paper II, tumour samples from 80 oesophageal cancer patients were investigated for the expression of EGFR and LRIG1-3. Based on a total score of intensity and expression fraction a trend towards survival differences was found for LRIG2 (p=0.18) and EGFR (p=0.09). Correlation analysis revealed a correlation between expression of EGFR and LRIG3 (p=0.0007). Significant correlations were found between LRIG1 mRNA expression levels and sensitivity to cisplatin (r = –0.74), docetaxel (r = –0.69), and vinorelbine (r = –0.82). In paper III, microtubule targeting drugs podophyllotoxin (PPT), vincristine and docetaxel inhibited survival and proliferation of oesophageal cancer cells. Unexpectedly, experiments showed that microtubule destabilising agents inhibited EGFR phosphorylation and signalling. A tyrosine phosphatase inhibitor, sodium orthovanadate, was able to reverse the EGFR dephosphorylation. In paper IV, imetelstat, a telomerase antagonist, inhibited telomerase activity, colony formation ability and decreased proliferation of oesophageal cancer cells. Inhibition of telomerase activity by imetelstat led to an increase of 53BP1 foci indicating induction of DSBs. Furthermore, the fraction and size of radiation-induced 53BP1 foci were increased by imetelstat pre-treatment. In conclusion, Hsp90 and telomerase represent potential therapeutic targets in oesophageal cancer. And, the implication of EGFR and LRIG as prognostic factors is limited. Furthermore, disruption of the microtubule network may activate a protein tyrosine phosphatase that can regulate EGFR phosphorylation.

La Crisis ambiental como proceso. Un análisis reflexivo sobre su emergencia, desarrollo y profundización desde la perspectiva de la teoría crítica

La investigación se focaliza en el análisis de la crisis ambiental considerada como un fenómeno complejo de evolución incierta producto de un proceso socio histórico, cuyos orígenes se remontan a los albores del proyecto moderno y deben rastrearse en un cúmulo de circunstancias que expresan las contradicciones de un modelo socioeconómico identificado con el crecimiento sostenido de la producción y las magnitudes económicas. Partiendo de esta base, se propone abordar la relación entre saber sociológico y crisis ambiental, a partir de las contribuciones de la teoría crítica a la compresión de las causas profundas de la crisis ambiental y de sus elementos constitutivos. Para lo cual se propone profundizar en algunas categorías desarrolladas por la corriente social crítica, las cuales si bien no se refieren en forma directa a la problemática ambiental, se vislumbran como campos fértiles para comprender los procesos sociales generadores de la crisis ambiental así como las alternativas que han de adoptarse para su resolución, teniendo en cuenta los mecanismos que ejerce el sistema para neutralizar los cambios que implica la conformación de una racionalidad alternativa. Acorde con ello, la primera parte del trabajo se centra en el abordaje de la relación sociedad naturaleza en el marco de tres grandes procesos sociales identificados como cosmológico, teocéntrico y antropocéntrico, haciendo especial hincapié en la correlación que en cada una de las etapas consideradas se establece entre racionalidad dominante, producción de conocimiento y conceptualización de naturaleza. El segundo capítulo profundiza en los efectos de la racionalidad instrumental sobre la ruptura del proyecto moderno y su incidencia sobre la emergencia de la crisis ambiental, a partir de la posición adoptada por Heidegger y por los referentes de la Escuela de Frankfurt. Posteriormente el análisis se estructura en torno a las dimensiones críticas de la modernidad avanzada, considerada como contexto situacional de desarrollo y profundización de la crisis ambiental, teniendo en cuenta los aportes teóricos desarrollados por Zygmunt Bauman e Immanuel Wallerstein.Al finalizar se presentan las conclusiones generales de la tesis y las posibles líneas de investigación que se abren a partir del trabajo desarrollado. / This investigation focuses on the analysis of the environmental crisis, considering it as a complex phenomenon whose evolution is uncertain, as it is the outcome of a socio-historical process that goes back to the beginning of modernity and that must be traced to the circumstances that express the contradictions of a socio-economic model identified with the sustained growth of production and the economy. From this perspective, I approach the relationship between sociological knowledge and environmental crisis by looking at the contributions of critical theory to the understanding of the deep causes of both the environmental crisis and its constitutive elements. I seek to analyse some categories developed by critical social studies. Even though they do not refer directly to the environmental problematics, it is possible to find in them both a fertile field from which to understand the social situations generated by the environmental crisis and alternative policies that ought to be adopted for its resolution, considering at the same time the mechanisms that the system sets in place to neutralize the changes implied by the formation of an alternative rationality.Accordingly, the first part of this work centres on the study of the relationship between society and nature across three large social processes identified as cosmology, theocentrism and anthropocentrism, emphasizing the correlation, in each of the stages considered, between dominant rationality, knowledge production, and conceptualization of nature.The second chapter deals with the effects of instrumental rationality on the breakdown of the project of modernity and its impact upon the emergency of the environmental crisis. Here I consider the position adopted by Heidegger and by the best known figures of the Frankfurt School.Later, I structure the analysis around the critical dimensions of advanced modernity, considering it as the situational context for the development and deepening of the environmental crisis. Here I take into account the theoretical contributions developed by Zygmunt Bauman and Immanuel Wallerstein.Finally, I present the general conclusions of my thesis and the possible lines of research that open up from my work.

Towards reliable contacts of molecular electronic devices to gold electrodes

PhD / SYNOPSIS OF THIS THESIS The aim of this thesis is to more fully understand and explain the binding mechanism of organic molecules to the Au(111) surface and to explore the conduction of such molecules. It consists of five discreet chapters connected to each other by the central theme of “The Single Molecule Device: Conductance and Binding”. There is a deliberate concentration on azine linkers, in particular those with a 1,10-phenanthroline-type bidentate configuration at each end. This linker unit is called a “molecular alligator clip” and is investigated as an alternative to the thiol linker unit more commonly used. Chapter 1 places the work in the broad context of Molecular Electronics and establishes the need for this research. In Chapter 2 the multiple break-junction technique (using a Scanning Tunnelling Microscope or similar device) was used to investigate the conductance of various molecules with azine linkers. A major finding of those experiments is that solvent interactions are a key factor in the conductance signal of particular molecules. Some solvents interfere with the molecule’s interaction with and attachment to the gold electrodes. One indicator of the degree of this interference is the extent of the enhancement or otherwise of the gold quantized conduction peak at 1.0 G0. Below 1.0 G0 a broad range for which the molecule enhances conduction indicates that solvent interactions contribute to a variety of structures which could bridge the electrodes, each with their own specific conductance value. The use of histograms with a Log10 scale for conductance proved useful for observing broad range features. vi Another factor which affects the conductance signal is the geometric alignment of the molecule (or the molecule-solvent structure) to the gold electrode, and the molecular alignment is explored in Chapters 3 for 1,10-phenanthroline (PHEN) and Chapter 4 for thiols. In Chapter 3 STM images, electrochemistry, and Density Functional Theory (DFT) are used to determine 1,10-phenanthroline (PHEN) structures on the Au(111) surface. It is established that PHEN binds in two modes, a physisorbed state and a chemisorbed state. The chemisorbed state is more stable and involves the extraction of gold from the bulk to form adatom-PHEN entities which are highly mobile on the gold surface. Surface pitting is viewed as evidential of the formation of the adatom-molecule entities. DFT calculations in this chapter were performed by Ante Bilic and Jeffery Reimers. The conclusions to Chapter 3 implicate the adatom as a binding mode of thiols to gold and this is explored in Chapter 4 by a timely review of nascent research in the field. The adatom motif is identified as the major binding structure for thiol terminated molecules to gold, using the explanation of surface pitting in Chapter 3 as major evidence and substantiated by emergent literature, both experimental and theoretical. Furthermore, the effect of this binding mode on conductance is explored and structures relevant to the break-junction experiment of Chapter 2 are identified and their conductance values compared. Finally, as a result of researching extensive reports of molecular conductance values, and having attempted the same, a simple method for predicting the conductance of single molecules is presented based upon the tunneling conductance formula.

Tree training and managing complexity and yield in fig (Ficus carica L.)

Thesis (MscAgric (Horticulture))--University of Stellenbosch, 2010. / ENGLISH ABSTRACT: Commercial fig production with popular European cultivars, Bourjasotte Noire, Col de Damme Noire and Noire de Caromb, is new to the Western Cape. Little research on fig production has been conducted in South Africa and producers are struggling to implement effective commercial practices. In order to establish practices that will maximise yield of quality fruit, the most productive one-year-old shoot lengths were identified in a phenological study. All shoot length categories evaluated in ‘Bourjasotte Noire’ (10 – 15 cm, 25 – 40 cm, 50 – 65 cm, 75+ cm) yielded fruit and will probably yield well the following season. In ‘Col de Damme Noire’, shoots longer than 60 cm seem to be suited to reproduction, yet they might produce a poor yield the following season. Shoots 10 – 20 cm long in ‘Noire de Caromb’ are productive relative to their length, while shoots 30 – 50 cm and 60 – 80 cm long are also fairly productive. Shoots longer than 100 cm produced suitable shoot lengths for yield the following season. Two experiments were conducted on ‘Bourjasotte Noire’ and ‘Col de Damme Noire’ to establish the type (Experiment 1, different intensity heading cuts) and timing (Experiment 2) of pruning cuts required to stimulate the growth of shoots of the same length as the shoots selected to be optimal for yield in the phenology study, and to reduce the expression of distal branching. In ‘Bourjasotte Noire’, removing one third of the total length of one-year-old shoots on 21 July by heading stimulated the development of more growth and longer current season shoots compared to other treatments, while reducing yield slightly. Heading back to three nodes in ‘Col de Damme Noire’ stimulated the growth of current season shoots of the optimal length established in the phenology study, while heading cuts on 30 June produced the longest average current season shoot length in ‘Col de Damme Noire’. To further address the effects of distal branching (acrotony), an experiment was conducted to establish whether rest breaking agents (RBA’s) in combination with tip-pruning can increase tree complexity by improving bud break, and whether increased complexity would increase yield in all three cultivars. A second experiment was conducted to evaluate the effects of timing RBA applications on bud break and harvest scheduling. Lift® increased the number of buds breaking in ‘Bourjasotte Noire’, while in ‘Noire de Caromb’ Dormex® and oil increased bud break. Tip-pruning increased the average shoot length in ‘Bourjasotte Noire’ and ‘Col de Damme Noire’, while causing a reduction in the amount of new growth in ‘Noire de Caromb’. Lift® applied 3 August and Dormex® applied 30 June shortened the number of days to 50% bud break in ‘Bourjasotte Noire’ and ‘Col de Damme Noire’. Both Lift® and Dormex® applied on 30 June decreased the number of days to 50% bud break and 50% harvest of the breba crop in ‘Noire de Caromb’. These treatments increased the number of fruit in both the breba and main crop of ‘Noire de Caromb’, but reduced fruit size. In conclusion, different approaches with regards to pruning needs to be followed for each cultivar to establish or maintain the optimal shoots for reproduction, while RBA’s can be used to force earlier, increased- bud break and harvest of breba fruit. / AFRIKAANSE OPSOMMING: Kommersiele verbouing van drie Europese vykultivars, Bourjasotte Noire, Col de Damme Noire en Noire de Caromb is nuut tot die Wes-Kaap. Baie min navorsing oor die verbouing van vye is al in Suid-Afrika gedoen, met die gevolg dat produsente sukkel om effektiewe kommersiële verbouingspraktyke te implementeer. ‘n Fenologiese studie van die drie kultivars is uitgevoer om vas te stel wat die mees produktiewe een-jaar-oue lootlengte is, met die doel om die opbrengs van kwaliteit vrugte te maksimeer. Al vier kategorieë wat ge-evalueer is in ‘Bourjasotte Noire’ (10 – 15 cm, 25 – 40 cm, 50 – 65 cm, 75+ cm) is gevind om geskik te wees vir huidige en volgende seisoen opbrengs. Lote langer as 60 cm is geskik vir opbrengs in ‘Col de Damme Noire’ in die huidige seisoen, maar mag in die volgende seisoen swak presteer a.g.v. die gebrekkige lengte van nuwe lote wat daarop ontwikkel. In ‘Noire de Caromb’ is gevind dat lote 10 – 20 cm lank baie produktief is relatief tot hul lengte en dat lote 20 – 50 cm en 60 – 80 cm lank ook relatief produktief is. Lote langer as 100 cm was minder produktief, maar het nuwe lote gelewer wat geskik is vir opbrengs die volgende seisoen. Twee snoei eskperimente is uitgevoer op ‘Bourjasotte Noire’ en ‘Col de Damme Noire’ om vas te stel wat die mees geskikte tipe snoeisnit (Eksperiment 1, verskillende dieptes van topsnitte) en tydstip om te snoei (Eksperiment 2) is met die doel om lote te produseer soortgelyk in lengte aan die wat in die fenologie studie uitgewys is as die produktiefste, en om moontlik die voorkoms van “kaalnekke” te verminder. In ‘Bourjasotte Noire’ is gevind dat die wegsnoei van ‘n derde van die loot op 21 Julie aanleiding gee tot meer groei, langer een-jaar-oue lote en ‘n effense afname in opbrengs. Geskikte lote langer as 60 cm kan in ‘Col de Damme Noire’ verkry word deur lote te top sodat net drie nodes oorbly. Die uitvoer van topsnitte op 30 Junie het langer gemiddelde lootlengtes tot gevolg gehad. Om die probleem van “kaalnekke” (apikale dominansie) verder aan te spreek, is ‘n eksperiment uitgevoer om vas te stel of rusbreekmiddels gekombineerd met tip-snoei gebruik kan word om kompleksiteit te vermeerder deur knopbreek te verhoog, en indien wel, of dit sal aanleiding gee tot verhoogde opbrengs in al drie kultivars. ‘n Tweede eksperiment met verskillende toedieningstye van rusbreekmiddels is uitgevoer om vas te stel of oeste geskeduleer kan word. Lift® het knopbreek verhoog in ‘Bourjasotte Noire’, terwyl Dormex® en olie knopbreek verhoog het in ‘Noire de Caromb’. Tip-snoei het die gemiddelde lootlengtes verhoog in ‘Bourjasotte Noire’ en ‘Col de Damme Noire’, terwyl dit groei verminder het in ‘Noire de Caromb’.
