Nouveaux hydrogels à base de polysaccharide obtenus par voie biomimétique ou par photoréticulation. / New hydrogels based on polysaccharide obtained by biomimetics or UV crosslinking

Dans un contexte de démarche écoresponsable et pour répondre aux exigences de biocompatibilité notamment dans les applications cosmétiques et biomédicales, nous avons développé de nouveaux hydrogels à base de polysaccharides neutres et anioniques en utilisant deux voies originales. La 1ère approche est biomimétique et a consisté à mimer un phénomène d’élaboration naturelle d’hydrogels que l’on retrouve chez certains végétaux pour lesquels une enzyme, la laccase, permet de créer des liens de réticulation par dimérisation des composés phénoliques (en l’occurrence de l’acide férulique FA) présents sur les arabinoxylanes des mucilages des graines de céréales par exemple. Notre travail a ainsi consisté à greffer de l’acide férulique via deux chimies différentes de type imidazole et carbodiimide respectivement pour des polysaccharides neutres ou anioniques. Nous avons ainsi fonctionnalisé trois polysaccharides : le pullulane ou PUL (neutre modèle), le carboxyméthylpullulane ou CMP (anionique modèle) et l’acide hyaluronique ou HA (anionique d’intérêt). Des taux de greffage compris entre 2 et 25% ont été obtenus. L’étude physicochimique en régimes dilué et semi-dilué a permis de mettre en évidence un comportement associatif lié au caractère amphiphile des polysaccharides fonctionnalisés. La réticulation en présence de laccase, suivie in situ en rhéologie, a été réalisée avec succès sur les différents systèmes envisagés avec des contrôles possibles de la cinétique, des propriétés mécaniques finales ou encore du gonflement des hydrogels en fonction du caractère neutre ou chargé des polysaccharides, du degré de substitution en acide férulique, de la concentration en polymère ou de l’activité enzymatique fixée. Les dérivés synthétisés ont globalement démontré des activités biologiques (antioxydante et cytocompatible) intéressantes. La deuxième approche repose sur la photoréticulation possible de polysaccharides (PUL, CMP et HA) fonctionnalisés par le greffage d’amine/acide gras mono ou polyinsaturé (oleylamine, acide oléique et linoléique) via la chimie des imidazoles. Si le pullulane modifié par l’acide linoléique à 2% s’est avéré non hydrosoluble en raison de son caractère neutre, tous les autres dérivés avec des taux de greffages de 3 et 10% ont démontré une bonne solubilité dans l’eau. Les études physicochimiques mettent en évidence un très fort caractère associatif de ces dérivés amphiphiles avec la formation de gels physiques en régime semi-dilué. La photoréticulation a été démontrée en rhéologie sous irradiation UV in situ en présence d’un photoamorceur de type Darocur 1173®. Les résultats préliminaires obtenus selon cette approche en photoréticulation ouvrent ainsi des perspectives intéressantes. / In the framework of an eco-responsible context and to take advantage of biocompatibility, notably in cosmetic and biomedical applications, we have developed new hydrogels based on neutral and anionic polysaccharides using two original routes. The first approach is biomimetic and consists of mimicking a natural development of hydrogels that is found in certain plants for which an enzyme, laccase, allows to create crosslinks by dimerization of phenolic compounds, in occurrence of ferulic acid (FA) present on arabinoxylans mucilage of cereal seeds for example. Thus, our work consisted in grafting ferulic acid via two different chemical ways that means imidazole and carbodiimide respectively for neutral or anionic polysaccharides. We functionalized three polysaccharides: pullulan or PUL (neutral model), carboxymethylpullulane or CMP (model anionic) and hyaluronic acid or HA (anionic of interest) with grafting rates of between 2 and 25%. The physicochemical study in diluted and semi-diluted regimes evidenced an associative behavior due to the amphiphilic character of the functionalized polysaccharides. The crosslinking in the presence of laccase, followed in situ thanks to rheology, has been successfully performed on the various envisaged systems with possible controls of kinetics, the final mechanical properties or the swelling of the hydrogels as a function of the neutral or charged nature of the polysaccharides, the degree of substitution in FA, the polymer concentration or the enzymatic activity. The synthesized derivatives have generally demonstrated interesting biological activities (antioxidant and cytocompatibility). The second approach is based on the possible photocrosslinking of polysaccharides (PUL, CMP and HA) functionalized by the grafting of mono or polyunsaturated fatty amine/acid (oleylamine, oleic acid and linoleic acid) via imidazole chemistry. If pullulan grafted with 2% of linoleic acid was found to be water-insoluble due to its neutral character, all other derivatives (i.e. anionic ones) with grafting rates of 3 and 10% showed good solubility in water. The physicochemical studies show a very strong associative character of these amphiphilic derivatives with the formation of physical gels in semi-diluted regime. Photocrosslinking has been demonstrated in situ thanks to rheology/UV irradiation in the presence of a Darocur 1173® photoinitiator. The preliminary results according to this photocrosslinking approach thus open interesting perspectives.

Estudo clínico randomizado triplo cego do efeito da carbodiimida (EDC) no desempenho de restaurações em resina composta /

Orientador: Josimeri Hebling / Resumo: Objetivo: Avaliar o desempenho clínico de restaurações de resina composta em lesões cervicais não cariosas (LCNC) realizadas com ou sem a aplicação de um agente de ligação cruzada (carbodiimida, EDC) durante o período de 12 meses. Métodos: 21 pacientes com no mínimo duas LCNC participaram deste estudo. Um total de 142 LCNC foram distribuídas randomicamente em dois grupos (n=71 cada): controle e experimental. Todas as LCNC foram restauradas pelo mesmo operador e com os mesmos materiais (sistema adesivo Adper Single Bond 2 e resina composta Z350), sendo que, no grupo experimental a dentina recebeu a aplicação de EDC 0,5 mol/L por 60 segundos após o condicionamento ácido e no grupo controle apenas PBS. As restaurações foram avaliadas imediatamente após sua confecção (baseline) e após uma semana, 6 e 12 meses pelo critério USPHS/Ryge por apenas um avaliador calibrado. Os escores obtidos a partir da avaliação clínica, considerando-se os fatores de confundimento, foram analisados pelos testes de Woolf e Cochran-Mantel-Haenzel (p<0,05). Resultados: As taxas de falhas acumuladas na retenção das restaurações nos períodos 7 dias, 6 e 12 meses, foram de 0%; 3,0% e 4,5%, respectivamente, para o grupo controle e de 0%; 3,0% e 7,4% para o grupo experimental (EDC). Não houve diferença estatisticamente significante entre os dois grupos para todos os critérios e períodos avaliados. A taxa de retorno dos pacientes nos períodos de 7 dias, 6 e 12 meses foi de 100%, 90,5% e 85,7%, respectivamente... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: o evaluate the clinical performance of composite restorations placed in noncarious cervical lesions (NCCL) performed with or without the application of a cross-linking agent, over a period of 12 months. Methods: 21 patients with at least two NCCL, participated in this study. A total of 142 NCCL were randomly allocated into two groups (n=71): control and experimental. All NCCL were restored by the same operator and with the same materials (Single Bond 2 and Z350). In the experimental group the etched dentin was exposed to 0.5 mol/L EDC for 60 seconds while in the control group the etched dentin was exposed to Phosphate buffer solution (PBS). The restorations were evaluated at baseline, 7 days, 6 and 12 months using the USPHS/Ryge criteria, by one blinded calibrated examiner. The scores from the clinical evaluation were submitted to Woolf and CochranMantel-Haenzel tests, considering the confounding factors (p<0.05). Results: The rates of accumulated failures in the retention of restorations in the periods 7 days, 6 and 12 months were 0%; 3,0% and 4.5%, respectively, for the control group, and 0%; 3,0% and 7.4% for the experimental group. There was no statistically significant difference between the two groups for all criteria and periods. The return rate of patients in the 7-day, 6- and 12-month periods was 100%, 90.5% and 85.7%, respectively. Molekulare Silylcarbodiimide: Synthese, Struktur und Reaktivität

Im Rahmen der vorliegenden Arbeit konnte die Vielfalt der Anwendbarkeit eines Pseudowasser-Konzeptes für Silylcarbodiimide erneut aufgezeigt und darüber hinaus deutlich erweitert werden. So waren über Umsetzungen des Pseudowassers Bis(trimethylsilyl)carbodiimid (BTSC) mit Dichlorsiliciumverbindungen R2SiCl2 nicht die monomeren Zielverbindungen vom Typ R2Si(NCNSiMe3)2 zugänglich, da diese – in Analogie zu Silandiolen – unter Abspaltung von BTSC zu Pseudo-Polysiloxanen [R2Si-NCN-]n kondensieren. Aus einem der resultierenden komplexen Reaktionsgemische wurde das cyclische Oligomer [Ph2Si-NCN-]4 sowie durch verwandte Reaktionen das Oxo-Analogon [Ph2Si-O-SiPh2-NCN]2 in kristalliner Form erhalten. Umsetzungen von BTSC mit AlCl3 führten zu den neuartigen Pseudohydraten Me3SiNCN(SiMe3)(AlCl3) und Al[NCN(SiMe3)2]3[NCN(SiMe3)(AlCl3)]3, wobei in letzterem erstmals ein präparativer Nachweis für die Relevanz der Cyanamid-Form des BTSC ((Me3Si)2N-C≡N) erbracht werden konnte.

Étude de semi-conducteurs de type p nanostructurés à base de métaux de transition pour une application en DSSC-p / Study of nanostructured p-type semiconductors based on transition metals for p-DSSC applications

Dans le but d'améliorer le rendement des cellules à colorant de type p (DSSC-p), ces travaux s'attachent à la synthèse et la caractérisation de matériaux semi-conducteurs de type p (SCs-p) sous forme de nanoparticules. En ce sens, des SCs-p répondant à un cahier des charges (bande de valence basse en énergie, grande surface spécifique, bon conducteur et bonne transparence) ont été étudiés. Dans ce cadre, une stratégie a été développée pour améliorer les propriétés de NiO (l'actuel matériau de référence) en optimisant sa nanostructuration, sa forte non-stœchiométrie en nickel et par son dopage à l'azote, paramètres tous favorables à la stabilisation de la valence mixte Ni3+/Ni2+, origine de la conductivité de type p. Cette longue étude a été initiée à partir d'un précurseur de nickel original nanostructuré Ni3O2(OH)4, à forte valence mixte Ni3+/Ni2+. La décomposition sous air et sous ammoniac de ce précurseur à basse température (250 °C) a permis de préparer Ni1-xO nanostructuré, fortement non-stœchiométrique (VNi = 25 %), de grande surface spécifique (240 m2.g-1) et dopé azote (NiO:N). De plus, deux matériaux non oxydes à structure delafossite, que sont les carbodiimides de nickel (NiNCN) et de manganèse (MnNCN) ont été préparés et caractérisés comme de nouveaux semi-conducteurs de type p, permettant de monter la première DSSC-p à base de NiNCN. / To improve the performances of p-Dye Sensitized Solar Cell (p-DSSC), this thesis work focuses on the synthesis and the characterization of p-type semiconductors (p-SCs) nanomaterials. These p-SCs with some specifications (low energy valence band, high specific surface area, high conductivity and transparency) were thoroughly studied. In this context, a strategy was developed to improve the NiO nanoparticles properties (commonly used as a reference) with higher nickel non-stoichiometry and nitrogen doping to promote the stabilization of the Ni3+/Ni2+ mixed valence (origin of the p-typness). This study was initiated with a nanostructured mixed valent Ni3O2(OH)4 precursor. Its thermal decomposition in air and ammonia at low temperature (250 °C) allows the formation of nanostructured Ni1-xO with a large amount of Ni vacancies (VNi = 25 %), a high specific surface area (240 m2.g-1) and a nitrogen doping (NiO:N). Moreover, two non-oxides materials with delafossite structure type, namely - nickel carbodiimide (NiNCN) and manganese carbodiimide (MnNCN) - were prepared and characterized as new p-type semiconductors. Thus, the first p-DSSC with NiNCN material was built with success.

Chemical Derivatization in Combination with Liquid Chromatography Tandem Mass Spectrometry for Detection and Structural Investigation of Glucuronides

<p>This thesis presents novel approaches for structural investigation of glucuronides using chemical derivatization in combination with liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS<sup>n</sup>).</p><p>Today, LC-ESI-MS<sup>n</sup> is the dominant technique for quantitative as well as qualitative analyses of metabolites, due to its high sensitivity and selectivity. However, for compounds without an easily ionizable group, e.g., steroids, the sensitivity is limited. In the work presented in this thesis, a derivatization procedure forming a basic oxime significantly increased the detection sensitivity for the altrenogest glucuronide. </p><p>Furthermore, in structural evaluations of glucuronides, the limitation of LC-MS<sup>n</sup> becomes evident due to the initial neutral loss of 176 u, i.e. monodehydrated glucuronic acid, which often makes it impossible to elucidate the structures of the conjugates. To solve this problem, the main part of the work described in this thesis was devoted to chemical derivatization as a means of facilitating the determination of the site of conjugation. </p><p>For the first time, the isomeric estriol glucuronides were evaluated using a combination of three reagents 2-chloro-1-methylpyridinium iodide (CMPI), 1-ethyl-3-(3-dimethyl- aminopropyl)-carbodiimide (EDC), and 2-picolylamine (PA). Interestingly, the derivatization gave a selective fragmentation pattern leading to differentiation of the isomers. </p><p>Another derivatization reagent, 1,2-dimethylimidazole-4-sulfonyl chloride (DMISC), was also tested for the first time in structural investigations. The isomeric glucuronides of morphine, formoterol, and hydroxypropranolol were evaluated. They can all be conjugated in aliphatic as well as aromatic positions. DMISC was proven to be useful in two ways. Firstly, the morphine and formoterol glucuronides that contained a free phenol could be differentiated from those that were conjugated in the aromatic position based on different reactivity. Secondly, for the aromatic <i>O</i>-glucuronide of 4’-hydroxypropranolol, DMISC was proven to react with the amine. This product gave a different fragmentation pattern compared to the corresponding derivative of the aliphatic glucuronide. </p>

Optimizing Engineered Tendon Development via Structural and Chemical Signaling Cues

<p>The rotator cuff is a group of four muscles and tendons in the shoulder that function to lift and rotate the arm. Rotator cuff tendon tears are increasingly common: more than 545,000 rotator cuff surgeries occur annually in the US. However, treatment is often complicated by disorganized collagen matrix formed via fibrosis and results in high re-tear rates. Tendon tissue engineering seeks to solve the problem using biomaterials to promote neo-tendon formation to augment repair or regenerate tendon. However, while current biomaterials provide the opportunity to improve tendon healing, they frequently still exhibit fibrosis in preclinical studies. Therefore, a critical need exists to understand the mechanisms of aligned collagen formation when designing biomaterials for tendon tissue engineering. Matrix architecture and transient receptor potential cation channel subfamily V member 4 (TRPV4) regulate aligned collagen formation during tenogenesis in vitro, but the mechanism remains to be determined. Recently, TRPV4 stimulation was found to induce nuclear localization and activation of transcriptional co-activators Yes-associated protein (YAP). YAP expression is upregulated during tendon development, a process characterized by aligned collagen formation, and in response to physiological mechanical stimulation, suggesting it could play an important role in tendon. The objective of this work is to improve tissue engineering strategies and progress toward making a device that regenerate tendon after injury. Aim 1 incorporates tendon-derived matrix into synthetic polymer scaffolds to add biological signaling cues to induce tenogenesis. Aim 2 uses a 2D photolithography system (microphotopatterning) to optimize architectural and structural cues to promote stem cell differentiation toward tenogenic, chondrogenic, and osteogenic lineages. Aim 3 investigates dynamic tensile loading protocols to promote collagen matrix synthesis and improve engineered tendon mechanical function. Aim 4 investigates the role of TRPV4 and YAP in collagen alignment during engineered tendon development.</p>
