Le lait écrémé est utilisé depuis plus d’un demi-siècle comme diluant protecteur
des spermatozoïdes de mammifères. Depuis quelques années, il existe une demande
grandissante pour des diluants exempts de produits d’origine animale. Toutefois, le
mécanisme par lequel le lait protège les spermatozoïdes n’est pas connu, ce qui rend
difficile de lui trouver un substitut.
Les protéines majeures du plasma séminal de taureau, les protéines « Binder of
SPerm » (BSP), sont néfastes lors de la conservation de la semence. Les spermatozoïdes
sont en contact avec une grande concentration de protéines BSP qui stimulent une
extraction continuelle de cholestérol/phospholipides de leur membrane plasmique. Les
lipoprotéines de faible densité (LDL) du jaune d’oeuf, un autre composé utilisé dans les
diluants, empêcheraient les protéines BSP de se lier à la membrane des spermatozoïdes de
taureaux et de stimuler un efflux des lipides membranaires, ce qui les protégerait durant la
conservation. Notre hypothèse était que les protéines du lait protègent les spermatozoïdes
durant la conservation en séquestrant les protéines BSP.
Premièrement, nous avons démontré par filtration sur gel qu’il y a une interaction
entre les protéines BSP bovines et les protéines du lait. Le lait écrémé a été fractionné en
trois fractions : F1 (alpha-lactalbumine, bêta-lactoglobuline et caséine kappa), F2 (toutes les
protéines du lait) et F3 (sels, sucres et petits peptides). Les protéines BSP1 et BSP5 ont
une affinité plus grande pour F1 que BSP3, tandis que toutes les protéines BSP ont une
affinité pour F2. Le titrage calorimétrique isotherme a permis de confirmer l’interaction
entre les protéines BSP et les protéines du lait. L’association entre la protéine BSP1
bovine et les micelles de caséines est caractérisée par une constante d’affinité (Ka) de 3.5
× 10^5 M-1 et un paramètre stoichiométrique (n) de 4,5 BSP1 pour une caséine.
L’association entre la protéine BSP1 bovine et l’alpha-lactalbumine (une protéine du sérum
principale), est caractérisée par un Ka de 2.4 × 10^5 M-1 et une valeur “n” de 0,8. Ces
résultats indiquent que le lait protège les spermatozoïdes bovins en séquestrant les
protéines BSP grâce à une interaction protéine : protéine, tandis que le jaune d’oeuf les
protège grâce à une interaction protéine : lipoprotéine. Deuxièmement, nous avons
démontré par filtration sur gel que les protéines homologues aux BSP bovines retrouvées
dans le plasma séminal de porc, d’étalon et de bélier ont une affinité avec les protéines du
lait, ce qui suggère que le mécanisme de protection des spermatozoïdes par le lait pourrait
être le même chez ces espèces. Troisièmement, nous avons caractérisé l’interaction entre
BSP1 bovine et les LDL du jaune d’oeuf qui a un Ka de 3.4 ± 0.4 × 10^6 M-1 et une valeur
de « n » de 104 BSP1 pour une particule de LDL, indiquant qu’il existe des différences
entre le mécanisme de protection des spermatozoïdes par le lait et le jaune d’oeuf.
Nous croyons que les résultats présentés dans cette thèse aideront à créer de
nouveaux diluants ne contenant pas de produits d’origine animale afin de cryoconserver
les spermatozoïdes des mammifères. / Skim milk is being used as a protective agent for mammalian semen conservation
over half a century. Recently, there has been increased interest in developing extenders
free of animal products. However, it is difficult to find suitable component in order to
replace milk as an extender, because the mechanisms by which milk protect sperm against
cooling and freezing damages during the storage is unknown.
The Binder of SPerm (BSP) proteins are the major proteins of bull seminal plasma
and they are harmful during sperm storage. In fact, sperm would be in contact with a large
quantity of BSP proteins that induce a continuous cholesterol and phospholipids efflux
from the sperm membrane during storage. When bull sperm is diluted with an extender
containing egg yolk, another compound frequently used in extender, the low-density
lipoproteins (LDL) present in the egg yolk prevent the binding of the BSP proteins to the
sperm membrane, thus, preventing the lipid efflux from the sperm membrane induced by
the BSP proteins. Our hypothesis was that milk proteins would protect sperm during
storage by binding BSP proteins.
First, we demonstrated by gel filtration that bovine BSP proteins could bind the
milk proteins. Skim milk was fractionated into three fractions: F1 (alpha-lactalbumin and beta-
lactoglobulin, the major whey proteins and kappa-casein), F2 (mainly caseins and all other
milk proteins in small amounts) and F3 (salts, sugars and small peptides). Bovine BSP1
and BSP5 have more affinity for F1 as compared to BSP3 and all the BSP proteins have
affinity for F2. We confirmed the interaction between bovine BSP proteins and milk
proteins by isothermal titration calorimetry. The binding of BSP1 to casein micelles is
characterized by an affinity constant (Ka) of 3.5 × 10^5 M-1 and of a stoichiometric
parameter for the association (n) of 4.5 BSP1 per casein. The association between BSP1
and alpha-lactalbumin (one of the major whey proteins) is characterized by a Ka of 2.4 × 10^5
M-1 and a “n” value of 0.8. These results support our contention that milk can protect
sperm by preventing the BSP proteins’ binding to the sperm membrane attributable to a
protein : protein interaction, while egg yolk sperm protection is attributable to a protein :
lipoprotein interaction. Second, our studies showed that the homologous BSP proteins
found in the boar, stallion and ram seminal plasma can bind the milk proteins. These
results indicate that the mechanism of sperm protection by milk in these species should be
similar to the one in bovine species. Third, we characterized the interaction between
bovine BSP1 protein and LDL from hen’s egg yolk. The binding was characterized by a
Ka of 3.4 ± 0.4 × 10^6 M-1 and a « n » value of 104 BSP1 per LDL particle. Our results
indicated that there is difference between the mechanism of sperm protection by milk and
egg yolk.
We believe that the results presented in this thesis may help to create new
extenders free of animal product for mammal sperm preservation in liquid or frozen state.

Les micelles de caséines, composées de caséines, minéraux et eau, sont en équilibres dynamiques, elles échangent en permanence de leur matière avec leur environnement. Les micelles de caséines possèdent d’intéressantes fonctionnalités pour la fabrication de produits laitiers, telles que leur capacité à stabiliser des émulsions et à former des gels sous l’action de la présure. Des changements environnementaux, variations de pH, additions de sels ou d’agents chélatants, affectent les équilibres des micelles et induisent des modifications de leurs compositions et propriétés colloïdales. Ces changements modifient également leurs propriétés fonctionnelles, bien que cet aspect soit peu décri. Le but de ce projet était de comprendre les relations liant l’environnement, l’équilibre minéral, les propriétés colloïdales et fonctionnelles des micelles de caséines. L’impact de cinq facteurs (pH, Na3Cit, NaCl, CaCl2, MgCl2) modifiant les micelles fut étudié en focalisant sur leurs propriétés émulsifL’acidification et l’addition de Na3Cit ont causé les modifications minérales les plus importantes en solubilisant le phosphate de calcium micellaire. Le premier conduisit au « gonflement » des micelles alors que l’agent chélatant causa leur dissociation en petits agrégats. L’ajout de NaCl n’eut aucun impact sur le contenu minéral des micelles mais provoqua aussi la libération d’agrégats, révélée par cryo microscopie électronique et diffusion de rayons-X aux petits angles. Le rôle du phosphate de calcium sur les fonctionnalités fut confirmé et l’étude révéla l’importance de contrôler la libé / Caseins micelles, composed of caseins, minerals and water, are under dynamic equilibria, they constantly exchange materials with their surrounding environments. In addition, casein micelles possess valuable functionalities in regards to the formation of dairy products, such as the ability to stabilize emulsions or to form rennet gels. Environmental changes, such as variations in pH, additions of salts or chelating agents, affect the casein micelles equilibria and lead to modifications in their compositional and colloidal properties. Such changes also modify their functional properties, although this aspect is poorly described in the literature. This project aimed to understand the relationships that link the environmental modifications, the mineral balance, the colloidal and functional properties of the casein micelles. The impact of five modifying factors (pH, Na3Cit, NaCl, CaCl2, MgCl2) of the casein micelles were studied and the focus was placed on emulsion and rennet coagulation fuThe former only induce the swelling of casein micelles while the latter led to their disruption into smaller aggregates. NaCl had no impact on the micellar mineral content but also caused the release of small aggregates, as revealed by electronic microscopy and small angle X-ray scattering analyses. The decisive role of micellar calcium phosphate on the functionalities was confirmed and this study highlighted the importance of monitoring the release of small aggregates, as they strongly affected emulsions stability and gels firmness. Cette thèse étudie le procédé d’ultrafiltration tangentiel assisté par ultrasons aux échellesmacro et nanométriques. Différentes dispersions colloïdales ont été filtrées (argiles, micelle decaséine, nanocristaux d’amidon et de cellulose). Les propriétés d'écoulement et les changementsinduits par les ultrasons (US) ont été caractérisés. Les organisations structurelles à proximité de lamembrane ont été mises en évidence pour la première fois aux échelles nanométriques, lors de lafiltration par diffusion de rayons X aux petits angles in-situ. L’application des US a permis uneaugmentation significative des flux de perméation d’un facteur 1,6 à 13,5, selon l'organisationstructurale des colloïdes. Trois mécanismes induits par les US ont été identifiés : une érosioncomplète, une rupture partielle ou pas de changement (nanométrique) des couches de particulesaccumulées. Grâce aux profils de concentration obtenus, une approche de modélisation a permisune prévision du flux perméation. / This thesis studies an ultrasonic assisted cross-flow ultrafiltration process from macro tonano scales. Different types of colloids were investigated: synthetic and natural clay dispersions,casein micelles (skim milk) and starch or cellulose nanocrystal suspensions. Firstly, flowproperties and the changes due to ultrasound (US) were investigated. Secondly, structuralorganizations at nanometer length scales in the vicinity of the membrane during filtration havebeen revealed for the first time by real-time in-situ Small Angle X-ray Scattering. The applied USincreased significantly the permeate flux of ultrafiltration by an enhancement factor of 1.6 to13.5, depending on the structural organization of the colloids. The applied US has led to threemain effects: a removal of accumulated particle layer, a partial disruption or no change of thenano-organization. Thirdly, thanks to the obtained concentration profiles, a modeling approachhas allowed a prediction of the permeate flux.

Die praktische Arbeit des Monumentalmalers Hermann Prell (1854–1922) wurde zeitlebens von einer intensiven Auseinandersetzung mit Fragen der Maltechnik begleitet, die sich in seinen umfangreichen Aufzeichnungen niederschlug. Aufgrund seiner zahlreichen Kontakte zu Zeitgenossen, wie Hans von Marées und Arnold Böcklin, stellt sein schriftlicher Nachlass eine wichtige Quelle zur kunsttechnischen Entwicklung um 1900 dar.
Im Rahmen der vorliegenden Arbeit werden seine Forschungen zur Maltechnik und die Beschreibung der von ihm angewendeten Materialien sowie Techniken das erste Mal tiefgreifend untersucht und umfassend vorgestellt. Die erhaltenen Dokumente bieten die Grundlage für eine Aufschlüsselung der verwendeten Rezepturen und handelsüblichen Künstlermaterialien sowie deren Kontextualisierung. Des Weiteren ergänzen historische Abbildungen und Vorstudien zur Erarbeitung der Komposition und Maltechnik den Blick auf den Werkprozess. Ebenso wird die kunsttechnologische Untersuchung von ausgewählten Werken mit der Auswertung der Quellen kombiniert.
Während seiner monumentalen Aufträge galt sein besonderes Interesse der technischen und künstlerischen Umsetzung eines Gesamtkunstwerkes. Hermann Prell widmete sich eingehend kunsttechnologisch relevanter historischer und zeitgenössischer Quellenliteratur, zum Beispiel von Cennino Cennini und Ernst Berger. Die Freskotechnik nahm im breiten technischen Spektrum seines Œuvres einen hohen Stellenwert ein. Daneben spielte die Beschäftigung mit Tempera sowohl für das Staffeleibild als auch für die Wandmalerei eine große Rolle. Insgesamt wird erstmals ein Überblick über Hermann Prells angewendete Kunsttechniken in ihrer Vielfalt sowie deren Genese gegeben. / Throughout his life Hermann Prell (1854–1922) – known for his monumental paintings – accompanied his practical work with intense explorations of questions related to painting technique. This is reflected in his extensive writings. Prell was also engaged in a lively exchange with other artists, such as Hans von Marées and Arnold Böcklin. Hermann Prell’s writings combined with his contemporaries’ present an important source on painting technique around the turn of the 19th century in Germany.
This study provides the first in-depth examination and comprehensive presentation of Prell’s research into painting techniques and a description of the materials and techniques he utilised. The preserved documents provide the basis for analysing the recipes the artist used and commercially available artists’ materials as well as their contextualisation with current research. Our view of his working process is further supplemented by historical reproductions and preparatory studies that served Prell to develop his compositions and painting techniques. Furthermore, the results of recent technical examinations of selected artworks are combined with an evaluation of the written sources.
For his numerous large commissions for government buildings, Prell was particularly interested in the technical and artistic realisation of a Gesamtkunstwerk. He occupied himself extensively with related historical and contemporary sources, for example, the writings of Cennino Cennini and Ernst Berger. Amongst the broad technical range of his oeuvre the fresco technique assumed particular importance. In addition, his occupation with tempera’s use for easel paintings as well as wall paintings played a major role. In conclusion, for the first time Hermann Prell’s various applied techniques and materials as well as their genesis are given a comprehensive overview.
