<p>Quantum chemical methods have been used to provide a better understanding of the photochemistry of astaxanthin and phytochromobilin; the photoenzymic repair of UV-light induced DNA damages; and the formation of lignin. </p><p>The carotenoid astaxanthin (AXT) is responsible for the colouration of lobster shell. In solution, the electronic absorption spectra of AXT peak in the 470-490 nm region, corresponding to an orange-red colouration. Upon binding to the lobster-shell protein-complex α-crustacyanin, the absorption maximum is shifted to 632 nm, yielding a slate-blue colouration. Herein, the structural origin of this bathochromic shift is investigated on the basis of recent experimental work.</p><p>The tetrapyrrole phytochromobilin (PΦB) underlies the photoactivation of the plant photoreceptor phytochrome. Upon absorption of 660-nm light, PΦB isomerizes from a C15-<i>Z,syn</i> configuration (in the inactive form of the protein) to C15-<i>E,anti</i> (in the active form). In this work, a reaction mechanism for this isomerization is proposed. </p><p>DNA photolyases are enzymes that repair DNA damages resulting from far-UV-light induced [2+2] cycloaddition reactions involving pyrimidine nucleobases. The catalytic activity of these enzymes is initiated by near-UV and visible light, and is governed by electron transfer processes between a catalytic cofactor of the enzyme and the DNA lesions. Herein, an explanation for the experimental observation that the repair of cyclobutane pyrimidine dimers (CPD) – the major type of lesion – proceeds by electron transfer from the enzyme to the dimer is presented. Furthermore, the formation of CPD is studied.</p><p>Lignin is formed by dehydrogenative polymerization of hydroxycinnamyl alcohols. A detailed understanding of the polymerization mechanism and the factors controlling the outcome of the polymerization is, however, largely missing. Quantum chemical calculations on the initial dimerization step have been performed in order to gain some insight into these issues.</p>

Gegenstand der vorliegenden Arbeit ist die Synthese und Charakterisierung neuer chromophorer Catecholderivate mit ausgeprägten push-pull-pi-Systemen. Die solvatochromen Eigenschaften dieser Verbindungen werden in Abhängigkeit der Wasserstoffbrückenbindungsdonor- und -akzeptorfähigkeit sowie Lösungsmitteldipolarität diskutiert. Mit entsprechenden methoxy- und dimethoxyfunktionalisierten Catecholderivaten ist es möglich, vergleichende Struktur-Eigenschaftsbeziehungen aufzustellen. Durch Untersuchungen zu den Wechselwirkungen der chromophoren Catechole mit Schwermetallionen kann gezeigt werden, dass die synthetisierten Verbindungen als Sensoren eingesetzt werden können. In der vorliegenden Arbeit wird des Weiteren die Adsorption der Catecholderivate an Metalloxide beschrieben. Mit Farbstoffen sensibilisierte Oberflächen stellen derzeit ein interessantes Forschunggebiet dar. Ferner wird über die Umsetzung der Catecholderivate mit Trialkoxysilanen zu zwitterionischen, spirozyklischen, pentakoordinierten lambda5Si-Silicaten sowie mit Tetraalkoxysilanen zu dianionischen, hexakoordinierten lambda6Si-Silicaten berichtet. Besonderes Augenmerk lag dabei auf UV/vis-spektroskopischen Untersuchungen.

Studies on Correlation between Microstructures and Electronic Properties of Organic SemiconductorsMukhopadhyay, Tushita January 2017 (has links) (PDF)
The work carried out in this thesis systematically investigates the correlation between microstructures and electronic properties of organic semiconductors. The major directions that were pursued in this thesis are: (i) studies on structure-property relationship by rational design and synthesis of monodisperse oligomers with varying chain-lengths (ii) role of electronic properties and aggregation (microstructures) in governing singlet fission (SF). In the first part of the thesis, the optical, structural and charge transport properties of Diketopyrrolopyrrole (DPP)-based oligomers, as a function of the chain length, has been discussed. The energy bands became wider with an increase in chain length and a gain in backbone electron affinity was observed, with an offset in microstructural order. With an increase in chain length, the tendency to form intramolecular aggregates increased as compared to intermolecular aggregates due to the onset of backbone conformational defects and chain folding. An insight into the solid-state packing and microstructural order has been obtained by steady-state and transient spectroscopy, grazing incidence small angle x-ray scattering (GISAXS), atomic force microscopy (AFM) and transmission electron microscopy (TEM) studies. The charge-carrier mobilities varied in accordance with the degree of microstructural order as: dimer > trimer > pentamer. A library of DPP-DPP based trimers was also generated by modifying the donor chromophore (phenyl, thiophene and selenophene) in the oligomer backbone. Highest n-channel mobility of ~0.2 cm2V-1s-1 was obtained which validated that: (a) the effect of solid-state packing predominates the effect of backbone electronic structure on charge carrier mobility. Although oligomers possess lesser backbone defects than polymers in general, their charge carrier mobilities were not comparable to that of 2DPP-OD-TEG polymer, which forms highly oriented and isotropic edge-on crystallites/microstructures in the thin film, shows high n-channel mobility of 3 cm2V-1s-1 and band-like transport ;(b) although delocalized electronic states are achieved at greater chain lengths, the degree of solid-state microstructural order drastically reduces which leads to lower charge carrier mobilities; (c) conformational collapse resulted in lower electron mobilities and an increase in ambipolarity.
The later part of the thesis debates on the relative contribution of electronic structure and aggregation (microstructures) in governing singlet fission (SF). Motivated by the recent SF model in carotenoid aggregates, a DPP-DPP based oligomer was synthesized by incorporating a vinylene bridge to imbue “polyene” character in the chromophore. Transient Spectroscopy (TA) measurements were carried out to monitor the formation of triplet states in the oligomer and to probe the occurrence of singlet fission. Although the oligomer exhibits “polyene” character like a typical “carotenoid aggregate”, it did not show singlet fission because of the additional stabilization of the singlet (S1) state which reduces the ∆EST. This study rationalized the importance of judicious control of band structures as well as microstructures to observe the SF phenomenon in this category of chromophores. The novel synthetic protocol provides the scope to tailor DPP-DPP based materials with desired effective conjugation lengths and side chains and can foreshow great prospects for future generation of organic electronics.

La photothérapie dynamique (PTD) est un traitement alternatif du cancer qui nécessite l’utilisation de chromophores (photosensibilisateurs) capables d’induire la mort cellulaire suite à une irradiation lumineuse. Les nanoparticles d’or (AuNP), grâce à leur phénomène de résonance plasmon localisée, peuvent exalter les propriétés photophysiques des chromophores localisés à leur surface. L’utilisation d’une excitation biphotonique, dans le proche infrarouge, peut être utilisée pour améliorer l’action thérapeutique (PTD) ou diagnostique (imagerie de fluorescence) des chromophores en augmentant la profondeur de pénétration dans les tissus et la résolution tridimensionnelle de la microscopie biphotonique.Lors de ce travail, l’élaboration de nouvelles nanoparticules hybrides est proposée, présentant des applications potentielles en bioimagerie (sondes brillantes) et comme photosensibilisateurs pour la PTD. Ces nanoparticules sont composées d’un cœur d’or sur lequel sont greffés des conjugués polymère-chromophores biphotoniques. La stratégie de synthèse a consisté à : i) synthétiser des conjugués polymère-chromophores biphotoniques solubles dans l’eau ; ii) les greffer de manière orientée à la surface des AuNP. La synthèse des conjugués polymère-chromophores hydrosolubles on été synthétisés via le couplage efficace de chromophores hydrophobes en position latérale des copolymères P(NAM-co-NAS) bien-définis, obtenus par la techniques de polymérisation radicalaire contrôlée RAFT. Cette stratégie permet le contrôle à la fois de la longueur des chaînes polymère formées (2 000 g.mol-1 < Mn plus < 37 000 g.mol-1) et du nombre de chromophores couplés par chaîne (de 1 à 21). Le greffage orienté de ces conjugués à la surface des AuNP a été mis en évidence (TEM, ATG) et les densités de greffage se sont avérées élevées (~0.5chaîne/nm²). Un des rôles de la chaîne polymère étant de contrôler la distance entre les chromophores et la surface des AuNP. Il a été mis en évidence que l’augmentation de la longueur de chaine des conjugués induisait, outre une augmentation de la couronne greffée (par MET) sur les AuNP, une augmentation de l’émission de fluorescence de ces conjugués polymère-chromophores. Enfin, les propriétés biologiques des conjugués avant et après greffage sur les AuNP ont été évaluées in cellulo, mettant en valeur leur potentiel pour des applications thérapeutiques et diagnostiques. / Photodynamic therapy (PDT) is an alternative treatment of cancer requiring the use of chromophore molecules (photosensitizers), which can induce cell death after light excitation. Gold nanoparticles (AuNP), exhibiting localized Surface Plasmon Resonance, can enhance the photophysical response of chromophores located in their vicinity, and thus improve their therapeutic action. Moreover, the use of highly localized two-photon chromophores (photosensitizers and fluorophores), capable to undergo a localized excitation by light in the Near InfraRed region, should increase the penetration depth into tissues, thus improve the treatment efficiency (by PDT) and the imaging (by fluorescence microscopy) of cancer tissues.In this work, we describe the elaboration of water-soluble hybrid nano-objects for PDT and fluorescence bioimaging applications, composed of two-photon chromophore-polymer conjugates grafted onto gold nanoparticles. In order to obtain these nano-objects we follow a multistep strategy: i) the synthesis of a well-defined water-soluble chromophore-polymer conjugates; ii) the end-group oriented grafting of chromophore-polymer conjugates onto 20 nm AuNP. The coupling of hydrophobic two-photon chromophores on linear water-soluble copolymer chains (poly(N-acryloylmorpholine-co-N-acryloxysuccinimide)), obtained by controlled/living RAFT polymerization, resulted in well-defined water-soluble chromophore-polymer conjugates, with different polymer lengths (2 000 g.mol-1 < Mn < 37 000 g.mol-1) and architectures (random or block), and a controlled number of chromophores per chain (varying between 1 and 21). Their grafting onto 20 nm AuNP gave water-soluble hybrid nano-objects with high grafting densities (~0.5 chains/nm²). The role of the polymer chain being to tune the distance between chromophores and AuNP surface, we have evidenced the increase in the polymer corona thickness of grafted AuNP (estimated by TEM) with the increasing polymer Mn, corroborating with the corresponding distance-dependent fluorescence properties of those. Finally, the in cellulo biological properties of two-photon chromophore-polymer conjugates, before and after grafting onto AuNP, have been investigated, highlighting their potential for two-photon bioimaging and PDT applications.

Bright and stable near-infrared (NIR) and infrared (IR) emitting chromophores are in high demand for applications in telecommunication, solar cells, security barcodes, and as fluorescent reporters in bioimaging studies. The best choice for wavelengths >750 nm are semiconductor nanocrystals, especially ternary or alloy nanocrystals like CdHgTe, which enable size and composition control of their optical properties. Here, we report on the influence of growth time and surface chemistry on the composition and optical properties of colloidal CdHgTe. Up to now, these are the only NIR and IR emissive quantum dots, which can be synthesized in high quality in water, using a simple one-pot reaction. For this study we utilized and compared three different thiol ligands, thioglycolic acid (TGA), 3-mercaptopropionic acid (MPA), and glutathione (GSH). Aiming at the rational design of bright NIR- and IR-emissive alloy materials, special emphasis was dedicated to a better understanding of the role of the surface ligand and adsorption–desorption equilibria on the photoluminescence quantum yield and stability. In this respect, dilution and protonation studies were performed. Our results show that with this simple synthetic procedure, strongly fluorescent CdHgTe colloids can be obtained with MPA as stabilizing ligand revealing quantum yields as high as 45% independent of particle concentration.
