Posteriormente nós testamos a produção de B12 pelas cepas mutantes e selvagens já descritas, e incluímos neste estudo a cepa de S. Typhimurium ΔcobSΔcbiA, e sua respectiva cepa selvagem. Todas as cepas mutantes não tiveram produção de B12 detectada. As cepas selvagens mostraram produção de vitamina B12 em ambos os ensaios utilizados, com exceção da S. Gallinarum que não apresentou produção de cobalamina in vitro. Como conclusão, a produção de vitamina B12 in vitro diferiu entre os sorotipos de Salmonella testados, a deleção dos genes cbiA e cobS produziu alteração na relação parasita hospedeiro em diferentes níveis entre os sorotipos de Salmonella estudados, sendo que esta foi muito maior entre o sorotipo Gallinarum e as aves. / Salmonella enterica serovar Typhimurium only synthesizes cobalamin (Vitamin B12) during anaerobiosis. Two-percent of the S. Typhimurium genome is devoted to the synthesis and uptake of Vitamin B12 and to B12-dependent reactions. In order to understand the requirement from cobalamin synthesis better, we constructed Salmonella serovar Gallinarum, Salmonella serovar Enteritidis and Salmonella serovar Pullorum mutants that are double-defective in cobalamin biosynthesis (ΔcobSΔcbiA). We compared the virulence of these mutants to that of their respective wild type strains and found no impairment in S. Enteritidis ΔcobSΔcbiA and S. Pullorum ΔcobSΔcbiA ability to cause disease in chickens. S. Gallinarum ΔcobSΔcbiA mutant showed attenuated for chickens. We then assessed B12 production by these mutants and their respective wild type strains, as well as S. Typhimurium ΔcobSΔcbiA, and their respective wild type strain. All mutants were unable to produce detectable B12. B12 was detectable in wild type strains, but, S. Gallinarum demonstrated no in vitro cobalamin production. In conclusion, the production of vitamin B12 in vitro differed across the Salmonella serotypes that were tested. Furthermore, the deletion of the cbiA and cobS genes resulted in an alteration in the relationship between the serotype Gallinarum and the birds more stronger than did to the others serotypes.

Ash from homestead fireplaces and wood as possible sources of minerals for livestock

In this study the mineral composition of ash from fireplaces at rural homesteads was determined to establish if such ash could be a source of minerals to goats. The mineral composition of ash collected from homesteads in six different communal farming regions of South Africa was determined. The ash samples collected in the three northern regions of the country contained between 180 and 248 g Ca/kg dry ash, while those from the Eastern Cape Province contained low concentrations (8–45 g Ca/kg ash) and high concentrations of Si. It is concluded that wood was probably used as fire making material in the northern regions, while cattle manure was probably used in the Eastern Cape regions, though soil contamination could have contributed to the high Si concentrations. The concentration of the Fe and Zn in ash was high while that of the other elements in ash was relatively low and would probably contribute little to a goat’s diet, considering the proportion of ash in a total diet. It is concluded that in some regions of the country goats would be able to ingest a substantial proportion of Ca when scavenging on ash heaps or receiving ash as a dietary supplement, while in other regions this will not be the case. / Dissertation (MInstAgrar (Animal Production))--University of Pretoria, 2008. / Animal and Wildlife Sciences / unrestricted

Compréhension et optimisation d'un procédé de biodégradation d'effluents agricoles composés de pesticides en mélange / Optimization of a biodegradation process for agricultural effluents composed of mixed pesticides

Ces travaux ont consisté à développer un procédé de traitement biologique des eaux contaminées en produits phytosanitaires (PF) issues du lavage des matériels de pulvérisation de PF. Ce procédé s’appuie surl’inoculation d’une culture bactérienne mixte sélectionnée et d’un matériau sorbant constitué de rafles de maïs broyées.Tout d’abord, un mélange modèle de PF,constitué de 6 herbicides, 3 fongicides et 1 insecticide, a été créé. Après avoir mis au point la production de l’inoculum en présence de ce mélange et étudié les propriétés sorbantes des rafles de maïs, les paramètres de traitement ont été définis à petite échelle de laboratoire et validés en réacteur.Finalement, le procédé développé est un traitement en réacteur batch séquentiel de 28 jours qui permet de réduire de plus de 50 % la masse initiale des PF du mélange modèle. Malgré la réduction des teneurs en PF, la phytotoxicité due aux résidus d’herbicides des effluents traités est encore trop élévée et ne permet pas de valider l’épandage sur un lit planté de roseaux comme post-traitement de la fraction liquide. / Aim of this work was to treat effluentsobtained from rinsing pesticide (PF) spraying equipments by developing biological treatment process into bioreactor with selected mixed bacterial culture and adsorption support consisting of crushed corn cobs. First, a model PF mixture was designed to represent agriculturaleffluents at watershed scale. This mixture was composed by 6 herbicides, 3 fungicides and 1 insecticide. Then, inoculum production under PF pressure was developed and sorption properties of corn cobswere investigated. Process parameters were defined at small-scale laboratory and validated into bioreactors. Thus, developed processwas a28 days sequential batch reactortreatment, which reduced more than 50% of molecules weight of model mixture.Despite of this process significantely reduced treated effluents PF contents, remaining phytotoxicity due to herbicide residues failed validation of their post-treatment on a bed planted with reeds.

USE OF TRIAXIAL TESTING TO OBTAIN THE SHEAR FAILURE SURFACE IN THE MODIFIED DRUCKER-PRAGER CAP MODEL

<p dir="ltr">Biorefineries rely on compression feed screws to transport biomass for biofuel production in chemical reactors. However, flowability issues within these feedscrews often lead to production downtime, impacting profitability. Modeling biomass flow within the feedscrews is crucial to optimize processing parameters like torque and speed, reducing downtime. Biomass is a non-uniform granular material which faces flowability issues. The problems in flowability is influenced by factors such as particle size, moisture content, material composition, and processing methods. Identifying key parameters that can influence the material behavior is vital to minimize production downtimes. Feedscrews operate under high pressures which makes obtaining accurate material parameters at these high pressures challenging. Many methods used within the pharmaceutical industry to obtain material parameters are unable to reach the larger pressures that the material experiences within the feedscrew. However, Triaxial testing can be used to test the material at the high pressure of interest. Triaxial testing has been used within the civil engineering field to test granular materials such as soils, sand, and rocks. The Finite Element Method (FEM) using a continuum model is used for modeling systems with a large number of particles. The modified Drucker-Prager Cap (mDPC) continuum model is often used to capture complex material behavior, including densification and shear yielding in granular materials. This model seems well suited to capture the behaviors of biomass material. The focus of the thesis is to obtain the shear failure properties of corn stover using triaxial testing and the Drucker-Prager Cap continuum model. Simulations and experimental data are utilized to establish a criterion for identifying shear failure. While simulations depict ideal behavior of a DPC material with frictionless and frictional platens, experimental data shows trends of real-life corn stover. Simulation results effectively predict the material’s friction angle but show larger errors in estimating cohesion, potentially due to extrapolation or cohesion’s sensitivity to volumetric plastic strain. Further simulations at smaller hydrostatic unloading pressures are recommended to reduce this error. Experimental trends for shear failure seem to align with simulation trends for shear failure identification. However, the densification trends in experiments lack the clarity observed in the trends from the simulations. More triaxial experiments should be run to determine if the trends are consistent at other hydrostatic loading and unloading pressures. More than two experiments at the same hydrostatic loading pressure should also be run to estimate the shear failure line to obtain a better estimation. Experimentally there are a number of other factors that could contribute to errors such as the estimated material diameter used to calculate Mises stress, if corrections were made for items such as the moving piston, latex membrane, and more, and how far the shear failure line is extrapolated to the vertical axis.</p>

A 3D-printed Fat-IBC-enabled prosthetic arm : Communication protocol and data representation

The aim of this thesis is to optimize the design of the Fat-IBC-based communication of a novel neuroprosthetic system in which a brain-machine interface is used to control a prosthetic arm. Fat-based intra-body communication (Fat-IBC) uses the fat tissue inside the body of the bearer as a transmission medium for low-power microwaves. Future projects will use the communication system and investigate ways to control the prosthetic arm directly from the brain. The finished system was able to individually control all movable joints of multiple prosthesis prototypes using information that was received wirelessly through Fat-IBC. Simultaneous transmission in the other direction was possible, with the control data then being replaced by sensor readings from the prosthesis. All data packets were encoded with the COBS/R algorithm and the wireless communication was handled by Digi Xbee 3 radio modules using the IEEE 802.15.4 protocol at a frequency of 2.45 GHz. The Fat-IBC communication was evaluated with the help of so-called "phantoms" which emulated the conditions of the human body fat channel. During said testing, packet loss measurements were performed for various combinations of packet sizes and time intervals between packets. The packet loss measurements showed that the typical amount of transmitted data could be handled well by the fat channel test setup. Although the transmission system was found to be well-functioning in its current state, increasing the packet size to achieve a higher granularity of the movement was perceived to be viable considering the findings from the packet loss measurements.
