O segmento das comunicações, já há alguns anos, vem passando por significativas transformações exigindo a interação entre ambientes tecnológicos convergentes heterogêneos, com qualidade na continuidade de serviços, para se manter competitivo, pois é este mercado que exige, constantemente, que mais recursos tecnológicos lhe sejam colocados à disposição. O ineditismo da proposta desenvolvida neste trabalho é a elaboração de um modelo para assegurar a continuidade da qualidade de serviço percebida pelo usuário final através de handover (ou handoff) vertical, o que caracteriza-se como uma necessidade de solução global, isto é, o modelo é genérico e independente da tecnologia, o que permite sua adoção em qualquer ambiente de rede existente aproveitando a capilaridade já disponível das redes legadas. É analisada também a situação na qual um usuário final esteja acessando simultaneamente serviços gerenciados por Provedores de Serviço distintos, o que conduz a duas situações possíveis: a) o usuário está acessando serviços distintos contratados a Provedores de Serviço diferentes; b) o usuário está acessando o mesmo serviço contratado a Provedores de Serviço distintos. Nesta última situação pode surgir a necessidade de disparar um processo de handover exigindo a decisão de qual dos Provedores o executará, de acordo com o SPHDA Service Providers Handover Decision Agreement. A metodologia adotada para o desenvolvido do modelo foi a RM-ODP - Reference Model for Open Distributed Processing, por abranger todos os aspectos técnicos e comerciais necessários à sua construção. / The sector of communications, for some years, has undergone significant changes requiring interaction between converging heterogeneous technology environments, with quality and continuity of services to stay competitive, because this market is that requires constantly more technological resources available. The novelty of the proposal developed in this work is the development of a model to ensure the continued quality of service perceived by end users via vertical handover (or handoff), which characterizes itself as a need for a global solution, i.e., the model is generic and technology independent, allowing its adoption in any network environment taking advantage of the capillary already available from legacy networks. It is also analyzed the situation in which an end user is simultaneously accessing services managed by different Service Providers, which leads to two possible situations: a) the user is accessing different services contracted to different Service Providers; b) the user is accessing the same service contracted to different Service Providers. In this last situation may be necessary to trigger a handover process requiring the decision of which the Providers shall execute it in accordance with the SPHDA - Service Providers Handover Decision Agreement. The methodology adopted for the model development was the RM-ODP - Reference Model for Open Distributed Processing, as it includes all technical and commercial aspects necessary for its construction.

Evolutionary analysis of the β-lactamase families / Analyse évolutive des familles de β-lactamase

Les antibiotiques β-lactamines sont parmi les médicaments antimicrobiens les plus anciens et les plus utilisés. L'enzyme bactérienne β-lactamase hydrolyse l'antibiotique β-lactame en cassant la structure de base "anneau β-lactame". Pour identifier les nouvelles β-lactamases, une étude complète a été réalisée dans diverses bases de données biologiques telles que Human Microbiome Project, env_nr et NCBI nr. L'analyse a révélé que les séquences ancestrales putatives et les recherches de profil HMM jouaient un rôle important dans l'identification de la base de données homologue et métagénomique à distance dans l'enzyme β-lactamase existante comme matière noire. Les larges analyses phylogénétiques des β-lactamases existantes et nouvellement identifiées représentent les nouveaux clades dans les arbres. En outre, l'activité d'hydrolyse des antibiotiques β-lactamines de séquences nouvellement identifiées (provenant d'archées et d'humains) a été étudiée en laboratoire, ce qui montre l'activité de la β-lactamase. La deuxième phase de l'étude a été entreprise pour examiner l'évolution fonctionnelle des β-lactamases. Premièrement, des séquences de protéines ß-lactamase 1155 ont été extraites de la base de données ARG-ANNOT et des valeurs CMI la littérature correspondante. Les résultats ont révélé que l'activité fonctionnelle de la β-lactamase évoluait de manière convergente au sein de la classe moléculaire. La troisième phase de cette thèse représente le développement d'une base de données intégrative de β-lactamases. La base de données publique actuelle de β-lactamases a des informations limitées, par conséquence, une base de données intégrative a été développée. / The β-lactam antibiotics are one of the oldest and widely used antimicrobial drugs. The bacterial enzyme β-lactamase hydrolyzes the β-lactam antibiotic by breaking the core structure “β-lactam ring”. To identify the novel β-lactamases a comprehensive investigation was performed in different biological databases such as Human Microbiome Project, env_nr, and NCBI nr. The analysis revealed that putative ancestral sequences and HMM profile searches played a significant role in the identification of remote homologous and uncovered the existing β-lactamase enzyme in the metagenomic database as dark-matter. The comprehensive phylogenetic analyses of extant and newly identified β-lactamase represent the novel clades in the trees. Further, the β-lactam antibiotic hydrolysis activity of newly identified sequences (from archaea and human) was investigated in laboratory, which shows β-lactamase activity.The second phase of the investigation was undertaken to examine the functional evolution of β-lactamases. First, 1155 β-lactamase protein sequences were retrieved from ARG-ANNOT database and MIC values from the corresponding literature. The results revealed that the functional activity of β-lactamase evolved convergently within the molecular class.The third phase of this thesis presents development of an integrative β-lactamase database. The existing public database of β-lactamase has limited information, therefore, an integrative database was developed.

Um desafio que surge face a essa nova característica dos serviços é como identificar os requisitos de QoS (Qualidade de Serviço) fim-a-fim sob a ótica do usuário final em um ambiente convergente e heterogêneo com o intuito de evoluir as relações entre o usuário final e o Provedor de Serviços para o estabelecimento de um contrato com níveis de serviço personalizado. Under this perspective a challenge arises on how to identify the QoS (Quality of Service) requirements in a convergent and heterogeneous environment end-to-end user centric way, in order to evolve the relationship between users and service providers aiming the establishment of personalized demands through personalized agreements. This work presents a study for the development of an open and generic tool for mapping the user perceived quality in QoS requirements through the capture of the packet flow generated between the applications of a service and simulates delays, jitter, loss of packets and bandwidth in order to allow the end user to identify and express the values of these parameters making transparent all the technological aspects involved in this identification. The definition of a service simulation environment, the development of a QoS simulator and a new relationship between users and services providers are also addressed.

Fundamental and applied studies of Ixodes ricinus salivary proteins / Etudes fondamentales et appliquées de protéines salivaires de la tique Ixodes ricinus

De nombreuses études suggèrent que linhibition de la voie alterne du complément de lhôte est nécessaire aux parasites hématophages pour leur permettre daccomplir leur repas sanguin. Une revue décrivant ces études a été publiée dans Developmental and Comparative Immunology (Schroeder et al., (2009) Dev. Comp. Immunol. 33, 5-13). Plusieurs études suggèrent que linhibition de la voie alterne de lhôte par les protéines salivaires de tiques est importante pour lacquisition du repas sanguin et la transmission subséquente de pathogènes à lhôte infesté. Confortant cette hypothèse, une protéine salivaire capable dinhiber la voie alterne du complément a été clonée chez la tique américaine Ixodes scapularis (Valenzuela et al., (2000) J. Biol. Chem. 275, 18717-18723). Cette protéine, appelée Isac, ne présente aucune homologie avec les autres molécules anti-complément connues à ce jour, suggérant que cette protéine a été acquise au cours de lévolution par un mécanisme dévolution convergente. En plus de cet aspect fondamental, Isac représente un candidat antigénique prometteur pour le développement dun vaccin anti-tique potentiellement capable dinduire le rejet de la tique et/ou de prévenir la transmission des pathogènes. Le but initial de cette thèse était didentifier le ou les homologue(s) de la protéine Isac de la tique américaine Ixodes scapularis chez la tique européenne Ixodes ricinus. De façon intéressante, deux séquences différentes ont été isolées du transcriptome des glandes salivaires de la tique I. ricinus (Daix et al., (2007) Insect Mol. Biol. 16 (2), 155-166). Lexpression de ces séquences a révélé quelles codent pour des protéines secrétées capables dinhiber la voie alterne du complément. Ces protéines ont été appelées IRAC I et II pour « Ixodes ricinus anti-complement protein I and II ». La caractérisation des IRACs à laide danticorps monoclonaux a permis de révéler que ces deux protéines sont exprimées de façon constitutive au sein des glandes salivaires de la tique Ixodes ricinus et sont sur-exprimées au cours du repas sanguin. Létude de lexpression des protéines IRAC I et IRAC II au sein de la population dIxodes ricinus a révélé que ces deux protéines sont des paralogues codés par des gènes différents et non par des allèles dun même locus. Enfin, des analyses phylogéniques portant sur les différentes séquences codant pour les protéines homologues à Isac isolées chez les tiques Ixodes scapularis, Ixodes ricinus et Ixodes pacificus ont révélé que les tiques appartenant au complexe Ixodes ricinus codent pour une famille encore non décrite de molécules anti complément qui se sont diversifiées au cours de lévolution par un processus de sélection darwinienne positive. Les analyses phylogéniques des IRACs suggèrent que ces séquences ont subi une diversification par un processus de pression de sélection darwinienne positive, menant probablement à des molécules possédant des propriétés biologiques différentes. Dans la seconde étude, nous avons testé lhypothèse de travail selon laquelle chaque paralogue pourrait posséder des activités inhibitrices différentes à lencontre du complément de différentes espèces dhôtes naturels, contribuant ainsi à élargir le spectre dhôte des tiques I. ricinus. Les résultats obtenus démontrent que cette hypothèse est correcte (Schroeder et al., (2007) Microbes Infect. 9 (2), 247-250). Dans la troisième et dernière étude de ce manuscrit, nous avons testé le potentiel des protéines IRACs comme candidat antigénique pour le développement dun vaccin anti tique. Des recombinants de lherpèsvirus bovin 4 (BoHV-4) exprimant IRAC I ou IRAC II ont été produits. De façon intéressante, nous avons observé que bien que les recombinants BoHV-4 expriment de hauts taux de protéines IRACs fonctionnelles in vitro, les lapins immunisés à laide des recombinants BoHV-4 exprimant les IRACs ne développent pas de réponse humorale détectable à lencontre des transgènes. Dans le but daugmenter limmunogénicité des IRACs exprimés comme transgène, une seconde génération de recombinants BoHV-4 a été produite. Ceux-ci induisent lexpression des IRACs sous la forme de protéines de fusion transmembranaires à la surface des cellules infectées. Linoculation de ces recombinants à des lapins a engendré le développement dune forte réponse humorale à lencontre des IRACs. Néanmoins, cette réponse immune na pas engendré deffet majeur sur le repas sanguin de femelles Ixodes ricinus placées sur les lapins immunisés. / An increasing number of studies demonstrate that inhibition of host complement activation is crucial for completion of the blood feeding process of hematophagous parasites. A review of these studies has been published in Developmental and Comparative Immunology (Schroeder et al., (2009) Dev. Comp. Immunol. 33, 5-13). Several observations suggest that inhibition of the host complement alternative pathway by tick salivary proteins is crucial for the achievement of blood feeding and efficient transmission of the pathogens transmitted by the parasite. Strongly supporting this conclusion, a salivary protein able to inhibit the alternative pathway was cloned from the American tick Ixodes scapularis (Valenzuela et al., (2000) J. Biol. Chem. 275, 18717-18723). Interestingly, this molecule, termed Isac, has no similarity to any previously reported anti-complement molecules suggesting that it has been acquired through a mechanism of convergent evolution. In addition to this fundamental aspect, Isac is also a promising candidate antigen for the development of an anti-tick vaccine potentially able to induce the reject of the tick and/or to prevent the transmission of the pathogens. The initial goal of the present work was to clone the orthologue of Isac from the European tick Ixodes ricinus. Interestingly, two different sequences were isolated from the transcriptome of I. ricinus salivary glands (Daix et al., (2007) Insect Mol. Biol. 16 (2), 155-166). Expression of these sequences revealed that they both encode secreted proteins able to inhibit the complement alternative pathway. These proteins were called I. ricinus anticomplement (IRAC) protein I and II. Further characterization of IRACs using monoclonal antibodies revealed that both proteins are expressed constitutively in I. ricinus salivary glands and are up-regulated during blood feeding. Analysis of a series of individual ticks revealed that all ticks tested express both IRAC I and IRAC II, demonstrating that they are the products of different genes and not of alleles of the same locus. Finally, phylogenetic analyses of the I. ricinus IRAC I and II sequences together with homologues from I. scapularis and I. pacificus demonstrates that ticks belonging to the Ixodes ricinus complex encode a new family of relatively small anti-complement molecules undergoing diversification by positive Darwinian selection. Phylogenetic analyses of IRACs suggested that these sequences were diversifying by a process of positive Darwinian selection, possibly leading to molecules with different biological properties. In the second study, we tested the hypothesis that each paralogue may have different inhibitory activities against the complement of different natural host species, thereby contributing to broaden the host range of I. ricinus ticks. The data obtained demonstrated that this working hypothesis is correct (Schroeder et al., (2007) Microbes Infect. 9 (2), 247-250). In the third and last chapter of the present manuscript, we addressed the potential of IRAC I and II as candidate antigens for the development of an anti-tick vaccine. Bovine herpesvirus 4 (BoHV-4) recombinants expressing IRAC I or II were produced. Interestingly, we observed that although both recombinants expressed high levels of functional IRAC proteins in vitro, our attempts to immunize rabbits against IRACs via infection with these viruses invariably failed. In order to improve the immunogenicity of IRACs expressed as transgene, a second generation of BoHV-4 recombinants was produced. The latter expressed IRACs as transmembrane fusion proteins on cell surface. Comparison of the vaccine potential of BoHV-4 recombinant viruses expressing either secreted or transmembrane IRAC proteins revealed that while the former did not induce a detectable immune response against IRACs, the latter led to high titers of anti-IRAC antibodies. However, the immune response induced against IRACs did not lead to the reject of the tick but only slightly increased the duration of the blood feeding process.

Realstrukturanalyse von BaFe2As2-Dünnschichten durch Elektronenbeugung

In der vorliegenden Arbeit werden epitaktische BaFe2As2 (Ba122)- Dünnschichten, die auf Spinell-Substraten mit einer Pufferschicht aus Eisen mit gepulster Laserabscheidung hergestellt wurden, durch Transmissions- (TEM) und Rasterelektronenmikroskopie (REM) untersucht. Idealerweise weisen die Ba122-Dünnschichten eine biaxiale Zugdehnung von etwa 2% auf, die zur Supraleitung in ihnen führt. Der Schwerpunkt dieser Arbeit richtet sich auf die Defektanalyse und die Untersuchung des Dehnungszustandes auf Inhomogenitäten. Auf allen Dünnschichten werden Droplets des Targetmaterials beobachtet. Wenn die Höhe der Ba122-Schicht mehr als 60 nm beträgt, erfolgt eine Relaxation der Dehnung durch die Bildung von Rissen. Experimente mit konvergenter Elektronenbeugung in einem TEM sind problematisch, weil der Dehnungszustand durch die Probenpräparation (Krümmung der TEM-Lamelle) beeinflusst wird. Durch die Verwendung hochauflösender Beugung rückgestreuter Elektronen (HREBSD) in einem REM wird gezeigt, dass in allen Ba122-Dünnschichten kleine Orientierungsänderungen des Kristallgitters (≤ 0,2°) und Inhomogenitäten des Dehnungszustandes (≤ 2 × 10-4) auf Längenskalen von wenigen 100 nm oder darunter vorliegen. Auf einer Probe werden teilweise dehnungsrelaxierte Stellen nachgewiesen. Es wird außerdem belegt, dass die Eisenpufferschicht eine sehr wichtige Rolle bei der Realstruktur der Ba122-Schicht spielt. Abschließend wird HREBSD mit konventioneller Auswertung der Beugung rückgestreuter Elektronen (EBSD) verglichen. Dabei wird ermittelt, dass mit großem Aufwand und mit optimalen experimentellen Bedingungen auch durch konventionelle Auswertung der EBSD-Pattern Orientierungsunterschiede mit einer Genauigkeit, die besser ist als 0,1°, gemessen werden können. In der Gegenwart von Probenstellen mit unterschiedlichen Dehnungszuständen ergeben sich jedoch deutliche Abweichungen im Gegensatz zu der exakten Auswertung durch HREBSD.

Distributed cross-layer scalable multimedia services over next generation convergent networks : architectures and performances

Multimedia services are the killer applications on next generation convergent networks. Video contents are the most resource consuming part of a multimedia flux. Video transmission, video multicast and video conferencing services are the most popular types of video communication with increasing difficulty levels. Four main parts of the distributed cross-layer scalable multimedia services over next generation convergent networks are considered in this research work, both from the architecture and performance point of views. Firstly, we evaluate the performance of scalable multimedia transmissions over an overlay network. For that, we evaluate the performance of scalable video end-to-end transmissions over EvalSVC. It is capable of evaluating the end-to-end transmission of SVC bit-streams. The output results are both objective and subjective metrics of the video transmission. Through the interfaces with real networks and an overlay simulation platform, the transmission performance of different types of SVC scalability and AVC bit-streams on a bottle-neck and an overlay network will be evaluated. This evaluation is new because it is conducted on the end-to-end transmission of SVC contents and not on the coding performance. Next, we will study the multicast mechanism for multimedia content over an overlay network in the following part of this PhD thesis. Secondly, we tackle the problems of the distributed cross-layer scalable multimedia multicast over the next generation convergent networks. For that, we propose a new application-network cross layer multi-variable cost function for application layer multicast of multimedia delivery over convergent networks. It optimizes the variable requirements and available resources from both the application and the network layers. It can dynamically update the available resources required for reaching a particular node on the ALM's media distribution tree. Mathematical derivation and theoretical analysis have been provided for the newly proposed cost function so that it can be applied in more general cases of different contexts. An evaluation platform of an overlay network built over a convergent underlay network comprised of a simulated Internet topology and a real 4G mobile WiMAX IEEE802.16e wireless network is constructed. If multicast is the one-to-many mechanism to distribute the multimedia content, a deeper study on the many-to-many mechanism will be done in the next part of the thesis through a new architecture for video conferencing services. Thirdly, we study the distributed cross-layer scalable video conferencing services over the overlay network. For that, an enriched human perception-based distributed architecture for scalable video conferencing services is proposed with theoretical models and performance analysis. Rich theoretical models of the three different architectures: the proposed perception-based distributed architecture, the conventional centralized architecture and perception-based centralized architecture have been constructed by using queuing theory to reflect the traffic generated, transmitted and processed at the perception-based distributed leaders, the perception-based centralized top leader, and the centralized server. The performance of these three different architectures has been considered in 4 different aspects. While the distributed architecture is better than the centralized architecture for a scalable multimedia conferencing service, it brings many problems to users who are using a wireless network to participate into the conferencing service. A special solution should be found out for mobile users in the next part of the thesis. Lastly, the distributed cross-layer scalable video conferencing services over the next generation convergent network is enabled. Phylogenetic Analysis of Subtribe Alopecurinae (Poaceae)

Subtribe Alopecurinae (Poeae, Poaceae) sensu lato‘s seven genera share interesting morphological similarities (dense spicate panicles and one-flowered spikelets) that were widely thought to have a common origin. However, recent molecular evidence for three of the genera has suggested that the subtribe may be polyphyletic. To test this, five DNA regions were sequenced and analyzed using phylogenetic methods. Results confirm that Alopecurinae s.l. as presently treated is polyphyletic and should be dissolved. Additionally, the genus Cornucopiae may be just another Alopecurus. Limnas and Pseudophleum are not closely allied to Alopecurus or each other, and are even further from Phleum. Phleum is a distinct lineage that is not closely allied to any other included Alopecurinae genus. Evidence for revising infrageneric classifications of Alopecurus and Phleum is presented, as is evidence for separating A. magellanicus into two or more subspecies.

Creativity as a crucial process in the development of the young child

This study was conducted in order to assess the effects of creative stimulation in early childhood. The quantitative quasi-experimental study was set out to test a target group of 8 children in grade R, ranging in age from five to six years, to determine the importance of stimulation and intervention of creativity from a young age. A pre-test, evaluated the initial level of each child’s creative skills in fluency, flexibility and originality. This was followed by selecting one class as the experimental group and another class, from the same demographical location, as the control group. The experimental group underwent a six-week intervention programme which focused on the primary concepts in each area of fluency, flexibility and originality. Both groups continued with their normal school learning programmes. The results indicate that the impact of stimulation on young children was significant and that there was a great difference in the abilities and understanding of concepts in those children whohad been stimulated. / Teacher Education / M.
