Global Cerebral Ischemia in Male Long Evans Rats Impairs Dopaminergic/ΔFosB Signalling in the Mesocorticolimbic Pathway Without Altering Delay Discounting Rates

Global cerebral ischemia (GCI) in rats has been shown to promote exploration of anxiogenic zones of the Elevated-Plus Maze (EPM) and Open Field Test (OFT). This study investigated changes in impulsive choice and/or defensive responses as possible contributors of heightened anxiogenic exploration observed after ischemia. Impulsivity was assessed using delay discounting (DD) paradigms, while the Predator Odour Test (PO) served to assess changes in defensive responses towards a naturally aversive stimulus. Male Long Evans rats underwent 9 days of autoshaping training and 24 days of DD training prior to GCI or sham surgery (n= 9/group). Post-surgery, rats completed the OFT, EPM, and PO, followed by 6 days of DD sessions. Blood droplets served to evaluate corticosterone secretion associated with PO exposure. With impulsivity being regulated through mesocorticolimbic monoaminergic pathways, we also characterized post-ischemic changes in the expression of dopamine D2 receptors (DRD2), dopamine transporters (DAT), and ΔFosB in the basolateral amygdala (BLA), nucleus accumbens core (NAcC) and shell (NAcS), and ventromedial prefrontal cortex (vmPFC) using immunohistofluorescence. Our findings revealed no impact of GCI on delay discounting rates, while PO approach behaviours were minimally affected. Nonetheless, GCI significantly reduced DRD2 and ΔFosB-ir in the NAcS and NAcC, respectively, while DAT-ir was diminished in both NAc subregions. Collectively, our findings refine the understanding of cognitive-behavioural and biochemical responses following stroke or cardiac arrest. They support significant alterations to the dopaminergic mesocorticolimbic pathway after ischemia, which are not associated with altered impulsive choice in a DD task but may influence locomotor exploration of the OFT and EPM.

An Exploration of Female Reproductive Plasticity in the Context of Strange Male Exposure and Stress in Mice (Mus musculus)

<p>Females experience reproductive plasticity in several situations. Under stress, females can experience implantation failure. In the presence of unfamiliar males, females can also experience implantation failure (Bruce effect), or hastened sexual maturation (Vandenbergh effect). Sexual maturation and implantation failure are also induced by 17β-estradiol (E<sub>2</sub>). Male mice excrete E<sub>2</sub> in their urine; furthermore, stress can increase endogenous E<sub>2</sub> in females. Accordingly, I explored the role of male urinary E<sub>2</sub> in the Bruce and Vandenbergh effects, and examined E<sub>2</sub> and progesterone (P<sub>4</sub>) during stress-induced implantation failure in mice. I showed that a male mouse’s ability to induce the Bruce and Vandenbergh effects depends on his urinary E<sub>2</sub>, as castration removed this ability,and injections of E<sub>2</sub> restored it and urinary E<sub>2</sub> levels. I also developed urinary measurements of the stress hormone, corticosterone, for use in subsequent stress studies. I showed that serum and urinary corticosterone follow a similar circadian rhythm, and that urinary corticosterone responds appropriately to acute stress in male mice. Also, urinary and serum corticosterone, P<sub>4</sub>, and E<sub>2</sub> in female mice show similar responses to stress, with slightly different time courses. Lastly, I showed that rat-exposure is stressful and causes implantation failure in female mice. Furthermore, P<sub>4</sub> suppressed and E<sub>2</sub> is elevated in stressed females losing their pregnancies. Exogenous P<sub>4</sub> did not mitigate stress-induced pregnancy failure, unless it was combined with a low dose of E<sub>2</sub>. Taken together, I suggest that exogenous and endogenous E<sub>2</sub> contribute to reproductive plasticity in female mice, particularly their ability to abandon early pregnancy.</p> / Doctor of Philosophy (PhD)

Abiotic Factors Underlying Stress Hormone Level Variation Among Larval Amphibians

Anthropogenic disturbances can alter the abiotic composition of freshwater systems. These compositional changes can act as physiological stressors towards system inhabitants. However, little is known about how these altered abiotic factors influence stress hormones (corticosterone) in larval amphibians. Throughout the following chapters, I examined the effects of several abiotic factors on baseline and stress-induced corticosterone levels in the larvae of four amphibian species: Jefferson salamander (Ambystoma jeffersonianum), spotted salamander (A. maculatum), wood frog (Rana sylvatica), and grey treefrog (Hyla versicolor). Chapter II examined corticosterone level differences throughout development in A. jeffersonianum and R. sylvatica larvae under field, mesocosm, and laboratory venues. Baseline corticosterone levels in R. sylvatica increased near metamorphic climax in all venues, but not in A. jeffersonianum. Rather, baseline corticosterone levels differed with respect to venue throughout development in A. jeffersonianum. Chapter III examined corticosterone level differences among free-living A. jeffersonianum populations and possible abiotic factors underlying these hormone differences. Corticosterone levels significantly differed across populations. Increased baseline corticosterone levels significantly correlated to low pH. There was also a trend for increased baseline corticosterone levels to be positively correlated with chloride levels and negatively correlated with conductivity. Chapter IV examined the effects of laboratory manipulated pH on corticosterone levels in A. jeffersonianum, A. maculatum, R. sylvatica, and H. versicolor. There was a significant correlation between increased baseline corticosterone levels to low pH in all four species. Prey consumption (in both Ambystoma species) and survival (in A. jeffersonianum, A. maculatum, and R. sylvatica) were also negatively correlated to low pH. Chapter V examined the effects of increased conductivity on corticosterone levels in A. jeffersonianum, R. sylvatica, and H. versicolor. Increased conductivity exposure significantly correlated to increased baseline corticosterone levels in A. jeffersonianum and R. sylvatica. Prey consumption in A. jeffersonianum was also negatively correlated to increased conductivity. My dissertation shows that abiotic factors, such as pH and conductivity, can influence corticosterone levels in larval amphibians. These results suggest that corticosterone levels in larval amphibians may be a suitable biomarker reflective of altered freshwater habitat quality. However, my results also suggest that one should use a high degree of caution when using corticosterone levels in larval amphibians as a means to infer the health status of a population. / Ph. D.

Functional Dysregulation in Stress-Induced Modulation of Synaptic Plasticity in a Mouse Model of Fragile X Syndrome

The fragile X mental retardation protein (FMRP) is an important regulator of protein translation, and a lack of FMRP expression leads to a cognitive disorder known as fragile X syndrome (FXS). Clinical symptoms characterizing FXS include learning impairments and heightened anxiety in response to stressful situations. The Fmr1-/y mouse has previously been shown to have deficits in context discrimination and novel object recognition tasks, which primarily rely on the dentate gyrus (DG) region of the hippocampal formation, but not in the Morris water maze (MWM) or the elevated plus-maze tasks, which primarily depend on the Cornu Ammonis (CA1) region. Furthermore, previous research has demonstrated N-methyl-D-aspartate receptor (NMDAR)-associated synaptic plasticity impairments in the DG but not in the CA1. However, the impact of acute stress on synaptic plasticity in the Fmr1-/y hippocampus has not been examined. The current study sought to extend previous behavioural investigations in the Fmr1-/y mouse, as well as examine the impact of stress on activation of the hypothalamic-pituitary-adrenal (HPA)-axis and on hippocampal synaptic plasticity. To further characterize hippocampus-dependent behaviour in this mouse model, the DG-dependent metric change spatial processing and CA1-dependent temporal order discrimination tasks were evaluated. The results reported here support previous findings and demonstrate that Fmr1-/y mice have performance deficits in the DG-dependent task but not in the CA1-dependent task, suggesting that previously reported subregional differences in NMDAR-associated synaptic plasticity deficits in the hippocampus of the Fmr1-/y mouse model may also manifest as selective behavioural deficits in hippocampus-dependent tasks. In addition, following acute stress, mice lacking FMRP showed a faster elevation of the glucocorticoid corticosterone and a more immediate impairment in long-term potentiation (LTP) in the DG. Stress-induced LTP impairments were rescued by administering the glucocorticoid receptor (GR) antagonist RU38486. Administration of RU38486 also enhanced LTP in Fmr1-/y mice in the absence of acute stress to wild-type levels, and this enhancement was blocked by application of the NMDAR antagonist 2-amino-5-phosphonopentanoic acid. These results suggest that a loss of FMRP results in enhanced GR signalling that may adversely affect NMDAR-dependent synaptic plasticity in the DG. Finally, synaptic plasticity alterations reported in this work were found to be specific to the DG and were unidirectional, i.e., restricted to LTP, as NMDAR- and metabotropic glutamate receptor (mGluR)-LTD were both unaffected by acute stress in the DG or the CA1 regions. This study offers new insights into synaptic plasticity impairments in the Fmr1-/y mouse model, and suggests stress and GRs as important contributors to learning and memory deficits in FXS. / Graduate

Does a high fundamental corticosterone concentration negatively affect the ornaments and visible sexual traits of male house sparrows (Passer domesticus)?

The male house sparrows has two assumed visible sexual traits which are 1) the colouration (or brightness) of the beak and 2) the size of the black badge on the bird’s throat and chest. Earlier research has shown that the testosterone concentration play an important part in determine the beak colour, with a higher testosterone concentration giving the bird a darker beak and as such making the male more attractive to females.     Even though stress has been counted for as a changing factor of phenotypes and behaviour it is not certain how vast effect stress has on different animals. This study is focusing on how and if the individuals fundamental stress level (fundamental corticosterone concentration) is affecting the morphological appearance, in this case the visible sexual traits (beak colour and badge size) and mask (an indicator of age), within the house sparrows in the Lundy Island population. In this study there was no relationship between beak colour and corticosterone concentrations, stating that beak brightness is not determined by fundamental stress. Likewise there was no relationship between the corticosterone concentrations and the size of the badge. Mask measurements in relationship to corticosterone showed no significant effect. These results indicate that there is no relationship between the corticosterone concentration in the blood and the mask size or the beak colour and badge size as visible sexual traits. / Många forskare är ense om att gråsparvshannen använder sig av två synliga karaktärsdrag för att attrahera honor. Den första är mörkhetsgraden på näbben och den andra är storleken på hannens svarta bröstlapp, placerad på hals och bröst. Tidigare forskning har visat att näbbfärgen påverkas kraftigt av hannens testosteron nivå och att en högre testosteron nivå leder till en mörkare näbb, vilket är mer attraktivt för honorna.    Även om stress har visat sig påverka både fenotyp och beteende hos många olika djurgrupper är det fortfarande oklart hur stor effekt stress har på olika djur. Denna studie fokuserar på hur och om den individuella fundamentala stressnivån (den fundamentala kortikosteron nivån) påverkar de synliga attraherande karaktärsdragen (näbbfärg och bröstlappsstorlek) hos gråsparvshannarna. I denna rapport analyseras även hannarnas masker, vilka fungerar som åldersindikatorer, i relation till de individuella kortikosteron koncentrationerna. Inget samband kunde hittas mellan näbbfärg och kortikosteron nivå hos de analyserade individerna. Likaså kunde inget samband hittas mellan kortikosteron nivån och storleken på hannens bröstlapp. Inte heller maskstorleken visade samband med kortokosteron nivån. Resultatet indikerar således att stress inte påverkar hannens sexuellt attraktiva karaktärsdrag eller mask.

O N-metil- 3-4, metilenodioximetanfetamina (MDMA) ou ecstasy tem sido freqüentemente usado por jovens. Analisamos neste trabalho, dentro de uma perspectiva neuroimune, os efeitos da administração aguda de MDMA em parâmetros comportamentais, neuroendócrinos, hematatológicos e imunes de camundongos Balb/C, usando para esta última finalidade um modelo de asma experimental. O MDMA produziu: 1- aumento diferencial da atividade locomotora nas diferentes zonas do campo aberto; aumento na locomoção total e diminuição da atividade exploratória no hole-board; aumento da porcentagem de entradas e da taxa de permanência nos braços abertos do LCE; aumento no tempo gasto na caixa de saída e diminuição do número de acessos de risco em uma caixa de exposição a um predador; 2- aumento dos níveis séricos de corticoterona; 3- aumento dos níveis de noradrenalina no estriado e córtex frontal, aumento nos níveis de dopamina e de DOPAC no estriado, diminuição dos níveis de DOPAC corticais, aumento de 5-HT e 5-HIAA no estriado, diminuição dos níveis de 5- HIAA e do turnover de serotonina no hipotálamo e diminuição do \"turnover\" de dopamina no estriado e córtex frontal; 4- alteração na migração de leucócitos em camundongos alérgicos com diminuição da porcentagem de linfócitos e monócitos circulantes, diminuição do número de granulócitos no lavado bronco alveolar (LBA), efeitos estes que foram revertidos pelo pré-tratamento com RU-486; 5 redução da expressão de L-selectinas por monócitos e tendência de redução da expressão de L -selectinas por neutrófilos no pulmão; 6- diminuição das produções espontâneas de IL-4, IL-5 e IL-10 e de IL-4 em cultura estimulada com LPS; 7- redução da contração da traquéia isolada de animais alérgicos e 8- redução da desgranulação dos mastócitos em brônquios intrapulmonares. Sugeriu-se que o estresse/ansiedade induzidos pelo MDMA tenham ativado o eixo HHA e/ou do sistema nervoso autonômico simpático dos camundongos, alterando a resposta imune dos mesmos na vigência de um modelo de asma. A inflamação alérgica pulmonar desponta, assim, como importante ferramenta para o entendimento da ação de drogas de abuso em processos neuroimunológicos. / The N-metil- 3-4, metilenodioximetanfetamina (MDMA) or ecstasy is a drug widely used amongst young people. This study was undertaken to analyze, under a neuroimmune perspective, the effects of acute MDMA administration on behavioral, neuroendocrine, hematological and immune parameters on Balb/C mice, using for the latter purpose the allergic lung inflammatory response model. It was observed that MDMA produced in mice: 1- a differential increase on total locomotion in the different open-field zones; an increase on total locomotion and a decrease on exploratory activity in the hole-board; an increase on both percentage of entrances and time spent on plus-maze open arms; an increase on time spent in the starting box and a decrease of risk assessments in a predator exposition box; 2- an increase in corticosterone serum levels; 3- an increase in striatal and frontal cortex noradrenaline levels, an increase in striatal dopamine and DOPAC levels, a decrease in cortical DOPAC levels, an increase in striatal 5-HT and 5-HIAA levels, a decrease in both 5-HIAA levels and 5-HT turnover rates in hypothalamus and a decrease in striatal and cortical dopamine \"turnover\" rates; 4- an alteration on leukocyte migration in allergic mice, i.e., decreased percentage of peripheral blood lymphocytes and monocytes, decreased number of granulocytes on bronchoalveolar lavage fluid ( LBA); these effects were reverted by previous RU-486 treatment; 5- a decrease in L-selectin expression by monocytes and a tendency towards a decrease in L-selectin expression by lung neutrophils; 6- a decrease on expontaneous production of IL-4, IL-5 e IL-10 and IL-4 in LPS-stimulated cultures; 7- a decrease in the contraction of allergic mice isolated trachea; and, 8- a decrease in bronchial mastocytes degranulation. It was suggested that MDMA-induced anxiety/stress symptoms increasing HHA-axis and/or the autonomic nervous system activities this leading to the immune changes observed presently in the allergic lung inflammation model of asthma used. This model, thus, emerges as a useful tool for the understanding of neuroimmune effects of drugs of abuse.

Efeitos do estresse por calor sobre a imunidade e a migração de Salmonela enteritidis em frangos de corte / Effects of heat stress on immunity and Salmonella enteritidis invasion in broiler chickens

O estresse é uma realidade na produção avícola mundial. Sabe-se que ambientes estressores prejudicam o bem-estar, os parâmetros produtivos e a imunidade de frangos de corte. Sabe-se, também, que o estresse por calor diminui a atividade de macrófagos em frangos de corte e, que existem, inúmeros estressores ambientais que insidem sobre a produção animal e podem aumentar a susceptibilidade às doenças. A Salmonella spp. é uma das maiores zoonoses do mundo, causando mais de 1 bilhão de casos de infecção. Nesse sentido, o presente trabalho analisa os efeitos do estresse por calor (31±1°C) sobre os índices zootécnicos, a imunidade, a invasão bacteriana e a integridade intestinal em frangos de corte infectados com Salmonella enteritidis; os dados obtidos foram discutidos dentro de uma perspectiva neuroimune. Os frangos foram divididos em quatro grupos: 1) Controle (C); 2) Estresse por Calor a 31±1 °C (HS31°C); 3) Controle infectados com Salmonella enteritidis (Controle Positivo [PC]) e; 4) Estresse por calor a 31±1 °C e infectados com Salmonella (PHS31°C). Nossos resultados mostraram que o estresse por calor em uma situação de infecção experimental por Salmonella enteritidis (grupo PHS31°C) 1) diminuiu os índices zootécnicos; especificamente, diminuiu o ganho de peso, consumo de alimento e a conversão alimentar; 2) diminuiu os níveis plasmáticos de INF-&gamma; e IgA; 3) diminuiu a expressão de, IL-6 e IL-12 em baço e diminui IL1- &beta;, IL-10 e TGF-&beta; em tonsila cecal; 4) diminuiu a expressão de AvBD-4 e AvBD-6 em tonsila cecal e; 5) diminuiu a expressão de TLR-2 em baço e tonsila cecal. Observamos, também, 6) aumento dos níveis séricos de corticosterona nos animais dos grupos HS31°C e PHS31°C e; 7) piora no quadro de enterite produzida pela Salmonella enteritidis, quando os animais foram estressados por calor, caracterizando-se uma enterite moderada ao longo de todo o intestino delgado. Finalmente, 8) observamos que o estresse por calor aumentou a migração de Salmonella enteritidis para baço das aves do grupo PHS31°C, porém esse aumento não foi observado no fígado; observamos, também, presença de Salmonella na medula osssea dos animais estressados e infectado com essa bactéria. Os dados obtidos sugerem que a somatória dos fatores estresse por calor e infecção por Samonella prejudicou os parâmetros produtivos, a integridade intestinal, a imunidade e, em especial a ativação e atividade de macrófagos, possibilitando um aumento da migração de Salmonella enteritidis para o baço e medula óssea dos frangos de corte. Neste sentido, o estresse por calor teria prejudicado a qualidade da barreira imune intestinal, via ativação do eixo HPA e aumento dos níveis de corticosterona, diminuindo a imunidade inata proporcionando a migração das bactérias patogênicas através da mucosa intestinal para o baço e a medula óssea das aves estressadas. / Stress is a reality in the world poultry production. Environmental stressors impair both welfare, performance parameters and immunity in broiler chickens. Heat stress decreases macrophage activity in broiler chickens and many environmental stressors that impact animal production increases animal\'s susceptibility to diseases. Samonella spp is one of the most endemic zoonotic diseases of the world, inducing more than 1 billion infection cases per year. In this way, we studied the effects of 31±1°C heat stress on performance parameters, immunity, bacteria invasion and intestinal integrity in broiler chickens experimentally infected with Salmonella enteritidis; the data were discussed under a neuroimmune perspective. The broiler chickens were divided into four different groups: 1) Control group (C); 2) 31±1 °C heat stressed group (HS31°C); 3) Control group infected with Salmonella enteritidis Positive control (PC) and; 4) 31±1 °C heat stressed and Salmonella enteritidis infected group (PHS31°C). We showed the heat stress applied in the course of Salmonella enteritidis infection (PHS31°C group) decreased poultry performance parameters; specifically, it decreased the body weight gain, the feed intake and the food conversion; 2) decresead INF-&gamma; and IgA plasmatic levels; 3) decreased the mRNA expression of IL-6 and IL- 12 in spleen and the mRNA expression of IL1-&beta;, IL-10 and TGF-&beta; in cecal tonsil; 4) decreased the mRNA expression of AvBD-4 and AvBD-6 in cecal tonsil and; 5) decreased the mRNA expression of TLR-2 in spleen and cecal tonsil. We also observed 6) an increase in corticosterone serum levels in the animals of the HS31°C and the PHS31°C groups and, 7) more severe intestinal inflamation produced by Salmonella enteritidis in heat stressed chickens, characterized by a moderate enteritis throughout all the small intestine mucosa (PHS31°C group). Finally, 8) we showed that the heat stress increased splenic Salmonella enteritides invasion in PHS31°C broiler chickens; we also observed the presence of Salmonella in the bone marrow of stressed and infected broiler chickens. These data suggest that heat stress and Salmonella infection working together impair chicken\'s performance parameters, intestinal integrity and immunity (specially the macrophage activity), increasing ate the same time splenic and bone marrow Salmonella enteritidis invasion. Thus, heat stress could have impared the intestinal immunity barrier quality, via HPA axis activation and corticosterone serum levels release, decreasing the inate immunity and, providing pathogenic bacteria migration through the intestinal mucosa for spleen and bone marrow of the heat stressed chickens.

Papel da corticosterona na vigência do estresse sobre a função pineal em ratos. / The role of corticosterone in the presence of stress upon the pineal function in rats.

O objetivo geral da presente tese é testar a hipótese que a pineal faz parte integrante da resposta ao estresse, da mesma forma que está integrada ao processo inflamatório. Dois modelos de estresse, contenção e frio, foram aplicados aos ratos por 30 min ou 2 horas. Os resultados foram: ausência de úlceras gástricas e TNF em nível sérico por ambos os estresses, apenas aumento de corticosterona; somente em duas horas qualquer um dos estresses aumentou significativamente melatonina da pineal; tratamento tanto com metirapona como com mifepristone aboliram indistintamente os efeitos dos estresses; tratamento tanto com talidomida como com fenilefrina não modificaram os efeitos dos estresses. Concluímos que, o estresse moderado agudo, pela ação da corticosterona, promove modulação da pineal na dependência do estado fisiológico da mesma. Confirmamos a existência de uma relação entre as glândulas adrenais e pineal. Afirmamos que a glândula pineal exerce, além de suas clássicas funções cronobióticas, um papel de grande sensor do estado geral ao organismo inteiro. / The objective of this thesis is to test the hypothesis that the pineal is a player on stress response, likewise that it is one player in inflammatory process. Two stress models, restraint and cold, were applied to the rats for 30 min or 2 hous. The results were: absence of gastric ulcers and TNF serum levels in both stresses, just enhancement of corticosterone plasma levels; only in two hours anyone stress increased significantly pineal melatonin; metyrapone or mifepristone treatment abolish indistinctly the effects of the stresses; thalidomide or phenylephrine treatment did not modify the effects of the stresses. We conclude that the acute moderate stress by the corticosterone action promote modulation of pineal on the dependence of the physiological status of itself. We confirm the existence of a network between the adrenal and pineal glands. We affirm that the pineal gland performs, beyond of its chronobiotical classical functions, one role of the great sensor of internal body state to the whole organism.

A desnutrição no período gestacional pode comprometer o desenvolvimento fetal e a maturação do sistema imune. Ratos Wistar de 12 semanas com baixo peso ao nascer, oriundos de mães desnutridas durante a gestação, apresentam resposta inflamatória pulmonar reduzida, e concentração de corticosterona circulante elevada. É possível concluir que a corticosterona está envolvida com a reduzida resposta inflamatória pulmonar desenvolvida por animais de baixo peso ao nascer. / The malnutrition during pregnancy may impair the fetal development and the maturation of immune system. Wistar rats with 12 weeks-old with low birth weigt present reduced lung inflammatory response, and increased circulating levels of corticosterone. This model was subjected to adrenalectomy and corticosterone replacement in phyological levels, and the acute lung injury by LPS was induced. The results obtained showed an increase in neutrophil infiltration into lung tissue, and increase of inflammatory mediators after reduction of corticosterone in low birth weight rats. The lung expression of glucocorticoid receptors was reduced in low birth weight rats, however, after reduction of corticosterone, the expression of receptors was incerased. The data showed that high levels of corticosterone are related to reduced acute lung inflammatory response developed by low birth weght rats.

Investigating the role of corticosterone in meal anticipatory behaviour, metabolism and glucosetolerance

Daily rhythms in physiology and behaviour are orchestrated by theautonomously rhythmic cells of the suprachiasmatic nucleus (SCN).Restricting food intake to the rest phase of nocturnal rodents, leads to thedevelopment of meal anticipatory behaviour, corticosterone and bodytemperature. Given that lesions to the SCN fail to abolish meal anticipation, asecond oscillator of unknown location, referred to as the food-entrainableoscillator (FEO) is thought to exist. Although the site of the FEO is unknown,several hypothalamus nuclei, including the dorsomedial hypothalamus(DMH) are thought to play a role in meal anticipation. Given thatadrenalectomy is reported to abolish meal anticipation, an intact HPA axis isalso thought to contribute to the functioning of the FEO. Some forms ofobesity are characterised by high basal levels of circulating corticosterone. Inaddition, limited access to high fat diet, fails to induce the development ofrobust meal anticipation in rats. During our initial studies, the effect of a standard and 45% high fat diet onthe development of meal anticipatory behaviour and hypothalamic c-Fosexpression were investigated. Restricted access to high fat diet led toattenuation of meal anticipation compared to those fed standard diet. Thiswas concurrent with a failure to develop an anticipatory rise in DMH c-Fosexpression. A meal anticipatory rise in corticosterone is thought to benecessary for the presence of meal anticipation as well as adaptation ofmetabolism to daily restricted feeding. In the next set of studies, weconfirmed that restricted access to standard diet leads to the development ofa meal anticipatory rise in plasma corticosterone. In contrast we observed adramatic post-anticipatory rise in plasma corticosterone in rats givenrestricted access to the 45% high fat diet. We hypothesised that the highcorticosterone levels resulting from high fat diet were a contributing factor tothe lack of meal anticipation in high fat fed rats. With the aid of apharmacokinetic study, a suitable experiment was designed for daily dosingof a potent glucocorticoid receptor antagonist, RU486, with the aim ofrescuing meal anticipation in high fat fed rats. Interestingly, treatment withRU486 successfully rescued meal anticipation in high fat fed rats, butattenuated meal anticipation in standard diet restricted fed rats. In the finalseries of studies the effect of diet and feeding regime on glucose toleranceand metabolism were investigated. High fat feeding was found to reduceglucose tolerance in both ad lib and restricted fed rats, with RU486 treatmentimproving glucose tolerance in a time dependant manner. Restricted accessto food was found to induce post satiation lipogenesis in both standard dietfed and to a lesser extent in high fat fed rats, an effect which may bebeneficial in reducing obesity. Overall the results provide further insight intothe complex role of corticosterone in promoting or preventing mealanticipatory behaviour. An anticipatory rise in plasma corticosterone isrequired for meal anticipation, as repeated daily dosing of RU486 inhibit mealanticipation. The high basal levels of corticosterone in high fat fed rats mayprevent meal anticipation, insulin secretion and post-satiation lipogenesiswhich may in fact be a homeostatic mechanism to prevent obesity. Nonetheless, treatment with RU486 rescues behavioural meal anticipationand glucose tolerance.
