Identification of cellular factors involved in herpes simplex virus type 1 nucelar egressMaric, Martina 01 July 2012 (has links)
The herpesvirus life cycle involves a step where newly formed capsids leave the nucleus by translocating across the intact nuclear envelope (NE). Little is known about the role of cellular factors during nuclear egress. We sought to identify novel cellular proteins that interact with the conserved herpes simplex virus-1 (HSV-1) pUL34 by performing a yeast two-hybrid screen. pUL34 was chosen due to its crucial and multifunctional role during nuclear egress. From 42 cellular factors that interacted with pUL34 in yeast, twelve were further evaluated in mammalian cells by co-localization studies using immunofluorescence. No specific co-location between the tested cellular factors and pUL34 was observed in infected cells, thus the screen failed to convincingly identify novel pUL34 interactors. In the second part of the thesis we addressed the functional significance of the cellular protein torsinA (TA) in the HSV-1 life cycle. We became interested in TA due to its role in maintaining normal NE morphology. We showed that perturbing the normal function of TA through overexpression impaired HSV-1 replication and caused a defect in capsid nuclear egress. In mouse embryonic fibroblasts that failed to express TA (TA-/-MEFs), HSV-1 replication was also inhibited, but a defect in capsid nuclear egress was not apparent. Strikingly, infection in TA-null MEFs induced a NE breakdown, the extent of which was dependent on viral products involved in nuclear egress. The viral growth defect and NE envelope breakdown, however, seem to be TA-null cell line specific rather than a functional consequence of TA loss as indicated by TA-/-MEFs reconstituted with TA and 293T with reduced TA levels. In conclusion, overexpression and loss of TA have different effects on the HSV-1 life cycle.

Simulating ingress and egress motion for heavy earthmoving machinesKwon, Hyun Jung 01 December 2011 (has links)
Design of heavy earthmoving equipment is based primarily on feedback from drivers. Most design studies on ingress and egress focus on the motion itself and rely heavily on experimental data. This process requires physical construction of expensive (in terms of time and money) mockups before any feedback can be obtained. Post-feedback design changes and the analysis of those changes are again expensive processes. Although the design of heavy vehicles requires consideration of human safety and comfort, very little attention has been given to simulating ingress and egress movement for these vehicles. This thesis describes the development of a virtual model to perform ingress and egress motions for heavy equipment and its applications to study the responses of operators with different anthropometries to different cab designs. Different performance measures are suggested and used with predictive dynamics to study human performance since human motion is not governed by a single performance measure. Optimizing multiple performance measures allows the full range of motion for all 55 degrees of freedom to be considered for simulating the task. Once the relevant performance measure was established, case studies were performed on seven different cab designs and digital human models with three different anthropometries. Finally, several different cab design metrics for propensity of injury, comfort, and accessibility were proposed. These design metrics were evaluated for each of the case studies. Finally, each cab design was ranked based on the design metrics to identify the best design for a range of anthropometries. These results help designers make decisions and plan further design changes.

Recently, several major incidents occurred in worldwide metro and subway systems due to inadequate or inappropriate emergency procedures.
In this study, the traditional and innovative metro underground stations were analyzed utilizity the CFAST and FDS computer program. A comparative study has been conducted to evaluate the smoke descending rate, and temperature distribution, etc with addressable zoned smoke control system.
Dynamic egress analysis was followed to validate the NFPA 130 which was taken as the design guidelines to evaluate the evacuation time using SIMULEX program.
Finally, the smoke management design and egress system was integrated, which warrants its qpplication in actual engineering designs.

Almost people hurt or die because of heavy smoke when the building fires. Smoke Management is to keep the smoke in the limited zone or to change its direction and to keep the ways which the people escape bright. It is convenient for fireman to save them and to keep their life and to reduce the money loss.
In the developed country of the world, smoke management design is one of the most important factors to affect egress system of the building. Effective smoke management design is to avoid the smoke diffusing or descending and to make the people have enough time to escape. The combination of smoke management and egress system using performance-based fire safety design should be developed safely and economically.
In Taiwan,we haven¡¦t the proper law of smoke management and egress system about the atria of the large space. So we must progress the computer simulation about smoke management and egress system and then use full-scale experiment to perform.
The major terms of the content are:
1.the optimal distribution zone analysis of smoke management of the building.
2.to design and to analyze smoke management of the atria.
3.the combination of design analysis of smoke management and egress system.
4.full-scale experimental investigation.
We hope that the performance-based fire safety design method about smoke management and egress system of the large shopping mall could be the excellent model and enchance the safety of the building in Taiwan.

Validation of EvacuatioNZ Model for High-Rise Building AnalysisTsai, Wei-Li January 2007 (has links)
This thesis covers a variety of analytical approaches that validate the use of the EvacuatioNZ model on high-rise building analysis. Through performing a number of sensitivity analyses, several model deficiencies as well as functional limitations were improved upon and part of the model developments are continued based on the previous research done by two Master's students at the University of Canterbury. In this thesis, data from three evacuations were considered for different validating aspects. These evacuations were, a hypothetical 21-storey hotel building located in the United States of America, which was previously simulated using Simulex and EXIT89; a trial evacuation that was carried out in a 13-storey office building located in Canada; and a fire drill conducted at a 21-storey office building located in Australia. Overall, the results indicated that the EvacuatioNZ is able to produce reasonable predictions of the total evacuation time regardless of the number of floors involved. The component testing also showed satisfactory outcomes regarding the involvement of disabled occupants, complexity of node configurations, and different pre-movement time distributions. However, the current model still has a number of limitations that need to be verified and tested. These include the preferred route function and the connection problem for long stairs. Further research should also be carried out on the use of the Evacuation model on other types of building structures so as to increase the confidence level of utilizing the EvacuatioNZ model for general applications.

Pedestrian Leadership and Egress Assistance Simulation Environment (PLEASE)Feuz, Kyle D. 01 December 2011 (has links)
Over the past decade, researchers have been developing new ways to model pedestrian egress especially in emergency situations. The traditional methods of modeling pedestrian egress, including ow-based modeling and cellular automata, have been shown to be poor models of human behavior at an individual level, as well as failing to capture many important group social behaviors of pedestrians. This has led to the exploration of agent-based modeling for crowd simulations including those involving pedestrian egress. Using this model, we evaluate different heuristic functions for predicting good egress routes for a variety of real building layouts. We also introduce reinforcement learning as a means to represent individualized pedestrian route knowledge. Finally, we implement a group formation technique, which allows pedestrians in a group to share route knowledge and reach a consensus in route selection. Using the group formation technique, we consider the effects such knowledge sharing and consensus mechanisms have on pedestrian egress times.

Previous issue date: 2016-08-01 / A presente dissertação foi desenvolvida no âmbito do Mestrado Profissional em Gestão e Avaliação da Educação (PPGP) do Centro de Políticas Públicas e Avaliação da Educação da Universidade Federal de Juiz de Fora (CAEd/UFJF). Com o crescimento de investimentos públicos em educação profissional, com a expansão da Rede Federal de Educação Profissional e Tecnológica, faz-se necessário identificar se a proposta dessa política pública está de fato gerando oportunidades ao público que se destina. Um mecanismo de se monitorar tais efeitos é o acompanhamento de egresso, pois o ex-aluno é uma importante fonte de informação à gestão e avaliação de uma instituição de ensino. Porém, observou-se nos Institutos Federais uma falta desse acompanhamento – constatado em relatório emitido pelo Tribunal de Contas da União (2012) – e uma carência de estudos acerca do tema egressos no Brasil. Tal fato demonstra a fragilidade das Instituições de Ensino ao não gerenciarem e adequarem seu planejamento institucional. Diante desse contexto, esse trabalho propôs investigar a política de monitoramento de egressos de uma unidade do Instituto Federal no Ceará, na cidade de Caucaia. O estudo objetivou indagar aos atores institucionais quais os entraves existentes à aplicação da pesquisa de acompanhamento de egressos e suas implicações, além de coletar informações dos egressos que possibilitassem traçar um breve perfil destes. Para tanto, utilizou-se uma abordagem qualitativa e a técnica de estudo de caso. Foram utilizados como instrumentos de coleta de dados questionários e entrevistas semiestruturadas. A pesquisa demonstrou má formulação e entraves que dificultaram a implementação dessa política na unidade. A partir dessas considerações foi proposto um Plano de Ação Educacional (PAE) que possibilite um redesenho e implementação da política de monitoramento de egresso no IFCE Campus Caucaia. / The present dissertation was developed in the Professional Master in Management and Education Evaluation (PPGP) of the Center for Public Policy and Education Evaluation of Federal University of Juiz Fora (CAEd / UFJF). With the growth of public investment in vocational education, with the expansion of the Federal Vocational and Technological Education Network , it is necessary to identify whether the proposal that public policy is indeed generating opportunities to the public it is intended. A mechanism to monitor such effects is monitoring egress because the former student is an important source of information for the management and evaluation of an educational institution. However, there was a lack in the Federal Institutes of this monitoring - noted in a report issued by the Court of the Union (2012) - and a lack of studies on the topic graduates in Brazil. This shows the weakness of education institutions to manage and do not fit its institutional planning. On this context, this work proposed investigate the graduates monitoring policy of a unit of the Federal Institute in Ceara, in the city of Caucaia. The study aimed to investigate the institutional actors which existing barriers to the implementation follow-up survey of graduates and their implications, in addition to collecting information from graduates who made it possible to trace a brief profile of these. Therefore, if used a qualitative approach and case study technique. It used questionnaires as data collection instruments and semi-structured interviews. Research has shown poor formulation and barriers that hindered implementation of this policy in the unit. From these considerations proposed an Educational Action Plan (PAE) that enables a redesign and implementation of egress monitoring policy in IFCE in its Campus of Caucaia.

Plasmodium falciparum e Toxoplasma gondii são protozoários parasitas pertencentes ao filo Apicomplexa. Apirases são enzimas metabolizadoras de nucleotídeos extracelulares. Nesta tese mostramos pela primeira vez a presença de um membro desta família de enzimas em P. falciparum, o qual foi capaz de degradar ATP extracelular. Análises por RT-qPCR revelaram a expressão da apirase durante todo o ciclo intraeritrocítico. A adição de inibidores desta classe de enzimas foi capaz de prejudicar o desenvolvimento dos parasitas e a invasão de novas hemácias pelos merozoitos, sugerindo assim um papel da apirase nestes processos. A via de sinalização por Ca2+ é universal e vital para todas as células. Para melhor entender a fisiologia celular de P. falciparum construímos uma nova linhagem de parasitas transgênicos, PfGCaMP3, que nos tornam capazes de monitorar a dinâmica de Ca2+ sem o uso de protocolos invasivos de marcação. De modo semelhante utilizamos uma nova linhagem de T. gondii expressando de forma estável o indicador de Ca2+ GCaMP3 para estudar o papel deste íon na saída da célula. T. gondii possui o Ca2+ necessário para promover este processo, entretanto Ca2+ extracelular age como um fator intensificador neste passo essencial do ciclo lítico. / Plasmodium falciparum and Toxoplasma gondii are protozoan parasites that belong to phylum Apicomplexa. Apirases are metabolizing enzymes of extracellular nucleotides. In this work we show for the first time the presence of an apyrase in P. falciparum, which was able to degrade extracellular ATP. RTqPCR analysis revealed the expression of apyrase throughout the intraerythrocytic cycle. Addition of apyrase inhibitors was able to impair the development of the parasites and the invasion of new erythrocytes by merozoites, thus suggesting a role of apyrase in these processes. Calcium signaling is universal and vital to all cells. To better understand the cellular physiology of P. falciparum we construct a new strain of transgenic parasites, PfGCaMP3, which enable us to monitor the Ca2+ dynamics without using invasive protocols. Similarly we use a new strain of T. gondii that stably express the Ca2+ indicator GCaMP3 to study the role Ca2+ in parasite egress. T. gondii has the Ca2+ required to promote this process, however extracellular Ca2+ acts as an enhancer factor in this crucial step of the lytic cycle.

Previous issue date: 2012-09-12 / The Technological and Professional Education in Brazil, marked by the sign of the division
between manual and intellectual work, dating back to the early twentieth century, with the
establishment of Schools of Learning Crafters. This century of existence was marked by
advances and setbacks, struggles and hopes to make it effective like integral education. In the
last two decades, public policies, particularly, those arising from LDBEN/1996 brought new
incentives for this type of human and professional education, increasing the number of
institutions, courses and turns, finally, the prospect of effectuation as claimed extension and
access democratization and graduation. The Technological Courses gained centrality in the
expansion processes and many speeches indicate guarantees of employability. But in what
terms? This thesis, integrated to the Line of Research Education, Society and Culture,
proposed to interrogate which mediations are established between the technological training
and the employability, revealed by egress of the Systems Analysis and Technology
Development Course, of the IF Goiano Campus Urutaí, designed with an emphasis on the
notion of skills required to respond to the market conditions for the professional technologist
performance. Was the employability actually accomplished? To answer this question, it was
chosen to privilege the voices of egress, entering students from 2003 to 2008, 108 egresses,
which 65 accepted to participate of the research. The conducted reflections were addressed,
theoretically, in official documents and researchers like Antunes (2005), Cunha (1977, 2000),
Ciavatta (2005), Frigotto (1999, 2001, 2003, 2005, 2006, 2009, 2010), Kuenzer (2005, 2010),
Manfredi (2002), Ramos (2011), Saviani (2005), among others. In the research design of
quantitative and qualitative methodological matrix, the following procedures have been
defined for the empirical basis: analysis of official documents (national and institutional),
interviews (01 Course Coordinator, 04 managers and 02 Attendant) and a questionnaire
(Egress). Secondary data were provided by the Department of Higher Education of the
Institution, locus of the research. The egress revelations, by the purpose of employability,
proved the marketing optimism of the job security is not always effective. Among 60.19%
(65) egress, it was found that 93.84% (61) are in the labor market, which, at first, it revealed
high employability. However, the data presented have complained that the 61 egress who
reported being in the labor market, only 29.51% (18) are exerting activities inherent to their
training area. Thus, in reality, the marketing "promises" disseminated by the public policies
have not been obeyed in the Systems Analysis and Technology Development Course of the IF
Goiano Câmpus Urutaí. / A Educação Profissional e Tecnológica no Brasil, marcada pelo signo da divisão entre
trabalho manual e intelectual, remonta ao início do século XX, com a criação das Escolas de
Aprendizes Artífices. Esse século de existência foi marcado por avanços e retrocessos, lutas e
esperanças para efetivá-la como educação integral. Nas duas últimas décadas, políticas
públicas, em especial, as decorrentes da LDBEN/1996, trouxeram novos estímulos a esta
modalidade de formação humana e profissional, ampliando o número de instituições, cursos e
turnos, enfim, a perspectiva de efetivação da tão reivindicada ampliação e democratização do
acesso e diplomação. Os Cursos Tecnológicos ganharam centralidade nos processos de
expansão e muitos discursos apontam para garantias de empregabilidade. Mas, em que
termos? Esta tese, integrada à Linha de Pesquisa Educação, Sociedade e Cultura, propôs-se a
interrogar quais mediações são estabelecidas entre a formação tecnológica e a
empregabilidade, reveladas pelos egressos do Curso Superior de Tecnologia em Análise e
Desenvolvimento de Sistemas do IF Goiano Câmpus Urutaí, concebido com ênfase na noção
de competências, exigidas para responder às condições mercadológicas para atuação
profissional do Tecnólogo. A empregabilidade de fato se efetivou? Para responder esta
questão, optou-se por privilegiar as vozes dos egressos, ingressantes de 2003 a 2008, 108
diplomados dos quais 65 aceitaram participar da pesquisa. As reflexões realizadas foram
aportadas, teoricamente, em documentos oficiais e pesquisadores como Antunes (2005),
Cunha (1977, 2000), Ciavatta (2005), Frigotto (1999, 2001, 2003, 2005, 2006, 2009, 2010),
Kuenzer (2005, 2010), Manfredi (2002), Ramos (2011), Saviani (2005) dentre outros. No
delineamento da pesquisa de matriz metodológica qualitativa e quantitativa, foram definidos
os seguintes procedimentos para a base empírica: análise de documentos oficiais (nacionais e
institucionais), entrevistas (01 Coordenador de Curso, 04 Gestores e 02 Servidores) e
questionário (Egressos). Dados secundários foram fornecidos pela Secretaria de Ensino
Superior da Instituição, locus da pesquisa. As revelações dos egressos, a propósito da
empregabilidade, comprovaram que o otimismo mercadológico da garantia de emprego, nem
sempre é efetivado. Dos 60,19% (65) egressos, constatou-se que 93,84% (61) estão no
mercado de trabalho, o que, a princípio, revelou alta empregabilidade. No entanto, os dados
apresentados denunciaram que dos 61 egressos que declararam estar no mercado de trabalho,
somente 29,51% (18) estão exercendo atividades precípuas à sua área de formação. Desta
forma, na realidade, as promessas mercadológicas veiculadas pelas políticas públicas não
foram cumpridas no Curso Superior de Tecnologia em Análise e Desenvolvimento de
Sistemas do IF Goiano Câmpus Urutaí.

Previous issue date: 2017-05-22 / As empresas passaram por grandes transformações nos últimos anos, com a globalização e as novas tecnologias, que têm sido as principais responsáveis pelas mudanças e exigências do mercado de trabalho. Frente às mudanças no mundo do trabalho, muitos profissionais ainda não conseguiram perceber com clareza tais modificações, e muito menos conseguem se adaptar a elas. Sendo assim, torna-se fundamental o melhor entendimento do mercado, dos fatores facilitadores e limitantes da inserção de egressos no mercado contemporâneo. Sendo assim, esta dissertação teve como objetivo principal analisar quais são as percepções de egressos de cursos de graduação em Administração sobre fatores facilitadores e dificultadores para sua inserção no mercado de trabalho, propiciados por seus cursos. A investigação foi realizada por meio de entrevista semiestruturada, com o objetivo de coletar informações sobre as percepções dos egressos em duas Faculdades Privadas, denominadas aqui como X e Y, localizadas na cidade de Itaperuna, no Noroeste Fluminense. Os sujeitos da pesquisa foram os egressos das faculdades X e Y, escolhidos por acessibilidade, formados no máximo há três anos. As respostas coletadas nas entrevistas foram analisadas qualitativamente, através de categorização do conteúdo das informações. Os resultados demonstraram que as Instituições de Ensino precisam desenvolver programas de orientação de carreira para os acadêmicos, com a possibilidade de agregar um diferencial competitivo frente aos concorrentes. / Companies have undergone major changes in recent years, with globalization and new technologies, which have been primarily responsible for the changes and demands of the labor market. Faced with changes in the world of work, many professionals have still not been able to clearly perceive such changes, let alone adapt to them. Therefore, the best understanding of the market, of the facilitating and limiting factors of the insertion of graduates in the contemporary market, becomes fundamental. Thus, this dissertation had as main objective to analyze what are the perceptions of graduates of undergraduate courses in Administration on factors that facilitate and impede their insertion in the job market, provided by their courses. The research was carried out by means of a semi-structured interview, with the purpose of collecting information about the perceptions of the graduates in two Private Colleges, denominated here as X and Y, located in the city of Itaperuna, in the Northwest Fluminense. The subjects of the research were the graduates of the X and Y colleges, chosen for accessibility, formed at most three years ago. The answers collected in the interviews were analyzed qualitatively, through categorization of the content of the information. The results showed that the Institutions of Education need to develop programs of career guidance for the academics, with the possibility of adding a competitive differential to the competitors
