1962 a new swedish schoolsystem was accepted called Comprehensive school. The study observes how the discussion of the new schoolsystem looks like in the Union magazine Skolvärlden during the 1970-1969. The study shows how teachers’ thoughts about a schoolsystem which are including all children, no matter what economic statues the have got. Teachers’ need more time to connect with the pupils’. In the other schoolsystem they did not have that kind of issues that might occure now. Some children from a less education background find it difficult to fit in in the comprehensive school. Teachers’ do not know how to handle those children which are interrupting their classes. As a reason of that special-need education are developed during the decade. The reason of that is that all children in Sweden from now have to go to school for nine years according to the law. Teachers’ also believe that the politicians are interfering a lot in school matters. As an example of that Ingvar Carlsson are saying that school need time to reconnect to the new circumstances. Though the former minister of education Olof Palme in the beginning of the decade says that teachers actually need to be more aware of the importance of internationalization in every subject not just the social subjects. Instead teachers rather will prefer to work by them self to implement the new conditions from the curricula and new research. They should instead leave the education to the teachers that actually have got the education and the knowledge about how to teach children. Teachers’ are also irritated of the low wages according to their harder labor circumstances. They believe that other parts of the labor market have got better incomes and conditions. Skolvärlden thinks that LO as an organisation connected to the Socialdemokratic party has priveligies that they do not. In the 1975 teachers are prepared to fight for better circumstances and the are very angry at LO who do not want to support them. Skolvärlden thinks that the reason of that is because LO does not think that education is important, which they actually should have thought. The reason of that is because education from now on is very important for a society development.

Syftet med uppsatsen är att undersöka hur gender-swapping påverkar en films huvudkaraktärer och handling. Filmerna som analyseras är Ocean’s Eleven (2001) och Ocean’s Eight (2018) med hjälp av Jens Eders modell karaktärsklockan och Robert McKees teorier om protagonister. Uppsatsens resultat visar att filmerna är väldigt lika varandra men att huvudkaraktärerna, förutom det uppenbara bytet av kön, skiljer sig åt på sådant sätt att det påverkar filmernas handling på olika sätt.

This is a study that with a gender-conscious analysis compares four films with each other. There are two "original" films with men starring in them, Ocean's eleven from 2001 and Ghostbusters from 1984. As well as remakes of these films with women starring in them, Ocean's eight from 2018 and Ghostbusters from 2016. This trend to do remakes of films and replace men with women have become increasingly popular among the people who create them. Despite this, the result is not very positive among the audience. In this study, we will take a look into the movies and their reception. Here, websites (imdb.com & rottentomatoes.com) were used to analyze the comments about the four different films, both the positive and the negative ones. The film analysis itself consists of selected scenes that was compared with each other in the various films. To make a comparison of the whole movies would need a much longer examination. That is why only some scenes were selected. The conclusion shows that it is rather the same films over and over again that are the problem for the audience rather than the female aspect.

The Regulation of Plasma Gelsolin by DNA Methylation in Ovarian Cancer ChemoresistanceManzoor, Hafiza Bushra 20 September 2023 (has links)
Ovarian cancer (OVCA) is the most lethal gynecologic cancer. Chemoresistance remains a major hurdle to successful therapy and patient survival. The secreted isoform of the actin-associated protein, gelsolin (plasma gelsolin; pGSN), is highly expressed in chemoresistant than chemosensitive OVCA cells, although the mechanism underlying the differential expression is not known. Also, its overexpression significantly correlates with shortened survival of OVCA patients. DNA methylation plays a key role in the regulation of genes expression and contributing to cancer development and chemoresistance with the help of DNA methyltransferases (DNMTs) or Ten eleven translocation (TETs) enzymes. TET1 is the most studied isoform of TETs family and primarily responsible for 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC) oxidation to initiate demethylation and increase in the expression of methylated genes. Whether pGSN expression in OVCA cells is regulated by DNA methylation and TET1 regulates the differential pGSN expression between chemosensitive and resistant OVCA cells is not known. In this study, we hypothesized pGSN overexpression in chemoresistant OVCA cells is due to the hypomethylation at its promoter region by TET1. Our objective was to investigate whether DNA methylation and specifically TET1 plays a role in the regulation of differential pGSN expression and chemosensitivity in OVCA. Chemosensitive and resistant OVCA cell lines of different histological subtypes were used in this study to measure pGSN and TET1 mRNA abundance and protein contents by qPCR and Western blotting respectively. Cisplatin-induced chemoresponsiveness was morphologically assessed by Hoechst staining (apoptosis). Infinium HumanMethylation450 BeadChip assay was used for global methylation analysis of twelve (12) different OVCA cells and to investigate the role of DNA methylation specifically in pGSN regulation and pGSN-induced chemoresistance. DNMTs and TETs were pharmacologically inhibited in sensitive and resistant OVCA cell using specific inhibitors. Gain-and-loss-of-function assays were carried to identify the relationship between TET1 and pGSN in OVCA chemoresponsiveness. Differential protein and mRNA expressions of pGSN and TET1 were observed between sensitive and resistant OVCA cells and cisplatin reduced their expression in sensitive but not in resistant cells. Global methylation analysis revealed hypomethylation in resistant cells compared to sensitive cells. Pharmacological inhibition of DNMTs increased pGSN protein levels in sensitive OVCA cells and decreases their responsiveness to cisplatin, however we did not observe any difference in methylation level at pGSN promoter region. TETs inhibition resulted in hypermethylation at multiple CpG sites and decreased pGSN protein level in resistant OVCA cells which was also associated with enhanced response to cisplatin, findings that suggested the methylation role of TETs in the regulation of pGSN expression in OVCA cells. Further, we found that TET1 is inversely related to pGSN and positively related to chemoresponsiveness of OVCA cells. This project does not only broaden our knowledge about the mechanistic insights into the epigenetic regulation of pGSN in OVCA chemoresistance, but it also reveals a new potential target to re-sensitize chemotherapy resistant OVCA cells. This may provide a future strategy to improve overall OVCA patient survival.

The role of Ten Eleven Translocation enzymes in the hair follicle mesenchymeAhmed, Aqib January 2022 (has links)
Epigenetic mechanisms play an important role during the morphogenesis
of the hair follicle and the hair cycle. Work on hair regeneration is of
importance as no products are available which can provide complete
reversal of hair loss. Tet2 promotes DNA demethylation by the
hydroxylation of 5mC to 5hmC which in turn causes gene transcription
activation. Dermal papilla (DP) cells located within the hair follicle are
responsible for the regulation of development and the growth of hair
follicles. Fgf20 signalling controls commitment of the mesenchymal
precursor cells to the DP progenitor lineage. An immature DP cells is then
formed during maturation by Shh signalling which then stimulates these to
differentiate into a DP cell by BMP and Wnt signalling.
Methylated DNA can be bound by the proteins recruiting transcription corepressors.
DNA methyltransferases (DNMT’s) can be degraded by
decitabine which reverses gene silencing. Conditional knockout of Tet2 in
mouse DP cells results in a delay in anagen initiation, suggesting Tet2 is
involved in the telogen-anagen transition. Additionally, by using dermal
fibroblasts and RA-DPAC (Dermal Papilla activating medium
supplemented with retinoic acid), it was found that decitabine can increase
plasticity in dermal fibroblasts and RA-DPAC can be used to accelerate a
lineage change to DP cells which is supported by the significant increase in the DP specific gene expression. Examples include AlPl, LEF1,
BMP4/6/7, FGF10, BMPR1A and PDGFA. Additionally, by way of siRNA
and conditional Tet2 knockout data in dermal fibroblasts, it was found Tet2
regulates signature DP genes such as Bmpr1a, ALPL, Tcf4 and SOX2.

Hovedproblemstillinga i denne undersøkelsen er ” Hvordan oppfatter musikk- og kulturskolelærerne den stille eleven og hvordan legger de opp gruppeundervisningen for å nå denne typen elever i gruppesituasjoner?” For å finne svar på dette brukte jeg kvalitativt forskningsintervju som metode. Fem strykelærere med minimum ti års fartstid og med erfaring fra både individuell- og gruppeundervisning har deltatt. Disse ble håndplukket fra mitt kontaktnett. Resultatet av undersøkelsen viser at lærerne ser på den stille eleven som en som ikke sier så mye og er veldig forsiktig. Utover dette kan det se veldig ulikt ut. For å nå den stille eleven kom det fram at det er viktig å arbeide med hele gruppa for å skape en fellesskapsfølelse, og at de som lærere ser og bekrefter elevene. I diskusjonen har jeg med utgangspunkt i teorier om gruppeprosesser og lederskap, diskutert hvilken betydning musikkundervisning i gruppe og hvilken rolle læreren har i forhold til den stille elevens atferd. / The main question in this survey is "What are experienced music and art teachers’ opinions on the quiet learner, and how do they reach this group of students in group teaching?" To answer this, I used qualitative interviews. Five string teachers with a minimum of ten years of experience and with experience in both individual and group teaching participated. The teachers were handpicked from my contacts. The results of the survey show that teachers see the quiet student as a person who does not say much, and is very careful. Beyond this, every quiet student can look very different. To reach the quiet student, it became apparent that it is important to create a group atmosphere, and that the teachers see and acknowledge the students. During the discussion, I have raised the importance of group teaching in music and the vital part the teacher plays, in relation to the development of quiet students, based on theories about group processes and leadership.
