Potassium Permanganate/ Carboxylic Acid/ Organic Solvent: A Powerful Reagent For C-c Bond Formation, Aryl Coupling Reactions And Enone Oxidation Ipso-nitration Of Arylboronic Acids With Silver Nitrite/ Tmscl

The first part of the thesis presents the KMnO4/ carboxylic acid/ organic solvent which is a powerful reagent for C-C bond formation, aryl coupling reactions and enone oxidation. The a&cent / -acetoxylation of enones and the a-acetoxylation of aromatic ketones were carried out with potassium permanganate and acetic acid, in which acetoxylation products were obtained in 74-96% yields. The same reaction was carried out with carboxylic acids other than acetic acid, which furnished corresponding acyloxy ketones with the same regioselectivity. For the first time, formyloxylation products were synthesized in a 61-85% yield by using formic acid. The potassium permanganate and acetic acid method was also used for aryl coupling reactions. The reaction of arylboronic acids and aryl hydrazines in benzene with potassium permanganate and acetic acid in turn furnished biaryls in a 85-96% yield. We showed that potassium permanganate/carboxylic acid/organic solvent behaves as manganese(III) acetate. In the second part of the thesis, ipso-nitration of arylboronic acids with AgNO2/ TMSCl was performed. Nitration of aromatic compounds is one of the most extensively studied reactions, and nitroaryl moieties play key roles in the physical and chemical properties of many target molecules in organic synthesis. For electrophilic nitration of aromatic compounds, a wide variety of reagents are available to date. Most of them are very strong nitrating agents and often lead to further nitration and mixture of isomers. Since most nitrating agents are oxidants, oxidation of other functional groups can also occur, giving a mixture of products. Thus, a search for milder and selective nitrating agents is a good research goal. In this work, we aimed to apply AgNO2/ TMSCl system to ipso nitration of arylboronic acids.

Chiral cyclic polyoxo-ketones are important structural units in many natural products, biologically active compouds, such as prostaglandins, didemnenones, sarkomycin, punaglandin, clavulone, etc. In this work, a chemoenzymatic synthesis of both enantiomers of the &amp / #945 / &rsquo / -acetoxy-&amp / #945 / -methyl and &amp / #947 / -hydroxy-&amp / #945 / -methyl cyclic enones starting from &amp / #945 / -methyl-&amp / #946 / -methoxy cyclic enone is described. Manganese (III) acetate-mediated acetoxylation followed by the enzyme-mediated hydrolysis of &amp / #945 / &rsquo / -acetoxy enone provides acetoxy enones. The reduction of the hydroxy enone, obtained from hydrolysis, furnished both enantiomers of 4-hydroxy enone or &amp / #947 / -hydroxy enone by using LiAlH4. This study is a model for the synthesis of these type compounds

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The synthesis of thirteen 3-(R1), 4-(R2) and 5-(R3)-substituted 1-cyanoacetyl-5-hydroxy-4,5-dihydro-1H-pyrazoles from the cyclocondensation reaction of 4-alkoxy-3-alken-2-ones [R3C(O)C(R2)=C(R1)(OR), where R3 = CF3, CCl3, CHCl2, CO2Et; R2 = H, Me; R1 = H, Me, Et, Pr, Pentyl, c-Hexyl, Ph, and R = Me, Et] with cyanoacetohydrazide is reported. This reaction was carried out in different methodologies, firstly using water like solvent and following with ionic liquid ([BMIM]BF4). The results showed that when the ionic liquid was used as reaction media, the reaction time was drastically decreased and the yield was improved. 4,5-Dihydropyrazole (R1 = Me, R2 = H, R3 = CF3) was used as an important building block to the synthesis of cyanoenaminopyrazole from the condensation reaction with N,N-dimethylformamide dimethyl acetal. Benzilidene cyanoacetohydrazide was also employed to the synthesis of a series of seven 1- benzilidenepyrid-2-ones from the cyclocondensation reaction with 4-alkoxy-1,1,1-trifluoro-3-alken-2-ones [CF3C(O)C(R2)=C(R1)(OR), where R2 = H, Me; R1 = Me, Pr, Butyl, c-Hexyl, Ph, 4-MePh, e R = Me, Et]. The attainment of these compounds was only possible when the reaction was carried out in ionic liquid [BMIM]BF4. / Neste trabalho é descrita a síntese de uma série de treze 1-cianoacetil-5-hidróxi-4,5-diidro-1H-pirazóis 3-(R1), 4-(R2) e 5-(R3) substituídos a partir da reação de ciclocondensação de 4-alcóxi-3-alquen-2-onas [R3C(O)C(R2)=C(R1)(OR), onde R3 = CF3, CCl3, CHCl2, CO2Et; R2 = H, Me; R1 = H, Me, Et, Pr, Pentil, c-Hexil, Ph, e R = Me, Et] com cianoacetoidrazida. Esta reação foi realizada por duas metodologias diferentes, primeiramente utilizando água como solvente e, em um segundo momento, em líquido iônico ([BMIM]BF4). Os resultados obtidos mostraram que o líquido iônico [BMIM]BF4 demonstrou ser um importante meio reacional, proporcionando uma drástica redução no tempo da reação, bem como um aumento nos rendimentos. O 4,5-diidropirazol (R1 = Me, R2 = H, R3 = CF3) foi utilizado como um importante bloco precursor para a síntese do derivado cianoenaminopirazol através da reação de condensação com N,Ndimetilformamida dimetilacetal. A benzilideno-cianoacetoidrazida também foi empregada na síntese de uma série de sete 1-benzilidenopirid-2-onas através de uma reação de ciclocondensação com 4-alcóxi-1,1,1-trifluor-3-alquen-2-onas [CF3C(O)C(R2)=C(R1)(OR), onde R2 = H, Me; R1 = Me, Pr, Butil, c-Hexil, Ph, 4- MePh, e R = Me, Et]. A obtenção destes compostos foi possível apenas quando o líquido iônico [BMIM]BF4 foi utilizado como meio reacional.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The present work describes the synthesis of novel 1-alkyl(aryl)-2-alkoxy-5-trifluoroacetyl-1,2,3,4-tetrahydropyridines, aza-condensed tetrahydropyridines and ethyl-1H-pyrazoles-3(5)-carboxylate from the use of cyclic enones as precursors. The 1-alkyl(aryl)-2-alkoxy-5-trifluoroacetyl-1,2,3,4-tetrahydropyridines were synthesized using stoichiometric amounts of trifluoroacetyl dihydropyranes and different primary amines in 55 96% yield, under basic catalysis, whereas the aza-condensed tetrahydropyridines were synthesized using stoichiometric amounts of trifluoroacetyl dihydropyranes and different diamines, leading to the desired products in 81 96% yield. The acylation of 2,3-dihydrofuran, 3,4-dihydropyran and 2-alkoxy-3,4-dihydro-2H-pyran with ethyloxallyl chloride furnished esterified enones (4-(2-ethyl-oxoacetate)2,3-dihydrofuran, 5-(2-ethyl-oxoacetate)-3,4-dihydro-2H-pyran and 2-methoxy-5-(2-ethyl-oxoacetate)-3,4-dihydro-2H-pyran) in 47 81% yield. Finally, the cyclocondensation reaction between enones (4-(2-ethyl-oxoacetate)-2,3-dihydrofuran-2-oxoacetate, 5-(2-ethyl-oxoacetate)-3,4-dihydro-2H-pyran) with different hydrazines led to the synthesis of 1H-pyrazole-3(5)-carboxylate ethyl ester in 62 96% yield. The N1 H substituted ethyl-1H-pyrazole-3(5)-carboxylate esters were N-alkylated with alkyl , allyl , and benzyl halides leading to the desired N-substituted pyrazoles in 48 96% yield. / Este trabalho apresenta a síntese de três séries de compostos inéditos: 1-alquil(aril)-2-alcóxi-5-trifluoracetil-1,2,3,4-tetraidropiridinas, hexaidroimidazopiridinas e heterociclos análogos aza-condensados e 1H-pirazóis-3(5)-carboxilato de etila a partir da utilização de enonas cíclicas como precursores. As 1-alquil(aril)-2-alcóxi-5-trifluoracetil-1,2,3,4-tetraidropiridinas, foram obtidas com rendimentos entre 55 e 96%, utilizando-se quantidades estequiométricas dos 2-alcóxi-5-trifluoracetil-3,4-diidro-2H-piranos com diferentes aminas primárias, sob catálise básica de piridina, enquanto que, as hexaidroimidazopiridinas e heterociclos aza-condensados foram sintetizados através da reação entre os mesmos precursores, em relação estequiométrica, frente a diaminas conduzindo a formação dos produtos desejados em rendimentos de 81 e 96%. A acilação entre o 2,3-diidrofurano, 3,4-diidropirano e 2-alcóxi-3,4-diidro-2H-pirano com o cloreto de etiloxalila promoveu a obtenção das enonas esterificadas (4-(etil-2-oxoacetato)-2,3-diidrofurano, 5-(etil-2-oxoacetato)-3,4-diidro-2H-pirano e 2-metóxi-5-(etil-2-oxoacetato)-3,4-diidro-2H-pirano) com rendimentos de 47 e 81%. A reação de ciclocondensação entre as enonas (4-(etil-2-oxoacetato)-2,3-diidrofurano, 5-(etil-2-oxoacetato)-3,4-diidro-2H-pirano) com diferentes hidrazinas conduziu à síntese dos 1H-pirazóis-3(5)-carboxilato de etila com rendimentos entre 62 e 96%, sendo que os 1H-pirazóis-3(5)-carboxilato de etila que apresentam hidrogênio como substituinte na posição-1, foram submetidos à posterior reação de N-alquilação conduzindo aos produtos desejados com rendimentos entre 48 e 96%.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / An efficient method to obtain six 4-alkoxy-2-oxo-but-3-enoic acid ethyl esters [EtO2CC(O)C(R2)=C(R1)OR, where R1/R2/R = Me/H/Me (2a), Ph/H/Me (2b), 4-MeC6H4/H/Me (2c), 4-BrC6H4/H/Me (2d), 4-FC6H4/H/Me (2e), H/Me/Et (2f)] from acylation of enol ethers or acetals with ethyl oxalyl chloride is reported. The cyclocondensation reaction of these substrates and their trifluormethylated analogues (5a-f) [CF3C(O)C(R2)=C(OR)R1] with salicylic hydrazide furnished a series of ethyl 5-hydroxy-1-(2-hydroxybenzoyl)-4,5-dihydro-1H-pyrazole-5-carboxylates (6) and 5-hydroxy-5-trifluoromethyl-4,5-dihydro-1H-1-(2-hydroxybenzoyl)pyrazoles (7), respectively. The structure of the obtained compounds was determined by spectroscopy and confirmed by crystallographic studies. The design of these compounds explore the hypothesis that the hybridization of salicylic acid with an appropriate 4,5-dihydro-1Hpyrazole scaffold can supply novel potent analgesic agents. The oral administration of one compound of each series of pyrazoles (6a, 7a) caused significant analgesia in the writhing test in mice, which was similar to the analgesia caused by aspirin. / Este trabalho descreve um eficiente método para obter seis 4-alcóxi-2-oxo-3-butenoatos de etila [EtO2CC(O)C(R2)=C(R1)OR, onde R1/R2/R = Me/H/Me (2a), Ph/H/Me (2b), 4-MeC6H4/H/Me (2c), 4-BrC6H4/H/Me (2d), 4-FC6H4/H/Me (2e), H/Me/Et (2f)] a partir da acilação de enoléteres e acetais com cloreto de etil oxalila. A reação de ciclocondensação desses substratos e seus análogos trifluormetilados (5a-f) [CF3C(O)C(R2)=C(OR)R1] com salicil hidrazida forneceu respectivamente as séries de 5-etilcarboxilato-5-hidróxi-4,5-diidro-1H-1-(2-hidroxibenzoil)pirazóis (6) e 5-hidróxi-5-trifluormetil-4,5-diidro-1H-1-(2-hidroxibenzoil)pirazóis (7). Adicionalmente aos dados espectroscópicos, a estrutura dos compostos foi confirmada por experimentos de raios-X. O planejamento desses compostos explora a hipótese de que a hibridização molecular do ácido salicílico com um núcleo 4,5-diidro-1H-pirazol pode fornecer novos potentes agentes analgésicos. A administração oral de um exemplo de cada série de pirazóis (6a, 7a) causou um efeito analgésico significante no teste das contorções abdominais em camundongos, o qual foi similar ao efeito analgésico apresentado pela aspirina.

This work presents the synthesis of three new series of nitrogen-heterocycles containing the substituent trifluoromethyl, exploiting the synthetic versatility and regioselectivity of 4-alkoxy-5-bromo-1,1,1-trifluorpent-3-en-2-ones and 4-amino-5-bromo-1,1,1-trifluoropent-3-en-2-ones in reactions with compounds containing nucleophilic nitrogen. Two series of 1-(3-alkoxy-5-trifluoromethyl-2,3-dihydrofuran-3-yl)-4,5-alkyl-3-(trifluoromethyl)-1H-pyrazoles were synthesized by the N-functionalization reaction of pyrazoles with 4-alkoxy-5-bromo-1,1,1-trifluorpent-3-en-2-ones, by Michael s nucleophilic addition. In the first step, there is a nucleophilic addition of the pyrazol molecule to the beta position of enones (Cβ), followed by an intramolecular cyclization reaction, where the furan ring is formed by replacement of the bromine atom by the carbonyl oxygen of enone, resulting in thirteen novel compounds with yields between 55-86%. The other compounds series, (E)-4-(amino)-1,1,1-trifluoro-5-(5-methyl-3- (trifluoromethyl)-1H-pyrazol-1-yl)pent-3-en-2-ones, was synthesized by N-alkylation reaction, through a bimolecular nucleophilic substitution (SN2) mechanism, with replament of the bromine atom, at five position (Cγ) of 4-amino-5-bromo-1,1,1-trifluoropent-3-en-2-ones, by the nucleophilic nitrogen of the pyrazoline ring. Seven N-alkylated products were obtained, with yields among 65-85%. In addition, the regioselectivity study of N-functionalized pyrazoles reactions is described, including the evaluation of reaction conditions and how substituents present in the pyrazole structure can influence the product formation, because many different steric and electronic factors. The obtained compounds were characterized by nuclear magnetic resonance 1H and 13C, mass spectrometry, elementary analysis and X-ray diffractometry. / A presente dissertação relata a síntese de três séries inéditas de heterociclos nitrogenados trifluormetil substituídos, que exploram a versatilidade sintética e a regiosseletividade das 4-alcóxi-5-bromo-1,1,1-trifluorpent-3-en-2-onas e das 4-amino-5-bromo-1,1,1-trifluorpent-3-en-2-onas, em reações com nucleófilos nitrogenados. As primeiras duas séries dos compostos 1-(3-alcóxi-5-trifluorometil-2,3-diidrofuran-3-il)-4,5-alquil-3-(trifluorometil)-1H-pirazóis, foram sintetizadas através do processo de N-funcionalização de pirazóis, a partir da reação com as 4-alcóxi-5-bromo-1,1,1-trifluorpent-3-en-2-onas, cujo o caminho mecanístico se deu através de uma reação de adição nucleofílica de Michael. Inicialmente, ocorre a adição do pirazol nucleofílico na posição beta (Cβ) das enonas bromadas, seguida de uma reação de ciclocondensação intramolecular formando o anel furano, com a substituição átomo de bromo pelo oxigênio enólico, resultando na formação de treze compostos inéditos, com rendimentos entre 55 e 86%. Outra série de compostos (E)-4-(amino)1,1,1-triflúor-5-(5-metil-3-(trifluormetil)-1H-pirazóis-1-il)pent-3-en-2-onas, foi sintetizada através da reação de N-alquilacão, via substituição nucleofílica bimolecular (SN2), onde o átomo de bromo na posição cinco (Cγ), das 4-amino-5-bromo-1,1,1-trifluorpent-3-en-2-onas, foi substituído pelo nitrogênio nucleofílico do anel pirazolínico, promovendo a formação de sete produtos N-alquilados, com rendimentos que variam entre 65 e 85%. Além disso, um estudo de regiosseletividade das reações N-funcionalizadas de pirazóis está descrito, onde a avaliação das condições reacionais e também de fatores estéricos e eletrônicos dos substituintes presentes nos substratos, foram determinantes para formação do produto formado. Os produtos obtidos neste trabalho foram caracterizados por ressonância magnética nuclear de 1H e 13C, espectrometria de massas de baixa e alta resolução, análise elementar e difratometria de Raio X.

Conselho Nacional de Desenvolvimento Científico e Tecnológico / The solvent-free microwave-assisted regiospecific synthesis of a series of 5-trihalomethyl-4,5-dihydro-1H-pyrazole (7 - 10) from the cyclocondensation of 1,1,1- trihalo-4-alkoxy-3-alken-2-ones (5 - 6),[ CX3COC(R)2=C(R)1OR, where X = Cl, F; R = Me, Et; R2 = H, Me and R1 = H, Me, Et, Pr, iso-Pr, Bu, iso-Bu, tert-Bu, iso - Pen, Ph, 4-Me-Ph, 4-MeO-Ph, 4-NO2-Ph, 4-Cl-Ph, 4-Br-Ph, 4-F-Ph, H, OEt,] with carboxymethyl hydrazine or pentafluorophenyl hydrazine is described. The reaction was performed in a 1:1.2 molar ratio of alken-2-ones and hydrazine, in a single reaction step, for 6-12 minutes, in good yields (70-98%). The advantages obtained by using microwave irradiation under solvent-free conditions, rather than a conventional method, were also demonstrated. The 4-alkoxy-1,1,1-trihalo-3-alken-2-ones (4-5) were obtained from the reaction of cloreto de trichloroacetyl cloride or trifluoroacetic anhydride with enol ethers or acetals in presence of pyridine, solvent free and microwave induced techniques. / Este trabalho descreve a sintese rgeioespecifica de uma serie de 4,5-diidro-1Hpirazóis (7 - 10), por ciclocondensação de 4-alcóxi-1,1,1-trialo-3-alquen-2-onas, [ CX3COC(R)2=C(R)1OR, com X = Cl, F; R = Me, Et; R2 = H, Me; R1 = H, Me, Et, Pr, i-Pr, c-Pr, Bu, i-Bu, t-Bu, i-Pen, Pen, Ph, 4-Me-Ph, 4-MeO-Ph, 4-NO2-Ph, 4-Cl-Ph, 4-Br-Ph, 4-F-Ph, H, OEt] com carboximetil ou pentafluorfenil hidrazinas. As condições reacionais utilizadas para a síntese dos 4,5-diidropirazóis envolveu técnicas ambientalmente corretas, em um único passo e na ausência de solvente, usando 1:1.2 de relação molar das β-alcoxivinil cetonas e hidrazinas, em um único passo com um tempo de 6-12 minutos, com bons rendimentos (70-98%). Foram demonstradas as vantagens da condição, sem solvente e com uso de irradiação de microondas sobre o metodo convencional. A sintese das 4-alcóxi-1,1,1-trialo-3- alquen-2-onas (5 - 6) foram obtidas por reações de cloreto de tricloro acetila ou anidrido trifluoracetico com enoleteres ou acetais na presensa de piridina, livre de sonvente e tecnicas induzidas por microondas.

The pyrroles were obtained by the reaction of 3-trifluoroacetyl-4,5-dihydrofuran with aryl amines of general formula ArNH2, and Ar= C6H4, 2-MeO(C6H4), 3-MeO(C6H4), 4-MeO(C6H4), 2-Me(C6H4), 3-Me(C6H4), 4-Me(C6H4), 3-F(C6H4), 4-F(C6H4), 3-Cl(C6H4), 4-Cl(C6H4), 4-OH(C6H4), 2-OH-5-Me(C6H3), 3-OH-4-Me(C6H3), 4-Br(C6H4) generating 1,1,1-trifluoro-3-(2-hydroxyethyl)-4-arylamino-3-beten-2-ones intermediates that could not be isolated. These intermediaries were submitted to Swern oxidation reaction producing 1,1,1-trifluoro-3-(2-ethanal)-4-arylamino-3-buten-2-ones that underwent intramolecular cyclization followed by aromatization by dehydratation, producing N-aryl substituted 3-trifluoracetyl-1H-pyrroles in 30-56% yield. The pyrroles obtained in this study were identified by 1H NMR, 13C NMR, and GC-Mass Espectroscopy. / Este trabalho apresenta uma nova estratégia sintética simples e versátil para a preparação de uma série inédita de 3-trifluoracetil-1H-pirróis N-aril substituídos. Os pirróis foram obtidos a partir da reação do 3-trifluoracetil-4,5-diidrofurano com aril aminas de fórmula geral ArNH2, sendo Ar= C6H4, 2-MeO(C6H4), 3-MeO(C6H4), 4-MeO(C6H4), 2-Me(C6H4), 3-Me(C6H4), 4-Me(C6H4), 3-F(C6H4), 4-F(C6H4), 3-Cl(C6H4), 4-Cl(C6H4), 4-OH(C6H4), 2-OH-5-Me(C6H3), 3-OH-4-Me(C6H3), 4-Br(C6H4) gerando os intermediários 1,1,1-trifluoro-3-(2-hidroxietil)-4-arilamino-3-buten-2-onas, que não foram isolados. Esses intermediários foram submetidos a reação de oxidação de Swern produzindo 1,1,1-trifluoro-3-(2-etanal)-4-arilamino-3-buten-2-onas que sofreram reação de ciclização intramolecular, seguido de aromatização com a perda de uma molécula de água produzindo os 3-trifluoracetil-1H-pirróis N-aril substituídos, com rendimentos reacionais (30-56%). Os pirróis obtidos neste trabalho foram identificados por Ressonância Magnética Nuclear de Hidrogênio, Ressonância Magnética Nuclear de Carbono-13 e Espectroscopia de Massas.

Chiral Boro-Phosphates in Asymmetric Catalysis: 1,4-reduction of Enones and Reductive Aldol

The biological activity of the pharmaceutical drugs often depends on how it fits with a receptor making stereochemistry a key component. Selective reactions can limit or avoid the mixture of enantiomers obtained. One such reaction is the selective reduction of a carbon-carbon double bond in the presence of a carbonyl. Although efficient, current asymmetric synthesis methods have limitations such as harsh reaction conditions, the high costs of chiral catalysts and the toxicity of the metal-based catalysts. Catalysts derived from small organic molecules have become an attractive alternative which have been explored more rigorously in recent years. Using a BINOL-derived boro-phosphate catalyst, we have developed a methodology that selectively reduces the carbon-carbon double bond of linear α, β-unsaturated ketones, exclusively giving the corresponding saturated ketone. To the best of our knowledge, this reaction is the first of its kind to accomplish this transformation and results give high yields of >93% and enantioselectivities >90% at room temperature. Furthermore, the products of this novel reaction can be subjected to a choice electrophile, in example benzaldehyde, to afford diastereoselective tertiary alcohol products with enantioselectivities of >88% and diastereoselectivities of up to 99:1.

Acilenaminonas: Síntese e Aplicação na Obtenção de Pirazóis, Pirazolo[3,4-d]piridazinonas e Pirazolo[1,5-a]pirimidinas / Acylenaminones: Synthesis and Application in the Obtaining of Pyrazoles, Pyrazolo[3,4-d]pyridazinones and Pyrazolo[1,5-a]pyrimidines

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / The regioespecific synthesis of a series of 14 N-acylated enaminones (52-88%) from the acylation reaction of secondary β-enamino ketones [RC(O)CH=CHNR1R2; R = Ph, 4- FC6H4, 4-NO2C6H4, thien-2-yl, CCl3, CF3; R1 = H; R2 = Bn, Ph, 4-NO2C6H4] with trifluoroacetic anhydride or ethyl oxalyl chloride in pyridine is reported. On the other hand, when tertiary enaminone precursors [R = Ph, 4-MeC6H4, 4-MeOC6H4, 4-BrC6H4, 4-ClC6H4, 4-FC6H4, 4-O2NC6H4, thien-2-yl, benzofur-2-yl, CCl3, CF3; R1,R2 = Me] were used, the acylation reaction led to a series of 17 C-acylated enaminones (75-95%). A series of 4-substituted-1H-pyrazole-5-carboxylates (73-94%) were obtained regiospecifically from the cyclocondensation reaction of non symmetrical enaminodiketones [RC(O)C(=CHNMe2)C(O)CO2Et; R = Ph, 4-MeOC6H4, 4-ClC6H4, 4-FC6H4, 4-O2NC6H4, thien-2-yl, benzofur-2-yl, CCl3, CF3] with tert-butylhydrazine or carboxymethylhydrazine. The reaction of these pyrazole-5-carboxylates (R = 4-MeOC6H4, 4-FC6H4, benzofur-2-yl, CF3) with hydrazine lead to synthesis of 4- substituted-pyrazolo[3,4-d]pyridazinones (74-96%). In addition, the reaction of enaminodiketones (R = Ph, 4-MeC6H4, 4-MeOC6H4, 4-BrC6H4, 4-ClC6H4, 4-FC6H4, 4- O2NC6H4, thien-2-yl, benzofur-2-yl) with 3-amino-5-methylpyrazole was performed, where a series of pyrazolo[1,5-a]pyrimidine-7-carboxylates were obtained regiospecifically (53-79%). / A síntese de uma série de 14 enaminonas N-aciladas regioespecificamente (52-88%) foi realizada a partir da reação de acilação de enaminonas secundárias [RC(O)CH=CHNR1R2; R = Ph, 4-FC6H4, 4-NO2C6H4, tien-2-il, CCl3, CF3; R1 = H; R2 = Bn, Ph, 4-NO2C6H4] com anidrido trifluoracético e com cloreto de etil oxalila em piridina. Quando foram utilizados como precursores enaminonas terciárias [R = Ph, 4-MeC6H4, 4-MeOC6H4, 4-BrC6H4, 4-ClC6H4, 4-FC6H4, 4-O2NC6H4, tien-2-il, benzofur-2-il, CCl3, CF3; R1,R2 = Me] a reação de acilação levou à obtenção regioespecífica de 17 enaminonas C-aciladas (75-95%). Uma série de 5-carboxietil-1H-pirazol 4-substituídos foi obtida regioespecificamente (73-94%) a partir da ciclocondensação das enaminodicetonas não simétricas [RC(O)C(=CHNMe2)C(O)CO2Et; R = Ph, 4-MeOC6H4, 4-ClC6H4, 4-FC6H4, 4-O2NC6H4, tien-2-il, benzofur-2-il, CCl3, CF3] com tert-butilidrazina ou carboximetilidrazina. A reação destes pirazóis (R = 4-MeOC6H4, 4-FC6H4, benzofur-2-il, CF3) com hidrazina monoidrato levou à síntese de pirazolo[3,4-d]piridazinona 4- substituídas (74-96%). Também foi realizada a reação de ciclocondensação de enaminodicetonas (R = Ph, 4-MeC6H4, 4-MeOC6H4, 4-BrC6H4, 4-ClC6H4, 4-FC6H4, 4-O2NC6H4, tien-2-il, benzofur-2-il) com 3-amino-5-metilpirazol onde as 7-carboxietilpirazolo[1,5-a]pirimidinas 6-substituídas foram obtidas regioespecificamente (53-79%).
