As células dendríticas (DCs) são consideradas como apresentadoras de antígenos profissionais, iniciando e modulando as respostas imunológicas. O direcionamento de diferentes antígenos para essas células têm sido uma estratégia promissora para a avaliação das respostas imunológicas humoral e celular. Nesse trabalho, o alvo de direcionamento é o subtipo de DCs CD8α+ DEC205+, por apresentarem características importante, como por exemplo, a apresentação cruzada para linfócitos T CD8+ via molécula de MHC II. Os antígenos de escolha para serem direcionados foram diferentes domínios da proteína do envelope (E) do vírus da dengue tipo 2, assim como a proteína na sua forma inteira. Os resultados mostraram que o direcionamento dos diferentes domínios da proteína do envelope viral foi capaz de induzir uma celular robusta nos animais quando comparada com o não direcionamento das mesmas proteínas. Além disso, com o direcionamento para DCs CD8α+ DEC205+ foi possível mapear regiões importantes da proteína E durante a indução da resposta imune. Portanto, os dados obtidos nesse trabalho, sugerem que o direcionamento da proteína do E e dos seus domínios ( EDI/II e EDIII) foi eficiente em induzir resposta imune celular contra o vírus da dengue tipo 2. / Dendritic cells (DCs) are considered to present professional antigens, initiating and modulating immune responses. The targeting of different antigens to these cells has been a promising strategy for the evaluation of humoral and cellular immune responses. Thus, the antigen of interest is directed to the population of DCs desired. In this work, the targeting target is the subtype CD8α + DEC205 + DCs, because they present important characteristics, such as cross-presentation to CD8 + T lymphocytes via the MHC II molecule. The antigens of choice for targeting the CD8α + DEC205 + DCs subtype were different domains of the envelope protein (E) of dengue virus type 2 as well as the protein in its entire form. The results showed that the targeting of the different domains of the viral envelope protein was able to induce a robust cell in the animals when compared to the non-targeting of the same proteins. Furthermore, with the targeting for CD8α + DEC205 + DCs it was possible to map important regions of the E protein during the induction of the immune response. Therefore, the data obtained in this work suggest that the targeting of E protein and its domains (EDI / II and EDIII) was efficient in inducing cellular immune response against dengue virus type 2.

Redes neurais artificiais na predição de respostas e estimação de derivadas aerodinâmicas de aeronaves / Artificial neural networks for prediction of responses and estimation of aerodynamic derivatives of aircraft

A área de dinâmica de aeronaves atingiu um alto nível de desenvolvimento e devido à crescente disponibilidade de computadores cada vez mais rápidos e com maior capacidade de processamento; a aplicação de técnicas numéricas de identificação nesta área também teve grande avanço. Este trabalho apresenta uma metodologia para predição de respostas de aeronaves dentro de envelopes de vôo pré-estabelecidos usando redes neurais recorrentes e uma metodologia para estimação das suas derivadas aerodinâmicas usando redes neurais feedforward. Para obter os conjuntos de dados para treinar as redes neurais, foi implementado um modelo não linear de dinâmica de vôo e simulado o comportamento de uma aeronave de combate em nove pontos de um envelope de vôo. Foram usadas as respostas simuladas correspondentes a quatro pontos para treinar a rede neural e depois disto, esta capturou satisfatoriamente a dinâmica da aeronave, identificando com grande sucesso as respostas do movimento longitudinal da aeronave por todo o envelope de vôo considerado. Após a simulação e identificação das respostas da aeronave dentro do envelope de vôo, é apresentada a resolução do problema inverso, ou seja, usando velocidades escalares e angulares da aeronave juntamente com seus dados geométricos como entradas para a rede neural feedforward, é obtido um modelo neural estimador de derivadas aerodinâmicas. Para mostrar a capacidade deste modelo neural estimador, este é aplicado na estimação das derivadas da aeronave simulada e também aplicado na estimação das derivadas aerodinâmicas da aeronave militar a jato Xavante AT-26 da Força Aérea Brasileira. Estas metodologias propostas reduzem custo de obtenção das derivadas aerodinâmicas e mostram a eficácia das redes neurais em estimar as respostas de aeronaves dentre de um envelope de vôo pré-definido. / The area of aircraft dynamics has reached a high level of development and due to the increasing availability of computers continuously faster and with bigger processing capacity, the application of numerical identification techniques in this area also had great advance. This work presents two methodologies, one for prediction of aircraft responses within a pre-established flight envelope using recurrent neural networks and another one for estimation of its aerodynamic derivatives using feedforward neural networks. To get data sets to train the neural networks, a combat aircraft flight dynamics non-linear model was implemented and simulated in nine points of the flight envelope to obtain its behavior. The simulated responses corresponding to a four points of the flight envelope were used to train the neural network and after that, it was possible to verify that this net satisfactorily captured the dynamics of the aircraft, identifying with great success the longitudinal motion responses of the aircraft at all the considered flight envelope positions. After the simulation and identification of the aircraft responses inside the flight envelope, the solution of the inverse problem is presented, i.e., using scalar and angular aircraft velocities together with its geometric data as input to the feedforward neural network, a neural estimator model of aerodynamic derivatives is obtained. In order to show the capacity of this neural estimator model, this model is applied to the estimation of the derivatives of the simulated aircraft as well as to the estimation of the aerodynamic derivatives of a brazilian air force military jet aircraft, the Xavante AT-26. These proposed methodologies reduce the cost of obtaining the aerodynamic derivatives and show the estimation effectiveness of the neural networks to estimate the responses of an aircraft inside a pre-defined flight envelope.

Desenvolvimento de métodos sorológicos para diagnóstico de infecções pelos vírus Chikungunya e Mayaro / Development of methods for serological diagnose of Chikungunya and Mayaro infections

Devido a existência de 2 alphavírus artritogênicos no Brasil, os vírus Mayaro (MAYV) e Chikungunya (CHIKV) tornou-se importante desenvolver testes diagnósticos eficazes para discriminar suas infecções. No presente trabalho, desenvolvemos ELISAs indiretos para diagnóstico de CHIKV e MAYV utilizando proteínas de envelope viral E2 recombinantes, produzidas em Escherichia coli, as rE2-CHIKV e rE2-MAYV ELISAs. As proteínas E2 recombinantes tiveram suas antigenicidades verificadas nos ensaios utilizando anticorpos policlonais oriundos de camundongos hiperimunizados com CHIKV, MAYV e outros alphavírus. O rE2-CHIKV ELISA detectou anticorpos murinos de forma homotípica e não produziu reações cruzadas evidenciáveis utilizando anticorpos murinos específicos contra outros Alphavírus. O rE2-MAYV ELISA detectou anticorpos murinos homotípicos e também, reagiu cruzadamente com anticorpos murinos anti-CHIKV, mas não para outros Alphavírus. Esses ELISAs, também, foram usados na detecção de anticorpos em soros de pacientes com suspeita de infecção arboviral. Pelo o rE2-CHIKV ELISA, testaram-se 59 soros, resultando em 26 amostras IgG positivas. Resultados desse ELISA, quando comparados aos obtidos por teste de neutralização, demonstraram sensibilidade de 89,66% e especificidade de 100%. Soros humanos IgG positivos foram detectados em altas diluições pelo rE2-CHIKV ELISA. Quanto a detecção de IgM, o rE2- CHIKV ELISA apresentou moderada concordância com outros ensaios sorológicos. Com rE2- MAYV ELISA, testaram-se 68 soros resultando em 23 amostras IgG positivas, das quais 11 também mostraram-se positivas em teste de neutralização, demonstrando sensibilidade de 100% e especificidade de 78,95%. Portanto, os rE2-CHIKV e rE2 MAYV ELISAs, particularmente para detecção de IgG, mostraram-se adequadamente sensíveis e específicos para serem validados em estudos com maiores números de amostras e serem aplicados ao diagnóstico de pacientes infectados com CHIKV e MAYV. / Due the existence of 2 arthritogenic alphaviruses in Brasil, the viruses Mayaro (MAYV) and Chikungunya (CHIKV), it became important the development of efficient diagnose tests to discriminate their infections. In the present work, we developed indirect ELISAs for CHIKV and MAYV diagnosis using viral recombinant envelope proteins E2, produced in Escherichia coli, the rE2-CHIKV and rE2-MAYV. The recombinant E2 proteins had their antigenicity confirmed in the assay by using polyclonal antibodies produced in hyperimmunized mice with CHIKV, MAYV and other alphaviruses. The rE2-CHIKV ELISA detected homotypic murine antibodies and did not produced detectable cross-reactivity signal when using murine antibodies from other alphaviruses. The rE2-MAYV ELISA detected homotypic antibodies and also cross-reacted with murine anti-CHIKV antibodies, but not to other alphaviruses. These ELISAs were also tested for the detection of human antibodies, using patient sera suspected of arboviral infection. For rE2- CHIKV ELISA, it were tested 59 sera, resulting in 26 positive IgG samples. These ELISA results, when compared to those of a neutralizing assay, demonstrated a sensibility of 89.66% and specificity of 100%. The IgG positive human sera were detected in high dilutions by rE2-CHIKV ELISA. Regarding the detection of IgM, the rE2-CHIKV ELISA showed a moderate samples detection agreement when compared to other serologic assays. For rE2-MAYV ELISA, it were tested 68 sera, resulting in 23 positive IgG samples, of which 11 demonstrated to be positive by the neutralization assay, demonstrating a sensibility of 100% and specificity of 78.95%. Therefore, the rE2-CHIKV and rE2-MAYV ELISAs, especially for IgG detection, demonstrated to be properly sensitive and specific to be validated in studies using a greater number of samples, and also to be applied in the diagnosis of infected CHIKV and MAYV patients.

THE APPLICATION OF RADAR ENVIRONMENT SIMULATION TECHNOLOGY TO TELEMETRY SYSTEMS

This is a very expensive, time-consuming process and does not necessarily exercise all of the functions and capabilities available in new digital antenna controllers. This paper introduces Radar Environment Simulator technology and its application to testing of telemetry tracking systems. Measured results are shown, demonstrating that operational environment simulation is a valuable approach to quickly and effectively characterize the real time operation of a telemetry tracking system.

8PSK Signaling Over Non-Linear Satellite Channels

Budget reductions is another problem that the space agencies must deal with. This budget constraint results in simpler spacecraft carrying less communication capabilities and also the reduction in staff to capture data in the earth stations. It is then imperative that the most bandwidth efficient communication methods be utilized. This paper gives the results of a computer simulation study on 8 Level Phase Shift Keying (8PSK) modulation with respect to bandwidth, power efficiency, spurious emissions, interference susceptibility and the non-constant envelope effect through a non-linear channel. The simulations were performed on a Signal Processing Worksystem (SPW: software installed on a SUN SPARC 10 Unix Station and Hewlett Packard Model 715/100 Unix Station). This work was conducted at New Mexico State University (NMSU) in the Center for Space Telemetry and Telecommunications Systems in the Klipsch School of Electrical and Computer Engineering.

PERFORMANCE STUDY OF ENHANCED FQPSK AND CONSTRAINED ENVELOPE MODULATION TECHNIQUES

International Telemetering Conference Proceedings / October 21, 2002 / Town & Country Hotel and Conference Center, San Diego, California / This paper investigates the spectral properties and the bit error rate (BER) performance of enhanced FQPSK (EFQPSK) and constrained envelope modulation (CEM) techniques. Both the techniques are found to provide good spectral efficiencies. The EFQPSK signals are found to generate spectral lines for unbalanced data. An analytical spectral study for the spectral lines is presented. While the performance of CEM techniques has been presented in [6] for an ideal nonlinear amplifier, we present results for more realistic amplifiers with AM/AM and AM/PM effects. It is shown that such an amplifier generates spectral regrowth and a predistorter is required to reduce the adverse effects. A BER performance study with/without channel coding is also presented for the two techniques.

More recently, numerous terrestrial applications of OQPSK and similar waveforms (Feher-patented FQPSK) have been proposed. The present paper describes a simple non-proprietary modification of the MIL-STD SOQPSK waveform which offers spectral containment and detection efficiency comparable to or better than FQPSK-B (Revision A1), while preserving a constant envelope characteristic and backward compatibility with existing equipment.

Electron spin echo envelope modulation spectroscopy of radical pairs in photosynthetic bacteria

Electron spin echo envelope modulation (ESEEM) spectroscopy is widely used to study the radical pairs created during the primary steps of photosynthesis. In this thesis the analysis of ESEEM spectra is improved, and some new applications and variations of this experiment suggested. Experimental spectra from species such as P<sup>+</sup>Q<sup>-</sup><sub>A</sub>, the secondary radical pair formed in the reaction centre of the bacterium Rhodobacter sphaeroides, give information about the exchange and dipolar couplings between the radicals. The model used to analyse the data affects the results; this thesis suggests two improvements. First, the effect of anisotropic hyperfine couplings in the radicals is considered by the addition of a single spin-1/2 nucleus to the model. This approach suggests that previous models neglecting the effect of nuclei may have been slightly in error. Secondly, several model fittings are performed in the time domain. This approach avoids the Fourier transformation to the frequency domain so that experimental dead-time does not corrupt the data. An excellent fit to experimental data is found with a model containing one spin-1/2 nucleus on each radical. The hyperfine coupling parameters resulting from the fit are consistent with independent experimental results. Use is made of the method of Cramér-Rao lower bounds to assess the precision to which experimental parameters are determined from a time domain curve fitting. It is shown that the lower bounds may also be used to determine the optimum sampling strategy for the experiment. An example is given of the novel use of ESEEM to determine the distance between the radicals in the strongly coupled, uncorrelated radical pair Q<sup>-</sup><sub>A</sub>Q<sup>-</sup><sub>B</sub> ESEEM has not yet been used for this purpose, and the simulated spectra produced here indicate that the experiment could be used to evaluate the dipolar coupling and hence the inter-radical distance. This thesis considers the possibility of performing ESEEM at higher frequencies than are usually considered. Calculations show that the increased resolution of the g-tensors allow an experiment performed at the W-band frequency of 95 GHz to make a correlation between the relative orientations of the radicals and the dipolar axis, information which has previously been unavailable from a single experiment.
