Acarreta prejuízos econômicos com a condenação total ou parcial das carcaças, bem como reflexos em saúde pública, devido ao potencial zoonótico dos agentes causais. O presente estudo investigou a ocorrência e as principais espécies do gênero Mycobacterium, assim como marcadores de virulência plasmidial em linhagens de Rhodococcus equi (R. equi) isoladas de linfonodos e das fezes de suínos de abatedouros do interior do estado de São Paulo, com e sem linfadenite. Foram examinados 150 linfonodos (50 mesentéricos, 50 mediastínicos e 50 submandibulares) com lesões, 150linfonodos (50 mesentéricos, 50 mediastínicos e 50 submandibulares) sem lesões aparentes e 150 fezes de suínos, provenientes de animais destinados ao abate, em dois frigoríficos do interior do estado de São Paulo. As amostras de linfonodos e fezes foram submetidas ao cultivo microbiológico simultaneamente nos meios de ágar acrescido de sangue bovino (5%) desfibrinado, e meios seletivos de CAZ-NB, TCP e TVP para R. equi, e Stonebrink–Lesslie e Lowenstein-Jensen para micobactérias. As colônias sugestivas de R. equi e positivas no teste de CAMP, foram enviadas ao Japão para detecção de linhagens VapA ou VapB, associadas a virulência. As linhagens sugestivas no cultivo microbiano para o gênero Mycobacterium foram submetidas a caracterização de espécies por PCR pela técnica de PRA. Foram identificados nos linfonodos de suínos com lesões 48 (32,0%) linhagens de Mycobacterium spp.e 6 (4,0%) Rhodococcus equi. Nas fezes foram identificadas 40 (26,6%) linhagens de Rhodococcus equi e ... / The infectious lymphadenitis represents one of the most important diseases in pigs worldwide caused by bacterium, usually diagnosed on the slaughterhouses.The disease leads to economic losses due to total or partial condemnation of carcasses, as well as public health concern due to the zoonotic potential of microorganisms. The present study investigated the occurrence and the main species of the genus Mycobacterium as well as virulence markers of plasmid in Rhodococcus equi (R. equi) strains isolated from lymph nodes and feces from pigs of slaughterhouses in the state of São Paulo, with and without lymphadenitis. Were sampled 150 lymph nodes (50 mesenteric, 50 mediastinal and 50 submandibular) with lesions, 150 lymph nodes (50 mesenteric, 50 mediastinal and 50 submandibular) without visible lesions, and 150 feces from pigs of slaughterhouses of state of São Paulo, Brazil. The lymph nodes samples and feces were subjected to microbiological culture simultaneously indefibrinated bovine blood agar (5%), selective media of CAZ-NB, TCP, TVP for R. equi, and Stonebrink-Lesslie, and Lowenstein-Jensen for mycobacteria . The suggestive colonies of R. equi and positive to CAMP test were sent to Japan for evaluation of plasmid profile (VapA or VapB). The suggestive of Mycobacterium sp. were subjected to polymerase chain reaction (PCR) – based species identification using restriction enzyme pattern analysis (PRA). Among lymph nodes of pigs with lesions, 48 (32.0%) Mycobacterium spp. and 6 (4.0%) R. equi strains were identified. In the lymph nodes of pigs without lesions were identified 11 (7.3%) Mycobacterium spp. and none R. equi strain. O colostro é rico em imunoglobulinas, que constitui para esses animais a única fonte de proteção contra os agentes infecciosos durante o período neonatal, fase em que os animais apresentam maior susceptibilidade às doenças. Dentre estas, destaca-se a infecção ocasionada pelo Rhodococcus equi, responsável por elevadas taxas de mortalidade e grandes perdas econômicas. A infecção possui distribuição mundial e causa pneumonia supurativa e enterite associada a linfadenite em potros com menos de seis meses de idade. Apesar da sua grande importância no Brasil, a rodococose ainda é pouco estudada em nossas condições. O objetivo deste estudo foi comparar a resposta imune humoral entre éguas das raças Brasileiro de Hipismo (BH) e Bretã previamente imunizadas para R. equi, e avaliar o efeito da imunoprofilaxia ativa materna na transferência de anticorpos específicos protetores, via colostro, para neonatos eqüinos, utilizando na detecção de anticorpos o ensaio imunoenzimático indireto (ELISA teste), comparando dois diferentes antígenos (APTX e comercial). Os resultados demonstraram que ocorreu aumento na titulação de anticorpos anti-Rhodococcus equi nos grupos de éguas após a vacinação e transferência de anticorpos através do colostro para os potros neonatos. Não houve efeito da raça na produção de anticorpos. O antígeno comercial detectou títulos de anticorpos maiores e mais persistentes do que o antígeno APTX... . / The passive immunity transfer through colostrum is very important to new-born foals survival. The colostrum is immunoglobulin full, wich is responsable to protect against infectious agents during new-born period, stage that animals present higher susceptibility to diseases. Among these, the infection caused by Rhodococcus equi has the great importance, responsable for large economics damage and high mortality rates in animals illness. The infection has a world-wide distribuition and it causes suppurative pneumonia and enterits associated to limphadenitis in foals under 6 months old. Although its great importance in Brazil, this disease is not well studied. The aims of this study was to compare the humoral immune respost between the breeds Brasileiro de hipismo (BH) and Breton, previously vaccinated against Rhodococcus equi, and to evaluate the matern immunoprofilaxy effect in the specific antibodies transfer through colostrum to new-born foals, using the Enzyme linked immunosorbent assay (ELISA-test) comparing two differents antigens (APTX and comercial). The results demonstrated that there were an increase in mares antibodies after vaccination and also antibodies transfer through colostrum to new-born foals. There was no breed effect in the antibodies production. The Comercial antigen demonstrated antibodies titulation higher and more persistents than APTX antigen. The ELISA-test demonstrated sensible R. equi antibodies determination.

Molecular characterization of Babesia caballi and Theileria equi, the aetiological agents of equine piroplasmosis, in South Africa

In an attempt to develop quantitative real-time PCR (qPCR) assays for the detection of equine piroplasms, sequence heterogeneity in the V4 hypervariable region of the 18S ribosomal RNA (rRNA) gene sequences within both Theileria equi and Babesia caballi from South Africa was discovered. A molecular epidemiological survey of the protozoal parasites that cause equine piroplasmosis was therefore carried out using horse and zebra samples from different geographical locations around South Africa. We evaluated the ability of a recently developed T. equi-specific qPCR assay in detecting all T. equi 18S rRNA variants identified in South Africa. We further present the first report on the development and application of a TaqMan minor groove binder (MGB™) qPCR assay, targeting the 18S rRNA gene, for the detection of B. caballi infections in equine blood samples. Despite the ability of the 18S rRNA T. equi- and B. caballi-specific qPCR assays to detect all known 18S rRNA gene sequence variants thus far identified in South Africa, the existence of as yet undetected variants in the field cannot be overlooked. Other qPCR assays targeting alternative genes could be developed which, used in conjunction with the 18S rRNA qPCR assays, may provide better confirmation of test results. A T. equi-specific qPCR assay targeting the equi merozoite antigen gene (ema-1) was recently developed for the detection of T. equi parasites in the midgut of Rhipicephalus (Boophilus) microplus nymphs. This assay was not able to detect T. equi in all South African samples that were confirmed positive by other molecular and serological assays. Sequence characterization of the ema-1 gene from South African isolates revealed the existence of variation in the regions where the qPCR primers and probes had been designed. Based on these observations, a conserved region of the ema-1 gene was selected and targeted in the development of an ema-1-specific TaqMan MGB™ qPCR assay, which was shown to have a higher sensitivity than the previously reported ema-1 qPCR assay. The rhoptry-associated protein (rap-1) gene from South African B. caballi isolates was also characterized following the failure of a B. caballi-specific competitive-inhibition enzyme-linked immunosorbent assay (cELISA) to detect B. caballi antibody in the sera of infected horses from South Africa. The genome walking PCR technique was used to amplify the complete rap-1 gene sequence from two South African B. caballi isolates. Significant heterogeneity in the rap-1 gene sequences and in the predicted amino acid sequences was found. Marked amino acid sequence differences in the carboxy-terminal region, and therefore the probable absence of the monoclonal antibody binding site, explains the failure of the cELISA to detect antibody to B. caballi in sera of infected horses in South Africa. This is the first comprehensive molecular study of the parasites that cause equine piroplasmosis in South Africa. The Chondroid Conundrum: Transpharyngeal Removal of Guttural Pouch Chondroids in Horses

Master of Science / Strangles is an extremely infectious disease of horses that has negative financial and health consequences. After infection, some horses form balls of concrete-like pus in a structure in an air-filled sac of the ear canal called guttural pouches (GPs). These concretions, called chondroids, allow affected horses to continue to pass on infection to other horses. Removal of chondroids is needed to stop disease transmission but current non-surgical and surgical removal techniques have limitations. Surgical laser passed through an endoscope (medical camera) allows minimally invasive surgery of the guttural pouches. We performed laser surgery on cadaver horse heads to create access to the GP, then evaluated our ability to remove chondroid-like beads from the GP using a custom-made 3D-printed instrument. Our results showed efficient and complete removal of the beads compared to lavage without the 3D-printed instrument, with little damage to the heads. These findings are an initial step to development of a new chondroid-removal technique and may allow surgeons to more efficiently remove chondroids without the need for invasive surgery.

Purification and quantitative description of Rhodococcus equi IgG designed for aerosol nebulization to foals

The objective of this study was to purify IgG from commercially available hyperimmune Rhodococcus equi plasma and to assess the delivery of IgG as an aerosol to the equine lung. IgG was purified from plasma, and the IgG concentration of both the plasma and the purified IgG was determined by ELISA. The purified IgG was aerosolized using a vibrating mesh nebulizer and aerosol characterization was performed using cascade impaction. The purified IgG was nebulized to six healthy adult horses in order to assess the efficacy of pulmonary delivery and safety of administration. Bronchoalveolar fluid was retrieved endoscopically using a low volume technique prior to aerosolization (time 0) and at 0.5, 4 and 24 hours post aerosolization. The BAL fluid IgG concentration was determined and cytologic analysis was performed. The IgG concentrations of the plasma and purified IgG were 2,175 mg/dL and 1,145 mg/dL, respectively. The MMAD of the purified IgG aerosol was 4.7 microns. The mean BAL fluid IgG concentration increased 61% from 19.33 µg/dL at time 0 to 31.5 µg/dL at 0.5 hours, but this increase was not significant (P=0.603). No significant change was observed in inflammatory cell numbers over time or at any time point during the study. This study demonstrated that IgG antibodies were purified at a concentration acceptable for nebulization, and that the nebulization unit generated aerosol particles from the IgG solution of appropriate size for pulmonary delivery. Nebulization of purified IgG to adult horses was well tolerated and caused no local or systemic adverse effects. / Master of Science

Yes / Peptidyl-prolyl isomerase (PPIase) lipoproteins have been shown to influence the virulence of a number of Gram-positive bacterial human and animal pathogens, most likely through facilitating the folding of cell envelope and secreted virulence factors. Here, we used a proteomic approach to demonstrate that the Streptococcus equi PPIase SEQ0694 alters the production of multiple secreted proteins, including at least two putative virulence factors (FNE and IdeE2). We demonstrate also that, despite some unusual sequence features, recombinant SEQ0694 and its central parvulin domain are functional PPIases. These data add to our knowledge of the mechanisms by which lipoprotein PPIases contribute to the virulence of streptococcal pathogens.

Current antibiotic therapy is often ineffective and thus recent attention has focused on vaccine development. A systematic understanding of S. equi virulence, leading to the identification of targets to which protective immunity can be directed, is a prerequisite of the development of such a vaccine. Here, the virulence factors of S. equi are reviewed.

Com o objetivo de avaliar os equinos alojados no Jóquei Clube de São Paulo, Brasil, quanto a presença de anticorpos contra Theileria equi e Babesia caballi, foram testadas 180 amostras de soro sanguíneo por meio da técnica de C-ELISA (Competitive Enzyme-Linked Immunosorbent Assay), metodologia atualmente recomendada pela OIE (Organização Internacional de Epizootíases) por ter alta sensibilidade e especificidade. A frequência de animais com sorologia positiva para Theileria equi foi de 6,66% (12/180), para Babesia caballi foi de 22,3% (40/180) e para infecção concomitante foi de 6,66% (12/180). Os resultados sorológicos obtidos por este estudo revelam que 35,5% (64/180) dos animais possuem anticorpos contra a babesiose equina sendo que a maioria dos animais acometidos tem dois e três anos de idade e portanto estão há menos tempo no hipódromo. Fatores como a ausência de carrapatos vetores, o uso de terapias babesicidas repetidas e o longo tempo de permanência dos animais no Jóquei após o tratamento, favorecem a diminuição dos títulos de anticorpos sem que ocorra reinfecção. Esses fatores podem justificar o menor número de animais com sorologia positiva para a doença nos cavalos com idade acima de quatro anos. Considerando-se esses resultados sugere-se que os animais sejam avaliados sorologicamente ao ingressarem no Jóquei Clube de São Paulo para que o uso de medicamentos contra a doença seja feito de forma adequada e para que os sinais clínicos compatíveis com babesiose equina em animais sorologicamente negativos sejam melhor avaliados e considerados em diagnósticos diferenciais. / In order to evaluate the presence of antibodies against Theileria equi and Babesia caballi in horses kept at the Jockey Club in São Paulo city, Brazil, a total of 180 samples of blood serum was tested using the Competitive Enzyme-Linked Immunosorbent Assay (C-ELISA test). This methodology has been recommended by the International Organization of Epizooties (IOE) due to its high sensitivity and specificity. The frequency of seropositive animals for Theileria equi, Babesia caballi and for both was 6.66% (12/180), 22.3% (40/180) and 6.66% (12/180), respectively. Serological results showed that 35.5% of the animals (64/180) had antibodies against equine piroplasmosis; they were from two to three years old and were at the Jockey Club for a shorter period of time. Factors such as absence of thick vectors, repeated therapy using babesicidal drugs and the long period of time that the animals stayed in the Jockey Club after treatment favoured the lowering of antibody titers with no reinfection. These factors might be responsible for the fewer number of animals with positive serology for the disease in horses over four years of age. Based on these findings, animals should be serologically evaluated at the time of entrance into the Jockey Club so that the use of drugs against the disease be performed properly and clinical signs suggestive of equine babesiosis in serologically negative animals be better evaluated and considered for differential diagnosis.

Isolamento de bacteriófagos líticos para Rhodococcus sp

O Rhodococcus equi é uma das principais causas de pneumonia piogranulomatosa e linfadenite em potros de 1-6 meses de idade, resultando em alta morbidade e muitas vezes em óbito. Em propriedades endêmicas, a enfermidade é responsável por gerar altos custos para tratar e prevenir a doença. Com o objetivo de propor uma alternativa para o controle ambiental do Rhodococcus equi, foi aplicado um protocolo para isolamento de bacteriófagos. Foram isoladas 14 amostras de bacteriófagos, obtidas do ambiente de criação de cavalos. Estas apresentaram capacidade de lisar in vitro amostras de campo de Rhodococcus equi com diferentes intensidades. O resultado obtido nos possibilita sugerir que os fagos podem ser uma alternativa natural para a redução da bactéria no ambiente. / Rhodococcus equi is an important cause of pyogranulomatus pneumonia and lymphadenitis in foals 1-6 months of age, resulting in high morbidity and often mortality. In endemic properties, the disease is responsible for generating high costs to treat and prevent disease. In order to propose an alternative to the environmental control of Rhodococcus equi, a protocol for isolation of bacteriophages was applied. Fourteen bacteriophage samples were isolated obtained from horse breeding environment. They showed capacity to lyse in vitro Rhodococcus equi field samples with different sensitivities. The results obtained allows us to suggest the phage as a natural alternative to reducing R. equi population.

Monitoramento de potros por ultrasonografia torácica, cultura bacteriológica e pcr: diagnóstico de infecção subclínica por Rhodococcus equi / Monitoring foals by thoracic ultrasonography, bacterial culture and PCR : diagnostic of Rhodococcus equi subclinical pneumonia

Rhodococcus equi (R.equi), uma bactéria gram-positiva intracelular facultativa, é uma causa importante de pneumonia em potros com idade entre 3 semanas e 5 meses. A manifestação clinica mais comum da doença é a broncopneumonia piogranulomatosa com abscedação. Na pneumonia causada por R. equi os primeiros sinais clínicos podem não ser aparentes até que as alterações patológicas estejam bastante avançadas, por esse motivo, o diagnóstico precoce e acurado de potros com pneumonia por R. equi se torna fundamental. O diagnóstico definitivo baseia-se na detecção de R. equi na cultura bacteriológica e identificação molecular a partir da amostra de lavado traqueal; no entanto, essa técnica é invasiva, traz riscos para o animal e é relativamente cara. A ultrassonografia (US) para detecção precoce tem se tornado uma pratica de rotina em muitas fazendas endêmicas para rodococose equina. Com o advento dessa prática de triagem, a forma mais identificada de pneumonia por R. equi tem sido a subclínica, onde os animais apresentam presença de alterações pulmonares mas não apresentam sinais clínicos da doença. Atualmente, vapA é o gene com função demonstrada na virulência. Identificação de R. equi por Reação em Cadeia da Polimerase (PCR) em amostras de fezes tem se mostrado efetivo para o reconhecimento precoce do agente. Ultrassonografia torácica e PCR das amostras de fezes e swab nasal foram realizadas em 22 potros desde as 3 até as 16 semanas de idade (intervalos de 15 dias) de 3 fazendas endêmicas de criação de cavalos no sul do Brasil para identificar a ocorrência de doença subclínica. A associação entre a ultrassonografia torácica e PCR das amostras de fezes possibilitaram a detecção de doença subclínica e identificação de pontos críticos de controle dessa doença. Considerando o fato de que 95.4% dos potros apresentaram doença subclínica e que nenhum deles desenvolveu a doença clínica demonstra que o tratamento desses casos não é justificável para a população analisada. / Rhodococcus equi, a gram-positive facultative intracellular pathogen, is an important cause of pneumonia in foals between 3 weeks and 5 months of age. Pneumonia caused by R. equi is an insidious disease in which clinical signs may not be apparent until pathologic changes are well progressed. Because of the insidious progression of infection to severe clinical signs, early and accurate diagnosis of foals with R. equi pneumonia is important. Definitive diagnosis is based on R. equi detection by bacterial culture and molecular identification from tracheobronchial aspirate (TBA), this procedure is invasive, labor-intensive, requires skill, carries risks to foals, and is relatively expensive. The sequential thoracic ultrasonography (TUS) to early detection of the disease has been adopted as a screening method in many endemic farms; for this reason, subclinical disease has been the most frequently observed form. Nowadays, vapA is the only virulent gene identified. Fecal polymerase chain reaction (PCR) is a noninvasive technique with good diagnostic accuracy. Thoracic ultrasound screening (TUS) and PCR from fecal and nasal swab samples were performed in 22 foals from 3 to 16 weeks of age from 3 endemic farms at south of Brazil to identify the occurrence of R. equi subclinical disease. The association of TUS and fecal PCR detection of virulent R. equi provided a possibility of identification of critical points in disease control. Considering the fact that 95.4% of the foals showed evidence of subclinical disease and none of them developed any signs of clinical disease, the antibiotic treatment was not reasonable for the foals followed.
