Estratégias de seleção de genótipos de soja para resistência à podridão vermelha das raízes / Selection strategies of soybean genotypes resistant to sudden death syndromeBernardi, Walter Fernando 01 September 2008 (has links)
Nas últimas décadas, a podridão vermelha das raízes da soja (PVR), causada pelo Fusarium solani f.sp. glycines (FSG), tornou-se uma doença séria nas regiões brasileiras onde já foi constatada, sendo a utilização de cultivares resistentes um componente fundamental de um sistema integrado de controle. Este trabalho teve por objetivo pesquisar estratégias de seleção de genótipos resistentes a PVR. Várias metodologias foram implementadas: avanço de progênies até a geração F7:2 e seleção em campo infestado, de progênies selecionadas em F2. Avaliou-se o cruzamento IAC 4 x Conquista em casa de vegetação (geração F3:2) e em campo infestado (gerações F3:2, F4:2 e F5:2), através de estudo de estabilidade e adaptabilidade das progênies. As metodologias de infecção em vasos de barro e bandejas de isopor em casa de vegetação foram comparadas para melhorar a eficiência de seleção; também foi feita análise de repetibilidade dos sintomas foliares da PVR, para otimização do número de avaliações necessárias para classificar o genótipo de soja quanto à reação ao FSG. Cultivares brasileiras de soja foram avaliadas em campo infestado. Em campo, as plantas foram avaliadas no estádio R5-6, com notas variando de 1 (ausência de sintomas) a 5 (100% da raiz principal com sintomas). Em casa de vegetação, as plantas foram avaliadas aos 35 dias pós-semeadura para sintomas radiculares e da parte aérea. Houve eficiência da seleção para resistência a PVR tanto em F2 (sintomas foliares em casa de vegetação) quanto em F7:2 (sintomas radiculares em campo naturalmente infestado). A seleção praticada nas gerações intermediárias também foi eficiente para aumentar a produtividade de grãos das progênies. Para o cruzamento IAC 4 x Conquista nas gerações F3:2, F4:2 e F5:2 em campo naturalmente infestado e na F3:2 em casa de vegetação, a metodologia de Annicchiarico possibilitou a seleção de genótipos com maior estabilidade e adaptabilidade para os diferentes ambientes; com o estudo genético destas gerações, demonstrou que há efeitos gênicos aditivos e dominantes no controle genético da PVR. Além disso, constatou-se que os genes responsáveis pela resistência estão dispersos nos genitores, provavelmente agrupados em blocos gênicos. A presença de dominância indica que a seleção deve ser postergada para gerações com maior homozigose (linhagem pura); esta idéia foi reforçada pela baixa herdabilidade ao nível de plantas. O uso de bandejas de isopor foi significativamente mais eficiente do que vasos de barro para inoculação de FSG em casa de vegetação. Pela análise de repetibilidade, detectou-se que quatro avaliações foram suficientes para discriminar se o genótipo era realmente suscetível ao FSG. A correlação da geração F3:2 (IAC 4 x Conquista) entre campo infestado e casa de vegetação foi praticamente nula; no entanto, 54% das progênies selecionadas em campo também foram selecionadas em casa de vegetação. A seleção de genótipos superiores para resistência ao FSG não é tarefa fácil, mas pode ser aprimorada pelo uso conjugado de metodologias suplementares que aumentem a eficiência de seleção. / During the last decades, soybean sudden death syndrome (SDS), caused by Fusarium solani f.sp. glycines (FSG), has become a serious disease in the regions where it was encountered. The use of resistant soybean cultivars has been an important component of an integrated management system. The aim of this work was to research selection strategies of SDS-resistant genotypes. A number of methodologies were used: self pollination up to generation F7:2 and progeny selection in infested field of selected F2. The crossing IAC 4 x Conquista was studied in greenhouse (generation F3:2) and in a naturally infested field (generations F3:2, F4:2 and F5:2), where the progeny stability and adaptability study was carried out. The methodologies of infection in clay pots and polystyrene trays in greenhouse were compared to improve selection efficiency. An analysis of the repeatability of foliar SDS symptoms was also performed in order to optimize the number of evaluations needed to classify a soybean genotypes reaction to FSG. Brazilian soybean cultivars were evaluated in a naturally infested field, where plants were evaluated at stage R5-6 with scores varying from 1 (absence of symptoms) to 5 (main root 100% symptoms). In the greenhouse, the plants were evaluated 35 days after sowing for root symptoms, while the leaf symptoms was only evaluated after the appearance of symptoms. The selection done in F2 (leaf symptoms in greenhouse) as well as F7:2 (root symptoms in infested field) for FSG were efficient. The selections performed in intermediary generations were efficient in increasing the grain yield of the progeny. Studies of the F3:2, F4:2 and F5:2 generations of the IAC 4 x Conquista cross in an infested field and F3:2 in greenhouse, enabled through Annicchiaricos methodology, the selection of genotypes with greater stability and adaptability to different environments. The genetic study of these generations demonstrated there are additive and dominant genetic effects on the genetic control of SDS. Moreover, it was noted that the genes responsible for resistance are dispersed on the genitors, probably grouped into genic blocks. The presence of dominance indicates that the selection should be passed onto generations with greater homozygosity. There is also a clear indication that breeders should work with progeny lines in order to assist selection due to the low inheritability at the individual plant level. For the experiments with different FSG inoculation methodologies (clay pots and polystyrene trays) in greenhouse the use of trays was significantly more efficient; by studying the repeatability analysis, it was found that four evaluations is enough to discriminate if the genotype is really susceptible to FSG. The correlation of the F3:2 generation (IAC 4 x Conquista) between infested field and greenhouse was practically null; however, 54% of the progeny selected in the field were selected in greenhouse. As observed, the selection of superior genotypes for FSG resistance is not an easy task, but may be improved by means of new methodologies and the conjugated use of methodologies which improve selection efficiency.

Previous issue date: 2015-02-27 / Capes / The objective of this study was to evaluate the incidence and damage of stalk rot (SR) in corn hybrids grown in tillage rotation with beans and succession to oats; quantify the efficiency of detection and control of Fusarium verticillioides in seed and seedlings health test according to the time of incubation. The first study was carried out at Muitos Capões municipal district on Rio Grande do Sul State, during the agricultural seasons of 2012/13 and 2013/14.The hybrids tested were P30F53 Hx, P32R22 YHR, DKB 250 VTPRO2, AS1555 VTPRO2, AS 1656 VTPRO2, AG8025 VTPRO e AG 9045 VTPRO. A randomized block design with four replications was employed. The incidence of SR was determined during harvesting and the damage by threshing operation analyzed separately in healthy and unhealthy plants. The means were compared by Scott-Knott test with a significance level of 5% (P <0.05). Statistical interaction between corn hybrid and harvesting season was detected. For this reason, the data was analyzed by harvesting season. In the 2012/13 growing season the prevailing diseases were diplodia blight, Anthracnosis and Fusarium, and as opposed to 2013/14, when Gibberella, Diplodia blight and Fusarium wilting were the prevailing in 2013/14. The incidence of SR in 2012/13 and 2013/14 ranged from 7.3% to 100% (46.2% in average), and from 3.14% to 72.6% (34.8% in average). The average of damage in the yield of hybrids were 6,48% and 8,75%, ranging from 197 kg ha-1 to 1205 kg ha-1 and from 113 kg ha-1 to 2536 kg ha-1, with an average of 640 kg ha-1 and 768 kg ha-1, respectively in 2012/2013 and 2013/14. In both growing seasons the hybrids P32R22 YHR and AG9045 VTPRO showed the highest SR incidence. The lowest incidence of this disease was detected in the P30F53 Hx and AG8025 VTPRO hybrids in 2012/13 and AG8025 VTPRO and DKB 250 VTPRO2 in 2013/14. The highest damages were found in AS1555 VTPRO2 (14%) and AG9045 VTPRO (11.2%) in 2012/13, and AG9045 VTPRO (26.3%) in
2013/14. The yield ranged from 8.430 kg ha-1 to 10.667 kg ha-1 in 2012/13 and 7.175 kg ha-1 and 8.790 kg ha-1 in the 2013/14. Secondly, hybrid corn seeds Status TL TG treated with metalaxyl-m + fludioxonil fungicides and seeds without seed treatment were cultured in PDA Culture Media to obtain symptomatic seedlings (infected by F. verticullioides) and asymptomatic, with 7 days of age. The samples were separated and transferred to test tubes containing agar-water culture media and maintained in growth chamber at 25 °C under 12 hours of photoperiod until the completion of 20 days. The treatments were T1 - uninfected seedlings without TS; T2 - infected seedlings without TS; T3 - uninfected seedlings with TS; T4 - infected seedlings with TS. The fungus incidence was quantified in the mesocotyl, crown and remaining seed and shoot and root length and dry matter weight were measured. Randomized block experimental design with eight replications was used. The data was submitted to variance analysis and Tukey’s test at 5% of significance level. The incidence of F. verticillioides in the remaining seeds was 96% for T1 and T3, and 100% for T2 and T4, with no difference between treatments. The incidence of F. verticillioides was 88% and 93% for mesocotyl, and 88% and 95% in the crown in the T1 and T3 seedlings, respectively. The root length showed 14.1 cm and 10.7 cm for T3 and T4 respectively, which are higher than T1 (9.6 cm) and T2 (5.8 cm). Higher root weight was detected in T3 (0.25 g) and T4 (0.18 g) cultivars compared to T1 (0.12 g) and T2 (0.09 g). T3 showed the highest shoot length (14.2 cm) when compared to the other treatments. The corn seed treatment with metalaxyl-m + fludioxonil fungicides is not effective in controlling F. verticillioides. The detection of F. verticillioides in BDA media at the seventh day of incubation does not represent the real incidence of the fungus, once the pathogen was recovered from asymptomatic seedlings when they were re-evaluated at 20 days. In all treatments, the seed treatment enhanced the seedlings development / Os objetivos deste trabalho foram avaliar a incidência e os danos causados por podridões do colmo (PC) em híbridos de milho cultivados em sistema de semeadura direta com rotação ao feijão e sucessão à aveia; e quantificar a eficiência de detecção e controle de Fusarium verticillioides em meio batata-dextrose-ágar em sementes e plântulas de milho em função do tempo de incubação. Foram realizados dois estudos, sendo estes conduzidos no município de Muitos Capões, estado do Rio Grande do Sul, nas safras agrícolas 2012/13 e 2013/14. Foram avaliados os híbridos P30F53 Hx, P32R22 YHR, DKB250 VTPRO2, AS1555 VTPRO2, AS1656 VTPRO2, AG8025 VTPRO e AG9045 VTPRO. O delineamento experimental foi de blocos casualizados com quatro repetições. A incidência de PC foi determinada na colheita do milho, sendo o dano mensurado pela trilha em separado de espigas das plantas sadias e plantas doentes. A comparação das médias foi realizada pelo teste de Scott-Knott, em nível de significância de 5% (p<0,05). Houve interação entre híbridos e safras, sendo os dados analisados separadamente por safra. Em 2012/13, as doenças predominantes foram diplodia, antracnose e fusariose; em 2013/14, houve predomínio de giberela, diplodia e fusariose. A incidência de PC em 2012/13 e 2013/14 variou de 7,3% a 100% (média de 46,2%) e de 3,14% a 72,6% (média de 34,8%), respectivamente. O dano médio no rendimento de grãos dos híbridos foi de 6,8% e 8,7%, respectivamente para 2012/13 e 2013/14. Nas duas safras os híbridos P32R22 YHR e AG9045 VTPRO apresentaram maior incidência de PC. As menores incidências foram detectadas nos híbridos P30F53 Hx e AG8025 VTPRO na safra de 2012/13, e AG8025 VTPRO e DKB250 VTPRO2 na safra de 2013/14. Os maiores danos foram
encontrados nos híbridos AS1555 VTPRO2 (14%) e AG9045 VTPRO (11,2%) em 2012/13, e AG9045 VTPRO (26,3%) em 2013/14. No segundo estudo, sementes de milho tratadas com fungicida metalaxil-m + fludioxonil e sem tratamento foram semeadas em meio de cultura de batata-dextrose-ágar para obtenção de plântulas sintomáticas (infectadas por F. verticillioides) e assintomáticas. Aos sete dias as plântulas foram separadas e transferidas para tubos de ensaio contendo meio de cultura ágar-água e mantidas em câmara de crescimento com temperatura de 25ºC e fotoperíodo de 12 h, onde permaneceram até completarem 20 dias. Obtiveram-se os tratamentos: T1 - plântulas assintomáticas sem fungicida; T2 - plântulas sintomáticas sem fungicida; T3 – plântulas assintomáticas com fungicida; T4 – plântulas sintomáticas com fungicida. A incidência foi quantificada no mesocótilo, coroa e semente remanescente das plântulas e o comprimento e peso seco da parte aérea e raiz. Os dados foram submetidos à análise de variância e teste de Tukey a 5%. A incidência de F. verticillioides em sementes remanescentes foi de 96% para T1 e T3, e 100% para T2 e T4, não diferindo os tratamentos entre si. Foi observada incidência de F. verticillioides de 88 e 93 % para mesocótilo, e 88 e 95% para coroa em plântulas provenientes de T1 e T3, respectivamente. O comprimento de raiz foi de 14,1 cm e 10,7 cm para T3 e T4 respectivamente, sendo estes superiores ao T1 (9,6 cm) e T2 (5,8 cm). Foram detectados maiores pesos de raiz nos tratamentos T3 (0,25 g) e T4 (0,18 g), quando comparados a T1 (0,12 g) e T2 (0,09 g). O T3 apresentou o maior comprimento de parte aérea,14,2 cm, em relação aos demais tratamentos. O tratamento de sementes com metalaxil-m + fludioxonil não foi eficiente no controle de F. verticillioides. A detecção de F. verticillioides em meio BDA aos sete dias de incubação não representou a real incidência do fungo, pois o patógeno foi recuperado de plântulas até então assintomáticas quando essas foram reavaliadas aos 20 dias. seed deterioration and root rot, stalk rot and ear in corn. This study was performed in
laboratory and aimed to assess the sanity of maize seeds treated with fungicides
commercialized in Santa Catarina and Rio Grande do Sul states determining the incidence and
frequency of fungus associated with seed occurrence; identify fungicides in seeds treatment
with eradicating action against Fusarium verticillioides ; adjust fungicides doses to the
eradicative control of F. verticillioides ; assess the seed treatments with fungicides
eradicatives of F. verticillioides related with germination and emergence of seedlings and
quantify the F. verticillioides transmission rate from seeds to the mesocotyl, crown and
coleoptile of seedlings from seeds with and without the treatment, sowed in soil under five
different temperatures. From 224 samples of commercial seeds the highest frequencies of
occurrence and incidence were obtained for F. verticillioides, Penicillium spp. and
Aspergillus flavus fungi with occurrence medium values of respectively 86,6%, 69,6%, 63,8%
and incidence of 14,0%, 12,4% and 8,4%. The results showed that the most used fungicide by
seed companies for seed treatment was the mixture fludioxonil + metalaxil. It is concluded
that the seed treatment is not eradicating the main fungi of seed. Test with fungicides, blends
and doses, in a sample of maize seeds with 41% of F. verticillioides incidence showed better
control with carbendazim+tolyfluanid, carbendazim and carbendazim+thiram. The rate adjust
was done in these fungicides with AS 1535 hybrid with 37% of F. verticillioides incidence,
testing rate indicated by manufacturer, 25%, 50%,75% and 100% beyond than indicated. The
carbendazim active principle isolated did not eradicate the fungos. The fungicides
carbendazim+ thiram (rate 75% beyond than indicated) and carbendazim+ tolylfluanid (rate
beyond than indicated) eradicated the in vitro F. verticillioides fungus. In climatized grout
chambers was determined that the F. verticillioides transmission was efficient in the
temperature of 21 and 27°C to the mesocotyl and crown in treatment without fungicide. In the
temperatures higher than 21°C the control of transmission was not achieved by seed treatment,
supporting that the fungicide efficacy in eradicate F. verticillioides must be evaluated in vivo .
The highest intensity of root rot was verified in the check control in 21°C with 34.7%, whereas
in this temperature were not detected root rottenness symptoms in seeds treated with
tolylfluanid+carbendazim. The minor seedlings emergence was obtained at 15°C. At 15ªC and
18°C these fungicides proportionate increase in the emerged plant population. From 21°C on
the seed treatment did not influence in the plant population. The fungicides
tolylfluanid+carbendazim and carbendazim+thiram showed the best results of F.
verticillioides control of transmission. It is possible conclude that the seed is source of
primary inoculum to F. verticillioides, the transmission rate is influenced by soil temperature
and the seedling emergence in cold soil sowing conditions can be improved with treatment of
seeds / As sementes infectadas constituem importante fonte de inóculo primário para os
fungos causadores de deterioração de sementes, podridões radiculares, podridões da base do
colmo e da espiga em milho. Os trabalhos foram conduzidos em laboratório e tiveram como
objetivos avaliar a sanidade de sementes de milho comercializadas no estado de Santa
Catarina e Rio Grande do Sul, determinando a incidência e a freqüência de ocorrência de
fungos associados às sementes; identificar fungicidas em tratamento de sementes com ação
erradicante contra Fusarium verticillioides; ajustar doses de fungicidas para controle
erradicativo de F. verticillioides; avaliar tratamentos de sementes com fungicidas
erradicativos de F. verticillioides em relação à germinação e emergência de plântulas e
quantificar a taxa de transmissão de F. verticillioides da semente para mesocótilo, coroa e
coleóptilo de plântulas em sementes com e sem tratamento, semeadas em solo sob cinco
temperaturas. De 224 amostras de sementes comerciais avaliadas, as maiores freqüências de
ocorrência e incidências foram obtidas para os fungos F. verticillioides, Penicillium spp. e
Aspergillus flavus com valores médios de ocorrência de 86,6%, 69,6%, 63,8% e incidência de
14,0%, 12,4% e 8,4%, respectivamente. Os resultados demonstraram que o fungicida mais
utilizado pelas companhias de sementes para tratamento foi o principio ativo
fludioxonil+metalaxil. Concluiu-se que o tratamento de sementes não está erradicando os
principais fungos da semente. Teste com fungicidas, misturas e doses, em amostra de
sementes de milho com 41,0% de incidência de F. verticillioides, indicaram melhor controle
com carbendazim+tolilfluanida (99,0%), carbendazim (95,5%) e carbendazim+tiram (78,0%).
O ajuste de dose foi feito nesses fungicidas com o híbrido AS 1535 com 37% de incidência de
F. verticillioides, testando dose indicada pelo fabricante, 25%, 50%, 75% e 100% superior à
indicada. O principio ativo carbendazim isolado não erradicou o fungo. Os fungicidas
carbendazim+tiram (dose 75% superior à indicada) e carbendazim+tolilfluanida (doses
superiores à indicada) erradicaram o fungo F. verticillioides in vitro . Em câmaras
climatizadas a transmissão de F. verticillioides foi eficiente na temperatura de 21 e 27 ºC para
o mesocótilo e coroa em sementes não tratadas. Nas temperaturas superiores a 21 ºC, o
controle da transmissão não foi alcançado pelo tratamento de semente, demonstrando que a
eficácia do fungicida em erradicar F. verticillioides deve ser avaliada in vivo . A maior
intensidade de podridão radicular foi verificada na testemunha aos 21 ºC com 34,7%, sendo
que nesta temperatura não foram detectados sintomas de podridão de raízes em sementes
tratadas com tolilfluanida+carbendazim. A menor emergência de plântulas foi obtida na
testemunha a 15ºC. A 15ºC e 18 ºC, esses fungicidas proporcionaram incremento na
população de plantas emersas. A partir de 21ºC o tratamento de sementes não influenciou na
população de plantas. Os fungicidas tolilfluanida+carbendazim e carbendazim+tiram
apresentaram os melhores resultados no controle da transmissão de F. verticillioides. Pode-se
concluir que a semente é fonte de inóculo primário para F. verticillioides e que a taxa de
transmissão é influenciada pela temperatura do solo e que a emergência de plântulas em
condições de semeadura de solo frio pode ser melhorada com o tratamento de sementes

Previous issue date: 2015-06 / A cultura da melancia pode ser afetada por diversos patógenos que atuam no sistema radicular das plantas, comprometendo a absorção e o transporte de água e nutrientes, interferindo nos processos vitais para o seu desenvolvimento. Os experimentos foram conduzidos no CCTA/UFCG, Campus de Pombal. O primeiro avaliou o efeito de diferentes doses (0,14mL, 28mL, 56mL e 112mL/planta) do composto orgânico Bokashi no controle da podridão radicular ocasionada por Fusarium spp. em duas etapas simultâneas utilizando solo estéril e não estéril em casa de vegetação, as doses foram aplicadas semanalmente utilizandos e a cultivar ‘Crimson Sweet’ que foi semeada em vasos contendo seis kg de solo argiloso, constituído de seis tratamentos com cinco repetições. Foi avaliada a população microbiana total do solo, colhido a 0, 30, e 60 dias após o plantio da melancia e a uma profundidade de aproximadamente 10 cm e próximo ao sistema radicular além da incidência e severidade da podridão radicular, e matéria fresca seca da parte aérea das plantas. O segundo experimento em campo avaliou as duas melhores doses do composto orgânico ‘Bokashi, (112ml e 56ml) do experimento anterior e outras doses (224ml e 0ml), assim distribuídos (T1 112ml com Trichoderma, T2 112ml sem Trichoderma, T3 56ml com Trichoderma, T4 56ml sem Trichoderma, T5 224ml com Trichoderma, T6 224ml sem Trichoderma, T7 0ml com trichoderma, T8 0ml sem Trichoderma) totalizando 8 tratamentos e cinco repetições onde foram avaliadas a incidência e severidade a podridão radicular, além das características agronômicas da cultura como acúmulo de massa das plantas, e produção total dos frutos e fisiológica como taxa fotossintética (A), condutância estomática (gs), transpiração (E) e concentração intercelular de CO2 (Ci), No estudo em casa de vegetação os tratamentos com dose de 56ml de bokashi associado ao Trichoderma e o que utilizou dose de 112ml de bokashi obtiveram resultado significativo na redução da Incidência de patógenos fúngicos em melancieiras plantadas em solo não estéril e na severidade da podridão radicular causada por Fusarium spp em solo estéril. Em campo a dose 112ml de bokashi associado ao Trichoderma obteve redução tanto na incidência de patógenos fúngicos quanto na severidade da podridão radicular em melancieiras. O bokashi associado ao Trichoderma obteve melhor controle na incidência e severidade de podridão radicular em melancieira. O bokashi e Trichoderma não interferiram no crescimento e produção da melancieira. O Trichoderma interferiu de forma negativa nas análises fisiológicas da melancieira, para a taxa fotossintética, condutância estomática, transpiração e concentração intercelular de CO2. Os tratamentos que utilizaram dose de 112ml e 56ml de bokashi sem a presença do Trichoderma foi a que apresentou um melhor incremento para taxa fotossintética e condutância estomática respectivamente, evidenciando a importância do bokashi no intuito de melhorar a resposta fisiológica da planta. / The culture of watermelon can be affected by various pathogens that act on the root system of
plants, affecting the absorption and transport of water and nutrients, interfering in the processes
vital to their development.The experiments were conducted in the CCTA / UFCG, Campus de
Pombal. The first evaluated the effect of different doses (0,14mL, 28ml, 56ml and 112mL /
plant) of Bokashi organic compound in the control of root rot caused by Fusarium spp. in two
simultaneous steps using sterile soil and non-sterile in greenhouse doses were applied weekly
using the watermelon cultivar 'Crimson Sweet' that was sown in pots containing six kilograms
of clay soil, made up of six treatments with five repetitions. We evaluated total soil microbial
population, collected at 0, 30, and 60 days after planting watermelon and to a depth of about 10
cm and close to the root system beyond the incidence and severity of root rot, dry and fresh
matter of watermelon. The second field experiment evaluating the top two doses of organic
compost 'Bokashi, (112ml and 56ml) of the previous experiment and other doses (224ml and
0ml), distributed as follows (with Trichoderma 112ml, 112ml without Trichoderma, with
Trichoderma 56ml, 56ml without Trichoderma, with Trichoderma a 224ml, 224ml without
Trichoderma, 0ml with Trichoderma, 0ml without Trichoderma) totaling eight treatments and
five replications which were assessed the incidence and severity of root rot and the agronomic
characteristics of culture as a mass accumulation of plants, photosynthetic rate (a), stomatal
conductance (gs), transpiration (E) and intercellular CO2 concentration (Ci), fresh and dry
matter of shoot and total production of fruits. In the study of greenhouse treatments with 56ml
dose of Bokashi associated with Trichoderma and what used dose of 112ml of Bokashi
achieved significant results in reducing fungal pathogens incidence in watermelon planted in
non-sterile soil and severity of root rot Fusarium spp in sterile soil. In the field dose of 112ml
Bokashi associated with Trichoderma achieved a reduction in both the incidence of fungal
pathogens as the severity of root rot in watermelon. Bokashi associated with Trichoderma got
better control the incidence and severity of root rot in watermelon.

Estratégias de seleção de genótipos de soja para resistência à podridão vermelha das raízes / Selection strategies of soybean genotypes resistant to sudden death syndromeWalter Fernando Bernardi 01 September 2008 (has links)
Nas últimas décadas, a podridão vermelha das raízes da soja (PVR), causada pelo Fusarium solani f.sp. glycines (FSG), tornou-se uma doença séria nas regiões brasileiras onde já foi constatada, sendo a utilização de cultivares resistentes um componente fundamental de um sistema integrado de controle. Este trabalho teve por objetivo pesquisar estratégias de seleção de genótipos resistentes a PVR. Várias metodologias foram implementadas: avanço de progênies até a geração F7:2 e seleção em campo infestado, de progênies selecionadas em F2. Avaliou-se o cruzamento IAC 4 x Conquista em casa de vegetação (geração F3:2) e em campo infestado (gerações F3:2, F4:2 e F5:2), através de estudo de estabilidade e adaptabilidade das progênies. As metodologias de infecção em vasos de barro e bandejas de isopor em casa de vegetação foram comparadas para melhorar a eficiência de seleção; também foi feita análise de repetibilidade dos sintomas foliares da PVR, para otimização do número de avaliações necessárias para classificar o genótipo de soja quanto à reação ao FSG. Cultivares brasileiras de soja foram avaliadas em campo infestado. Em campo, as plantas foram avaliadas no estádio R5-6, com notas variando de 1 (ausência de sintomas) a 5 (100% da raiz principal com sintomas). Em casa de vegetação, as plantas foram avaliadas aos 35 dias pós-semeadura para sintomas radiculares e da parte aérea. Houve eficiência da seleção para resistência a PVR tanto em F2 (sintomas foliares em casa de vegetação) quanto em F7:2 (sintomas radiculares em campo naturalmente infestado). A seleção praticada nas gerações intermediárias também foi eficiente para aumentar a produtividade de grãos das progênies. Para o cruzamento IAC 4 x Conquista nas gerações F3:2, F4:2 e F5:2 em campo naturalmente infestado e na F3:2 em casa de vegetação, a metodologia de Annicchiarico possibilitou a seleção de genótipos com maior estabilidade e adaptabilidade para os diferentes ambientes; com o estudo genético destas gerações, demonstrou que há efeitos gênicos aditivos e dominantes no controle genético da PVR. Além disso, constatou-se que os genes responsáveis pela resistência estão dispersos nos genitores, provavelmente agrupados em blocos gênicos. A presença de dominância indica que a seleção deve ser postergada para gerações com maior homozigose (linhagem pura); esta idéia foi reforçada pela baixa herdabilidade ao nível de plantas. O uso de bandejas de isopor foi significativamente mais eficiente do que vasos de barro para inoculação de FSG em casa de vegetação. Pela análise de repetibilidade, detectou-se que quatro avaliações foram suficientes para discriminar se o genótipo era realmente suscetível ao FSG. A correlação da geração F3:2 (IAC 4 x Conquista) entre campo infestado e casa de vegetação foi praticamente nula; no entanto, 54% das progênies selecionadas em campo também foram selecionadas em casa de vegetação. A seleção de genótipos superiores para resistência ao FSG não é tarefa fácil, mas pode ser aprimorada pelo uso conjugado de metodologias suplementares que aumentem a eficiência de seleção. / During the last decades, soybean sudden death syndrome (SDS), caused by Fusarium solani f.sp. glycines (FSG), has become a serious disease in the regions where it was encountered. The use of resistant soybean cultivars has been an important component of an integrated management system. The aim of this work was to research selection strategies of SDS-resistant genotypes. A number of methodologies were used: self pollination up to generation F7:2 and progeny selection in infested field of selected F2. The crossing IAC 4 x Conquista was studied in greenhouse (generation F3:2) and in a naturally infested field (generations F3:2, F4:2 and F5:2), where the progeny stability and adaptability study was carried out. The methodologies of infection in clay pots and polystyrene trays in greenhouse were compared to improve selection efficiency. An analysis of the repeatability of foliar SDS symptoms was also performed in order to optimize the number of evaluations needed to classify a soybean genotypes reaction to FSG. Brazilian soybean cultivars were evaluated in a naturally infested field, where plants were evaluated at stage R5-6 with scores varying from 1 (absence of symptoms) to 5 (main root 100% symptoms). In the greenhouse, the plants were evaluated 35 days after sowing for root symptoms, while the leaf symptoms was only evaluated after the appearance of symptoms. The selection done in F2 (leaf symptoms in greenhouse) as well as F7:2 (root symptoms in infested field) for FSG were efficient. The selections performed in intermediary generations were efficient in increasing the grain yield of the progeny. Studies of the F3:2, F4:2 and F5:2 generations of the IAC 4 x Conquista cross in an infested field and F3:2 in greenhouse, enabled through Annicchiaricos methodology, the selection of genotypes with greater stability and adaptability to different environments. The genetic study of these generations demonstrated there are additive and dominant genetic effects on the genetic control of SDS. Moreover, it was noted that the genes responsible for resistance are dispersed on the genitors, probably grouped into genic blocks. The presence of dominance indicates that the selection should be passed onto generations with greater homozygosity. There is also a clear indication that breeders should work with progeny lines in order to assist selection due to the low inheritability at the individual plant level. For the experiments with different FSG inoculation methodologies (clay pots and polystyrene trays) in greenhouse the use of trays was significantly more efficient; by studying the repeatability analysis, it was found that four evaluations is enough to discriminate if the genotype is really susceptible to FSG. The correlation of the F3:2 generation (IAC 4 x Conquista) between infested field and greenhouse was practically null; however, 54% of the progeny selected in the field were selected in greenhouse. As observed, the selection of superior genotypes for FSG resistance is not an easy task, but may be improved by means of new methodologies and the conjugated use of methodologies which improve selection efficiency.

Recherche de déterminants génétiques impliqués dans la résistance à la Fusariose de l'épi chez le blé tendre / Identification of loci involved in the genetic resistance to Fusarium head blight in wheatLe Couviour, Fabien 28 June 2011 (has links)
La fusariose des épis des céréales est due à un champignon pathogène (Fusarium spp.) qui entraine non seulement une perte directe de rendement en interrompant le remplissage des grains, mais également une perte indirecte par la production de mycotoxines, responsables de perturbations de procédés industriels et d'intoxications alimentaires. Parmi les différents moyens de lutte, la création de variétés résistantes semble la stratégie la plus efficace et la plus durable. Plusieurs variétés exotiques ont ainsi été identifiées comme résistantes mais présentent la particularité d'être difficilement cultivables sous nos latitudes. L'obtention de variétés résistantes adaptées à nos climats passe donc par une meilleure connaissance des mécanismes de résistance disponibles. La détection de QTL (Quantitative Trait Loci) chez le blé tendre a déjà permis de localiser certains des facteurs génétiques impliqués dans des mécanismes de résistance. Cependant l'utilisation de ces résultats en amélioration des plantes se heurte à deux contraintes majeures : la position des QTL reste le plus souvent imprécise et ces QTL, dérivant de populations biparentales, ne représentent qu'une fraction de la diversité génétique potentiellement intéressante pour ce caractère. L'objectif de la thèse était d'étudier la résistance du blé à la fusariose en réalisant une approche de génétique d'association. L'utilisation de génotypes faiblement apparentés et d'une densité élevée de marqueurs a permis de détecter des associations entre le polymorphisme génétique et des variations phénotypiques. Après avoir réalisé une synthèse sur le pathogène et sur l'approche par génétique d'association, le premier travail a été de réaliser une carte génétique fiable afin de pouvoir localiser les associations mises en évidence. Un panel de 195 variétés de blé tendre élites a été évalué pour leur résistance globale à la fusariose, pour leur résistance à la progression des symptômes et pour leur résistance à l'accumulation des mycotoxines. Des notations morphologiques (précocité, taille, aristation, nombre d'épillets par épi et extrusion des anthères) ont également été réalisées afin d'évaluer leur influence dans l'infestation. Ces variétés ont été génotypées avec 3016 marqueurs SNP, 200 marqueurs SSR et 1400 marqueurs DArT. Préalablement à la réalisation des tests d'association, l'étude de la structure a mis en évidence trois origines géographiques (française, anglaise et allemande) des variétés du panel. De même, l'étude du déséquilibre de liaison a montré que celui-ci était conservé sur une distance comprise entre 2 et 6 cM. L'étude d'association réalisée sur les notations d'infestation a permis d'identifier plusieurs zones associées, dont plusieurs sont colocalisées avec des Meta-QTL et/ou avec des zones identifiées avec les notations morphologiques. La densification en marqueurs de 6 zones associées avec les différentes notations d'infestation, localisées sur les chromosomes 1A, 1D, 2A, 2B, 5A et 7A, ont permis de confirmer ces régions, de restreindre fortement l'intervalle d'intérêt et de mettre en évidence de potentiels gènes candidats. Cette analyse a ainsi permis de mieux comprendre les mécanismes génétiques et morphologiques impliqués dans la résistance à la fusariose des épis. La décomposition en trois partie de cette résistance (résistance globale, résistance à la progression des symptômes, et résistance à l'accumulation des mycotoxines) montre l'existence de mécanismes génétiques indépendants et donc complémentaire à prendre en compte dans une stratégie de création de variétés résistantes à la fusariose des épis. / Fusarium head blight (FHB), caused by the fungal pathogen Fusarium spp., is a major cereals disease that not only cause direct yield losses, by interrupting grain filling, but also indirect quality losses by the production of mycotoxins, responsible of industrial processes disturbance and food poisoning. Most wheat breeding programs in FHB-affected areas of the world have been screening germplasm to identify sources of improved tolerance. Unfortunately, these sources of genetic resistance are often of exotic origin and not adapted to West European growing conditions. The selection of adapted varieties with improved tolerance therefore needs a better characterization of resistance mechanisms. Several QTL for FHB resistance in wheat have been identified in European germplasm, but the use of these information in marker-assisted selection is constrained by the precision of the QTL and the low diversity tested by using biparental population.The aim of the present study is to use association mapping, also called linkage disequilibrium mapping, to identify loci involved in the resistance to FHB. This method refers to the analysis of statistical associations between genotypes determined in a collection of individuals, and the phenotypes of the same individuals. A dense genetic map was compiled to localize precisely the association results. Resistance to F. graminearum was studied in a panel of 195 elite wheat varieties by the evaluation of three components: resistance to global infection, resistance to symptom progression and resistance to accumulation of mycotoxins. Morphological factors (plant height, heading date, awnedness, spikelets per ears, anther extrusion), known to influence resistance to FHB, were also recorded. All the varieties have been genotyped with around 3300 SNP markers, 200 SSR markers and 1400 DArT markers. We first investigated the structure of the panel, which could generate bias in the estimate of allele effects, if not included explicitly in the association models. We showed that the structure is based on geographical origin (French, German and UK). Study of the linkage disequilibrium (LD) showed an extent of LD between 2 and 6 cM. Results of association studies permitted to identify several loci for each of the evaluated components of resistance. Some of these loci colocalized with the results of the MetaQTL analysis and/or with loci associated with morphological traits. We selected more specifically 6 loci, located on chromosome 1A, 1D, 2A, 2B, 5A and 7A. Marker saturation of the regions, allowed to confirm the genome wide association results and to increase the accuracy of the loci of interest. This analysis allowed to better understand the many factors that influenced FHB resistance, whether genetic or morphological. Results show that the genetic mechanisms are independent between the three components and therefore, information obtained for each component are to be used complementary to create varieties with increased resistance to FHB.

Towards new roles for cytochrome P450s and strigolactones in Fusarium Head Blight of Brachypodium distachyon / Vers de nouveaux rôles pour les cytochromes P450 et les strigolactones dans la fusariose des épis de Brachypodium distachyonChangenet, Valentin 01 October 2018 (has links)
La fusariose des épis est l’une de maladies les plus dommageables des céréales tempérées et est principalement causée par le champignon toxinogène Fusarium graminearum (Fg). Ces dix dernières années, de nombreuses études ont rapporté l’induction transcriptionnelle de gènes de la plante codant pour des cytochromes P450 (P450) en réponse l’infection par Fg. Les P450s constituent une famille enzymatique impliquée dans de nombreuses voies métaboliques, certaines avec des intérêts potentiels dans la résistance face aux maladies. Nous avons utilisé la petite graminée modèle Brachypodium distachyon (Bd) pour caractériser fonctionnellement le premier gène codant pour un P450 induit chez la plante au cours de la fusariose des épis par l’utilisation de lignées altérées dans la séquence ou l’expression du gène Bradi1g75310 codant le P450 BdCYP711A29. Nous avons montré qu’en plus d’être un facteur de sensibilité à la maladie, le gène Bradi1g75310 est impliqué dans une voie de biosynthèse hormonale chez Bd, celle des strigolactones (SLs). En effet, en plus de complémenter génétiquement les phénotypes aériens de la lignée mutante max1-1 d’Arabidopsis thaliana altérée dans le gène homologue MAX1 (AtCYP711A1), une lignée de Bd surexprimant Bradi1g75310 (lignée OE) exsude davantage d’orobanchol, une SL spécifique, que la lignée sauvage ou mutante. Une analyse préliminaire de l’impact direct de l’orobanchol sur la croissance de Fg semble indiquer une activation des étapes précoces du développement du champignon (germination) qui pourrait être à l’origine de l’induction plus rapide de gènes de défenses observée chez une lignée OE de Bradi1g75310. Nous avons également montré que les 4 paralogues de Bradi1g75310 chez Bd, qui codent également pour des CYP711A, sont tous capable de complémenter la lignée max1-1 et avons généré du matériel végétal fondamental pour la poursuite de l’étude de la diversification des SLs chez la plante monocotylédone modèle Bd. Au global, ce projet constitue une première étape dans la caractérisation de l’implication des P450 dans la réponse de la plante face à l’infection par Fg en plus de donner de nouveaux indices concernant le rôle des SLs dans les interactions plante-pathogène. Les résultats obtenus au cours de ce travail de thèse pourront permettre l’amélioration de caractères tant développementaux que de résistance à la fusariose chez les céréales cultivées. / Fusarium Head Blight (FHB) is one of the most important diseases of temperate cereals and is mostly caused by the toxin producing-fungus Fusarium graminearum (Fg). This last decade, several studies reported the transcriptional activation of cereal cytochrome P450-encoding genes (P450s) in response to Fg infection. P450s constitute an enzymatic family participating in very diverse metabolic pathways with potential interest for disease resistance. We used the model temperate cereal Brachypodium distachyon (Bd) to functionally characterize the first FHB-induced P450- encoding gene using Bd lines altered in the locus or gene expression of the Bradi1g75310 gene encoding the BdCYP711A29 P450. We showed that in addition to be a plant susceptibility factor towards the disease, the Bradi1g75310 gene is involved in the hormonal biosynthetic pathway of strigolactones (SLs) in Bd. Indeed, in addition to genetically complement the shoot phenotypes of the Arabidopsis thaliana mutant line for the homologous gene MAX1 (AtCYP711A1, max1-1 line), a Bd linewhich overexpresses the Bradi1g75310 gene (OE) exudes more orobanchol, a specific SL, compared to wild-type or mutant lines. Preliminary analysis of the direct impact of orobanchol on Fg growth suggests an activation of early fungal development (germination) likely to induce faster induction of defense-related genes during FHB, observed in Bradi1g75310 OE line. We showed that the four paralogs of Bradi1g75310 encoding BdCYP711A P450s are all able to genetically complement max1-1 line and provide important plant material for studying SLs diversification in the model monocot B. distachyon. Overall, this project constitutes a first step in the characterization of P450s involvement in plant response towards Fg infection in addition to give new evidences about the role of SLs in plant-pathogen interactions. Results obtained during this Ph.D. project will allow the improvement of both developmental and FHB-related traits in cereal crops.

La fusariose est une des maladies les plus importantes altérant le blé en Bourgogne. L’espèce fongique Fusarium graminearum est l’un des agents principaux de la maladie. L’interaction hôte-pathogène peut entrainer la production de mycotoxines toxiques pour l’homme et l’animal.La seule alternative pour prévenir le développement de la maladie est de contrôler l’inoculum primaire dans son habitat naturel : les adventices, le sol et les résidus de culture. En raison de la tendance à la réduction du travail du sol, une attention particulière doit être portée au rôle des résidus de culture dans la survie et le développement de F. graminearum. Dans ce travail de thèse, deux approches ont été choisies pour mieux comprendre le développement saprotrophe du champignon et ses conséquences. La première, à travers un essai en champ dans le contexte pédoclimatique Bourguignon, avait pour but de définir la part relative des différentes sources d’inoculum dans le développement de la fusariose et l’accumulation des mycotoxines dans les grains. Cet essai devait en outre permettre de déterminer si des indicateurs précoces du développement de la maladie sur épi et de l’accumulation de mycotoxines pouvaient être identifiés. La seconde, à travers un essai en microcosmes, avait pour but de suivre le développement de F. graminearum dans le sol et les résidus.Ce travail a permis de souligner l’importance de la gestion des résidus de culture dans le développement de la fusariose du blé. Favoriser une décomposition biologique rapide des résidus de culture et/ou introduire des cultures intermédiaires assainissantes constituent des perspectives de recherche sur lesquelles doivent porter nos efforts / Fusarium Head Blight (FHB), mainly caused by the fungal species Fusarium graminearum, is one of the most important disease altering wheat crops in Burgundy. Moreover the plant-pathogen interaction leads to the production of mycotoxins potentially toxic for humans and animals.The only alternative to date to prevent the development of the disease is to control the saprotrophic development of F. graminearum in its natural habitat, i.e. weeds, soil and crops residues. Due to the trend of reduced tillage, special attention should be paid to the role of crop residues in the survival and development of F. graminearum.Two approaches were chosen to better understand the saprotrophic development of F. graminearum and its consequences towards FHB. i) The first through a field experiment in the Burgundian pedoclimatic context aimed at defining the relative importance of the different sources of inoculum in the development of FHB and the accumulation of mycotoxins in grains. The field experiment was also to determine whether early indicators of disease development on ears and accumulation of mycotoxins could be identified. ii) The second, through test microcosms, was to follow the development of F. graminearum in the soil and crop residues.This work highlighted the importance of crop residues management in the development of FHB and gave new understanding about the survival of the fungus on these residues. Improve the biological decomposition of crop residues at the soil surface or/and using suppressive intermediate crops could be the next prospective to investigate to limit the soil inoculum potential of saprotrophic F. graminearum

O presente trabalho avaliou a atividade antifúngica in vitro de óleos essenciais de espécies de Piper ricos em fenilpropanóides frente a Fusarium solani f. sp. piperis, agente causal da fusariose. A inibição do crescimento micelial pelo método de difusão em Agar na concentração de 5 mg.Ml-1 foi considerada: baixa para P. aduncum (20,3%) e P. krukoffii (31,4%); moderada para P. callosum (55,7%) e P. marginatum (70,3%) e alta para P. divaricatum (93,3%). Os componentes majoritários identificados por CG-EM foram: dilapiol (92,0%), safrol (78,0%), metileugenol (75,2%) e eugenol (7,9%), apiol (80,0%), Z-isoosmorizol (44,0%) e E-anetol (22,0%), respectivamente. O óleo de P. divaricatum e seus compostos majoritários apresentaram valores CIM de 0,75 mg.mL-1. A avaliação dos efeitos combinados do eugenol e metileugenol apontou o eugenol como principal responsável pela atividade. / The culture of black pepper (Piper nigrum L.) is one of the main agricultural activities in Pará State and it suffers serious damages caused by fusariosis, disease restrict in Brazil. This paper evaluated the antifungal activity in vitro of essential oils of species of Piper rich in phenylpropanoid before the Fusarium solani f. sp. piperis, which causes the fusariosis. The inhibition of mycelial growth by the method of agar dissemination in concentration of 5 mg.mL-1 was considered: decrease to P. aduncum (20.3%) and P. Krukoffii (31.4%); moderated to P. callosum (55.7%) and P. marginatum (70.3%) and high to P. divaricatum (93.3%). The major components identified by CG-EM were dilapiol (92.0%), safrole (78.0%), methyleugenol (75.2%) and eugenol (7.9%), apiol (80.0%), Z-isoosmorizol (44.0%) and E-anethole (22.0%), respectively. The oil of P. divaricatum and its major compounds present CIM values of 0.75 mg.mL-1. The evaluation of the effects combined of eugenol and methyl eugenol pointed eugenol as the main responsible by the activity.
