O presente estudo teve como objetivo realizar a triagem de PHA sintases numa biblioteca metagenômica de solo de Mata Atlântica, empregando duas técnicas de busca: o método de detecção fenotípica e a técnica de PCR. Resultados positivos foram obtidos com este último, empregando iniciadores descritos na literatura e iniciadores descritos neste trabalho. Em 10.67% dos clones da biblioteca foram obtidos amplicons, dos quais 7 foram seqüenciados, apresentando similaridade para os genes phaC dos tipos II e IV. Adicionalmente, 67 clones positivos para a classe III foram obtidos e 4 destes foram seqüenciados. Duas das seqüências obtidas mostraram alta similaridade com o gene codificador da enzima glutamina sintetase tipo I, outro deles para a proteína hipotética conservada pertencente à família de enzimas oxidoredutases e a outra para o componente D do gene hidrogenase-4. A partir da análise dos resultados, iniciadores mais específicos são propostos. Assim, a técnica de PCR foi mais eficiente na detecção de genes da biossíntese de PHA na biblioteca metagenômica estudada. / To perform a PHA synthase screening in a metagenomic library from Atlantic forest soil two search methods were applied: phenotypic detection and PCR. Positive results with PCR were obtained by using primers described in the literature and proposed in this study. Amplicons were obtained in 10.67% of the library, 7 of them were sequenced showing similarity with class II and IV phaC genes. In addition, 67 positive clones for class III were obtained and 4 of them were sequenced. Two of these sequences showed high similarity to the glutamine synthase gene type I, the third one showed similarity to the conserved hypothetical protein of the reductase family, and the forth presented similarity to the component D of the hidrogenase-4. According to the results, more specific primers are suggested. Therefore, PCR was more efficient in the detection of PHA biosynthesis genes in the studied metagenomic library.

Previous issue date: 2016-04-28 / Dentre os enfoques que podem ser dados aos estudos climáticos destaca-se a análise rítmica, que por considerar a sequência diária com que os elementos climáticos se manifestam, assim como os sistemas atmosféricos e suas correlações com o ambiente, permite uma análise integrada do clima. Esta, vêm contribuindo para que a sociedade se torne menos vulnerável às intempéries climáticas, à medida que considera tanto a sucessão habitual dos tipos de tempo quanto os eventos extremos em suas análises diárias. Partindo-se da necessidade de se conhecer essa complexa relação entre clima - ambiente – sociedade, o presente estudo teve por objetivo fazer uma investigação acerca do ritmo climático, através da quantificação da participação dos sistemas atmosféricos e dos fatores geográficos na gênese e no ritmo das chuvas na bacia hidrográfica do Rio Preto, MG/RJ, para os anos de 2006, 2007 e 2008. A partir daí se tornou possível estabelecer correlações entre o fenômeno climático e o meio físico, identificando sua dinâmica e principais unidades espaciais e temporais, estabelecendo um modelo integrado e a criação de mapas e gráficos de ritmo que subsidiassem a compreensão do sequenciamento dos tipos de tempo. A escala temporal de analise é a diária e em anos padrão (MONTEIRO, 1971, 1991) e a metodologia de escolha desses foi com base em técnicas estatísticas (box plot, desvio padrão, Sturgers e quantil). Os mapeamentos foram realizados no software ArcGIS 10, inserindo informações obtidas junto ao Hidroweb (ANA) e imagens de satélite (LANDSAT e SRTM). A construção dos gráficos de ritmo foi feita a partir de dados meteorológicos coletados no site Hidroweb da ANA, de analise sinótica de cartas sinóticas da Marinha do Brasil e dos boletins Climanalise. Como resultados, constatouse que os três anos estudados apresentaram características distintas quanto a distribuição espacial e temporal das chuvas, havendo maior destaque na alternância do regime destas. Em termos de participação dos sistemas atmosféricos é a mTa a que atuou no maior número de dias para os três anos e em 2008 (ano úmido) a atuação da ZCAS foi mais que o dobro dos demais anos (19%). Sob a atuação desta última, em quase 100% dos dias constatou-se a presença de chuvas. Também em dias sob atuação das frentes frias constatou-se a presença de chuvas. Os eventos extremos não seguiram um padrão de distribuição espacial. / Among the approaches that can be given to climate studies stands out the rhythmic analysis, what is set as the daily sequence with which the climatic elements manifest themselves, as well as the atmospheric systems they relate with the environment, providing an analysis integrated the climate. This contributed to what the society be less vulnerable to climatic conditions, as it considers both the usual succession of weather types as extreme events in their daily analysis. Starting from the need to understand the complex relationship between climate - environment - society, this study aimed to make an investigation about the climate pace, by quantifying in the participation of atmospheric systems and geographical factors in the genesis and rhythm of rain in the river watershed Preto, MG / RJ, for the years 2006, 2007 and 2008. Thenceforth, it became possible to establish correlations between the climatic phenomenon and the physical environment, identifying your dynamics and main spatial and temporal units, to establishing an integrated model and the creating maps, rhythm graphics that subsidize the understanding of the sequencing of the types of time. The temporal scale of analysis is daily and in years "pattern" (MONTEIRO, 1971, 1991) and the methodology of choice of these was based on statistical techniques (box plot, standard deviation, Sturgers and quantile). The maps were made in the ArcGIS 10 software by entering information obtained from the Hidroweb (ANA) and satellite imagery (Landsat and SRTM). The construction of the rhythm graphics was made from collected meteorological data in Hidroweb (ANA) website, synoptic analysis of synoptic maps of the Navy of Brazil and Climanalise newsletters. As a result, it was found that the three years studied showed different characteristics as the spatial and temporal distribution of rainfall, with greater emphasis on the annual alternation of these arrangements. In terms of participation of atmospheric systems is the Atlantic Tropical mass that acted in the greater number of days and in 2008 (wet year) the performance of South Atlantic convergence zone was more than double the other years (19%). Under the action of the latter, nearly 100% of the days found for the presence of rain. Also in action on day of cold fronts is it found the presence of rain. Extreme events have not followed a pattern of spatial distribution.

A study of 1 Peter 3:18- 4:6 : an investigation into the historical background of the doctrine of Christ’s descent into HadesDu Toit, Marietjie 11 August 2008 (has links)
The aim of this study is to prove that neither 1 Peter 3:19 nor 1 Peter 4:6 refers to the Christian doctrine the ‘Descensus Christi ad Inferos’. The meaning of these two verses has long been debated (cf. Dalton 1989:27-28), and is very often seen as a reference to Christ’s descent into Hades (cf. Feinberg 1986:309). This study will be done by means of a parallel study. The first part of this study will involve the doctrine of the ‘Descensus’; looking at its origin and its development. It will be argued in this section that the roots of this doctrine can be found in Jewish-Christianity and not pagan mythology as has been suggested (cf. Bousset 1907:224ff&Beare 1945:145). The discussion of the doctrine is necessary, since we do need to know more about the doctrine to see whether it is the referent in 1 Peter. The second part of the study will then engage in the meaning of 1 Peter 3:19 and 4:6. This section will be very context driven. We will start with an introduction to 1 Peter, discussing all the preliminary questions (i.e. author, date, audience&form). This will be followed by a structural analysis of 1 Peter. Here it will be argued that the letter should be understood in terms of metaphors, with the ‘Diaspora’ as the controlling metaphor (cf. Martin 1992). The verses under discussion, form part of the third metaphor-cluster namely the ‘Sufferers of the Dispersion’, while the name of our subsection is’ The Righteous Sufferer’. The Light, for Two Narrators and Chamber EnsembleFeezell, Mark Brandon 05 1900 (has links)
The Light is a twenty-four minute composition for two narrators and chamber orchestra. The two narrators perform the roles of the Apostle John and Moses. After an overview of the piece and a brief history of pieces incorporating narrators, the essay focuses on my compositional process, describing how orchestration, drama, motive, and structure work together in the piece. The Light is organized as a series of five related scenes. In the first scene, God creates light. In the second scene, God places Adam and Eve into the Garden of Eden to tend it, allowing them to eat from any tree except the tree of the knowledge of good and evil. The serpent appears, Adam and Eve succumb to his evil influence, and God banishes them from the Garden of Eden. Many generations have passed when Scene Three begins. Moses relates a story from Israel's journey in the wilderness after leaving Egypt. The people had become frustrated with Moses and with God. When God sent serpents among them as punishment, they appealed to Moses to pray for them. God's answer was for Moses to make a bronze serpent and place it on a pole. Whoever looked at the serpent would live. In Scene Four, John relates his vision of final redemption. New Jerusalem descends from heaven, with the River of Life and the Tree of Life ready to bring healing to the nations. Sadly, some people are not welcomed into the city, and the drama pauses to give respectful consideration to their fate. Finally, the fifth scene celebrates the eternal victory over sin, death, and the serpent of Eden. As I composed The Light, I had in mind the dramatic profile, the general motivic progression and the fundamental structural progression. However, most of the intricate interrelationships among orchestration, drama, motive, and structure were the result of informed intuition. Throughout the piece, each of these four elements interacts with the others, sometimes influencing and sometimes responding to them. My hope is that these subtle tensions propel the composition forward toward its ultimate resolution.

'Spirited bodies' as a prerequisite for an earth-keeping ethos : a juxtaposition on the first creation story of Genesis with ubuntu cosmogonyNalwamba, Kuzipa January 2013 (has links)
Multidisciplinary contemporary discourse involving science, philosophy and theology has explored themes of creation and human identity. Contemporary critiques of anthropocentricism stem from such discourse. The understanding of human beings as ‘spirited bodies’ rather than embodied spirits, arises from a non-reductionist physicalist standpoint. This is the point of departure for this thesis. The study attempts to explore the understanding of human beings as ‘spirited bodies’ from a non-reductionist physicalist view and as a metaphor for ‘fresh’ perspectives and insights that could potentially inform and/or shape a theologically grounded earth-keeping ethos on a different premise from the traditional dualistic hierarchical viewpoint.
Methodologically, this study attempts to reflect a unitary approach to knowledge. The study views the subject through three prisms. Firstly it takes a retrospective look to account for perspectives that have shaped hierarchical views of creation based on a dualistic principle that in turn have shaped the human power-dominion relationship with the rest of creation that is deemed to have led to the devastating eco-crisis the world faces today. Secondly, it considers a non-reductionist physicalist viewpoint that has challenged dualistic anthropological views of being in favour of the conception of human beings as ‘spirited bodies’ and which places human beings in a continuum with the rest creation. Thirdly, it picks up on Moltmann’s Trinitarian and pneumatological views of creation which orient the theological framework anchored on the community and communion within the triune relationship. Human solidarity with the rest of creation is then posited as the nexus that converges the strands of these different perspectives. The juxtaposition of the Genesis 1 creation story with Zambian cosmogony constitutes ‘case studies’ that illustrate how the fresh perspectives on creation and human identity open up an ‘interpretive space’ that could locate human beings in a continuum with the rest of creation and offer insight for an alternative earth-keeping ethos. Jephthah's Daughter: A Hebrew Foreground for Jesus' Passion in MatthewSeariac, Hanna Elizabeth 12 August 2022 (has links)
While there are key differences between Jephthah's daughter's story and that of Jesus, not least the difference in gender of the sacrificial victims, this thesis posits that both the similarities and differences between these two accounts can enrich readings of Jesus' death in the gospel of Matthew. A careful comparison of the narrative of Jephthah's daughter with Jesus' Passion narrative in the gospel of Matthew leads to the conclusion that Jesus' death should be interpreted as a human sacrifice. Reading Jesus' death as a human sacrifice and locating it in that socio-religious context makes his death indicative of a transactional, covenantal relationship between him and the Father. These two accounts also share archetypes that come from the Hebrew Bible. Foregrounding Jesus' narrative with Jepthah's daughter's narrative intricately reveals Jesus' connections with the Hebrew Bible, shedding light on the interpretation of his Passion.

The object matter of the study is psychological correlates of hardiness in adolescents.
The master's thesis consists of an introduction, two chapters, conclusions, a list of references (95 sources) and an annex including the blank forms of the applied methods. The volume of the master`s thesis is 122 pages, which include 14 figures and 3 tables.
The introduction reveals the relevance of the chosen research problem, the level of development of the problematics; the objective and the tasks of the study are set, the subject matter and the scope of the research are determined, the basic and additional hypotheses are formulated, the methods and the empirical base are defined, as well as the stages of the research, its scientific novelty, theoretical and practical significance of the work.
The first chapter includes a review of foreign and domestic literature on the research topic, a therminological and multidisciplinary analysis of comprehension is provided. Chapters, describing the correlations of comprehension presented.
Conclusions on the first chapter are the results of the study of theoretical material.
The second chapter is devoted to the empirical part of the study. It provides a description of the organization and methods of the study and the results obtained from all the methods used:
Also in the chapter the comparative and correlation analysis of the research results is presented.
The conclusions of Chapter 2 include the main results of the empirical study.

Analysis of Reconstructed Mine Soils on Reclaimed Coal and Mineral Sands Mined Lands in VirginiaAngel, Hannah Zoe 01 September 2022 (has links)
Post-mining land use potentials are greatly influenced by mine soil properties. I analyzed and classified coal mine soils in SW VA and mineral sands mine soils in SE VA to aid development of appropriate post-mining land use interpretations. For coal mine soils, long-term pedogenesis was assessed in sandstone (SS) and/or siltstone (SiS) spoils with and without surface amendments. For mineral sands mined lands, I evaluated their spatial variability with respect to underlying relic mining influences and rowcrop productivity. Coal mine subsoils densified as root-limiting layers formed that were not observed initially and all soils had weak pedogenic development (^Bw; cambic horizons), particularly SS derived soils. Initial rock spoil type strongly influenced coal mine soils over time (i.e., SS had coarser textures and lower subsoil pH vs. SiS). However, most soil chemical properties (e.g., pH, EC, CEC) became similar in ^A horizons over time. Fe-oxides increased, and extractable-P decreased with weathering, raising concerns for long-term P-availability. Organic amendments applied to rock spoil surfaces sustained higher total-N (sawdust and biosolids) and extractable P (biosolids). Soil carbon sequestration rates did not differ among rock types or amendments (0.16 to 0.28 Mg ha yr-1). Further, mineral sands mine soils were limited by densic contacts and short-range variability of important properties (texture) appeared related to underlying relic mining features (pits and berms). Yields in reclaimed soils were more variable than in adjacent undisturbed farmland, but mean yields were only slightly reduced for soybean in 2020. Coloration and Fe-oxide spectral indices had weak to moderate negative correlations with yield. Berm positions supported better soybean growth (three out of five sites), while pit positions were redder/darker (one out of five sites). Current Soil Taxonomy conveys anthropogenic origins at the family (e.g., spolic) and subgroup (e.g., Anthroportic) levels. Existing taxa and proposed Artesols order criteria both acknowledge effective soil depth x compaction limitations (Anthrodensic subgroup), but Artesols more effectively recognizes pedogenesis (Inceptic subgroup) and acknowledges mine soils at the highest level. The biggest management limitation for both mine soil types is root-limiting compaction; thus, future efforts should focus on defining depth ranges and taxonomic interpretations for densic contacts. / Doctor of Philosophy / Surface mining results in drastic disturbances to the original soil, land, and water features. Actual impacts vary due to local site conditions and methods used for mineral extraction and reclamation. Advancing reclamation science requires an understanding of the resulting mine soil properties and how they vary spatially and develop over time. This research program involved two very different mining scenarios and resultant mine soil types in Virginia that were both evaluated in terms of soil properties, taxonomic placements, and land use interpretations. First, I assessed Appalachian coal mine soils in southwestern Virginia following 34 years of soil development in sandstone (SS) and/or siltstone (SiS) derived overburden with and without surface amendments. Over time, rocky mine spoils transformed into weakly developed soil profiles, commonly with four to five total distinct layers (horizons), along with apparent "self-compacted" layers at depth. SS soils were coarser textured with somewhat better subsoil structure, while SiS soils were finer textured and had higher subsoil pH. However, many chemical properties were similar in the soil surface. Organic amendments and topsoil return imparted only subtle differences over time. A secondary objective was to assess mineral sands mine soils, which originate from recombined Coastal Plain sediments. Due to the nature of their wet deposition during final reclamation, these mine soils exhibited high variability across local landforms. Soil profiles over former berm positions were composed mostly of sandier materials, while pit positions had more variable materials and features. Reclaimed rowcrop yields were moderately correlated with bare soil remotely sensed variables. Soils in pit positions were redder in color due to higher clay. While yields were variable, especially in drier years, berms appeared to support better soybean growth, but results were inconsistent across fields. For both mine soil types, root-limiting compaction is their biggest impediment to productive use. Forestry is the predominant post-mining land use on former coal mines, while rowcrop production is becoming more common on former minerals sands mines. Since these two different post-mining environments present unique challenges for correctly interpreting their post-disturbance land-uses, it is important to convey their special properties (e.g., densic contacts) and limitations in their classification.
